本发明公开了一种利用黄精药渣制备水不溶性膳食纤维的方法,包括如下步骤:预处理、脱脂、盐酸处理、碱提、乙醇洗、冷冻干燥和粉碎,本发明以黄精药渣为原料,经脱脂处理、盐酸处理和碱提处理,有效的分解黄精药渣中蛋白质、淀粉和脂肪等成分,制备出纯度高的水不溶性膳食纤维产品,实现了黄精药渣的综合利用;制备的不溶性膳食纤维的纯度高,具有较好的感官品质,同时持水力和膨胀力较高;本发明水不溶性膳食纤维制备方法简单,可操作性强,易于实现工业化操作。本方法与传统的化学分离法相比,提取条件温和,膳食纤维损失少,色泽好,产生的废液较少。

技术领域
本发明涉及功能性食品领域,具体是一种利用黄精药渣制备水不溶性膳食纤维的方法。
背景技术
膳食纤维是当前国际上公认的一种功能性食品基料,被称为除了水、蛋白质、糖类、脂类、维生素、矿物质6大营养素之外的“第七营养素”,并被誉为21世纪食品工业中最受关注、具有特殊保健功能和食品改性作用的一种食品添加剂。膳食纤维可以作为一种有效的功能性保健食品预防多种疾病,并具有保水性、保油性、改进食品色泽及风味等特殊功效,能够改善人体消化吸收,增强人体排泄功能,促进人体的膳食结构平衡,阻碍人体对一些富营养的过多吸收,保持机能平衡,具有抗癌、降压、降血脂、减肥及预防各类结石等作用。因此,对于具有抗氧化作用的膳食纤维中的研究无疑对维系人体健康有着重要的意义。
膳食纤维可分为水溶性纤维(SDF)和水不溶性纤维(IDF)两大类,SDF主要有寡糖(低聚异麦芽寡糖、菊粉等)、抗性糊精、改性纤维素、合成多糖以及植物胶体等,IDF主要包括纤维素、半纤维素和木质素等。膳食纤维在蔬菜、水果、粗粮杂粮、豆类及菌藻类食物中含量丰富。
发明内容
本发明的目的在于提供一种利用黄精药渣制备水不溶性膳食纤维的方法,以解决上述背景技术中提出的问题。
为实现上述目的,本发明提供如下技术方案:
一种利用黄精药渣制备水不溶性膳食纤维的方法,包括如下步骤:
(1)预处理:黄精经水提取后得到的药渣经干燥、粉碎、过筛后得到黄精药渣粉;
(2)脱脂:向黄精药渣粉中加入乙酸乙酯脱脂,黄精药渣粉与乙酸乙酯的质量比为1:5-8,萃取,固液分离,同时回收乙酸乙酯;固相干燥后得脱脂药渣a;
(3)盐酸处理:在步骤(2)所得脱脂药渣a中加入20-30倍的0.35%HCl溶液,处理120-150min,过滤,将所得固相水洗至中性,得滤渣b;
(4)碱提:在步骤(3)所得滤渣b中加入浓度为0.1-0.3mol/L的NaoH溶液,搅拌均匀,其中药渣a与NaoH溶液的比即料液比为1:12-1:30g/mL,将固液混合物浸提50-120min,浸提温度35-55℃,固液分离,将所得固相水洗至中性,得滤渣c;
(5)乙醇洗:在步骤(4)所得滤渣c用乙醇洗60-90min,至无过氧化氢残留,真空抽滤;
(6)冷冻干燥和粉碎:将滤渣在冷冻干燥机中干燥,干燥至含水量低于5%,再经超微粉碎机粉碎至8O-100目的粉状,即得黄精不溶性膳食纤维。
优选地,步骤(1)中所述的黄精药渣的干燥是在温度40℃-80℃下热风干燥6-10h,干燥至水份含量低于10%。
步骤(2)中萃取温度50-65℃,萃取次数为2-4次,每次萃取时间为1-3h。
