Verfahren zur Herstellung eines neuen Benzol-sulfonamidderivates.- Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man ein 2-Benzolsul- fonamido-5-n-butyl-thiodiazol-(1,3,4), das in p-Stellung einen durch Hydrolyse in die Aminogruppe überfiihrbaren Substituenten aufweist, mit einem hydrolysierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsulfon- amido)-5-n-butyl-thiodiazol-(1,3,4) schmilzt bei 186 . Die neue Verbindung soll als Arz neimittel sowie als Zwischenprodukt Ver wendung finden.
Das 2-Benzolsulfonamido-5-n-butyl-thio- diazol-(1,3,4), das in p-Stellung zur Sulfon- amidgruppe einen durch Hydrolyse in die Aminogruppe überführbaren Substituenten enthält, kann auf verschiedene Art und Weise gewonnen werden.
Besonders geeignet ist die Umsetzung der entsprechenden reak tionsfähigen Benzolsulfonsäurederivate, ins besondere der Benzolsulfonsäurehalogenide, mit 5-n-Butyl-thiodiazol-(1,3,4)-verbindun- gen, die in 2-Stellung eine Gruppe enthalten, die mit dem Benzolsulfonsäurederivat ein 2 - Benzolsulfonamido - 5 - n-butyl-thiodiazol- (1,3,4) zu bilden vermag,
wie mit 2-Amino- 5 - n - butyl - thiodiazol - (1,ä,4). Man kann auch entsprechende Sulfonamide der Formel RSO,ZNHY, in der Y einen bei der nachfol genden Reaktion sich abspaltenden Rest be deutet, mit 2-Halogen-5-n-butyl-thiodiazolen- (1,3,4) umsetzen. Auch können andere dem Fachmann geläufige Herstellungsmethoden benutzt werden.
<I>Beispiel 1:</I> 26,4 g p - Carbäthogyamino - benzolsulfo- chlorid werden mit 15,7 g 2-Amino-5-n- butyl-thiodiazol-(1,3,4) (F. 193,5 C; her gestellt durch Einwirkung von n-Valeryl- chlorid auf Thiosemicarbazid) und 50 cm' Pyridin am Rückflusskühler eine Stunde auf dem Wasserbad erhitzt.
Nach dem Erkalten wird mit Äther im Überschuss gefällt, das ausgefällte Reaktionsprodukt zweimal mit Äther angerieben., und nach dem Abdekantie- ren des Äthers werden 400 cm-" Wasser hinzugefügt.
Die auskristallisierende Carb- äthoxyverbindung vom F. 198,7 liefert nach Verseifung mit zweifach normaler Natron lauge das 2-(p-Amino-benzolsulfonamido)-5- n-butyl-thiodiazol-(1,3,4). Das durch zwei maliges Umkristallisieren aus Alkohol ge reinigte Produkt schmilzt bei 186 C.
Das gebildete p-Aminobenzolsulfonamid- derivat lässt sich auch in Form seiner Salze, z. B. des Natriums oder des Caleiums, isolieren.
Beispiel <I>2:</I> 15,7 g 2-Amino - 5--n-butyl-thiodiazol- (1,3,4) werden in 25 cm"> Pyridin aufgenom men und unter Rühren allmählich mit 23,5 g p - Acetylamino - benzolsulfochlorid versetzt. Hierauf wird noch eine Stunde auf 95 er hitzt. Durch Eingiessen der erhaltenen Lö sung in Eiswasser wird das Kondensations- produkt in fester Form erhalten. Es wird ab gesogen und durch Kochen in verdünnter Natronlauge verseift.
Das mit Essigsäure ausgefällte 2-(p-Amino-benzolsulfonamido)- 5-n-buty 1-thiodiazol-t.1,3,4) hat nach dem Umkristallisieren aus wässerigem Alkohol den F. 186 .
Die Hydrolyse kann man auch mit an dern alkalischen Mitteln, beispielsweise Erd- allr.a.lihydroxyden, wie Calciumhydroxyd oder mit sauren Mitteln, z. B. Salzsäure, in Ge genwart von Wasser oder organischen Lö sungsmitteln, wie Alkohol, durchführen.
Process for the preparation of a new benzene sulfonamide derivative.- The subject of the present patent is a process for the preparation of a new benzene sulfonamide derivative, which is characterized in that a 2-benzenesulfonamido-5-n-butyl-thiodiazole- (1,3 , 4), which has a substituent which can be converted into the amino group by hydrolysis, is treated with a hydrolyzing agent in the p-position.
The 2- (p-amino-benzenesulfonamido) -5-n-butyl-thiodiazole- (1,3,4) obtained in this way melts at 186. The new compound is to be used as a medicinal product and as an intermediate product.
The 2-benzenesulfonamido-5-n-butyl-thiodiazole- (1,3,4), which contains a substituent which can be converted into the amino group by hydrolysis in the p-position to the sulfonamide group, can be obtained in various ways .
The reaction of the corresponding reactive benzenesulfonic acid derivatives, in particular the benzenesulfonic acid halides, with 5-n-butyl-thiodiazole- (1,3,4) compounds which contain a group in the 2-position which corresponds to the benzenesulfonic acid derivative is particularly suitable a 2 - benzenesulfonamido - 5 - n-butyl-thiodiazole- (1,3,4) is able to form,
as with 2-amino-5 - n - butyl - thiodiazole - (1, ä, 4). Corresponding sulfonamides of the formula RSO, ZNHY, in which Y denotes a radical which is split off in the subsequent reaction, can also be reacted with 2-halo-5-n-butyl-thiodiazolen- (1,3,4). <I> Example 1: </I> 26.4 gp - Carbäthogyamino - benzenesulfochloride are mixed with 15.7 g of 2-amino-5-n-butyl-thiodiazole- (1,3,4) (F. 193, 5 C; produced by the action of n-valeryl chloride on thiosemicarbazide) and 50 cm 'pyridine heated in a reflux condenser for one hour on a water bath.
After cooling, it is precipitated with ether in excess, the precipitated reaction product is rubbed twice with ether, and after the ether has been decanted off, 400 cm water is added.
The carbethoxy compound which crystallizes out, having a melting point of 198.7, provides 2- (p-amino-benzenesulfonamido) -5-n-butyl-thiodiazole- (1,3,4) after saponification with twice normal sodium hydroxide solution. The product, which has been purified by recrystallizing twice from alcohol, melts at 186 C.
The p-aminobenzenesulfonamide derivative formed can also be used in the form of its salts, e.g. B. of sodium or calcium, isolate.
Example <I> 2: </I> 15.7 g of 2-amino - 5 - n-butyl-thiodiazole- (1,3,4) are taken up in 25 cm "> of pyridine and gradually added to 23, while stirring 5 gp-acetylamino-benzenesulphonyl chloride are then added, and the mixture is then heated for a further hour to 95. The condensation product is obtained in solid form by pouring the resulting solution into ice-water.
2- (p-Amino-benzenesulfonamido) -5-n-buty-1-thiodiazol-t.1,3,4) precipitated with acetic acid has a melting point of 186 after recrystallization from aqueous alcohol.
