Abstract

"DERIVADOS DE AMIDA E SEU USO COMO INIBIDORES DE 11-BETA-HIDROXIESTERóIDE DESIDROGENASE TIPO 1". A presente invenção refere-se a compostos de fórmula (I) que proporcionam agentes farmacológicos que reduzem concentrações intracelulares de glicocorticóide em mamíferos, em particular, níveis intracelulares de cortisol em humanos. Portanto, os compostos da presente invenção aperfeiçoam sensibilidade à insulina no músculo e no tecido adiposo, e reduz lipólise e produção livre de ácidos graxos no tecido adiposo. Os compostos da invenção reduzem concentração hepática de glicocorticóide em mamíferos, em particular, concentração hepática de cortisol em humanos, resultando na inibição de gliconeogênese hepática e redução de níveis plasmáticos de glicose. Desse modo, os compostos da presente invenção poderão ser particularmente úteis em mamíferos como agentes hipoglicêmicos para o tratamento e prevenção de condições em que hiperglicemia e/ou resistência à insulina são envolvidas, tais como diabetes tipo 2. Os compostos da invenção poderão ser também usados para tratar outros distúrbios associados a glicocorticóide, tais como Síndrome X, dislipidemia, hipertensão e obesidade central. A invenção além disso refere-se ao uso dos compostos de acordo com a invenção para a preparação de medicamentos, em particular de medicamentos úteis para o tratamento e prevenção de distúrbios associados a glicocorticóide, aperfeiçoando sensibilidade à insulina, reduzindo níveis plasmáticos de glicose, reduzindo lipólise e produção livre de ácidos graxos, e diminuindo formação de tecido adiposo visceral.
