Abstract

发明背景
植物和植物部分中的乙烯产生被多种外部因素和应激物诱导,包括创伤、施加激素(如生长素)、厌氧条件、寒冷、热、干燥和病原侵染。还在多种植物发育过程(包括果实或蔬菜成熟、种子萌发、叶脱落和花衰老)中观察到乙烯产生增加。
植物中乙烯生物合成通常被描述为牵涉三种酶、习惯上称为"Yang循环"的酶促方案,其中S-腺苷-L-甲硫氨酸(SAM)合成酶催化甲硫氨酸转化为S-腺苷-L-甲硫氨酸(AdoMet);1-氨基环丙烷-1-羧酸(ACC)合成酶催化AdoMet转化为ACC;且ACC氧化酶催化ACC转化为乙烯和副产品二氧化碳和氰化氢。参见例如,Srivastava(2001)Plant Growth and Development:Hormones and Environment(植物生长和发育:激素和环境)(Academic Press,New York)对植物中乙烯生物合成和受乙烯调节的植物发育过程的总体描述。

因此,本领域对于延迟植物发育过程的安全的方法和装置仍有显著需要。这种方法和装置可提供更好地控制果实成熟、蔬菜成熟、花衰老、叶脱落和种子萌发,并延长各种农产品(如果实、蔬菜、和切花)的贮存期限,从而允许更长距离地运输这些产品而不需要冷藏,增加产品对消费者的合意性,并减少因不合时宜地成熟和衰老的产品损失而带来的金钱消耗。
发明概述
提供延迟包括但不限于果实成熟、蔬菜成熟、花衰老和叶脱落的植物发育过程的方法。本发明方法通常包括将植物或植物部分暴露于足以延迟目标植物发育过程的量的一种或多种细菌。在本发明某些方面,细菌选自红球菌属种(Rhodococcus spp.)、绿针假单胞菌(Pseudomonas chloroaphis)、酮戊二酸短杆菌(Brevibacteriumketoglutamicum)、及其混合物组成的组。用于实践本发明方法的细菌可进一步被包括例如天冬酰胺、谷氨酰胺、钴、尿素、及其混合物的诱导剂处理以诱导细菌延迟目标植物发育过程的能力。

本发明提供延迟目标植物发育过程的方法,包括将植物或植物部分暴露于一种或多种细菌。在具体实施方案,所述方法引申为延迟植物发育过程,包括将植物或植物部分暴露于选自红球菌属种、绿针假单胞菌、酮戊二酸短杆菌、及其混合物组成的组的一种或多种细菌,其中所述一种或多种细菌以足以延迟植物发育过程的量暴露于植物或植物部分。进一步提供延迟目标植物发育过程和实践本文所述方法的装置。可使用本发明方法和装置来例如,延迟果实/蔬菜成熟或花衰老并增加果实、蔬菜或花的贮存期限,从而便于这种植物产品的运输、分配和销售。

在本发明的一些方面,通过暴露于合适的诱导剂或以其处理,一种或多种细菌被"诱导"以展示期望的特征(如,延迟植物发育过程诸如果实成熟的能力)。诱导剂包括但不限于天冬酰胺、谷氨酰胺、钴、尿素、或其任何混合物。在具体实施方案,细菌暴露于诱导剂天冬酰胺或以其处理,更尤其是暴露于包含天冬酰胺、钴和尿素的诱导剂混合物或以其处理。诱导剂可在培养期望的细胞期间的任何时间加入。例如,关于细菌,培养基可在开始培养细菌之前补充诱导剂。或者,细菌可在培养基上培养预先确定的时间量以生长细菌,并可在一种或多种预先确定的时间加入诱导剂以诱导细菌中期望的酶活性。此外,可在细菌完成生长后向生长培养基(或向包含预先生长的细菌的单独混合物)加入诱导剂以诱导细菌中期望的活性。

在本发明的其它方面,在天冬酰胺存在时产生天冬酰胺酶I活性的绿针假单胞菌(ATCC保藏号43051)、和也已经显示产生天冬酰胺酶活性的革兰氏阳性细菌酮戊二酸短杆菌(B.kletoglutamicum)(ATCC保藏号21533)用于公开的方法。表达腈水合酶、酰胺酶、和/或天冬酰胺酶的真菌细胞,诸如镰刀菌属(Fusarium)的真菌、植物细胞和动物细胞也可用于本文公开的方法和装置,作为全细胞或作为分离一种或多种上述酶的来源。
在其它实施方案,已被遗传改造以表达腈水合酶、酰胺酶、和/或天冬酰胺酶的宿主细胞可用于根据本发明方法和装置暴露于植物或植物部分以延迟植物发育过程。具体地,编码腈水合酶、酰胺酶、或天冬酰胺酶的多核苷酸(或各编码腈水合酶、酰胺酶、或天冬酰胺酶的多种多核苷酸)可通过标准分子生物学技术引入宿主细胞以产生表达本发明的一种或多种酶的转基因细胞。使用术语"多核苷酸"、"多核苷酸构建体"、"核苷酸"或"核苷酸构建体"不意为将本发明限制为包含DNA的多核苷酸或核苷酸。本领域技术人员将认识到,多核苷酸和核苷酸可包含核糖核苷酸以及核糖核苷酸和脱氧核糖核苷酸的组合。这种脱氧核糖核苷酸和核糖核苷酸包括天然产生的分子和合成类似物。本发明的多核苷酸还包括所有形式的序列,包括但不限于单链形式、双链形式和类似物。

在本发明某些方面,固定的催化剂或一个或多个催化剂组件元件放在"物理结构"中、放在其上、或附加于其。物理结构包括但不限于能够容纳一个或多个催化剂组件元件的膜、片、包被层、盒、袋、包、或有槽箱。在一些实施方案,物理结构包括适于运输或储存果实、蔬菜或花的容器。物理结构可进一步包含多于一个单独结构,所有单独结构藉以连接于中间的催化剂或催化剂组件元件。上述物理结构可任选地被外部元件冷藏或在物理结构本身内包括冷藏单元。
监测催化剂延迟目标植物发育过程的效力(如,以评估何时应替换催化剂或催化剂组件)、或测量或控制气流、含湿量/湿度、和二氧化碳水平的元件可任选地包括在本发明的装置中。延迟植物发育过程的任何装置可进一步包括允许气流流至或流经催化剂或催化剂组件元件的一个或多个元件。本领域技术人员将易于想到对本文所述装置的其它可能修饰以监测和控制催化剂、催化剂组件元件、或物理结构的大气条件(如,气流、湿度、和二氧化碳水平)。条件诸如温度、大气组成(如,相对湿度、O2和CO2水平、物理应激、光、化学应激、辐射、水应激、生长调节素和病原攻击对呼吸率起重要作用并显著影响果实、蔬菜、花和其它植物-相关产品的贮存期限。尽管储存的温度和大气条件根据果实、蔬菜或其它目标植物产品而变化,推荐的储存温度通常在约0℃至约20℃的范围,O2和CO2水平的大概范围分别为1-10%和0-20%。储存果实、蔬菜和相关植物产品通常推荐的相对湿度为约50%至约100%、尤其是85%至约95%、更具体地约90%至约95%。由于在植物产品的呼吸率和贮存期限之间有显著相关性,控制上述因素对于延迟这种产品劣化是重要的。因此,装置中可设有二氧化碳清除剂以减少二氧化碳含量。
在本发明的具体实施方案,提供延迟植物发育过程的包含多层的可透气的催化剂装置。例如,如图1所示,催化剂装置10可包括外层12和14,和位于外层12和14之间的中间催化剂层16。催化剂层16包含一种或多种细菌(如,红球菌属种、绿针假单胞菌、酮戊二酸短杆菌、及其混合物)或酶(腈水合酶、酰胺酶、天冬酰胺酶、及其混合物),其中所述一种或多种细菌或酶以足以延迟目标植物发育过程的量提供,以及第三层。在该实施方案,一个或多个外层12和14为催化剂装置10提供结构完整性。外层12和14通常允许空气流入催化剂层16,然而在一些实施方案,具有不可透气的外层可为有利的,如,如果装置形成盒的侧面且期望不允许盒的最外层将催化剂层暴露于环境。根据本发明,催化剂装置10可设在可重复使用的或一次性的包或袋中。在一个实施方案中,催化剂层16包含红球菌属种的细胞,尤其是紫红红球菌DAP96253菌株、红球菌属种DAP96622菌株、红串红球菌、或其混合物。在本发明的装置中用作催化剂的细菌细胞可被如上所详述的一种或多种诱导剂(如,天冬酰胺、谷氨酰胺、钴、尿素、或其混合物)诱导。

实施例
现在将具体参考多个实施例对本发明进行描述。以下实施例并非旨在限制本发明,而是提供示例性实施方案。
实施例1:暴露于诱导的红球菌属种之后延迟果实成熟
将以天冬酰胺、丙烯腈或乙腈诱导的红球菌属种的细胞固定在戊二醛-交联的DEAE-纤维素基质中。诱导细胞和制备上述基质的方法在以下详细描述。
将交联的DEAE-纤维素催化剂基质放在三个独立的纸包中(每包大约1-2克成团湿重(pack wet weight)的细胞),每包中包含未成熟的香蕉、桃或鳄梨。作为负对照,将同样的果实置于独立的纸包而没有催化剂基质。将纸包置于室温,每天观察产品的果实成熟和降解迹象。
暴露于催化剂基质的所有产品展示果实成熟的显著延迟。尤其是,催化剂基质存在下桃的坚硬和果皮完整度保持更长时间。类似地,对于香蕉,相对于负对照褐色斑点延迟出现且坚硬保持更长时间。
实施例2:一般发酵和诱导方案
发酵过程
以下一般方案和培养基用于发酵用于其它实验的红球菌属种菌株红球菌属种DAP96622和紫红红球菌DAP96523:
发酵罐配置有测量溶解氧(DO)和pH的探测器、以及测量葡萄糖浓度(线下)的取样设备。另外的口用于加入矫正物(如,酸、碱或防沫剂)、诱导物、营养和补充物。将预先清洁的罐在原处灭菌。使用合适的基础培养基(1或1.5X)R2A或R3A。这些培养基的具体成分列于以下。在一些实验中对培养基的成分进行了某些代替。例如,(Trader's Protein,Memphis,TN)有时用于代替蛋白胨和/或酪蛋白水解物。此外,在某些实验中,使用Hy-Cotton(Quest International,Hoffman Estates,IL)、棉籽水解物、超滤的棉籽水解物(Marcor Devolpment Corp.,Carlstadt,NJ)代替(Trader's Protein,Memphis,TN)。
设定营养补充的供应方案以逐渐地以更有营养的培养基即2X YEMEA替换R2A或R3A基础培养基,2X YEMEA的成分也在以下详述。其它任选营养补充物包括麦芽糖50%(w/v)和右旋糖50%(w/v)。有时使用含有右旋糖等价物(葡萄糖、麦芽糖、和更高级多糖)的商业产品代替麦芽糖和右旋糖。
从红球菌属种DAP96622和紫红红球菌DAP96523菌株在合适的固体培养基上的培养物制备接种物并以其适当温度(如,30℃)接种。在具体实施方案,细胞生长在YEMEA琼脂平板上4-14天,优选地7天。可选地,从此前发酵运转的冷冻细胞浓缩物制备接种物。通常以发酵罐中存在浓度的20X浓度制备细胞浓缩物。此外,有时从合适的两相培养基(即,液体培养基铺在相同或不同组成的固体培养基上的组合)制备接种物。当使用两相培养基时,培养基通常在液体和固体层包含YEMEA。
对于诱导腈水合酶,在t=0小时,加入无菌的CoCl2·6H2O和尿素以达到5-200ppm的CoCl2和750mg/l-10g/l的尿素的浓度,通常优选10-50ppm的CoCl2和7500mg/l-7.5g/l的尿素。在具体实施方案,在发酵期间再次加入尿素和/或钴。例如,在4-6小时或24-30小时加入等体积的尿素和150ppm的CoCl2。除了尿素,在不同时间开始,逐步或以恒定速率加入终浓度为300-500ppm的丙烯腈/乙腈或0.1M-0.2M的天冬酰胺。当细胞质量和酶浓度可接受时发酵运转终止,通常在24-96小时。
随后以任何可接受的方法收集细胞,包括但不限于分批离心或连续离心、倾析或过滤。收集的细胞重悬于20X浓缩体积的合适的缓冲液,诸如补充有发酵过程中使用的诱导物的50mM磷酸盐缓冲盐水(PBS)。随后冷冻细胞浓缩物,尤其是通过快速冷冻。冷冻的细胞储存在-20℃-80℃或液氮中备用。

基于具体液体诱导物的密度,计量体积地加入易挥发的诱导物液体(如,丙烯腈/乙腈),作为过滤灭菌的液体诱导物。在固体诱导物(如,天冬酰胺/谷氨酰胺)的情况,称重固体并直接加入培养基。将所得培养基高压灭菌。当使用过滤灭菌的液体诱导物时,加入液体诱导物之前将培养基单独高压灭菌并冷却至40℃。目标诱导物的典型浓度为:500ppm丙烯腈/乙腈;500ppm天冬酰胺/谷氨酰胺;和50ppm丁二腈。随后将细胞生长在具体培养基并进一步分析具体酶活性和生物量。
实施例3:分析亚硝酸盐水合酶、酰胺酶和天冬酰胺酶活性和天冬酰胺-诱导的红球菌属种的细胞的生物量
在红球菌属种的菌株红球菌属种DAP96622和紫红红球菌DAP96523的天冬酰胺-诱导的细胞评价腈水合酶、酰胺酶和天冬酰胺酶活性和生物量。分析培养基成分、施加方法、施加速率和向细胞提供的天冬酰胺浓度、和细胞来源的各种改变对于上述酶活性和生物量的影响。本实施例的A至G部分描述每组测试条件的细节并提供在各种具体条件下获得的酶活性和生物量的概述。
A.基本如上实施例2中所述的,使用从固体培养基收集的紫红红球菌DAP96253细胞接种的20-升发酵罐连续补充诱导物天冬酰胺(120μl/分钟的0.2M溶液)。使用Hy-Cotton代替上述R3A培养基中的蛋白胨#3。在发酵运转结束时,根据本领域已知的标准技术测量丙烯腈-特异性腈水合酶活性、酰胺酶活性和生物量。
腈水合酶活性、酰胺酶活性和生物量的结果提供在下表3,活性以单位/mg cdw(细胞干重)提供。一单位的腈水合酶活性涉及在每毫克细胞(干重)、7.0的pH、30℃的温度,每分钟转化1μmol的丙烯腈为其相应酰胺的能力。一单位的酰胺酶活性涉及在每毫克细胞(干重)、7.0的pH、30℃的温度,每分钟转化1μmol的丙烯酰胺为其相应酸的能力。生物量以成团细胞g/lcww(细胞湿重)报告。

B.基本如上实施例3A中所述的,培养基变化如下指出,以紫红红球菌DAP96523细胞评价酶活性和生物量。具体地,向修改的R3A培养基加入YEMEA、右旋糖或麦芽糖,该培养基进一步含有Hy-Cotton代替蛋白胨#3。在t=8小时开始,以120μl/分钟的持续速率加入0.2M天冬酰胺溶液。在发酵运转结束时,测量丙烯腈-特异性腈水合酶活性、酰胺酶活性和生物量。结果概述于表4。随着向培养基加入YEMEA、右旋糖或麦芽糖,观察生物量产量的增加。

C.来自固体培养基的红球菌属种DAP96622细胞用作20-升发酵运转的接种物来源(发酵过程细节参见实施例2)。在t=24小时开始,每6小时半连续地加入0.2M的天冬酰胺溶液,以2ml/分钟的速率持续50-70分钟。在修改的R3A培养基中使用Hy-Cotton代替蛋白胨#3。在发酵运转结束时,测量丙烯腈-特异性腈水合酶活性、酰胺酶活性和生物量。结果概述于表5。

D.来自固体培养基的红球菌属种DAP96622细胞用作20-升发酵运转的接种物来源。在t=12小时开始,每6小时半连续地加入0.2M的天冬酰胺溶液,以2.5ml/分钟的速率持续12-85分钟。在修改的R3A培养基中使用棉籽水解物代替蛋白胨#3。在发酵运转结束时,测量丙烯腈-特异性腈水合酶活性、酰胺酶活性和生物量,结果概述于表6。

E.此前冷冻的紫红红球菌DAP96253细胞用作20-升发酵运转的接种物来源。向修改的R3A培养基加入YEMEA、右旋糖或麦芽糖,该培养基进一步含有Hy-Cotton代替蛋白胨#3。在t=8小时开始,以120μl/分钟的持续速率加入0.15M的天冬酰胺溶液。在发酵运转结束时,测量丙烯腈-特异性腈水合酶活性、酰胺酶活性和生物量。结果概述于表7。

F.生长在两相培养基上的紫红红球菌DAP96253细胞用作20-升发酵运转的接种物来源。使用通过加入糖类(即,YEMEA、右旋糖或麦芽糖)并进一步含有棉籽水解物代替蛋白胨#3而被补充的修改的R3A培养基。在t=10小时开始,以1000μl/分钟的持续速率加入0.15M的天冬酰胺溶液。在发酵运转结束时,测量丙烯腈-特异性腈水合酶活性、酰胺酶活性、天冬酰胺酶I活性和生物量。结果概述于表8。

G.生长在两相培养基上的紫红红球菌DAP96253细胞用作20-升发酵运转的接种物来源。使用含有麦芽糖(代替右旋糖)和Hy-Cotton代替蛋白胨#3的修改的R3A培养基。在t=8小时开始,以476μl/分钟的持续速率加入0.15M的天冬酰胺溶液。在发酵运转结束时,测量丙烯腈-特异性腈水合酶活性、酰胺酶活性和生物量,且结果概述于表9。

实施例4:在与戊二醛交联的DEAE-纤维素中固定红球菌属种的细胞
源自美国专利第4,229,536号和Lopez-Gallego等人(2005)J.Biotechnol.119:70-75所述方法的修饰方法用于在包含戊二醛交联的DEAE-纤维素的基质中固定红球菌属种的细胞。
制备细胞
将红球菌属细胞生长在适当培养基(如,YEMEA-麦芽糖+诱导物、两相培养、等)并在8,000rpm离心10分钟而收集。将所得的细胞小团重悬在100ml的50mM磷酸盐缓冲液(pH7.2)并在8,000rpm离心10分钟。将重悬细胞小团和在8,000rpm离心10分钟的过程重复两次。记录最终细胞样品的成团湿重(ww)。对细胞小样品进行腈水合酶活性以评价全细胞的酶活性。
固定细胞
获得与收集的红球菌属种的细胞等量的DEAE-纤维素,将细胞和DEAE-纤维素重悬于100ml的去离子水。搅拌下将足以实现终浓度为0.5%的体积的25%戊二醛溶液加入细胞/DEAE-纤维素混合物。搅拌混合物1小时,之后加入400ml去离子水并进一步混合。搅拌时加入50%(溶液重量)的聚乙烯亚胺(PEI;MW750,000)。继续搅拌直到完成絮凝。过滤絮凝的混合物并挤压通过适当大小的注射器。固定的细胞破裂成小片、过夜干燥、并在使用前切成大约2-3mm的颗粒。
实施例5:在藻酸钙中固定红球菌属种的细胞并硬化藻酸钙微珠
从Bucke(1987)Methods in Enzymology(酶学方法),第135(B)卷(Academic Press,Inc.,San Diego,California;Mosbacn编辑)的"Cell Immobilization in Calcium Alginate(在藻酸钙中固定细胞)"中所述的方法修改的方法用于在藻酸钙中固定红球菌属种的细胞。
制备细胞
如以上实施例4所述的制备红球菌属种的细胞。
固定细胞
通过在24ml的50mM Tris-HCl(pH7.2)中溶解1g藻酸钠产生25g的4%藻酸钠溶液。向藻酸盐溶液加入25mg的高碘酸钠并在25℃搅拌1小时或直到藻酸盐完全溶解。将如上述制备的细胞重悬于50ml终体积的50mM Tris-HCl(pH7.2),随后伴随搅拌加入到藻酸钠溶液。将所得的微珠挤压通过27号(gauge)针头到500ml的0.1M CaCl2溶液中。针头通常放置在溶液上方大约两英寸高以防止空气进入微珠并防止微珠粘附。在CaCl2溶液中固化微珠1小时,随后以水润洗微珠并在4℃、0.1MCaCl2溶液中储存备用。
硬化包含红球菌属种的细胞的藻酸钙微珠
如上述制备的藻酸钙微珠通过与PEI交联可被进一步强化。将微珠培养在2L的0.5%PEI的0.1M CaCl2溶液(20g的50%PEI在0.1M CaCl2溶液中)。如果需要,以HCl或NaOH调整最终溶液的pH到7.0,并培养微珠24小时。随后以水润洗微珠并在4℃、0.1M CaCl2溶液中储存备用。
受益于在前文描述中所呈现的教导,本发明所属领域内的技术人员会想到此处所阐述的本发明的许多改进和其他实施方案。因此,应当理解本发明并不局限于所公开的具体实施方案,改进和其他实施方案也意欲包括在所附权利要求的范围内。尽管此处采用了具体的术语,它们也仅是以普遍和描述性的意义进行使用,而并非是为了限制的目的。
Claims (16)
1.一种延迟植物发育过程的方法,其包括将植物或植物部分暴露于一种或多种酶,其中所述一种或多种酶选自由腈水合酶、酰胺酶、天冬酰胺酶及其混合物组成的组,且其中所述一种或多种酶以足以延迟所述植物发育过程的量暴露于所述植物或植物部分。
2.根据权利要求1所述的方法,其中所述一种或多种酶由一种或多种细菌提供。
3.根据权利要求2所述的方法,其中所述一种或多种酶从所述一种或多种细菌中分离、纯化或半纯化。
4.根据权利要求2或3所述的方法,其中所述一种或多种细菌选自由红球菌属种(Rhodococcus spp.)、绿针假单胞菌(Pseudomonas chloroaphis)、酮戊二酸短杆菌(Brevibacterium ketoglutamicum)及其混合物组成的组。
5.根据权利要求4所述的方法,其中所述一种或多种细菌包括红球菌属种(Rhodococcus spp.),可选的是,其中所述红球菌属种包括紫红红球菌DAP96253菌株、红球菌属种DAP96622菌株、红串红球菌、或其混合物。
6.根据权利要求2或3所述的方法,其中所述一种或多种细菌通过暴露于选自天冬酰胺、谷氨酰胺、钴、尿素及其混合物组成的组的诱导剂而被诱导。
7.根据权利要求1所述的方法,其中所述植物或植物部分间接或直接暴露于所述一种或多种酶。
8.根据权利要求1所述的方法,其中所述植物发育过程是果实成熟、蔬菜成熟或叶脱落。
9.根据权利要求1所述的方法,其中所述植物部分是果实、蔬菜或花;可选的是,其中所述果实是呼吸跃变型果实;可选的是,所述呼吸跃变型果实选自香蕉、桃、梅、油桃、苹果、番茄、梨和鳄梨组成的组。
10.根据权利要求1所述的方法,其中所述植物部分是花且所述植物发育过程是花衰老、萎蔫、脱落或花瓣闭合,可选的是,其中所述花是康乃馨、玫瑰、兰花、马齿苋、锦葵或秋海棠。
11.根据权利要求1所述的方法,其中延迟所述植物发育过程导致增加的贮存期限或有助于所述植物或植物部分的长距离运输。
12.根据权利要求1所述的方法,其中所述一种或多种酶被固定并放入物理结构中、放在物理结构上、或附加于物理结构。
13.一种延迟植物发育过程的装置,其包含多层,其中至少一层包含催化剂,所述催化剂包括选自由腈水合酶、酰胺酶、天冬酰胺酶及其混合物组成的组的一种或多种酶,所述催化剂放入物理结构中、放在物理结构上、或附加于物理结构,其中所述一种或多种酶以足以延迟所述植物发育过程的量提供。
14.根据权利要求13所述的装置,其中所述一种或多种酶固定在包含交联的DEAE-纤维素的基质、包含藻酸盐的基质、包含角叉菜的基质、包含聚丙烯酰胺的基质、或藻酸钙微珠中。
15.根据权利要求13所述的装置,其中所述装置是延迟植物发育过程的可透气的催化剂装置,其包括:
第一层;和
第二层,其包含催化剂,所述催化剂包括选自由腈水合酶、酰胺酶、天冬酰胺酶及其混合物组成的组的一种或多种酶,其中所述一种或多种酶以足以延迟植物发育过程的量提供;
其中所述第一层为所述装置提供结构完整性。
16.根据权利要求13~15中任一项所述的装置,其中所述物理结构为膜、片、包被层、盒、袋、包、或有槽箱。
