Abstract

The coptis root contains a plurality of active ingredients such as coptisine, jatrorrhizine and the like. Has remarkable antibacterial effect on staphylococcus aureus, streptococcus pneumoniae producing ultra-broad-spectrum-beta lactamase and other gram-positive bacteria, pseudomonas aeruginosa, escherichia coli, drug-resistant acinetobacter baumannii, neisseria gonorrhoeae and other gram-negative bacteria, candida albicans and other fungi, and can balance grease.
The centella asiatica extract is rich in terpenoid, can obviously improve the content of collagen and cell layer fibronectin (the fibronectin is widely involved in the processes of cell migration, adhesion, proliferation, hemostasis, skin tissue repair and the like), can stimulate and promote the generation of type I collagen and type III collagen, activate fibroblast genes, and improve and repair the barrier function of skin; can promote wound healing and scar repair, and reduce inflammatory reaction. In addition, centella asiatica extract can reduce the production of pro-inflammatory mediators (IL-1, MMP-1) and improve and repair the self barrier function of skin.
In another aspect, the present application provides a method for preparing the above acne-removing composition, comprising the steps of:
(1) Drying radix Scutellariae, cortex Phellodendri, rhizoma Coptidis, and herba Centellae, and pulverizing to obtain plant powder;
(2) Adding water, butanediol and 1, 2-pentanediol into the plant powder obtained in the step (1), extracting, and filtering.
Preferably, in the step (1), the pulverization means pulverization to 10 to 50 mesh.
Preferably, in the step (2), the mass ratio of the plant powder to the water, the butanediol and the 1, 2-pentanediol is 10-30:30-70:30-70:1-10, and more preferably 17:50:50:5.
Preferably, in the step (2), the extraction adopts a low-temperature high-pressure difference method, specifically: regulating the temperature to 30-50deg.C, pressurizing to 50-100MPa, and soaking and extracting under stirring for 30-60 min.
The low temperature and high pressure difference method is characterized in that the loss of thermosensitive components can be avoided under the low temperature condition, the solvent is easier to permeate into the plant powder under the high pressure condition, the active ingredients are rapidly dissolved in the solvent, and the test proves that the active ingredients of the extract obtained by the low temperature and high pressure difference method are higher and the effect is better.
In still another aspect, the application also provides application of the acne-removing composition in cosmetics.
Preferably, in the application, the content of the acne removing composition in the cosmetic is 2-10wt%.
Finally, the application provides an oil-control acne-removing gel, which comprises the composition.
Preferably, the oil-control acne-removing gel comprises a phase A and a phase B, wherein the phase A comprises water, allantoin, disodium EDTA, polyacrylate crosslinked polymer-6, 1, 2-pentanediol, menthol and the composition; the phase B comprises hydroxyethyl acrylate/sodium acryloyldimethyl taurate copolymer, cyclopentadimethicone, cyclohexasiloxane and dimethicone.
Further preferably, the oil control acne removal condensation comprises the following components:
the beneficial effects of the application are as follows:
(1) The acne-removing composition provided by the application has the effects of removing acne marks, removing acne scars, controlling oil and removing acne. The purpose, the action route and the mechanism of each component in the composition are different, and the components show the best synergistic effect when the dosage proportion is proper.
(2) The effective components are plant sources, have no irritation to skin, and are mild and safe.
(3) The low temperature and high pressure difference method is characterized in that the loss of thermosensitive components can be avoided under the low temperature condition, the solvent is easier to permeate into the plant powder under the high pressure condition, the active ingredients are rapidly dissolved in the solvent, and the test proves that the active ingredients of the extract obtained by the low temperature and high pressure difference method are higher and the effect is better.

In order to make the technical means, the creation features, the achievement of the purpose and the effect of the present application easy to understand, the present application will be further elucidated with reference to the specific embodiments, but the following embodiments are only preferred embodiments of the present application, not all of them. Based on the examples in the embodiments, those skilled in the art can obtain other examples without making any inventive effort, which fall within the scope of the application. In the following examples, unless otherwise specified, the methods of operation used were conventional, the equipment used was conventional, and the materials used in the examples were the same. For process parameters or conditions not specifically noted, reference may be made to conventional techniques.
In the following examples, the term "Sanhuang acne-removing agent" is a composition prepared by the application, and comprises a baical skullcap root extract, a cortex phellodendri extract, a rhizoma coptidis root extract and a centella asiatica extract, and is abbreviated as "Sanhuang" because of containing the baical skullcap root extract, the cortex phellodendri extract and the rhizoma coptidis root extract.
Example 1
A preparation method of a Sanhuang acne-removing agent comprises the following steps:
(1) Drying radix Scutellariae, cortex Phellodendri, rhizoma Coptidis and herba Centellae, wherein the mass ratio of radix Scutellariae, cortex Phellodendri, rhizoma Coptidis and herba Centellae is 1:10:1:1; pulverizing to 50 mesh to obtain plant powder;
(2) Adding water, butanediol and 1, 2-pentanediol into plant powder, wherein the mass ratio of the plant powder to the water, the butanediol and the 1, 2-pentanediol is 17:50:50:5; mixing, placing into an extraction kettle, adjusting temperature to 30deg.C, pressurizing to 50MPa, soaking under stirring, extracting for 60 min, centrifuging, collecting supernatant, and filtering with 1000 mesh filter cloth.

Test example 1 Propionibacterium acnes-resistance effect
Propionibacterium acnes is the primary pathogen responsible for causing acne. Antibacterial experiments to verify the antibacterial effect of the composition of the application by using propionibacterium acnes as a bacterial model, antibacterial performance determination is performed on the compositions of examples 1-3 and comparative examples 1-7 according to GB15979-2002 "antibacterial Performance test method for dissolution antibacterial (antibacterial) products of Disposable sanitary articles sanitary Standard" appendix C4 ".
1) Test sample: the compositions described in examples 1-3 and comparative examples 1-7 were diluted to 2% dilutions, respectively.
2) Preparation of bacterial suspension: the propionibacterium acnes 24h culture is washed by 0.03mol/L phosphate buffer solution to prepare 104-106CFU/mL bacterial suspension.
3) 1g of test sample is respectively taken, 0.1mL of the bacterial suspension is added for uniform coating and mixing, timing is started, 9mL of 0.03mol/L phosphate buffer solution is added after 20min for full mixing, after proper dilution is carried out, 3 dilutions are taken, 1mL of the solution is respectively taken and placed in two parallel plates, and then the corresponding culture medium is added for culturing for 24 hours under the anaerobic condition at 36+/-1 ℃ for colony counting. The above test was repeated 3 times, averaged, and a blank control group was set.
The calculation formula of the bacteriostasis rate is as follows: x= (a-B)/a×100%, where X is the antibacterial rate, a is the average recovered colony of the blank control sample, and B is the average recovered colony of the test sample.
The efficacy evaluation criteria were: the antibacterial rate is more than or equal to 90 percent, which is obvious in effect; (2) the bacteriostasis rate is 40-90% and the effect is general; and (3) the antibacterial rate is less than 40 percent, which is no effect. The results are shown in the following table.

As can be seen from Table 1, the compositions of examples 1-3 have remarkable antibacterial effects on Propionibacterium acnes, and the antibacterial effects of comparative examples 1-7 are general; experiments show that the radix scutellariae, cortex phellodendri, rhizoma coptidis and centella asiatica can obviously inhibit the proliferation of propionibacterium acnes when being combined and used by proper proportions, and the inhibition effect is better than that of the single use of the components; can effectively prevent or improve acne symptoms of skin.

Test example 2 oil control and acne removal test
The oil control acne-removing gel prepared in examples 4 to 6, comparative examples 8 to 13 and blank control group was subjected to an evaluation of the oil control acne-removing efficacy. The blank was different from example 6 in that it did not contain a three-yellow anti-acne agent.
The oil control effect tester is a skin grease tester. The working principle of the adhesive tape is that a special adhesive tape is adopted, so that grease on the surface of the skin cuticle can be absorbed rapidly, the transmittance of the adhesive tape can be changed, and the grease content of the skin cuticle can be obtained by measuring the transmittance of the adhesive tape before and after use through a photometer built in an instrument.
The testing method comprises the following steps: 100 volunteers with oily skin and acne are summoned, and the volunteers are divided into 10 groups of 10 people, wherein one group uses oil control acne-removing gel of a blank control group, three groups use the oil control acne-removing gels prepared in examples 4-6 respectively, and six groups use the oil control acne-removing gels prepared in comparative examples 8-13 respectively, and the oil control acne-removing gel is used once a day in the morning and evening, the use amount is 0.2 g/time, and the oil control acne-removing gel is smeared on the face and is gently massaged until absorption. The fat content of the forehead part of the volunteer was measured with a skin fat tester every other week, each part was tested 3 times, and the average value was taken, and specific test records are shown in table 2. The acne removal condition is recorded every other week, the acne number is reduced by more than 1/3 by comparison with the photo before the product is used, and the specific test record is shown in the table below.

From tables 11 and 12, the oil-control acne-removing gel prepared by the embodiment of the application has good oil-control acne-removing effect; and the effects of acne removal of the baikal skullcap root, the cortex phellodendri, the rhizoma coptidis root and the centella asiatica are better than those of acne removal of the baikal skullcap root, the cortex phellodendri, the rhizoma coptidis root and the centella asiatica when the baikal skullcap root, the cortex phellodendri and the rhizoma centella asiatica are combined and used by proper proportions.

The oil control acne-removing gel prepared in examples 4 to 6 and comparative examples 8 to 13 and the oil control acne-removing gel prepared in the blank control group and the commercial acne-removing products of comparative examples 14 to 15 were subjected to acne mark and acne scar removal efficacy evaluation, wherein the blank control group is different from example 6 in that the three-yellow acne-removing agent is not contained.
The testing method comprises the following steps: 120 volunteers with new acne marks and acne scars (the acne marks and the acne scars are generated within one month) are summoned, the volunteers are divided into 10 groups, 10 people are respectively used in one group, the oil control acne-removing condensation of a blank control group is used, the oil control acne-removing condensation prepared in examples 4-6 is respectively used in three groups, the oil control acne-removing condensation prepared in comparative examples 8-13 is respectively used in six groups, the commercial acne-removing products in comparative examples 14-15 are respectively used in two groups, the use amount is 0.2 g/time in the morning and evening, and the volunteers are smeared on the face and gently massaged until absorption. Comparison was made by volunteers for signs of acne marks and scars after one month of use of the product, and specific test recordings are shown in the following table.

From 13 and 14, the acne-removing conditioning essence (containing salicylic acid) of comparative example 14 has better acne mark removing effect and poorer acne mark removing effect; the centella asiatica acne-removing paste of comparative example 15 has good acne-removing effect and poor acne mark-removing effect; the oil-control acne-removing gel prepared by the embodiment of the application has good acne mark removing and acne scar removing effects, and has better acne removing effects than the independent use of each component.
Test example 4 irritation test (Patch test)
Test sample: the oil-control acne-removing gel prepared in examples 4-6 and the commercial acne-removing products of comparative examples 14-15.
The subject: 30 volunteers (each test sample used the same batch of subjects) were selected;
the test method comprises the following steps: a closed test is adopted, 0.02g of test sample is placed in a patch device, the patch device is applied to the bent side of the arm of a tested object by using a hypoallergenic adhesive tape, the test area is at least 5X 5cm, the arm of the tested object is straightened when the patch device is applied, the test sample is applied from the lower part to the upper part, the adhesive tape is slightly pressed by the palm to discharge air, the patch device is tightly adhered to the skin for 24 hours, and a blank control group is a test sample which is not used.
Observation results: after removing the patch device for 30min, the patch device was interpreted according to the following interpretation standard, and the statistical test results are shown in the following table.
Interpretation criteria: (one) negative reaction; suspicious reaction: only mild erythema; (+) weak positive: erythema, infiltration, and possibly small papules; (++) strong positive: erythema, infiltration, papules, blisters; (+ ++ + electrode) strong positive: erythema, infiltration, papules, blisters and bullae.

As is clear from the table, the oil-control acne-removing gel prepared in the examples 4-6 has low skin sensitization, and no positive reaction is detected in 30 subjects, so that the composition has no stimulation to skin, mild ingredients and safety.
In summary, the three-yellow acne-removing agent compositions obtained in examples 1 to 3 have more outstanding skin oil secretion reduction and better acne-removing effects than the comparative examples, and especially the effect of example 3 is most ideal; experimental results show that the composition can show the optimal oil control and acne removal effects through proper proportion collocation combination, and simultaneously has the optimal acne mark removal and acne scar removal effects. The analysis shows that the efficacy of the composition is due to the fact that the purpose of action, the action route and the mechanism of each component in the composition are different, and the components show the best synergistic effect when the dosage proportion is proper; and experiments prove that the composition has no stimulation to skin, mild ingredients and safety.

1. An acne-removing composition with acne mark and acne scar removing effects is characterized by comprising radix scutellariae extract, cortex phellodendri extract, rhizoma coptidis extract and centella asiatica extract in a mass ratio of 1-10:1-10:1-10:1-10;
the preparation method comprises the following steps:
(1) Drying radix Scutellariae, cortex Phellodendri, rhizoma Coptidis, and herba Centellae, and pulverizing to obtain plant powder;
(2) Adding water, butanediol and 1, 2-pentanediol into the plant powder obtained in the step (1), extracting, filtering,
in the step (2), the extraction adopts a low-temperature high-pressure difference method, and specifically comprises the following steps: regulating the temperature to be 30-50 ℃, pressurizing to be 50-100MPa, soaking and extracting for 30-60 minutes under stirring, wherein the mass ratio of the plant powder to water, butanediol and 1, 2-pentanediol is 10-30:30-70:30-70:1-10.
2. The acne-removing composition according to claim 1, wherein the composition comprises the raw materials of scutellaria root, cortex phellodendri, coptis root and centella asiatica in a mass ratio of 5:3:3:6.
3. The method for preparing the acne-removing composition according to any one of claims 1 to 2, comprising the steps of:
(1) Drying radix Scutellariae, cortex Phellodendri, rhizoma Coptidis, and herba Centellae, and pulverizing to obtain plant powder;
(2) Adding water, butanediol and 1, 2-pentanediol into the plant powder obtained in the step (1), extracting, filtering,
in the step (2), the extraction adopts a low-temperature high-pressure difference method, and specifically comprises the following steps: regulating the temperature to be 30-50 ℃, pressurizing to be 50-100MPa, soaking and extracting for 30-60 minutes under stirring, wherein the mass ratio of the plant powder to water, butanediol and 1, 2-pentanediol is 10-30:30-70:30-70:1-10.
4. The method according to claim 3, wherein in the step (2), the mass ratio of the plant powder to water, butanediol, and 1, 2-pentanediol is 17:50:50:5.
5. Use of an anti-acne composition according to any one of claims 1-2 or prepared according to the preparation method of any one of claims 3-4 for the preparation of an anti-acne cosmetic.
6. The use according to claim 5, characterized in that the acne-removing composition is present in the cosmetic in an amount of 2-10wt%.
7. An oil-controlling acne-removing gel characterized by comprising the acne-removing composition according to any one of claims 1 to 2 or the acne-removing composition prepared by the preparation method according to any one of claims 3 to 4.
8. The oil-controlling anti-acne gel according to claim 7, comprising a phase a and a phase B, wherein the phase a comprises water, allantoin, disodium EDTA, polyacrylate crosslinked polymer-6, 1, 2-pentanediol, menthol and the anti-acne composition of any one of claims 1-2 or the anti-acne composition prepared according to the preparation method of any one of claims 3-4; the phase B comprises hydroxyethyl acrylate/sodium acryloyldimethyl taurate copolymer, cyclopentadimethicone, cyclohexasiloxane and dimethicone.
9. The oil control acne-removing gel according to claim 8, wherein the oil control acne-removing gel comprises the following components:
