Abstract
本发明涉及发酵饮料技术领域,本发明公开了一种增强免疫力的乳酸菌发酵饮料,包括药食同源中草药、蛹虫草、雨生红球藻、磷脂酰丝氨酸、果蔬、牛奶、同型发酵乳酸菌、溶菌酶、红糖、食用香精和纯化水。本发明还公开了一种增强免疫力的乳酸菌发酵饮料的制备方法,包括以下制备步骤:S1、称重;S2、臭氧清洗;S3、草药破碎;S4、菌种驯化;S5、联合破壁;S6、同型乳酸发酵;S7、离心;S8、调配灌装;S9、灭菌。本发明通过发酵产物对肠道微生态的调节,增强了人体对营养成分和功效成分的吸收效果,并通过雨生红球藻中的虾青素清除自由基,维护人体机体生态平衡,有效促进人体机能健康,增强人体的免疫力,抵御致病菌入侵。

技术领域
本发明涉及发酵饮料技术领域,具体为一种增强免疫力的乳酸菌发酵饮料及其制备方法。
背景技术
随着我国经济的快速增长,人们生活方式和生活节奏都在不断变化,生活质量和身体健康意识不断增强,但同时高压力、高强度、长时间的不健康工作和生活方式带来的身体过度透支引起的体虚、疲惫、机体免疫力弱及常伴有不同程度的失眠等问题,一直困扰着较多的人群。纵观目前市场上在售的各型各色、功能繁多的药品及保健品,虽有一定效果,但短时间只能治其表而不能固其本,长期食用可能对身体机能产生负担引发副作用且价格昂贵、食用普遍不方便。
国家推行中医“治未病”理论,提倡“药补不如食补”的健康养生方式,但一般家用基本上都是一种或少数几种中药或药食同源原料搭配煲汤或用做配料食用,食用次数和加工方式简单、有限,很难起到食补效果。而现在市场上的绝大多数功能性食品,基本都是采用中草药提取物粉,喷粉过程易导致部分营养成分流失且需添加较多糊精或其他食品添加剂方可喷粉,导致有效成分含量降低,功效不明显。
虾青素具有抗氧化、着色、抗癌及增强免疫等功能,特别是抗氧化功能超强,被誉为“超级抗氧化剂”,有着广阔的应用前景。天然虾青素更是具有重要的医疗保健价值,目前雨生红球藻中天然虾青素含量最高,但由于其细胞壁很厚,导致溶剂难以渗透到细胞里,难以直接提取。
授权公告号为CN104961668B的发明公开了一种从雨生红球藻中快速提取虾青素的方法,具体工艺为:取雨生红球藻粉,加入有机溶剂,混合成悬浊液,进防爆型高压均质机,进行破壁、萃取;将破壁后藻粉与溶剂进行分离,得到虾青素提取液及藻渣;将虾青素提取液在惰性气体保护性下浓缩并脱除溶残,得到虾青素。该发明采用有机溶剂对破壁后的雨生红球藻进行萃取,制备的虾青素存在有机溶剂,影响最终产品的品质,此外,对于取雨生红球藻的厚壁孢子而言,高压均质机的破壁效果差。
发明内容
本发明的目的在于提供一种增强免疫力的乳酸菌发酵饮料及其制备方法,以解决上述背景技术中提出的问题。
为实现上述目的,一方面,本发明提供如下技术方案:一种增强免疫力的乳酸菌发酵饮料,包括由以下重量份配比的原料制备而成:药食同源中草药9~18份、蛹虫草2~4份、雨生红球藻2~4份、磷脂酰丝氨酸2~4份、果蔬15~20份、牛奶15~20份、同型发酵乳酸菌0.01~0.05份、溶菌酶0.01~0.05份、红糖8~10份、食用香精0.1~0.5份和纯化水50~70份;
其中,所述药食同源中草药包括人参、益智仁、枸杞和黄精。
可选的,所述药食同源中草药中各组分的重量份配比为人参3~6份、益智仁2~4份、枸杞2~4份、黄精2~4份。
可选的,所述磷脂酰丝氨酸由天然软磷脂采用酶转化法制备。
可选的,所述果蔬包括蓝莓、葡萄、苹果、菠萝、番茄、黑豆、胡萝卜、韭菜、芋头、雪梨、柠檬中的一种或多种。
可选的,所述同型发酵乳酸菌包括由以下重量份配比的原料:乳链球菌2份,德氏乳杆菌2份,乳酸乳杆菌2份和保加利亚乳杆菌2份。
另一方面,本发明还提供如下技术方案:一种增强免疫力的乳酸菌发酵饮料,包括以下制备步骤:
S1、称重:按要求称取各重量份的原料;
S2、臭氧清洗:将药食同源中草药、蛹虫草、雨生红球藻和果蔬浸泡在流动的50~60℃水中3~5min,再用臭氧清洗机清洗5~10min;
S3、草药破碎:将药食同源中草药、蛹虫草和果蔬投入万能破碎机中,按照0.5~1.0cm规格进行破碎后,添加纯化水,搅拌均匀,得到混合物料A;
S4、菌种驯化:将同型发酵乳酸菌接种于牛奶培养基,待常温活化后,接入混合物料A:牛奶=1:1的培养基,于15~28℃下培养增值,得到驯化后的同型发酵乳酸菌;
S5、联合破壁:将雨生红球藻投入万能破碎机中,破碎成0.1~0.2cm的颗粒后,与牛奶、溶菌酶一同投入反应釜中,连续机械搅拌4~8h后,开启超声波,连续超声搅拌10~20min,得到混合物料B;
S6、同型乳酸发酵:将混合物料A、混合物料B、驯化后的同型发酵乳酸菌、红糖和纯化水投入发酵釜中,连续乳酸发酵6~8d,得到发酵原液;
S7、离心:将发酵原液先投入卧螺离心机进行粗分离,再投入碟片离心机进行精细分离,得到浆液;
S8、调配灌装:向浆液中投入磷脂酰丝氨酸、食用香精和纯化水,控制混合物料体系温度在70~80℃至物料完全溶解,无氧灌装,得到灌装饮料;
S9、灭菌:将灌装饮料放入水浴式杀菌锅中,于105℃恒温条件下连续灭菌20~40min,冷却至常温,即得到最终产品。
可选的,S4中,所述发酵釜的控制温度在15~28℃。
可选的,S5中,所述机械搅拌的转速控制为10~30r/min;所述超声搅拌的转速控制为90~120r/min。
与现有技术相比,本发明提供了一种增强免疫力的乳酸菌发酵饮料及其制备方法,具备以下有益效果:
1.本发明通过发酵产物对肠道微生态的调节,增强了人体对营养成分和功效成分的吸收效果,并通过雨生红球藻中的虾青素清除自由基,维护人体机体生态平衡,有效促进人体机能健康,增强人体的免疫力,抵御致病菌入侵;
2.本发明通过先用溶菌酶破坏雨生红球藻的厚壁孢子上的多糖和糖脂成分,使得雨生红球藻表面松动,再通过超声波产生的空化作用,对松动的破坏雨生红球藻表面直接破坏,使得虾青素直接流出雨生红球藻细胞并融入牛奶中,制备过程无需大量使用溶菌酶,相比超声波直接破壁,联合破壁超声作用时间短,经济效益好。

下面将对本发明实施例中的技术方案进行清楚、完整地描述,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例1:本申请实施例1提供的乳酸菌发酵饮料,包括由以下重量份配比的原料制备而成:人参3.2份、益智仁2份、蛹虫草2份、黄精2份、雨生红球藻2份、磷脂酰丝氨酸2.3份、果蔬18份、牛奶16份、同型发酵乳酸0.04份、溶菌酶0.02份、红糖9份、食用香精0.2份和纯化水60份。
实施例2:本申请实施例2提供的乳酸菌发酵饮料,包括由以下重量份配比的原料制备而成:人参5.6份、益智仁4份、蛹虫草4份、黄精3.8份、雨生红球藻4份、磷脂酰丝氨酸4份、果蔬18份、牛奶16份、同型发酵乳酸0.04份、溶菌酶0.02份、红糖9份、食用香精0.2份和纯化水60份。
实施例3:本申请实施例3提供的乳酸菌发酵饮料,包括由以下重量份配比的原料制备而成:人参4份、益智仁3份、蛹虫草3份、黄精3份、雨生红球藻3份、磷脂酰丝氨酸3份、果蔬18份、牛奶16份、同型发酵乳酸0.04份、溶菌酶0.02份、红糖9份、食用香精0.2份和纯化水60份。
其中,磷脂酰丝氨酸由天然软磷脂采用酶转化法制备;果蔬包括番茄、黑豆、胡萝卜、韭菜、芋头、雪梨、柠檬;同型发酵乳酸菌包括由以下重量份配比的原料:乳链球菌2份,德氏乳杆菌2份,乳酸乳杆菌2份和保加利亚乳杆菌2份。
本发明的乳酸菌发酵饮料由人参、益智仁、蛹虫草、黄精、雨生红球藻、磷脂酰丝氨酸、果蔬、牛奶、同型发酵乳酸、溶菌酶、红糖、食用香精和纯化水组成,将这些成分组合使其发酵生成发酵饮料。
其中,人参具有大补元气,复脉固脱,补脾益肺,生津,安神,用于体虚欲脱,肢冷脉微,脾虚食少。益智仁具有温脾止泻摄涎,暖肾缩尿固精之功效,常用于脾胃虚寒,呕吐,泄泻,腹中冷痛,口多唾涎,肾虚遗尿,尿频,遗精,白浊。蛹虫草具有既能补肺阴,又能补肾阳,主治肾虚,阳痿遗精,腰膝酸痛,病后虚弱,久咳虚弱。枸杞具有补虚益精,清热明目。黄精具有滋肾润肺,补脾益气。雨生红球藻含有丰富的虾青素,虾青素是一种优良的抗氧化剂,降低膜通透性、限制过氧化物启动子进入细胞内,从而保护细胞内重要分子免受氧化损伤,可以清除自由基,提高人体抗衰老能力。磷脂酰丝氨酸主要是改善神经细胞功能,调节神经脉冲的传导,增进大脑记忆功能,由于其具有很强的亲脂性,吸收后能够迅速通过血脑屏障进入大脑,起到舒缓血管平滑肌细胞,增加脑部供血的作用。
本发明的乳酸菌发酵饮料的制备方法,包括如下步骤:
S1、称重:对每种药食同源中草药随机抽样,参照《中华人民共和国药典(2015版)》和新食品原料检测方法对抽取的样品进行标志性成分含量检测,以确定原料真假,再筛选出发霉、变质等不符合要求的原料,按要求称取各重量份的原料;
S2、臭氧清洗:将药食同源中草药、蛹虫草、雨生红球藻和果蔬浸泡在流动的48℃水中4min,再用臭氧清洗机清洗8min;
S3、草药破碎:将药食同源中草药、蛹虫草和果蔬投入万能破碎机中,按照0.5~1.0cm规格进行破碎后,添加纯化水,搅拌均匀,得到混合物料A;
S4、菌种驯化:将同型发酵乳酸菌接种于牛奶培养基,待常温活化后,接入混合物料A:牛奶=1:1的培养基,于25℃下培养增值,得到驯化后的同型发酵乳酸菌;
S5、联合破壁:将雨生红球藻投入万能破碎机中,破碎成0.1~0.2cm的颗粒后,与牛奶、溶菌酶一同投入反应釜中,开启机械搅拌并控制其转速为20r/min,连续机械搅拌6h后,开启超声波,开启超声搅拌并控制其转速为90~120r/min,连续超声搅拌15min,得到混合物料B;
S6、同型乳酸发酵:将混合物料A、混合物料B、驯化后的同型发酵乳酸菌、红糖和纯化水投入发酵釜中,发酵釜的控制温度在25℃,连续乳酸发酵7d,得到发酵原液;
S7、离心:将发酵原液先投入卧螺离心机进行粗分离,再投入碟片离心机进行精细分离,得到浆液;
S8、调配灌装:向浆液中投入磷脂酰丝氨酸、食用香精和纯化水,控制混合物料体系温度在72℃至物料完全溶解,无氧灌装,得到灌装饮料;
S9、灭菌:将灌装饮料放入水浴式杀菌锅中,于105℃恒温条件下连续灭菌30min,冷却至常温,即得到最终产品。
在上述制备过程中,为提高雨生红球藻中虾青素的提取量,联合破壁采用先用溶菌酶破坏雨生红球藻的厚壁孢子上的多糖和糖脂成分,使得雨生红球藻表面松动,再通过超声波产生的空化作用,对松动的破坏雨生红球藻表面直接破坏,使得虾青素直接流出雨生红球藻细胞并融入牛奶中。联合破壁无需大量使用溶菌酶,相比超声波直接破壁,联合破壁超声作用时间短,经济效益好。
实施例1~3采用上述制备方法,均可以制备出本发明的乳酸菌发酵饮料。
试验例
试验目的:研究本发明的乳酸菌发酵饮料对免疫力改善的效果。
试验材料:实施例2制备出的乳酸菌发酵饮料。
试验分组和剂量选择:动物随机分组,每组10只,各取4组分别进行5个实验项目:碳廓清实验(I);ConA诱导的小鼠淋巴细胞转化实验、NK细胞的活性测定(II);脏体比值测定、半数溶血值(HC50)的测定、和抗体生成细胞数的测定(III);迟发型变态反应实验(IV);小鼠腹腔巨噬细胞吞噬鸡红细胞实验(V)。
据人体口服推荐量,设实施例2制备出的乳酸菌发酵饮料低、中、高剂量分别为9.1g/kg、18.2g/kg、54.6g/kg(分别相当于人体推荐量的5、10、30倍)。试验时,分别取本发明产品浸泡浓缩液10mL、20mL、60mL加蒸馏水定容至200予以等体积的溶剂,每天一次,对照组投喂等体积生理盐水,连续灌胃至少30d。
试验方法:小鼠碳廓清实验;ConA诱导小鼠脾淋巴细胞转化实验采用MTT法;DNFB诱导小鼠迟发型变态反应采用耳肿胀法;抗体生成细胞检测采用Jeren改良玻片法;血清溶血素测定采用半数溶血值;小鼠腹腔巨噬细胞吞噬鸡红细胞试验采用半体内法;NK细胞活性测定采用乳酸脱氢酶测定法。
试验结果:统计记录于表1、表2和表3。

根据表1、表2和表3,经口灌胃给予小鼠9.1g/kg、18.2g/kg、54.6g/kg剂量的实施例2制备出的乳酸菌发酵饮料30d,与对照组比较,18.2g/kg、54.6g/kg剂量能显著提高小鼠迟发型变态反应能力、NK细胞活性、半数溶血值,54.6g/kg剂量能显著提高小鼠抗体生成细胞数(p<0.05),提示本发明产品具有一定增强免疫力功能的作用。
尽管已经示出和描述了本发明的实施例,对于本领域的普通技术人员而言,可以理解在不脱离本发明的原理和精神的情况下可以对这些实施例进行多种变化、修改、替换和变型,本发明的范围由所附权利要求及其等同物限定。
Claims (8)
1.一种增强免疫力的乳酸菌发酵饮料,其特征在于,包括由以下重量份配比的原料制备而成:药食同源中草药9~18份、蛹虫草2~4份、雨生红球藻2~4份、磷脂酰丝氨酸2~4份、果蔬15~20份、牛奶15~20份、同型发酵乳酸菌0.01~0.05份、溶菌酶0.01~0.05份、红糖8~10份、食用香精0.1~0.5份和纯化水50~70份;
其中,所述药食同源中草药包括人参、益智仁、枸杞和黄精。
2.根据权利要求1所述的一种增强免疫力的乳酸菌发酵饮料,其特征在于,所述药食同源中草药中各组分的重量份配比为人参3~6份、益智仁2~4份、枸杞2~4份、黄精2~4份。
3.根据权利要求1所述的一种增强免疫力的乳酸菌发酵饮料,其特征在于,所述磷脂酰丝氨酸由天然软磷脂采用酶转化法制备。
4.根据权利要求1所述的一种增强免疫力的乳酸菌发酵饮料,其特征在于,所述果蔬包括蓝莓、葡萄、苹果、菠萝、番茄、黑豆、胡萝卜、韭菜、芋头、雪梨、柠檬中的一种或多种。
5.根据权利要求1所述的一种增强免疫力的乳酸菌发酵饮料,其特征在于,所述同型发酵乳酸菌包括由以下重量份配比的原料:乳链球菌2份,德氏乳杆菌2份,乳酸乳杆菌2份和保加利亚乳杆菌2份。
6.一种如权利要求1~5任意一条所述的增强免疫力的乳酸菌发酵饮料,其特征在于,包括以下制备步骤:
S1、称重:按要求称取各重量份的原料;
S2、臭氧清洗:将药食同源中草药、蛹虫草、雨生红球藻和果蔬浸泡在流动的50~60℃水中3~5min,再用臭氧清洗机清洗5~10min;
S3、草药破碎:将药食同源中草药、蛹虫草和果蔬投入万能破碎机中,按照0.5~1.0cm规格进行破碎后,添加纯化水,搅拌均匀,得到混合物料A;
S4、菌种驯化:将同型发酵乳酸菌接种于牛奶培养基,待常温活化后,接入混合物料A:牛奶=1:1的培养基,于15~28℃下培养增值,得到驯化后的同型发酵乳酸菌;
S5、联合破壁:将雨生红球藻投入万能破碎机中,破碎成0.1~0.2cm的颗粒后,与牛奶、溶菌酶一同投入反应釜中,连续机械搅拌4~8h后,开启超声波,连续超声搅拌10~20min,得到混合物料B;
S6、同型乳酸发酵:将混合物料A、混合物料B、驯化后的同型发酵乳酸菌、红糖和纯化水投入发酵釜中,连续乳酸发酵6~8d,得到发酵原液;
S7、离心:将发酵原液先投入卧螺离心机进行粗分离,再投入碟片离心机进行精细分离,得到浆液;
S8、调配灌装:向浆液中投入磷脂酰丝氨酸、食用香精和纯化水,控制混合物料体系温度在70~80℃至物料完全溶解,无氧灌装,得到灌装饮料;
S9、灭菌:将灌装饮料放入水浴式杀菌锅中,于105℃恒温条件下连续灭菌20~40min,冷却至常温,即得到最终产品。
7.根据权利要求6所述的一种增强免疫力的乳酸菌发酵饮料的制备方法,其特征在于,S4中,所述发酵釜的控制温度在15~28℃。
8.根据权利要求6所述的一种增强免疫力的乳酸菌发酵饮料的制备方法,其特征在于,S5中,所述机械搅拌的转速控制为10~30r/min;所述超声搅拌的转速控制为90~120r/min。
