Abstract
ENRIQUECIMENTO DE CLASSIFICAÇÃO DE CÉLULAS ATIVADAS POR FLUORESCÊNCIA (FACS) PARA GERAR PLANTAS. A invenção descreve uma Plataforma de Integração de Transgenes Manipulados Geneticamente (ETIP) que pode ser inserida randomicamente ou em locais alvo nos genomas de plantas para facilitar a rápida seleção e detecção de um GOI que é perfeitamente direcionado (em ambas as extremidades 3' e 5') para o local genômico na ETIP. Um elemento da invenção é a introdução de rupturas de filamento duplo específicas na ETIP. Em algumas modalidades, a invenção descreve uma ETIP usando sítios fixadores da nuclease dedos-de-zinco, mas pode utilizar outras tecnologias de vetorização tais como meganucleases, TALs, CRISPRs, ou zíperes de leucina. Também estão descritas composições de plantas transgênicas, e método para produzir plantas transgênicas, onde o DNA doador ou de "payload" expressa um ou mais produtos de uma sequência de ácidos nucleicos exógena (por exemplo, proteína ou RNA) que fora estavelmente integrada em uma ETIP em uma célula vegetal. Nas modalidades, a ETIP facilita a testagem de genes candidatos e vetores de expressão vegetais da invenção através das fases de desenvolvimento.
