背景技术
在过去10至15年间,通过在癌症基因组测序方面的努力,已经鉴定出了许多导致人类癌症的发展和维持的癌基因。尽管鉴定出了500多种经过验证的癌症基因,但三种RAS基因HRAS、NRAS和KRAS仍然构成了人类癌症中最常见的突变癌基因家族。
当RAS被输入信号“打开”时,它随后打开其他蛋白,这些蛋白最终启动涉及细胞生长、分化和存活的基因。ras基因的突变可导致产生永久激活的RAS蛋白。结果,即使在没有输入信号的情况下,这也可以导致细胞内部意外的和过度活跃的信号传导。
因为这些信号导致细胞生长和分裂,所以过度活跃的RAS信号传导最终会导致癌症。3种RAS基因(HRas、KRas和NRas)是人类癌症中最常见的癌基因。在20%至25%的所有人类肿瘤和高达90%的某些类型的癌症中,发现了永久激活RAS的突变。
在最初发现癌基因后的30多年里,具有RAS突变的癌症基本上无法治愈。因此,许多年来,RAS被认为是“不可成药性(undruggable)”。
在HRAS、NRAS和KRAS中,KRAS是最常见的突变RAS亚型,已经表明在90%的胰腺癌、45%的结肠直肠癌和35%的肺腺癌中发生突变。KRAS突变与增加的致瘤性和不良预后有关。

药物制剂

当R3为C(O)NHR3a时,即,当n3为0,Y3为O,X3为NH,m3为1,k3为0时,使用作为偶联剂的1,1-羰基二咪唑和合适的碱(例如三乙胺),将胺K(方案1)或胺N(方案2)与R3aNH2偶联,分别得到相应的脲L和脲O(WO 2015/089337)。
当R3=C1-2烷基-C(Y3)-(X3)m 3-(CH2)k 3-R3a时,即,当n3为1或2时,使用Cl-C1-2-烷基-C(Y3)-(X3)m 3-(CH2)k 3-R3a或Br-C1-2-烷基-C(Y3)-(X3)m 3-(CH2)k 3-R3a和合适的碱(例如三乙胺),通过亲核取代制备胺L(方案1)或胺O(方案2)。

N-(恶唑-4-基甲基)-3,4-二氢异喹啉-2(1H)-羧酰胺
步骤1:2-(3-(苄氧基)苯基)乙醇的合成
向2-(3-羟基苯基)乙醇(2.2g,15.6mmol)在无水二甲基甲酰胺(40mL)的溶液中,添加碳酸钾(4.3g,31.1mmol)。在室温下搅拌10分钟后,添加苄基溴(1.9mL,15.6mmol),并在50℃下搅拌反应。2小时后,将反应混合物在乙酸乙酯和水之间分配。用盐水洗涤有机层,经无水MgSO4干燥,过滤并真空浓缩,得到产物,为黄色油(2.7g,77%收率)。1H NMR(400MHz,CDCl3)δppm 7.33-7.45(m,5H),7.22-7.26(m,1H),6.83-6.87(m,3H),5.06(s,2H),3.88(t,J=6.2Hz,2H),2.85(t,J=6.3Hz,2H)。

步骤1:2-(3-溴-4-甲氧基苯基)乙胺的合成

步骤11:1-(2,4-二甲基苯基)-2-((恶唑-4-基甲基)氨基甲酰基)-7-(2-(吡啶-3-基)乙氧基)-1,2,3,4-四氢异喹啉-6-羧酸盐酸盐的合成
在甲基1-(2,4-二甲基苯基)-2-((恶唑-4-基甲基)氨基甲酰基)-7-(2-(吡啶-3-基)乙氧基)-1,2,3,4-四氢异喹啉-6-羧酸盐(90mg,0.166mmol)在THF(8.3mL)和水(830μL)的溶液中,添加氢氧化锂(8mg,0.33mmol)。将反应混合物在室温下搅拌。添加水2小时(8mL)以稀释反应混合物后,真空蒸发有机溶剂,并通过加入1N HCl酸化含水残余物(pH=5)。用乙酸乙酯进行萃取,得到产物,为氯水合物盐(90mg,96%收率)。1H NMR(400MHz,CDCl3)δppm11.0(bs,1H),8.41-8.43(m,2H),7.95(s,1H),7.75(s,1H),7.67-7.69(m,2H),7.25(t,J=7.5Hz,1H),7.17(s,1H),6.92-6.98(m,3H),6.28(s,1H),6.01(bs,1H),4.27(t,J=7.1Hz,2H),4.10(s,2H),3.44-3.54(m,2H),2.90-3.00(m,4H),2.34(s,6H)。
步骤12:1-(2,4-二甲基苯基)-N2-(恶唑-4-基甲基)-7-(2-(吡啶-3-基)乙氧基)-3,4-二氢异喹啉-2,6(1H)-二甲酰胺的合成
将1-乙基-3-(3-二甲基氨基丙基)碳二亚胺盐酸盐(61mg,0.32mmol)和N,N-二异丙基乙胺(84μL,0.48mmol)添加到氯化铵(43mg,0.8mmol)、1-(2,4-二甲基苯基)-2-((恶唑-4-基甲基)氨基甲酰基)-7-(2-(吡啶-3-基)乙氧基)-1,2,3,4-四氢异喹啉-6-羧酸盐酸盐(90mg,0.16mmol)和1-羟基苯并三唑水合物(22mg,0.16mmol)在无水N,N-二甲基甲酰胺中的溶液中。24小时后,将反应混合物在乙酸乙酯和水之间分配。将有机层经无水MgSO4干燥,过滤并真空浓缩。将残余物通过反相色谱法纯化(乙腈/水0-100%梯度),得到标题产物,为白色固体(49mg,58%收率)。1H NMR(400MHz,CDCl3)δppm8.41-8.43(m,2H),7.95(s,1H),7.67-7.69(m,2H),7.57(s,1H),7.50(bs,2H),7.25(t,J=7.5Hz,1H),6.92-6.98(m,3H),6.28(s,1H),6.01(bs,1H),4.27(t,J=7.1Hz,2H),4.10(s,2H),3.44-3.54(m,2H),2.90-3.00(m,4H),2.34(s,6H)。

实施例3-在肿瘤细胞系中的活性
·细胞系#1:A549。携带KRasG12S致癌突变的肺癌细胞系
·细胞系#2:H358。携带KRasG12C致癌突变的非小细胞肺癌系
·细胞系#3:PANC-1。携带KRasG12D致癌突变的胰腺细胞系上皮样癌
·细胞系#4:RPMI。携带KRasG12A致癌突变的骨髓瘤细胞系
将细胞系在补充有10%FBS的DMEM或RPMI-1640中培养。为了评估化合物的抗增殖作用,将细胞分别以1.8×103、6.2×103、7.8×103、21×103和2×103个细胞/cm2的密度接种在组织培养微孔板中,并在5%CO2的潮湿环境中孵育。24小时后,添加溶于100%DMSO中的化合物,最终浓度范围在0.1至50μM之间,最终的DMSO浓度为0.5%,然后将板再孵育72小时。孵育后,按照制造商的使用说明,使用水性非放射性细胞增殖试验-MTS(Promega#G5421)对增殖进行定量。490nm的吸光度与活细胞的数量成正比。用BMGFluostar Optima微孔板读数器记录吸光度,并进行归一化与溶剂进行对照。
IC50值(μM):细胞增殖抑制
化合物1-53中显示的数据是实验结果的中位数。化合物54-63显示的数据是基于估计和/或初步实验结果。
实施例4-在小鼠异种移植中的活性
化合物18在治疗NOD/SCID小鼠皮下NCI-H358人肺癌异种移植模型中的疗效评价。
实验设计
当平均肿瘤大小达到141mm3时开始治疗。下面的实验设计表中显示了每个研究组中的测试物质给药和动物数量。

研究终点:该研究的主要终点包括以下:
肿瘤生长抑制(TGI):TGI(%)是抗肿瘤疗效的指标,表示为:TGI(%)=100×(1-T/C)。T和C分别是在给定的一天中治疗组和对照组的平均肿瘤体积。
荷瘤小鼠的体重变化结果如图1所示。组2(化合物18,10mg/kg)中只有一只小鼠的体重减轻(BWL)达到10%,而组3(顺铂,3.5mg/kg)中有4只小鼠的BWL达到10%或甚至更低。结果表明,携带皮下NCI-H358人肺癌异种移植模型的小鼠可耐受10mg/kg、每日两次的化合物18。
不同组的肿瘤生长曲线如图2所示。
接种后第24天(PG-D22,首次给药后第22天),组1(溶剂)的平均肿瘤体积达到630mm3。在PG-D22,组2(化合物18,10mg/kg)的平均肿瘤体积达到238mm3,TGI为约62%。在PG-D22,组3(顺铂,3.5mg/kg)的平均肿瘤体积达到231mm3,TGI为约63%。与溶剂组相比,组2和组3均表现出显著的抗肿瘤作用(组2:p=0.026,组3:p=0.019)。
测试化合物18在皮下NCI-H358人肺癌异种移植模型中显示出显著的抗肿瘤活性,并且10mg/kg、每日两次的化合物18对携带小鼠是安全的。
实施例5:使用表面等离子体共振测定平衡解离常数(KD)
化合物18的KD为8.8nM(Ka=1.17×105M-1·s-1;Kd=1.03×10-3s-1)
测定KD的方案如下:
在40%湿度下,将溶于水中的各种浓度的KRas手动印刷到裸露的镀金(厚度47nm)的PlexArray纳米捕获传感器芯片(Plexera Bioscience,西雅图,华盛顿州,美国)上。将每个浓度重复印刷,每个斑点包含0.2μL的KRas溶液。将芯片在80%湿度中于4℃孵育过夜,并用10×PBST冲洗10分钟,用1×PBST冲洗10分钟,用去离子水冲洗两次达10分钟。然后在使用前,在氮气流下干燥之前,将芯片用5%(w/v)的脱脂牛奶在水中封闭过夜,并用10×PBST洗涤10分钟,用1×PBST洗涤10分钟,用去离子水洗涤两次达10分钟。使用PlexAray HT(Plexera Bioscience,西雅图,华盛顿州,美国)进行SPRi测量。
准直光(660nm)穿过耦合棱镜,从SPR活性金表面反射回来,并被CCD摄像机接收。通过非脉冲式活塞泵将缓冲液和样品注入安装在耦合棱镜上的30μL流通池(flowcell)中。每个测量周期包含四个步骤:用PBST运行缓冲液以2μL/s的恒定速率洗涤以获得稳定的基线,以5uL/s注入化合物18进行结合,用PBST以2μL/s洗涤表面300s,并用0.5%(v/v)的H3PO4以2μL/s再生300s。所有测量均在4℃进行。将结合和洗涤后的信号变化(任意单位)记录为测定值。分析了SPR图像中选定的蛋白质移植区域,并将选定区域的平均反射率变化绘制为时间的函数。通过数据分析模块(DAM,Plexera Bioscience,西雅图,华盛顿州,美国)记录和分析实时结合信号。使用BIAevaluation 4.1软件(Biacore,Inc.)进行动力学分析。
实施例6:对NIH-H358细胞系的3D活力测定中的疗效测试
进行3D CellTiter-GloTM细胞活力测定的方案如下:
第1天细胞的平板接种
-用相应的培养基将细胞浓度调整至1×105个细胞/ml。(根据数据库或密度优化测定法调整细胞浓度)。将3.5mL细胞悬液和6.5mL的1%甲基纤维素混合。混合并等待气泡分散,然后移液。该步骤在0.65%甲基纤维素溶液中产生10ml细胞悬液。根据具有最终细胞密度的板图,将99.5μL细胞悬液添加到96孔板中。
-将设置两个重复的板。一个用于在第0天进行读数(T0),另一个将在培养器中培养用于在终点进行读数。
-将板在具有5%CO2的加湿培养器中于37℃孵育过夜。
第0天:T0板读数和化合物处理
-取T0板,向每个孔中加入0.5μL培养基以进行T0读数。
-在轨道式振荡器上将内容物混合2分钟,以促进细胞裂解。
-让板在室温下孵育10分钟以稳定发光信号。
-使用EnVision多标记读数仪(EnVision Multi Label Reader)记录发光。
-以“测试物质稀释”指示的浓度稀释测试物质。根据板接种图,添加0.5μL每种200X化合物工作溶液。
第7天:7天的化合物处理的板读数
-在轨道式振荡器上将内容物混合2分钟,以促进细胞裂解。
-让板在室温下孵育10分钟以稳定发光信号。
-使用EnVision多标记读数仪(EnVision Multi Label Reader)记录发光。
