本发明提供一种具有辅助降血糖作用的固体饮料及其制备方法,所述固体饮料以重量份计包含:芦荟凝胶0.5~2份、水溶性膳食纤维60~75份、L‑阿拉伯糖10~25份、茶多酚0.05~0.1份、苦瓜提取物1.0~5.0份、酵母提取物0.01~0.05份、桑叶提取物5~15份、植物甾醇酯3~8份以及α‑环糊精1.0~5.0份。本发明提供的组合物在降血糖效果方面显著优于单独使用芦荟凝胶、不溶性膳食纤维、L‑阿拉伯糖、茶多酚、苦瓜提取物或者其中的任意组合物,具有明显的有益效果。

技术领域
本发明涉及一种具有辅助降血糖作用的固体饮料及其制备方法,属于食品生产领域。
背景技术
据国际糖尿病组织(IDF)最新数据显示,全世界有3亿7千多万人患有糖尿病,每年有480万人死于糖尿病,其中90%以上的糖尿病患者患的是II型糖尿病。在中国,总糖尿病患病率为9.7%(在男性和女性中分别为10.6%和8.8%),另外,我国糖尿病前期患病率已达到15.5%。中国的II型糖尿病患者占到了总糖尿病患者的95%。柳叶刀评论认为中国糖尿病的发展随着中国经济的快速发展,生活条件的改善使得人们吃到更多高脂高糖食物,同时久坐不动的生活方式和生活压力的增大加剧了II型糖尿病这种富贵病的增多,而人们对它的认知还远远不够。
对糖尿病患者,除需控制血糖外,尚需控制高血压、血脂异常和凝血功能异常。糖尿病患者发生以动脉粥样硬化疾病为特征的大血管病变的危险,是非糖尿病人群的3-4倍,而且病变发生早,进展快,是糖尿病患者死亡的最主要原因。这种大血管病变导致的死亡,与糖尿病人的血脂代谢异常密切相关。糖尿病患者血脂异常的特点是:1、甘油三酯升高(有30-40%的病人甘油水平>2.25mmol/L);2、餐后血脂水平高于普通人群;3、高密度脂蛋白胆固醇(可防止血管硬化的脂蛋白)下降;4、致病性很强的LDL由于发生糖化和氧化而使消除速度减慢,故其对糖尿病大血管病变的危害性最大。
作为一种常见的代谢性疾病,II型糖尿病常常会伴有冠心病、动脉粥样硬化、肾脏病变、神经病变、视网膜病变以及足部病变等并发症。糖尿病人主要死于糖尿病的并发症。有研究表明,糖尿病风险系数主要包含年龄增长、肥胖、高血压、高血脂和种族因素。如果这些条件同时得到满足,则表明这些人具有高发糖尿病的风险。此外,糖尿病家族史、缺乏体力运动也会提高糖尿病人的患病率。II型糖尿病的发病机制主要包括由胰岛素抵抗引起的炎症反应、线粒体功能受损和氧化应激、内质网应激、能量代谢紊乱、SIRT1信号通路下调以及中枢调控紊乱等。
糖尿病前期治疗主要依赖于保养,需要以营养治疗为基础长期严格执行。然而,糖尿病饮食配制是一项繁琐复杂的过程,患者不易掌握和做到,由于传统饮食习惯,有很大部分病人出院后很快就放弃饮食治疗。所以开发一种既能有效控制糖尿病病人的血糖血脂情况,又能被病人很好地理解掌握、并在其日常生活中运用,便于病人能长期坚持的保健品显得尤为重要。大量研究显示合理有效的饮食干预对于血糖稳定性的维持有良好作用,可减少和预防血糖骤升骤降而引起的并发症,并能减少药物维持血糖而造成的经济负担。
发明内容
针对上述需求,本发明提供一种具有辅助降血糖作用的固体饮料及其制备方法。本发明的固体饮料可以冲饮的方式在中晚餐前食用,预防和缓减糖尿病症状和高血脂症状。
本发明的目的通过以下技术方案予以实现:
在一方面,本发明提供一种具有辅助降血糖作用的固体饮料,所述固体饮料以重量份计包含:芦荟凝胶0.5~2份,优选0.1~1.5份、更优选0.1~1份;水溶性膳食纤维60~75份;L-阿拉伯糖10~25份;茶多酚0.05~0.1份;苦瓜提取物1.0~5.0份;酵母提取物0.01~0.05份;桑叶提取物5~15份;植物甾醇酯3~8份;以及α-环糊精1.0~5.0份。
在优选的实施方式中,所述芦荟凝胶提取自天然芦荟,其中芦荟多糖含量为50wt%以上;所述茶多酚提取自绿茶,其中多酚含量为98wt%以上;所述桑叶提取物提取自天然桑叶,其中桑叶黄酮含量大于10wt%;和/或所述苦瓜提取物来自于苦瓜,其中苦瓜皂苷大于0.2wt%。
在另一方面,本发明提供上述固体饮料的制备方法,所述方法包括以下步骤:将膳食纤维与L-阿拉伯糖过筛,选取40-80目筛下细粉,然后按照配比称取各成分,混合均匀。
优选地,本发明的方法具体包括:(1)预处理:对各原料进行干燥灭菌;(2)称重配料:按配方比例称取各原料;(3)混合搅拌:将配好后的粉料装入三维混合机内搅拌30~45min;(4)灌装、包装入库。
在另一方面,本发明还提供了本发明的固体饮料在制备防治高血糖的保健食品中的用途。
本发明提供的组合物在降血糖效果方面显著优于单独使用芦荟凝胶、不溶性膳食纤维、L-阿拉伯糖、茶多酚、苦瓜提取物或者其中的任意组合物,具有明显的有益效果。
具体实施方式
本发明人发现,以天然芦荟提取物为主要活性成分,添加水溶性膳食纤维、L-阿拉伯糖、茶多酚、苦瓜提取物、酵母提取物、桑叶提取物、植物甾醇酯和α-环糊精制成固体饮料,可在保证良好风味口感的同时具有优异的降血糖效果,并且还能在一定程度上缓解高血脂症状。
本发明配方中的芦荟凝胶来源于芦荟(Aloe spp.),即百合科芦荟属植物,其应用范围遍及内、外、妇、儿、皮肤、五官科等,如可用于通便、抗炎、调节免疫、抗菌、促进伤口。芦荟凝胶中的有效成分主要是多糖类成分和芦荟叶表中的黄色液汁(主要活性成分为蒽醌类成分)。一般认为起降血糖作用的成分主要可能是叶皮中的成分,但是也有研究发现凝胶中的多糖类物质对小鼠有降血糖作用。另外,从库拉索芦荟晶状体中提取的苦味素,对四氧嘧啶诱导的糖尿病小鼠的空腹血糖都起明显的降低作用。凝胶部分中的糖蛋白,也能快速降低葡萄糖,叶皮制备物也有降血糖作用,并且可使人工诱导糖尿病动物恢复正常胰岛素分泌量。同时,有研究对大鼠皮下注射库拉索芦荟制备物,发现其能促进伤口愈合,降低对疼痛的敏感性,并减少由芥子气引起的水肿。在接下来的研究发现,芦荟凝胶和其中所含的赤霉素对化学诱导的糖尿病小鼠有几乎相等的消炎作用,说明芦荟中的赤霉素或相似物质是抗炎的活性成分。此外,芦荟对由糖尿病引起的神经、肾脏病变的并发症也有缓解和治疗作用。
本发明中以芦荟凝胶粉的形式使用芦荟凝胶,优选地,该芦荟凝胶粉为库拉斯芦荟和/或好望角芦荟的混合物。最优选地,所述芦荟凝胶粉由30-70wt%库拉索芦荟和30-70wt%好望角芦荟组成。
可溶性纤维,能抑制餐后血糖值的上升,其原因是延缓和抑制对糖类的消化吸收,并改善末梢组织对胰岛素的感受性,降低对胰岛素的要求。水溶性膳食纤维随着凝胶的形成,阻止了糖类的扩散,推迟了在肠内的吸收,因而抑制了糖类吸收后血糖的上升胰岛素升高的反应。此外膳食纤维能改变消化道激素的分泌,如胰汁的分泌减少,从而抑制了糖类的消化吸收,并减少小肠内糖类与肠壁的接触,从而延迟血糖值的上升。因此提高可溶性膳食纤维的摄入量可以防止II型糖尿病的发生。可溶性纤维在胃肠中能形成一种粘膜,使食物营养素的消化吸收过程减慢,从而降低血糖水平。有研究还发现可溶性膳食纤维可以恢复受损的胰岛结构和功能,与茶多酚搭配效果更好。
L-阿拉伯糖是重要的医药中间体,可用来合成抗癌、抗病毒和治疗心血管疾病的药物,而近年来发现L-阿拉伯糖可抑制胃肠道对蔗糖的吸收,使它在减肥、控制糖尿病等方面的应用前景被广泛看好,日本厚生省的特定保健用食品清单中已将L-阿拉伯糖列入调节血糖的专用特殊保健食品添加剂,而美国医疗协会也已将L-阿拉伯糖列入抗肥胖剂的营养补充剂或非处方药。
本发明中所使用的苦瓜提取物和桑叶提取物是通过本领域常规手段以水提或水醇浸提方法分别从苦瓜和桑叶中提取后干燥获得的提取物。例如,专利CN102579296B与CN1520838A中公开的苦瓜提取物、CN1981814A与CN102370708A中公开的桑叶提取物等。
苦瓜为葫芦科苦瓜属植物苦瓜的果实,味苦性寒,无毒。其根、茎、叶、花、果实和种子在世界各地均有药用记载。苦瓜多糖由鼠李糖、L-阿拉伯糖、甘露糖、葡萄糖和半乳糖组成的,含β-D-吡喃糖苷键,是一种活性多糖。苦瓜醇提物也能显著增加正常小鼠肝糖原含量,明显改善正常小鼠的糖耐量,显著降低链唑霉素、四氧嘧啶糖尿病大小鼠的血糖。另外,从苦瓜种子中分离得到的苦瓜皂甙有明显降血糖作用,有类似胰岛素的作用,被誉为“植物胰岛素”。
《本草纲目》等资料记载:桑叶性味甘、平、寒;功能:清肝明目聪耳,镇静神经,润肺热,止咳,手足麻木,赤眼涩痛,解蛇虫毒,可代茶止渴,通关节。现代中药学大都认为桑叶性味甘、苦、寒,入肺、肝经,具有清肺止咳、清肝明目的作用。现代大量药理学研究证明桑叶具降血糖、降血压、清除自由基、抗菌和抗病毒抑制癌细胞等诸多药理作用。自古以来,中医就将桑叶作为治疗消渴证的中药应用于临床。日本古书《吃茶养生记》也有记载桑叶有改善“饮水病”(即糖尿病)的作用。近代医家也常将桑叶配伍于中药复方中应用于临床。研究表明桑叶降血糖作用是通过两个方面实现的:一是通过桑叶生物碱DNJ对二糖类分解酶活性产生抑制作用,从而抑制小肠对双糖吸收,降低食后血糖的高峰值;二是桑叶生物碱Fagomine促进胰细胞分泌胰岛素,而胰岛素可以促进细胞对糖的利用、肝糖原合成以及改善糖代谢,最终达到降血糖效果。
本发明的固体饮料中的植物甾醇酯是一种利用大豆油等植物油馏分或者塔罗油为原料,通过皂化、萃取、结晶等工艺生产制得的天然活性物质。植物甾醇酯可竞争性地阻碍小肠吸收胆固醇,促进胆固醇排出体外,可显著降低血清TC、LDL-C水平,达到降血脂的作用。
本发明中茶多酚是一种较强的抗氧化剂,与可溶性膳食纤维结合后,对糖尿病人的血糖指数有明显的降低效果。
酵母提取物:酵母具有独特的营养及保健价值,不仅直接被开发为营养食品,还可进一步制备成多种营养活性物质,作为营养食品的载体,进一步深加工则成为更具营养和保健价值的食品。酵母在食品领域中的应用历史悠久,以酿酒和面制品发酵为人们所熟悉,其应用已扩大到药品、食品添加剂、保健食品、调味品、饲料、培养基成分等方面。市场上已经有了比较丰富的各类酵母食品。其中,酵母抽提物就是以酵母为原料开发的食品,它有着极其浓郁的肉香味,可改善食物的风味和口感,已经广泛应用在调味品领域,因其营养丰富,也可用作营养补充剂。以酵母为载体,补充一些微量营养素的保健食品和特殊营养食品,如富铬酵母。即在生产酵母的培养基中,增加铬浓度,从而使酵母中含有较多的这些物质,供人食用。有研究表明,对II型糖尿病患者进行临床试验后发现,糖尿病患者通过摄取一定量的铬微量元素后,其血糖有明显降低。
α-环糊精是一种常见的环糊精(Cyclodextyin,CD),由环糊精葡萄糖残基转移酶(Cyclodextrin glycosyltransferase,CGTase)作用于淀粉、糖原、麦芽寡聚糖等葡萄糖聚合物而形成的,由6~12个D-吡喃葡萄糖基以α-1,4-葡萄糖苷键连接而成的环状低聚糖。这种独特的结构可与许多无机、有机分子结合成主客体包合物,并能改变被包合物的化学和物理性质,具有保护、稳定、增溶客体分子和选择性定向分子的特性,广泛应用于食品中以掩盖一些食品原料的不良气味。
芦荟、水溶性膳食纤维、L-阿拉伯糖、茶多酚、酵母提取物、桑叶提取物六种原料均有降低血脂、调节血糖、延缓衰老、增强免疫等功效,该发明配方中的L-阿拉伯糖既改善了本品的口感,而又能让喜欢甜食的糖尿病人得到满意,吃到甜味。该固体饮料处方和剂量配伍合理,有较高的创新性。临床试用表明,这种降血糖的固体饮料具有降血糖、降血脂、益气提神、修复胰岛、有效缓解“三多一少”症状的作用;并且无毒副作用,是降血糖、降血脂的理想药食两用的天然保健食品。
对18-65岁糖尿病史前期(空腹血糖值100-126mg/dl,2小时糖耐量>140-199mg/dl)人群,每天给予两次本发明所述的固体饮料,餐前服用,每次10g,经2-6个月后,精力很好,血糖值下降的人群比例占总人数的90%。
以下通过具体实施例来对本发明进行进一步的描述,本发明实施例中所使用的试验方法如无特殊说明,均为常规方法;所使用的原料、试剂等,如无特殊说明,均为可从常规市购等商业途径所得的原料和试剂。
实施例
接下来,通过实施例对本发明进行进一步详细地说明,但本发明不仅限于这些实施例。
如无其它说明,在以下实施例中所使用的原料如下所列:水溶性膳食纤维为日本松谷生产;L-阿拉伯糖为山东福田公司生产;茶多酚为福建大闽生产、酵母提取物为湖北安琪酵母公司生产(铬含量不低于20ppm)、植物甾醇酯为中粮天科生物工程(天津)有限公司生产,α-环糊精由山东西王集团生产。
本发明实施例中,芦荟凝胶生产方法为:芦荟叶片蒸馏水洗净,切成小块,组织捣碎机捣碎匀浆5min;匀浆液加10-20倍体积的蒸馏水于60-80℃浸提4-6h,2000-3000rpm离心20-30min,收集上清,沉淀再加10-20倍体积的蒸馏水,60-80℃热水浸提2-3h,2000-3000rpm离心15min,收集上清,合并两次上清,旋转蒸发仪55℃减压浓缩至原体积的70-80%。浓缩液加3-5倍体积95%乙醇,充分混匀,静置过夜,收集沉淀,得芦荟凝胶,室温干燥,得粉状样品,其中多糖含量为50%以上。
桑叶提取物:将桑叶捣碎,70-90℃热水浸提4-6h,过滤,温度在70℃以下进行真空浓缩,浓缩到原来体积的50-70%时,90℃进行灭菌20-30min,然后喷雾干燥得到桑叶提取物,其中桑叶黄酮含量不低于10%。
苦瓜提取物:将苦瓜洗净、切碎成1-2cm小块,按照原料6倍量40%乙醇回流提取2次,每次2h,合并过滤过60目筛;在温度≤60℃,-0.06MPa≤真空度≤-0.07Mpa条件下,浓缩液相对密度1.10~1.12(60℃)下减压浓缩;然后进行喷雾干燥。
实施例1:
原料配方组成(按重量份计):芦荟凝胶粉0.5份、水溶性膳食纤维60份、L-阿拉伯糖15份、茶多酚0.5份、苦瓜提取物5.0份、酵母提取物0.05份、桑叶提取物10份、植物甾醇酯4份和α-环糊精5.0份。
将上述原料芦荟凝胶粉、水溶性膳食纤维、L-阿拉伯糖、茶多酚、苦瓜提取物、酵母提取物、桑叶提取物、植物甾醇酯和α-环糊精分别进行干燥灭菌,按配方比例称取原料装入三维混合机内搅拌30min,然后进行分装、入库。
实施例2:
原料配方组成(按重量份计):芦荟凝胶粉0.8份、水溶性膳食纤维65份、L-阿拉伯糖15份、茶多酚0.5份、苦瓜提取物5.0份、酵母提取物0.05份、桑叶提取物10份、植物甾醇酯4份和α-环糊精5.0份。
将上述原料芦荟凝胶粉、水溶性膳食纤维、L-阿拉伯糖、茶多酚、苦瓜提取物、酵母提取物、桑叶提取物、植物甾醇酯和α-环糊精分别进行干燥灭菌,按配方比例称取原料装入三维混合机内搅拌35min,然后进行分装、入库。
实施例3:
原料配方组成(按重量份计):芦荟凝胶粉1份、水溶性膳食纤维70份、L-阿拉伯糖15份、茶多酚0.5份、苦瓜提取物5.0份、酵母提取物0.05份、桑叶提取物10份、植物甾醇酯4份和α-环糊精5.0份。
将上述原料芦荟凝胶粉、水溶性膳食纤维、L-阿拉伯糖、茶多酚、苦瓜提取物、酵母提取物、桑叶提取物、植物甾醇酯和α-环糊精分别进行干燥灭菌,按配方比例称取原料装入三维混合机内搅拌40min,然后进行分装、入库。
实施例4:
原料配方组成(按重量份计):芦荟凝胶粉0.5份、水溶性膳食纤维75份、L-阿拉伯糖15份、茶多酚0.5份、苦瓜提取物5.0份、酵母提取物0.05份、桑叶提取物10份、植物甾醇酯4份和α-环糊精5.0份。
将上述原料芦荟凝胶粉、水溶性膳食纤维、L-阿拉伯糖、茶多酚、苦瓜提取物、酵母提取物、桑叶提取物、植物甾醇酯和α-环糊精分别进行干燥灭菌,按配方比例称取原料装入三维混合机内搅拌40min,然后进行分装、入库。
效果例
将实施例4的固体饮料与比较例1、比较例2配方的固体饮料提供给受试动物,在投食28日后进行血糖检测。其中,比较例1的配方为:水溶性膳食纤维80份、L-阿拉伯糖15份、茶多酚0.5份和α-环糊精5.0份;比较例2的配方为:水溶性膳食纤维80份、L-阿拉伯糖15份、苦瓜提取物5.0份、酵母提取物0.05份和α-环糊精5.0份。
选用体重152-175g,SPF级雄性Wistar大鼠40只作为受试动物进行实验,每组实验大鼠10只。结果如表1所示。

以实施例4、样品1、样品2和安慰剂灌胃大鼠28天后,各组动物生长活动良好,各剂量组动物增重与安慰剂对照组相比,差异无统计学意义(P>0.05);对于28日后的空腹血糖值和餐后血糖值,实施例4的效果优于样品1、样品2和安慰剂组,且差异具有统计学意义(P<0.05)。可见,本发明的固体饮料具有明显更优异的降血糖效果。
Claims (3)
1.一种具有辅助降血糖作用的固体饮料,所述固体饮料以重量份计包含:芦荟凝胶0.5~1份;水溶性膳食纤维60~75份;L-阿拉伯糖15~25份;茶多酚0.5份;苦瓜提取物1.0~5.0份;酵母提取物0.01~0.05份;桑叶提取物10~15份;植物甾醇酯4~8份;以及α-环糊精1.0~5.0份;
其中,所述芦荟凝胶提取自天然芦荟,其中芦荟多糖含量为50wt%以上;所述茶多酚提取自绿茶,其中茶多酚含量为98wt%以上;和/或所述桑叶提取物提取自天然桑叶,其中桑叶黄酮含量为10wt%以上;
所述固体饮料的制备方法包括以下步骤:将膳食纤维与L-阿拉伯糖过筛,选取40-80目筛下细粉,然后按照配比称取各成分,混合均匀;
所述固体饮料的制备方法具体包括:(1)预处理:对各原料进行干燥灭菌;(2)称重配料:按配方比例称取各原料;(3)混合搅拌:将配好后的粉料装入三维混合机内搅拌30~45min;(4)灌装、包装入库。
2.如权利要求1所述的固体饮料,其中酵母提取物中铬含量不低于20ppm,植物甾醇酯纯度为90wt%以上。
3.权利要求1或2所述的固体饮料在制备辅助降血糖的保健食品中的应用。
