Description
Die vorliegende Erfindung betrifft neue substituierte Pyri-midinyl(thio)-phosphor(phosphon)-säureester bzw.- esteramide, ein Verfahren zu ihrer Herstellung und ihre Verwendung als Insektizide, Akarizide und Nematizide.
Es ist bereits bekannt, dass Pyrimidinylthionophosphonsäu-reester, z.B. 0-(2,4-DimethyIpyrimidin(6)yl)-0-methyl- bzw. -O-äthyl-thionomethan- bzw. -äthan-phosphonsäureester, in-

R2, R3, X und Y die oben angegebene Bedeutung haben und 5o Hai für Halogen, insbesondere Chlor steht, gegebenenfalls in Gegenwart eines Lösungs- oder Verdünnungsmittels umsetzt.
Uberraschenderweise zeigen die erfindungsgemässen substituierten Pyrimidinyl(thio)-phosphor(phosphon)-säureester bzw. -esteramide eine bessere insektizide, akarizide und nemati-zide Wirkung als die vorbekannten Pyrimidinylthionophos-phonsäureester analoger Konstitution und gleicher Wirkungsrichtung. Die erfindungsgemässen Stoffe stellen somit eine echte Bereicherung der Technik dar.
Verwendet man beispielsweise 6-Hydroxy-4-n-propoxyme-thyl-pyrimidin und S-Äthyl-thiolothionoäthanphosphonsäure-esterchlorid als Ausgangsstoffe, so kann der Reaktionsverlauf durch das folgende Formelschema wiedergegeben werden:

Die zu verwendenden Ausgangsstoffe sind durch die Formeln (II) und (III) allgemein definiert. Vorzugsweise stehen darin jedoch:
R für Wasserstoff, geradkettiges oder verzweigtes Alkyl mit 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen, geradkettiges oder verzweigtes Alkoxy, Alkylthio oder Dialkylamino mit 1 bis 4, insbesondere 1 oder 2 Kohlenstoffatomen je Alkylrest,
R1 für Chlor, geradkettiges oder verzweigtes Alkoxy mit 1 bis 4, insbesondere 1 bis 3 Kohlenstoffatomen, geradkettiges oder verzweigtes Alkylthio mit 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen oder geradkettiges oder verzweigtes Dialkylamino mit 1 bis 4, insbesondere 1 oder 2 Kohlenstoffatomen je Alkylrest, insbesondere die genannten Alkoxy-, Al-kylthio- und Dialkylaminoreste.
R2 für Phenyl, geradkettiges oder verzweigtes Alkyl mit 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen, geradkettiges oder verzweigtes Alkoxy bzw. Monoalkylamino mit 1 bis 5, insbesondere 1 bis 4 Kohlenstoffatomen je Alkylrest,
R3 für geradkettiges oder verzweigtes Alkyl mit 1 bis 6, insbesondere 1 bis 4 Kohlenstoffatomen,
X für Schwefel und Y für Sauerstoff oder Schwefel.
Die als Ausgangsstoffe zu verwendenden substituierten 6-Hydroxypyrimidine (II) sind nach literaturbekannten Verfahren leicht herstellbar, z.B. indem man Acetessigsäurealkylester halogeniert, anschliessend mit Alkylmercaptiden umsetzt und mit Amidinen den Ring schliesst.

Die weiterhin als Ausgangsstoffe zu verwendenden (Thio) Phosphor(phosphon)-saureester bzw. -esteramidhalogenide (III) sind literaturbekannt und nach üblichen Verfahren herstellbar.

Das Verfahren zur Herstellung der erfindungsgemässen Verbindung wird bevorzugt unter Mitverwendung geeigneter Lösungs- oder Verdünnungsmittel durchgeführt. Als solche kommen praktisch alle inerten organischen Solventien infrage. 15 Hierzu gehören insbesondere aliphatische und aromatische, gegebenenfalls chlorierte Kohlenwasserstoffe, wie Benzol, Toluol, Xylol, Benzin, Methylenchlorid, Chloroform, Tetrachlorkohlenstoff, Chlorbenzol, oder Äther, wie Diäthyl- und Dibutyl-äther, Dioxan, ferner Ketone, beispielsweise Aceton, Methyl-20 äthyl-, Methylisopropyl- und Methylisobutylketon, ausserdem Nitrile, wie Aceton- und Propionitril.
Als Säureakzeptoren können alle üblichen Säurebindemittel Verwendung finden. Besonders bewährt haben sich Alkalicar-bonate und -alkoholate, wie Natrium- und Kaliumcarbonat, Na-25 trium- und Kaliummethylat bzw. äthylat oder Kalium-tert.-bu-tylat, ferner aliphatische, aromatische oder heterocyclische Amine, beispielsweise Trimethylamin, Triäthylamin, Dimethyl-anilin, Dimethylbenzylamin und Pyridin.
Die Reaktionstemperatur kann innerhalb eines grösseren 30' Bereiches variiert werden. Im allgemeinen arbeitet man zwischen 0 und 120 °C, vorzugsweise bei 20 bis 80 °C.
Die Umsetzung lässt man im allgemeinen bei Normaldruck ablaufen.
Zur Durchführung des Verfahrens rührt man das 6-Hydr-35 oxy-pyrimidin (II) vorzugsweise in 10%igem molaren Über-schuss in Gegenwart eines Säureakzeptors in einem Lösungsmittel bei erhöhter Temperatur einige Zeit und fügt zu der abgekühlten Lösung die Phosphorsäurekomponente (III). Nach beendeter Reaktion wird die Mischung mit Wasser verdünnt, 40 mit einem organischen Lösungsmittel ausgeschüttelt und die organische Phase in üblicher Weise durch Trocknen und Abdestil-lieren des Lösungsmittels aufgearbeitet.
Die neuen Verbindungen fallen meist in Form von Ölen an, die sich zum Teil nicht unzersetzt destillieren lassen, jedoch 45 durch sogenanntes «Andestillieren», d.h. längeres Erhitzen unter vermindertem Druck auf mässig erhöhte Temperatur von den letzten flüchtigen Anteilen befreit und auf diese Weise gereinigt werden können. Zu ihrer Charakterisierung dient der Brechungsindex. Einige Produkte erhält man in kristalliner so Form ; sie sind durch ihren Schmelzpunkt charakterisiert.
Wie bereits mehrfach erwähnt, zeichnen sich die erfindungsgemässen substituierten Pyrimidinyl(thio)-phosphor(phos-phon)-säureester bzw. -esteramide durch eine hervorragende insektizide, akarizide und nematizide Wirkung aus. Sie wirken 55 gegen Pflanzen-, Hygiene- und Vorratsschädlinge sowie auf dem veterinär-medizinischem Sektor und besitzen bei geringer Phytotoxizität sowohl eine gute Wirkung gegen saugende als auch fressende Insekten und Milben.
Aus diesem Grunde können die erfindungsgemässen Verso bindungen mit Erfolg im Pflanzenschutz sowie auf dem Hygiene-, Vorratsschutz- und Veterinärsektor als Schädlingsbekämpfungsmittel eingesetzt werden.
Die Wirkstoffe eignen sich bei guter Pflanzenverträglichkeit und günstiger Warmblütertoxizität zur Bekämpfung von tieri-65 sehen Schädlingen, insbesondere Insekten, Spinnentieren und Nematoden, die in der Landwirtschaft, in Forsten, im Vorratsund Materialschutz sowie auf dem Hygienesektor vorkommen. Sie sind gegen normal sensible und resistente Arten sowie gegen
alle oder einzelne Entwicklungsstadien wirksam. Zu den oben erwähnten Schädlingen gehören:

Die Wirkstoffe können in die üblichen Formulierungen übergeführt werden, wie Lösungen, Emulsionen, Spritzpulver, Suspensionen, Pulver, Stäubemittel, Schäume, Pasten, lösliche Pulver, Granulate, Aerosole, Suspensions-Emulsionskonzentra-te, Saatgutpuder, Wirkstoff-imprägnierte Natur- und synthetische Stoffe, Feinstverkapselungen in polymeren Stoffen und in Hüllmassen für Saatgut, ferner in Formulierungen mit Brennsätzen, wie Räucherpatronen, -dosen, -spiralen u.ä. sowie TJLV-Kalt- und Warmnebel-Formulierungen.
Diese Formulierungen werden in bekannter Weise hergestellt, z.B. durch Vermischen der Wirkstoffe mit Streckmitteln, also flüssigen Lösungsmitteln, unter Druck stehenden verflüssigten Gasen und/oder festen Trägerstoffen, gegebenenfalls unter Verwendung von oberflächenaktiven Mitteln, also Emulgiermitteln und/oder Dispergiermitteln und/oder schaumerzeugenden Mitteln. Im Falle der Benutzung von Wasser als Streckmittel können z.B. auch organische Lösungsmittel als Hilfslösungsmittel verwendet werden. Als flüssige Lösungsmittel kommen im wesentlichen infrage: Aromaten, wie Xylol, Toluol,
oder Alkylnaphthaline, chlorierte Aromaten oder chlorierte aliphatische Kohlenwasserstoffe, wie Chlorbenzole, Chloräthylene oder Methylenchlorid, aliphatische Kohlenwasserstoffe, wie Cy-clohexan oder Paraffine, z.B. Erdölfraktionen, Alkohole, wie Butanol oder Glycol sowie deren Äther und Ester, Ketone, wie Aceton, Methyläthylketon, Methylisobutylketon oder Cyclo-hexanon, stark polare Lösungsmittel, wie Dimethylformamid und Dimethylsulfoxid, sowie Wasser; mit verflüssigten gasförmigen Streckmitteln oder Trägerstoffen sind solche Flüssigkeiten gemeint, welche bei normaler Temperatur und unter Normaldruck gasförmig sind, z.B. Es können in den Formulierungen Haftmittel wie Carboxy-methylcellulose, natürliche und synthetische pulverige, körnige oder latexförmige Polymere verwendet werden, wie Gummiar-abicum, Polyvinylalkohol, Polyvinylacetat.

Die Anwendung der erfindungsgemässen Wirkstoffe erfolgt in Form ihrer handelsüblichen Formulierungen und/oder den aus diesen Formulierungen bereiteten Anwendungsformen.
Der Wirkstoffgehalt der aus den handelsüblichen Formulierungen bereiteten Anwendungsformen kann in weiten Bereichen variieren. Die Wirkstoffkonzentration der Anwendungsformen kann von 0,0000001 bis zu 100 Gew.-% Wirkstoff, vorzugsweise zwischen 0,01 und 10 Gew.-% liegen.
Die Anwendung geschieht in einer den Anwendungsformen angepassten üblichen Weise.
Bei der Anwendung gegen Hygiene- und Vorratsschädlinge zeichnen sich die Wirkstoffe durch eine hervorragende Residualwirkung auf Holz und Ton sowie durch eine gute Alkalistabilität auf gekälkten Unterlagen aus.
Die Anwendung der erfindungsgemässen Wirkstoffe geschieht im Veterinärsektor in bekannter Weise, wie durch orale Anwendung in Form von beispielsweise Tabletten, Kapseln, Tränken, Granulaten, durch dermale Anwendung in Form beispielsweise des Tauchens (Dippen), Sprühens (Sprayen), Auf-giessens (pour-on and spot-on) und des Einpuderns sowie durch parenterale Anwendung in Form beispielsweise der Injektion.

Phorbia antiqua-Maden im Boden 3 Gewichtsteile Aceton 1 Gewichtsteil Alkylarylpolyglykoläther mischt. Dabei spielt die Konzentration des Wirkstoffs in der Zubereitung praktisch keine Rolle, entscheidend ist allein die Wirkstoffgewichtsmenge pro Volumeneinheit Boden, welche in ppm (= mg/1) angegeben wird. Man füllt den Boden in Töpfe 5 und lässt diese bei Raumtemperatur stehen.
Nach 24 Stunden werden die Testtiere in den behandelten Boden gegeben und nach weiteren 2 bis 7 Tagen wird der Wirkungsgrad des Wirkstoffs durch Auszählen der toten und leben-Zur Herstellung einer zweckmässigen Wirkstoffzubereitung den Testinsekten in % bestimmt. Zur Herstellung einer zweckmässigen Wirkstoffzubereitung vermischt man 1 Gewichtsteil Wirkstoff mit der angegebenen Menge Lösungsmittel, gibt die angegebene Menge Emulgator zu und verdünnt das Konzentrat mit Wasser auf die gewünschte Konzentration.
Die Wirkstoffzubereitung wird innig mit Boden vermischt,
der mit den Testnematoden stark verseucht ist. Dabei spielt die Konzentration des Wirkstoffs in der Zubereitung praktisch keine Rolle, entscheidend ist allein die Wirkstoffmenge pro Volumeneinheit Boden, welche in ppm angegeben wird. Man füllt den behandelten Boden in Töpfe, sät Salat ein und hält die 60 Töpfe bei einer Gewächshaus-Temperatur von 27 °C.
Nach vier Wochen werden die Salatwurzeln auf Nemato-denbefall (Wurzelgallen) untersucht und der Wirkungsgrad des Wirkstoffs in % bestimmt. Der Wirkungsgrad ist 100%, wenn der Befall vollständig vermieden wird, er ist 0%, wenn der Be-65 fall genau so hoch ist wie bei den Kontrollpflanzen in unbehandeltem, aber in gleicher Weise verseuchtem Boden.

Die zur Herstellung der erfindungsgemässen Verbindungen benötigten Ausgangsstoffe (II) werden z.B. wie im folgenden beschrieben hergestellt:
a) Cl-CH2-CO-CH2-CO-OC2H5 In einer Lösung von 84 g (1 Mol) Diketen in trockenem Tetrachlorkohlenstoff leitet man bei — 20 °C 72 g (1 Mol)
Chlor ein, tropft die Reaktionslösung anschliessend unter Rühren in 200 ml Äthanol, wobei die Temperatur 0 °C nicht über-65 steigen sollte, zieht anschliessend das Solvens am Rotationsverdampfer ab und destilliert den Rückstand. Es werden 155 g (94% derTheori) Y-Chloracetessigsäureäthylester mit dem Siedepunkt 90 °C/7 Torr gewonnen.

b) c2h5o-ch2-co-ch2-co-oc2h5
2,2 Mol Natriumäthylat in Äthanol - hergestellt aus 55,2 g (2,2 Mol) Natrium in 500 ml Äthanol - werden mit 500 ml Te-trahydrofuran verdünnt. Man versetzt die Lösung bei Raumtemperatur so schnell mit 164,5 g (1 Mol) y-Chloracetatessig-säureäthylester, dass die Reaktionstemperatur auf 50 °C ansteigt, kühlt die Mischung anschliessend auf Raumtemperatur ab, fügt 72 g (1,2 Mol) Eisessig hinzu, dampft das Lösungsmittel am Rotationsverdampfer ab, schüttelt den Rückstand mit 250 ml Wasser, extrahiert ihn 2 X mit je 250 ml Methylenchlorid und trocknet die vereinigten organischen Phasen über Magnesiumsulfat. Das Lösungsmittel wird unter vermindertem Druck abgezogen und der Rückstand destilliert. Man erhält so 123,7 g (71 % der Theorie) v-Äthoxyacetessigsäureäthylester mit dem Siedepunkt von 75 °C/3 Torr.
Analog wird der 7-Methoxyacetessigsäuremethylester in einer Ausbeute von 63 % der Theorie mit dem Siedepunkt 76 °C/ 7 Torr hergestellt.
c) n-C3H7S-CH2-CO-CH2-CO-OC2H5
Zu einer Lösung von 300 g (2,3 Mol) Acetessigsäureäthyl-ester in 350 ml Äther tropft man bei 0 °C innerhalb von 90 Minuten 367 g (2,3 Mol) Brom, rührt die Mischung anschliessend 1 Stunde bei Raumtemperatur nach, versetzt sie unter Eiskühlung mit 500 ml Wasser, trennt die Phasen und wäscht die Ätherlösung einmal mit 100 ml einer 10%igen Natriumbicarbo-natlösung. Die über Magnesiumsulfat getrocknete organische Phase tropft man bei Raumtemperatur in eine äthanolische Lösung von 2 Mol Natriuirtpropylmercaptid- hergestellt aus 46 g (2 Mol) Natrium, 600 g Äthanol und 152 g (2 Mol) n-Propyl-mercaptan - rührt den Ansatz noch 1 Stunde bei Raumtemperatur nach, schüttelt ihn anschliessend mit 2 Mol Natronlauge (1334 g einer 6%igen Lösung) aus, verwirft die Ätherschicht, säuert die wässrige Lösung mit konzentrierter Salzsäure bis pH~2 an, schüttelt sie 3 X mit je 300 ml Methylenchlorid aus, trocknet die vereinigten organischen Phasen über Magnesiumsulfat und destilliert das Lösungsmittel ab. Nach der Destillation des Rückstandes werden 200 g (50% der Theorie) y-n-Propyl-mercaptoacetessigester mit dem Siedepunkt 85 °C/0,7 Torr gewonnen.
Analog werden hergestellt:
Y-Methylmercaptoacetessigester in 49%iger Ausbeute mit dem Siedepunkt von 100 °C/ 2 Torr.
Y-Äthylmercaptoacetessigsäureester in 56%iger Ausbeute mit dem Siedepunkt von 99 °C/1 Torr

2) Abweichend von der oben angegebenen Vorschrift wird in diesem Fall die Reaktion bei — 5 bis 0 °C durchgeführt; man 40 rührt die Mischung anschliessend noch 12 Stunden bei 0 °C nach.

Eine Lösung von 187 g (2 Mol) Acetamidinhydrochlorid und 292 g (2 Mol) y-Methoxyacetessigsäuremethylester in 1000 ml Methanol wird bei 5 °C mit 968 g (4 Mol) Natriumme-thylat in Methanol versetzt, das Gemisch 3 Stunden auf 50 °C erwärmt und dann unter vermindertem Druck das Lösungsmittel abgezogen. Der Rückstand wird in 500 ml Eiswasser gelöst, unter Aussenkühlung mit konzentrierter Salzsäure auf pH~6 gebracht, der Niederschlag abgesaugt, mit Äther gewaschen und anschliessend im Exsikkator über Phosphorpentoxid getrocknet. Man erhält 180 g (58% der Theorie) 2-Methyl-4-methoxy-methyl-6-hydroxy-pyrimidin in Form eines weissen Pulvers mit einem Schmelzpunkt von 168-170 °C.

Ein Gemisch aus 75 g des wie unter d) beschrieben hergestellten 4-Chlormethyl-2-methyl-6-hydroxy-pyrimidins, 73 g (1 Mol) Diäthylamin, 300 ml Wasser und 300 ml Äthanol wird zwei Stunden unter Rückfluss erhitzt, anschliessend zur Trockne eingedampft, mit 500 ml Aceton versetzt und durch Filtration vom ausgeschiedenen Hydrochlorid befreit. Danach wird die Acetonlösung eingeengt, der feste Rückstand mit Äther verrührt, abgesaugt und der Feststoff unter vermindertem Druck über Phosphorpentoxid getrocknet. Es verbleiben 60 g (62% der Theorie) 4-Diäthylaminomethyl-2-methyl-6-hydroxypyri-midin in Form eines weissen Pulvers mit einem Schmelzpunkt von 112 °C.

- 635 3502PATENTANSPRÜCHE 1. Substituierte Pyrimidinyl(thio)-phosphor(phosphon)-säureester bzw. -esteramide der Formel r -ch sektizide und akarizide Eigenschaften haben (vergleiche US-Patentschrift 3 216 894).Es wurde nun gefunden, dass die neuen substituierten Pyri-midinyl(thio)-phosphor(phosphon)-säureester bzw. -esteramide s der Formel(I)Rr1-ch,N•O-Pin welcherR für Wasserstoff, Alkyl, Alkoxy, Alkylthio oder Alkyl-amino,R1 für Halogen, Alkoxy, Alkylthio oder Alkylamino,R2 für Alkyl, Alkoxy, Alkylamino oder Phenyl,R3 für Alkyl undX und Y, welche gleich oder verschieden sind, für Sauerstoff oder Schwefel stehen.
- 2. Verfahren zur Herstellung von substituierten Pyrimidi-nyl(thio)-phosphor(phosphon)-säureestern bzw. -esteramiden der Formel I, dadurch gekennzeichnet, dass man 6-Hydroxy-pyrimidine der Formel(I)r(II)in welcherR und R1 die in Anspruch 1 angegebene Bedeutung haben, gegebenenfalls in Form der entsprechenden Alkali-, Erdalkalibzw. Ammoniumsalze mit (Thio)Phosphor-(phosphon)-säure-ester- bzw. esteramidhalogeniden der Formel in welcher15 R für Wasserstoff, Alkyl, Alkoxy, Alkylthio oder Alkylamino,R1 für Halogen, Alkoxy, Alkylthio oder Alkylamino,R2 für Alkyl, Alkoxy, Alkylamino oder Phenyl,R3 für Alkyl und 20 X und Y, welche gleich oder verschieden sind, für Sauerstoff oder Schwefel stehen,eine ausgezeichnete insektizide, akarizide und nematizide Wirkung besitzen.Weiterhin wurde gefunden, dass die neuen substituierten Pyrimidinyl(thio)-phosphor(phosphon)-säureester bzw. -esteramide der Formel (I) erhalten werden, wenn man 6-Hydroxy-pyrimidine der Formel r1-ch35 in welcher(ii)Hal-P/YR3\r2(III)40R und R1 die oben angegebene Bedeutung haben, gegebenenfalls in Gegenwart eines Säureakzeptors oder gegebenenfalls in Form der Alkali-, Erdalkali- bzw. Ammoniumsalze, mit (Thio)Phosphor-(phosphon)-säureester- bzw. -esteramidhalo-geniden der Formel in welcherR2, R3, X und Y die oben angegebene Bedeutung haben und Hai für Halogen steht, umsetzt.
- 3. Insektizide, akarizide und nematizide Mittel, gekennzeichnet durch einen Gehalt an Verbindungen der Formel I.
- 4. Verwendung von Verbindungen der Formel I zur Bekämpfung von Insekten, Milben und Nematoden.X11 /Hal-P.45 \YR3R2(HI)
