背景技术
益生菌是一类对宿主有益的活性微生物,是定植于人体肠道、生殖系统内,能产生确切健康功效,从而改善宿主微生态平衡、发挥对肠道有益作用的活性有益微生物的总称。现在广泛的应用于食品、生物、医药等领域。
中老年慢性便秘是困扰老年人的一个常见疾病,其严重影响患者生活质量,还增加高血压、心脑血管疾病的致死率。服用益生菌是缓解老年性便秘的一种常规疗法,但其也存在着疗效低,作用效果不佳的问题。
发明内容
为了解决以上技术问题,本发明提供了一种治疗老年性便秘的一种发酵复方饮品,并提供了其制备方法。
人参自古誉为“百草之王”,“具有滋阴补肾,扶正固本”的功效,其含多种皂甙和多糖类成分,浸出液可被皮肤缓慢吸收、医药不良刺激,能扩张皮肤毛细血管,促进皮肤血液循环,增加皮肤营养,调节皮肤的水油平衡,防止皮肤脱水、硬化、起皱,人参活性物质抑制黑色素的还原性能,使皮肤洁白光滑,能增强皮肤弹性,使细胞获得新生,是护肤美容的极品。人参还是一种强壮滋补药,适用于调整血压、恢复心脏功能、神经衰弱及身体虚弱等症,也有祛痰、健胃、利尿、兴奋等功效。
本发明的发酵饮品是一种以人参,山药,大枣,为主要原料的滋阴补肾,润肠通便的益生菌发酵复方饮品,能达到滋阴补肾,用于习惯性便秘,尤其中老年人慢性便秘,并兼具有美容养颜,促进新陈代谢的功效。
本发明是采用如下制备方法制备而成的:
一、制备乳酸菌种子液
1)准备种子培养基材料:20 g葡萄糖、10g蛋白胨、10g牛肉膏、5g酵母膏,5g乙酸钠,2g柠檬酸氢二铵,0.25g硫酸锰,0.6g硫酸镁, 2g磷酸二氢钾,1ml吐温80,加热水至完全溶解,然后用自来水定容至1L,调节pH6.5-7.5,200ml瓶分装,每瓶加0.7g碳酸钙,0.1 MPa 121℃灭菌20min,然后冷却备用;
2)菌种活化培养:将干酪乳杆菌、植物乳杆菌和鼠李糖乳杆菌三个菌种分别在MRS平板培养基上划出单菌落,在培养箱中37℃,培养48小时,得到扩大培养所需的菌种;
3)种子液:用步骤1)中准备好的冷却至室温的培养基,在无菌条件下等量接种三种菌种,摇匀后放置于培养箱中,37℃,静止发酵24h:检测发酵液的pH,pH在4.0左右时即可终止发酵,以此发酵液作为种子液;
二、制备中药复方提取物
按与种子液质量比取:1%人参、2.3%山药、1.7%大枣、1.3%枸杞子、2%桑椹、2.7%鲜芦根、1.3%芦荟、1.7%荷叶、2%薏苡仁,称取以上物质加入体积/质量比8倍的水,浸泡40min后,于115摄氏度提取1h,然后过滤,即可得到中药复方提取液;
三、制备益生菌发酵中药提取物底物:
按与种子液质量比取:0.7%葡萄糖、0.07%酵母膏、0.3%蛋白胨、0.07%硫酸镁、0.13%牛肉膏、0.1%磷酸氢二钾、0.1%磷酸二氢钾,溶解,加入中药复方提取液,即可制得发酵中药提取液,转移至50L发酵罐中,调节pH6.5-7.5,定容,121℃,30min灭菌,然后冷却至36℃;
四、接种发酵
在无菌条件下,按照预培养体积的2%将步骤一中的种子液接入发酵罐中静置培养,控制发酵温度37℃,培养24h,PH降到3.5-4.0之间,即可终止发酵;
五、调配
向步骤四中所得到的发酵液中加入20%的低聚果糖,加入过程中发酵液应处于均匀搅拌状态,搅拌速度不宜过快;
六、 灌装:
将调配好的发酵液通过管道输送至灌装生产线,进行自动化灌装;
七、 灭菌:
将灌装好的益生菌中药发酵复方饮品放入灭菌柜中115度,30分钟灭菌。
本发明采用上述解决方案所能达到的有益效果是:
1、本发明利用益生菌对中草药提取液进行发酵,经过发酵中草药提取液,益生菌的活动提取了发酵底物中的活性成分,经过发酵后的中草药将里面的蛋白质、多肽、多糖等有效成分小分子化,更容易被人体吸收。对于病人有更显著的辅助治疗作用。且在产品中的益生菌,有利于调节肠道菌群结构,增强机体的免疫力。一个突出的优点是本发明的材料主要使用一些生活中常见的食品,或一些药食同源性的中草药,食用安全,无任何不良的副作用,用于辅助治疗滋阴补肾,润肠通便有明显效果。

实施例1
一、制备乳酸菌种子液

3)种子液:用步骤1)中准备好的冷却至室温的培养基,在无菌条件下等量接种三种菌种,摇匀后放置于培养箱中,37℃,静止发酵24h:检测发酵液的pH,pH在4.0左右时即可终止发酵,以此发酵液作为种子液;
二、制备中药复方提取物
按与种子液质量比取:1%人参、2.3%山药、1.7%大枣、1.3%枸杞子、2%桑椹、2.7%鲜芦根、1.3%芦荟、1.7%荷叶、2%薏苡仁,称取以上物质加入体积/质量比8倍的水,浸泡40min后,于115摄氏度提取1h,然后过滤,即可得到中药复方提取液;
三、制备益生菌发酵中药提取物底物:
按与种子液质量比取:0.7%葡萄糖、0.07%酵母膏、0.3%蛋白胨、0.07%硫酸镁、0.13%牛肉膏、0.1%磷酸氢二钾、0.1%磷酸二氢钾,溶解,加入中药复方提取液,即可制得发酵中药提取液,转移至50L发酵罐中,调节pH6.5-7.5,定容,121℃,30min灭菌,然后冷却至36℃;
四、接种发酵
在无菌条件下,按照预培养体积的2%将步骤一中的种子液接入发酵罐中静置培养,控制发酵温度37℃,培养24h,PH降到3.5-4.0之间,即可终止发酵;
五、调配
向步骤四中所得到的发酵液中加入20%的低聚果糖,加入过程中发酵液应处于均匀搅拌状态,搅拌速度不宜过快;
六、 灌装:
将调配好的发酵液通过管道输送至灌装生产线,进行自动化灌装;
七、 灭菌:
将灌装好的益生菌中药发酵复方饮品放入灭菌柜中115度,30分钟灭菌。
本发明是以灵芝,人参,山药,大枣为主要原料,以三株益生菌为菌种,以中草药复合配方为辅助的益生菌发酵中药发酵复方饮品。本品有滋阴补肾,润肠通便的功效,虽然此功能食品主要以中草药中的滋阴补肾,润肠通便的功效为主,但发酵好的发酵液益生菌活菌数也能达到约109CFU/ml以上。本发明所用的益生菌菌株分别为干酪乳杆菌、植物乳杆菌和鼠李糖乳杆菌,种子培养基为普通的液体MRS培养基,生产发酵培养基为中草药16.7%及1.47%的益生菌生长因子。
本发明的工艺为发酵罐三级静态液体深层发酵,控制发酵温度37℃,发酵时间24h。
试验资料
将诊断为慢性中老年便秘的患者共60例,随机分为两组,一组30例,第一组为治疗组给与本发明制备的中药益生菌发酵液,另一组为对比组给与益生菌治疗(仅仅没有加中药提取物,其他完全与第一组相同),连续服用四周,治疗组有效26例,治愈率为:86.7%,对比组有效21例,治愈率为:70%,所以治疗组制备的益生菌发酵饮品对老年性便秘的功效明显大于一般的益生菌。
Claims (2)
1.一种发酵复方饮品,其特征在于,由以下制备方法制备而成:
一)制备乳酸菌种子液
1)准备种子培养基材料:20 g葡萄糖、10g蛋白胨、10g牛肉膏、5g酵母膏,5g乙酸钠,2g柠檬酸氢二铵,0.25g硫酸锰,0.6g硫酸镁, 2g磷酸二氢钾,1ml吐温80,加热水至完全溶解,然后用自来水定容至1L,调节pH6.5-7.5,200ml瓶分装,每瓶加0.7g碳酸钙,0.1 MPa 121℃灭菌20min,然后冷却备用;
2)菌种活化培养:将干酪乳杆菌、植物乳杆菌和鼠李糖乳杆菌三个菌种分别在MRS平板培养基上划出单菌落,37℃,培养48小时,得到扩大培养所需的菌种;
3)种子液:用步骤1)中准备好的冷却至室温的培养基,在无菌条件下等量接种三种菌种,摇匀后放置于培养箱中,37℃,静止发酵24h:检测发酵液的pH,pH在4.0时即可终止发酵,以此发酵液作为种子液;
二)制备中药复方提取物

2.一种权利要求1所述的发酵复方饮品的制备方法,其特征在于,具体制备方法如下:
一)制备乳酸菌种子液
