Description

Sebum biomimetic composition and cosmetic containing same
Technical Field
The invention relates to the technical field of cosmetics, in particular to a sebum biomimetic composition and a cosmetic containing the same.
Background
The skin of a human body is damaged from outside at any moment, and due to dryness, aging, improper skin care products or excessive cleaning, the sebum on the surface of the skin is produced too little or is deprived in a large amount, so that the barrier function of the skin is reduced, the moisture retaining capacity of the skin is reduced, and dryness, sensitivity, pruritus and the like are easily caused. Meanwhile, harmful bacteria can also increase and propagate to cause infection, the cell differentiation capacity is affected, the cutin is not regularly arranged, subcutaneous collagen is lost, and various skin problems are caused. Research shows that most skin problems are caused by the damage of the barrier function of the skin, the barrier function of the horny layer is damaged, the tolerance or the defense capability of the skin is reduced, and the skin problems of dry skin, fragile and sensitive skin, easy external infection and the like are caused.
The skin epidermis barrier is formed by skin surface sebum membrane, intercellular lipid forming brick wall structure, dermis mucopolysaccharide, mucopolysaccharide and other components, and resists the entry of harmful and irritants outside, wherein the sebum is used as the outermost layer component of the skin, and the main components of the sebum comprise triglyceride, free fatty acid, wax ester, squalene, cholesterol ester, cholesterol and other components, and the skin epidermis barrier is vital to the function of the skin barrier, and the skin epidermis barrier mainly has the following functions: 1. the water loss of the skin is prevented, and a lipid film is formed on the surface of the skin by the lipid layer, so that the horny layer is soft and moist, excessive evaporation of water in the skin can be prevented, and the moisture-keeping effect is achieved; 2. the damage of ultraviolet rays to the skin is reduced, and as the outermost layer of the skin, the integrity of the sebum layer plays a vital role in reducing the damage of the ultraviolet rays to the skin; 3. protecting against biological attack, triglyceride secreted from sebum layer is decomposed under the action of esterase to generate free fatty acid (such as undecylenic acid) which can inhibit the growth and reproduction of Staphylococcus aureus, hemolytic streptococcus, Candida albicans, etc. The sebaceous layer therefore plays a crucial role in the skin barrier function.
At present, a variety of skin barrier function repairing cosmetics are provided, inflammation of damaged skin is reduced by enhancing the moisture retention of the skin surface layer or adding a soothing component, and the effects of barrier function damage and sebum loss cannot be fundamentally solved.
CN105748315B discloses a composition for repairing skin barrier, which is prepared from the following raw materials by weight: 0.1-1% of sodium hyaluronate, 0.5-3% of trehalose, 5-25% of humectant and 71-94.4% of water. Experiments prove that the composition for repairing the skin barrier can effectively relieve the symptoms of red swelling, hot pain, skin fading, pruritus and the like caused by skin after the beauty means such as laser beauty, tartaric acid skin changing and the like are treated, has good absorption effect, simple preparation method, short production period, simple prescription and low cost, but can only play the roles of relieving and moisturizing, does not fundamentally repair the skin barrier and reduce the loss of sebum.
CN108685741A discloses a composition for promoting skin barrier repair and an application thereof, wherein the composition for promoting skin barrier repair comprises the following components in parts by weight: 10-20 wt% of trehalose, 10-30 wt% of sodium hyaluronate, 5-15 wt% of centella asiatica extract, 5-15 wt% of spirulina polysaccharide, 5-15 wt% of collagen, 5-20 wt% of aloe extract, 1-5 wt% of ceramide, 1-5 wt% of sodium polyglutamate, 1-2 wt% of ascorbyl glucoside and 0.5-2 wt% of acetyl chitosamine. The composition prepared by the invention can be used for promoting the repair of the damaged skin barrier, and is particularly suitable for promoting the repair of the damaged skin barrier after laser beauty treatment, but the composition only can play the roles of moisturizing, relieving and diminishing inflammation, and the problem of damaged skin barrier is not fundamentally solved.
CN108498371A discloses a composition with skin barrier repairing effect and its preparation method, wherein the raw materials in the composition include mannitol and glycosphingolipids, and the composition is further researched to be: mannitol, glycosphingolipids, glycerophosphoinositide choline salt, tetrahydromethylpyrimidine carboxylic acid, oligopeptide-1; researches show that the composition has good effect of repairing skin barriers, but the composition only can temporarily keep moisture and relieve the skin, and does not fundamentally repair the skin barriers and slow the loss of sebum.
Therefore, in view of the fundamental problem of repairing skin barrier, those skilled in the art should develop a composition and cosmetic, which not only has better effects of relieving inflammation, but also has better effects of repairing skin barrier and promoting cell regeneration.

Then adding 1mL of sample to be tested (namely the sebum bionic composition and the single-component natural vegetable oil provided in the examples and the comparative examples) and 0.4mL of 25mmol/L pyrogallol solution, mixing uniformly, reacting for 5min in a water bath at 25 ℃, and adding 1.0mL of 8mol/L hydrochloric acid to terminate the reaction. Absorbance values were measured at 299nm with Tris-HCl buffer as a reference.
Blank control experiment: only 1mL of ethanol was used in place of the test sample, and the other steps were not changed.
The clearance (D) was calculated according to equation (1).
D(%)=[1-(A2/A1)]×100% (1)
Wherein A1 is the absorbance value of the blank control; a2 is the absorbance value of the sample to be tested.
Test example 2
Measurement of hydroxyl radical scavenging ability
A sample to be tested: sebum biomimetic compositions and one-component natural vegetable oils of examples 1-10 and comparative examples 1-10.
The specific test method is as follows:
3mL of FeSO with the concentration of 2mmol/L are sequentially added into a 25mL colorimetric tube43mL of H with a concentration of 1mmol/L2O2Shaking, adding 3mL of salicylic acid 6mmol/L, shaking, heating in 37 deg.C water bath for 15min, and measuring absorbance; respectively adding samples to be tested (namely the sebum bionic composition and the single-component natural vegetable oil provided in the examples and the comparative examples) with certain concentration, shaking up, continuing to heat in water bath for 15min, and taking out to test the absorbance.
Blank control experiment: only 1mL of ethanol was used in place of the test sample, and the other steps were not changed.
The formula (2) is a formula for calculating the clearance rate of the sample to be tested to the carboxyl free radical (OH).
OH(%)=(A3-A2)/A1×100% (2)
In the formula, A1 is the absorbance value of the reaction system before adding the sample to be tested; a2 is the absorbance value of the system after OH is removed from the sample to be tested; a3 is the absorbance value of the system after blank control is cleared of OH.
The test results of test examples 1 and 2 are shown in table 4.
TABLE 4
Figure BDA0001970777100000201
Figure BDA0001970777100000211
As can be seen from Table 4, the sebum-mimicking compositions of examples 1 to 10 all had better radical-scavenging ability, while the effects of the examples were not achieved using either single-component natural vegetable oils (comparative examples 1 to 5) or compositions lacking one component (comparative examples 6 to 10).
Comparing examples 1-10, it can be seen that optimizing the formulation and selecting five natural vegetable oils in a specific ratio combination further improves the free radical scavenging ability of the composition.
Test example 3
Skin prick test
A sample to be tested: the sebum-stimulating composition, the single-component natural vegetable oil and the cosmetic provided in examples 1 to 16 and comparative examples 1 to 13 were each blended.
The specific test mode is as follows:
100 volunteers (50 women 50 men) were selected to evaluate the effect of the composition on resisting itch and red swelling of skin caused by histamine, and histamine hydrochloride (10mg/mL) was dropped on the skin test site with a test blade on the inner side of forearm of the subject, and after 5 minutes, the skin developed red swelling and itching. The test samples to be tested are respectively smeared, and the relieving effects of different samples on skin itch and red swelling caused by histamine resistance are compared.
Blank control: the only difference from the above test method is that nothing is coated, waiting for the skin to repair itself.
Evaluation method of skin redness and swelling degree: visual assessment of skin redness on 5 point scale, 0: non-red and non-swollen, 1: slight red swelling, 2: marked redness, 3 severe redness, 4 very severe redness; the average of 9 volunteers was calculated.
The test results are shown in tables 5 and 6.
TABLE 5
Figure BDA0001970777100000221
Figure BDA0001970777100000231
As can be seen from Table 5, the samples of examples 1 to 10 were excellent in the effect of relieving the red swelling and itching caused by histamine, as compared with the blank control and the comparative examples, in which the effect of example 1 was most remarkable, and the red swelling and itching condition was substantially eliminated 2.5 hours after application, while the effects of examples 1 to 10 could not be achieved by using one component of the natural vegetable oils (comparative examples 1 to 5) or the compositions lacking one component (comparative examples 6 to 10).
Comparing examples 1-10, it can be seen that optimizing the formulation and selecting five natural vegetable oils in a specific ratio combination can further enhance the efficacy of the composition in combating the red swelling and itching associated with histamine.

Example 16 4.01 2.60 2.11 1.70 1.61 1.10
As can be seen from Table 6, the samples provided in examples 11-16 have obvious effects of relieving and repairing skin irritation and redness caused by histamine, particularly, the samples of example 11 have obvious effects of relieving skin redness and swelling stimulated by histamine after being applied to the samples for half an hour, the skin redness and itching are relieved, the discomfort basically disappears after 2.5 hours, and the blank control and comparative examples 11-13 have obviously poorer test results than the samples of examples 11-16 and still have redness and swelling and itching to a certain extent after 2.5 hours, so that the sebum biomimetic composition provided by the invention can provide good relieving and repairing effects for cosmetics after being added to the cosmetics.
It is understood from comparative examples 11 to 15 that the effect is better when the sebum-mimicking composition is added to the cosmetic in an amount of 0.01 to 30%, and that the effect is deteriorated when the amount is too large (example 15) or too small (example 14).
Fig. 1 is a graph comparing the improvement effect of the samples of example 11 and comparative example 11 and the blank sample against the symptom of skin itch caused by histamine, and it is apparent that the moisturizing cream of example 11 has better soothing and repairing effects.
Test example 4
