Verfahren zur Herstellung eines neuen Benzol-sulfonamidderivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man 2-Benzolsulfon- amido-5-isobutyl-thiodiazol-(1,3,4), das in p Stellung einen durch Hydrolyse in die Amino- gruppe überführbaren Substituenten aufweist, mit einem hydrolysierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsulfon- amido)-5-isobutyl-thiodiazol-(1,3,4) schmilzt bei 225 . Die neue Verbindung soll als Arznei mittel sowie als Zwischenprodukt Verwen dung finden.
Das 2-Benzolsulfonamido-5-isobutyl-thio- diazol-(1,3,4), das in p-Stellung zur Sulfon- amidgruppe einen durch Hydrolyse in die A minogruppe überführbaren Substituenten enthält, kann auf verschiedene Art und Weise gewonnen werden.
Besonders geeignet ist die Umsetzung der entsprechenden reaktionsfähi gen Benzolsulfonsäurederivate, insbesondere der Benzolsulfonsäurehalogenide mit 5-Iso- butyl-thiodiazol-(1,3,4)-verbindungen, die in 2-Stellung eine Gruppe enthalten, die mit dem Benzolsulfonsäurederivat ein 2-Benzol- sulfonamido-5-isobutyl-thiodiazol-(1,3,4) zu bilden vermag, wie mit 2-Amino-5-isobutyl- thiodiazol-(1,3,4)
. Man kann auch entspre chende Sulfonamide der Formel R SO2NH Y, in der Y einen bei der nachfolgenden Reak tion sich abspaltenden Rest bedeutet, mit 2-Halogen-5-isobutyl-thiodiazolen-(1,3,4) um setzen. Auch können andere, dem Fachmann geläufige Herstellungsmethoden benützt wer den.
<I>Beispiel 1:</I> 26,4 g p-Carbäthogyamino-benzolsulfo- chlorid und 15,7 g 2-Amino-5-isobutyl-thiodi- azol-(1,3,4) (F. 234-235 ; erhalten durch Einwirkung von iso-Valerylchlorid auf Thio- semicarbazid) werden in 50 eins Pyridin eine Stunde am Rückflusskühler auf dem Wasser bad erhitzt.
Nach dem Erkalten wird mit Äther im Überschuss gefällt, das ausgefällte Reaktionsprodukt zweimal mit Äther ange rieben und nach dem Abdekantieren des Äthers werden 400 cm' Wasser hinzugefügt. Die auskristallisierende Carbäthoxyverbindung schmilzt nach Umkristallisieren aus Alkohol bei 175" C. Das daraus durch Verseifung mit 2n-Natronlauge erhaltene 2-(p-Amino-berrzol- sulfonamido)-5-isobutyl-thiodiazol-(1,3,4) hat nach dem Umlösen aus Alkohol den F. 225".
Das gebildete p-Amino-benzolsulfonamid- derivat lässt sich auch in Form seiner Salze, zum Beispiel des Natriums oder des Calciums, isolieren.
<I>Beispiel 2:</I> 15,7g 2-Amino-5-isobutyl-thiodiazol-(1,3,4) werden in 25 cm' Pyridin aufgenommen und unter Rühren allmählich mit 23,5 g p-Acetyl- amino - benzolsulfäehlorid versetzt. Hierauf wird noch eine Stunde auf 95" erhitzt. Durch Eingiessen der erhaltenen Lösung in Eis wasser wird das Kondensationsprodukt in fester Form erhalten. Es wird äbgesogen und durch Kochen in verdünnter Natronlauge verseift.
Das mit Essigsäure ausgefällte 2- (p -Amino - benzolsulfonamido)- 5 -isobuty 1-thio- diazol-(1,3,4) hat nach dem Umkristallisieren aus wässerigem Alkohol den F. 225".
Die Hydrolyse kann man auch mit andern alkalischen Mitteln, beispielsweise Erdalkali- 15 hydroxyden, wie Calciumhydroxyd, oder mit sauren Mitteln, zum Beispiel Salzsäure in Gegenwart von Wasser, oder organischen Lö sungsmitteln, wie Alkohol, durchführen.

The hydrolysis can also be carried out with other alkaline agents, for example alkaline earth hydroxides, such as calcium hydroxide, or with acidic agents, for example hydrochloric acid in the presence of water, or organic solvents such as alcohol.
