Abstract
本发明涉及微生物制剂技术领域,具体涉及一种抗氧化和抗衰老的益生菌组合物及其制备方法,包括以下质量份数的原料乳杆菌5~15份、双歧杆菌5~15份、鼠李糖乳杆菌5~15份、植物乳杆菌5~15份、嗜酸乳杆菌5~15份、番茄10~30份、木瓜10~30份、中药添加剂5~10份、β胡萝卜素2~7份、载菌溶液20~40份、氯化钙溶液80~100份、辅料添加剂10~20份。本发明制得的益生菌组合物维持肠道菌落环境平衡,改善氧化应激、较少炎症,抑制病原菌,维持新陈代谢活性,有利于细胞清除自由基,维持自由基的平衡,提高免疫力,延长寿命,增效发挥抗氧化、抗衰老的效果。

技术领域
本发明涉及微生物制剂技术领域,具体涉及一种抗氧化和抗衰老的益生菌组合物及其制备方法。
背景技术
衰老是指机体对环境的生理和心理适应能力进行性降低的现象。由于内在或外在的原因,或随着年龄的增长,身体新陈代谢减弱,身体内自由基逐渐增多,细胞清除自由基的能力逐步下降,各种损伤积累,直接引发人体衰老。除此之外,饮食不健康也是加速人体衰老的重要原因之一。抗衰老旨在延长人类高质量的生命及健康长寿为目标,随着生活水平的提高,人们对防治衰老、延年益寿和提高生命活力有更加迫切的要求。
衰老的自由基学说是能指导抗衰老实践的学说之一,氧自由基是皮肤与身体加速衰老的第一杀手,抗氧化也即抗氧化自由基成为减缓衰老的有效途径。衰老过程往往伴随着多种慢性疾病的发生,而这些疾病中很多与肠道菌群失调有关。研究发现,肠道菌群通过参与氧化应激、炎性反应、免疫反应、代谢过程等多种途径参与了衰老的进程。因此,肠道微生物是影响衰老相关生活质量的重要指标之一。补充合适的益生菌可以减轻炎症,维持肠道内微生物平衡,在一定程度上可以延缓机体的衰老,增加个体的寿命。然而,目前市售的抗衰老的益生菌中含有众多化学物质或者重金属,长时间使用,会危害人类的皮肤健康,抗衰老和抗氧化作用差,功效单一、生物利用度方面不高,仍有待改进。
发明内容
本发明的目的在于提供一种抗氧化和抗衰老的益生菌组合物及其制备方法,维持肠道菌落环境平衡,改善氧化应激、较少炎症,抑制病原菌,维持新陈代谢活性,有利于细胞清除自由基,维持自由基的平衡,提高免疫力,延长寿命,增效发挥抗氧化、抗衰老的效果。
为了达到上述目的,本发明提供如下技术方案:
一种抗氧化和抗衰老的益生菌组合物,包括以下质量份数的原料:
所述抗氧化和抗衰老的益生菌组合物的制备方法,包括以下步骤:
步骤一:取质量份数的番茄、木瓜,洗净并去皮切块,用30~50℃的温水浸泡番茄和木瓜3~5min,捞出后,立即放入榨汁机中进行榨汁,得到混合榨汁;
步骤二:取质量份数的中药添加剂,放入超高速打粉机中,打成粉末,过筛200目,中药混合物;
步骤三:将步骤一中得到的混合榨汁、步骤二中得到的粉末中药混合物加入到发酵罐中,再分别加入质量份数的植物乳杆菌、嗜酸乳杆菌,进行发酵,设置发酵温度为30~40℃,发酵5~48h,过滤颗粒物,得到混合悬浊液;
步骤四:取步骤三中得到的混合悬浊液,再分别加入质量份数的乳杆菌、双歧杆菌、鼠李糖乳杆菌、β胡萝卜素、辅料添加剂,混合均匀后,将其加入到载菌溶液中,水合1~2h后,用注射器吸入并将其缓慢挤压滴入氯化钙溶液中,固化30min~1h后,使用生理盐水洗涤并过滤得到湿基益生菌组合物颗粒,冷冻干燥后得到海藻酸钠-蛋白质微胶包覆的益生菌组合物。
所述中药添加剂为灵芝、当归、三七、山楂的混合物;质量比为灵芝:当归:三七:山楂=1:1:1:1~1:1:2:3。
所述载菌溶液为海藻酸钠、酪蛋白酸钠和乳清蛋白的混合,质量比为海藻酸钠:酪蛋白酸钠:乳清蛋白=1:1:1,所述载菌溶液进行灭菌处理,于120℃下,高温灭菌15min~30min后使用。
所述氯化钙溶液为体积分数是2%的氯化钙溶液。
所述辅料添加剂为低聚半乳糖、纯牛奶、蔗糖、人参多糖、寡肽和玉米淀粉中的一种或多种混合。
所述步骤三中,发酵前需要调整发酵罐中液体含量,向发酵罐中加入经过灭菌处理的超纯水,液面高度小于发酵罐高度的一半,发酵过程需要对发酵罐进行密封处理。
所述步骤四中,生理盐水洗涤为:固化后,立马过滤,之后向滤饼和滤纸上倒入生理盐水,继续使用生理盐水过滤2~3次,完成冲洗和过滤;冷冻干燥前需要在-4~-10℃的冰箱中预冻4h以上,过滤后在-40℃冷冻干燥。
本发明的有益效果:
1、本发明使用产生的番茄发酵物中,含有多种维生素和活性物质,具有良好的抗衰老、抗氧化、降低胆固醇的作用;木瓜发酵物中含有还原糖、氨基酸、黄酮等活性物质,能够增强免疫力、抗氧化、美容的作用;同时,对中药添加剂物质的发酵,不仅保留灵芝、当归、三七、山楂中的抗氧化和抗衰老物质,同时增加益生菌的含量,有利于发挥益生菌保健作用和充分发挥番茄和木瓜中的营养物质对抗氧化和抗衰老的作用。
2、β胡萝卜素和辅料添加剂中的低聚半乳糖、蔗糖等,能够协助益生菌对维持肠道菌落平衡,维持新陈代谢活性,有利于细胞清除自由基,维持自由基的平衡,缓解衰老的发生。
3、载菌溶液的使用,载菌溶液可包覆益生菌、中药添加剂、番茄和木瓜中有利于缓解衰老的活性物质,益生菌在经过胃液时,可缓慢释放,能够保证进入体内的益生菌的活性,益生菌的代谢活动可通过清除氧化剂化合物或阻止其在肠道中的生成发挥抗氧化作用,最终保证益生菌组合物抗氧化和抗衰老的作用。
4、本发明采用乳杆菌、双歧杆菌、鼠李糖乳杆菌相互配合、相互促进,共同维持肠道菌落环境平衡,改善氧化应激、较少炎症、抑制病原菌、提高免疫力、延长机体衰老,延长寿命。

下面将对本发明的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有作出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1
一种抗氧化和抗衰老的益生菌组合物,包括以下质量份数的原料:乳杆菌5份,双歧杆菌5份,鼠李糖乳杆菌5份,植物乳杆菌5份,嗜酸乳杆菌5份,番茄10份,木瓜10份,中药添加剂5份,β胡萝卜素2份,载菌溶液20份,氯化钙溶液80份,辅料添加剂10份;
所述抗氧化和抗衰老的益生菌组合物的制备方法,包括以下步骤:
步骤一:取质量份数的番茄、木瓜,洗净并去皮切块,用30℃的温水浸泡番茄和木瓜3min,捞出后,立即放入榨汁机中进行榨汁,得到混合榨汁;
步骤二:取质量份数的中药添加剂,放入超高速打粉机中,打成粉末,过筛200目,中药混合物;
步骤三:将步骤一中得到的混合榨汁、步骤二中得到的粉末中药混合物加入到发酵罐中,再分别加入质量份数的植物乳杆菌、嗜酸乳杆菌,进行发酵,设置发酵温度为30℃,发酵5h,过滤颗粒物,得到混合悬浊液;
步骤四:取步骤三中得到的混合悬浊液,再分别加入质量份数的乳杆菌、双歧杆菌、鼠李糖乳杆菌、β胡萝卜素、辅料添加剂,混合均匀后,将其加入到载菌溶液中,水合1h后,用注射器吸入并将其缓慢挤压滴入氯化钙溶液中,固化30min后,使用生理盐水洗涤并过滤得到湿基益生菌组合物颗粒,冷冻干燥后得到海藻酸钠-蛋白质微胶包覆的益生菌组合物。
实施例2
一种抗氧化和抗衰老的益生菌组合物,包括以下质量份数的原料:乳杆菌15份,双歧杆菌15份,鼠李糖乳杆菌15份,植物乳杆菌15份,嗜酸乳杆菌15份,番茄30份,木瓜30份,中药添加剂10份,β胡萝卜素7份,载菌溶液40份,氯化钙溶液100份,辅料添加剂20份;
所述抗氧化和抗衰老的益生菌组合物的制备方法,包括以下步骤:
步骤一:取质量份数的番茄、木瓜,洗净并去皮切块,用50℃的温水浸泡番茄和木瓜5min,捞出后,立即放入榨汁机中进行榨汁,得到混合榨汁;
步骤二:取质量份数的中药添加剂,放入超高速打粉机中,打成粉末,过筛200目,中药混合物;
步骤三:将步骤一中得到的混合榨汁、步骤二中得到的粉末中药混合物加入到发酵罐中,再分别加入质量份数的植物乳杆菌、嗜酸乳杆菌,进行发酵,设置发酵温度为40℃,发酵48h,过滤颗粒物,得到混合悬浊液;
步骤四:取步骤三中得到的混合悬浊液,再分别加入质量份数的乳杆菌、双歧杆菌、鼠李糖乳杆菌、β胡萝卜素、辅料添加剂,混合均匀后,将其加入到载菌溶液中,水合2h后,用注射器吸入并将其缓慢挤压滴入氯化钙溶液中,固化1h后,使用生理盐水洗涤并过滤得到湿基益生菌组合物颗粒,冷冻干燥后得到海藻酸钠-蛋白质微胶包覆的益生菌组合物。
实施例3
一种抗氧化和抗衰老的益生菌组合物,包括以下质量份数的原料:乳杆菌10份,双歧杆菌10份,鼠李糖乳杆菌10份,植物乳杆菌10份,嗜酸乳杆菌10份,番茄20份,木瓜20份,中药添加剂7份,β胡萝卜素5份,载菌溶液30份,氯化钙溶液90份,辅料添加剂15份;
所述抗氧化和抗衰老的益生菌组合物的制备方法,包括以下步骤:
步骤一:取质量份数的番茄、木瓜,洗净并去皮切块,用40℃的温水浸泡番茄和木瓜4min,捞出后,立即放入榨汁机中进行榨汁,得到混合榨汁;
步骤二:取质量份数的中药添加剂,放入超高速打粉机中,打成粉末,过筛200目,中药混合物;
步骤三:将步骤一中得到的混合榨汁、步骤二中得到的粉末中药混合物加入到发酵罐中,再分别加入质量份数的植物乳杆菌、嗜酸乳杆菌,进行发酵,设置发酵温度为35℃,发酵20h,过滤颗粒物,得到混合悬浊液;
步骤四:取步骤三中得到的混合悬浊液,再分别加入质量份数的乳杆菌、双歧杆菌、鼠李糖乳杆菌、β胡萝卜素、辅料添加剂,混合均匀后,将其加入到载菌溶液中,水合1.5h后,用注射器吸入并将其缓慢挤压滴入氯化钙溶液中,固化43min后,使用生理盐水洗涤并过滤得到湿基益生菌组合物颗粒,冷冻干燥后得到海藻酸钠-蛋白质微胶包覆的益生菌组合物。
以秀丽隐杆线虫为研究用动物模型,研究益生菌组合物对秀丽隐杆线虫寿命、产卵量、运动能力、热应激和氧化应激抵抗能力的影响,探究益生菌组合物的抗衰老作用,对比实施例为所用培养板使用同等剂量(与实施例1~3相同)的生理盐水,实施例1~3所用培养板使用同等剂量的益生菌组合物,在NGM培养基上培养,所有测试平行进行3次。
每组含有秀丽隐杆线虫50条,每天将其转移到新板上,20℃培养,当成虫不再产卵后,每2d挑取线虫1次,直到线虫全部死亡时停止,该时间为秀丽隐杆线虫寿命。
每组10条秀丽隐杆线虫,产卵后以5h为周期转移,直至线虫不再具有生殖能力。培养所有留有线虫的平板,统计虫30h后的成虫数量,即待测线虫的产卵量。
随机选取培养至8d的秀丽隐杆线虫,观察20s内秀丽隐杆线虫运动次数,记为运动能力。
每组30条秀丽隐杆线虫,35℃热敷16h,统计其存活情况,记为抗热激能力。
每组30条秀丽隐杆线虫,将其转入溶液浓度为20mmol/L的过氧化氢中静置2h,随后将培养基放在培养箱内20℃处理16h,每小时统计线虫存活率,记为抗氧化应激能力。
测试结果如下表:
由表可知,与对比实施例相比,按照本发明方法制备的益生菌组合物培养秀丽隐杆线虫,具有更长的寿命、更强的产卵能力,运动能力、热应激抵抗力、氧化应激抵抗能力显著增强,说明按照本发明方法制备的益生菌组合物对抗氧化和抗衰老有着积极的作用。

Claims (7)
1.一种抗氧化和抗衰老的益生菌组合物,其特征在于,包括以下质量份数的原料:

步骤二:取质量份数的中药添加剂,放入超高速打粉机中,打成粉末,过筛200目,中药混合物;
步骤三:将步骤一中得到的混合榨汁、步骤二中得到的粉末中药混合物加入到发酵罐中,再分别加入质量份数的植物乳杆菌、嗜酸乳杆菌,进行发酵,设置发酵温度为30~40℃,发酵5~48h,过滤颗粒物,得到混合悬浊液;
步骤四:取步骤三中得到的混合悬浊液,再分别加入质量份数的乳杆菌、双歧杆菌、鼠李糖乳杆菌、β胡萝卜素、辅料添加剂,混合均匀后,将其加入到载菌溶液中,水合1~2h后,用注射器吸入并将其缓慢挤压滴入氯化钙溶液中,固化30min~1h后,使用生理盐水洗涤并过滤得到湿基益生菌组合物颗粒,冷冻干燥后得到海藻酸钠-蛋白质微胶包覆的益生菌组合物。
2.根据权利要求1所述的一种抗氧化和抗衰老的益生菌组合物,其特征在于,所述中药添加剂为灵芝、当归、三七、山楂的混合物;质量比为灵芝:当归:三七:山楂=1:1:1:1~1:1:2:3。
3.根据权利要求1所述的一种抗氧化和抗衰老的益生菌组合物,其特征在于,所述载菌溶液为海藻酸钠、酪蛋白酸钠和乳清蛋白的混合,质量比为海藻酸钠:酪蛋白酸钠:乳清蛋白=1:1:1,所述载菌溶液进行灭菌处理,于120℃下,高温灭菌15min~30min后使用。
4.根据权利要求1所述的一种抗氧化和抗衰老的益生菌组合物,其特征在于,所述氯化钙溶液为体积分数是2%的氯化钙溶液。
5.根据权利要求1所述的一种抗氧化和抗衰老的益生菌组合物,其特征在于,所述辅料添加剂为低聚半乳糖、纯牛奶、蔗糖、人参多糖、寡肽和玉米淀粉中的一种或多种混合。
6.根据权利要求1所述的一种抗氧化和抗衰老的益生菌组合物,其特征在于,所述步骤三中,发酵前需要调整发酵罐中液体含量,向发酵罐中加入经过灭菌处理的超纯水,液面高度小于发酵罐高度的一半,发酵过程需要对发酵罐进行密封处理。
