Abstract
本发明涉及保健食品领域,具体为一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料,按重量份计包括:药食同源中草药水提液7‑12份、果蔬15‑20份、植物乳杆菌0.01‑0.05份、白砂糖8‑10份、食用香精0.1‑0.5份和纯化水50‑70份。本发明还包括上述乳酸菌保健饮料的制备方法,包括制备药食同源中草药水提液,将药食同源中草药水提液与果蔬、白砂糖和水混合均匀后加入植物乳杆菌发酵,最后依次经过滤、调配、灌装和灭菌制得该保健饮料。本发明复合保健饮料通过对人体肾、肝、脾等器官的综合调理,引入高效的植物乳杆菌进行发酵,其发酵产物对肠道微生态的调节,增强了人体对营养成分和功效成分的吸收效果,在抗疲劳和耐缺氧等方面均具有显著功效。

技术领域
本发明涉及保健食品领域,具体为一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料及其制备方法。
背景技术
随着我国经济的快速增长,人们生活方式和生活节奏都在不断变化,生活质量和身体健康意识不断增强,但同时高压力、高强度、长时间的不健康工作和生活方式带来的身体过度透支引起的体虚、疲惫、机体免疫力弱及常伴有不同程度的便秘等问题,一直困扰着较多的人群。纵观目前市场上在售的各型各色、功能繁多的药品及保健品,虽有一定效果,但短时间只能治其表而不能固其本,长期食用可能对身体机能产生负担引发副作用且价格昂贵、食用普遍不方便。
国家推行中医“治未病”理论,提倡“药补不如食补”的健康养生方式,但一般家用基本上都是一种或少数几种中药或药食同源原料搭配煲汤或用做配料食用,食用次数和加工方式简单、有限,很难起到食补效果。而现在市场上的绝大多数功能性食品,基本都是采用中草药提取物粉,喷粉过程易导致部分营养成分流失且需添加较多糊精或其他食品添加剂方可喷粉,导致有效成分含量降低,功效不明显。
为此,我们提出了一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料及其制备方法以良好的解决上述弊端。
发明内容
本发明的目的在于提供一种能够提高机体抗疲劳和耐缺氧能力的具有抗疲劳和耐缺氧功能的药食同源复合保健饮料。
本发明复合保健饮料各原料的用量为在下述重量份范围都具有较好效果:
药食同源中草药水提液7-12份、果蔬15-20份、植物乳杆菌0.01-0.05份、白砂糖-10份、食用香精0.1-0.5份、纯化水50-70份;
其中药食同源中草药水提液各原料的用量为在下述重量份范围都具有较好疗效:
人参3-4份、黄精6-8份、益智仁6-7份、龙眼肉10-12份、肉桂3-4份、山药3-5份、莲子8-10份、枸杞9-10份、覆盆子9-10份、桑葚9-10份、生姜3-4份。
优选为:所述果蔬是蓝莓、葡萄、苹果、菠萝、番茄、黑豆、胡萝卜、韭菜、西红柿、芋头、雪梨、柠檬中的一种或多种。
优选为:人参3份、黄精7份、益智仁7份、龙眼肉11份、肉桂3份、山药4份、莲子9份、枸杞10份、覆盆子10份、桑葚10份、生姜3份。
本发明的另一个目的在于提供该保健饮料的制备方法。
本发明采用以下方法制备成保健饮料:首先制备药食同源中草药水提液,备用;其次对果蔬与药食同源中草药水提液混合物进行发酵;最后将发酵原液、食用香精和纯化水按比例进行调配,经过灭菌冷却即可得到该保健饮料;为了使药食同源中草药更好地发挥药效,优选对远原料水提液进行醇沉。但是这些不能用于限制本发明的保护范围。
优选的,本发明复合保健饮料的制备方法如下:
S1,制备药食同源中草药水提液
a)原料鉴定、筛选:对每种药食同源中草药随机抽样,参照《中华人民共和国药典(2015版)》和新食品原料检测方法对抽取的样品进行标志性成分含量检测,以确定原料真假,再筛选出发霉、变质等不符合要求的原料;将人参、黄精、益智仁、龙眼肉、肉桂、山药、莲子、枸杞、覆盆子、桑葚和生姜按照相应的重量配比进行称取,干燥处理,备用;
b)将干燥后的各原料经万能粉碎机破碎,再用纯化水清洗干净;
c)蒸煮浓缩,根据原料的量,按照一定的料液比进行蒸煮三次,合并三次收集的滤液;
d)将步骤c)中的滤液加5%的乙醇进行反复数次沉降,除去不溶物质;
e)将步骤d)中的滤液倒入三足式离心机,采用200目筛网,在转速为4000r/min条件下离心分离,去除滤渣,收集离心液,制得药食同源中草药水提液,备用;。
S2,发酵
f)将果蔬用水清洗干净,并用粗碎机进行破碎分散,并与步骤e)中的药食同源中草药水提液混合,加入白砂糖,混合均匀,在140℃温度下保持10s进行超高温瞬时灭菌处理;
g)将步骤f)中的浆液控制温度在40-43℃之间,加入植物乳杆菌,混合均匀,发酵时间2-15天;
h)过滤,将步骤g)中的浆液浆液投入三足式离心机,采用200目筛网,在转速为4000r/min条件下离心分离,去除滤渣,取上清液得中药材和果蔬的发酵原液,备用;
S3,调配
i)将步骤h)制备的中药材和果蔬发酵原液与食用香精及纯化水按规定比例称量、混合,控制混合物料体系温度在70-80℃至物料完全溶解;
S4,灌装
j)采用全自动饮料灌装生产线,将步骤i)调配的混合物料灌装成净含量为50ml的瓶装或铝塑复合膜袋装饮料,灌装过程控制混合物料温度为60-70℃;
S5,灭菌
k)将步骤j)灌装的混合物料在100-110℃温度下进行熟化、灭菌处理30-50min,制得成品。
上述制备方法中,优选为:其中步骤c)的蒸煮浓缩中,第一次按照料液比1:10将步骤b)中清洗干净的原料和纯化水投入提取罐,100℃提取2h,过150目筛网,收集得到第一滤液;第二次按照料液比1:8将纯化水注入提取罐,100℃提取1.5h,过100目筛网,收集得到第二滤液;第三次按照料液比1:6将纯化水注入提取罐,100℃提取1.5h,过100目筛网,收集得到第三滤液。
上述制备方法中,优选为:其中步骤k)的熟化灭菌操作是采用分段阶梯式降温熟化灭菌,把温度迅速升到110℃并保温15min,然后降温到105℃维持10min,再降温到100℃保持15min。
与现有技术相比,本发明 提供了一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料及其制备方法,具备以下有益效果:
本发明复合保健饮料通过对人体肾、肝、脾等器官的综合调理,特别是引入高效的植物乳杆菌进行发酵,其发酵产物对肠道微生态的调节,增强了人体对营养成分和功效成分的吸收效果,在抗疲劳和耐缺氧等方面均具有显著功效。
具体实施方式
以下通过实施例来进一步阐述本发明的有益效果,显然,所描述的实施例仅仅是本发明一部分实施例,而不是全部的实施例。基于本发明中的实施例,本领域普通技术人员在没有做出创造性劳动前提下所获得的所有其他实施例,都属于本发明保护的范围。
实施例:按下述重量配比称取中药原料:人参3份、黄精7份、益智仁7份、龙眼肉11份、肉桂3份、山药4份、莲子9份、枸杞10份、覆盆子10份、桑葚10份、生姜3份。
生产方法如下:将上述配比的中药原料干燥处理,经万能粉碎机破碎后,加水冲洗;根据中药的量按照一定的料液比进行蒸煮三次,具体为:第一次按照料液比1:10将清洗干净的原料和纯化水投入提取罐,100℃提取2h,过150目筛网,收集得到第一滤液;第二次按照料液比1:8将纯化水注入提取罐,100℃提取1.5h,过100目筛网,收集得到第二滤液;第三次按照料液比1:6将纯化水注入提取罐,100℃提取1.5h,过100目筛网,收集得到第三滤液,合并三次收集的滤液;在滤液中加5%的乙醇进行醇沉;将滤液倒入三足式离心机,采用200目筛网,在转速为4000r/min条件下离心分离,收集滤液并得到药食同源中草药水提液;记药食同源中草药水提液的重量份为10,将重量份为18的果蔬破碎分散,与药食同源中草药水提液混合,加入重量份为10的白砂糖,混合均匀后,在140℃温度下保持10s进行超高温瞬时灭菌处理;待灭菌后的浆液冷却并控制在40-43℃之间,在浆液中加入重量份为0.05的植物乳杆菌,混合均匀,发酵时间12天;浆液投入三足式离心机,采用200目筛网,在转速为4000r/min条件下离心,制得中药材和果蔬的发酵原液(记为实验组)。将上述中药材和果蔬发酵原液与重量份为0.5的食用香精进行混合,加入纯化水至其累积量为60份,再充分搅匀调配。将调配的混合物料采用全自动饮料灌装生产线灌装成净含量为50ml的瓶装或铝塑复合膜袋装饮料,灌装过程控制混合物料温度为70℃;最后将灌装的混合物料在100-110℃温度下进行熟化、灭菌处理30-50min,制得成品。
1.本发明发酵原液的抗疲劳验证实验
试验试剂和仪器:生理盐水(记为空白组);红牛功能饮料(记为对照组);中药材和果蔬的发酵原液(实验组);游泳恒温箱(60 cm×50 cm×50 cm)
试验方法:按《保健食品功能学评价程序和检验方法》进行。取SPF级昆明小鼠40只,雌雄各半,6-8周龄,随机分配成4组, 每组10只;分别为空白组、对照组、给药低剂量(实验组1)和给药高剂量(实验组2);然后对上述小鼠进行灌胃给药。剂量分组根据日常功能饮料人体摄入量120ml/d,按60kg体重计,相当于2ml/kg,折和小鼠的等效剂量换算系数为9.1,相当于18.2ml/kg,发酵原液组按0.5、2倍等效量设计低、高两剂量组,空白组和对照组均按1倍等效量设计。分组如下:
(1)空白组:18.2ml/kg(生理盐水)
(2)对照组:18.2ml/kg(红牛功能饮料)
(3)实验组1:9.1ml/kg
(4)实验组2:36.4ml/kg
连续给予受试物15d,测试末次给样30min后,小鼠负重5%体重的铅皮,置于水温25℃、水深35~40cm 的水箱中游泳,记录小鼠自游泳开始至死亡的时间,作为小鼠的游泳时间(min)。用SPSS19.0 for windows 进行数据统计分析。计量资料以 x±s 表示,组间比较采用 t 检验,P < 0.05 为差异有统计学意义。
实验结果
1.1酵素对小鼠体重的影响

由表1.1可知,小鼠服用酵素15d 后,小鼠体重与空白组和对照组比较无显著差异(P>0.05)。各组均不会引起体重过快增长,也不会抑制小鼠的正常发育。

由表1.2可知,灌服酵素15d后,实验组小鼠负重游泳时间与空白组及对照组相比有显著延长(p<0.01),显示酵素对小鼠负重游泳时间的影响要明显优于红牛。
体能是机体对体力活动和运动的适应能力。体力活动对机体的影响主要反应在两个基本生理、生化过程中,即能量代谢消耗及机体各系统生理功能的调节和适应。就能量而言,肌肉活动时主要靠肌细胞中的ATP迅速分解提供能量,而ATP的补充主要靠糖原、葡萄糖的无氧酵解生成乳酸过程中释放能量合成ATP和有氧情况下线粒体内糖、脂肪酸及氨基酸氧化分解产生能量合成ATP,另外肌肉中CP分解合成ATP,因为体力活动时主要靠糖原酵解来获得能量,当糖原被大量消耗时,机体活动能力就降低,糖酵解的产物—乳酸的堆积就会引起疲劳。在消耗肌糖原的同时从血液中摄取血糖,后者可由肝糖原分解,加以补充维持血糖水平。一旦肌、肝糖原大量被消耗,则血糖下降可导致中枢神经系统供能不足,从而导致全身性疲劳的发生,体能下降。
在一系列抗疲劳实验中,酵素给药组结果均为阳性,且差异极显著(P<0.01)。由表2可见,酵素给药组小鼠在负重游泳试验中,其负重游泳时间均明显长于空白组及对照组,显示天然酵素高低剂量组可增加肝糖原的储备量,肝糖原贮备的增加可维持较长时间的血糖水平,为机体提供更多的能量,减少剧烈体力劳动后糖原的消耗,从而具有增进体能,增强耐力、抗疲劳的作用。
根据实验结果可知:酵素具有增强机体抗疲劳的功效,且其作用效果明显优于红牛功能饮料。
2.本发明发酵原液的耐缺氧验证实验
试验试剂和仪器:生理盐水(记为空白组);维生素C(河南九州通医药有限公司,记为对照组);中药材和果蔬的发酵原液(实验组);钠石灰(上海纳辉干燥试剂厂)、凡士林(分析纯)、250mL广口瓶、注射器、电子天平(上海箐海仪器有限公司,型号FA1104N)
试验方法:按《保健食品功能学评价程序和检验方法》进行。取昆明小鼠40只,雌雄各半,6-8周龄,随机分配成4组,每组10只;分别为空白组、对照组、给药低剂量(实验组1)和给药高剂量(实验组2);然后对上述小鼠进行灌胃给药。剂量分组根据日常功能饮料人体摄入量120ml/d,按60kg体重计,相当于2ml/kg,折和小鼠的等效剂量换算系数为9.1,相当于18.2ml/kg,发酵原液组按0.5、2倍等效量设计低、高两剂量组,空白组和对照组均按1倍等效量设计,每日一次,连续14天。分组如下:
(1)空白组:18.2ml/kg(生理盐水)
(2)对照组:18.2ml/kg(维生素C)
(3)实验组1:9.1ml/kg
(4)实验组2:36.4ml/kg
末次灌胃后1h,将各组小鼠分别放入盛有15g钠石灰的250ml磨口瓶内(用前均用水校正容量,每瓶放一只小鼠),加盖密封(盖口涂上凡士林),立即计时,以腹部不再起伏,呼吸停止为指标,记录小鼠的存活时间。用SPSS19.0forwindows进行数据统计分析。计量资料以x±s表示,组间比较采用t检验,P<0.05为差异有统计学意义。

由表2.2可知,灌服酵素14d后,实验组小鼠耐缺氧能力与空白组相比有显著提高(p<0.01),同时也高于对照组(p<0.01)。
通过实验研究发现酵素低、高剂量组在提高小鼠的耐缺氧能力上与空白组和对照组均有明显差异,说明酵素可提高机体的耐缺氧能力。随着小鼠在密闭广口瓶中不断呼吸,广口瓶中的氧气会越来越少,而二氧化碳则由于钠石灰的吸收而不会明显增加,小鼠因外环境供氧减少而发生了乏氧性缺氧,表现为竖毛、转圈、后肢向外后方伸直、抽搐等,最后因严重缺氧而死亡。酵素低、高组较空白对照组均能够延长小鼠的存活时间,其中酵素高剂量组效果最为明显。
缺氧是一种劣性刺激,可使组织细胞得不到所需的氧,进而影响机体的供氧功能,最终会导致机体心、脑等重要器官损伤,严重时会发生机体供氧不足而死亡。缺氧是一种紧张性的刺激,可能会引起机体的各种应激性反应。轻度缺氧时,主要引起机体代偿性反应,比如表现为呼吸加深加快,心跳加速,心输出量增加,血液重新分配,以保证重要器官的血液供应;而严重缺氧时,机体出现代谢机能障碍,甚至会导致机体死亡;急性缺氧时,机体来不及完成代偿反应,各种缺氧情况都会造成机体器官不同程度的损伤。目前,缺氧是造成细胞损伤的最常见原因,可以诱发体内各系统的功能改变,
根据实验结果可知:酵素具有增强机体耐缺氧的功效,且其作用效果明显优于维生素C。

Claims (7)
1.一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料,其特征在于,按下列重量份计,所述保健饮料包括下述组分:药食同源中草药水提液7-12份、果蔬15-20份、植物乳杆菌0.01-0.05份、白砂糖8-10份、食用香精0.1-0.5份和纯化水50-70份;
所述药食同源中草药水提液是由以下重量份的中药原料制备而成:人参3-4份、黄精6-8份、益智仁6-7份、龙眼肉10-12份、肉桂3-4份、山药3-5份、莲子8-10份、枸杞9-10份、覆盆子9-10份、桑葚9-10份、生姜3-4份。
2.根据权利要求1所述的一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料,其特征在于:所述果蔬是蓝莓、葡萄、苹果、菠萝、番茄、黑豆、胡萝卜、韭菜、西红柿、芋头、雪梨、柠檬中的一种或多种。
3.根据权利要求1所述的一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料,其特征在于,所述药食同源中草药水提液是由以下重量份的中药原料制备而成:人参3份、黄精7份、益智仁7份、龙眼肉11份、肉桂3份、山药4份、莲子9份、枸杞10份、覆盆子10份、桑葚10份、生姜3份。
4.根据权利要求1-3任意一项所述的一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料的制备方法,其特征在于:所述制备方法的步骤如下:
S1,制备药食同源中草药水提液
a)取药食同源中草药各原料进行干燥处理,备用;
b)将干燥后的各原料经万能粉碎机破碎,再用纯化水清洗;
c)蒸煮浓缩,根据原料的量,按照一定的料液比进行蒸煮三次,合并三次收集的滤液;
d)将步骤c)中的滤液加5%的乙醇进行反复数次沉降,除去不溶物质;
e)将步骤d)中的滤液离心分离,收集离心液,制得药食同源中草药水提液,备用;
S2,发酵
f)将果蔬破碎分散,并与步骤e)中的药食同源中草药水提液混合,加入白砂糖,混合均匀,在140℃温度下保持10s进行超高温瞬时灭菌处理;
g)将步骤f)中的浆液控制温度在40-43℃之间,加入植物乳杆菌,混合均匀,发酵时间2-15天;
h)过滤,将步骤g)中的浆液离心,取上清液得中药材和果蔬的发酵原液,备用;
S3,调配
i)将步骤h)制备的中药材和果蔬发酵原液与食用香精及纯化水按规定比例称量、混合;
S4,灌装
j)将步骤i)调配的混合物料灌装,灌装过程控制混合物料温度为60-70℃;
S5,灭菌
k)将步骤j)灌装的混合物料在100-110℃温度下进行熟化、灭菌处理30-50min,制得成品。
5.根据权利要求4所述的一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料的制备方法,其特征在于:其中步骤c)的蒸煮浓缩中,第一次按照料液比1:10将步骤b)中清洗干净的原料和纯化水投入提取罐,100℃提取2h,过150目筛网,收集得到第一滤液;第二次按照料液比1:8将纯化水注入提取罐,100℃提取1.5h,过100目筛网,收集得到第二滤液;第三次按照料液比1:6将纯化水注入提取罐,100℃提取1.5h,过100目筛网,收集得到第三滤液。
6.根据权利要求4所述的一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料的制备方法,其特征在于:其中步骤k)的熟化灭菌操作是采用分段阶梯式降温熟化灭菌,把温度迅速升到110℃,然后分段降到100℃,熟化灭菌时间为30-50min。
7.根据权利要求6所述的一种具有抗疲劳和耐缺氧功能的药食同源复合保健饮料的制备方法,其特征在于:其中步骤k)的熟化灭菌操作是将灌装的混合物料在110℃下保温15min,然后降温到105℃维持10min,再降温到100℃保持15min。
