Description

Whitening and moisturizing skin care product
Technical Field
The invention belongs to the technical field of skin care products, and particularly relates to a whitening and moisturizing skin care product.
Background
Tyrosinase is a key enzyme for melanin synthesis, melanin cells are distributed among basal layer cells of human epidermis, and the tyrosinase contained in the melanin can oxidize tyrosine into polysaccharide, and finally melanin can be generated through a series of metabolic processes. The more melanin production, the darker the skin; otherwise, the whiter the skin. If the activity of tyrosinase can be inhibited, the synthesis of melanin can be inhibited, and the whitening effect can be achieved. The tyrosinase inhibitor can be used for treating pigmentation dermatoses such as freckle, chloasma, and senile plaque.
Hesperetin (hesperetin), chemical formula C16H14O6302.29 molecular weight, light yellow needle crystal, melting point 228 deg.C, is one of flavanones, and is the main effective component of fruit of Citrus in Rutaceae. Research has shown that the compound has various medicinal activities, such as treating bone diseases related to diabetes, reducing blood fat, reducing permeability of capillary, resisting allergy and dermatitis, and the like. In addition, hesperetin also has effects of resisting oxidation and scavenging free radicals. In addition to the above effects, hesperetin also has tyrosinase activity inhibiting effect, so that the compound has effectsHas theoretical basis for application in whitening cosmetics. However, at present, the application of hesperetin in whitening skin care products is less, and the compounding of hesperetin and other skin care ingredients is more rarely studied.

(I) detection method
1. Whitening effect
The whitening effect of the skin care product is evaluated by detecting the inhibition effect of the skin care product on tyrosinase activity. The lower the residual activity of tyrosinase, the better the inhibition effect of the skin care product on the tyrosinase activity and the better the whitening effect.
The test is to measure the inhibition of tyrosinase and the activity of tyrosinase by the sample by ultraviolet spectrophotometry (analysis is performed by the selective absorption of light by substances). The experiment is carried out by using tyrosinase and dopa to form oxidation product dopachrome under the catalysis of enzyme in an ultraviolet region, wherein the dopachrome has maximum absorption at 475nm, and the absorption coefficient epsilon is 3700(cm & mol/L).
Using a hand-held pipette, 1.0mL of 2 mmoL/L-dopa and 2.6mL of a phosphate buffer (pH 6.8) were pipetted into cuvettes. The mixture is kept in a constant temperature water bath at 30 ℃ for 10min, and then 0.2mL of the skin care product (sample) obtained in the embodiment 1-6 and 0.2mL of a tyrosinase solution with the concentration of 100 mu g/mL are added to form a 4mL solution system. The absorbance of the solution system was measured at a wavelength of 475nm as a function of time over 8 min. The tyrosinase residual activity (U) is calculated as follows:
U=[(OD1-OD3)/OD2-OD4]×100%
in the formula, OD1The absorbance of a living test system containing a substrate, tyrosinase and a sample solution is measured; OD3The absorbance of a living system only containing a substrate and a sample solution is measured; OD2The absorbance of a activity measuring system only containing a substrate and a tyrosinase solution; OD4Refers to the absorbance of the assay system containing only the substrate solution.
2. Moisture absorption and retention effects
The moisture absorption and retention efficiency of the skin care product is evaluated through the moisture absorption rate and the moisture retention rate of the skin care product, and the higher the moisture absorption rate and the moisture retention rate is, the better the moisture absorption and retention effect is.
Will contain saturated (NH)4)2SO4The inner cavity of the dryer for the aqueous solution is used as an environment with a relative humidity of 81% (RH: 81%), and saturated K is filled in the dryer2SO4The inner cavity of the dryer of the aqueous solution was set to an atmosphere with a relative humidity of 43% (RH ═ 43%). 0.5g of the skin care products (samples) of examples 1 to 6 dried to a constant weight were accurately weighed in small weighing bottles and placed in the 2 dryers described above. Placing 2 dryers in a constant temperature environment of 20 ℃, standing for 24 hours, weighing the mass of each sample, and calculating the moisture absorption rate:
moisture absorption rate (m)2-m1)/m1×100%
m1Is the initial mass of the sample, m2Is the sample mass after 24 h.
Accurately weighing 0.5g of the skin care product of the embodiment 1-6 dried to constant weight, adding deionized water with the mass of 10% of the sample, placing the mixture in a small weighing bottle, and placing the small weighing bottle and the small weighing bottle in the 2 dryers respectively. Placing 2 dryers in a constant temperature environment of 20 ℃, standing for 24 hours, weighing the mass of each sample, and calculating the moisture retention rate:
moisture retention rate m4/m3×100%
m3Is the initial moisture mass of the sample, m4Is the moisture mass of the sample after 24 h.
(II) the result of the detection
The results of the skin care products of examples 1-6 for whitening and moisturizing are shown in table 1:
table 1 test results of whitening and moisturizing effects of skin care products in examples 1 to 6
Figure BDA0001593257020000081
From the above results, the skin care products in embodiments 1 to 6 of the present invention all have good whitening and moisturizing effects, can effectively inhibit tyrosinase activity, and have good hygroscopicity and moisturizing properties. Among them, the skin care product in example 6 had the best whitening and moisturizing effects.
Finally, it should be noted that the above embodiments are only used for illustrating the technical solutions of the present invention and not for limiting the protection scope of the present invention, and although the present invention is described in detail with reference to the preferred embodiments, it should be understood by those skilled in the art that modifications or equivalent substitutions can be made on the technical solutions of the present invention without departing from the spirit and scope of the technical solutions of the present invention.
