Abstract
본 발명은, 신규한 3-하이드록시프로피오네이트-락테이트의 블록공중합체[P(3HP-b-LA)]{Method for preparing poly(3-hydroxypropionate-b- lactate) block copolymer by using microorganisms} 및 그 제조방법에 관한 것으로, 더욱 상세하게는 락트산을 생합성하지 못하도록 개량한 재조합 대장균을 이용하여, 배양 초기에 글리세롤을 탄소원으로 하여 3-하이드록시프로피오네이트를 생산하는 유전자들과 개량된 PHA 신타아제를 통해 P(3HP)를 생합성하는 제1 배양단계; 및 탄소원으로 글리세롤과 글루코오스가 함께 공급시 글루코오스만 선택적으로 대장균 내로 유입시키는 탄소 이화대사억제 시스템(Carbon catabolic repression system)을 이용하여 P(3HP) 생산을 억제시키고, IPTG 유도 시스템을 통해 락테이트 생성 효소와 락틸-CoA 전환 효소가 발현되도록 하여 중단된 P(3HP) 말단에 폴리락테이트를 생합성시키는 제2 배양단계;를 포함하는 3-하이드록시프로피오네이트-락테이트의 블록 공중합체를 제조하는 방법 및 이에 의해 제조된 3-하이드록시프로피오네이트-락테이트의 블록 공중합체를 제공한다.
