Description

Asparagus polyphenol moisturizing skin care product and preparation method thereof
Technical Field
The invention belongs to the technical field of cosmetics, and particularly relates to a asparagus polyphenol moisturizing skin care product and a preparation method thereof.
Background
The skin separates the internal environment of the human body from the external environment, and is similar to a protective wall constructed between the internal environment of the human body and the external environment, so that the stability and the balance of the internal environment of the human body are kept. The skin can be divided into epidermis, dermis, subcutaneous tissue and skin appendage, wherein the epidermis is located at the outermost layer of the skin and sequentially comprises a basal layer, a spinous layer, a granular layer, a transparent layer and a cuticle from inside to outside.
Skin moisturizing is a basic nursing work, and the skin moisturizing is well done, so that the skin aging can be delayed, and the occurrence of skin diseases can be reduced and prevented. Skin moisturization depends primarily on the water content of the stratum corneum, which should be maintained at 10% to 20% for healthy skin, and when the water content of the stratum corneum is sufficient, the skin is soft, smooth, tender and elastic, and less than 10% of the skin is dry, rough and even chapped. The protection and maintenance of stratum corneum moisture relies on the barrier and hydration of the epidermis. The barrier function of the skin relies primarily on the "brick wall structure" of the stratum corneum. The 'brick wall structure' of the horny layer is composed of 'brick walls' (keratinocytes) and 'mortar' (lipid in the gaps between the keratinocytes), and the compact and firm structure can block the invasion of external harmful factors and the loss of water and nutrient substances in the body and maintain the important barrier function of the epidermis. In addition, the lipid (ceramide, squalene, linoleic acid, linolenic acid, etc.) in the sebum membrane on the skin surface can lock water, prevent loss of nutrients, moisturizing factors and water, maintain the water content of the stratum corneum, and play a role in moistening and moisturizing the skin. Hydration of the skin epidermis is mainly dependent on Natural Moisturizing Factors (NMF) in the keratinocyte matrix. NMF is a small molecular compound with water absorption property existing in skin, is formed by amino acid assembly formed by degradation of filaggrin, can effectively absorb and lock water, and mainly comprises amino acid, lactate, urea and other unknown substances. The natural moisturizing factor and the compact keratin cellulose increase the strength of the inside of the cell, and simultaneously combine a large amount of water to ensure that the stratum corneum keeps a certain water content, which is an essential condition for stabilizing the hydration state of the skin and maintaining the normal physiological function of the stratum corneum. The inherent hydration ability of skin depends on hyaluronic acid and moisture transport within the skin, in addition to the presence of NMF in keratinocytes. The proteins in the epidermis of human skin, which play a role in water transport, are mainly Aquaporin 3 (AQP 3), AQP3 can transport water and substances such as urea and glycerol into and out of the skin, and is a key factor for maintaining skin hydration.
In the related art, the cosmetics can achieve the effects of moisturizing water and moisture by adding the moisture-keeping wetting agent. The moisturizing humectant is similar to NMF, human protein, polysaccharide and the like in the epidermis, and when the moisture of the skin is lost, the moisturizing humectant enters the skin through pores, or carries water, or wraps up the water which is not lost in the skin to prevent the water from being lost, so that the water in the skin is retained.
The plant components are natural in source and rich in effect, and are widely used in food and medicines. In cosmetics, plant ingredients can be used in place of various functional materials or as auxiliary materials. The addition of plant components in the product can conform to the idea that consumers adopt nature, so more and more merchants begin to develop products with the center of gravity which is inclined to plant skin care.
Gracilaria lemaneiformis is temperate red algae and is mainly distributed in coastal areas such as Shandong, Fujian and Guangdong. In recent years, researches on asparagus are mostly focused on saccharides, such as asparagus polysaccharide, asparagus oligosaccharide, agar prepared by using asparagus, and the like, while related researches on asparagus polyphenol are few and mainly focused on the antioxidant capacity, and at present, researches or reports on the moisture absorption and retention performance of asparagus polyphenol are not found.
The above-mentioned contents are only the techniques known by the inventor, and are only used for assisting understanding of the technical scheme of the invention, and do not represent an admission that the above-mentioned contents are the prior art.

Firstly, extracting asparagus polyphenol in asparagus by an ethanol reflux method:
(1) weighing appropriate amount of thallus Gracilariae dry powder, and placing into a container (such as round bottom flask);
(2) adding 70% ethanol into a container, wherein the liquid-solid ratio of the ethanol to the dry powder of the asparagus is 50 mL/g;
(3) heating the container to 80 ℃, and refluxing for 2h for extraction; wherein the average extraction rate of asparagus polyphenol is 2.20 +/-0.04 mg/g of dry asparagus.
Next, preferred examples of the present invention will be described in detail, and experimental methods in which specific conditions are not specified in the examples are generally conventional conditions.
Example 1
Determination of asparagus polyphenol in vitro moisture absorption capacity:
accurately weighing 0.5g of each of the asparagus polyphenol and glycerol which are dried to constant weight at room temperature of about 20 ℃, respectively putting the asparagus polyphenol and glycerol into a culture dish, putting the culture dish into a constant-temperature and constant-humidity environment with the temperature of 20 ℃ and the Relative Humidity (RH) of 81%, standing for a period of time (1, 2, 4, 8, 12, 24 and 48 hours), accurately weighing the mass of each sample, and calculating the moisture absorption rate. Wherein the moisture absorption rate is (m)1-m0)×100%/m0In the formula, m0To weigh the mass of the sample before moisture absorption, m1For the quality of the sample after moisture absorption, data analysis is carried out by using SPSS 12.0 statistical software, and if the moisture absorption rate P is less than 0.05, the difference is considered to be significant.
As can be seen from fig. 1, in both the asparagus polyphenol and the humectant glycerin commonly used in skin care products, the moisture absorption rate gradually increases with time under a high relative humidity (81%) environment. The moisture absorption rates of asparagus polyphenol and glycerol are similar at 1h, and the moisture absorption capacity of asparagus polyphenol is obviously higher than that of glycerol (P is less than 0.05) at 2h and 4 h. After 8h, the moisture absorption capacity of the glycerol is higher than that of asparagus polyphenol. In real life, the frequency of face cleaning is often several hours, so that the moisture absorption capacity of asparagus polyphenol is stronger than that of glycerin at 2h and 4h, thereby showing the feasibility of the asparagus polyphenol as a skin care product moisture absorbent.
Example 2
Testing the in-vitro moisturizing capability of asparagus polyphenol:
weighing 0.5g each of the asparagus polyphenol and glycerol which are dried to constant weight at room temperature of about 20 ℃, adding the asparagus polyphenol and glycerol into a weighing bottle with the diameter of 3cm, adding 0.2g of deionized water, and placing the weighing bottle in a dryer filled with silica gel. The standing time is 1, 2, 4, 8, 12, 24 and 48 hours, the mass of each sample is accurately weighed, and the moisture retention rate is calculated. WhereinMoisture retention rate ═ m1×100%/m0In the formula, m0To add the amount of water, m1The data analysis is carried out by SPSS 12.0 statistical software for the moisture content after the sample is placed, and the difference is considered to be significant if the moisture retention rate P is less than 0.05.
As can be seen from fig. 2, both the asparagus polyphenol and the glycerin gradually decreased the moisture retention rate with time. The moisturizing rates of asparagus polyphenol and glycerol are similar at 1h and 2h, and after 4h, the moisturizing rate of asparagus polyphenol is higher than that of glycerol (P is less than 0.05).
Example 3
Influence of asparagus polyphenol on human skin moisture change rate (detection of asparagus polyphenol in vivo moisture retention capacity):
the influence of the asparagus polyphenol water solution with the mass fraction of 1% and 3% on the change rate of the skin moisture of a human body is measured by a human body method, and water is used as a negative control. 26 healthy female volunteers without skin diseases are selected, aged between 18 and 25 years, and tested by a CM825 type skin moisture tester under the conditions that the ambient temperature is 22 ℃ and the relative humidity is 60 percent. Any cosmetics or external medicines can not be used for 2 to 3 days before the test of the tested part, and water can not be contacted for 1 to 3 hours before the test. Before the test, the testee needs to clean the inner sides of the forearms of the hands uniformly, and the cleaning method is to wipe the forearms clean by using dry facial tissue. The inner sides of the forearms of both hands of the testee are marked with measuring areas, the area of the testing area is 4cm multiplied by 4cm, the same arm can simultaneously mark a plurality of areas, and each testing area is at least 1cm apart. Before formal test, people should sit still in a room meeting the standard for more than 20min without drinking water and beverages. The forearms were exposed and left in the test condition to remain relaxed. The test sample was evenly spread over the test area using a latex finger cuff at a rate of 2.0mg/cm 2. The skin moisture content was measured before and 30, 60, 90, 120, 150, 180, 210, 240min after sample application, and the skin moisture change rate was calculated. The skin moisture content testing operation is completed by the same person, and the skin moisture change rate calculation formula is as follows:
rate of change of skin moisture ═ mn-m0)×100%/m0
In the formula: m isnRepresents the measurement value at the n hour after the sample is coated; m is0The measured values before coating the sample are shown. Data analysis was performed using SPSS 12.0 statistical software, and differences were considered significant if the skin moisture change rate P < 0.05.
As can be seen from FIG. 3, the rate of change of skin moisture caused by the application of the aqueous solution of Gracilaria lemaneiformis polyphenol, whether the aqueous solution is a 1% or 3% solution, is significantly higher than that of the application of blank control water (P < 0.05), thereby indicating that the Gracilaria lemaneiformis polyphenol has better moisturizing performance.
