Verfahren zur Herstellung eines neuen Benzol-sulfonamidderivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man ein 2-Benzolsul- fonamido-5-diäthylmethyl-thiodiazol-(1,3,4), das in p-Stellung einen durch Hydrolyse in die Amiuogruppeüberführbaren Substituenten aufweist, mit einem hydrolysierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsul- f onamido)-5-diäthylmethyl-thiodiazol-(1,3,4) schmilzt bei 230 . Die neue Verbindung soll als Arzneimittel sowie als Zwischenprodukt Verwendung finden.
Das 2 - Benzolsulfonamido - 5 - diäthyl - methyl-thiodiazol-(1,3,4), das in p-Stellung zur Sulfonamidgruppe einen durch Hydro lyse in die Aminogruppe überführbaren Sub- stituenten enthält, kann auf verschiedene Art und Weise gewonnen werden.
Besonders geeignet ist die Umsetzung der entsprechen den reaktionsfähigen Benzolsulfonsäurederi- vate, insbesondere der Benzolsulfonsäure- halogenide mit 5-Diäthylmethyl-thiodiazol- (1,3,4)-verbindungen, die in 2-Stellung eine Gruppe enthalten, die mit dem Benzolsulfon- säurederivat ein 2-Benzolsulfonamido-5-di- äthylmethyl-thiodiazol-(1,3,4) zu bilden ver mag,
wie mit 2-Amino-5-diäthylmethyl-thio- diazol-(1,3,4). Man kann auch entsprechende Sulfonamide der Formel R S02NH Y, in der Y einen bei der nächstfolgenden Reaktion sich abspaltenden Rest bedeutet mit 2-Halo- gen-5-diäthylmethyl-thiodiazolen-(1,3,4) um setzen. Auch können andere dem Fach mann geläufige Herstellungsmethoden be nützt werden.
<I>Beispiel Z:</I> 26,4 g p-Carbäthogyamino-benzolsulfo- cblorid, 17,1 g 2-Amino-5-diäthylmethyl- thiodiazol-(1,3,4) (F. 168 C; hergestellt durch Einwirkung von Diäthylessigsäure- chlorid auf Thiosemicarbazid) werden in 50 cm' Pyridin eine Stunde am RückfluB- kühler auf dem Wasserbad erhitzt.
Nach dem Erkalten wird mit Äther im ÜberSChuss ge- fällt, das ausgefällte Ueaktionsprodukt zwei mal mit Äther angerieben und nach dem Ab dekantieren des Äthers werden 400 cm' Wasser hinzugefügt. Die auskristallisierende Carbäthogyverbindung vom F. 209 C wird durch Verseifung mit 2n-Natronlauge in das 2 - (p-Amino - benzolsulfonamido) - 5 - diäthyl- methyl-thiodiazol-(1,3,4) übergeführt.
Es schmilzt nach dem Umkristallisieren aus Al kohol bei 230 C.
Das gebildete p-Amino-benzolsulfonamid- derivat lässt sich auch in Form seiner Salze, z. B. des Natriums oder des Calciums, isolieren.
<I>Beispiel 2:</I> 17,1 g 2-Amino-5 -diäthylmethyl-thio- diazol-(1,3,4) werden in 25 cm3 Pyridin auf genommen und unter Rühren allmählich mit 23,5 g p-Acetylamino-benzolsulfochlorid ver setzt. Hierauf wird noch eine Stunde auf 95 erhitzt. Durch Eingiessen der erhaltenen Lösung in Eiswasser wird das Kondensa tionsprodukt in fester Form erhalten. Es wird abgesogen und durch Kochen in ver dünnter Natronlauge verseift.
Das mit Essigsäure ausgefällte 2-(p-Amino-benzol- sulf onamido) - 5 - diäthylmethyl - thiodiazol - (1,3,4) hat nach dem Umkristallisieren aus wässerigem Alkohol den F. 230 .
Die Hydrolyse kann man auch mit an dern alkalischen Mitteln, beispielsweise Erd- alkalihydrogyden, wie Calciumhydrogyd oder mit sauren Mitteln, z. B. Salzsäure, in Ge genwart von Wasser oder organischen Lö sungsmitteln, wie Alkohol, durchführen.
Process for the production of a new benzene sulfonamide derivative. The p-amino-benzenesulfonamide derivative formed can also be used in the form of its salts, e.g. B. of sodium or calcium isolate.
<I> Example 2: </I> 17.1 g of 2-amino-5-diethylmethyl-thio-diazole- (1,3,4) are taken up in 25 cm3 of pyridine and gradually increased with 23.5 g of p -Acetylamino-benzenesulfochloride ver sets. It is then heated to 95 for another hour. The condensation product is obtained in solid form by pouring the resulting solution into ice water. It is sucked off and saponified by boiling in dilute sodium hydroxide solution.
