Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man ein 2-Benzolsulfon- amido - 5 - diäthylmethyl - thiodiazol - (1,3,4), das in p-Stellung einen durch Reduktion in die Aminogruppe überführbaren Substituen- ten aufweist, mit einem reduzierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsulfon- amido) - 5 - diäthylmethyl - thiodiazol- (1,3,4) schmilzt bei 230 . Die neue Verbindung soll als Arzneimittel sowie als Zwischenprodukt Verwendung finden.
Das 2 - Benzolsulfonamido - 5 - diäthyl - methyl-thiodiazol-(1,3,4), das in p-Stellung zur Sulfonamidgruppe einen durch Reduk tion in die Aminogruppe überführbaren Sub- stituenten enthält, kann auf verschiedene Art und Weise gewonnen werden.
Besonders ge eignet ist die Umsetzung der entsprechenden reaktionsfähigen Benzolsulfonsäurederivate, insbesondere der Benzolsulfonsäurehalogenide mit 5-Diäthylmethyl-thiodiazol-(1,3,4)-ver- bindungen, die in 2-Stellung eine Gruppe enthalten, die mit dem Benzolsulfonsäure- derivat ein 2-Benzolsulfonamido-5-diäthyl- methyl-thiodiazol-(1,3,4) zu bilden vermag, insbesondere mit 2-Amino-5-diäthylmethyl- thiodiazol-(1,3,4)
. Man kann aber auch die entsprechenden Sulfonamide der Formel RSO,NHY, in der Y einen bei der nachfol genden Reaktion sich abspaltenden Rest be deutet, mit 2-Halogen-5-diäthylmethyl-thio- diazolen-(1,3,4) umsetzen oder andere, dem Fachmann geläufige Herstellungsmethoden benutzen.
<I>Beispiel</I> In eine Suspension von<B>17,1</B> g 2-Amino- 5-diäthylmethyl-thiodiazol-(1,3,4) in 45 cm' trockenem Pyridin werden 22,2 g p-Nitro- benzolsulfochlorid eingetragen. Nach ein stündigem Erwärmen der Mischung auf dem Dampfbad wird die Nitroverbindung mit Wasser ausgefällt und durch Umkristallisie- r en aus Alkohol gereinigt. Die Reduktion zur Aminoverbindung erfolgt mit Zinnchlorür in konzentrierter Salzsäure.
Nach dem Um- kristallisieren aus Äthylalkohol schmilzt das ?\ - (p -Amino - benzolsulfonamido) - 5 - diäthyl- methyl-thiodiazol-(1,3,4) bei 230 .
Das gebildete p-Aminobenzolsulfonamid- derivat lässt sich auch in Form seiner Salze, z. B. des Natriums oder des Calciums, isolieren.
Beispiel <I>2:</I> 17,1 g \? - Amino - 5 - diäthylmethyl - tliio- diazol-(1,3,4) werden in 25 ein' Pyridin mit 22 g p-Nitro-benzolsulfochlorid kondensiert. Die Reaktion wird beendigt durch einstiin- diges Erwärmen auf dem Wasserbad.
Das Kondensationsprodukt wird durch Eingiessen in Eiswasser ausgefällt und abgenutscht. Es wird hierauf unter Rühren in ein siedendes Gemisch von 20 g Eisenspänen, 300 cm' :50%igem Alkohol und 1 cm' konzentrierter Salzsäure eingetragen und mehrere Stunden im Sieden gehalten. Der Alkohol wird ab destilliert und das Reaktionsgemisch mit Natronlauge alkalisiert und heiss filtriert.. Aus dem Filtrat wird mit Essigsäure das - (p -Amino - benzolsulf onamido) - 5 - diäthyl- niethyl-thiodiazol-(1,3,4) kristallin gefällt.
Aus wässerigem Alkohol umkristallisiert ist es ein weisses Pulver vom F. 230".
Die Reduktion kann auch mit Ferro- hydroxyd in Gegenwart von Natrium- oder Calciumhydroxyd und Wasser durchgeführt werden, wobei sich die Natrium- bezw. Caleiumverbindung des Benzolsulfonamids bildet.

The condensation product is precipitated by pouring into ice water and suction filtered. The alcohol is distilled off and the reaction mixture is made alkaline with sodium hydroxide solution and filtered hot. From the filtrate, the - (p -amino-benzenesulfonamido) -5-diethylniethyl-thiodiazole (1,3,4) is precipitated in crystalline form with acetic acid .
Recrystallized from aqueous alcohol it is a white powder of 230 ".
The reduction can also be carried out with ferro hydroxide in the presence of sodium or calcium hydroxide and water, the sodium or Forms calcium compound of benzene sulfonamide.
