特許文献1には、1-イソブチル-3-メチルベンゼンに対して、加圧下で一酸化炭素と反応させ、4位にホルミル基を直接導入することが記載されている。
本発明は、1,4-ジアルキルベンゼンを含む反応原料を用いて、m-ジアルキルベンズアルデヒドを製造する方法を提供することを目的とする。
すなわち、本発明は、以下の通りである。
<3> 式(1)で表される化合物が、下記式(1-1)~(1-7)から選択される、<1>又は<2>に記載の製造方法。
<5> ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HF/BF3であり、かつ、反応原料に対するHFのモル比(HF/反応原料)が、5.0以上25.0以下である、<1>~<4>のいずれかに記載の製造方法。
<6> ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HF/BF3であり、かつ、反応原料に対するBF3のモル比(BF3/反応原料)が、0.2以上2.5以下である、<1>~<5>のいずれかに記載の製造方法。
<7> ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HCl/AlCl3であり、かつ、反応原料に対するHClのモル比(HCl/反応原料)が、0.0001以上0.3以下である、<1>~<4>のいずれかに記載の製造方法。

本発明の式(3)で表されるm-ジアルキルベンズアルデヒドの製造方法(以下、単に「本発明の製造方法」ともいう)は、ブレンステッド酸及びルイス酸の存在下で、式(1)で表される1,4-ジアルキルベンゼンを含む反応原料に、一酸化炭素を作用させる工程を有し、前記反応原料が、式(1)で表される1,4-ジアルキルベンゼンであるか、又は式(1)で表される1,4-ジアルキルベンゼンを10モル%以上含有する、式(1)で表される1,4-ジアルキルベンゼンと式(2)で表される1,3-ジアルキルベンゼンとの混合物である。
反応原料がp-ジアルキルベンゼンであるときの本発明の製造方法は、以下の式(I)の通りであり、反応原料がp-ジアルキルベンゼンとm-ジアルキルベンゼンとの混合物であるときの本発明の製造方法は、以下の式(II)の通りである。
また、式(3)で表される化合物は、下記式(3-1)で表される2-R1-4-R2ベンズアルデヒドと、式(3-2)で表される4-R1-2-R2-ベンズアルデヒドとの混合物であるが、式(3-1)で表される化合物が主生成物であり、式(3-2)で表される化合物が副生成物である。
本発明者等は、特定構造のp-ジアルキルベンゼンを含む出発原料に、ブレンステッド酸及びルイス酸の存在下に、一酸化炭素を作用させることにより、アルキル基が転位して異性化した、m-ジアルキルベンズアルデヒドが得られることを見出した。
上記の反応が生じる詳細な理由は不明であるが、式(1)中のR2が、ベンジル位が3級炭素である炭素数3以上6以下の鎖状又は環状のアルキル基である、p-ジアルキルベンゼンを出発原料として使用することにより、ベンジル位の炭素上で陽電荷が安定化し、より熱力学的に安定なm-ジアルキルベンゼンへ異性化した後、ホルミル化が進行したものと考えられる。
なお、得られるm-ジアルキルベンズアルデヒドにおけるホルミル基の置換位置は特に限定されないが、立体障害及び電子密度により、2,4-ジアルキルベンズアルデヒドの収率が高い傾向にあり、上述したように、2位がR1、4位がR2である、2,4-ジアルキルベンズアルデヒドの収率がより高い傾向にある。
以下、本発明について詳細に説明する。
本発明において、反応原料として、p-ジアルキルベンゼン(式(1)で表される化合物)、又はp-ジアルキルベンゼンを10モル%以上含有する、p-ジアルキルベンゼンとm-ジアルキルベンゼン(式(2)で表される化合物)との混合物(以下、単に「混合物」ともいう)を使用する。
〔式(1)で表される化合物〕
上記式(1)中、R1はメチル基又はエチル基を示し、R2はベンジル位が3級炭素である炭素数3以上6以下の鎖状又は環状のアルキル基を示す。
R1は、反応性の観点から、メチル基であることが好ましい。
R2が鎖状のアルキル基であるとき、R2は炭素数3以上5以下であることが好ましい。R2としては、イソプロピル基、1-メチルプロピル基、1-エチルプロピル基、1-メチルブチル基、1,2-ジメチルプロピル基が例示される。
R2が環状のアルキル基であるとき、R2は炭素数4以上6以下であることが好ましく、5以上6以下であることがより好ましい。
出発原料として、p-ジアルキルベンゼンとm-ジアルキルベンゼンとの混合物を使用する場合、p-ジアルキルベンゼンの含有量は10モル%以上である。
本発明の製造方法は、p-ジアルキルベンゼンを含む反応原料を使用する点に特徴がある。
混合物中のp-ジアルキルベンゼンの含有量は、好ましくは15モル%以上、より好ましくは20モル%以上、更に好ましくは25モル%以上、更に好ましくは30モル%以上である。混合物中のp-ジアルキルベンゼンの含有量が多くても、m-ジアルキルベンズアルデヒドを効率的に得ることができる。
混合物中のp-ジアルキルベンゼンの含有量の上限は特に限定されず、100モル%未満である。
本発明の製造方法は、ブレンステッド酸及びルイス酸の存在下で、p-ジアルキルベンゼンを含む反応原料に、一酸化炭素を反応させる。
本発明は、Gattermann-Koch反応を利用したものであり、Gattermann-Koch反応は、ブレンステッド酸及びルイス酸の存在下に、アルキルベンゼンと一酸化炭素から、芳香族アルデヒドを合成する方法であり、古くは、ブレンステッド酸としてHCl、ルイス酸としてAlCl3を使用した、HCl/AlCl3の組合せが使用されてきた。ブレンステッド酸及びルイス酸の組合せは、これに限定されず、例えば、HF/SbF5、CF3SO3H/SbF5、HF/BF3であってもよい。
これらの中でも、ブレンステッド酸及びルイス酸の組合せ(ブレンステッド酸/ルイス酸)として、HF/BF3又はHCl/AlCl3が好ましく、反応性、収率、及び選択率の観点から、HF/BF3がより好ましい。
本発明において、ブレンステッド酸及びルイス酸の存在下で、m-ジアルキルベンゼンを含む反応原料に一酸化炭素を反応させる。
反応時の一酸化炭素を作用させる温度は、反応性を向上させ、副反応を抑制し、ホルミル基が導入される位置選択性を向上させる観点から、好ましくは-30℃以上、より好ましくは-27℃以上であり、そして、好ましくは30℃以下、より好ましくは15℃以下、更に好ましくは5℃以下である。
ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)の組合せとして、HF/BF3を使用する場合、前記反応温度は、反応性を向上させ、かつ、副反応を抑制する観点から、好ましくは-30℃以上、より好ましくは-27℃以上であり、そして、好ましくは10℃以下、より好ましくは-5℃以下、更に好ましくは-15℃以下である。
また、ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)の組合せとして、HCl/AlCl3を使用する場合、前記反応温度は、反応性を向上させ、かつ、副反応を抑制する観点から、好ましくは-15℃以上、より好ましくは-10℃以上、更に好ましくは-5℃以上であり、そして、好ましくは30℃以下、より好ましくは15℃以下、更に好ましくは5℃以下である。
反応時の圧力は、反応性を向上させ、かつ、副反応を抑制する観点から、好ましくは1.0MPaG以上、より好ましくは1.5MPaG以上、更に好ましくは1.8MPaG以上であり、そして、好ましくは3.0MPaG以下、より好ましくは2.5MPaG以下、更に好ましくは2.2MPaG以下である。
溶媒の使用量は、特に限定されず、反応の均一性、反応速度、及び溶媒除去の観点から、適宜選択すればよい。
なお、例えば、ブレンステッド酸としてHFを使用する場合には、HFは溶媒としても機能するため、溶媒は使用しなくてもよく、溶媒を用いない方が、溶媒除去が不要となるため、好ましい。
また、製造方法に使用する装置は、加圧下で、温度を調整しつつ、液相と気相とが十分に混合できる反応装置である。
例えば、回分式では、撹拌装置付反応器に、反応原料、ブレンステッド酸、ルイス酸、及び必要に応じて溶媒を仕込み、内容物を撹拌し液温を好ましくは-30℃以上30℃以下に保った後、一酸化炭素により、好ましくは1.0~3.0MPaGに昇圧し、その後そのままの圧力、液温を保ち、一酸化炭素が吸収されなくなるまで10分~5時間保持した後、反応生成液を抜き出し、m-ジアルキルベンズアルデヒドを得ることができる。
転化率(モル%)={1-(反応後に残存する反応原料の物質量(モル数))/(反応原料の仕込み物質量(モル数))}×100
選択率(モル%)=目的とする化合物の物質量(モル数)/反応により消費した反応原料の物質量(モル数)×100
収率(モル%)=転化率×選択率/100
なお、各物質量(モル数)は、GC分析によるピーク面積を分子量で除し、その比を取ることで算出した。
装置:GC-2010 Plus((株)島津製作所製)
検出器:FID
カラム:DB-1(Agilent社製キャピラリーカラム)
(0.32mmφ×30m×0.50μm)
昇温条件:100℃から310℃まで5℃/分で昇温し、310℃で20分保持
装置1:Bruker Avance2 600MHz-NMR
(5mm Cryo-CPDUL Probe)(BRUKER社製)
装置2:Bruker Avance3 HD 500MHz-NMR
(5mm BBO CryoProbe)(BRUKER社製)
溶媒:重クロロホルム(CDCl3)
測定モード:1H、13C、HSQC、Dept(90deg)、HMBC
内部標準物質:テトラメチルシラン(TMS)
なお、1H、13C、Dept(90deg)の測定には装置1を使用し、HSQC、HMBCの測定には装置2を使用した。
ナックドライブ式撹拌機と上部に3個の入口ノズル、底部に1個の出口ノズルを備え、ジャケットにより内部温度を制御できる500mLのオートクレーブをホルミル化反応器として使用した。
ジャケットに冷媒を流し、-25℃に冷却したオートクレーブにフッ化水素126.5g(6.32モル)を仕込んだ。
その後、撹拌させながら、温度が-25℃を超えないように調整しながら、三フッ化ホウ素42.6g(0.63モル)を加えた。
三フッ化ホウ素を加えた後、オートクレーブ内の温度を-25℃に保ったまま一酸化炭素により2MPaGまで昇圧し、m,p-混合シメン(イソプロピルトルエン、m:p(モル比)=68.3:31.7)56.7g(0.42モル)を加えた。
温度-25℃、圧力2MPaGを保ちながら45分撹拌した後、オートクレーブ内の反応混合液を氷水中に抜液した。抜液したものをよく振り混ぜた後、油層を分液した。得られた油層部を水洗した後、ガスクロマトグラフィー(GC)により分析したところ、m,p-混合シメン転化率は98.9モル%であり、4-イソプロピル-2-メチルベンズアルデヒド選択率は85.9モル%、2-イソプロピル-4-メチルベンズアルデヒド選択率は11.0モル%で、2異性体合計での選択率は96.9モル%の結果であった。
反応条件、及び使用する反応原料の種類を表1のように変更した以外は実施例1と同様にして、m-ジアルキルベンズアルデヒドを製造した。
結果を表1に示す。
NMR分析により、主生成物は、4-イソプロピル2-メチル-ベンズアルデヒドと、2-イソプロピル-4-メチルベンズアルデヒドであることが同定された。1H-NMRのピーク面積の結果と、GC分析の結果はよく整合していた。
NMRスペクトル分析の結果を以下に示す。

13C NMR (150 MHz, CDCl3) δ 19.8 [10], 23.6 [9], 34.3 [8], 124.4 [7], 130.0 [6], 132.3 [5], 132.6 [4], 140.8 [3], 155.3 [2], 192.4 [1]
1H NMR (600 MHz, CDCl3) δ 1.28-1.30 (6H, d, J = 7.2Hz, [9]), 2.41 (3H, s, [10]), 3.93-4.00 (1H, sep, J = 7.2Hz, [8]), 7.14-7.15 (1H, d, J = 7.8Hz, [6]), 7.24 (1H, s, [7]), 7.70-7.72 (1H, m*1, [4]), 10.29 (1H, s, [1])
13C NMR (150 MHz, CDCl3) δ 22.0 [10], 23.8 [9], 27.5 [8], 126.8 [7], 126.9 [6], 130.7 [5], 132.0 [4], 144.9 [3], 151.5 [2], 192.0 [1]
*1:4-イソプロピル-2-メチル-ベンズアルデヒドのピークと被っているため、十分に読み取ることが困難である。
反応条件、及び使用するp-ジアルキルベンゼンの種類を表2のように変更した以外は実施例1と同様にして、ジアルキルベンズアルデヒドを製造した。
結果を表2に示す。
ナックドライブ式撹拌機と上部に3個の入口ノズル、底部に1個の出口ノズルを備え、ジャケットにより内部温度を制御できる500mLのオートクレーブをホルミル化反応器として使用した。
ジャケットに冷媒を流し、0℃に冷却したオートクレーブに塩化アルミニウム74.5g(0.56モル)、1,2-ジクロロエタン(1,2-DCE)147.4g(1.49モル)、及び35%塩酸1mL(0.01モル)を仕込んだ。
その後、撹拌させながら、オートクレーブ内の温度を0℃に保ったまま一酸化炭素により2MPaGまで昇圧し、p-シメン50.0g(0.37モル)を加えた。
温度0℃、圧力2MPaGを保ちながら60分撹拌した後、オートクレーブ内の反応混合液を氷水中に抜液した。抜液したものをよく振り混ぜた後、油層を分液した。得られた油層部を水洗した後、ガスクロマトグラフィーにより分析したところ、m,p-混合シメン転化率は79.7モル%であり、4-イソプロピル-2-メチルベンズアルデヒド選択率は22.7モル%、2-イソプロピル-4-メチルベンズアルデヒド選択率は4.5モル%で、2異性体合計での選択率は27.2モル%の結果であった。
結果を表3に示す。
Claims (10)
- ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HF/BF3又はHCl/AlCl3である、請求項1に記載の製造方法。
- 一酸化炭素を作用させる温度が、-30℃以上30℃以下である、請求項1~3のいずれかに記載の製造方法。
- ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HF/BF3であり、かつ、反応原料に対するHFのモル比(HF/反応原料)が、5.0以上25.0以下である、請求項1~4のいずれかに記載の製造方法。
- ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HF/BF3であり、かつ、反応原料に対するBF3のモル比(BF3/反応原料)が、0.2以上2.5以下である、請求項1~5のいずれかに記載の製造方法。
- ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HCl/AlCl3であり、かつ、反応原料に対するHClのモル比(HCl/反応原料)が、0.0001以上0.3以下である、請求項1~4のいずれかに記載の製造方法。
- ブレンステッド酸及びルイス酸(ブレンステッド酸/ルイス酸)が、HCl/AlCl3であり、かつ、反応原料に対するAlCl3のモル比(AlCl3/反応原料)が、0.2以上2.5以下である、請求項1~4、7のいずれかに記載の製造方法。
- 反応圧力が1.0MPaG以上3.0MPaG以下である、請求項1~8のいずれかに記載の製造方法。
- 前記反応原料が、式(1)で表される1,4-ジアルキルベンゼンを10モル%以上含有する、式(1)で表される1,4-ジアルキルベンゼンと式(2)で表される1,3-ジアルキルベンゼンとの混合物である、請求項1~9のいずれかに記載の製造方法。
