Description

Method for promoting generation of new water-cultured Chinese chestnut roots
Technical Field
The invention relates to the field of Chinese chestnut cultivation, in particular to a method for promoting the generation of new water-cultured Chinese chestnut roots.
Background
At present, most of basic scientific research experimental materials of Chinese chestnuts adopt tissue culture nursery stocks of solid culture media or nursery stocks cultured by grass beach vermiculite in fields, and have a plurality of limitations in the aspect of scientific research treatment. On the one hand, many researches in the aspect of the Chinese chestnut root system need to take out the root system from the growing solid medium, but the solid medium of the root system growth is relatively hard, easily damages and stresses the root system, destroys the integrity of the root system, and can influence the physiological characteristics and biochemical indexes of the root system to a certain extent. On the other hand, in the salt and alkali stress treatment test of the Chinese chestnut, the Chinese chestnut root systems at different growth stages need to be set with different salt stress concentrations, pH values and different plant hormones for treatment, and the development of different test treatments needs to completely and completely return the Chinese chestnut root systems growing to a certain stage to a solid growth medium, so that the practical operation is basically infeasible. In addition, the components of the solid medium for the root growth of the Chinese chestnut and the environmental regulation are relatively difficult. These all greatly limit the progress of basic scientific research on chestnuts.
At present, no relevant report about the success of Chinese chestnut water culture exists, and mainly due to the growth habit, the physiological characteristics, the special environmental adaptability and the like of Chinese chestnuts, other plant water culture technical systems cannot be applied to Chinese chestnuts. Therefore, how to provide a water culture technology suitable for Chinese chestnuts, in particular to a method for promoting the generation of new water culture roots of Chinese chestnuts is a problem to be solved.

Table 2 shows the influence of pH value on chlorophyll content of chestnut leaves
Figure BDA0003248396130000072
Table 3 shows the influence of pH on the activity of chestnut root system
Figure BDA0003248396130000081
Table 4 shows the influence of pH on the malondialdehyde content in the roots and leaves of Castanea mollissima

Table 5 shows the effect of pH on SOD activity of chestnut roots and leaves
Figure BDA0003248396130000083
Table 1 shows that the high pH value severely inhibits the growth and development of Chinese chestnuts, the growth of overground part tissues and underground part tissues is inhibited, and particularly the growth amount of underground part root systems is severely inhibited. Table 2 shows that under the stress of high pH value, the content of chlorophyll a, chlorophyll b and carotenoid of the chestnut is reduced. Table 3 shows that root vigor is significantly reduced under high pH stress and is more severe with stress time. As can be seen from tables 4 and 5, the malondialdehyde content of the chestnut roots and leaves was increased under stress conditions, while the SOD activity was increased. Therefore, the growth of the Chinese chestnut is remarkably inhibited under the stress of high pH value.
