Abstract
本发明提供在促进积蓄的脂肪组织的分解并发挥瘦身作用的在抑制或防止肥胖、改善肥胖体质方面有效的脂肪分解剂、瘦身剂和脂肪团改善剂。脂肪分解促进剂以黄花菜或其提取物和黄嘌呤衍生物为有效成分。
Description
技术领域
本发明涉及在肥胖的抑制或防止、肥胖体质的改善等方面有效的脂肪分解促进剂和瘦身剂。
背景技术
在各发达国家中因为运动不足和卡路里过度摄取而存在容易变得肥胖的环境要素。该肥胖是由于摄取的能量比消耗的能量过剩且过剩的能量在白色脂肪细胞中作为中性脂肪积蓄而引起的。
并且,因为在各发达国家,肥胖容易引发心肌梗塞、动脉硬化、抗胰岛素性等很多疾病,所以,从预防这些疾病的观点出发,在各发达国家中也成为问题。
另外,中性脂肪特别容易局部地在例如靠近肠部的腹部、腰、臀部、大腿部等处作为所谓皮下脂肪而积蓄,形成体型非常不理想的状态。如果水分和体内排泄物在脂肪细胞中贮留,则皮肤表面生成成为凸凹的脂肪团(ce1lulite),脂肪团在外观上给予皮肤柔软膨松感,在美容上不理想。
作为促进体内脂肪分解的机理,已知是抑制由腺苷酸环化酶的活化和PDE(磷酸二酯酶,Phosphodiesterase)的阻碍引起的cAMP(环状三磷酸腺苷,Cyclic adenosine monophosphate)分解,从而在脂肪细胞的水平上提高cAMP的比例。
例如,报告了通过摄取具有PDE阻碍作用的咖啡因和茶碱等黄嘌呤衍生物,从而提高作用于交感神经的去甲肾上腺素和肾上腺素等的作用,具有促进脂肪分解的作用(例如,参照专利文献1和2)。
另外,已知含有咖啡因的咖啡豆、绿茶、乌龙茶、马黛茶等植物也有促进脂肪分解的作用。另一方面,已知黄花菜是百合科的折叶萱草(Hemerocallis plicata Stepf)的花,已知具有利尿作用和改善浮肿作用,而近年来报告了具有促进脂肪分解的作用(例如,参照专利文献3)。

发明内容
本发明提供以下1)~10)相关的发明。
1)以黄花菜和黄嘌呤衍生物或者黄花菜的提取物和黄嘌呤衍生物为有效成分的脂肪分解促进剂。
2)以黄花菜和黄嘌呤衍生物或者黄花菜的提取物和黄嘌呤衍生物为有效成分的瘦身剂。
3)以黄花菜和黄嘌呤衍生物或者黄花菜的提取物和黄嘌呤衍生物为有效成分的脂肪团减少剂。
4)黄花菜和黄嘌呤衍生物或者黄花菜的提取物和黄嘌呤衍生物作为脂肪分解促进剂的用途。

图2表示以对照品为100%时的茶碱、黄花菜提取物、黄花菜茶碱混合物1和黄花菜茶碱混合物2的脂肪分解促进活性。黄花菜茶碱混合物1是(黄花菜提取物各浓度+茶碱1μg/mL)、黄花菜茶碱混合物2是(黄花菜提取物各浓度+茶碱3μg/mL)。
图3表示以对照品为100%时的氨茶碱、黄花菜提取物、黄花菜氨茶碱混合物1和黄花菜氨茶碱混合物2的脂肪分解促进活性。黄花菜氨茶碱混合物1是(黄花菜提取物各浓度+氨茶碱1μg/mL)、黄花菜氨茶碱混合物2是(黄花菜提取物各浓度+氨茶碱3μg/mL)。
具体实施方式
本发明涉及提供促进积蓄的脂肪组织、特别是促进皮下脂肪的分解从而能够更有效地实现抑制或防止肥胖、改善肥胖体质、减少脂肪团的化妆品、医药部外品、医药品或食品。
本发明人等发现:相比于分别单独使用时,在组合使用黄花菜或其提取物和咖啡因、茶碱等黄嘌呤衍生物时,可以大大地促进在脂肪细胞组织中积蓄的中性脂肪的分解,这些能够成为在抑制或防止肥胖、改善肥胖体质方面发挥优异效果的化妆品、医药部外品、医药品、食品。
根据本发明的脂肪分解促进剂等,能够抑制或防止肥胖、改善肥胖体质、减少脂肪团。
“黄花菜”是以百合科的折叶萱草(Hemerocallis plicata Stepf(中药大辞典,上海科学技术出版社和小学馆编,第1卷,株式会社小学馆,昭和60年,p114))、本萱草(Hemerocallis fulva L.(中药大辞典,上海科学技术出版社和小学馆编,第1卷,株式会社小学馆,昭和60年,p378))、薮萱草(Hemerocallis fulva L(中药大辞典,上海科学技术出版社和小学馆编,第1卷,株式会社小学馆,昭和60年,p381、p539))、黄花萱草(Hemerocallis flava L(中药大辞典,上海科学技术出版社和小学馆编,第1卷,株式会社小学馆,昭和60年,p378、p539))、小黄花菜(Hemerocallis minor Mill.(中药大辞典,上海科学技术出版社和小学馆编,第1卷,株式会社小学馆,昭和60年,p378、p539))和这些类似植物为基原的植物的花、蕾的总称。
在本发明中,所谓“黄花菜”是包含这一系列全部植物的黄花菜。可以良好地直接或干燥后使用在市场上流通的黄花菜的蕾。另外,干燥可以应用在植物的干燥中通常使用的方法,日晒干燥、风干、加热干燥或冷冻干燥中的任意一种都可以。
作为本发明的黄花菜提取物,可以列举在常温或加温下提取上述“黄花菜”而得到的溶剂提取液、其稀释液、其浓缩液或其干燥物等。提取方法可以是浸渍、煎出、浸出、回流提取、超临界提取、微波提取等中的任意一种。
作为在提取中使用的提取溶剂,可以使用极性溶剂或非极性溶剂的任意一种,可以将这些混合使用。
作为极性溶剂或非极性溶剂,例如可以列举水、醇类、酮类、酯类、链状和环状醚、聚醚类、烃类、芳香族烃类、吡啶类、超临界二氧化碳和油脂、蜡、其它油等。
在这里,作为醇类例如可以列举甲醇、乙醇、丙醇、丁醇、丙二醇、丁二醇等;作为酮类例如可以列举丙酮、甲乙酮等;作为酯类例如可以列举醋酸甲酯、醋酸乙酯等;作为链状和环状醚例如可以列举四氢呋喃、二乙醚等;作为聚醚类例如可以列举聚乙二醇等;作为烃类例如可以列举己烷、环己烷、石油醚等;作为芳香族烃类例如可以列举苯、甲苯等。
其中,优选单独使用水或醇类或使用水—醇类混合液,更优选水—醇类混合液,特别优选水—甲醇或水—乙醇混合液。在水—醇类混合液的情况中,混合比例(V/V)优选是0~80%、更优选是0~60%。
在本发明的黄花菜提取物中的提取溶剂的使用量,根据溶剂的种类而不同,但优选相对1质量份黄花菜干燥物是1~100质量份、更优选是5~20质量份。
另外,提取温度是5~70℃、优选是5~50℃、更优选是10~40℃。提取时间是1小时~30日、优选是7~14日。
上述黄花菜提取物也可以提取·配制后液体直接使用或稀释使用,但也可以将该提取物的液体减压浓缩、冷冻浓缩或膜浓缩或风干燥、加热干燥或冷冻干燥,调制成浓缩物、糊状或干燥物(粉末)而使用。
另外,上述黄花菜提取物还优选使用通过液液分配、固液分配、过滤膜、活性碳、吸附树脂、离子交换树脂等公知的技术除去了非活性杂质后的黄花菜提取物。还可以根据需要在施加了脱臭、脱色等处理后使用。
在本发明中,所谓黄嘌呤衍生物,指的是具有PDE阻碍活性且作为具有中枢兴奋作用、利尿作用等的物质而公知的化合物群。具体的,可以列举黄嘌呤和以下述通式(1)表示的化合物或其盐(例如盐酸、硫酸等无机酸盐,乙二胺、胆碱、醋酸、柠檬酸等有机酸盐等),
[式中,R1表示甲基或氢原子,R2表示氢原子或可以由1~2个羟基或碳原子数1~10的烷酰基(例如甲酰基、乙酰基、丙酰基和丁酰基等)取代的碳原子数1~12的烷基(例如,甲基、乙基、丙基、丁基、己基等)]。
更具体地,可以列举黄嘌呤、咖啡因、茶碱、氨茶碱、胆茶碱(cholinetheophylline)(胆茶碱(Oxtriphylline))、可可碱、二羟丙茶碱和羟丙茶碱等,优选氨茶碱、茶碱和咖啡因,这些黄嘌呤衍生物也可以组合2种以上。
在本发明中使用的黄嘌呤衍生物可以使用市售品,但可以通过从含有该衍生物的植物中进行提取和公知的化学合成法得到。
如在后述实施例中所示,本发明的黄花菜或其提取物和黄嘌呤衍生物在大鼠腹部皮下脂肪组织中,协同地增强由去甲肾上腺素引起的脂肪分解活性,所以,通过将其给药可以使身体苗条,因为即使在内脏组织中也发挥本活性,所以,可以作为发挥抑制、防止肥胖或改善体质等瘦身效果的脂肪分解促进剂、瘦身剂和脂肪团减少剂(以下为脂肪分解促进剂等)使用,另外还可以用于生产该脂肪分解促进剂等。即,本发明的脂肪分解促进剂等是促进在脂肪组织中积蓄的中性脂肪、特别是皮下脂肪的分解、更促进脂肪团的分解并发挥抑制、防止肥胖或改善体质等的瘦身效果的物质,可以作为使身体苗条的化妆品、医药部外品、医药品、食品等使用。
另外,脂肪分解促进剂等可以作为以抑制、防止肥胖或改善体质等生理功能为概念且配合在各种食品中而具有脂肪分解促进作用和瘦身效果的食品而利用,特别是作为表示了用于脂肪分解促进、瘦身或减少脂肪团的意图的食品、例如功能性食品、病人用饮食品、特定保健用食品而利用。
作为将本发明的脂肪分解促进剂等作为食品而使用时的形态,可以列举面包类、蛋糕类、面类、点心类、果冻类、冷冻食品、冰激凌类、乳制品、饮料、汤类等各种食品,除此以外,可以列举片剂、胶囊剂、糖浆剂等。
作为将本发明的脂肪分解促进剂等作为医药品使用时的给药形态,例如,可以列举片剂、胶囊剂、颗粒剂、散剂、糖浆剂等经口给药,或注射剂、坐剂、吸入剂、经皮吸收剂、外用剂等非经口给药。
作为将本发明的脂肪分解促进剂等作为医药部外品和化妆品使用时,可以制成皮肤外用剂、洗净剂、浴用剂、装扮用化妆品等,根据使用方法,可以以洗液、乳液、凝胶、乳脂、软膏剂、粉末、颗粒等各种剂型进行提供。
这样的化妆品、医药部外品、医药品、食品等可以分别由一般的制造方法,直接或和制剂上能够允许的载体一起混合、分散后,加工成所希望的形态而进行配制。在这样的制剂中,根据需要可以适当组合各种粉体、油或油状物质、防腐剂、抗氧化剂、湿润剂、缓冲剂、表面活性剂、增粘剂、活性增强剂、着色剂、香料而进行配合,除此以外还可以适当组合紫外线吸收剂、抗炎症剂、杀菌剂、血液流动促进剂、维生素类、具有脂肪代谢促进作用或脱共轭蛋白质表达促进作用的药物或天然物等的药效成分而进行配合。
作为粉体,例如可以列举白垩、滑石粉、漂白土、高岭土、云母、淀粉、橡胶、胶体二氧化硅、聚丙烯酸钠、纤维素粉、尼龙粉、交联型聚硅酮粉末、交联型甲基聚硅氧烷、多孔纤维素粉、多孔尼龙粉、无水二氧化硅、氧化锌、氧化钛等。作为油或油状物质,可以列举油脂类、蜡类、烃类、高级脂肪酸类、高级醇类、酯类、精油类、硅油类、氟油类等。

作为具有脂肪代谢促进作用或脱共轭蛋白质表达促进作用的药物或天然物等药效成分,例如可以列举β-肾上腺素作用兴奋药、α-肾上腺素作用抑制药、联吡啶衍生物、异黄酮酸、海藻提取物(rodisterol)、二十八烷醇、3,4-二羟基苯乙醇(Hydroxytyrosol)、圆柚油(grapefruit oil)、覆盆子酮(Raspberry ketone)、姜油酮、蓟属、胡椒科、芸香科、防己科、吊灯树属植物、甘茶蔓、苍术、安息香、薏苡仁、小豆、茴香、黄钟木属(Tabebuia rosea)、童氏老鹳草(Geranium thunbergii)、黄芩、桃、百里香、芍药、茶、可乐树、日本獐牙菜(Swertia japonica)、桂皮、ジユ、鼠尾草、艾菊、枇杷、岩藻、人参、香菇、虎耳草、银杏等其它植物的提取物。
在本发明的脂肪分解促进剂等中的黄花菜或其提取物和黄嘌呤衍生物的含量优选为,黄花菜或其提取物换算为干燥物是0.0001~30质量%、黄嘌呤衍生物是0.001~2质量%,更优选黄花菜或其提取物换算为干燥物是0.0002~5质量%、黄嘌呤衍生物是0.005~1质量%。
另外,制成为外用剂时,本发明的黄花菜或其提取物和黄嘌呤衍生物的含量是0.005质量%以上、优选是0.01~30质量%、更优选是0.05~20质量%、特别优选是0.1~10质量%。
另外,制成为经口给药用制剂时,本发明的黄花菜或其提取物和黄嘌呤衍生物的含量一般是0.01~80质量%、优选是0.05~70质量%、特别优选是0.1~50质量%。
黄花菜或其提取物和黄嘌呤衍生物的含有比率为,相对1质量份换算为干燥物的黄花菜或其提取物,黄嘌呤衍生物至少是0.0001质量份以上、优选是0.001~10质量份。
将本发明的脂肪分解促进剂等作为食品或医药品给予时,其给予量根据这些有效成分的添加形态和给予形态而不同,可以从广泛范围中进行选择。例如,从效果方面出发优选,黄花菜或其提取物给予量是成人每日0.01~100g、优选是0.1~20g、特别优选是1.0~10g;黄嘌呤衍生物给予量是成人每日0.001~10g、优选是0.01~5g、特别优选是0.05~5g。
另外,所谓本发明的瘦身方法,是用于使外表苗条且看上去漂亮的方法,例如,包括非医师的美容师等进行的美容行为,除此以外,也包含所谓的医师等进行的医疗行为。

[制造例黄花菜提取物的制造]
在40g黄花菜(新和物产(株))中加入400mL50%(v/v)乙醇-水混合溶液,在室温(20~30℃)下提取7日后,过滤,得到340mL提取液。另外,从该提取液中减压馏去溶剂后,得到18.4g干燥物。
[试验例1]
黄花菜提取物使用上述制造例的提取物,咖啡因使用市售品(和光纯药工业株式会社)。
按照罗德贝尔的方法(Rodbell,M.,J.Biol.Chem.,239,375(1964)),从1~3只维斯塔尔类雄性大鼠(体重170~230g)的腹部皮下脂肪组织中,使用胶原酶溶液配制游离脂肪细胞。
作为被试验物,使用黄花菜提取物(单独物)、咖啡因(单独物)、黄花菜提取物和咖啡因混合物(以下称为黄花菜咖啡因混合物)1(黄花菜提取物+咖啡因10μg/mL)、以及黄花菜咖啡因混合物2(黄花菜提取物+咖啡因30μg/mL)。
在去甲肾上腺素被配制成为90nM的含有牛血清白蛋白的Hanks缓冲液(反应液)中添加被试验物,将上述细胞在37℃培养2小时,利用酶法(Free Glycerol Reagent,シグマアルドリツチ社)测定游离的甘油。
此时,在反应液中添加各被试验物,以使黄花菜提取物的最终浓度分别成为0,3,10,30和100μg/mL,咖啡因的最终浓度分别成为0,10和30μg/mL。
对照是仅用不含被试验物的去甲肾上腺素培养。
脂肪分解促进活性以对照(control)为100%,用下式算出,在表1和图1中表示这些结果。
脂肪分解促进活性(%)=(在各组中的游离甘油值/在对照组中的游离甘油值)×100

***黄花菜提取物是黄花菜提取物各浓度,黄花菜咖啡因混合物1是(黄花菜提取物各浓度+咖啡因10μg/mL),黄花菜咖啡因混合物2是(黄花菜提取物各浓度+咖啡因30μg/mL)。
如在表1和图1中所示,咖啡因、黄花菜提取物、黄花菜咖啡因混合物显示了大于100%的脂肪分解活性,但组合咖啡因和黄花菜提取物而得到的黄花菜咖啡因混合物的情况通过协同效果显示了高的脂肪分解活性。另外,黄花菜咖啡因混合物的咖啡因含量增多则促进脂肪分解。
[试验例2]
将咖啡因取代为茶碱,和上述试验例1同样进行试验。作为被试验物,使用黄花菜提取物(单独物)、茶碱(单独物)、黄花菜提取物和茶碱的混合物(以下称为黄花菜茶碱混合物)1(黄花菜提取物+茶碱1μg/mL)、以及黄花菜茶碱混合物2(黄花菜提取物+茶碱3μg/mL)。
茶碱使用市售品(シグマアルドリツチジヤパン株式会社制)。
此时,在反应液中添加各被试验物,以使得黄花菜提取物的最终浓度分别成为0,10,30与100μg/mL,茶碱的最终浓度分别成为0,1与3μg/mL。

如在表2和图2中所示,茶碱、黄花菜提取物、黄花菜茶碱混合物显示了大于100%的脂肪分解活性,但组合茶碱和黄花菜提取物而得到的黄花菜茶碱混合物的情况通过协同效应而显示了高的脂肪分解活性。另外,黄花菜茶碱混合物的茶碱含量如果增多,则促进脂肪分解。
[试验例3]
将咖啡因取代为氨茶碱,和上述试验例1同样进行试验。作为被试验物,使用黄花菜提取物和氨茶碱的混合物(以下称为黄花菜氨茶碱混合物)1(黄花菜提取物+氨茶碱1μg/mL)、和黄花菜氨茶碱混合物2(黄花菜提取物+氨茶碱3μg/mL)。
氨茶碱使用市售品(シグマアルドリツチジヤパン株式会社制)。
此时,在反应液中添加各被试验物,以使得黄花菜提取物的最终浓度分别成为0,10,30与100μg/mL,使氨茶碱的最终浓度分别成为0,1与3μg/mL。

Claims (8)
1.一种脂肪分解促进剂,其特征在于,
以黄花菜和氨茶碱、黄花菜和茶碱、黄花菜和咖啡因、黄花菜的提取物和氨茶碱、黄花菜的提取物和茶碱、或者黄花菜的提取物和咖啡因为有效成分。
2.如权利要求1所述的脂肪分解促进剂,其特征在于,
相对于1质量份换算成干燥物的黄花菜或其提取物,含有0.0001~10质量份的氨茶碱、茶碱或者咖啡因。
3.一种瘦身剂,其特征在于,
以黄花菜和氨茶碱、黄花菜和茶碱、黄花菜和咖啡因、黄花菜的提取物和氨茶碱、黄花菜的提取物和茶碱、或者黄花菜的提取物和咖啡因为有效成分。
4.一种脂肪团减少剂,其特征在于,
以黄花菜和氨茶碱、黄花菜和茶碱、黄花菜和咖啡因、黄花菜的提取物和氨茶碱、黄花菜的提取物和茶碱、或者黄花菜的提取物和咖啡因为有效成分。
5.如权利要求1或2所述的脂肪分解促进剂,其特征在于,
是使身体变得苗条的脂肪分解促进剂。
6.黄花菜和氨茶碱、黄花菜和茶碱、黄花菜和咖啡因、黄花菜的提取物和氨茶碱、黄花菜的提取物和茶碱、或者黄花菜的提取物和咖啡因在制造脂肪分解促进剂中的用途。
7.黄花菜和氨茶碱、黄花菜和茶碱、黄花菜和咖啡因、黄花菜的提取物和氨茶碱、黄花菜的提取物和茶碱、或者黄花菜的提取物和咖啡因在制造瘦身剂中的用途。
8.黄花菜和氨茶碱、黄花菜和茶碱、黄花菜和咖啡因、黄花菜的提取物和氨茶碱、黄花菜的提取物和茶碱、或者黄花菜的提取物和咖啡因在制造脂肪团减少剂中的用途。
