Abstract
本发明提供了一种中药抗菌草莓酒的制备方法,本发明选取的中草药都含有丰富的抗菌成分,用其代替酿酒过程中的SO2,可以避免因为摄入过量SO2而导致的过敏、哮喘等问题,彻底消除安全隐患,符合安全环保趋势;将壳聚糖、明胶通过复合、交联固化形成微球,其吸附效果良好,一方面,通过与中药提取物低压浸泡,吸附大量的中药提取物并达到饱和,发酵期加入到草莓酒中,可以达到缓释的目的,延长中药提取物杀菌的作用时间,保证发酵过程达标;另一方面,复合微球释放中草药后可以重新吸附草莓酒中有机大分子、不溶物等,起到澄清降酸的作用,解决了酒体浑浊、酸味突出的问题。

本发明属于酒饮料及其制备方法技术领域,具体涉及一种中药抗菌草莓酒的制备方法
背景技术
在果酒的传统酿造工艺中,通常应用SO2作为添加剂,SO2在果酒酿造中的作用是抗菌和两个方面。虽然SO2在酿造中有如此重要的作用,但是它也会给人的健康带来问题。大量研究结果表明SO2对人及动物具有毒性,能使动物出现胃部发生病变、硫胺素缺乏及生长缓慢等几个方面的问题。人体对过量SO2会出现过敏反应,过敏出现在低摄入量时(大约1mg),主要表现为哮喘(占总人数的 4%~10%)。随着人们对SO2安全性的重视,找到SO2的天然无毒的替代品已成为果酒酿造安全方面的重要研究课题,然而中草药的抗菌效果表现趋势为先是效果较好,随着发酵时间的增加,抗菌效果则不明显,无法保证发酵期内抗菌的需求。
发明内容
针对上述问题,本发明旨在提供一种利用壳聚糖/明胶复合微球作为中药提取物载体的方法,以解决现存的抗菌时间短,效果不足的问题。
本发明通过以下技术方案实现:
一种中药抗菌草莓酒的制备方法,其特征在于,包括以下步骤:
(1)将以重量份计的黄连3-5份,黄柏1-2份,金银花3-5份,板蓝根4-8份,砂仁1-3份,紫苏5-10份,花生壳6-10份充分研磨,加入50-100重量份的沸水进行煎煮1-2h后,用3-5层纱布过滤药渣,得到中药提取物备用;
(2)将10-15份壳聚糖溶于100-300份醋酸溶液中,20-30份明胶溶于80-120份水中得明胶溶液,将上述两种溶液混合搅拌均匀形成壳聚糖/明胶混合溶液;将20-40份液体石蜡和2-4份表面活性剂span-80搅拌条件下水浴加热至50-70℃后,加入上述壳聚糖/明胶混合溶液,乳化10-30min,然后将水浴变为冰水浴,加入3-5份戊二醛进行交联固化反应,40-80min后结束反应,离心,并用乙醇洗涤产物3-5次,将产物干燥备用;
(3)将步骤(2)所得物加入到步骤(1)所得物中,密封,于真空度0.02-0.04MPa下超声震荡20-40min,得到吸附中药提取物的壳聚糖/明胶悬浮液备用;
(4)将80-120份草莓洗净、烘干后破碎,加入步骤(3)所得物、20-40份白砂糖、0.1-0.2份活性干酵母,置于橡木桶内,盖上木桶盖,置于15-20℃下,湿度为60-80%环境下发酵5-10天;
(5)将步骤(4)所得物上清液倒出,除去下层沉淀后重新装入橡木桶内,密封,置于步骤(4)所述发酵条件下继续发酵3-6月即可。
进一步的,步骤(2)所述的醋酸溶液浓度为2-5%,所述戊二醛溶液浓度为30-60%。
进一步的,步骤(3)所述离心条件为转速800-1500 r/min,8-12min。
进一步的,步骤(3)所述干燥温度为40-60℃。

具体实施方式
下面用具体实施例说明本发明,但并不是对本发明的限制。
实施例1
一种中药抗菌草莓酒的制备方法,其特征在于,包括以下步骤:
(1)将以重量份计的黄连4份,黄柏2份,金银花3份,板蓝根4份,砂仁2份,紫苏6份,花生壳7份充分研磨,加入100重量份的沸水进行煎煮2h后,用3层纱布过滤药渣,得到中药提取物备用;
(2)将10份壳聚糖溶于150份醋酸溶液中,20-30份明胶溶于80-120份水中得明胶溶液,将上述两种溶液混合搅拌均匀形成壳聚糖/明胶混合溶液;将20份液体石蜡和2份表面活性剂span-80搅拌条件下水浴加热至50℃后,加入上述壳聚糖/明胶混合溶液,乳化20min,然后将水浴变为冰水浴,加入3份戊二醛进行交联固化反应,1h后结束反应,离心,并用乙醇洗涤产物3次,将产物干燥备用;
(3)将步骤(2)所得物加入到步骤(1)所得物中,密封,于真空度0.02MPa下超声震荡20min,得到吸附中药提取物的壳聚糖/明胶悬浮液备用;
(4)将100份草莓洗净、烘干后破碎,加入步骤(3)所得物、20份白砂糖、0.1份活性干酵母,置于橡木桶内,盖上木桶盖,置于20℃下,湿度为60%环境下发酵10天;
(5)将步骤(4)所得物上清液倒出,除去下层沉淀后重新装入橡木桶内,密封,置于步骤(4)所述发酵条件下继续发酵6月即可。

每天提取经步骤(4)操作处理的实施例1和对比实施例1、2所得上清液,采用紫外可见分光光度计在波长为 570nm 条件下测定果酒的吸光度值用来表征果酒中含菌量,具体对比数据如下表2所示:

由表1表2可以看出,利用壳聚糖/明胶微球吸附中草药添加到草莓酒中,不仅可以使酒体澄清,同时可以保持较长时间的杀菌效果,且杀菌效果良好,本发明值得推广。
Claims (5)
1.一种中药抗菌草莓酒的制备方法,其特征在于,包括以下步骤:
(1)将以重量份计的黄连3-5份,黄柏1-2份,金银花3-5份,板蓝根4-8份,砂仁1-3份,紫苏5-10份,花生壳6-10份充分研磨,加入50-100重量份的沸水进行煎煮1-2h后,用3-5层纱布过滤药渣,得到中药提取物备用;
(2)将10-15份壳聚糖溶于100-300份醋酸溶液中,20-30份明胶溶于80-120份水中得明胶溶液,将上述两种溶液混合搅拌均匀形成壳聚糖/明胶混合溶液;将20-40份液体石蜡和2-4份表面活性剂span-80搅拌条件下水浴加热至50-70℃后,加入上述壳聚糖/明胶混合溶液,乳化10-30min,然后将水浴变为冰水浴,加入3-5份戊二醛进行交联固化反应,40-80min后结束反应,离心,并用乙醇洗涤产物3-5次,将产物干燥备用;
(3)将步骤(2)所得物加入到步骤(1)所得物中,密封,于真空度0.02-0.04MPa下超声震荡20-40min,得到吸附中药提取物的壳聚糖/明胶悬浮液备用;
(4)将80-120份草莓洗净、烘干后破碎,加入步骤(3)所得物、20-40份白砂糖、0.1-0.2份活性干酵母,置于橡木桶内,盖上木桶盖,置于15-20℃下,湿度为60-80%环境下发酵5-10天;
(5)将步骤(4)所得物上清液倒出,除去下层沉淀后重新装入橡木桶内,密封,置于步骤(4)所述发酵条件下继续发酵3-6月即可。
