Verfahren zur Herstellung eines neuen Benzol-sulfonamidderivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man ein 2-Benzolsulfon- amido-5-methyl-thiodiazol-(1,3,4), das in p- Stellung einen durch Hydrolyse in die Amino- gruppe überführbaren Substituenten aufweist, mit einem hydrolysierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsulfon- amido)-5-methyl-thiodiazol-(1,3,4) schmilzt bei 202 . Die neue Verbindung soll als Arznei mittel sowie als Zwischenprodukt Verwen dung finden.
Das 2-Benzolsulfonamido-5-methyl-thiodi- azol-(1,3,4), das in p-Stellung zur Sulfon- amidgruppe einen durch Hydrolyse in die Aminogruppe überführbaren Substituenten enthält, kann auf verschiedene Art und Weise gewonnen werden.
Besonders geeignet ist die Umsetzung der entsprechenden reaktionsfähi gen Benzolsulfonsäurederivate, insbesondere der Benzolsulfonsäurehalogenide mit 5-Methyl- thiodiazol-(1,3,4)-verbindungen, die in 2-Stel- lung eine Gruppe enthalten, die mit dem Benzolsulfonsäurederivat ein 2-Benzolsulfon- amido-5-methyl-thiodiazol-(1,3,4) zu bilden vermag, wie mit 2-Amino-5-methyl-thiodiazol- (1,3,4)
. Man kann auch entsprechende Sulfon amide der Formel R # SO2NHY, in der Y einen bei der nachfolgenden Reaktion sich abspaltenden Rest bedeutet, mit 2-Halogen- 5-methyl-thiodiazolen-(1,3,4) umsetzen. Auch können andere, dem Fachmann geläufige Herstellungsmethoden benützt werden.
Beispiel <I>1:</I> In eine Suspension von 46 g 2-Amino-5- methyl-thiodiazol-(1,3,4) in etwa 300 cm' trockenem Pyridin werden 94,4 g reines p- Acetylamino - benzolsulfochlorid eingetragen, wobei unter Erwärmung alles in Lösung geht, und anschliessend wird 1 Stunde am Rückfluss- kühler auf dem Wasserbad erhitzt.
Nach dem Erkalten wird mit Äther im Überschuss gefällt, das ausgefällte Reaktionsprodukt zwei mal mit Äther angerieben, und nach dem Abdekantieren des Äthers werden 400 cm3 Wasser hinzugefügt, wobei zunächst der Niederschlag in Lösung geht, dann aber nach etwa 5 Minuten aus der Lösung das 2-(p- Acetylamino - benzolsulfonamido) - 5 - inethyl - thiodiazol-(1,3,4) auskristallisiert.
Die so er haltene Acetylaniinoverbindung wird mit etwa 600 cm' 2n-Salzsäure so lange gekocht, bis alles in Lösung gegangen ist, die Lösung mit einer gesättigten Natriumacetatlösuug bis pH 6,8 versetzt, wobei das 2-(p-Amino-benzol- sulfonamido)-5-methyl-thiodiazol-(1,3,4) kri stallin ausfällt. Ausbeute 80-90 g. F. 202 (aus 96"/o Alkohol).
Das gebildete p- Amino-benzolsulfonamid- derivat lässt sich auch in Form seiner Salze, zum Beispiel des Natriums oder des Calciunis, isolieren.
Beispiel <I>2:</I> 11,5 g 2-Amino-5-methyl-thiodiazol-(1,3,4) werden in 25 cm' Pyridin aufgenommen und unter Rühren allmählich mit 23,5 g p-Acetyl- amino-benzolsulfochlorid versetzt. Hierauf wird noch eine halbe Stunde auf 95' erhitzt. Durch Eingiessen der erhaltenen Lösung in Eiswasser wird das Kondensationsprodukt in fester Form erhalten. Es wird abgesogen und durch Kochen in verdünnter Natronlauge verseift.
Das mit Essigsäure ausgefällte 2- (p -Amino-benzolsulfonamido)-5-methyl-thiodi- azol-(1,3,4) hat nach dem 1Jmkristallisieren aus wässerigem Alkohol den F. 202 .
Die Hydrolyse kann man auch mit andern alkalisehen Mitteln, beispielsweise Erdalkali- hy droxyden, wie Calciumhydroxyd, oder mit sauren Mitteln, zum Beispiel Salzsäure, in Gegenwart von Wasser, oder organischen Lösungsmitteln, wie Alkohol, durchführen.
Process for the production of a new benzene sulfonamide derivative. The reaction of the corresponding reactive benzenesulfonic acid derivatives, in particular the benzenesulfonic acid halides, with 5-methylthiodiazole (1,3,4) compounds which contain a group in the 2-position which, with the benzenesulfonic acid derivative, is a 2-benzenesulfone, is particularly suitable - Amido-5-methyl-thiodiazole- (1,3,4) is able to form, as with 2-amino-5-methyl-thiodiazole- (1,3,4)
. Corresponding sulfonamides of the formula R # SO2NHY, in which Y is a radical which is split off in the subsequent reaction, can also be reacted with 2-halo-5-methyl-thiodiazolen- (1,3,4). Other production methods familiar to the person skilled in the art can also be used.
Example <I> 1: </I> In a suspension of 46 g of 2-amino-5-methyl-thiodiazole- (1,3,4) in about 300 cm 'dry pyridine, 94.4 g of pure p-acetylamino - Benzenesulfochloride added, everything dissolving when heated, and then heated for 1 hour on the reflux condenser on the water bath.
After cooling, an excess of ether is used to precipitate, the precipitated reaction product is rubbed twice with ether, and after decanting off the ether, 400 cm3 of water are added, whereby the precipitate first dissolves, but then after about 5 minutes the 2 - (p-Acetylamino-benzenesulfonamido) -5-ynethyl-thiodiazole- (1,3,4) crystallized out.
The acetylaniino compound thus obtained is boiled with about 600 cm 'of 2N hydrochloric acid until everything has dissolved, and a saturated sodium acetate solution to pH 6.8 is added to the solution, the 2- (p-amino-benzenesulfonamido ) -5-methyl-thiodiazole- (1,3,4) crystalline precipitates. Yield 80-90g. F. 202 (from 96 "/ o alcohol).
The p-amino-benzenesulfonamide derivative formed can also be isolated in the form of its salts, for example of sodium or calcium.
Example <I> 2: </I> 11.5 g of 2-amino-5-methyl-thiodiazole- (1,3,4) are taken up in 25 cm 'pyridine and gradually added to 23.5 g of p-acetyl with stirring - Amino-benzenesulfochloride added. This is then heated to 95 'for another half hour. The condensation product is obtained in solid form by pouring the resulting solution into ice water. It is suctioned off and saponified by boiling in dilute sodium hydroxide solution.
The 2- (p -amino-benzenesulphonamido) -5-methyl-thiodiazole- (1,3,4) precipitated with acetic acid has a melting point of 202 after crystallization from aqueous alcohol.
The hydrolysis can also be carried out with other alkaline agents, for example alkaline earth metal hydroxides, such as calcium hydroxide, or with acidic agents, for example hydrochloric acid, in the presence of water or organic solvents such as alcohol.
