Abstract
Description
タマネギ加工品の原料生タマネギ換算量に対して、ピルビン酸含有量が3.8μmol/g未満であり、ヘキサナール含有量が0.025ppm(wt/wt)以上である
ことを特徴とする、タマネギ加工品。
(2)原料生タマネギ換算量に対するピルビン酸含有量が1.5μmol/g未満である、(1)に記載のタマネギ加工品。
(3)アリイナーゼ遺伝子の発現が従来品種と比べて50分の1未満に抑制されているタマネギの加工品である、(1)又は(2)に記載のタマネギ加工品。
(4)タマネギ加工品の製造方法であって、
タマネギを切断する工程と、
切断したタマネギを、加熱価が900以上10,000未満となるように50℃以上で加熱処理する工程と、
加熱処理後のタマネギを加工してタマネギ加工品を製造する工程と
を含み、
前記加熱価が、タマネギの品温(A)[℃]に対して10{(A-40)/10}で求められる値を加熱時間[分]で積分した値である、
タマネギ加工品の製造方法。
(5)タマネギが、アリイナーゼ遺伝子の発現が従来品種と比べて50分の1未満に抑制されているタマネギである、(4)に記載の方法。
本明細書に開示するタマネギ加工品の製造方法によれば、上記の特徴を有するタマネギ加工品を製造することができる。
本明細書で開示するタマネギ加工品の実施形態は、
タマネギ加工品の原料生タマネギ換算量に対して、ピルビン酸含有量が3.8μmol/g未満であり、ヘキサナール含有量が0.025ppm(wt/wt)以上である
ことを特徴とする。
タマネギ加工品は、袋状の容器、瓶、缶、PET容器等に収容された容器詰タマネギ加工品の形態であることができる。
好ましくは「アリイナーゼ遺伝子」とは、辛み成分及び催涙成分の生成に主に寄与する特定のアリイナーゼ遺伝子である。
(a)配列番号1で示されるアミノ酸配列を含むか、当該アミノ酸配列よりなるポリペプチド;
(b)配列番号1で示されるアミノ酸配列において、一又は複数個のアミノ酸の欠失、置換、挿入及び/又は付加を有するアミノ酸配列を含むか当該アミノ酸配列よりなり、かつアリイナーゼ活性を有するポリペプチド;
(c)配列番号1で示されるアミノ酸配列と、70%以上、80%以上、90%以上、95%以上、96%以上、97%以上、98%以上、又は99%以上の同一性を有するアミノ酸配列を含むか当該アミノ酸配列よりなり、かつアリイナーゼ活性を有するポリペプチド。
(a)配列番号1で示されるアミノ酸配列を含むポリペプチド;
(b)配列番号1で示されるアミノ酸配列と、95%以上、96%以上、97%以上、98%以上、又は99%以上の同一性を有するアミノ酸配列を含み、かつアリイナーゼ活性を有するポリペプチド。

本明細書で開示するタマネギ加工品の製造方法の実施形態は、
タマネギを切断する工程と、
切断したタマネギを、加熱価が900以上10,000未満となるように50℃以上で加熱処理する工程と、
加熱処理後のタマネギを加工してタマネギ加工品を製造する工程と
を含み、
前記加熱価が、タマネギの品温(A)[℃]に対して10{(A-40)/10}で求められる値を加熱時間[分]で積分した値である
ことを特徴とする。
(式):CV値=10{(品温-基準温度)/Z値}
様々な加熱条件での加熱量を比較するために基準温度とZ値は固定する必要があるが、その値は対象により異なる。本明細書におけるタマネギの加熱では、Z値を10℃とし、基準温度を40℃とした。
生タマネギとして、NCIMB 42219として代表的な種子が国際寄託されているタマネギを用いた。
生タマネギの鱗茎部の皮を剥き、天地カットした後に10mm角のダイス状にカットした。
タマネギピューレ試料の官能評価(辛み、苦み、フレッシュ感)、ピルビン酸量測定、ヘキサナール量測定は以下の条件で行った。
粉砕から30分経過後にタマネギピューレ試料を喫食し辛み、苦み、フレッシュ感を評価した。
0:全く辛みを感じない
1:新タマネギ未満の辛みを感じる
2:新タマネギ以上に辛みを感じる
0:全く苦みを感じない
1:新タマネギ未満の苦みを感じる
2:新タマネギ以上に苦みを感じる
0:フレッシュ感を感じない
1:フレッシュ感を感じる
試料をミルミキサーで破砕して作製したピューレを、15000rpm、4℃、10分間遠心し上澄みをサンプル(抽出サンプル)とした。サンプル20μLを96ウェルプレートに入れ、粉砕から30分後に水43μL、DNPH(2,4-ジニトロフェニルヒドラジン)溶液66μLを添加し、37℃で10分間反応させた。10分後、1.5M NaOHを66μL添加し、反応を停止させ、ピペッティングを3回以上行い混合した。分光光度計で515nmの吸光度を測定した。抽出サンプルのかわりにピルビン酸Na溶液の希釈系列を用いて、同様に測定した値から、検量線を作成し、抽出サンプルでの吸光度測定値をピルビン酸量に換算した。こうして測定されたタマネギピューレの抽出サンプル中のピルビン酸量と、タマネギピューレの調製に用いた原料生タマネギの重量とから、タマネギピューレの、原料生タマネギ換算重量あたりのピルビン酸量(μmol/g)を算出した結果を下記表に示す。
以下の手順に従いタマネギピューレ中のヘキサナール濃度を測定した。測定されたタマネギピューレ中のヘキサナール濃度と、タマネギピューレの調製に用いた原料生タマネギの重量とから、タマネギピューレの、原料生タマネギ換算重量あたりのヘキサナール濃度(ppm(wt/wt))を算出した結果を下記表に示す。
(内部標準液作成)
内部標準試薬:サフロール CAS No.94-59-7
サフロールをメタノールに溶解し、約100ppmとなるよう調製し、内部標準液とする。
(標準添加サンプル作成)
標準試薬を添加する試験品(標準添加用サンプル)として、ヘキサナールが観測されない条件(加熱価が十分に高い条件、例えば80℃、3分間(加熱価30000)の加熱条件)で加熱処理したタマネギピューレ試料を使用する。
標準試薬:ヘキサナール CAS No.66-25-1
ヘキサナールをメタノールに溶解し、タマネギピューレ試料のヘキサナールが定量範囲に入るようメタノールで希釈して、数段階作成する。これを標準液とする。
20mLバイアル瓶に標準添加用サンプルをマイクロピペットで2.0gサンプリングする。
20mLバイアル瓶に標準液50μLを入れる。
20mLバイアル瓶に内部標準液50μLを入れる。
塩化ナトリウムを0.5g添加する。
マイクロピペットで緩やかに混和する。
密栓したものを標準添加サンプルとし、測定機器に供する。
標準液によるタマネギピューレ試料の重量増はわずかであるので、考慮しないこととする。
(測定サンプル作成)
20mLバイアル瓶にタマネギピューレ試料試験品をマイクロピペット2.0gサンプリングする。
20mLバイアル瓶にメタノール50μLを入れる。
20mLバイアル瓶に内部標準液50μLを入れる。
塩化ナトリウムを0.5g添加する。
マイクロピペットで緩やかに混和する。
密栓したものを測定サンプルとし、測定機器に供する。
(測定方法)

(計算方法)
標準添加サンプルにおいて、標準添加サンプル中の既知ヘキサナール濃度に対して、あらかじめ定めたM/Zにおけるイオンクロマトグラムより得られたヘキサナールピークとサフロールピークの面積比の検量線を作成する。
測定サンプルにおいて、観測されたヘキサナールピークとサフロールピークの面積比を検量線にあてはめ、測定サンプルのヘキサナール濃度を算出する。

- タマネギ加工品であって、
タマネギ加工品の原料生タマネギ換算量に対して、ピルビン酸含有量が3.8μmol/g未満であり、ヘキサナール含有量が0.025ppm(wt/wt)以上である
ことを特徴とする、タマネギ加工品。 - 原料生タマネギ換算量に対するピルビン酸含有量が1.5μmol/g未満である、請求項1に記載のタマネギ加工品。
- アリイナーゼ遺伝子の発現が従来品種と比べて50分の1未満に抑制されているタマネギの加工品である、請求項1又は2に記載のタマネギ加工品。
- タマネギ加工品の製造方法であって、
タマネギを切断する工程と、
切断したタマネギを、加熱価が900以上10,000未満となるように50℃以上で加熱処理する工程と、
加熱処理後のタマネギを加工してタマネギ加工品を製造する工程と
を含み、
