Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates. Gegenstand des vorliegenden Patentes ist ein Verfahren zur Herstellung eines neuen Benzolsulfonamidderivates, das dadurch ge kennzeichnet ist, dass man ein 2-Benzolsulfon- amido-4-methyl-pyrimidin, das in p-Stellung einen durch Reduktion in die Aminogruppe überführbaren Substituenten aufweist, mit einem reduzierenden Mittel behandelt.
Das so erhaltene 2-(p-Amino-benzolsulfon- amido)-4-methyl-pyrimidin schmilzt bei 234 . Die neue Verbindung soll als Arzneimittel sowie als Zwischenprodukt Verwendung finden.
Das 2-Benzolsulfonamido-4-methyl-pyri- midin, das in p-Stellung zur Sulfonamid- gruppe einen durch Reduktion in die Amino- gruppe überführbaren Substituenten enthält, kann auf verschiedene Art und Weise ge wonnen werden.
Besonders geeignet ist die Umsetzung der entsprechenden reaktions fähigen Benzolsulfonsäurederivate, insbeson dere der Benzolsulfonsäurehalogenide mit 4-3vIethylpyrimidinverbindungen, die in 2-Stel- lung eine Gruppe enthalten, die mit dem Benzolsulfonsäurederivat ein 2-Benzolsulfon- amido-4-methyl-pyrimidin zu bilden vermag, insbesondere 2 - Amino - 4 - methy 1- pyrimid in.
Man kann auch entsprechende Sulfonamide der Formel RSO=NHY, in der Y einen bei der nachfolgenden Reaktion sich abspalten den Rest bedeutet, mit 2-Halogen-4-methyl- pyrimidinen umsetzen oder andere, dem Fachmann geläufige Herstellungsmethoden benutzen.
<I>Beispiel 1:</I> In eine Suspension von 10,9 g 2-Amiuo- 4-methyl-pyrimidin in der vierfachen Menge Pyridin werden 22,2 g p-Nitro-benzolsulfo- chlorid eingetragen. Die Reaktionsmischung wird 24 Stunden stehen gelassen und sodann noch eine Stunde auf dem Wasserbad erhitzt.
Nach Beendigung der Umsetzung wird das Pyridin im Vakuum abdestilliert. Nach dem Abkühlen wird der Rückstand mit Wasser versetzt, wobei das gebildete 2-(p=Nitro- benzolsulfonamido)-4-methy 1-pyriinidin a;i:s- fällt.
29,7 g 2-(p-Nitro-benzolsulfonamido)-4- methyl-pyrimidin werden in der 50fachen 3lenge absolutem Alkohol gelöst und mit 10 Gewichtsprozent eines Nickel-Träger-Ka- talysators im Rührautoklaven mit Wasser stoff unter einem Überdruck von 50 Atm. bis zur Beendigung der Wasserstoffaufnahme bei 100-125 behandelt. Nach dem Erkalten wird vom Katalysator abfiltriert und die alkoholische Lösung eingedampft.
Dabei fällt das gebildete 2-(p-Amino-benzolsulfon- aniido)-4-metliyl-pyrimidin kristallin an. Die Verbindung kann durch Reinigung mit ver dünntem Alkohol, gegebenenfalls unter Zu satz von Tierkohle, umkristallisiert werden und fällt dann in Form farbloser Kristalle an. Schmelzpunkt 23.1 .
Das gebildete p-Amino-benzolsulfonamid- derivat lässt sich auch in Form seiner Salze. z. B. des Natriums oder des Ca.leiums, isolieren.
<I>Beispiel 2:</I> <B>10,9</B> g ? Amino-4-methyl-pyrimidin wer den wie in Beispiel 1 mit 24,2 g 4-Nitro- benzolsulfochlorid umgesetzt, worauf das gebildete 2- (p-Nitro - l)enzolsitlfonaniido) -4- niethvl-pyrimidin isoliert wird.
<B>29,7</B> g der Nitroverbindung werden mittels<B>67,7</B> - Zinnchlorür (Hydrat) und der doppelten G ewiehtsmenge konzentrierter Salzsäure reduziert. Das gebildete 2-(p- Amino-benzolsulfonamido) -4-methyl-pyrimi- din wird isoliert und gegebenenfalls um kristallisiert.
<I>Beispiel 3:</I> 109 g 2-Amino-4-methyl-py rimidin wer den in 250 cm'"' trockenem Pvridin bei 45 bis 55' mit 222 g p-Nitro-benzolsulfoclilorid unter Rühren versetzt. Man erwärmt die Mi schung noch 1 Stunde auf 55-60 und ver setzt mit verdünnter Salzsäure. Das abge schiedene 2-(p-Nitro-benzolsulfonamido)-4- methyl-pyrimidin kann durch Lösen in 2n Sodalösung, Filtrieren und Fällen mit Salz säure gereinigt werden.
In die kochende Reduktionsmischung von 20 g Eisenpulver, 10 cm3 2n-Salzsäure und 350 cm' 50%igem Alkohol trägt man nach und nach 29,4 g 2-(p-Nitro-benzolsulfori- amido)-4-methyl-pyrimidin ein.
Nach vier stündigem Kochen am Rückfluss wird der Alkohol abdestilliert. Nun gibt man 100 ein' 2n-Natronlauge zu, erwärmt zum Sieden, nutscht ab und fällt aus dem Filtrat mit ver dünnter Essigsäure das 2-(p-Amino-benzol- sulfona,mido)-4-methyl-pyriinidin kristallin aus. Zur Reinigung kristallisiert man unter Benutzung von Tierkohle aus verdünntem Alkohol um. F. 234 .
Die Reduktion kann auch mit Ferro- hydroxyd in Gegenwart von Natrium- oder Caleiumhydroxyd und Wasser durchgeführt werden, wobei sich die Natrium- bezw. Caleiumverbindung des Benzolsulfonamids bildet.

<I> Example 2: </I> <B> 10.9 </B> g? Amino-4-methyl-pyrimidine is reacted with 24.2 g of 4-nitrobenzenesulfochloride as in Example 1, whereupon the 2- (p-nitro-1) enzolsitlfonaniido) -4-niethvl-pyrimidine is isolated.
