Abstract
【課題】本発明の目的は、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上を含む外用組成物において、経時的に生じる褐変を抑制できる製剤技術を提供することである。
【解決手段】外用医薬組成物において、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上と共に、モノテルペンを含有させることにより、経時的に生じる褐変を効果的に抑制できる。
Description
本発明は、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上を含み、経時的に生じる褐変を抑制できる外用組成物に関する。
ロキソプロフェンナトリウム、ジクロフェナクナトリウム、インドメタシン等に代表される非ステロイド性抗炎症薬は、外用医薬組成物の有効成分として広く使用されている。また、ノナン酸バニリルアミドは、温感付与、血行促進等の効能が知られており、外用医薬組成物の有効成分として広く使用されている。更に、ニコチン酸ベンジルエステルは、吸収促進、血行促進等の効能が知られており、外用医薬組成物の有効成分として広く使用されている。
従来、非ステロイド性抗炎症薬、ノナン酸バニリルアミド、ニコチン酸ベンジルエステル等を有効成分として配合した外用組成物の処方が種々提案されている。例えば、特許文献1には、ロキソプロフェン0.5~5重量%、ノナン酸バニリルアミド、及び水を含有する外用消炎鎮痛剤組成物は、ロキソプロフェンの消炎鎮痛作用を向上させ得ることが記載されている。また、特許文献2には、インドメタシンと、ノナン酸バニリルアミド及び/又はニコチン酸ベンジルエステルとを含む外用剤は、インドメタシンの保存安定性を向上させ得ることが記載されている。
本発明者は、非ステロイド性抗炎症薬、ノナン酸バニリルアミド、及びニコチン酸ベンジルエステルを含む外用組成物において、ノナン酸バニリルアミド及びニコチン酸ベンジルエステルの効能を効果的に発揮させるべく、これら両成分の含有量を高めた処方について検討を進めた。その結果、非ステロイド性抗炎症薬と共に、ノナン酸バニリルアミド0.015重量%以上を共存させると、経時的な褐変が生じることを知得した。更に、非ステロイド性抗炎症薬と共に、ノナン酸バニリルアミド0.015重量%以上及びニコチン酸ベンジルエステル0.01重量%以上を共存させると、経時的に生じる褐変が顕著になることを知得した。
そこで、本発明の目的は、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上を含む外用組成物において、経時的に生じる褐変を抑制できる製剤技術を提供することである。
本発明者は、前記課題を解決すべく鋭意検討を行ったところ、外用医薬組成物において、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上と共に、モノテルペンを含有させることにより、経時的に生じる褐変を効果的に抑制できることを見出した。発明は、かかる知見に基づいて、更に検討を重ねることにより完成したものである。
即ち、本発明は、下記に掲げる態様の発明を提供する。
項1. 非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、ニコチン酸ベンジルエステル0.01重量%以上、及びモノテルペンを含有する、外用組成物。
項2. 前記非ステロイド性抗炎症薬が、ロキソプロフェン、ジクロフェナク、及びこれらの塩よりなる群から選択される少なくとも1種である、項1に記載の外用組成物。
項3. 前記モノテルペンが、メントールである、項1又は2に記載の外用組成物。
項4. 液剤、クリーム剤、ローション剤、ジェル剤、又は乳液剤である、項1~3のいずれかに記載の外用組成物。

本発明の外用組成物は、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上を含んでいながらも、経時的に生じる褐変を効果的に抑制でき、優れた製剤安定性を備え、保存中に含有成分を安定に保持し、良好な外観形状を維持することができる。
1.外用組成物
本発明の外用組成物は、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、ニコチン酸ベンジルエステル0.01重量%以上、及びモノテルペンを含有することを特徴とする。以下、本発明の外用組成物について詳述する。

[非ステロイド性抗炎症薬]
本発明の外用組成物は、非ステロイド性抗炎症薬を含有する。非ステロイド性抗炎症薬とは、グルココルチコイド以外の抗炎症薬の総称である。

本発明で使用される非ステロイド性抗炎症薬の種類については、特に制限されないが、例えば、ロキソプロフェン、ジクロフェナク、インドメタシン、フェルビナク、サリチル酸、アセチルサリチル酸、アスピリン、サルサラート、サリチルアミド、エテンザミド、イブプロフェン、ケトプロフェン、ナプロキセン、フルルビプロフェン、ピロキシカム、チアプロフェン酸、スプロフェン、トルメチン、アルミノプロフェン、ベノキサプロフェン、ベンジダミン、スリンダク、アセメタシン、プログルメタシン、アンフェナク、モフェゾラク、ロルノキシカム、アンピロキシカム、フェノプロフェン、チアプロフェン酸、オキサプロジン、メフェナム酸、フルフェナム酸、アザプロパゾン、フェンブフェン、エトドラク、ロフェコキシブ、ジフルニサル、ゾメピラク、セレコキシブ、ザルトプロフェン、ケトロラック、ニメスリド、アセクロフェナクが挙げられる。
また、非ステロイド性抗炎症薬は、塩の形態をとり得る場合には、塩の形態であってもよい。塩の形態の非ステロイド性抗炎症薬としては、例えば、ロキソプロフェンの塩、ジクロフェナクの塩等が挙げられる。
ロキソプロフェンの塩としては、具体的には、ナトリウム塩、カリウム塩等のアルカリ金属塩;カルシウム塩等のアルカリ土類金属塩等が挙げられる。これらの中でも、好ましくはアルカリ金属塩、更に好ましくはナトリウム塩が挙げられる。また、ロキソプロフェンの塩は、水和物であってもよい。
また、ジクロフェナクの塩としては、具体的には、ナトリウム塩、カリウム塩等のアルカリ金属塩;カルシウム塩等のアルカリ土類金属塩;アンモニアとの塩;ジメチルアミン、ジエチルアミン、トリメチルアミン、トリエチルアミン等の第1級、第2級若しくは第3級のアルキルアミンとの塩等が挙げられる。これらの中でも、好ましくはアルカリ金属塩、更に好ましくはナトリウム塩が挙げられる。
これらの非ステロイド性抗炎症薬は、1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。
これらの非ステロイド性抗炎症薬の中でも、好ましくは、ロキソプロフェン、ジクロフェナク、及びこれらの塩が挙げられる。とりわけ、非ステロイド性抗炎症薬として、ロキソプロフェン及びその塩を使用した場合には、経時的に生じる褐変をより一層効果的に抑制することが可能になる。
本発明の外用医薬組成物における非ステロイド性抗炎症薬の含有量は、使用する非ステロイド性抗炎症薬の種類、備えさせるべき薬効等に応じて適宜設定すればよいが、例えば0.1~15重量%、好ましくは0.5~10重量%、より好ましくは0.5~5重量%、更に好ましくは1~1.5重量%が挙げられる。
[ノナン酸バニリルアミド]
本発明の外用組成物は、ノナン酸バニリルアミドを0.015重量%以上含有する。ノナン酸バニリルアミドは、カプサイシノイドの一種であり、温感付与、血行促進等の効能が知られている公知の成分である。

本発明の外用組成物におけるノナン酸バニリルアミドの含有量は、0.015重量%以上であればよいが、具体的には、0.015~0.3重量%、好ましくは0.015~0.15重量%、より好ましくは0.02~0.15重量%、更に好ましくは0.05~0.1重量%が挙げられる。非ステロイド性抗炎症薬及びニコチン酸ベンジルエステル0.01重量%以上と共に、ノナン酸バニリルアミド0.02重量%以上を共存させると、経時的に生じる褐変が顕著になる傾向が現れるが、本発明の外用組成物では、ノナン酸バニリルアミド0.02重量%以上含まれていても、経時的に生じる褐変を効果的に抑制することができる。
本発明の外用組成物において、非ステロイド性抗炎症薬とノナン酸バニリルアミドの比率については、前述する各含有量を充足する範囲であればよいが、例えば、非ステロイド性抗炎症薬1重量部当たり、ノナン酸バニリルアミドが0.001~1重量部、好ましくは0.01~0.3重量部、より好ましくは0.015~0.1重量部が挙げられる。
[ニコチン酸ベンジルエステル]
本発明の外用組成物は、ニコチン酸ベンジルエステルを0.01重量%以上含有する。ニコチン酸ベンジルエステルは、ナイアシンとベンジルアルコールとのエステルであり、吸収促進、血行促進等の効能が知られている公知の成分である。

本発明の外用組成物におけるニコチン酸ベンジルエステルの含有量は、0.01重量%以上であればよいが、具体的には、0.01~0.3重量%、好ましくは0.01~0.1重量%、より好ましくは0.01~0.1重量%、更に好ましくは0.02~0.1重量%、特に好ましくは0.04~0.1重量%が挙げられる。従来技術では、非ステロイド性抗炎症薬及びノナン酸バニリルアミド0.015重量%以上と共に、ニコチン酸ベンジルエステル0.01重量%以上を共存させると、経時的に褐変が生じるが、本発明の外用組成物では、ニコチン酸ベンジルエステルが0.01重量%以上の高含有量で含まれていても、経時的に生じる褐変を効果的に抑制することができる。
本発明の外用組成物において、非ステロイド性抗炎症薬とニコチン酸ベンジルエステルの比率については、前述する各含有量を充足する範囲であればよいが、例えば、非ステロイド性抗炎症薬1重量部当たり、ニコチン酸ベンジルエステルが0.001~1重量部、好ましくは0.005~0.3重量部、より好ましくは0.008~0.1重量部が挙げられる。
[モノテルペン]
本発明の外用組成物は、前述する成分に加えて、モノテルペンを含む。本発明の医薬組成物では、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上と共に、モノテルペンを含有させることにより、経時的に生じる褐変を効果的に抑制することが可能になる。
本発明の外用組成物は、前述する成分に加えて、モノテルペンを含む。本発明の医薬組成物では、非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上と共に、モノテルペンを含有させることにより、経時的に生じる褐変を効果的に抑制することが可能になる。
モノテルペンとは、分子内にイソプレン単位が2個含まれる構造を有し、清涼化作用等を有する公知の成分である。本発明で使用されるモノテルペンの種類については、薬学的に許容されることを限度として、特に制限されないが、例えば、メントール、チモール、ゲラニオール、リナロール、ボルネオール、シネオール、テルピネオール等のアルコール系モノテルペン;シトラール、シトロネラール、ペリルアルデヒド、サフラナール等のアルデヒド系モノテルペン;カンフル、メントン、カルボメントン、ヨノン等のケトン系モノテルペン等が挙げられる。これらのモノテルペンは、光学異性体が存在する場合には、d体、l体、dl体のいずれであってもよい。これらのモノテルペンは、1種単独で使用してもよく、2種以上を組み合わせて使用してもよい。
また、本発明では、モノテルペンとして、モノテルペンを含む精油を使用してもよい。モノテルペンを含む精油は、公知のものから適宜選択して使用することができるが、例えば、メントールを含む精油としては、ハッカ油、ペパーミント油、スペアミント油等が挙げられる。なお、本明細書におけるモノテルペンの含有量や比率に関する記載は、モノテルペンを含む精油を使用する場合は、当該精油に含まれるモノテルペン量に換算した値である。
これらのモノテルペンの中でも、好ましくはメントール、より好ましくはl-メントールが挙げられる。
本発明の外用組成物におけるモノテルペンの含有量については、使用するモノテルペンの種類等に応じて適宜設定すればよいが、例えば、モノテルペンの総量で、0.1~15重量%、経時的に生じる褐変をより一層抑制させる観点から、好ましくは1~10重量%、より好ましくは2~7重量%が挙げられる。
本発明の外用組成物において、非ステロイド性抗炎症薬とモノテルペンの比率については、前述する各含有量を充足する範囲であればよいが、例えば、非ステロイド性抗炎症薬1重量部当たり、モノテルペンが0.01~500重量部、経時的に生じる褐変をより一層抑制させる観点から、好ましくは0.1~50重量部、より好ましくは1~5重量部が挙げられる。
[1価低級アルコール]
本発明の外用組成物は、前述する成分に加えて、1価低級アルコールを含んでいてもよい。本発明において、1価低級アルコールとは炭素数1~5の1価アルコールを指す。

1価低級アルコールの種類については、薬学的に許容されることを限度として特に制限されないが、例えば、エタノール、n-プロパノール、イソプロパノール等が挙げられる。これらの1価低級アルコールの中でも、好ましくはエタノールが挙げられる。
本発明の外用医薬組成物に1価低級アルコールを含有させる場合、その含有量については、特に制限されないが、例えば、0.1~90重量%、好ましくは25~80重量%、より好ましくは50~80重量%が挙げられる。
[水]
本発明の外用組成物は、更に水を含んでいてもよい。本発明の外用医薬組成物に水を含有させる場合、その含有量については、特に制限されないが、例えば、0.1~80重量%、好ましくは1~50重量%、より好ましくは10~40重量%、更に好ましくは15~30重量%が挙げられる。

[その他の成分]
本発明の外用組成物は、前述する成分の他に、必要に応じて、通常使用される他の添加剤が含まれていてもよい。このような添加剤としては、例えば、界面活性剤、植物油、動物油、鉱物油、脂肪酸アルキルエステル、脂肪酸、多価アルコール、高級アルコール、pH調節剤、緩衝剤、可溶化剤、防腐剤、保存剤、酸化防止剤、安定化剤、香料等が挙げられる。本発明の外用組成物において、これらの添加剤を含有させる場合、その含有量については、使用する添加剤の種類等に応じて適宜設定すればよい。
本発明の外用組成物は、前述する成分の他に、必要に応じて、通常使用される他の添加剤が含まれていてもよい。このような添加剤としては、例えば、界面活性剤、植物油、動物油、鉱物油、脂肪酸アルキルエステル、脂肪酸、多価アルコール、高級アルコール、pH調節剤、緩衝剤、可溶化剤、防腐剤、保存剤、酸化防止剤、安定化剤、香料等が挙げられる。本発明の外用組成物において、これらの添加剤を含有させる場合、その含有量については、使用する添加剤の種類等に応じて適宜設定すればよい。
また、本発明の外用組成物は、前述する成分の他に、薬理成分が含まれていてもよい。このような薬理成分としては、例えば、抗ヒスタミン剤、局所麻酔剤、保湿剤、殺菌剤、抗菌剤、鎮痒剤、皮膚保護剤、血行促進成分、ビタミン類等が挙げられる。これらの薬理成分は、1種単独で使用してもよく、また2種以上を組み合わせて使用してもよい。また、本発明の外用組成物において、これらの薬理成分を含有させる場合、その濃度については、使用する薬理成分の種類、期待する効果等に応じて適宜設定すればよい。
[製剤形態]
本発明の外用組成物は、経皮適用できる剤型である限り、その製剤形態については、特に制限されず、液状又は半固形状(ゲル状、軟膏状、ペースト状)いずれであってもよいが、好ましくは液状が挙げられる。

また、本発明の外用組成物の製剤形態として、具体的には、液剤、クリーム剤、ローション剤、ジェル剤、乳液剤、エアゾール剤等が挙げられる。これらの中でも、好ましくは液剤、クリーム剤、ローション剤、ジェル剤、乳液剤が挙げられる。これらの製剤形態への調製は、第十七改正日本薬局方 製剤総則等に記載の公知の方法に従って、製剤形態に応じた添加剤を用いて製剤化することにより行うことができる。
以下に実施例を示して本発明をより具体的に説明するが、本発明はこれらに限定されるものではない。
試験例1:ロキソプロフェンナトリウム水和物を含む外用組成物の評価
表1及び2に示す組成の外用組成物(液剤)を調製した。得られた外用組成物は、いずれも、調製直後は無色透明の外観を呈していた。得られた外用組成物を透明ガラス瓶に充填して、50℃、相対湿度60%RHの暗所に2カ月間保存した。保存後の外用組成物の外観を目視にて観察し、以下の判定基準に従って褐変の程度を評価した。
<褐変の程度の判定基準>
◎ :褐変が認められず、無色透明である。
〇 :ごく僅かに褐変が認められるが、実用上は問題ないレベルである。
△ :やや褐変が認められる。
× :明らかな褐変が認められる。
××:顕著な褐変が認められ、濃い褐色を呈している。
表1及び2に示す組成の外用組成物(液剤)を調製した。得られた外用組成物は、いずれも、調製直後は無色透明の外観を呈していた。得られた外用組成物を透明ガラス瓶に充填して、50℃、相対湿度60%RHの暗所に2カ月間保存した。保存後の外用組成物の外観を目視にて観察し、以下の判定基準に従って褐変の程度を評価した。
<褐変の程度の判定基準>
◎ :褐変が認められず、無色透明である。
〇 :ごく僅かに褐変が認められるが、実用上は問題ないレベルである。
△ :やや褐変が認められる。
× :明らかな褐変が認められる。
××:顕著な褐変が認められ、濃い褐色を呈している。
結果を表1及び2に示す。ノナン酸バニリルアミド0.01重量%を単独で含む場合には、保存後に褐変がやや認められた(参考例1)。また、ロキソプロフェンナトリウム水和物とノナン酸バニリルアミド0.01重量%を含む場合には、保存後には褐変は殆ど認められなかった(参考例2)。一方、ロキソプロフェンナトリウム水和物と共に、ノナン酸バニリルアミドを0.015重量%以上を含む場合には褐変が生じ、ノナン酸バニリルアミドの含有量を0.02重量%以上を含む場合には更に褐変が生じた。更にニコチン酸ベンジルエステルを0.01重量%以上含有させた場合には、褐変が顕著になっていた(比較例1~7)。これに対して、ロキソプロフェンナトリウム水和物、ノナン酸バニリルアミド0.015重量%以上、及びニコチン酸ベンジルエステル0.01重量%以上と共にl-メントールを含有させた場合には、褐変を十分に抑制できており、保存後でも無色透明な外観を維持できていた。また、実施例1~12のl-メントールの含有量を6重量%に変更した場合においても、実施例1~12と同様に褐変を十分に抑制できており、保存後でも無色透明な外観を維持できていた。
試験例2:ジクロフェナクナトリウムを含む外用組成物の評価
表3及び4に示す組成の外用組成物(液剤)を調製した。得られた外用組成物は、いずれも、調製直後は無色透明の外観を呈していた。得られた外用組成物の保存後の褐変の程度を前記試験例1と同様の方法で評価した。

結果を表3及び4に示す。非ステロイド性抗炎症薬としてジクロフェナクナトリウムを使用した場合であっても、ノナン酸バニリルアミドを0.015重量%以上を含む場合には褐変が生じ、ノナン酸バニリルアミドの含有量が0.02重量%以上を含む場合には更に褐変が生じた。更にニコチン酸ベンジルエステル0.01重量%以上を共存させた場合には著しい褐変が認められたが(比較例8~14)、これらの成分に加えてl-メントールを含有させることにより、褐変を抑制でき、保存後でも実用上問題ない程度の外観を維持できていた(実施例13~24)。また、実施例13~24のl-メントールの含有量を6重量%に変更した場合においては、実施例13~24と比較して顕著に褐変を抑制できており、保存後でも無色透明な外観を維持できていた。

- 非ステロイド性抗炎症薬、ノナン酸バニリルアミド0.015重量%以上、ニコチン酸ベンジルエステル0.01重量%以上、及びモノテルペンを含有する、外用組成物。
- 前記非ステロイド性抗炎症薬が、ロキソプロフェン、ジクロフェナク、及びこれらの塩よりなる群から選択される少なくとも1種である、請求項1に記載の外用組成物。
- 前記モノテルペンが、メントールである、請求項1又は2に記載の外用組成物。
- 液剤、クリーム剤、ローション剤、ジェル剤、又は乳液剤である、請求項1~3のいずれかに記載の外用組成物。
