Abstract
本发明属于中药技术领域,涉及一种制备治疗老年性骨折的中药组合物的方法。该制备方法是以鹰嘴豆豆芽提取物、黄芪、党参、红花、川芎、当归、熟地、黄瓜子、三七、甘草、乳香、没药和白胶香为原料制得中药组合物。本发明的制备方法简便、操作方便,适合现代制药工艺批量、快速生产的特点。而且,制得的中药组合物在治疗老年人骨折方面,具有补气行血、活血散瘀、接骨续筋、强筋壮骨等显著的治疗优势。

技术领域
本发明属于中药技术领域,尤其涉及一种制备治疗老年性骨折的中药组合物的方法。
背景技术
骨折,外伤或疾病所致之骨部分或完全折断者,见《外台秘要》卷二十九,又名折骨、折伤、伤折、折疡。骨折是指由于外伤或病理等原因致使骨质部分或完全断裂的一种疾病。其主要临床表现为:骨折部有局限性疼痛和压痛,局部肿胀和出现瘀斑,肢体功能部位或完全丧失,完全性骨质尚可出现肢体畸形及异常活动。骨折是临床上的常见病症,多由外力作用下引发,骨折不仅给患者带来疼痛,还会在临床表现上出现一系列并发症,如发热症状,及患者因广泛的软组织损伤、大量出血、剧烈疼痛等而引起的周围组织损伤,重要血管损伤,周围神经损伤,骨筋膜室综合征,损伤性关节炎等,常见于儿童和老年人。
老年人由于腿脚不便,身体不灵活等原因,常常容易发生骨折,而且老年人由于组织愈合能力差,经常合并全身性疾病等,骨折愈合能力较差,愈合的时间较长。目前市场上专门用于治疗老年人骨折的中药配方甚少,使用较多的是活血化瘀、续筋接骨、滋养肝肾、补气非血的中药,而老年人由于机体功能衰退,造血能力下降,血液不流通等导致骨折愈合的时间更长,因此,寻找一种益气养血,补气行血,促进老年人血气运行的中药配方尤其重要。
鹰嘴豆享有很高的声誉和广泛的药用价值,在维吾尔语的医学中已沿用了2500多年。它含有丰富的易被人体吸收和消化的植物蛋白质、赖氨酸、粗纤维、亚油酸、维生素和镁、钙、铁等人体必需的营养成份。具有补中益气、温肾壮阳、润肺止咳、强身健体、增强记忆力、治疗糖尿病和肺病等功效。尤其对胸痛、胸膜炎的治疗有独到的疗效,并有解毒、消炎、养颜、抗衰老的作用,是适合高血糖、糖尿病患者以及中老年人长期食用的一种高蛋白、低淀粉、纯天然的食品,被誉为“营养之花、豆中之王”。
鹰嘴豆在医学领域的药用价值越来越被人们重视,为了进一步提高鹰嘴豆产品的附加值,新疆理化所科研人员通过与外国专家合作,对鹰嘴豆的化学成份进行了分析研究。研究结果证明,鹰嘴豆的全豆或纤维部分成份很适合制做低血糖指数食品,并有望研制成具有降糖功能的鹰嘴豆医药产品。新疆理化所植物资源化学实验室还与上海新药创新中心合作开展了鹰嘴豆粉降糖功能动物实验。结果表明,鹰嘴豆对四氧嘧啶所致小鼠血糖升高有一定的降糖作用,并且至今没有文献报道鹰嘴豆及其提取物在骨折方面的用途。
中国发明专利201210325056.5公开了一种治疗骨折的药物,该药物包括以下重量份数的原料:当归2-20份、淫羊藿2-20份、鹿角胶2-20份、地龙2-20份、川断10-30份、骨碎补2-20份、煅自然铜1-10份、脆蛇2-20份、儿茶2-20份、土鳖虫1-11份,龙骨5-25份和薏苡仁5-25份,该发明的药物在治疗骨折方面起到一定的疗效,但对于治疗老年人骨折方面效果不是十分理想。
发明内容
本发明要解决的技术问题是提供一种制备治疗老年性骨折的中药组合物的方法,该中药组合物能有效的益气补血,活血化瘀,促进老年人的血气运行,本发明提供相应的制造工艺将该中药在保证疗效的前提下制成方便易用的成品制剂。
为了解决该技术问题,发明人在配伍组方的同时,充分考虑到现代制药工艺批量、快速生产的特点,通过临床实践的反复验证,调整药材的量和种类,最终发明了一个全新的中药组合物和其制备工艺,其制备工艺简便、操作方便。该中药组合物包括以下重量份数的制备原料:鹰嘴豆豆芽提取物5-30份、黄芪6-15份、党参9-30份、红花3-9份、川芎3-9份、当归6-12份、熟地9-15份、黄瓜子3-10份、三七3-9份和甘草1.5-15份。
进一步的,该中药组合物包括以下重量份数的制备原料:鹰嘴豆豆芽提取物20份、黄芪10份、党参20份、红花6份、川芎6份、当归9份、熟地12份、黄瓜子7份、三七6份和甘草9份。
进一步的,该中药组合物还包括以下重量份数的制备原料:乳香3-10份和没药3-10份。
进一步的,该中药组合物还包括以下重量份数的制备原料:白胶香1.5-5份。
进一步的,该中药组合物包括以下重量份数的制备原料:鹰嘴豆豆芽提取物20份、黄芪10份、党参20份、红花6份、川芎6份、当归9份、熟地12份、黄瓜子7份、三七6份、甘草9份、乳香6份、没药6份和白胶香3份。
再进一步的,该中药组合物被制成中药冲剂、颗粒剂、散剂、胶囊剂、水丸剂或片剂。
本发明制备治疗老年性骨折的中药组合物的方法,包含下述步骤:
(1)制备鹰嘴豆豆芽提取物:将鹰嘴豆经3-4天发芽,采摘豆芽,晾干,粉碎成粗粉,加入重量20倍浓度为70%的乙醇水溶液,浸泡至透心,加热至90℃,回流提取3次,每次1.5小时,过滤提取液,得到滤液;把滤液减压回收乙醇至药液含醇浓度为5-20%时,经大孔吸附树脂柱吸附8h,用蒸馏水和乙醇洗脱至无色,在60℃、-0.1Mpa条件下减压浓缩,粉碎,过80-100目筛,得到鹰嘴豆豆芽提取物;
(2)将重量份当归6-12份、熟地9-15份、黄芪6-15份和党参9-30份经超声常规水提取工艺提取,制成浸膏细粉;
(3)将重量份三七3-9份、乳香3-10份、红花3-9份、川芎3-9份、没药3-10份、白胶香1.5-5份、黄瓜子3-10份和甘草1.5-15份分别研成细粉,与上述鹰嘴豆豆芽提取物5-30份、浸膏细粉混合,搅拌均匀,即得中药组合物。
上述超声常规水提取工艺包括以下步骤:
(1)称取原料加水浸泡过夜,常温超声提取3次,每次30分钟;
(2)将步骤(1)所得的提取液抽滤,得到滤液后合并滤液,用水和80%乙醇溶液洗脱至无色,减压回收,烘干得到粗品;
(3)将步骤(2)所得的粗品用95%乙醇溶液超声溶解,抽滤,获得滤液后将滤液减压回收至干,即得浸膏细粉。
本发明所用组分的来源、性味、归经及功效:
红花:本品为菊科植物红花的花;味辛、苦,性温;归肝、心脾经;活血通经、利气、祛瘀止痛。
川芎:本品为伞形科植物川芎的干燥根茎;味辛,性温;归肝、胆、心包经;活血行气、祛风止痛。
乳香:本品为橄榄科乳香树属植物低温树干皮部伤口渗出的油胶树脂;味辛、苦,性温;入心、肝、脾经;活血止痛。
没药:本品为橄榄科没药属植物的树干皮部渗出的油胶树脂;味苦,性平;入肝经;散瘀止痛,外用消肿生肌。
白胶香:本品为金缕梅科植物枫香的树脂;味辛、苦,性平;活血、凉血、解毒、止痛。
当归:本品为伞形科植物当归的干燥根;味甘、辛,性温;补血活血、调经止痛。
熟地:本品为生地黄的炮制加工品;味甘,性微温;归肝、肾经;滋阴补血、益精填髓。
黄瓜子:本品为葫芦科植物黄瓜的种子;味甘、凉,无毒;补钙、接骨、壮骨。
三七:本品为植物三七的根茎;味甘微苦,性平;归肝、胃、大肠经;止血、活血化瘀、消肿定痛、滋补强壮。
黄芪:本品为豆科植物黄芪的根;味甘,微温;归肺、脾、肝、肾经;补气固表、托毒排脓、利尿、生肌。
党参:本品为桔梗科植物党参的干燥根;味甘,性平;归脾、肺经;补中益气、健脾益肺。
甘草:本品为豆科植物甘草、胀果甘草或光果甘草的干燥根。味甘,性平;归心、肺、脾、胃经;补脾益气,清热解毒,祛痰止咳,缓急止痛,调和诸药。
本发明中药组方的组方分析:
本发明中药组方是以黄芪、党参、鹰嘴豆豆芽提取物为君药,养气活血,补血,补中益气;以当归、红花、三七、川芎、熟地为臣药,补气固表,活血通经,祛瘀止痛,以黄瓜子、乳香、没药、白胶香为佐药,凉血解毒,补钙强身,祛风止痛,消肿生肌;以甘草为使药,调和诸药性,使诸药搭配,协同作用,共同达到益气养血,活血化瘀,接骨止痛、补气行血、活血散瘀、接骨续筋、强筋壮骨等功效,对老年性骨折有十分显著的治疗效果。
本发明中黄芪和党参均有补气益气的作用,两者一起使用效果更佳;鹰嘴豆豆芽富含叶酸、钾、镁、磷、锌、铜和维生素B1,还含有一定数量的烟酸、维生素B6、泛酸、钙和纤维,在补血、补钙等方面作用明显。
与现有技术相比,本发明具有如下技术优势:
1、本发明的制备方法简便、操作方便,适合现代制药工艺批量、快速生产的特点。
2、本发明中药组合物采用了鹰嘴豆豆芽提取物,通过鹰嘴豆豆芽提取物与其他中药组分的协同作用,在益气补血和补钙强身方面效果更佳。
3、本发明中药组合物在治疗骨折方面具有疗效好,治愈时间快等显著的治疗优势,尤其针对老年人骨折方面效果显著;本发明中药组合物主要针对老年人气血不足导致骨折愈合程度慢的问题,具有益气补血,活血止痛,促进老年人气血运行的显著效果。

实施例1:
本发明实施例1中药组合物包括以下重量份数的原料:鹰嘴豆豆芽提取物20份、黄芪10份、党参20份、红花6份、川芎6份、当归9份、熟地12份、黄瓜子7份、三七6份、甘草9份、乳香6份、没药6份和白胶香3份。
制备方法如下:
S1:制备鹰嘴豆豆芽提取物:将鹰嘴豆经3-4天发芽,采摘豆芽,晾干,粉碎成粗粉,加入重量20倍浓度为70%的乙醇水溶液,浸泡至透心,加热至90℃,回流提取3次,每次1.5小时,过滤提取液,得到滤液;把滤液减压回收乙醇至药液含醇浓度为5-20%时,经大孔吸附树脂柱吸附8h,用蒸馏水和乙醇洗脱至无色,在60℃、-0.1Mpa条件下减压浓缩,粉碎,过80-100目筛,得到鹰嘴豆豆芽提取物;
S2:将当归、熟地、黄芪和党参经超声常规水提取工艺提取,制成浸膏细粉;
S3:将三七、乳香、红花、川芎、没药、白胶香、黄瓜子和甘草分别研成细粉,与上述鹰嘴豆豆芽提取物、浸膏细粉混合,搅拌均匀,即得本发明的中药组合物;
上述超声常规水提取工艺包括下述以下步骤:
S21:称取原料加水浸泡过夜,常温超声提取3次,每次30分钟;
S22:将步骤S21所得的提取液抽滤,得到滤液后合并滤液,用水和80%乙醇溶液洗脱至无色,减压回收,烘干得到粗品;
S23:将步骤S22所得的粗品用95%乙醇溶液超声溶解,抽滤,获得滤液后将滤液减压回收至干,即得浸膏细粉。
实施例2:
本发明实施例2中药组合物包括以下重量份数的制备原料:鹰嘴豆豆芽提取物5份、黄芪6份、党参9份、红花3份、川芎3份、当归6份、熟地9份、黄瓜子3份、三七3份、甘草1.5份、乳香3份、没药3份和白胶香1.5份。

本发明中药组合物药效学研究
试验例一本发明中药组合物对骨折临床志愿者患者的临床疗效观察
一、资料与方法
1.一般资料:本组130例封闭式骨折临床志愿者,其中男78例,女52例,52-79岁,平均年龄68.5岁。
2.疗效标准:
2.1有效:疼痛得到较大的缓解,并且骨折部位红、肿、热均减轻;
2.2无效:疼痛未有缓解,骨折部位红、肿、热均未获得明显减轻。
3.分组与治疗方法
选择骨折患者90例,随机分为6组,每组15人,具体分组与给药如下:
独一味组:口服独一味胶囊(甘肃独一味生物制药有限责任公司,0.3g*50粒/瓶),每日3次,每次3粒;
中药组合物A组:给药本发明实施例1制备的胶囊(0.4g*100粒/瓶),每日3次,每次4粒;
中药组合物B组:给药本发明实施例2制备的胶囊(0.4g*100粒/瓶),每日3次,每次4粒;
中药组合物C组:给药本发明实施例3制备的胶囊(0.4g*100粒/瓶),每日3次,每次4粒;
中药组合物D组:给药本发明实施例4制备的胶囊(0.4g*100粒/瓶),每日3次,每次4粒;
中药组合物E组:给药本发明实施例5制备的胶囊(0.4g*100粒/瓶),每日3次,每次4粒。
备注:本发明实施例1-5的中药组合物按照常规工艺封闭于胶囊壳获得实施例1-5所述的胶囊。
4.数据处理
用药后记录半年内有效人数、痊愈人数和治愈时间。计算有效率=有效人数/病例总人数×100%。应用统计软件SPSS15.0进行各治疗组治愈率和有效率的χ2检验。若χ2>χ2 0.05,1=3.84,以p<0.05具有统计学差异性。并对治愈时间进行t检验比较,结果见表1。
二、试验结果
临床试验结果(见表1)表明:
(1)对于骨折的治疗,与独一味组相比,各治疗组在有效率方面更高,尤其以实施例1的治愈效果最好;
(2)对于骨折的的治疗,本发明考察了不同重量份数配比的中药组合物对骨折的疗效,结果发现,其中尤其以本发明实施例1提供的中药组合物胶囊治愈时间最短。也就是说当本发明中药组合物的制备原料包括鹰嘴豆豆芽提取物20份、黄芪10份、党参20份、红花6份、川芎6份、当归9份、熟地12份、黄瓜子7份、三七6份、甘草9份、乳香6份、没药6份和白胶香3份时,本发明中药组合物治疗骨折效果最好。

1、仪器:日立7020全自动生化分析仪,日本Hitachi公司;骨密度测定仪(Piximusbonedensito-meter,美国LUNAR公司);
2、试验动物:大白兔,雌雄各半,1.5-3.0kg,由中山大学中心提供;
3、试验药物:本发明实施例1制备的中药组合物;独一味胶囊,甘肃独一味生物制药有限责任公司,0.3g*50粒/瓶。
二、试验方法:
1、动物分组:取白兔70只,随机分为5组,每组14只,分别为模型组(等体积蒸馏水),独一味组(1.0g/kg﹒d),本发明中药组合物低剂量组(0.3g/kg﹒d)、中剂量组(0.9g/kg﹒d)和高剂量组(1.8g/kg﹒d);本发明中药组合物制备同实施例1。
2、动物模型的制备:家兔适应性饲养一周后,家兔用3%戊巴比妥钠溶液按照1ml/kg剂量静脉麻醉后,两侧前肢剪毛,75%酒精消毒,在无菌条件下用医用手术牙科钻在两侧桡骨中上段1/3交界处横钻成宽约为3mm,深入骨髓腔形成骨损伤,缝合皮肤,手术后次日开始灌胃给药,连续给药,给药后一周到五周取各组家兔中的7只,测定各项指标。
3、骨密度测定:分别于给药后第三周、第五周截取两侧桡骨,其长度包括损伤及上下各正常段骨质,用美国LUNAR公司Piximusbonedensito-meter测定仪测定骨密度,观察骨折愈合情况。
三、试验结果
试验结果(具体见表2)表明:连续给予本发明的中药组合物三周后,独一味组和本发明中药组合物低中高三个剂量组的骨密度增加,与模型组相比具有显著性差异(p<0.05)。给药五周后,独一味组与模型组相比具有极显著性差异(p<0.01),本发明中药组合物低剂量组、中剂量组和高剂量组与模型组相比有显著性差异(p<0.05),并且五周后的骨密度比三周后的骨密度增大,本发明中药组合物的中高剂量组对骨密度的升高作用与独一味组相当。结果表明本发明的中药组合物具有加快骨矿沉积速率,提高骨密度,促进骨折部位愈合的功能。

Claims (4)
1.一种制备治疗老年性骨折的中药组合物的方法,其特征在于包括下述步骤:
(1)制备鹰嘴豆豆芽提取物:将鹰嘴豆经3-4天发芽,采摘豆芽,晾干,粉碎成粗粉,加入重量20倍浓度为70%的乙醇水溶液,浸泡至透心,加热至90℃,回流提取3次,每次1.5小时,过滤提取液,得到滤液;把滤液减压回收乙醇至药液含醇浓度为5-20%时,经大孔吸附树脂柱吸附8h,用蒸馏水和乙醇洗脱至无色,在60℃、-0.1Mpa条件下减压浓缩,粉碎,过80-100目筛,得到鹰嘴豆豆芽提取物;
(2)将重量份当归6-12份、熟地9-15份、黄芪6-15份和党参9-30份经超声常规水提取工艺提取,制成浸膏细粉;
(3)将重量份三七3-9份、乳香3-10份、红花3-9份、川芎3-9份、没药3-10份、白胶香1.5-5份、黄瓜子3-10份和甘草1.5-15份分别研成细粉,与上述鹰嘴豆豆芽提取物5-30份、浸膏细粉混合,搅拌均匀,即得中药组合物。
2.如权利要求1所述的制备治疗老年性骨折的中药组合物的方法,其特征在于,所述超声常规水提取工艺包括以下步骤:
(1)称取原料加水浸泡过夜,常温超声提取3次,每次30分钟;
(2)将步骤(1)所得的提取液抽滤,得到滤液后合并滤液,用水和80%乙醇溶液洗脱至无色,减压回收,烘干得到粗品;
(3)将步骤(2)所得的粗品用95%乙醇溶液超声溶解,抽滤,获得滤液后将滤液减压回收至干,即得浸膏细粉。
3.如权利要求1所述的制备治疗老年性骨折的中药组合物的方法,其特征在于,所述中药组合物包括以下重量份数的制备原料:鹰嘴豆豆芽提取物20份、黄芪10份、党参20份、红花6份、川芎6份、当归9份、熟地12份、黄瓜子7份、三七6份、甘草9份、乳香6份、没药6份和白胶香3份。
4.如权利要求1所述的制备治疗老年性骨折的中药组合物的方法,其特征在于:所述中药组合物被制成中药冲剂、颗粒剂、散剂、胶囊剂、水丸剂或片剂。
