Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Erythromycinderivate der Formeln Ia und Ib wie in Patentanspruch 1 definiert.

Aus der Publikation von E. H. Massey et al. in J. med. Chem. 17 (1974) 105—107, sind Kondensationsprodukte von Arylaldehyden mit Erythromycylamin bekannt. Die Autoren berichten dort über Versuche, eine Reihe von Aldehyden mit Erythromycylamin zu kondensieren. Dabei entstanden mit Arylaldehyden N-Arylidenerythromycylamine, die unter Einbeziehung des Cu- oder Cc-Sauerstoffs eine Tetrahydro-1,3-oxazin- oder eine Hexahydro-l,3-oxazepin-Struktur besitzen, eine genaue Zuordnung der Strukturen erfolgte nicht.
Die Verbindungen der Formeln Ia und Ib werden mit Hilfe des in Patentanspruch 1 definierten Verfahrens hergestellt.
Welcher der Formeln Ia und Ib die im Einzelfall entstehenden Verfahrensprodukte zuzuordnen sind, konnte bisher trotz eingehender Untersuchungen mittels IR-Spektro-skopie und NMR-Spektroskopie noch nicht geklärt werden.
Die Umsetzung wird vorzugsweise in einem Lösungsoder Suspensionsmittel bei Temperaturen zwischen 0 und 150° C durchgeführt. Als Lösungs- oder Suspensionsmittel kommen vorzugsweise polare Lösungsmittel, wie Wasser, Alkohole, Dioxan, Dimethylformamid, Dimethylsulfoxyd, sowie Gemische dieser Lösungsmittel in Betracht.
Das Verfahren lässt sich mit besonderem Vorteil auf Ausgangsverbindungen der Formel III anwenden, wie man sie aus Acetalen der Formel IV

worin Ru und R12, die gleich oder voneinander verschieden sein können, je eine Alkylgruppe bedeuten oder zusammen mit der dazwischenliegenden Gruppe

einen 5- bis 7-gliedrigen Ring bilden, durch Spaltung mit Hilfe von Säuren oder sauren Ionenaustauschern erhält. Dabei ist es besonders vorteilhaft, die in situ gebildete Verbindung der Formel III direkt, d. h. ohne Zwischenisolierung, der Kondensationsreaktion zu unterwerfen.
Bedeutet R in den Verbindungen der Formel III bzw. IV eine freie oder monosubstituierte Aminogruppe, so empfiehlt es sich, diese Aminogruppe vor der Umsetzung mit Erythromycylamin mit einer Schutzgruppe zu versehen, die nach Beendigung der Umsetzung wieder abgespalten wird. Als Schutzgruppen kommen insbesondere die in der Peptid-chemie bekannten Aminosäure-Schutzgruppen, beispielsweise die Benzyl-, Trityl- oder Carbobenzoxycarbonylgruppe, in Betracht.
Das als Ausgangsverbindung dienende Erythromycylamin der Formel II ist literaturbekannt. Seine Herstellung ist beispielsweise in J. med. Chem. 17 (1974) 105—107, beschrieben. Die Aldehyde und Acetale der Formeln III und IV sind ebenfalls literaturbekannt oder lassen sich in Anlehnung an literaturbekannte Methoden herstellen.
Für/ die nachträgliche Überführung der Verbindungen der Pormeln Ia und Ib in ihre Säureadditionssalze mit anorganischen oder organischen Säuren eignen sich allgemein zur Bildung physiologisch verträglicher Säureadditionssalze verwendbare Säuren, wie beispielsweise Salzsäure, Bromwas-sersföffsäure, Schwefelsäure, Essigsäure, Zitronensäure oder Laurylschwefelsäure.
Die Erythromycinderivate der Formeln Ia und Ib und ihre Salze besitzen wertvolle pharmakologische Eigenschaften; sie sind insbesondere wirksam gegen gram-positive und gramnegative Bakterien.
Die Untersuchungen auf die antibakterielle Wirksamkeit wurden nach dem Agar-Diffusionstest und nach dem Reihen-Verdünnungstest in Anlehnung an die von P. Klein in «Bakteriologische Grundlagen der chemotherapeutischen Laboratoriumspraxis», Springer-Verlag, 1957, Seiten 53 bis 76 und 87 bis 109, beschriebenen Methoden durchgeführt.
Besonders gut antibakteriell noch in Konzentrationen von 0,3 bis 5 «g/ml gegen Staphylococcus aureus SG 511 und von 10 bis 40 /ig/ml gegen Escherichia coli wirken folgende Substanzen:

Die akute Toxizität der vorstehend genannten Verbindungen wurde an der Maus bestimmt. Bei oraler und subkutaner Applikation zeigen alle diese Verbindungen LDso-Werte > 1 g/kg Maus.
Die folgenden Beispiele sollen die Erfindung näher erläutern.
Beispiel 1
Glykolaldehyd-Erythromycylamin-Kondensationsprodukt 42 g Erythromycylamin und 4,2 g Glykölaldehyd wurden in 600 ml absolutem Äthanol 24 Stunden bei Raumtemperatur gerührt. Nach Verdampfen des Äthanols wurde der Rückstand in 500 ml heissem Acetonitril aufgenommen. Beim Erkalten schieden sich 35 g farblose Kristalle ab, die bei 80° Celsius im Vakuum getrocknet wurden.

Beispiel 2
Methoxyacetaldehyd-Erythromycylamin-Kondensationsprodukt
2,3 g (0,03 Mol) Erythromycylamin und 1,2 g (0,01 Mol) Methoxyacetaldehyddimethylacetal wurden in 20 ml Dioxan und 2 ml Wasser mit 12 g Dowex 50 W bei Raumtemperatur gerührt. Nach 6 Stunden wurde der Ionenaustauscher abfiltriert und mit Dioxan gewaschen. Das Filtrat wurde eingedampft und der Rückstand säulenchromatographisch gereinigt (Aluminiumoxid basisch, Aktivitätsstufe 3, Chloroform/Methanol = 40:1). Die Fraktion mit Rf = 0,6 wurde eingedampft und durch Behandeln mit Äther/Petroläther zur Kristallisation gebracht. Man erhielt 0,8 g (33 % der Theorie) farblose Kristalle vom Schmelzpunkt 191° C.

Beispiel 3
Methylthioacetaldehyd-Erythromycylamin-Kondensationsprodukt
1,3 g (0,008 Mol) Methylthioacetaldehyd-diäthylacetal werden in einem Gemisch von 20 ml Dioxan und 4 ml Wasser mit 4,0 g Amberlite IR-120 (H-Form) 15 Minuten bei Raumtemperatur gerührt. Anschliessend gibt man eine Lösung von 3,0 g (0,004 Mol) Erythromycylamin in 10 ml Dioxan zu und rührt das Reaktionsgemisch weitere 4 Stunden bei derselben Temperatur.
Der Ionenaustauscher wird abfiltriert und mit Dioxan gewaschen. Das Filtrat wird eingedampft, der ölige Rückstand in Äther aufgenommen, die Lösung filtriert und mit Petroläther bis zur beginnenden Kristallisation versetzt.
Das weisse, feinkristalline Produkt wird abgesaugt, mit einem Äther/Petroläther-Gemisch gewaschen und getrocknet. Ausbeute: 2,5 g (80 °/o der Theorie)
Schmelzpunkt: 100—105° C (Zers.).

Beispiel 4
Carbäthoxymethylthioacetaldehyd-Erythromycylamin-Kondensationsprodukt
Eine Emulsion von 13 g (0,055 Mol) Carbäthoxymethyl-thioacetaldehyd-diäthylacetal in 130 ml Wasser wird mit 55 g Amberlite IR 120 (H-Form) versetzt und 4 Stunden bei Raumtemperatur kräftig gerührt.
Die klare Lösung wird vom Ionenaustauscher abfiltriert, mit Kochsalz gesättigt und 4mal mit je 100 ml Äther extrahiert. Nach Trocknen über Natriumsulfat wird das Lösungsmittel abdestilliert. Der verbleibende Aldehyd ist chromatographisch rein und wird roh weiter umgesetzt.
Ausbeute: 7,6 g (85 % der Theorie)
5,5 g (0,0075 Mol) Erythromycylamin und 7,5 g (0,045 Mol) Carbäthoxymethylthioacetaldehyd werden in 50 ml Methanol p. a. gelöst und 24 Stunden bei Raumtemperatur gerührt. Nach Abdestillieren des Lösungsmittels im Vakuum verbleibt ein dunkelbraunes Öl, aus dem durch Behandeln mit einem Chloroform/Äther-Gemisch dunkelgefärbte Nebenprodukte abgetrennt werden. Die Lösung wird eingedampft, der hellbraune flüssige Rückstand in Chloroform aufgenommen, mit Aktivkohle entfärbt und filtriert. Das Filtrat versetzt man bis zur beginnenden Kristallisation mit Äther.
Die ausgefallene, kristalline Substanz wird abgenutscht, mit Äther gewaschen und getrocknet.
Ausbeute: 6,2 g (64 % der Theorie),
Schmelzpunkt: 150—155° C (Zers.).

Beispiel 5
Acetylglykolaldehyd-Erythromycylamin-Kondensationsprodukt
2,3 g (0,003 Mol) Erythromycylamin und 1,1 g (0,006 Mol) Acetylglykolaldehyd-diäthylacetal werden in 20 ml Dioxan und 2 ml Wasser mit 12 g Dowex 50 W bei Raum-, temperatur gerührt. Nach 6 Stunden wird der Ionenaustauscher abfiltriert und mit Dioxan gewaschen. Das Filtrat wird eingedampft und der Rückstand säulenchromatographisch ge reinigt (Aluminiumoxid basisch, Aktivitätsstufe 3, Chloroform/Methanol = 4:1). Die Fraktion mit Rf = 0,6 wird eingedampft und durch Behandeln mit Chloroform/Äther zur Kristallisation gebracht. Man erhält 2,4 g farblose Kristalle vom Schmelzpunkt 105—110° C.

2,3 g (0,003 Mol) Erythromycylamin und 1,0 g (0,006 Mol) Benzoylglykolaldehyd werden in 60 ml absolutem Dioxan 24 Stunden bei Raumtemperatur gerührt. Nach Verdampfen des Dioxans wird der Rückstand in Chloroform aufgenommen und mit Äther versetzt. Es scheiden sich 0,8 g 15 farblose Kristalle ab, die bei 20° C im Vakuum getrocknet werden.

Beispiel 7
Diäthylphosphonoacetaldehyd-Erythromycylamin-Kondensationsprodukt
2,3 g (0,003 Mol) Erythromycylamin und 1,4 g (0,006 Mol) Diäthylphosphonoacetaldehyd-dimethylacetal werden in 20 ml Dioxan und 2 ml Wasser mit 12 g Dowex 50 W bei Raumtemperatur gerührt. Nach 6 Stunden wird der Ionenaustauscher abfiltriert und mit Dioxan gewaschen. Das Filtrat wird mit Wasser bis zur beginnenden Kristallisation versetzt. Das weisse Produkt wird abgesaugt, mit wässrigem Dioxan gewaschen und getrocknet.
Ausbeute: 1,4 g (52 % der Theorie).
Schmelzpunkt: 110—113° C (Zers.).

Beispiel 8
Diäthylphosphonoacetaldehyd-Erythromycylamin-Kondensationsprodukt
23 g (0,03 Mol) Erythromycylamin und 10 g (0,055 Mol) Diäthylphosphonoacetaldehyd werden in 300 ml absolutem Dioxan 24 Stunden bei Raumtemperatur gerührt. Nach Verdampfen des Lösungsmittels wird der Rückstand in 150 ml heissem Acetonitril aufgenommen und die Lösung mit 750 ml Wasser versetzt. Beim Erkalten scheiden sich 13 g (43 % der Theorie) farblose Kristalle ab, die bei 80° C im Vakuum getrocknet werden.
F.: 110—113° C (Zers.).
Die Verbindungen der Formeln Ia und Ib lassen sich in an sich bekannter Weise in die üblichen pharmazeutischen Zubereitungsformen, zum Beispiel in Lösungen, Supposito-rien, Tabletten, einarbeiten. Die Einzeldosis beträgt für Erwachsene bei peroraler Applikation 50 bis 500 mg, die bevorzugte Einzeldosis 100 bis 250 mg, die Tagesdosis 0,5 bis 4 g, die bevorzugte Tagesdosis 1 bis 2 g.

3. Verfahren nach Patentanspruch 1 oder 2, dadurch gekennzeichnet, dass man in einer Ausgangsverbindung der Formel III, sofern R eine freie oder monosubstituierte Aminogruppe bedeutet, diese intermediär schützt.
