Abstract
本发明的目的是提供一种具有改善视力功效的组合物及其制剂,即一种以黑果枸杞、枸杞或其提取物作为有效成分的具有改善视力功效的组合物。本发明所提供的组合物兼顾缓解视力疲劳,预防和辅助治疗因黄斑病变、视网膜色素变、糖尿病视网膜病变引起的视力衰退、夜盲症、飞蚊症、青光眼、白内障等视力问题的功能,能够最大程度的改善视力。并且从中医理论来讲,枸杞和黑果枸杞都具有清肝功效,清肝即明目。
Description
技术领域
本发明属于活性物质提取应用技术领域,具体涉及一种具有改善视力功效的组合物。
背景技术
眼睛是心灵的窗户,它带给人们明亮的世界。随着科技的不断进步,电视、电脑、手机普及大大地增加了人们使用眼睛的频率,因此越来越多的眼健康问题开始缠绕在我们生活中。眼睛的干涩、痒痛等症状以及黑眼圈已经成为现今白领们的家常便饭;而过重的生活压力也让“四眼”群体也成为社会的“强势群体”。根据有关部门的统计,我国近视发病率早在1998年就已经排名世界第四;2002年时迅速上升到了世界第二,仅次于日本。近期公布的最新数据显示:我国青少年近视发病率为50%~60%,依然稳居世界第二,但是发病人数已经跃居世界之首,局部地区甚至达到了80%的惊人比例。更该引起重视的是我国的近视人群还存在严重的低龄化问题。造成这种现象的原因除了少部分是遗传因素所致外,大部分是由于生活负担过重加上不良的用眼习惯导致近距离用眼时间过长而引起的。研究表明,重度近视是很容易致盲的,严重的视力问题甚至可能会影响到大脑的发育而导致智力下降。因此,开发缓解视力疲劳,预防和辅助治疗因黄斑病变、视网膜色素变、糖尿病视网膜病变引起的视力衰退、夜盲症、飞蚊症、青光眼、白内障等视力问题的功能食品、保健食品、特殊医学用途食品甚至药品显得尤为重要。
发明内容
本发明的目的是提供一种具有改善视力功效的组合物及其制剂,即一种以黑果枸杞、枸杞或其提取物作为有效成分的具有改善视力功效的组合物。本发明所制备的新组合物具有协同增效作用,可用于改善视力的食品、保健食品、特殊医学用途食品和药品。
本发明首先提供一种具有改善视力的组合物,所述的组合物包含有枸杞和黑果枸杞;
所述的组合物,其中枸杞全部或部分用枸杞糖肽提取物替代;所述的枸杞糖肽提取物的制备方法如下:
1)将清洗后的枸杞子高速剪切后,用水进行提取,然后将获得的提取液离心;
2)用分子量为3500Da的膜系统浓缩,干燥后得到枸杞糖肽提取物,将硫酸苯酚法检测,其糖肽含量>50%。
所述的组合物中,黑果枸杞全部或部分用黑果枸杞提取物替代;所述的黑果枸杞提取物,其制备方法如下:
1)将清洗后的黑果枸杞高速剪切后,按照料液比1:5~8加入添加了5%的Vc和1%的柠檬酸的60%乙醇水溶液进行提取;提取温度为60℃,提取时间为1~3h,管式离心得到提取液;
2)将提取液减压浓缩,上样至大孔吸附树脂混合树脂模拟床,用纯水洗脱后,再用60%~80%乙醇溶液洗脱,回收乙醇洗脱液,干燥、灭菌得到黑果枸杞提取物。经HPLC检测,原花青素含量大于30%。
所述的大孔树脂混合模拟床包含有2~4根MAR柱;每根MAR柱之间即可串联也可并联,其中至少有2根大孔吸附树脂柱串联,大孔吸附树脂柱径高比为1:8~1:12;每根柱子所用大孔吸附树脂均为至少2种或几种以不同比例混合的树脂;流速为4~10BV/HR(柱体积,简称BV)。
所述的树脂,为NKA-9、DA201、ADS-7、S-8、XDA-1、AB-8、HPD-100、XDA-8、D101、LX-68、LX-68B、中的2种或几种。
上述方步骤2)中,所述的回收乙醇洗脱液的方法为膜过滤系统或渗透汽化膜分离设备。
所述的膜过滤系统,具体方法为将管式离心得到的提取液泵入分子量为80的纳滤膜系统浓缩,一旦发现透过纳滤膜的液体有颜色,即可停止浓缩,直接干燥即得。
上述方步骤2)中,所述的干燥方法为冷冻干燥、冷冻喷雾干燥或波态干燥。
作为优选,所述的组合物中还添加有叶黄素;
本发明的组合物用于制备用于改善视力功效的制剂;
所述的制剂为口服制剂,包括如普通片剂、泡腾片、咀嚼片、口含片等,或是如胶囊剂、颗粒剂、合剂、口服剂、丸剂、滴丸剂等。
本发明所提供的组合物兼顾缓解视力疲劳,预防和辅助治疗因黄斑病变、视网膜色素变、糖尿病视网膜病变引起的视力衰退、夜盲症、飞蚊症、青光眼、白内障等视力问题的功能,能够最大程度的改善视力。并且从中医理论来讲,枸杞和黑果枸杞都具有清肝功效,清肝即明目。
具体实施方案
枸杞子是我国传统名贵中药材,在我国已有2000多年的食用历史,也是享誉海内外的滋补食疗珍品。其味甘,性平,有滋补肝肾,益精明目的功效。现代医学、营养学和药理学研究发现,枸杞子中对视网膜具有保护作用的成分主要是枸杞糖肽。枸杞糖肽的生物学作用与其视网膜保护作用密切相关。研究发现,枸杞糖肽能够通过调节视网膜小胶质细胞的免疫活性增加慢性高眼压损伤视网膜节细胞的存活率,从而对青光眼具有视神经保护作用。
黑果枸杞是一种珍稀野生植物,产于戈壁、沙漠等地。具有滋补肝肾、益精明目等功效,被《维吾尔药志》、《晶珠本草》和《四部医典》等著名藏药经典著作所记载。黑果枸杞被称为花青素之王,富含原花青素和花色苷,现代药理学表明,原花青素和花色苷是黑果枸杞缓解视力疲劳、改善视力的物质基础。原花青素主要通过增强眼底视网膜毛细血管的微循环,降低脂质过氧化水平,减轻晶状体的氧化损伤来改善视力。而花青素可促进视网膜细胞中视紫质的再生成,从而预防重度近视及视网膜剥离。
叶黄素是存在于植物中的类胡萝卜素,其对眼睛的显著保护能力受到了科学界的认可和推崇。现代药理学研究表明,叶黄素改善视力的机制主要有以下几点:1.形成视网膜黄斑部中最主要的抗氧化成分;2.过滤有害短波蓝光;3.抑制感光过程中所形成的自由基;4.增加黄斑色素密度,改善强光造成的短暂眩光失能现象,降低光刺激恢复时间。由于人体无法自行合成叶黄素,若无法从食物中得到足量,就需要额外补充。
以下为本发明的实施例,对本发明的上述内容做进一步的详细说明。但不应该就此理解为本发明上述主体的范围仅限于以下的实施例。应当指出,对于本技术领域的普通技术人员来说,在不脱离本发明的核心技术的前提下,还可以做出改进和润饰,这些改进和润饰也应属于本发明的专利保护范围。与本发明的权利要求书相当的含义和范围内的任何改变,都应认为是包括在权利要求书的范围内。
实施例1:
1)将挑拣、清洗过的枸杞子用高速剪切机以转速10000rpm,剪切30min对其进行剪切处理,在60℃下提取2次,每次2h。第一次提取加入10倍纯化水,第二次提取加入8倍纯化水。管式离心后用分子量为3500Da的膜过滤设备进行浓缩,得到枸杞提取液,喷雾干燥后的枸杞糖肽提取物。所得枸杞糖肽用国标检测方法(硫酸苯酚法),测得枸杞多糖含量>50%。
2)将挑选、清洗过得黑果枸杞用高速剪切机以转速10000rpm,剪切30min对其进行剪切处理,在60℃下提取2次,每次2h,每次加5%的Vc和1%的柠檬酸。第一次提取加入12倍70%乙醇,第二次提取加入10倍70乙醇。管式离心机离心得到的提取液减压浓缩,上样至大孔吸附树脂混合模拟床纯化,所用树脂为XDA-8、D101、AB-8的比例为1:1:2。用4BV纯水洗脱后,再用4BV的60%乙醇溶液洗脱,收集60%乙醇洗脱液,洗脱液泵入渗透汽化膜设备回收溶剂至无醇味,冷冻干燥,收集干粉,得黑果枸杞提取物。经检测,原花青素含量大于30%,花色苷含量大于20%,总多酚大于70%。
本发明对黑果枸杞提取物的提取方法进行了优化,从而使提取物中原花青素含量明显提高,从而使获得的黑果枸杞提取物具有更好的明目效果。
实施例2
1)取上述枸杞提取物20kg、黑果枸杞提取物4kg,山梨糖醇0.1kg,叶黄素3g,牛磺酸5g,低聚木糖500g,过80目筛。
2)上述物料均质后,喷雾干燥,灭菌,包装,即得枸杞提取物的固体饮料。
对制得的固体饮料进行质量检测,结果如下:
1.感官指标:
所得固体饮料色泽均匀、口感香醇、带有枸杞特有的清香味道,不结块,不吸潮。
2.理化指标:
所得固体饮料净含量5g/袋,含量差异<3%。按照中国药典的规定对枸杞提取物固体饮料的重金属含量进行测定,测定结果为铅<0.2mg/kg,砷<0.1mg/kg。
依据中国药典规定的微生物限度检测法对枸杞提取物固体饮料进行微生物检测,检测结果如下:细菌总数<1000cfu/g,霉菌总数<20,大肠菌群、金黄色葡萄球菌和沙门氏菌均为检出。
实施例3:
将1200g枸杞糖肽提取物、300g黑果枸杞提取物、0.53g叶黄素与750g甘露醇、500g木糖醇、750乳糖,8.5g枸橼酸混匀,加入聚维酮2.5%和羧甲基纤维素钙3%造粒,60℃真空干燥,得到第一颗粒。干颗粒过12目尼龙筛整理,然后将此颗粒与10.5g的硬脂酸镁,10.5g的滑石粉充分混合,再过12目尼龙筛,颗粒经含量测定合格后,用12mm冲压片,灭菌、包装后即得咀嚼片。
对制得的咀嚼片进行质量检测,结果如下:

2.理化指标:
咀嚼片平均片重为1g/片,片重差异<3%。按照中国药典的规定对咀嚼片的重金属含量进行测定,测定结果为铅<0.2mg/kg,砷<0.1mg/kg。
依据中国药典规定的微生物限度检测法对咀嚼片进行微生物检测,检测结果如下:细菌总数<1000cfu/g,霉菌总数<20,大肠菌群、金黄色葡萄球菌和沙门氏菌均为检出。
3.硬度指标:
对咀嚼片的硬度进行测试,测试结果为4kg左右,完全符合片剂规定。
实施例4:
取枸杞糖肽提取物500g,黑果枸杞提取物100g,叶黄素0.12g,D-甘露糖醇120g,山梨醇粉50g,阿斯巴甜1.8g,L-薄荷脑3.0g,放入多维混合机中混合30分钟。待充分混合均匀后,缓慢加入适量的水将混合好的物料制成软材,软材的干湿度应适宜,以用手紧握能成团但不粘手,用手指轻压能裂开为宜。将制好的软材投入摇摆式颗粒机中以14目筛网进行制粒,然后将湿颗粒置于热风循环干燥箱中在60℃以下干燥,0.5h翻动一次,以加快干燥速度,当颗粒中水分降至4.0-6.0%时停止干燥。干燥颗粒用整粒机16目整粒,得均匀颗粒,然后加入适量微粉硅胶和硬脂酸镁,置多维混合机中混合30分钟。混合好后取出,用压片机进行压片,共压制1000片,即得含片。
本次实例所得含片的感官指标、理化指标和硬度指标均符合要求。
Claims (6)
1.一种具有改善视力的组合物,所述的组合物包含有枸杞和黑果枸杞;所述的黑果枸杞全部或部分用黑果枸杞提取物替代;
所述的黑果枸杞提取物的其制备方法,包括如下的步骤:
1)将清洗后的黑果枸杞高速剪切后,按照料液比1:5~8加入添加了5%的Vc和1%的柠檬酸的60%乙醇水溶液进行提取;提取温度为60℃,提取时间为1~3h,管式离心得到提取液;
2)将提取液减压浓缩,上样至大孔吸附树脂混合树脂模拟床,用纯水洗脱后,再用60%~80%乙醇溶液洗脱,回收乙醇洗脱液,干燥、灭菌得到黑果枸杞提取物;
所述的大孔树脂混合模拟床包含有2~4根MAR柱;每根MAR柱之间串联和/或并联,其中至少有2根大孔吸附树脂柱串联,大孔吸附树脂柱径高比为1:8~1:12;每根柱子所用大孔吸附树脂均为至少2种或几种以不同比例混合的树脂;流速为4~10BV/HR;
所述的树脂为XDA-8、D101、AB-8,其比例为1:1:2。
2.如权利要求1所述的组合物,其特征在于,所述的组合物,其中枸杞全部或部分用枸杞糖肽提取物替代;所述的枸杞糖肽提取物的制备方法如下:
1)将清洗后的枸杞子高速剪切后用水进行提取,然后将获得的提取液离心;
2)用分子量为3500Da的膜系统浓缩,干燥后得到枸杞糖肽提取物,将硫酸苯酚法检测,其糖肽含量>50%。
3.一种黑果枸杞提取物的其制备方法,其特征在于,所述的方法包括如下步骤:
1)将清洗后的黑果枸杞高速剪切后,按照料液比1:5~8加入添加了5%的Vc和1%的柠檬酸的60%乙醇水溶液进行提取;提取温度为60℃,提取时间为1~3h,管式离心得到提取液;

4.如权利要求1所述的组合物,其特征在于,所述的组合物中还添加有叶黄素。

6.一种改善视力功效的制剂,其特征在于,所述的制剂中包含有权利要求1或4所述的组合物。
