Abstract

技术领域
本发明涉及从流体样品中分离固体组分。具体而言,本发明涉及用于从诸如骨髓样品等生物样品中分离治疗性细胞组分的装置和方法。
背景技术
祖细胞在分化时具有多向潜能,并因此潜在地使其参与到不同的治疗性疗法中。已确定将这些细胞的富集组分输送至患病的或者受损的组织部位,通过加速组织愈合而具有最佳治疗效果。祖细胞通常是在一些哺乳动物组织和体液内包含于非治疗性细胞的异质混合物中的非常罕见的细胞群(Caplan 2005)。
在文献中已报道各种俘获细胞的技术和设备,包括沉淀(Takazawa &Tokashiki,1989)、离心(Apelman等,1992;Jaeger,1992)、旋转过滤(Himmerlflab等,1969)和横向流微滤(Maiorella等1991)。然而,这些技术都存在若干缺点。沉淀过程受到细胞的低沉淀速度和缓慢分离过程的影响。离心装置依赖于离心力驱使细胞偏向流体外侧,尽管能实现快速回收,但这些设备通常资产成本很高。旋转过滤器和横向流过滤器采用膜以过滤细胞悬浮液,但其易于积垢而导致性能损失。使用这些过滤器的其他方法包括可控剪切过滤(Vogel和Kroner,1999)、切向流过滤(Radlett,1972)和动态过滤。分离的速率和效率取决于积垢程度。动态过滤依赖于膜和外壳之间的相对运动从而产生独立于流经过滤器的液流的剪切流(Castilho & Medrohno,2002)。涡流过滤器和转盘过滤器是两类常见的动态过滤设备(Stromberg等,1989)。Karumanchi等(2002)提供了用于细胞和生物大分子的场协助分离技术的综述,包括电学、磁学和声学现象实现所需要的分离。介电电泳(Dielectrophoresis)是由于非均匀电场产生的极化效应而导致的侧向运动(Docoslis等,1997,1999)。这仅仅能应用于非常低导电性介质否则将造成流体的过量发热。磁分离分为两类。第一种,待分离的细胞内在具有磁性质(例如红血细胞、趋磁细菌),第二类,所需要的混合物的非磁性组分必须通过磁响应实体而被赋予磁性。在两种情况下,流体必须受作用于磁场而实现分离。磁性分离已广泛用于各种细胞分离,包括骨髓处理以富集祖细胞(Roath等,1990)。声学分离器已广泛使用以分离微生物体和细胞,但几乎普遍地依赖于使用超声波。由于超声波在与声波传播方向相对的方向上反射时形成驻波从而实现超声细胞保持。细胞陷入驻波的压力节点面。Kilbum等,(1989)和WO95/001214首先报道了使用这些波作为从悬浮液中分离细胞的手段的可能性,并且从此以后出现了许多采用该技术的报道(Coakley等,1994;Dobelhoof-Dier,1994;Gaida等,1996;Gorenflo等,2003,2004)。超声分离器需要仔细地调校并通常具有受限制的上样容量,因此仅能回收小量的细胞。此外,该技术用于从流体相中除去均质细胞群并因此不能从异质混合物中俘获特定的细胞群。
本发明涉及用于从流体样品中分离固体组分的装置和方法。具体而言,本发明涉及从流体样品中的异质混合物中分离细胞组分。该方法是单步骤方法,且细胞分离基于仅使用物理手段的样品机械过滤。
发明内容
根据本发明的一个方面提供了用于从流体样品中分离固体组分的装置,所述装置包括过滤单元,所述过滤单元包括:
声波发生元件;
至少一个过滤器,所述过滤器将所述单元分隔为用于接收流体样品的过滤前腔室和用于接收能传导声波的流体的过滤后腔室;和
经设置与过滤后腔室相关连的基板,所述基板能在向其施加声波时共振;
其中,所述声波发生元件经设置与基板相关连从而使得所述声波发生元件引起所述基板的共振,所述基板随之传导声驻波穿过过滤后腔室里的流体和过滤前腔室里的流体样品。
在分离过程中,声驻波在所述过滤单元里的流体中的传导导致流体的扰动,或者为连续方式或者为间歇方式。该扰动的特别优点在于使过滤器最小程度地积垢和阻塞。
在其中所述装置用于从流体样品中分离细胞组分的本发明的实施方式中,在所述单元中的流体的扰动还具有另一个优点,在于使过滤前腔室中细胞与过滤器的接触或驻留时间最短。这是令人所希望的,因为如果细胞与过滤器相接触或者足够接近,则存在细胞在周围流体的压力下受力穿过孔的倾向。可通过细胞的变形实现如此穿过孔,但在穿越过程中细胞可能暴露于不希望的压力,诸如剪切力,其对于细胞有不利影响。具体而言,已表明白细胞功能显著受到剪切力的影响(Carter等,2003)。因此所希望的是防止令人感兴趣的细胞穿过过滤器。在这点上,过滤器经设计使得在将令人感兴趣的细胞保留在过滤前腔室中的同时允许流体和其他细胞组分通过。
声波发生元件定义为能响应动力信号而产生声波的结构。例如,在单一扬声器的情况下,动力信号将以确定的波幅和频率使薄膜偏转从而产生声驻波。其他声学元件包括压电传感器,其响应于施加的AC电压产生振动能量而所述物理振动作为声学力传导至流体。压电元件的例子是在两侧具有金属膜电极的陶瓷片,这些压电薄膜通常是氧化锌。
声波发生元件与能共振的基板的结合相比单独使用声学元件具有许多优势。声学元件并不与流体直接接触,该元件经设计使得在贴于基板材料上时能提供更强的共振,所述基板材料具有比元件本身的锥体更大的面积。这意味着在使用小声学元件时仍能向流体输送更多的能量。总体上这显著提高了所述设备的能量效率和足迹(footprint),这在处理大量流体时尤其有利。适合的声学元件可由NXT Technology Limited(HongKong)获得,型号RM-ETN0033K19C-2K01。
在本发明的实施方式中,声波发生元件产生频率为约300~700Hz的声驻波。在流体体积和所需的最佳频率之间存在相互关系。因此,在本发明的具体实施方式中有利的是用户能输入流体样品的体积,则声驻波的频率将被计算得到。例如,对于体积为约5ml~15ml的样品,声驻波的最佳频率为约300Hz~700Hz。
在本发明的具体实施方式中,声波发生元件是扬声器,其具有0.4W的功率、4Ohm的阻抗、峰峰间振幅为约4.2V~7.36V、频率为约300~700Hz。
在本发明的实施方式中,基板基本上平行于过滤器放置。
在本发明的实施方式中,基板位于过滤单元的底部。
基板可由任意适合的材料制成,所述材料能响应于声学发生元件的刺激而共振。例如,金属、陶瓷或聚合物。有利的是,由于基板可与装置中所用的流体相接触,因此基本由医疗级材料制得。这减少了任何毒性物质污染的风险。
在本发明的实施方式中,声波发生元件与基板可逆相连。该相连可由本领域技术人员已知的各种手段实现,例如使用粘合剂。这在过滤单元如果设计为一次性使用时特别有利,因为声学元件可被拆下并在其他单元中再次使用。
流体样品穿过过滤器的运动可以是重力导致的被动运动。另外一种方式,可以通过施加正压力或负压力使流体样品主动流过过滤器。流体样品穿过过滤器的流速可以是恒定的或者是可变的。
在本发明的实施方式中,在过滤后腔室中提供的能够传导声驻波的流体也用作清洗流。在该实施方式中,提供正压力泵,其推动清洗流由过滤后腔室穿过过滤器进入过滤前腔室中。同时,负压力泵抽取流体由过滤前腔室进入过滤后腔室。因此,流体样品和清洗流以相反方向连续通过过滤器,从而使得流体样品的净移动为进入过滤后腔室,而被分离的固体组分保留在过滤前腔室中。
流体样品和清洗流以相反方向穿过过滤器的连续运动防止了过滤器的积垢和阻塞,进一步提高了分离过程的效率。该连续运动可以是循环的,循环中的一相是使用清洗流的过滤器的快速回流,循环中的第二相为迫使流体样品向下穿过过滤器。该循环过程持续进行以充分地减少样品体积而同时也能在过滤器上保持足够体积的流体从而使得颗粒组分保持在溶液之中。通常,样品体积减少约10倍。
在本发明的其他实施方式中,过滤器本身在分离过程中受扰动。例如可以通过将过滤器与可移动的固定器相关连从而实现该扰动。扰动可以在纵向上或者横向上或者同时在两个方向上。在本发明的另外一些实施方式中,过滤器可旋转。
额外扰动所述清洗流体和/或流体样品的其他手段包括使用旋转元件(例如流体中的叶轮或者插入物)、旋转腔室壁或者旋转腔室壁,其中腔室壁配备有隔板或者使用主动翼片。
“流体样品”是从其中将分离固体组分的任何流体。样品可以来自任何来源,诸如生物体、来自相同物种或不同物种的生物体群、来自环境诸如来自身体或者水或者来自土壤,或者来自食物来源或者工业来源。样品可以是经加工或未经加工的样品。样品可以是气体或液体。样品可以是萃取物,例如土壤或食物样品的液体萃取物。
所述装置特别适用于粘滞的并且更容易倾向于通过在过滤器表面上形成膜或覆层而使过滤器积垢的液体。清洗流流经过滤器的回洗,以及任选的流体样品和/或过滤器的扰动显著地减少了过滤器的积垢,并且同时也将溶液中的颗粒组分保留在过滤前腔室中。
样品可由受试对象获得。受试对象可以是任何生物体诸如动物或者人。动物包括任何动物诸如野生动物或者家畜。家畜也可以包括例如伴侣动物,诸如狗或者猫。
样品可以是生物样品诸如血液样品、渗出液、尿液样品、精液、骨髓抽取物、脊髓液、来自组织的细胞悬浮液、粘液、痰液或唾液。生物样品可由任何动物获得并且不限于由人类获得的生物样品。
本文使用的“血液样品”指经加工或未经加工的血液样品,包括脐带血样品、骨髓抽取物、内部血液或者外周血液,可以是任意体积,并且能来自于任何受试对象诸如动物或人。优选的受试对象是人。
将从生物流体样品中分离的固体组分可以包含至少一个细胞组分。所述细胞组分优选为包括治疗性细胞,其可以是具有治疗或治病效果的任何细胞。
细胞组分可包含或者构成于至少一种白血细胞。“白血细胞”是白血球或者非网织红细胞或血小板的造血细胞,可在动物或人类的血液中发现。白血球可包括自然杀伤细胞(“NK细胞”)、淋巴细胞诸如B淋巴细胞(“B细胞”)或T淋巴细胞(“T细胞”)。白血球也可包括噬菌细胞诸如单核细胞、巨噬细胞和粒细胞,包括嗜碱细胞、嗜酸细胞(eosinophil)和嗜中性细胞。白血球也可以包括肥大细胞。
细胞组分可包含或者构成于至少一种红血细胞。“红血细胞”是红血球。
细胞组分可包含或者构成于至少一种赘生性细胞。“赘生性细胞”指具有不受控制的细胞增殖并在诱导新生的刺激消失以后持续生长的异常细胞。赘生性细胞倾向于表现出部分地或者全部地缺失结构组织以及与正常组织的功能性协调,并且可以是良性的或者恶性的。
细胞组分可包含或构成于至少一种恶性细胞。“恶性细胞“指具有局部入侵性和破坏性生长和转移的性质的细胞。恶性细胞的例子包括但不限于在包括血液、骨髓、腹水、尿液、支气管冲洗液在内的各种体液中的白血病细胞、淋巴瘤细胞、实体瘤的癌细胞、转移实体瘤细胞(例如乳腺癌细胞、前列腺癌细胞、肺癌细胞、结肠癌细胞)。
细胞组分可以包含或者构成于至少一种癌细胞。“癌细胞”指表现出不受调控的生长的细胞,并且其在绝大多数情况下已经失去其至少一种分化性质诸如但不限于特征性形态、非迁移性行为、细胞-细胞相互作用和细胞信号传递行为、蛋白表达和分泌模式等等
细胞组分可以包含或者构成于至少一种干细胞。“干细胞”是未分化的细胞,其能够通过一个或多个细胞分裂周期而产生至少一种分化细胞类型。
细胞组分可包含或者构成于至少一种祖细胞。“祖细胞”是定型的但未分化的细胞,其能够通过一个或多个细胞分裂周期而产生至少一种分化细胞类型。通常干细胞响应于一条具体刺激或者一组刺激而通过一个或多个细胞分裂产生祖细胞,祖细胞响应于一条具体刺激或一组刺激而产生一种或多种已分化的细胞。
骨髓(或者髓骨质)是在中空的骨内部发现的软组织。有两种类型的骨髓“红骨髓(也称之为骨髓组织)和黄骨髓。红血细胞、血小板和绝大部分白血细胞产生于红骨髓,一些白血细胞在黄骨髓中出现。
骨髓含有两种干细胞:造血干细胞和间质干细胞。干细胞是所有保留了通过细胞分裂更新自身的能力的多细胞生物体共有的原始细胞,并且能分化为各种特化细胞类型。造血干细胞产生在血液循环中找到的三类血细胞:白血细胞(白血球)、红血细胞(红血球)和血小板(凝血细胞)。间质干细胞被发现排列在骨髓中的中心窦周围并且具有分化为造骨细胞、软骨细胞、肌细胞和许多其它类型的细胞的能力。
胚胎干细胞是真正的干细胞,在于它们是全能的或者多能的并且显示出不受限制的自身更新能力,而位于骨髓之内的成人干细胞更恰当地称为祖细胞,其类似于干细胞而具有自身更新和分化的能力,但远受到更多限制。祖细胞通常是单能的或者专能的,而不是多能的。
典型地获得自骨髓的间质干细胞或MSC是能分化为各种细胞类型的专能干细胞。MSC表现出的能分化为的细胞类型包括造骨细胞、软骨细胞、肌细胞、脂肪细胞、神经细胞。
骨髓密度在患者之间并不相同,并且骨髓没有均一的粘度。更年轻的患者通常具有更致密的更稠的骨髓,这是空穴中更多小梁组织的结果。此粘性骨髓倾向于使过滤设备中的过滤器积垢。
在本发明的实施方式中,流体样品是骨髓抽取物。
在本发明的其他实施方式中,流体样品是骨髓抽取物,而固体组分是祖细胞。
本文中使用的“组织”包括存在于所有动物中的基本类型的组织:上皮、结缔组织、肌肉组织和神经组织。
结缔组织的例子包括皮肤、肌肉、软骨、骨骼、肌腱、韧带、关节囊和脂肪组织。
本文中使用的“脂肪组织”意指身体中的平均脂肪和其他微脉管组织来源。脂肪组织是含有多种细胞类型的复杂组织,包括脂肪细胞、周细胞、纤维原细胞、巨噬细胞、干细胞和微脉管细胞。由此,脂肪组织是身体中祖细胞的最便捷来源之一。
本文中使用的“微脉管细胞”意指含有微脉管结构的平均细胞,诸如内皮细胞、平滑肌细胞和周细胞。
脂肪组织可以由位于体内的“脂肪”储藏部位获取。合适的储藏部位包括附睾、肩胛间脂肪垫或髌下脂肪垫(霍法(Hoffas)脂肪垫)。另外一种方式,潜在地更方便的是,所述脂肪组织可以是吸脂过程中获得的吸脂物。
虽然吸脂物能直接导入本发明的设备,脂肪组织碎片需要预加工。组织碎片被粉碎和/或经酶消化而释放出组织的细胞组分。该细胞组分然后可以悬浮在合适的载体中并导入所述设备中。
能设想的是如上所得的脂肪抽取物和/或细胞悬浮液在导入设备前进行再加工。例如,可以使用重力沉淀和/或离心以分离更大的脂肪小球和来自基质组分(包括干细胞、内皮细胞和周细胞)的脂肪细胞。
在其中过滤生物样品的本发明的实施方式中,过滤单元可以与吸气器相连从而使得流体直接由受试对象转移到离心单元中。过滤单元可以在无菌环境中使用,而该设置降低了在从患者获取样品并导入过滤单元之间样品污染的风险。
在另一个本发明的实施方式中,固体组分由能感染受试对象的致病剂诸如细菌、真菌、原生动物、病毒、寄生虫或朊病毒等构成。
样品可以是活体外细胞悬浮液。
过滤器,也能被称为薄片,可以由适用于在本发明所描述的方法中从流体样品中分离固体组分的材料制造。过滤器可以由天然材料或合成材料或者两种材料的组合制成。合适的材料包括但不限于金属、金属合金、陶瓷或聚合材料。实例包括聚碳酸酯(PLC)、聚对苯二甲酸乙二醇酯(PET)、聚酰亚胺(polyidide)(PI)、镍和不锈钢。材料优选为医疗级材料。合适的履带式蚀刻过滤器由it4ip(比利时)提供。合适的镍箔过滤器由Tecan Limited(U.K)提供。
在本发明的实施方式中,过滤器基本是平面的。也就是,过滤器具有这样的二维模式,即其中过滤器的直径大于过滤器的高度。这样的模式提高了过滤器暴露于流体样品的潜在过滤面积,由此提高了过滤速率。该模式也使任何固体物质在过滤器中阻塞的可能性降至最低。
过滤器的合适厚度的例子为11、23和50微米。过滤器越薄,通过其的流体流速越快。
可以设想的是过滤器配备有具有相同几何形状的相同直径或不同直径的孔。另外一种方式,过滤器可配备有具有不同几何形状的相同直径的孔。另外一种方式,过滤器可配备有具有不同直径和不同几何形状的孔。
适宜的孔形状包括但不限于横截面为圆形、椭圆形、方形、矩形或三角形。
孔可以是锥形的。孔的锥形有助于真空下细胞的变形。根据在锥形体的每一端的孔径和锥形的取向,可以实现基于尺寸的细胞优先选择。在本发明的有利实施方式中,锥形孔的最狭窄点位于过滤器的上表面。这种设置使得较小的细胞流过孔而较大的细胞保留在过滤器之上。相反,似乎使得在清洗流向上回流通过过滤器时过滤后腔室中的细胞更难地返回通过孔。已发现如果孔的最狭窄点位于过滤器的下表面,细胞倾向于进入孔并变形从而挤过最狭窄点。
在本发明的具体实施方式中,孔具有约1微米~12微米的直径。
在本发明的其他实施方式中,孔可以为圆柱形。
在其中由骨髓抽取物中分离祖细胞的本发明的其他实施方式中,适合的过滤器由PET制成,具有23微米的厚度,3微米的孔径和400,000孔/cm2的孔密度。由真空泵产生的能有效地将流体“拉”过过滤器的最佳负压为约-0.1~-0.5psi,更具体为-0.2~-0.3psi。另外一种方式,可以产生正压以有效地将流体“推”过过滤器。由该泵产生的最佳正压为约+0.1~+0.5psi,更具体为+0.2~+0.3psi。
过滤单元的设计可经过修改使得用于流体样品的过滤前腔室的高宽比(aspect ratio)减小,因而提供更大的每单元体积过滤表面积。
在本发明的其他实施方式中,过滤单元的过滤前腔室分隔为多个腔室,可用于接受孔板格式的批次流体样品。
在过滤后,含有被分离的(也称为纯化的、富集的、浓缩的)固体组分的保留流体样品可通过使用例如移液器等进行吸取而从过滤器单元的上部腔室中取出,并储存或者使用。在本发明的其他实施方式中,尤其是在其中保留流体样品含有治疗性细胞组分的实施方式中,保留流体样品可以与例如水凝胶或骨结合剂等相混合。在这些实施方式中,水凝胶或骨结合剂用作细胞储库。
根据本发明的一个方面提供了从流体样品中分离固体组分的方法,所述方法包括步骤:
(i)将流体样品导入本发明的装置;
(ii)过滤所述流体样品;和
(iii)从过滤前腔室中取出已分离的组分。
根据本发明的另一个方面,提供了使用本发明的装置从流体样品中隔离或分离治疗性细胞的方法。
在本发明的实施方式中,流体样品可以是诸如血液样品、渗出液、尿液样品、精液、骨髓抽取物、脊髓液、来自组织的细胞悬浮液、粘液、痰液或唾液等生物样品。
在本发明的实施方式中,治疗性细胞是祖细胞。
根据本发明的再一个方面提供了使用本发明的装置从骨髓抽取物中隔离或分离治疗性细胞的方法。
在本发明的实施方式中,治疗性细胞是祖细胞。
系统可手动运行。然而,为了提高分离过程的整体效率和准确性,尤其是当装置在手术室中由医务工作者使用时,装置有利地为自动运行。可编程逻辑控制器(PLC)可经编程以时控顺序周期旋转切换真空泵和回洗泵的开关。真空泵在负压下吸取流体样品向下流过过滤器。回流泵迫使清洗流向上通过过滤器。
过滤腔室和/或过滤器可以是一次性使用的。控制单元可以是一次使用的或者是独立专用单元。
在本发明的其他方面,经分离的治疗性细胞可以作为悬浮剂直接施用至对其有需要的位点。另外一种方式,细胞可以与适当的载体材料相结合或者相联合,例如凝胶、糊剂、粘合剂、胶水、支撑架、膜、植入物或敷料。
可设想的是所分离的治疗性细胞可用于针对人类和/或非人类动物的多种医疗用途以修复、再生和/或增强组织功能。
医疗应用的实例包括整形外科、神经病学、心血管、皮肤病学、美容手术和牙科。
可设想的是在本发明的某些实施方式中,所分离的治疗性细胞包括间质干细胞。这些细胞能分化为造骨细胞、软骨细胞、肌细胞和脂肪细胞。含有间质干细胞的治疗性组分可用于疾病或损伤造成的整形缺陷中,诸如软骨修复、骨骼修复(包括骨折修复)、脊柱融合术、椎间盘退变治疗(包括环修复、髓核数量增强(nucleous populous augmentation)、椎间盘增强(disc augmentation))。
功能性内皮祖细胞(EPC)是血管生成和血管新生的核心。已表明EPC发展自成体动物的骨髓单核细胞。因此可设想的是在本发明的某些实施方式中,所分离的治疗性细胞包括EPC,并且该治疗性组分可用于其中受损的或者缺血的组织需要修复、再生或血管发生的缺陷,诸如外周血管疾病。
因此提供了方法用于i)在受试对象的组织中形成新血管,ii)提高受试对象组织中的血流,iii)治疗受试对象的患病组织,iv)提高患病组织中的血管新生或v)防止受试对象的心力衰竭,所有的所述方法包括步骤:
a)使用本发明的装置分离骨髓单核细胞,
b)将有效量的骨髓单核细胞局部植入组织中以在组织中形成新血管。
在本发明的实施方式中,骨髓单核细胞来自于自体。
骨髓单核细胞植入其中的组织包括患病的或者受损的组织以及任何需要修复或再生的组织,包括但不限于灌流(underpurfused)组织,诸如在慢性贫血中发现的组织,以及心肌组织、骨骼肌组织、脑组织(例如受到中风或AV畸形影响)、冠状血管、肾、肝、胃肠道器官、受萎缩(包括神经类肌肉萎缩)影响的肌肉组织。
本发明的实施方式中,新血管是毛细血管和/或侧支血管。

多孔过滤器2将过滤单元1分隔为两个腔室:上层(过滤前)腔室3和下层(过滤后)腔室5,需要进行细胞分离的液体样品4导入上层(过滤前)腔室3,能传导声驻波的流体6导入下层(过滤后)腔室5。声学元件8与基板7相连,所述基板7在下层腔室内并位于底部,并且响应于发声元件而共振并产生驻波穿过两个流体相和过滤器从而扰动样品。同时,真空吸取9的循环过程导致样品向下移动通过过滤器。由装有合适泵和阀门的远程单元控制真空压力、流体流速和震动频率。所需要的较大细胞的浓缩组分保留在过滤器的顶部,同时较小的细胞穿过过滤器进入废物容器(未示出)。
在本发明的具体实施方式中,声学元件是扬声器,其具有0.4W的功率、4Ohm的阻抗、峰峰间振幅为约4.2V~7.36V、频率为约300~700Hz。

该图描述了包括本发明的过滤单元(如图1所示)和控制单元的装置。在控制单元19中的PLC控制器(Mitsubuishi AL2-24MR-04)可编程切换真空泵15和回流泵14(均为Koge KPV14A-6A)的开关。它也用于控制电磁阀(Cole Parmer 98302-02)17和18的触动。这可编程为以时控顺序运行阀门和泵或者手动控制泵。内建在控制单元中的放大器和信号产生器芯片使得可以通过PLC设定声学元件18的频率和振幅。收集来自设备的排出的流体的容器12和输送回洗流的容器13通过适宜的流管与过滤器单元和控制单元相连。

以下描述涉及本发明的实施方式,其中流体持续流动通过过滤器。
在正常运行中,过滤器单元中没有流体。回洗泵打开将流体输送至过滤单元13的下层腔室直至流体通过过滤器(12)进入上层(过滤前)腔室。声学元件的峰峰间振幅为4.2V~7.36V,频率为300~700Hz。该元件设定为驱动驻波穿过流体并且观察到流体处于持续扰动之中。真空泵打开抽取液面下降刚好至通过过滤器。通过针型阀22调节通过过滤器的流速。样品流输送到上层(过滤前)腔室中并观察到持续扰动。由PLC启动缓慢减少样品体积的快速回洗和真空抽取循环过程。样品体积通常减小10倍,并由PLC停止所述过程。从上层(过滤前)腔室中取出样品。
图7描述了用于本发明的装置之中的过滤器固定器。过滤器固定器的设计使得过滤器可在纵向或横向或者同时两个方向上的移动。固定器55基本为圆形。马蹄形或U形孔56将固定器分为两个部分,外环部分57和中心环形岛58。部分57与部分58通过桥元件59a和59b相连。部分58配备有多个孔。所述孔以统一的方式分布。所述孔基本上为具有约3微米直径的圆形。栓元件60配备在部分57的外表面上并与桥元件59b成一直线。该栓元件60由在装置之外的马达和凸轮(未示出)机械驱动。
合并的过滤器固定器和过滤器制造为单一塑料单元。
实施例
从流体样品中分离固体部分的实施例
1.人骨髓抽取物样品
由Cambrex购买得到人骨髓抽取物,使用前经过70μm细胞过滤器以除去任何脂肪细胞和大块细胞聚集体。用coulter计数器计数白血细胞(WBC)和红血细胞(RBC)。将2ml细胞悬浮液加入到设备中,所述设备已经装填有用作具有约3μm孔径的过滤器的光电成形的网筛。使用设备的控制单元启动分离过程并持续进行直至在设备的上层腔室中于过滤器上游保留约200μl的浓缩流体部分。后处理,从设备中过滤器的顶部取出100μl的浓缩物然后通过coulter计数器,所述coulter计数器在每次实验前经过日常标准校准。得到所得样品中的WBC和RBC计数。
2.猪骨髓样品
猪细胞群落的分离和培养扩展
由屠宰场获得新鲜猪后腿。由每条腿的股骨头中抽取蕈体柄(Trabeculum)骨髓并收集到盛有PBS的锅中。用剪刀将骨髓细微地剪碎,并将所得浆液放置于37℃/5%CO2的定轨振荡器中晃动,此后将上清液通过70μm的细胞过滤器进入分离管。然后将试管在1500rpm旋转5分钟并弃去上清液,所得小球再悬浮于10ml的α-MEM+15%FCS培养基中。
如上分离并处理骨髓,然后将100μl的骨髓悬浮液通过coulter计数器,所述coulter计数器在每次实验前经过日常标准校准。得到的WBC和RBC计数用于通过将提取物用猪血浆稀释从而使骨髓提取物的WBC为每毫升1×107个细胞。将2ml该流体加入到设备中进行分离
猪血浆的制备
从屠宰场收集新鲜猪血放入盛有4%(w/v)柠檬酸三钠溶液(sigma)的储存瓶中,比例为1∶9(柠檬酸钠∶血)。将血液等分到50ml离心管中并在2500rpm旋转30分钟,然后小心地将上清液转移到另一离心管中。上清液再次于2500rpm旋转30分钟并将上清液转移到新的50ml离心管中,并在-20℃保存直至准备使用。
猪骨髓抽取物的葡聚糖分离
如上所述从新鲜猪后腿中抽取猪骨髓,最终细胞小球再悬浮于25ml的α-MEM+15%FCS培养基中。
新鲜猪血用磷酸缓冲盐水稀释,以使RBC数目减少至大约每ml中3.5×109个细胞。一旦完成,通过加入早先获得的骨髓将WBC数目增加至大约每ml中2.5×107个细胞。血液随后稀释2∶1(补齐盐溶液∶血液),然后将35ml的该稀释血液放置于15ml葡聚糖(Amersham biosciences)顶部。试管随后于1800rpm旋转30分钟,制动被关闭(Megafuge 1.0R SOP/CB/021.)。然后抽取WBC层并制成小球然后再次悬浮于低葡萄糖DMEM培养基中,对RBC和WBC产品进行计数。
3.猪血样品
从屠宰场收集新鲜猪血放入盛有4%(w/v)柠檬酸三钠溶液(sigma)的储存瓶中,比例为1∶9(柠檬酸钠∶血)。通常,将20ml的等分试样通过70μm细胞过滤器进入收集试管。该过程重复三次,每次使用新过滤器。将2ml最终过滤样品放入设备进行分离。
结果
图8:具有约3微米孔径的过滤器被证实为最佳的(p<0.05)从全猪血中回收MNC:采用的统计测试为单向ANOVA和Tukey事后分析以及95%的置信区间。
图9描述了当使用孔径约3微米的过滤器时,可以在6分钟之内从全猪血中将MNC富集超过六倍。
图10描述了当在装置中采用的运行条件包括:(i)真空抽取流体样品向下穿过过滤器,(ii)回洗驱使清洗流向上通过过滤器和(iii)声学元件产生并传到驻波向上通过清洗流和样品流的时候,可得到约10倍的猪骨髓组织的体积减少。
图11:确定了流体通过过滤器的最佳流速。流体样品来自于猪骨髓。过滤前流体体积为2ml。过滤后流体样品体积为200μl。6滴/分钟时实现流体样品体积减少的时间为8分钟,12滴/分钟时为4分钟,25滴/分钟时为2分钟,全部采用3微米网筛过滤器。最佳流速确定为6滴/分钟,因为相比12滴/分钟或25滴/分钟流速,这对应于改善的-MNC富集。
图12描述了在过滤后流体样品中WBC相比RBC的富集。小于1的数值指示了RBC通过了过滤器。
图13描述了过滤前和过滤后全猪血样品中的RBC∶WBC比率。过滤前,RBC∶WBC的比率为约375∶1,过滤后,RBC∶WBC的比率为约50∶1。
图14描述了使用具有23μm厚度、400k孔密度和3.5μm孔径的PET过滤器,人骨髓中骨髓成核细胞的浓缩倍数和骨髓成核细胞的保留百分比%。
图15描述了具有400,000孔密度的PET过滤器的孔径分布。该图进一步描述了当考虑处理来自骨髓抽取物的细胞时的该过滤器的性能。

Claims (27)
1.用于从流体样品中分离固体组分的装置,所述装置包括过滤单元,所述过滤单元包括:
声波发生元件;
至少一个过滤器,所述过滤器将所述单元分隔为用于接收流体样品的过滤前腔室和用于接收能传导声波的流体的过滤后腔室;和
经设置与过滤后腔室相关连的基板,所述基板能在向其施加声波时共振;
其中,所述声波发生元件经设置与基板相关连从而使得所述声波发生元件引起所述基板的共振,所述基板随之传导声驻波穿过过滤后腔室里的流体和过滤前腔室里的流体样品。
2.如权利要求1所述的装置,其中所述声波发生元件与所述基板可逆相连。
3.如权利要求1或2所述的装置,其中在过滤前和过滤后腔室中的流体以相反方向连续移动通过所述过滤器,从而使得流体样品的净移动为进入过滤后腔室。
4.如权利要求1所述的装置,其中所述至少一个过滤器在分离过程中被扰动。
5.如权利要求3所述的装置,其中所述过滤器与能够在纵向或横向或同时两个方向上振动所述过滤器的固定器相关连。
6.如前述权利要求中任一项所述的装置,其中所述流体样品是液体。
7.如权利要求6所述的装置,其中所述液体是生物样品。
8.如权利要求7所述的装置,其中所述生物样品是血液样品、渗出液、尿液样品、精液、骨髓抽取物、脊髓液、来自组织的细胞悬浮液、粘液、痰液或唾液。
9.如前述权利要求中任一项所述的装置,其中所述固体组分是细胞组分。
10.如权利要求9所述的装置,其中所述细胞组分是白血细胞组分。
11.如权利要求10所述的装置,其中所述细胞组分是选自干细胞或祖细胞的未分化细胞。
12.如前述权利要求中任一项所述的装置,其中所述过滤器由合成材料制造。
13.如权利要求14所述的装置,其中所述孔径为约2~4微米。
14.如权利要求15所述的装置,其中所述孔径为约3微米。
15.如前述权利要求中任一项所述的装置,其中所述装置是自动化的。
16.从流体样品中分离固体组分的方法,所述方法包括步骤:
(i)将流体样品导入如权利要求1所述的装置;
(ii)过滤所述流体样品;和
(iii)从过滤前腔室中取出已分离的组分。
17.如权利要求16所述的方法,其中由所述声波造成的过滤前和过滤后腔室中的扰动在分离过程中持续进行。
18.如权利要求16所述的方法,其中由所述声波造成的过滤前和过滤后腔室中的扰动在分离过程中间歇进行。
19.如权利要求16-18中任一项所述的方法,其中所述方法进一步包括使过滤前和过滤后腔室中的流体以相反方向连续移动的步骤,从而使得流体样品的净移动为进入过滤后腔室。
20.如权利要求16-19中任一项所述的方法,其中所述方法进一步包括在分离过程中扰动过滤器的步骤。
21.如权利要求20所述的方法,其中所述过滤器与能够在纵向或横向或同时两个方向上振动所述过滤器的固定器相关连。
22.如权利要求16-21中任一项所述的方法,其中所述流体样品是液体。
23.如权利要求22所述的方法,其中所述液体是生物样品。
24.如权利要求23所述的方法,其中所述生物样品是血液样品、渗出液、尿液样品、精液、骨髓抽取物、脊髓液、来自组织的细胞悬浮液、粘液、痰液或唾液。
25.如权利要求16-24中任一项所述的方法,其中所述固体组分是细胞组分。
26.如权利要求25所述的方法,其中所述细胞组分是白血细胞组分。
27.如权利要求26所述的方法,其中所述细胞组分是选自干细胞或祖细胞的未分化细胞。
