Description

중증 열성 혈소판 감소 증후군(SFTS) 바이러스 감염 질환 예방 또는 치료용 백신 조성물
본 발명은 중증 열성 혈소판 감소 증후군(Severe Fever with Thrombocytopenia Syndrome; SFTS) 바이러스 질환에 의한 감염병을 예방 또는 치료하기 위한 백신 조성물에 관한 것이다.
중증 열성 혈소판 감소 증후군(SFTS; Severe Fever with Thrombocytopenia Syndrome)에 의한 심한 열병은 중국, 한국 및 일본에서 흔히 나타나는 신흥 바이러스 질환이다. SFTS에 대한 효과적인 백신이나 특정 치료법은 현재 제공되지 않고 있는 실정이다. SFTS는 고열, 구토, 설사, 혈소판감소, 백혈구감소 및 다발성 장기부전과 같은 증상을 동반하며, 치사율이 6-30%에 이르는 심각한 질환이다 (Yu XJ et al., N. Engl. J. Med. 2011; 364:1523-32; Ding F et al Clin Infect Dis 2013; 56: 1682-3).
또한, SFTS 바이러스의 혈청전환 및 바이러스혈증이 염소, 양, 소, 돼지 및 개와 같은 사육동물에서 발견되었으며, 이러한 동물도 SFTS 바이러스가 퍼진 지역에서 중간 매개체로 작용하는 것으로 생각된다 (Zhao L et al. Emerg Infect Dis 2013; 18: 963-5; Niu G et al. Emerg Infect Dis 2013; 19: 756-63).
한편, 중국 공개 특허 공보 제102070704호는 SFTS 바이러스를 이용한 SFTS 바이러스 증폭 및 검출 키트를 개시하고 있다.
따라서, 본 발명자들은 SFTS에 대한 효과적인 백신을 고안하였다.
본 발명의 목적은 개체에서 면역 반응을 효과적으로 유도하는 SFTS 바이러스 항원의 재조합 DNA 또는 펩티드; 및 이를 포함하는 SFTS 바이러스 백신을 제공하는 것이다.
구체적으로는, 본 발명은 중증 열성 혈소판 감소 증후군(Severe Fever with Thrombocytopenia Syndrome; SFTS) 바이러스에 의한 감염병을 예방 또는 치료하기 위한 백신 조성물을 제공하는 것이다.
그러나 본 발명이 이루고자 하는 기술적 과제는 이상에서 언급한 과제에 제한되지 않으며, 언급되지 않은 또 다른 과제들은 아래의 기재로부터 당업계에서 통상의 지식을 가진 자에게 명확하게 이해될 수 있을 것이다.

본 발명은 바이러스 항원의 재조합 DNA 또는 펩티드를 포함하는 바이러스 백신 및 백신 방법에 관한 것이고, 또한 항원 단백질 또는 펩티드인 면역원을 코딩하는 뉴클레오티드 서열을 포유류 체내에 도입하는 단계(여기서, 상기 단백질 또는 펩티드는 포유류 체내에서 발현되어 항원 단백질 또는 펩티드에 대한 면역 반응을 야기한다)를 포함하는 백신 방법의 개선에 관한 것이다. 이러한 백신 방법은 공지되어 있다.

서열번호 295로 표시되는 아미노산 서열을 포함하거나, 서열번호 294로 표시되는 염기서열을 포함하는 재조합 제 5 DNA에 의해 코딩되는 재조합 제 5 펩티드로 이루어진 군에서 선택된 어느 하나 이상의 재조합 펩티드를 유효성분으로 포함하는 항원용 조성물을 제공한다.
본 발명의 항원용 조성물은 서열번호 287로 표시되는 아미노산 서열을 포함하거나, 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA에 의해 코딩되는 재조합 제 1 펩티드; 및 서열번호 289로 표시되는 아미노산 서열을 포함하거나, 서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA에 의해 코딩되는 재조합 제 2 펩티드; 중 적어도 하나를 포함할 수 있다.
본 발명의 항원용 조성물은 서열번호 287로 표시되는 아미노산 서열을 포함하거나, 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA에 의해 코딩되는 재조합 제 1 펩티드; 및 서열번호 289로 표시되는 아미노산 서열을 포함하거나, 서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA에 의해 코딩되는 재조합 제 2 펩티드;를 포함할 수 있다.

본 발명의 항원용 조성물 또는 백신은 본 발명이 속하는 기술 분야에서 통상적으로 이용되는 방법으로 제조할 수 있다. 본 발명의 항원용 조성물 또는 백신은 경구형 또는 비경구형 제제로 제조할 수 있고, 바람직하게는 비경구형 제제인 주사액제로 제조하며, 진피 내, 근육 내, 복막 내, 정맥 내, 피하 내, 비강 또는 경막 외(eidural) 경로로 투여할 수 있다.
본 발명의 항원용 조성물 또는 백신은 면역학적 유효량으로 개체에 투여할 수 있다. 상기 "면역학적 유효량"이란 중증 열성 혈소판 감소 증후군(Severe Fever with Thrombocytopenia Syndrome; SFTS) 바이러스 또는 SFTS 바이러스의 예방 또는 치료 효과를 나타낼 수 있을 정도의 충분한 양과 부작용이나 심각한 또는 과도한 면역 반응을 일으키지 않을 정도의 양을 의미하며, 정확한 투여 농도는 투여될 특정 면역원에 따라 달라지며, 예방 또는 치료 대상자의 연령, 체중, 건강, 성별, 개체의 약물에 대한 민감도, 투여 경로, 투여 방법 등 의학 분야에 잘 알려진 요소에 따라 당업자에 의해 용이하게 결정될 수 있으며, 1회 내지 수회 투여 가능하다.
본 발명의 백신은 약제학적 유효량으로 투여한다. 용어 "약제학적 유효량"이란 백신효과를 나타낼 수 있을 정도의 충분한 양과 부작용이나 심각한 또는 과도한 면역 반응을 일으키지 않을 정도의 양을 의미하며, 정확한 투여 농도는 투여될 항원에 따라 달라지며, 대상자의 연령, 체중, 건강, 성별, 대상자의 약물에 대한 민감도, 투여 경로, 투여 방법 등 의학 분야에 잘 알려진 요소에 따라 당업자에 의해 용이하게 결정될 수 있으며, 1회 내지 수회 투여 가능하다.
본 발명의 DNA 백신은 약제학적으로 허용되는 담체 내에 본 발명의 항원결정기 펩티드를 코딩하는 뉴클레오티드가 포함된 DNA 백신으로 바람직하게는 DNA 플라스미드 형태이며, 가장 바람직하게는 mock 플라스미드(pVax-1 유래) 형태이지만, 이에 한정되는 것이 아니다. 따라서, 상기 뉴클레오티드들이 공지의 다양한 재조합 발현 벡터에 삽입된 것이 바람직하다.
본 발명의 또 다른 구현 예에 따르면,

본 발명의 발현 벡터는 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA; 및 서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA; 중 적어도 하나의 재조합 DNA를 포함할 수 있다.
본 발명의 발현 벡터는 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA; 및 서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA; 를 포함할 수 있다.
본 발명에서 상기 "벡터(vector)"는 숙주 세포에서 목적 유전자를 발현시키기 위한 수단을 의미한다. 상기 벡터는 목적 유전자 발현을 위한 요소 (elements)를 포함하는 것으로, 복제원점 (replication origin), 프로모터, 작동 유전자 (operator), 전사 종결 서열 (terminator) 등을 포함할 수 있고, 숙주 세포의 게놈 내로의 도입을 위한 적절한 효소 부위 (예컨대, 제한 효소 부위) 및/또는 숙주 세포 내로의 성공적인 도입을 확인하기 위한 선별 마커 및/또는 단백질로의 번역을 위한 리보좀 결합 부위 (ribosome binding site; RBS), IRES (Internal Ribosome Entry Site) 등을 추가로 포함할 수 있다. 상기 벡터는 프로모터로서 상기한 융합 폴리뉴클레오타이드 (융합 프로모터)를 갖도록 통상적인 유전공학적 방법으로 조작된 것일 수 있다. 상기 벡터는 상기 프로모터 이외의 전사 조절 서열 (예컨대 인핸서 등)을 추가로 포함할 수 있다.
본 발명에 있어서, ”발현 벡터”는 목적하는 숙주세포에서 목적 펩티드를 발현할 수 있는 재조합 벡터로서, 유전자 삽입물이 발현되도록 작동하게 연결된 필수적인 조절 요소를 포함하는 유전자 제작물을 의미한다. 상기 발현 벡터는 개시 코돈, 종결 코돈, 프로모터, 오퍼레이터 등의 발현조절 요소들을 포함하는데, 상기 개시 코돈 및 종결 코돈은 일반적으로 폴리펩티드를 암호화하는 뉴클레오티드 서열의 일부로 간주되며, 유전자 제작물이 투여되었을 때 개체에서 반드시 작용을 나타내야 하며 코딩 서열과 인프레임(in frame)에 있어야 한다. 벡터의 프로모터는 구성적 또는 유도성일 수 있다. 자체적으로 발현되는데 필요한 모든 요소를 포함하는 유전자 구조체인 발현 카세트 (expression cassette)의 형태로 숙주세포에 도입될 수 있다. 상기 발현 카세트는 통상 발현될 유전자 삽입물에 작동 가능하게 연결되어 있는 프로모터 (promoter), 전사 종결신호, 리보좀 결합부위 및 번역 종결신호를 포함할 수 있다. 상기 발현 카세트는 자체 복제가 가능한 발현 벡터 형태일 수 있다. 본 발명에서 상기 발현 벡터는 바이러스성 또는 비바이러스성 벡터일 수 있으며, 상기 바이러스성 벡터는 아데노바이러스 벡터, 렌티바이러스를 포함하는 레트로바이러스 벡터, 아데노-부속 바이러스 벡터 또는 헤르페스 심플렉스 바이러스 벡터 등일 수 있으나, 이에 한정되지 않는다. 또한, 상기 비바이러스성 벡터로는 플라스미드 벡터, 박테리오파지 벡터, 리포솜, 세균인공염색체, 효모인공염색체 등일 수 있으나, 이에 한정되지 않는다.
본 발명에서 상기 발현 벡터에서 상기 목적 유전자는 상기 융합 폴리뉴클레오타이드에 작동 가능하게 연결될 수 있다. 용어 "작동 가능하게 연결된(operatively linked)"은 유전자 발현 조절 서열과 다른 뉴클레오타이드 서열 사이의 기능적인 결합을 의미한다. 상기 유전자 발현 조절 서열은 "작동 가능하게 연결(operatively linked)"됨으로써 다른 뉴클레오타이드 서열의 전사 및/또는 해독을 조절할 수 있다. 상기 발현 벡터에 있어서, 상기 융합 폴리뉴클레오타이드가 상기 목적 유전자에 작동 가능하게 연결되기 위해서, 상기 융합 폴리뉴클레오타이드는 상기 목적 유전자의 5' 말단에 연결된 것일 수 있다. 본 발명의 발현 벡터는, 발현시키고자 하는 목적 단백질의 암호화 유전자가 작동 가능하게 연결될 경우, 적절한 숙주 세포에서 상기 목적 단백질을 높은 효율로 발현시킬 수 있는 목적 단백질의 발현 벡터로 사용될 수 있다.
본 발명의 상기 발현 벡터는 면역증강제로 IL-7를 코딩하는 유전자(서열번호 298) 및 IL-33를 코딩하는 유전자(서열번호 299) 중 적어도 하나, 바람직하게는 IL-33을 코딩하는 유전자를 추가로 포함할 수 있다.
본 발명의 상기 발현 벡터는 전사 조절 서열을 추가로 포함할 수 있다. 상기 전사 조절 서열은 폴리아데닐화 서열(pA)과 같은 전사 종결 서열; f1 복제원점, SV40 복제원점, pMB1 복제원점, 아데노 복제원점, AAV 복제원점, BBV 복제원점 등의 복제 원점; 번역(translation) 과정의 개시점(ATG)에서 라이보좀의 인식률을 높임으로써 유전자 발현을 높일 가능성이 높다고 알려진 코작 서열(Kozak sequence, AACAATGGC); 또는 IgE 선도 서열(leader 서열) 등으로 이루어진 군에서 선택된 1종 이상일 수 있으나, 이에 제한되는 것은 아니다.
본 발명의 상기 발현 벡터는 제한효소 절단 부위를 추가로 더 포함할 수 있다. 상기 제한효소 절단 부위는 제한효소에 의해 특이적으로 인식되어 절단되는 특정한 염기 서열을 지칭한다. 상기 절단 부위는 예를 들면, EcoRⅠ, BamHⅠ, HindⅢ, kpnⅠ, SalⅠ, NotⅠ, NcoⅠ, PstⅠ, SmaⅠ, 및 Xho I과 같은 제한효소에 의해 특이적으로 인식되는 서열일 수 있다.
또한, 본 발명에서 상기 발현 벡터는 선별 마커를 추가로 포함할 수 있다. 상기 선별 마커는 발현 벡터가 숙주 세포 내에 성공적으로 도입되었는지 여부를 확인하거나 안정적인 세포주 구축을 위한 유전자로서, 예컨대, 항생제와 같은 약물 저항 유전자, 대사 관련 유전자, 유전자 증폭 유전자 등으로 이루어진 군에서 선택된 1종 이상일 수 있다.
본 발명의 상기 발현 벡터는 상기 재조합 DNA와 함께 IL-7를 코딩하는 유전자(서열번호 298)를 포함할 수 있고, 이때 상기 발현 벡터(바람직하게, 발현 플라스미드)는 제한효소 절단 부위로 BamHI, NcoI, 및 NotI로 이루어진 군으로부터 선택된 어느 하나 이상을 포함할 수 있으며, 일 예시로 상기 발현 벡터는 상기 재조합 DNA와 함께 서열번호 300으로 표시되는 염기 서열을 포함할 수 있다.
본 발명의 상기 발현 벡터는 상기 재조합 DNA와 함께 IL-33를 코딩하는 유전자(서열번호 299)를 포함할 수 있고, 이때 상기 발현 벡터(바람직하게, 발현 플라스미드)는 제한효소 절단 부위로 BamHI, NcoI, 및 NotI로 이루어진 군으로부터 선택된 어느 하나 이상을 포함할 수 있으며, 일 예시로 상기 발현 벡터는 상기 재조합 DNA와 함께 서열번호 301로 표시되는 염기 서열을 포함할 수 있다.
본 발명에서 상기 발현 벡터는 숙주 세포에서 발현 가능한 것일 수 있다. 예를 들어, 상기 숙주 세포는 동물 세포에서 전사 개시를 강력하게 유도할 수 있는 것일 수 있으며, 구체적으로 포유동물 세포, 예를 들어 인간으로부터 유래된 세포와 같은 동물세포에서 전사 개시를 유도하는 것일 수 있다.
본 발명의 상기 발현 벡터는 당업계에 공지된 다양한 방법을 통해 구축될 수 있다.
본 발명의 또 다른 구현 예에 따르면, 본 발명에서 제공하는 발현 벡터가 숙주세포에 도입되어 형질 전환된 형질 전환체에 관한 것이다.
본 발명에서 상기 발현 벡터의 세포 내로의 운반(도입)은, 당업계에 널리 알려진 운반 방법을 사용할 수 있다. 상기 운반 방법은 예컨대, 미세 주입법, 칼슘 포스페이트 침전법, 전기 천공법(electroporation), 초음파 천공법(sonoporation), 자기장을 이용한 자기주입법(magnetofection), 리포좀-매개 형질감염법, 유전자 밤바드먼트 (gene bombardment), 덴드리머 및 무기(inorganic) 나노 입자의 사용 등을 사용할 수 있으나, 이에 제한되는 것은 아니다.
상술한 발현 벡터를 세포에 형질전환시켜 형질전환체를 제조할 수 있다.
본 발명에서 "형질전환체"란 프로모터와 작동 가능하게 연결되며, 유용물질을 코딩하는 DNA 서열로 이루어지는 DNA 구조물(DNA construct)에 의해 형질전환된 세포 또는 식물체를 의미한다. 본 발명에서 형질전환체는 형질전환된 미생물, 동물세포, 식물세포, 형질전환된 동물 또는 식물체 및 이들로부터 유래된 배양세포 등을 포함하는 의미이다.
본 발명의 또 다른 구현 예에 따르면, 본 발명은 상기한 바와 같은 항원용 조성물; 상기한 바와 같은 백신; 상기한 바와 같은 발현 벡터; 또는 상기한 바와 같은 형질 전환체의 유효량을 개체에게 투여하는 단계를 포함하는, 중증 열성 혈소판 감소 증후군(SFTS) 바이러스 감염에 대한 예방 또는 치료 방법을 제공한다.
본 발명의 또 다른 구현 예에 따르면, 본 발명은 상기한 바와 같은 항원용 조성물; 상기한 바와 같은 백신; 상기한 바와 같은 발현 벡터; 또는 상기한 바와 같은 형질 전환체를 유효 성분으로 포함하는, 중증 열성 혈소판 감소 증후군(SFTS) 바이러스 감염에 대한 예방 또는 치료용 약학 조성물을 제공한다.
본 발명에 있어서, 상기 약학 조성물은 캡슐, 정제, 과립, 또는 주사제, 연고제, 분말 또는 음료 형태임을 특징으로 할 수 있으며, 상기 약학 조성물은 인간을 대상으로 하는 것을 특징으로 할 수 있다.
본 발명의 약학 조성물은 이들로 한정되는 것은 아니지만, 각각 통상의 방법에 따라 산제, 과립제, 캡슐, 정제, 수성 현탁액 등의 경구형 제형, 외용제, 좌제 및 멸균 주사용액의 형태로 제형화하여 사용될 수 있다. 본 발명의 약학 조성물은 약제적으로 허용 가능한 담체를 포함할 수 있다. 약제학적으로 허용되는 담체는 경구 투여 시에는 결합제, 활탁제, 붕해제, 부형제, 가용화제, 분산제, 안정화제, 현탁화제, 색소, 향료 등을 사용할 수 있으며, 주사제의 경우에는 완충제, 보존제, 무통화제, 가용화제, 등장제, 안정화제 등을 혼합하여 사용할 수 있으며, 국소투여용의 경우에는 기제, 부형제, 윤활제, 보존제 등을 사용할 수 있다. 본 발명의 약제학적 조성물의 제형은 상술한 바와 같은 약제학적으로 허용되는 담체와 혼합하여 다양하게 제조될 수 있다. 예를 들어, 경구 투여시에는 정제, 트로키, 캡슐, 엘릭서(elixir), 서스펜션, 시럽, 웨이퍼 등의 형태로 제조할 수 있으며, 주사제의 경우에는 단위 투약 앰플 또는 다수회 투약 형태로 제조할 수 있다. 기타, 용액, 현탁액, 정제, 캡슐, 서방형 제제 등으로 제형할 수 있다.
한편, 제제화에 적합한 담체, 부형제 및 희석제의 예로는, 락토즈, 덱스트로즈, 수크로즈, 솔비톨, 만니톨, 자일리톨, 에리스리톨, 말디톨, 전분, 아카시아 고무, 알지네이트, 젤라틴, 칼슘 포스페이트, 칼슘 실리케이트, 셀룰로즈, 메틸 셀룰로즈, 미정질 셀룰로즈, 폴리비닐피롤리돈, 물, 메틸하이드록시벤조에이트, 프로필하이드록시벤조에이트, 탈크, 마그네슘 스테아레이트 또는 광물유 등이 사용될 수 있다. 또한, 충진제, 항응집제, 윤활제, 습윤제, 향료, 유화제, 방부제 등을 추가로 포함할 수 있다.
본 발명에 따른 약학 조성물의 투여 경로는 이들로 한정되는 것은 아니지만 구강, 정맥내, 근육내, 동맥내, 골수내, 경막내, 심장내, 경피, 피하, 복강내, 비강내, 장관, 국소, 설하 또는 직장이 포함된다. 경구 또는 비경구 투하가 바람직하다.
본 발명에서, "비경구"는 피하, 피내, 정맥내, 근육내, 관절내, 활액낭내, 흉골내, 경막내, 병소내 및 두개골내 주사 또는 주입기술을 포함한다. 본 발명의 약학 조성물은 또한 직장 투여를 위한 좌제의 형태로 투여될 수 있다.
본 발명의 약학 조성물은 사용된 특정 화합물의 활성, 연령, 체중, 일반적인 건강, 성별, 정식, 투여시간, 투여경로, 배출율, 약물 배합 및 예방 또는 치료될 특정 질환의 중증을 포함한 여러 요인에 따라 다양하게 변할 수 있고, 상기 약학 조성물의 투여량은 환자의 상태, 체중, 질병의 정도, 약물 형태, 투여경로 및 기간에 따라 다르지만 당업자에 의해 적절하게 선택될 수 있고, 1일 0.0001 내지 50mg/kg 또는 0.001 내지 50mg/kg으로 투여할 수 있다. 투여는 하루에 한번 투여할 수도 있고, 수회 나누어 투여할 수도 있다. 상기 투여량은 어떠한 면으로든 본 발명의 범위를 한정하는 것은 아니다. 본 발명에 따른 의약 조성물은 환제, 당의정, 캡슐, 액제, 겔, 시럽, 슬러리, 현탁제로 제형될 수 있다.
본 발명에서 제공하는 SFTS 바이러스의 재조합 DNA 또는 펩티드 및 이를 포함하는 SFTS 바이러스 백신은 개체에서 SFTS 바이러스에 대한 면역 반응을 효과적으로 유발하여, SFTS 바이러스 감염에 대한 예방 및 치료 효과가 우수하다.

본 발명의 상기 항원용 조성물 또는 백신은 면역증강제를 추가로 포함할 수 있고, 이때 상기 면역증강제는 IL-7 및 IL-33 중 적어도 하나일 수 있고, 바람직하게는 IL-33일 수 있지만, 이에 한정되는 것은 아니다.
이하, 본 발명을 하기의 실시예에 의해 상세히 설명한다. 단, 하기 실시예는 본 발명을 예시하는 것일 뿐, 본 발명의 내용이 하기 실시예에 의해 한정되는 것은 아니다.
SFTS 바이러스 항원 5종 및 면역증강제 2종 유전자 합성
목적유전자 (SFTSV 항원 또는 면역증강제 유전자) 5' 말단에 IgE 리더 및 코작(kozak) 서열을 삽입하였고, 3' 말단에 종결코돈을 삽입하였다. 최종적으로 유전자의 양끝에 제한효소 서열 (5' BamH I과 3' Not I)을 삽입한 후 유전자의 합성을 진행하였다.
고효율 backbone 플라스미드 (pGX0001) 클로닝
삽입유전자의 합성을 완료한 후, BamH I과 Not I으로 절단하고, 같은 제한효소로 절단된 고효율 backbone 플라스미드 (pGX0001)에 삽입하여 후보물질 플라스미드를 제작하였다(도 33 내지 도 39 참고). 유전자 합성 및 클로닝 결과는 염기서열 분석을 통해 확인하였다.
체내 항원 고발현 DNA 백신 서열 최적화
최적의 항원 5종으로 Gc(glycoprotein C), Gn(glycoprotein N), NP(nucleocapsid protein), NS(non-structural protein), RdRp(RNA dependent RNA polymerase))와, 최적의 면역증강제 2종으로 IL-7 및 IL-33을 선정하였다. 상기 항원 5종에 대해 한국, 중국, 일본 사람에서 분리된 SFTS 바이러스 균주 27~32개로부터 도출된 공통 서열을 확보하였다. 공통 서열은 다양한 SFTS 바이러스 서브타입 및 변종 바이러스의 공통적인 항원 아미노산 서열을 이용하여 교차면역능이 있는 범용 항원 서열로 디자인하였다. In silico 면역정보학 기법을 활용하여 다양한 서브타입 및 변종 SFTS 바이러스에 존재하는 인간의 주요 MHC 클래스 I 및 II 에피톱을 발굴하고, 이를 항원에 포함하도록 서열을 디자인하였다. 이후, 최적화된 SFTS 바이러스 항원 아미노산 서열을 바탕으로 DNA 백신을 위한 염기서열을 최종적으로 도출하였다.
이하의 실험에서 사용되는 본 발명의 일 실시예에 따르는 발현 카세트 구조는 고발현 프로모터(플라스미드 backbone 서열), 코작(kozak) 서열, IgE 선도(leader) 서열, poly A 신호 서열 (플라스미드 backbone 서열)을 포함하도록 하였고, 이때 체내에서의 유전자 발현율을 높이기 위해 kozak, IgE leader 서열을 목적 유전자 (SFTSV 항원 또는 면역증강제) 앞에 삽입하였다. 한편, 체내에서의 항원 유전자 발현율을 높이기 위해 SFTS 바이러스 항원 5종(Gc, Gn, NP, NS, RdRp)의 서열을 인간 코돈으로 최적화하였다.

목적 유전자 (SFTSV 항원 또는 면역증강제 유전자) 5' 말단에 IgE 선도 서열과 코작 서열을 삽입하였고, 3' 말단에 스톱 코돈을 삽입하였다. 최종적으로 유전자의 양끝에 제한효소 서열 (5' BamH I과 3' Not I)을 삽입한 후 유전자의 합성을 진행하였다. 삽입 유전자의 합성을 완료한 후, BamH I과 Not I로 절단하고, 같은 제한효소로 절단된 고효율 backbone 플라스미드(pGX0001)에 삽입하여 후보물질 플라스미드를 제작하였다. 유전자 합성 및 클로닝 결과는 염기서열 분석을 통해 확인하였다.
마우스 모델을 이용한 SFTS 바이러스 항원 발현 DNA 백신의 면역원성 평가
면역원성 평가용 OLP(overlapping peptide) 풀을 제작하였다. 구체적으로는 SFTS 바이러스의 항원 5종에 대한 면역원성을 평가하고자, 각 항원의 서열을 8개의 아미노산이 중복되도록 하여 15mer의 펩티드로 단편화하였다. 각 펩티드는 제작 과정에서 모두 고성능액체크로마토그래피와 전자분무 질량분석기를 이용하여 정성, 정량하여 순도를 확인하였다. 이 과정을 통하여 Gn 항원에서 총 76개의 펩티드를 확보하여, 이 펩티드를 38개씩 혼합하여 Gn에 대한 OLP1, 및 OLP2(각 펩티드 25ug/ml)를 제조하였고(표 1), Gc 항원에서는 총 76개의 펩티드를 제작하여, 이 펩티드를 38개씩 혼합하여 OLP3, 및 OLP4를 제조하였다(표 2). NP 항원에서는 총 34개의 펩티드를 확보하고 이를 혼합하여 OLP5를 제조하였고(표 3), NS 항원에서는 총 41개의 펩티드를 얻어 이를 혼합하여 OLP6를 제조하였으며(표 4), RdRp 항원에서는 총 58개의 펩티드를 29개씩 혼합하여 OLP7, 및 OLP8를 제조하였다(표 5).
보다 상세하게는, 표 1 내지 표 5에 나타내는 바와 같이, 서열번호 1 내지 서열번호 38을 혼합한 것인 OLP1이다. 또한, 서열번호 39 내지 서열번호 76을 혼합한 것인 OLP2이다. 서열번호 77 내지 서열번호 114를 혼합한 것인 OLP3이다. 서열번호 115 내지 서열번호 152를 혼합한 것인 OLP4이다. 서열번호 153 내지 서열번호 186을 혼합한 것인 OLP5이다. 서열번호 187 내지 서열번호 227을 혼합한 것인 OLP6이다. 서열번호 228 내지 서열번호 256을 혼합한 것인 OLP7이다. 서열번호 257 내지 서열번호 285을 혼합한 것인 OLP8이다.
이렇게 제작된 OLP 풀은 이하의 T세포의 면역 반응 평가에 사용하였다.

SFTS 바이러스 항원 5종 및 면역증강제 2종의 면역원석 검증
BALB/c 마우스를 이용하여, 나이브 그룹, DNA 백신을 근육 내 주사 (intramuscular injection)한 그룹, DNA 백신의 근육 내 주사 후 전기천공법 (electroporation)을 실시한 그룹, DNA 백신과 IL-7 면역증강제를 근육 내 주사 후 전기천공법을 실시한 그룹, DNA 백신과 IL-33 면역증강제를 근육 내 주사 후 전기천공법을 실시한 그룹, 총 5가지의 그룹으로 나누어 각 그룹당 6마리씩 접종을 진행하였다. 나이브 그룹의 각 마우스에는 SFTS 바이러스의 유전자가 삽입되지 않은 플라스미드를 200 ug로 접종하였고, 다른 4개의 그룹의 마우스에는 SFTS 바이러스 항원 발현 DNA 5종(Gn, Gc, NP, NS, 및 RdRp의 DNA 서열, 각각의 DNA 서열번호는 286, 288, 290, 292, 및 294이며, 아미노산 서열은 각각 287, 289, 291, 293, 및 295이다.)을 각 40 ug씩 총 200 ug이 되도록 접종하였다. 또한, IL-7과 IL-33 면역증강제를 접종하는 그룹의 마우스에는 추가적으로 DNA 접종과 함께 각 면역증강제를 50 ug씩 접종하였다. 나이브 그룹과 DNA 백신을 근육 내 주사한 그룹을 제외한 3개의 그룹의 마우스에는 근육 내 주사 직후 접종 부위에 전기천공기를 사용하여 0.2A로 전기천공법을 실시하였다. 첫 번째 접종 21일 후 동량으로 두 번째 접종을 진행하였으며, 두 번째 접종 21일 후 마우스를 희생시켜 비장과 서혜 림프절을 분리하여 면역원성 평가에 사용하였다.
SFTSV 백신 후보물질의 T세포 면역 반응 검증
SFTS 바이러스에 특이적인 T세포 반응을 측정하기 위해 ELISpot 분석을 진행하였다(도 1). 96-웰 여과 플레이트에 PBS로 희석한 항-인간 IFN-γ 항체(2μg/ml; endogen)를 각 웰당 100 ul씩 분주해주고 4℃에서 밤새 인큐베이션한 후, 마우스의 비장세포(5×105cells/well)를 SFTS 바이러스의 펩티드로부터 제작한 8종의 OLP로 자극을 주며 37℃에서 24시간 인큐베이션을 실시하였다. 이 후 플레이트를 세척하여 PBS/Tween20/1%BSA로 희석한 비오티닐화된 항-인간 IFN-γ 항체(0.5 μg/ml; endogen)를 각 웰당 100 ul씩 분주해주고 4℃에서 밤새 인큐베이션하였다. 4번의 세척후, PBS/Tween20/1%BSA에 5000:1로 희석한 스트렙타아비딘-알칼리성 인산가수 분해효소(BD)를 각 웰당 100 ul씩 분주해주고 37℃에서 1시간 인큐베이션하였다. AP 접합 기질 키트(AP conjugate substrate kit, BIO-RAD사)를 사용하여 10분 동안 반응시키고 나서, 세척하여 반응을 멈춘 후 ImmunoSpot(Cellular Technology Limited)을 통해 SFTS 바이러스 항원에 따른 SFTS 바이러스에 특이적인 T세포의 IFN-γ 생성을 관찰하였다. 이를 통해 DNA 백신 접종 후 마우스 모델에서 SFTS 바이러스에 특이적인 T세포의 면역 반응이 성공적으로 유도되었음을 확인하였다. 이러한 면역 반응은 DNA 백신의 근육 내 주사 후 전기천공법(electroporation)을 실시한 그룹과 추가적으로 IL-33 면역증강제를 함께 주사한 그룹에서 뚜렷하게 관찰할 수 있었으며, 특히 IL-33 면역증강제를 SFTS 바이러스 항원 발현 DNA 백신과 함께 주사한 그룹에서 확연하게 T 세포의 면역 반응이 증가하는 것을 확인하였다(도 2a 내지 도 2e).
SFTSV DNA 백신 후보물질의 T세포 기능 다기능성 평가
백신 접종이 완료된 쥐에서 수득한 비장 세포에 SFTS 바이러스 OLP 자극을 준 후, 다색 FACS를 이용한 세포내 사이토카인 염색법(intracellular cytokine staining; ICS)으로 분석하였다.
표 1 내지 표 5에 나타내는 바와 같은 8종류의 SFTS 바이러스 OLP로 각각 자극을 주고 IFN-γ, TNF-α, IL-2를 분비하는 T세포 서브세트를 변환하였다. 그 결과 각각의 사이토카인을 합성하는 T세포의 비율을 확인하여 도 3a 내지 도 3c에 나타내었다.
도 3a 내지 도 3c에서 보는 바와 같이, 전반적으로 근육내 주사만 실시한 그룹(IM)보다 전기천공법(electroporation)을 병행한 그룹(IMEP)에서 더 높은 면역 반응이 나오는 것을 확인하였다. 또한 전기천공법과 더불어 면역증강제로 IL-33을 사용한 그룹(IL-33)에서 가장 강한 면역 반응이 나오는 경향성이 있음을 확인하였다. 이러한 경향성은 CD8+ T세포에서 더욱 잘 확인되었다(도 3a 내지 도 3c).
SFTS 바이러스의 NS 단백질에 해당하는 OLP인 OLP6에서 가장 강한 면역 반응이 유도되는 것을 확인했으며, 특히 IFN-γ와 TNF-α에서 매우 강한 반응이 나타남을 확인하였다. 더불어, IM 그룹보다 IMEP 그룹이 더 강한 반응을 보이고, IL-33을 면역증강제로 사용했을 때 가장 강한 면역 반응이 유도되는 것을 OLP6를 처리한 CD8+ T세포에서 매우 명확하게 확인할 수 있었다.
이는 전기천공법과 IL-33을 사용할 경우 백신에 의해 유도되는 SFTS 바이러스 특이 T세포의 비율이 더욱 증가된다는 것을 의미하였다.
백신 후보물질에 의해 유도되는 T세포의 다기능성 분석
FACS 데이터를 기반으로 각 그룹에서 T세포의 다기능성을 분석하였다. FACS에서의 결과를 토대로, 면역 반응이 가장 강하게 유도되는 OLP6 자극을 받은 CD8+ T세포의 다기능성을 그룹 별로 정리하여 결과를 얻었다. IM 그룹에 비해 IMEP 그룹이 다기능 T세포의 비율이 더 높고, 전기천공법과 더불어 면역증강제로 IL-33을 사용한 그룹에서 그 비율이 가장 높아지는 결과를 나타내었다. 이는 백신에 의해 유도된 T세포 면역 반응이 전기천공법과 IL-33을 사용할 경우 질적으로도 더 좋아진다는 것을 의미하였다. 이러한 경향성은 전체 작동 T세포 중 다기능성 T세포의 비율에서도 유사한 것으로 나타났다(도 4). 또한, CD4+ T세포의 경우 다기능성에서 큰 차이를 보이지 않았다.
SFTSV 백신 후보물질의 항체 형성능 평가
백신에 의해 유도된 SFTSV 특이적 항체 생성 반응 검증
SFTSV 백신에 의해 유도되는 SFTS 바이러스에 특이적인 항체 생성 반응을 측정하기 위해 ELISA(enzyme-linked immunosorbent assay) 어세이 기법을 실시하였다. 재조합 SFTSV NP 항원 단백질에 대한 ELISA 기법을 확립하기 위해 백신을 통해 면역원성을 갖추었다고 평가되었던 마우스의 혈청을 이용해 실험을 진행하였다. 상기한 바와 같이 ELISA 어세이 기법을 이용하여, 백신에 의해 유도된 항체면역 반응을 정량적으로 확인하였다. 도 5a 내지 도 5f에 나타난 바와 같이, 근육주사 + 전기천공법(IMEP) 그룹의 마우스에서 다른 그룹에 비해 강한 항체 면역 반응이 생성되었음을 확인하였다. 도 5a 내지 도 5f에서, CrMN은 chromium precursor를 이용하여 MN(미세침) 표면을 처리하여 미세침 표면에 nano-pattern을 형성시킨 미세침을 사용하였다.
SFTSV DNA 백신에 의해 유도된 중화항체 효능의 정량 평가
DNA 백신 후보물질과 다양한 면역증강제에 의해 마우스에서 생산된 33개 항체의 중화항체가를 PRNT50 방법으로 측정하였다.
본 발명자들이 개발한 마우스 표준항체를 양성대조군으로 사용하였고, 실험 결과 실험군 개체별로 20~160 SN 적정농도(titer) 값을 나타내어 중화항체가 형성된 것을 확인하였다. 특히, 근육주사 + 전기천공 기법으로 DNA 백신을 주입한 그룹(IMEP)에서 보다 강한 중화항체 반응이 관찰되었으며, 미세침 그룹(Microneedle)에서는 매우 약한 중화항체 효능이 검출되었다(도 6).
SFTSV 감염 동물모델로써의 중형동물 모델의 확립
SFTSV 백신의 감염억제 효능을 검증하기 위한 새로운 동물 모델을 개발하였다. 중형동물에 SFTS 환자에게서 분리한 SFTSV를 감염시킨 결과, 혈액 내 바이러스가 검출되며, 감염 8일까지 혈소판 수가 계속 감소하는 것으로 관찰되었다(도 7). 또한, 체온이 4일째 2℃ 이상 증가하고, 이러한 증상이 지속됨에 따라 감염 후 9일 전, 후로 감염된 개체가 모두 폐사하는 것을 확인하였다. 이러한 결과는 환자의 임상 경과와도 매우 유사하였다. SFTSV 감염 후 회복기에 들지 못하면 약 10일 가량 이후 사망하는 양상을 보였다. 본 발명자들에 의해 확립된 중형동물 모델은 SFTSV 감염시 고열, 바이러스 수치(viral load) 증가, 혈소판 및 혈액내 구성성분 변화 등 SFTS 환자와 매우 유사한 임상적 소견을 보이기 때문에, 백신 효능 검증을 위한 SFTSV 감염 동물 모델로써 뛰어난 적합성을 보인다고 판단하였다.
SFTSV 감염 동물 모델을 이용한 SFTSV 백신의 면역원성 규명
본 발명자들은 확립한 SFTSV 감염 중형동물 모델을 SFTSV 백신의 감염억제 효능을 검증하는 모델로 사용하였다. 백신처리군(N = 6)에는 SFTS 바이러스 항원 발현 DNA 5종을 각 200 ug씩 총 1mg이 되도록 혼합(표 6 참고)하여 양쪽 대퇴 부위에 500ug씩 피 내 주사로 접종하였다. 대조군에는 SFTS 바이러스의 유전자가 삽입되지 않은 mock 플라스미드(pVax-1 유래) 1mg을 양쪽 대퇴 부위에 500ug씩 피 내 주사로 접종하였다. 두 그룹 모두 피내투여 직후 전기천공기를 사용하여 0.2A로 전기천공법을 실시하였다. 백신은 2주 간격으로 총 5회 투여하였다(0일차, 14일, 28일, 42일, 및 56일차에 투여).

SFTSV 감염 중형동물 모델에서 백신에 의해 유도된 T세포 면역 반응 평가
백신 투여 전, 2회 / 4회 / 5회 백신 투여 각 2주 후에 5ml의 혈액을 채취하고, PBMC 및 혈청을 분리하여 SFTSV-특이 T세포 면역 반응(ELISpot 어세이) 및 항체 면역 반응(ELISA 및 중화항체 어세이)을 측정하였다. 백신에 의해 형성된 SFTSV-특이 T세포 면역 반응을 ELISpot 분석법을 통해 평가하였다. 도 8a 내지 도 8d에 나타내는 바와 같이, SFTSV 백신 후보물질에 의해 중형동물에서 매우 강한 SFTSV-특이 T세포 면역 반응이 유도되는 것을 확인하였으며, 2회 투여 이후 안정적인 면역 반응이 관찰되는 것을 확인하였다.
SFTSV 감염 중형동물 모델에서 백신에 의해 유도된 항체 면역 반응 및 중화항체 유도능 정량 평가
DNA 백신에 의해 형성된 SFTSV 특이 반응 항체 형성을 ELISA 방법으로 측정 평가하였다. 도 9에 나타내는 바와 같이, 공벡터 백신을 접종한 6마리의 MOCK 그룹의 경우 음성 대조군 혈청과 비슷한 정도의 ELISA 값을 보여 특이 항체가 형성되지 않은 것으로 판단되었다. 하지만, 다른 6마리의 SFTSV DNA 백신 접종 그룹에서 생산된 SFTSV 특이 반응 항체가 양성 대조군 혈청에서와 비슷하거나 더 높게 나타나는 것을 관찰하였다. 이러한 결과는 백신 후보물질이 중형동물에서 항체 면역 반응을 효과적으로 유도할 수 있음을 보여주는 결과이다.
DNA 백신에 의해 유도된 항체의 중화항체가를 PRNT50 방법으로 측정하였다. 도 10에 나타내는 바와 같이 SFTSV DNA 백신을 접종한 6마리의 동물에서 생산된 중화항체 역가는 양성 대조군과 비슷한 정도로 나타나는 것을 확인하였다. 이에 비해, 공벡터 백신을 접종한 6마리 백신 그룹에서는 중화항체가 전혀 생성되지 않은 것을 확인할 수 있었다. 이러한 결과는 DNA 백신 후보물질에 의해 SFTSV-특이 중화항체가 효과적으로 유도될 수 있음을 의미하였다.
SFTSV 감염 중형동물 모델에서 SFTSV 예방 DNA 백신의 방어 효능 검증
백신의 방어 효능을 검증하기 위해, SFTSV 백신을 투여한 개체에 치사량(lethal dose)의 SFTSV를 감염시 킨 후 생존률, SFTSV 바이러스 수치, 혈소판 수, 체온, 체중 변화 등과 같은 임상 증상을 평가하였다. 도 11에 나타내는 바와 같이, 생존률을 평가한 결과, 대조군에서 감염 이후 7일에 2마리, 8일에 3마리, 9일에 1마리 등 6마리의 대조군 개체 모두 폐사하였다. 이에 반해, SFTSV 백신을 투여한 개체는 6마리 모두 생존하였다.
실시간 PCR을 통해 SFTSV 바이러스 수치를 측정하였다. 도 12a 내지 도 12c에 나타내는 바와 같이, 대조군에서는 감염 후 2일차에 바이러스 수치가 증가하고 감염 4일차에 가장 높은 바이러스 수치를 보이는 것으로 관찰되었다. 이와 비교하여 백신 그룹의 4마리의 중형동물에서는 바이러스 수치가 측정되지 않았다. 1마리의 백신 그룹 개체에서 대조군과 유사한 정도의 바이러스 수치가 측정되었지만, 감염 4일차에 감소한 후 감염 6일차에 완전히 제거된 것을 확인하였다. 이 개체는 혈소판이 증가했던 개체와 같은 개체로 바이러스 수치가 감소함에 따라 혈소판 수치도 정상으로 회복된 것을 관찰하였다.
혈소판 수를 측정한 결과, 대조군의 급격한 혈소판의 감소가 SFTSV 감염에 의해 관찰되었다(도 13a 내지 도 13c). 이와 비교하여 백신 그룹의 혈소판 수치는 정상적으로 유지됨을 확인하였다. 백신 그룹의 한 개체에서 감염 후 4일차에 혈소판 수치가 약 120x103/ul까지 감소함을 관찰했지만, 감염 6일 후에 혈소판 수치가 정상으로 회복한 것을 확인하였다.
한편, 도 14a에 나타내는 바와 같이, SFTSV 감염 후 대조군의 개체들은 체중이 유의하게 감소하지만, 백신 그룹의 개체들은 체중이 전혀 감소하지 않음을 확인하였다. 또한, 도 14b에 나타내는 바와 같이, SFTSV 감염 후 대조군의 개체들은 체온이 약 2℃ 정도 증가하지만, 백신 그룹 개체들은 뚜렷한 체온 변화가 관찰되지 않았다.
이러한 결과를 종합하면, 본 발명자들이 개발한 SFTSV 예방 DNA 백신 후보물질은 중형동물 모델에서 다양한 임상 지표(생존률, 혈소판 수치, 체온, 체중) 검증을 통해 확인한 바와 같이 SFTSV 감염을 효과적으로 예방할 수 있었다.
마우스 및 중형동물에서 유도된 중화항체의 교차반응(cross-reactivity) 효능검사
SFTSV DNA 백신 투여 후 마우스에서 형성된 SFTSV 중화항체의 교차반응을 평가하기 위해 다른 SFTS 바이러스와의 PRNT50 테스트를 실시하였다. 도 15a에 나타내는 바와 같이, SFTSV/2014 바이러스에는 중화항체 역가가 40~80 정도 형성된 반면, 다른 바이러스인 SFTSV/2015에 대한 중화항체 형성이 뚜렷하게 나타나지 않았다.
또한, SFTSV DNA 백신 투여 후 중형동물에서 형성된 SFTSV 중화항체의 교차반응을 평가하기 위해 다른 SFTS 바이러스와의 PRNT50 테스트를 실시하였다. 도 15b에 나타내는 바와 같이, 중화항체 역가가 SFTSV/2014 바이러스에서 160~320 정도의 값으로 나타났으며, 다른 바이러스인 SFTSV/2015에 비해 2배 정도 높게 형성한 것을 확인하였다.
SFTSV 감염 중동물 모델에서 SFTSV 예방 DNA 백신의 면역원성 검증
상기한 바와 같이 확립한 SFTSV 감염 중형동물 모델을 사용하여 SFTSV 백신의 감염 억제 효능을 검증하였다. 각 백신의 방어 효능을 검증하기 위하여, 각각 Gn 및 Gc가 조합된 Gn/Gc, NP, NS, 및 RdRp에 대한 SFTSV 백신 처리군(각각 N = 4, N = 3, N = 3, N = 3)에는 SFTS 바이러스 항원 발현 DNA 5종을 각 200 ug씩 총 1mg이 되도록 혼합하여 양쪽 대퇴 부위에 피 내 주사로 접종하였다. 대조군에는 SFTS 바이러스의 유전자가 삽입되지 않은 mock 플라스미드(pVax-1 유래) 1mg을 양쪽 대퇴 부위에 500ug씩 피 내 주사로 접종하였다. 두 그룹 모두 피내투여 직후 전기천공기를 사용하여 0.2A로 전기천공법을 실시하였다. 백신은 2주 간격으로 총 3회 투여하였다(0일차, 14일, 28일에 투여).
SFTSV 감염 중형동물 모델에서 백신에 의해 유도된 T세포 면역 반응 평가
백신 투여 전, 및 3회 백신 투여 각 2주 후에 5ml의 혈액을 채취하고, PBMC 및 혈청을 분리하여 SFTSV-특이 T세포 면역 반응(ELISpot 어세이) 및 항체 면역 반응(ELISA 및 중화항체 어세이)을 측정하였다. 백신에 의해 형성된 SFTSV-특이 T세포 면역 반응을 ELISpot 분석법을 통해 평가하였다. SFTSV 백신 후보물질에 의해 중형동물에서 접종한 백신에 따라 각 SFTSV 항원에 대해 매우 강한 SFTSV-특이 T세포 면역 반응이 유도되는 것을 확인하였다. 도 16에 나타난 바와 같이, Gn/Gc SFTSV 백신 처리군에서 가장 우수한 SFTSV-특이 면역 반응을 확인할 수 있었다.
SFTSV 감염 중형동물 모델에서 백신에 의해 유도된 항체 면역 반응 및 중화항체 유도능 정량 평가
DNA 백신에 의해 유도된 항체의 중화항체가를 PRNT50 방법으로 측정하였다. 도 17에 나타난 바와 같이, SFTSV Gn/Gc에 대한 DNA 백신을 접종한 4마리의 동물에서 효과적으로 중화항체 역가가 생성되는 것을 확인하였다. 이에 비해, 공벡터 백신을 접종한 6마리 백신 그룹을 비롯하여 각각 NP, NS, 및 RdRp에 대한 백신을 접종한 그룹의 개체에서는 중화항체가 전혀 생성되지 않은 것을 확인할 수 있었다. 이러한 결과는 Gn/Gc에 대한 DNA 백신 후보물질에 의해 SFTSV-특이 중화항체가 효과적으로 유도될 수 있음을 의미하였다.
SFTSV 감염 중형동물 모델에서 SFTSV 예방 DNA 백신의 방어 효능 검증
각 백신의 방어 효능을 검증하기 위해, 각각 Gn/Gc, NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 개체에 치사량(lethal dose)의 SFTSV를 감염시 킨 후 생존률, SFTSV 바이러스 수치, 혈소판 수, 체온, 체중 변화, ALT, 및 AST 변화 등과 같은 임상 증상을 평가하였다. 도 18에 나타난 바와 같이, 생존률을 평가한 결과, 대조군에서 감염 이후 6마리의 개체 모두 폐사하였으며 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서 모두 3마리의 개체 중 1마리만이 생존하였다. 이에 반해, Gn/Gc에 대한 SFTSV 백신을 투여한 개체는 4마리 모두 생존하였다.
또한, 실시간 PCR을 통해 SFTSV 바이러스 수치를 측정하였다. 그 결과를 도 19 및 도 20에 나타내었다. 대조군에서는 감염 후 2일 차에 바이러스 수치가 증가하고 감염 6일차에 가장 높은 바이러스 수치를 보이는 것을 관찰하였다. 이와 비교하여 Gn/Gc백신 그룹의 4마리의 중형동물에서는 바이러스 수치가 측정되지 않았다. 1마리의 Gn/Gc 백신 그룹 개체에서 감염 후 2일차에 미량의 바이러스 수치가 측정되었지만, 감염 4일차에 완전히 제거된 것을 확인하였다. 반면 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서는 대조군의 바이러스 수치와 유사한 정도로 이가 증가함을 확인하였다.
또한, 도 21 및 도 22에 나타난 바와 같이, 혈소판 수를 측정한 결과, 대조군에서 SFTSV 감염에 의해 급격한 혈소판의 감소를 관찰하였다. 이와 비교하여 Gn/Gc백신 그룹의 혈소판 수치는 정상적으로 유지됨을 확인하였다. 반면 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서는 감염 6일차까지 혈소판 수치가 약 300x103/ul까지 감소함을 관찰하였다.
도 23 및 도 24에 나타난 바와 같이, 백혈구 수를 측정한 결과, 대조군에서 SFTSV 감염에 의해 급격한 백혈구의 감소를 관찰하였다. 이와 비교하여 Gn/Gc백신 그룹의 백혈구 수치는 정상적으로 유지됨을 확인하였다. 반면 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서는 감염 6 혹은 4 일차까지 백혈구 수치가 감소함을 관찰하였다.
한편, SFTSV 감염 후 대조군의 개체들은 약 80% 이하까지 체중이 유의하게 감소하지만, 백신 그룹의 개체들은 90% 이하의 체중 감소를 보이지 않음을 확인하였다(도 25 및 도 26 참고).
또한, 도 27 및 도 28에 나타난 바와 같이, SFTSV 감염 후 대조군의 개체들은 체온이 약 2℃ 정도 증가하지만, Gn/Gc 백신 그룹 개체들은 뚜렷한 체온 변화가 관찰되지 않았다. 반면 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서는 0.5-1℃ 정도 체온이 증가함을 관찰하였다.
도 29 및 도 30에 나타난 바와 같이, 혈청 내 ALT를 측정한 결과, 대조군에서 SFTSV 감염에 의해 급격한 ALT 수치의 증가를 관찰하였다. 이와 비교하여 Gn/Gc백신 그룹의 ALT 수치는 정상적으로 유지됨을 확인하였다. 반면 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서는 감염 6일차까지 ALT 수치가 급격하게 증가하는 것을 관찰하였다.
또한, 도 31 및 도 32에 나타난 바와 같이, 혈청 내 AST를 측정한 결과, 대조군에서 SFTSV 감염에 의해 급격한 AST 수치의 증가를 관찰하였다. 이와 비교하여 Gn/Gc백신 그룹의 AST 수치는 정상적으로 유지됨을 확인하였다. 반면 NP, NS, 및 RdRp에 대한 SFTSV 백신을 투여한 세 그룹에서는 감염 6일차까지 AST 수치가 급격하게 증가하는 것을 관찰하였다.
이로부터 Gn/Gc, NP, NS, 및 RdRp 각각 항원의 방어효능을 중형동물(패럿) 감염모델에서 검증한 결과, Gn/Gc DNA 백신이 다른 항원-발현 DNA 백신에 비해 월등이 높은 방어효능을 보이는 것을 확인할 수 있었다.
이상으로 본 발명의 특정한 부분을 상세히 기술하였는 바, 당업계의 통상의 지식을 가진 자에게 있어서 이러한 구체적인 기술은 단지 바람직한 구현 예일 뿐이며, 이에 본 발명의 범위가 제한되는 것이 아닌 점은 명백하다. 따라서, 본 발명의 실질적인 범위는 첨부된 청구항과 그의 등가물에 의하여 정의된다고 할 것이다.
본 발명은 중증 열성 혈소판 감소 증후군(Severe Fever with Thrombocytopenia Syndrome; SFTS) 바이러스 질환에 의한 감염병을 예방 또는 치료하기 위한 백신 조성물에 관한 것이다.

Claims (16)

  1. 서열번호 287로 표시되는 아미노산 서열을 포함하거나, 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA에 의해 코딩되는 재조합 제 1 펩티드;
    서열번호 289로 표시되는 아미노산 서열을 포함하거나, 서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA에 의해 코딩되는 재조합 제 2 펩티드;
    서열번호 291로 표시되는 아미노산 서열을 포함하거나, 서열번호 290으로 표시되는 염기서열을 포함하는 재조합 제 3 DNA에 의해 코딩되는 재조합 제 3 펩티드;
    서열번호 293으로 표시되는 아미노산 서열을 포함하거나, 서열번호 292로 표시되는 염기서열을 포함하는 재조합 제 4 DNA에 의해 코딩되는 재조합 제 4 펩티드; 및
    서열번호 295로 표시되는 아미노산 서열을 포함하거나, 서열번호 294로 표시되는 염기서열을 포함하는 재조합 제 5 DNA에 의해 코딩되는 재조합 제 5 펩티드로 이루어진 군에서 선택된 어느 하나 이상을 포함하는 재조합 펩티드를 유효성분으로 포함하는 항원용 조성물.
  2. 제 1 항에 있어서,
    상기 항원용 조성물은 근육내, 피내, 피하, 표피하, 경피, 또는 정맥내에서 선택된 경로를 통해 체내 주사되는, 항원용 조성물.
  3. 제 1 항에 있어서,
    상기 항원용 조성물은 근육 내 주사를 통해 대상체에 주입되는, 항원용 조성물.
  4. 제 1 항에 있어서,
    상기 항원용 조성물은 피 내 주사를 통해 대상체에 주입되는, 항원용 조성물.
  5. 제 2 항에 있어서,
    상기 항원용 조성물은 체내 주사 시 추가로 전기천공법을 이용하여 대상체에 주입되는, 항원용 조성물.
  6. 서열번호 287로 표시되는 아미노산 서열을 포함하거나, 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA에 의해 코딩되는 재조합 제 1 펩티드;
    서열번호 289로 표시되는 아미노산 서열을 포함하거나, 서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA에 의해 코딩되는 재조합 제 2 펩티드;
    서열번호 291로 표시되는 아미노산 서열을 포함하거나, 서열번호 290으로 표시되는 염기서열을 포함하는 재조합 제 3 DNA에 의해 코딩되는 재조합 제 3 펩티드;
    서열번호 293으로 표시되는 아미노산 서열을 포함하거나, 서열번호 292로 표시되는 염기서열을 포함하는 재조합 제 4 DNA에 의해 코딩되는 재조합 제 4 펩티드; 및
    서열번호 295로 표시되는 아미노산 서열을 포함하거나, 서열번호 294로 표시되는 염기서열을 포함하는 재조합 제 5 DNA에 의해 코딩되는 재조합 제 5 펩티드로 이루어진 군에서 선택된 어느 하나 이상을 포함하는 재조합 펩티드를 유효성분으로 포함하는 백신.
  7. 제 6 항에 있어서,
    상기 백신은 근육내, 피내, 피하, 표피하, 경피, 또는 정맥내에서 선택된 경로를 통해 체내 주사되는, 백신.
  8. 제 6 항에 있어서,
    상기 백신은 근육 내 주사를 통해 대상체에 주입되는, 백신.
  9. 제 6 항에 있어서,
    상기 백신은 피 내 주사를 통해 대상체에 주입되는, 백신.
  10. 제 7 항에 있어서,
    상기 백신은 체내 주사 시 추가로 전기천공법을 이용하여 대상체에 주입되는, 백신.
  11. 제 6 항에 있어서,
    상기 백신은 면역증강제를 추가로 포함하는, 백신.
  12. 제 11 항에 있어서,
    상기 면역증강제는 IL-7, 및 IL-33 중 적어도 하나인, 백신.
  13. 서열번호 286으로 표시되는 염기서열을 포함하는 재조합 제 1 DNA;
    서열번호 288로 표시되는 염기서열을 포함하는 재조합 제 2 DNA;
    서열번호 290으로 표시되는 염기서열을 포함하는 재조합 제 3 DNA;
    서열번호 292로 표시되는 염기서열을 포함하는 재조합 제 4 DNA; 및
    서열번호 294로 표시되는 염기서열을 포함하는 재조합 제 5 DNA로 이루어진 군에서 선택된 어느 하나 이상의 재조합 DNA를 포함하는 발현 벡터.
  14. 제 13 항의 발현 벡터가 숙주세포에 도입되어 형질 전환된 형질 전환체.
  15. 제 1 항 내지 제 5 항 중 어느 한 항의 항원용 조성물, 제 6 항 내지 제 12 항 중 어느 한 항의 백신, 제 13 항의 발현 벡터, 또는 제14항의 형질 전환체의 유효량을 개체에게 투여하는 단계를 포함하는, 중증 열성 혈소판 감소 증후군(SFTS) 바이러스 감염에 대한 예방 또는 치료 방법.
  16. 제 1 항 내지 제 5 항 중 어느 한 항의 항원용 조성물, 또는 제 6 항 내지 제 12 항 중 어느 한 항의 백신, 제 13 항의 발현 벡터, 또는 제14항의 형질 전환체를 유효 성분으로 포함하는, 중증 열성 혈소판 감소 증후군(SFTS) 바이러스 감염에 대한 예방 또는 치료용 약학 조성물.
