Description
La présente invention a pour objet un procédé de préparation de nouveaux composés actifs contre l'hypertension en relation avec l'angiotensine chez des espèces de mammifères, ou un de leurs sels. Ces composés sont de formule suivante:

Les produits de formule VI peuvent être utilisés pour la préparation des dérivés mercapto de formule:
(VII) IjL3 R2 Ri
HS—CH2 —CH—CO—N—CH—COOH
par ammonolyse.
Dans les formule ci-dessus, les symboles Ri, R2, R3 et Rô ont la même signification que précédemment. Dans ces symboles, les groupes alcoyle inférieurs sont des groupes hydrocarbure à chaîne droite ou ramifiée ayant jusqu'à 7 atomes de carbone tels que les groupes méthyle, éthyle, propyle, isopropyle, butyle, isobutyle t-butyle, pentyle, isopentyle, hexyle, heptyle, etc. Les groupes alcoyle inférieurs en C1-C4, et notamment les groupes alcoyle inférieurs en Ci ou C2 sont préférés. Les groupes phénylalcoyle inférieurs comprennent de tels groupes alcoyle inférieurs auxquels est fixé un groupe phényle. On préfère particulièrement les groupes benzyle et phénéthyle et surtout le groupe benzyle.
Les composés représentés par les formules VI et VII qui comprennent la structure des aminoacides qui sont l'alanine, la leucine, la phénylalanine, l'arginine, la sarcosine, la sérine, l'asparagine, la glutamine, l'histidine, le tryptophane, la cystéine, la méthionine, la thréonine, la lysine, la tyrosine, la leucine, la valine ou l'acide aspartique sont préférés, et notamment l'arginine, l'alanine, la phénylalanine et la leucine. De préférence, R5 est un atome d'hydrogène.
L'étude expérimentale indique que la Na-(3-mercapto-2-méthylpropanoyl)-L-arginine, la N"-(3-mercaptopropanoyl)-L-arginine, la N-(3-mercaptopropanoyl)-L-phénylalanine et la N-(3-mercaptopropanoyl)-L-leucine sont les membres les plus puissants du groupe des composés de formule VII, et on les préfère particulièrement.
Les composés de formules VI et VII forment aussi des sels avec diverses bases inorganique et organique, dont la préparation entre aussi dans la portée de l'invention. Ces sels comprennent des sels d'ammonium, des sels de métal alcalin tels que les sels de sodium et de potassium, des sels de métal alcalino-terreux tels que les sels de calcium et de magnésium, des sels avec des bases organiques, par exemple des sels de dicyclohexylamine, de benzathine, d'hydra-bamine et de N-méthyl-D-glucamine.
Les composés de formules VI et VII ont un atome de carbone asymétrique ou même deux si R3 est autre que l'hydrogène. Ces atomes de carbone sont indiqués par un astérisque dans la formule VI. Ainsi, les composés existent dans des formes diastéréo-isomères ou en leurs mélanges racémiques. La préparation de toutes ces formes entre dans la portée de l'invention.
On a trouvé que, pour atteindre un degré important d'activité biologique, l'atome de carbone asymétrique portant le reste Ri doit avoir la configuration L—, une stéréospécificité qui n'est pas apparente dans les applications rapportées antérieurement des mercaptoalcylaminoacides. Ainsi, ce sont ceux qu'on préfère.
L'inhibition de l'enzyme convertissant l'angiotensine au moyen de composés de formules VI et VII peut être mesurée in vitro avec l'enzyme convertissant l'angiotensine isolée du poumon du lapin après avoir procédé comme il est décrit par Cushman et Cheung, «Biochem. Pharmacol.», 20,1637 (1971), et avec un essai de muscle lisse excisé [E. O'Keefe et al., «Fédération Proc.» 31, 511 (1972)] où on a montré que ces composés sont des agents inhibiteurs puissants de l'activité contractile de l'angiotensine I et des agents de potentialisation de l'activité contractile de la brady-kinine.
En administrant une composition contenant un agent inhibiteur de l'enzyme convertissant l'angiotensine ou une association de ces agents ou un de leurs sels physiologiquement acceptables au mammifère hypertendu atteint d'hypertension qui dépend de l'angiotensine, cela gêne la séquence:
rénine-*angiotensine I->angiotensine II, et la condition d'hypertension est réduite ou soulagée.
Une dose unique, ou de préférence deux à quatre doses quotidiennes partagées, données à raison d'environ 1 à 1000 mg/kg/j, avantageusement d'environ 10 à 500 mg/kg/j et de préférence de 30 à 300 mg/kg/j conviennent pour réduire la pression sanguine élevée associée à l'angiotensine. Les expériences sur des modèles animaux décrites par S.L. Engel, T.R. Schaefer, M. H. Waugh et R. Rubin, «Proc. Soc. Exp. Biol. Med.» 143, 483 (1973) sont un guide utile. De préférence, on administre le composé ou la composition par voie orale, mais on peut aussi utiliser des voies paren-térales telles que la voie sous-cutanée, intramusculaire, intraveineuse ou intrapéritonéale.
Les exemples ci-après illustrent l'invention. Les températures sont en degrés centigrades.
Exemple 1 :
N*- (3-acétylthio-2-méthylpropanoyl j -L-arginine
On dissout 2,61 g de L-arginine dans un mélange de 3,2 g de carbonate de sodium et 30 ml d'eau et on réfrigère la solution dans un bain de glace. On ajoute 2,7 g de chlorure de 3-acétylthio-2-méthylpropanoyle et on agite le mélange réactionnel à la température ordinaire pendant 1,5 h. On ajoute 50 ml d'une résine échan-geuse d'ions (AG 50 W), puis on applique la suspension à une colonne de 80 ml de la même résine. Après lavage à l'eau, on élue la Na-(3-acétylthio-2-méthylpropanoyl)-L-arginine avec un tampon à la pyridine/acide acétique de pH = 6,5, on sépare le solvant sous vide, on dissout le résidu dans le méthanol et on le précipite avec l'éther pour obtenir 3,86 g de Na-(3-acétylthio-2-méthyl-propanoyl)-L-arginine qui fondent à 133° C.
Exemple 2:
Na- (3-mercapto-2-méthylpropanoyl) -L-arginine.
On dissout 1 g de Na-(3-acétylthio-2-méthylpropanoyl)-L-arginine dans un mélange de 5 ml d'eau et 5 ml d'ammoniaque concentré. Après 1 h à la température ordinaire, on concentre la solution jusqu'à un volume de 3 ml sous vide (sans chauffage) et on ajoute une résine échangeuse d'ions (AG-50 W) jusqu'à un pH d'approximativement 4. On applique la suspension à une colonne de la même résine et on élue la Na-(3-mercapto-2-méthylpropa-noyl)-L-arginine avec un tampon de pyridine/acétate de pH = 6,5. On sépare le solvant sous vide et on lyophylise le résidu pour obtenir 0,86 g qui fondent à 100°C.
Exemple 3:
N-(3-acétylthio-2-méthylpropanoyl)-L-valine.
On dissout 88 g de L-valine et 40 g de carbonate de sodium dans 11 d'eau et on réfrigère la solution dans un bain de glace en agitant vigoureusement. On ajoute 135 g de chlorure de 3-acétylthio-2-méthylpropanoyl et 120 g d'une solution de carbonate de sodium dans 500 ml d'eau en 5 portions égales au cours de 15 mn. Après 1,5 h, on extrait le mélange réactionnel à l'acétate d'éthyle, on

acidifie la phase aqueuse avec de l'acide chlorhydrique concentré et on l'extrait à l'acétate d'éthyle. On lave la phase organique avec de l'eau, on la sèche sur du sulfate de magnésium et on la concentre à sec pour obtenir 190 g de N-(3-acétylthio-2-méthylpropanoyl)-L-valine.
Exemple 4:
N-(3-mercapto-2-méthylpropanoyl)-L-valine.
On dissout 20 g de N-(3-acétylthio-2-méthylpropanoyl)-L-valine dans un mélange de 46 ml d'eau et 31 ml d'ammoniaque concentré. Après 1 h à la température ordinaire, on extrait le mélange réactionnel à l'acétate d'éthyle, on acidifie la phase aqueuse avec de l'acide chlorohydrique concentré et on l'extrait à l'acétate d'éthyle. On sèche la phase organique sur du sulfate de magnésium et on la concentre à sec sous vide pour obtenir 19 g de N-(3-mercapto-2-méthylpropanoyl)-L-valine.
Exemple 5:
N-(3-mercapto-2-méthylpropanoyl)-L-phénylalanine.
En substituant la L-phénylalanine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la N-(3-acétylthio-2-méthylpropanoyl)-L-phénylalanine et la N-(3-mercapto-2-méthylpropanoyl)-L-phényl-alanine.
Exemple 6:
N-(3-mercapto-2-méthylpropanoyl)-L-thréonine.
En substituant la L-thréonine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la N-(3-acétylthio-2-méthylpropanoyl)-L-thréo-nine et la N-(3-mercapto-2-méthylpropanoyl)-L-thréonine.
Exemple 7:
N-(3-mercapto-2-méthylpropanoyl)-L-glutamine.
En substituant la L-glutamine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la N-(3-acétylthio-2-méthylpropanoyl)-L-gluta-mine et la N-(3-mercapto-2-méthylpropanoyl)-L-glutamine.
Exemple 8:
Na-(3-mercapto-2-méthylpropanoyl)-L-lysine.
En substituant la Ne-t-butyloxycarbonyl-L-lysine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la Na-(3-acétylthio-2-méthyl-propanoyl)-N£-t-butyloxycarbonyl-L-lysine et la Na-(3-mercapto-2-méthylpropanoyl)-NE-t-butyloxycarbonyl-L-lysine. En traitant ces produits avec l'acide trifluoroacétique à la température ordinaire pendant 15 mn, on obtient la Na-(3-acétylthio-2-méthyl-propanoyl)-L-lysine et la N2-(3-mercapto-2-méthylpropanoyl)-L-lysine.
Exemple 9:
N- (3-mercapto-2-méthylpropanoyl) -L-ty rosine.
En substituant la L-tyrosine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la N-(3-acétylthio-2-méthylpropanoyl)-L-tyrosine et la N-(3-mercapto-2-méthylpropanoyl)-L-tyrosine.
Exemple 10:
N- (3-mercapto-2-méthylpropanoyl)-L-tryptophane.
En substituant le L-tryptophane à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient le N-(3-acétylthio-2-méthylpropanoyl)-L-tryptophane et le N-(3-mercapto-2-méthylpropanoyl)-L-trypto-phane.
Exemple 11 :
N- (3-mercapto-2-méthylpropanoyl)-L-méthionine.
En substituant la L-méthionine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la N-(3-acétylthio-2-méthylpropanoyl)-L-mé-thionine et la N-(3-mercapto-2-méthylpropanoyl)-L-méthionine.
Exemple 12:
N-(3-mercapto-2-méthylpropanoyl)-N-méthyl-L-phénylalanine. En substituant la N-méthyl-L-phénylalanine à la L-valine dans le procédé de l'exemple 3, puis en traitant le produit selon le procédé de l'exemple 4, on obtient la N-(3-acétylthio-2-méthyl-propanoyl)-N-méthyl-L-phénylalanine.
Exemple 13:
Chlorure d'acide 3-acétylthio-2-benzylpropanoïque.
On mélange 8,1 g d'acide 2-benzylacrylique et 5,3 g d'acide thiolacétique et on chauffe au bain-marie pendant 1 h. Après avoir refroidi jusqu'à la température ordinaire, on ajoute 9,75 g de chlorure de thionyle et on garde le mélange pendant une nuit à la température ordinaire. On sépare sous vide l'excès de chlorure de thionyle et on distille le résidu pour obtenir le chlorure d'acide 3-acétylthio-2-benzylpropanoïque qui bout (0,05) à 120-122° C.
Exemple 14:
N*-(3-acétylthio-2-benzylpropanoyl)-L-arginine.
En substituant le chlorure d'acide 3-acétylthio-2-benzylpropa-noïque au chlorure d'acide 3-acétylthio-2-méthylpropanoïque dans le procédé de l'exemple 1, on obtient la Na-(3-acétylthio-2-benzyl-propanoyl)-L-arginine qui fond à 253-295° C.
Exemple 15:
jV*- (3-mercapto-2-benzylpropanoyl)-L-arginine.
En substituant la Na-(3-acétylthio-2-benzylpropanoyl)-L-arginine à la Na-(3-acétylthio-2-méthylpropanoyl)-L-arginine dans le procédé de l'exemple 2, on obtient la Na-(3mercapto-2-benzylpropanoyl)-L-arginine qui fond à 135°C.
Exemple 16:
N- (3-mercaptopropanoyl) -L-alanine.
On dissout 4,2 g de N-(3-benzoylthiopropanoyl)-L-alanine dans un mélange de 7,5 ml d'eau et 6 ml d'hydroxyde d'ammonium concentré. Après 1 h, on dilue le mélange avec de l'eau, on le filtre et on extrait le filtrat à l'acétate d'éthyle. On acidifie la phase aqueuse avec de l'acide chlorhydrique concentré et on l'extrait à l'acétate d'éthyle. On lave la phase organique avec de l'eau, on la sèche et on la concentre à sec sous vide. On cristallise le résidu à partir d'acétate d'éthylhexane pour obtenir 1,87 g de N-(3-mer-captopropanoyl)-L-alanine qui fond à 79-81°C.
Exemple 17:
N-(3-mercaptopropanoyl)-L-leucine.
En substituant 6,46 g de N-(3-benzoylthiopropanoyl)-L-leucine à la N-(3-benzoylthiopropanoyl)-L-alanine dans le procédé de l'exemple 16, on obtient 2,75 g de N-(3-mercaptopropanoyl)-L-leucine qui fondent à 131-132° C. On recristallise cette substance à partir de l'acétonitrile.
Exemple 18:
N- ( 3-mercaptopropanoyl)-L-phénylalanine.
On dissout 1,78 g de N-(3-benzoylthiopropanoyl)-L-phényl-alanine dans un mélange de 20 ml d'eau et de 5,5 ml d'hydroxyde de sodium N. On ajoute à cette solution 20 ml d'hydroxyde d'ammonium concentré, puis 20 ml d'eau. Après 3 h, on extrait le mélange réactionnel avec l'acétate d'éthyle, on acidifie avec de

l'acide chlorhydrique concentré, puis on extrait de nouveau à l'acétate d'éthyle. On lave le deuxième extrait d'acétate d'éthyle à l'eau, on le sèche sur du sulfate de magnésium et on le concentre à sec sous vide. On Chromatographie le résidu sur une colonne de Silicagel avec un mélange de benzène/acide acétique pour obtenir 0,47 g de N-(3-mercaptopropanoyl)-L-phénylalanine qui fond à 106-107° C.
Exemple 19:
N(3-mercaptopropanoyl)-L-arginine.
On dissout 1 g de Na-(3-benzoylthiopropanoyl)-L-arginine en agitant vigoureusement dans un mélange de 5 ml d'eau et 5 ml d'ammoniaque concentré. Après 1 h, on extrait la solution à l'acétate d'éthyle et on la concentre à sec sous vide. On Chromatographie le résidu sur une colonne de 85 ml de DEAE Sephadex [résine échangeuse d'anions dérivée du destran (Mikes, supra, p. 328)] avec un tampon de bicarbonate d'ammonium 0,005M. On réunit les fractions contenant la Na-(3-mercaptopropanoyl)-L-arginine (comme indiqué par une réaction positive du thiol et la réaction de Sakaguchi) et on les lyophylise pour enlever le bicarbonate d'ammonium; on obtient 200 mg qui fondent à 230°C (la décomposition commence à 200° C).
Exemple 20:
N- (3-mercaptopropanoyl) sarcosine.
En substituant 2,8 g de N-(3-benzoylthiopropanoyl)-sarcosine à la N-(3-benzoylthiopropanoyl)-L-alanine dans le procédé de l'exemple 16, on obtient 1,65 g de N-(3-mercaptopropanoyl)sar-cosine brute. On convertit cette substance en son sel de dicyclo-hexylammonium (2,7 g) qui fond à 157-158° C, puis on convertit le sel purifié en l'acide libre par répartition entre l'acétate d'éthyle et du bisulfate de potassium à 10%.
Exemple 21 :
N- (3-mercaptopropanoyl)-L-sérine.
En substituant la L-sérine à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthyl-propanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 16, on obtient la N-(3-ben-zoylthiopropanoyl)-L-sérine et la N-(3-mercaptopropanoyl)-L-sérine.
Exemple 22:
N- (3-mercaptopropanoyl)-L-asparagine.
En substituant la L-asparagine à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthyl-propanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 2, on obtient la N-(3-benzoyl-thiopropanoyl)-L-asparagine et la N-(3-mercaptopropanoyl)-L-asparagine.
Exemple 23:
N1- ( 3-mercaptopropanoylj -L-lysine.
En substituant la NE-tert-butyloxycarbonyl-L-lysine [R. Schwy-zer et W. Rittel, «Helv. Chim. Acta», 44,159 (1961)] à la L-argine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétyl-thio-2-méthylpropanoyle dans le procédé selon l'exemple 1, puis en traitant le produit obtenu par le procédé selon l'exemple 16, on obtient la Na-(3-benzoylthiopropanoyl)-NE-t-butyloxycarbonyl-L-lysine et la Na-(3-mercaptopropanoyl)-NE-t-butyloxycarbonyl-L-lysine. En traitant cette substance avec l'acide trifluoroacétique à la température ordinaire pendant 15 mn, on obtient la Na-(3-benzoylthiopropanoyl)-L-lysine et la Na-(3-mercaptopropanoyl)-L-lysine.
Exemple 24:
N®- (3-mercaptopropanoyl)-L-histidine.
En substituant la L-histidine à L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthyl-propanoyle dans le procédé de l'exemple 1, puis en traitant le produit par le procédé de l'exemple 16, on obtient la Na-(3-ben-zoylthiopropanoyl)-L-histidine et la Na-(3-mercaptopropanoyl)-L-histidine.
Exemple 25:
N- (3-mercaptopropanoyl)-L-tryptophane.
En substituant le L-tryptophane à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthyl-propanoyle dans le procédé de l'exemple 1, puis en traitant le produit par le procédé de l'exemple 16, on obtient le N-(3-benzoyl-thiopropanoyl)-L-tryptophane et le N-(3-mercaptopropanoyl)-L-tryptophane.
Exemple 26:
N- (3-mercaptopropanoyl)-L-cystèine.
En substituant la L-cystéine à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthyl-propanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 16, on obtient la N-(3-ben-zoylthiopropanoyl)-L-cystéine et la N-(3-mercaptopropanoyl)-L-cystéine.
Exemple 27:
N- (3-mercaptopropanoyl) -L-méthionine.
En substituant la L-méthionine à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthyl-propanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 16, on obtient la N-(3-ben-zoylthiopropanoyl)-L-méthionine et la N-(3-mercaptopropanoyl)-L-méthionine.

N- ( 3-mercaptopropanoyl) -N-méthyl-L-leucine.
En substituant la N-méthyl-L-leucine à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthylpropanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 16, on obtient la N-(3-benzoylthiopropanoyl)-N-méthyl-L-leucine et la N-(3-mer-captopropanoyl)-N-méthyl-L-leucine.

N- ( 3-mercaptopropanoyl) -N-benzyl-L-leucine.
En substituant la N-benzyl-L-leucine à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-méthylpropanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 16, on obtient la N-(3-acétylthiopropanoyl)-N-benzyl-L-leucine et la N-(3-mercapto-propanoyl)-N-benzyl-L-leucine.
Exemple 30:
Acide N-(3-mercaptopropanoyl)-L-aspartique.
En substituant l'acide L-aspartique à la L-arginine et le chlorure de 3-benzoylthiopropanoyle au chlorure de 3-acétylthio-2-méthylpropanoyle dans le procédé de l'exemple 1, puis en traitant le produit selon le procédé de l'exemple 16, on obtient l'acide N-(3-benzoylthiopropanoyl)-L-aspartique et l'acide N-(3-mer-captopropanoyl)-L-aspartique.

Bien que la conversion de l'angiotensine I en angiotensine II par une enzyme convertissant l'angiotensine soit probablement la

réaction la plus importante dans la pathologie de l'hypertension, on dose de manière plus commode et plus précise l'activité de l'enzyme isolée en mesurant la vitesse de clivage sur un substrat d'un peptide plus simple, c'est-à-dire l'hippuryl-L-histidyl-L-leucine. Pour déterminer les valeurs de I50 (concentrations des s composés exprimées en microgrammes/millilitres produisant une inhibition de 50% de l'enzyme convertissant l'angiotensine), on ajoute diverses concentrations de chaque composé à des tubes de 13 x 100 mm en même temps que les composants suivants aux concentrations finales indiquées dans un volume final de 0,25 ml : 10 100 mM de tampon au phosphate de potassium, pH=8,3 ; 300 mM de chlorure de sodium; 5 mM d'hippuryl-L-histidyl-L-leucine et 5 milliunités d'enzyme convertissant l'angiotensine de poumon de lapin. On prépare de manière analogue des témoins ne contenant pas d'inhibiteur (activité = 100%) et on les acidifie avant d'ajouter 15 l'enzyme (activité=0%). Toutes les réactions enzymatiques sont commencées en ajoutant le composant qui est l'enzyme; les tubes sont incubés pendant 30 mn à 37° C et les réactions sont terminées en ajoutant 0,25 ml d'HCl IN. L'acide hippurique formé par l'action de l'enzyme convertissant l'angiotensine sur l'hippuryl-L- 20 histidyl-L-leucine est extrait dans l'acétate d'éthyle, évaporé à sec, redissous dans l'eau, puis dosé d'après son absorbance à 228 nm. Le pourcentage d'inhibition par chaque concentration de composé est calculé par comparaison avec les témoins ayant une activité de 0% et de 100%. Les concentrations des composés selon la 25 présente invention qui inhibent l'activité de l'enzyme convertissant l'angiotensine de 50% sont indiquées dans le tableau suivant:

Le procédé pour évaluer l'inhibition de l'enzyme convertissant l'angiotensine I dans l'iléon excisé d'un cobaye est conduit comme suit: on ajoute diverses concentrations du composé à un bain d'iléon excisé du cobaye dans une solution Krebs à 37° C, on aère avec un mélange de 95% O2 et 5% CO2. Après 2 mn, on ajoute l'angiotensine I (25 mg/ml) et on mesure les contractions isotoniques. Claims (7)
1. Procédé de préparation d'un nouveau composé de formule:
(VI) R3 R2 Ri RtìCO—S—CH2 —CH—CO—N—CH—COOH
ou un de ses sels, dans laquelle Ri est un groupe alcoyle inférieur, phénylalcoyle inférieur, hydroxyphénylalcoyle inférieur, amino-alcoyle inférieur, guanidinoalcoyle inférieur, imidazolylalcoyle inférieur, indolylalcoyle inférieur, mercaptoalcoyle inférieur, alcoyle inférieur-mercaptoalcoyle inférieur, carbamoylalcoyle inférieur ou carboxyalcoyle inférieur; R2 et R3 sont chacun un atome d'hydrogène, un groupe alcoyle inférieur ou phénylalcoyle inférieur, et Rß est un groupe alcoyle inférieur ou phényle, caractérisé en ce qu'on fait réagir un composé de formule:
(IV) R3 R6-CO-S-CH2-CH-COOH
dans laquelle Rô a la signification ci-dessus avec un aminoacide de formule:
(V) R2 RI
HN-CH-COOH
2. Procédé selon la revendication 1, caractérisé en ce que Ri est un groupe guanidinoalcoyle inférieur, aminoalcoyle inférieur, phénylalcoyle inférieur ou alcoyle inférieur; R2 est un atome d'hydrogène et R3 est un atome d'hydrogène, un groupe alcoyle inférieur ou phénylalcoyle inférieur.

REVENDICATIONS
3. Procédé selon la revendication 1, caractérisé en ce que Ri est un groupe guanidinopropyle, aminopropyle, aminobutyle, phényl-méthyle ou isobutyle et R2, R3 et Rô ont chacun la signification indiquée dans la revendication 1.
4. Procédé selon la revendication 1, caractérisé en ce que Ri est un groupe phénylméthyle et R3 est un atome d'hydrogène.
5. Procédé selon la revendication 1, caractérisé en ce que Ri est un groupe isobutyle et R3 est un atome d'hydrogène.
6. Procédé selon la revendication 1, caractérisé en ce que Ri est un groupe guanidinopropyle et R3 est un atome d'hydrogène.
7. Utilisation des composés de formule VI définis dans la revendication 1 pour la préparation des composés de formule:
(VII) R3 R2 Ri
HS—CH2-CH—CO—N—CH—COOH par ammonolyse.
L'angiotensine II est un agent vasoconstricteur puissant qu'on a impliqué comme agent causal principal dans l'étiologie de l'hypertension rénovasculaire.
L'angiotensine II est formée à partir de l'angiotensine I par l'action d'une enzyme convertissant l'angiotensine. L'angiotensine I est un décapeptide biologiquement inerte scindé de l'angio-tensinogène, qui est une protéine du sang, par l'action de la résine qui est une enzyme [Oparil et al., «New England J. of Med.» 291, 389-457 (1974]. L'angiotensinogène et la rénine sont aussi inertes biologiquement.
L'enzyme qui convertit l'angiotensine est aussi responsable de l'inactivation de la bradykinine qui est un agent vasodilatateur qu'on a impliqué dans la régulation de la fonction rénale [Erdos, «Circulation Research», 36, 247 (1975)].
Des agents qui inhibent l'enzyme convertissant l'angiotensine peuvent donc contrecarrer l'effet hypertenseur de l'angiotensine I
du moment que cela n'est dû qu'à sa conversion en angiotensine II. On peut utiliser ces agents thérapeutiquement pour le traitement de formes d'hypertension rénovasculaire et maligne ainsi que pour d'autres formes d'hypertension qui dépendant de l'angiotensine [Gavras et al., «New England J. of Med.», 291, 817 (1974)].
Selon Oparil et al. ci-dessus, l'angiotensine II a un rôle majeur pour maintenir l'homéostase circulatoire chez l'animal appauvri en sodium mais, chez l'animal normal qui absorbe une quantité normale de sel, l'angiotensine II n'est pas nécessaire pour le maintien aigu de la pression sanguine. Dans une variété d'états qui soumettent à une contrainte (stress) le système rénine/angiotensine, l'administration aiguë d'un inhibiteur de l'enzyme convertissant l'angiotensine ou un agent bloquant l'angiotensine II abaisse la pression sanguine et cause une augmentation de l'activité de la rénine du plasma.
Certains mercaptoacylaminoacides sont divulgués dans la littérature. Le brevet US N° 3246025 montre des dérivés de mer-captopropionylglycine qui sont utiles pour renforcer la fonction du foie comme antidote pour des poisons tels que le mercure et des composés d'organo-arsenic. (Voir aussi le brevet allemand N° 2349707.) Le brevet US N° 3897480 présente des N-(a-mer-captoacyl)aminoacides utiles pour la prophylaxie et la thérapie pour le traitement d'un désordre métabolique tel que la noso-toxicose due à un métal lourd, un désordre d'irradiation, le diabète ou l'hépatite. Le brevet US N° 3857951 montre l'utilisation de la 2-mercaptopropionylglycine et de ses sels de métal alcalin pour le traitement de maladies respiratoires.
On a rapporté que la 2-mercaptopropionylglycine, connue pour être un agent protecteur du foie, produisait un abaissement de la pression sanguine lors d'une injection intraveineuse à des rats à tension normale anesthésiés [Schulze, «Arzneim. Forsch.», 22, 1433 (1972)], modèle peu sûr [Schwarts, «Methods in Pharma-cology», vol. 1, 125 (1971); Berger, «Selected Pharmacological Testing Methods», vol. 3,171, 194 (1968); tandis que d'autres n'ont pas rapporté d'effets observables sur la pression sanguine, etc. (Fijimura et al., «Nippon Yakurigaku Zasshi», 60, 278-92 (1964)]. Voir aussi Ripa, «Proc. Int. Symp. Thiola», Osaka, Japon, 1970, pp. 226-230, qui ont rapporté que, chez les rats à tension normale, l'a-mercaptopropionylglycine augmente le taux en angio-tensinogène et abaisse le taux de rénine dans le sang par un mécanisme homéostatique de rétroaction.
On a maintenant trouvé que les mercaptoacylaminoacides répondant aux formules VI et VII ci-après sont des inhibiteurs de l'enzyme convertissant l'angiotensine et que, lorsqu'on les administre à des espèces de mammifères souffrant d'hypertension, ils réduisent ou soulagent l'hypertension en relation avec l'angiotensine.
