技术领域
本发明涉及从植物中提取有效成分技术,具体是一种柿子籽中黄酮的提取方法。
背景技术
柿子是人们比较喜欢食用的果品,甜腻可口,营养丰富,不少人还喜欢在冬季吃冻柿子,别有味道。柿子营养价值很高,含有丰富的胡萝卜素、核黄素、维生素等微量元素。所含维生素和糖分比一般水果高1-2倍左右。假如一个人一天吃1个柿子,所摄取的维生素C基本上就能满足一天需要量的一半。柿叶中含有多种活性成分,如维生素C、多种黄酮甙类、二萜类、胆碱、β-胡萝卜素等。柿叶提取物具有抗氧化性,可与茶多酚、橙皮甙等这些天然抗氧化物相媲美,有望开发为新型天然食品抗氧化剂。近几年来,柿皮的综合利用大幅度提高。柿皮中类胡萝卜素、总酚、维生素C等活性物质含量极为丰富。陈栓虎等致力于对柿子皮综合利用和深加工的研究,先后成功研制了柿子果胶、柿皮膳食纤维添加剂、柿皮果冻、柿皮果酒及柿皮软糖等产品,提高了柿子的经济价值。然而每个柿子中一般含有7~12颗柿子籽,柿子籽的重量占比较大,现在对柿子籽的深加工研究较少。柿子籽的黄酮含量较高,黄酮类化合物具有良好的降血糖、调血脂、抗脂质过氧化、增加胰岛素与受体结合力等功能和作用,并能降低空腹血糖,改善糖耐量,能降低血清和肝组织中脂质过氧化产物丙二醛含量,以及增加血清中超氧化物歧化酶活力,增加胰岛素敏感指数等很多生理功能。但目前尚无有效提取方法的报道和未见有此类产品上市的信息。
发明内容
本发明针对现有技术的不足,提供一种柿子籽中黄酮的提取方法。
为了实现上述目的,本发明采用了以下技术方案:
一种柿子籽中黄酮的提取方法,包括如下步骤:
(1)将柿子籽从柿子果实中剥离出来后,晒干并粉碎,得到柿子籽粉末;
(2)将柿子籽粉末、体积分数不低于70%的乙醇以及月桂酸甲酯按照1∶20~30∶5~10的重量份数之比混合,用碱液将pH调节至8~10,然后置于40~60℃的水浴中加热,在搅拌的同时施加超声场作用,最后冷却至室温后过滤去渣,滤液为黄酮粗提液;
(3)利用超滤膜对黄酮粗提液进行过滤,再将超滤膜的透过液用反渗透膜进行浓缩,取反渗透膜截留液,
(4)向反渗透膜截留液中加入硅胶进行吸附,再用正己烷和氯仿的混合物为洗脱剂进行洗脱,正己烷进行重结晶,即得。
进一步的,所述步骤(1)中,柿子籽晒干后水分含量低于5%。
进一步的,所述步骤(1)中,柿子籽粉末小于100目。
进一步的,超声场的参数为:超声功率为30~45W/cm2,超声频率为35~45KHz,超声作用时间为2~3h。
进一步的,所述反渗透膜的分子截留量为400~500Da。

(3)利用超滤膜对黄酮粗提液进行过滤,再将超滤膜的透过液用反渗透膜进行浓缩,取反渗透膜截留液,
(4)向反渗透膜截留液中加入硅胶进行吸附,再用正己烷和氯仿的混合物为洗脱剂进行洗脱,正己烷进行重结晶,即得。

实施例3
一种柿子籽中黄酮的提取方法,包括如下步骤:
(1)将柿子籽从柿子果实中剥离出来后,晒干并粉碎,得到柿子籽粉末;
(2)将柿子籽粉末、体积分数85%的乙醇以及月桂酸甲酯按照1∶25∶8的重量份数之比混合,用碱液将pH调节至9,然后置于50℃的水浴中加热,在搅拌的同时施加超声场作用,最后冷却至室温后过滤去渣,滤液为黄酮粗提液;
(3)利用超滤膜对黄酮粗提液进行过滤,再将超滤膜的透过液用反渗透膜进行浓缩,取反渗透膜截留液,
(4)向反渗透膜截留液中加入硅胶进行吸附,再用正己烷和氯仿的混合物为洗脱剂进行洗脱,正己烷进行重结晶,即得。
进一步的,所述步骤(1)中,柿子籽晒干后水分含量低于5%。
进一步的,所述步骤(1)中,柿子籽粉末75目。
进一步的,超声场的参数为:超声功率为38W/cm2,超声频率为40KHz,超声作用时间为2.5h。
进一步的,所述反渗透膜的分子截留量为450Da。
进一步的,所述超滤膜0.012μm。
Claims (6)
1.一种柿子籽中黄酮的提取方法,其特征在于,包括如下步骤:
(1)将柿子籽从柿子果实中剥离出来后,晒干并粉碎,得到柿子籽粉末;
(2)将柿子籽粉末、体积分数不低于70%的乙醇以及月桂酸甲酯按照1∶20~30∶5~10的重量份数之比混合,用碱液将pH调节至8~10,然后置于40~60℃的水浴中加热,在搅拌的同时施加超声场作用,最后冷却至室温后过滤去渣,滤液为黄酮粗提液;
(3)利用超滤膜对黄酮粗提液进行过滤,再将超滤膜的透过液用反渗透膜进行浓缩,取反渗透膜截留液,
(4)向反渗透膜截留液中加入硅胶进行吸附,再用正己烷和氯仿的混合物为洗脱剂进行洗脱,正己烷进行重结晶,即得。
2.如权利要求1所述的柿子籽中黄酮的提取方法,其特征在于,所述步骤(1)中,柿子籽晒干后水分含量低于5%。
3.如权利要求1所述的柿子籽中黄酮的提取方法,其特征在于,所述步骤(1)中,柿子籽粉末小于100目。
4.如权利要求1所述的柿子籽中黄酮的提取方法,其特征在于,超声场的参数为:超声功率为30~45W/cm2,超声频率为35~45KHz,超声作用时间为2~3h。
5.如权利要求1所述的柿子籽中黄酮的提取方法,其特征在于,所述反渗透膜的分子截留量为400~500Da。
6.如权利要求1所述的柿子籽中黄酮的提取方法,其特征在于,所述超滤膜0.01~0.015μm。
