体内のアンモニアは、主にタンパク質代謝または運動による筋肉の分解などにおけるアミノ酸の異化の過程で発生する。アンモニアは、ピルビン酸からアセチルCoAへの酸化を阻害して疲労を起こさせる(非特許文献1参照)。また、アンモニアが神経系の機能低下をもたらして中枢性疲労の原因になることや、筋肉での乳酸の蓄積を促進して筋肉疲労を促すこと(特許文献1)、および高アンモニア血症が脳障害などの様々な障害を引き起こすこと(非特許文献2)が知られている。慢性肝臓病患者におけるアンモニアの蓄積は、肝性脳症および多臓器不全(呼吸不全、心血管不全、腎不全)の進行に重要な役割を果たしていると考えられる(特許文献2)。
体内のアンモニアは、肝臓の尿素回路により無毒な尿素へと変換され、尿中に排出される。アルギニン、オルニチンおよびシトルリンは、アンモニアを無毒な尿素に変換する肝臓の尿素回路に関与している。尿素回路に関与するアミノ酸であるオルニチン、シトルリンまたはアルギニンにより血中アンモニアの上昇が抑制されることが報告されている(特許文献3、4、及び非特許文献1)。生体内ではオルニチンからシトルリンが合成され、シトルリンはアルギニンへと変換される。オルニチンにより血中アンモニアの上昇が抑制されることや、疲労が改善するという報告がある(特許文献3参照)。またシトルリン摂取により血中アルギニンが上昇することや、脳疲労が改善することが報告されている(特許文献4、5)。
持久力は運動競技において必要とされるだけでなく、我々の日常生活において、歩く、走る等の単純な動作や、筋肉労作を繰り返し行う労働においても必要不可欠なものである。持久力の低下は、特に高齢者において、日常動作の困難の原因となり、生活の質(quality of life:QOL)に重大な悪影響をもたらす。持久力の向上には運動トレーニングが有効であることは広く認識されているが、時間の確保やモチベーションの維持の困難さから、また高齢者においては怪我の恐れがあるため、トレーニングによる持久力の維持または向上は決して容易なことではない。
カテキンは、従来、持久力向上または疲労を防止する作用を有する成分として報告されている(特許文献6、7)。しかし、アンモニア代謝に対するカテキンの作用に関する報告はない。
その他の持久力向上または疲労を防止する作用を有する成分として、各種アミノ酸、例えば、オルニチン(特許文献3)、シトルリン(非特許文献1)、シトルリンとアルギニンの併用(特許文献4)、アルギニン、オルニチンおよびシトルリンの併用(非特許文献3)などが報告されている。

本発明は、カテキン類を有効成分とするアンモニア代謝促進剤を提供する。
また本発明は、カテキン類を有効成分とする高アンモニア血症の予防もしくは改善剤を提供する。
また本発明は、カテキン類を有効成分とする肝性脳症の予防もしくは改善剤を提供する。
また本発明は、カテキン類を有効成分とする慢性疲労症候群の予防もしくは改善剤を提供する。
さらに本発明は、カテキン類、シトルリンおよびアルギニンを有効成分とするアンモニア代謝促進剤を提供する。
さらに本発明は、カテキン類、シトルリンおよびアルギニンを有効成分とする持久力向上剤を提供する。

(1.定義)

本明細書において、「非治療的」とは、医療行為を含まない、すなわち人間を手術、治治療または診断する方法を含まない、より具体的には医師、または医療従事者もしくは医師の指示を受けた者が人間に対して手術、治療または診断を実施する方法を含まない概念である。
本明細書において、「予防」とは、個体における疾患もしくは状態の発症の防止、抑制または遅延、あるいは個体の疾患もしくは状態の発症の危険性を低下させることをいう。また本明細書において、「改善」とは、疾患もしくは状態の好転、疾患もしくは状態の悪化の防止、抑制または遅延、あるいは疾患もしくは状態の進行の逆転、防止、抑制または遅延をいう。
本明細書において、「運動」とは、例えば、スポーツ、トレーニング、有酸素運動等の運動競技(狭義の運動)、および筋肉労作を伴う労働、日常の動作、等を含む広義の身体運動をいう。本明細書においては、「運動」は、アスリートまたは狭義の運動を行う者に関して使用される場合、好ましくは狭義の運動を意味し得、一方「運動」が中高年、高齢者、病弱者、疾病者、疾病からの回復期にある者に関して使用される場合、好ましくは筋肉労作を伴う労働または日常の動作を意味し得る。
本明細書において、「持久力」とは、上記「運動」に対する持久力をいい、「持久力向上」とは、持久力の増進および持久力の低下の抑制を含む概念である。また本明細書において、「抗疲労」とは、上記「運動」によって生じる疲労を抑制またはその回復を促進することをいう。
本明細書において、「カテキン類」とは、カテキン(C)、ガロカテキン(GC)、カテキンガレート(Cg)、ガロカテキンガレート(GCg)、エピカテキン(EC)、エピガロカテキン(EGC)、エピカテキンガレート(ECg)、およびエピガロカテキンガレート(EGCg)からなる群より選択される少なくとも1種を意味する。
カテキン類は、Camellia属、例えばC. sinensis var. sinensis、C. sinensis var. assamica、やぶきた種、またはそれらの雑種から得られる葉から製造された茶葉から、水または熱水、場合によってはこれに抽出助剤を添加したもので抽出することができる。当該茶葉には、(1)煎茶、番茶、玉露、てん茶、釜入り茶等の緑茶;(2)総称して烏龍茶と呼ばれる鉄観音、色種、黄金桂、武夷岩茶などの半発酵茶;(3)紅茶と呼ばれるダージリン、ウバ、キーマン等の発酵茶の茶葉が含まれる。当該茶葉からのカテキン類の抽出は、撹拌抽出などの通常の方法により行うことができる。抽出用の水または熱水には、予めアスコルビン酸ナトリウムなどの有機酸または有機酸塩類を添加してもよい。必要に応じて、抽出には、煮沸脱気や窒素ガス等の不活性ガスを通気して溶存酸素を除去しつつ、いわゆる非酸化的雰囲気下で抽出する方法を併用してもよい。
あるいは、カテキン類を茶葉から直接抽出するかわりに、茶抽出物の濃縮物または精製物が使用されてもよい。茶抽出物の濃縮物とは、例えば茶葉から熱水もしくは水溶性有機溶剤により抽出された抽出物を濃縮したものであり、茶抽出物の精製物とは、例えば当該抽出物を溶剤やカラム等を用いて精製したものである。当該茶抽出物の濃縮物または精製物の例としては、特開昭59-219384号、特開平4-20589号、特開平5-260907号、特開平5-306279号等に詳細に例示されている方法で調製したものが挙げられる。茶抽出物、およびその濃縮物または精製物は市販品を使用してもよく、その例としては、三井農林(株)「ポリフェノン」、(株)伊藤園「テアフラン」、太陽化学(株)「サンフェノン」、サントリー(株)「サンウーロン」等が挙げられる。茶抽出物の濃縮物または精製物の形態としては、固体、液体、スラリー、ならびにそれらを水、炭酸水または通常の手順で抽出された茶類抽出液等に溶解または希釈したものが挙げられるが、特に限定されない。
さらに、本発明におけるカテキン類は、茶葉以外の他の原料起源、例えばブドウおよびその加工品(ワイン、ジュース類等)またはカカオ豆およびその加工品に由来するもの、あるいは化学合成品であってもよい。
本発明のカテキン類は、好ましくは茶抽出物の濃縮物または精製物の形態で使用され、より好ましくは緑茶抽出物の濃縮物または精製物の形態で使用される。
本発明において、シトルリンおよびアルギニンは、いずれも遊離体またはその塩の形態で使用することができる。シトルリンおよびアルギニンは、L-体、D-体、DL-体、およびそれらの混合物のいずれであってもよいが、好ましくはL-体である。
シトルリンおよびアルギニンの塩としては、酸付加塩、金属塩、アンモニウム塩、有機アミン付加塩、アミノ酸付加塩等が挙げられる。当該酸付加塩としては、塩酸塩、硫酸塩、硝酸塩、リン酸塩等の無機酸塩、および酢酸塩、マレイン酸塩、フマル酸塩、クエン酸塩、リンゴ酸塩、乳酸塩、α-ケトグルタル酸塩、グルコン酸塩、カプリル酸塩等の有機酸塩が挙げられる。当該金属塩としては、ナトリウム塩、カリウム塩等のアルカリ金属塩、マグネシウム塩、カルシウム塩等のアルカリ土類金属塩、アルミニウム塩、亜鉛塩等が挙げられる。当該アンモニウム塩としては、アンモニウム、テトラメチルアンモニウム等の塩が挙げられる。当該有機アミン付加塩としては、モルホリン、ピペリジン等の塩が挙げられる。当該アミノ酸付加塩としては、グリシン、フェニルアラニン、リジン、アスパラギン酸、グルタミン酸等の塩が挙げられる。
当該シトルリンおよびアルギニンの遊離体またはそれらの塩は、それらを含む動植物から単離精製する方法、化学合成、発酵生産等により得ることができる。あるいは市販品を購入してもよい。
(2.カテキン類によるアンモニア代謝促進)
一態様において、本発明は、体内のアンモニア代謝を促進する作用を有する素材の提供に関する。本発明者らは、鋭意検討の結果、カテキン類が、優れた体内アンモニア代謝促進作用を有することを見出した。本発明によれば、カテキン類は、体内のアンモニア代謝を促進し、高アンモニア血症、肝性脳症、慢性疲労症候群などのアンモニアの過剰な蓄積によって生じる疾患または状態を改善することができる。
一態様において、本発明は、体内のアンモニア代謝を促進する作用を有する素材の提供に関する。本発明者らは、鋭意検討の結果、カテキン類が、優れた体内アンモニア代謝促進作用を有することを見出した。本発明によれば、カテキン類は、体内のアンモニア代謝を促進し、高アンモニア血症、肝性脳症、慢性疲労症候群などのアンモニアの過剰な蓄積によって生じる疾患または状態を改善することができる。
後述の実施例に示すとおり、カテキン類は、肝臓細胞におけるアンモニア代謝関連因子の遺伝子発現を亢進し、尿素産生量を増加させる(図1~3参照)。またカテキン類の摂取により、血中および肝臓内アンモニア濃度が低下する(図4、5参照)。したがって本発明によれば、カテキン類は、体内のアンモニア代謝の促進に使用される。
本発明によるアンモニア代謝促進のためのカテキン類の使用は、治療的使用および非治療的使用を含み得る。例えば、非治療的使用としては、アンモニア蓄積による神経系の機能低下に伴う中枢性疲労(特許文献1参照)の抑制、さらには該中枢性疲労による眠気もしくは倦怠感の抑制への適用が挙げられる。また例えば、非治療的使用としては、他者に、医療行為としてではなくカテキン類を投与または摂取させるために、上記中枢性疲労抑制、眠気抑制または倦怠感抑制などの効果を謳ってカテキン類を提供することが挙げられる。
治療的使用としては、慢性疲労症候群、高アンモニア血症、肝性脳症などアンモニアの過剰な蓄積によって生じる疾患または状態の予防または改善への適用が挙げられる。高アンモニア血症は、呼吸性アルカローシス(Respiratory alkalosis)、言語不明瞭(Slurred speech)、振戦(Tremors)、衰弱(Weakness)、筋緊張の増加または低下(Increased or decreased muscle tone)、運動失調(Ataxia)、低体温(Hypothermia)、発作(Seizure)、脳浮腫(Brain oedema)、昏睡(Coma)、脳幹圧迫(Brain stem compression)、頭痛を伴う錯乱(Confusion with headache)、悪心(Nausea)、嘔吐(Vomiting)、興奮(Agitation)、妄想と幻覚(Delusions and delirium)、過敏(irritability)、攻撃性(Aggression)、多動、奇行または自己傷害性行動(Hyperactive, bizarre or self-injurious behavior)、認知障害(Cognitive deficits)、タンパク質嫌悪(Protein aversion)、拒食(Anorexia)、成長遅延(Delayed growth)などの様々な状態を引き起こす。したがって、本発明による高アンモニア血症の予防または改善は、上記に挙げた高アンモニア血症によって引き起こされる状態の予防または改善を含み得る。
したがって、一態様において、本発明は、カテキン類を有効成分とするアンモニア代謝促進剤を提供する。また本発明は、カテキン類を有効成分とする高アンモニア血症の予防もしくは改善剤を提供する。また本発明は、カテキン類を有効成分とする肝性脳症の予防もしくは改善剤を提供する。また本発明は、カテキン類を有効成分とする慢性疲労症候群の予防もしくは改善剤を提供する。
別の態様において、本発明は、アンモニア代謝促進剤の製造のためのカテキン類の使用を提供する。また本発明は、高アンモニア血症、肝性脳症もしくは慢性疲労症候群の予防もしくは改善剤の製造のための、カテキン類の使用を提供する。
一実施形態において、当該本発明の剤は、カテキン類から本質的に構成され得る。別の一実施形態において、当該本発明の剤は、少なくともカテキン類を含有する組成物であり得る。
また別の態様において、本発明は、アンモニア代謝促進のためのカテキン類の使用を提供する。また本発明は、高アンモニア血症、肝性脳症もしくは慢性疲労症候群の予防もしくは改善のための、カテキン類の使用を提供する。
さらに別の態様において、本発明は、アンモニア代謝促進に使用するためのカテキン類を提供する。また本発明は、高アンモニア血症、肝性脳症もしくは慢性疲労症候群の予防もしくは改善に使用するためのカテキン類を提供する。
本発明において、カテキン類は、ヒトおよび非ヒト動物のいずれに対しても使用することができる。非ヒト動物としては、非ヒト哺乳動物、鳥類、爬虫類、両生類および魚類が挙げられ、非ヒト哺乳動物としては、例えば類人猿、その他霊長類、マウス、ラット、ウマ、ウシ、ブタ、ヒツジ、イヌ、ネコ、ハムスター、およびコンパニオン動物等が挙げられる。
なお別の態様において、本発明は、対象におけるアンモニア代謝促進方法を提供する。また本発明は、対象における高アンモニア血症の予防もしくは改善方法を提供する。また本発明は、対象における肝性脳症の予防もしくは改善方法を提供する。また本発明は、対象における慢性疲労症候群の予防もしくは改善方法を提供する。当該方法は、対象に、カテキン類を有効量で投与することを含む。好ましい実施形態において、投与は経口投与である。
当該本発明の方法を適用する対象の例としては、アンモニア代謝促進、または高アンモニア血症、肝性脳症もしくは慢性疲労症候群の予防もしくは改善、またはアンモニア蓄積による神経系の機能低下に伴う中枢性疲労の抑制、または該中枢性疲労による眠気もしくは倦怠感の抑制などを必要とする、上述したヒトおよび非ヒト動物が挙げられる。

当該対象のさらなる例としては、血中アンモニア濃度の上昇に伴う症状または状態、例えば、不明瞭言語、無気力、こん睡、意識障害、視力不鮮明、発達障害、タンパク質誘導性の嘔吐、乳幼児における哺乳力低下など、の予防もしくは改善を必要とする上述したヒトおよび非ヒト動物が挙げられる。
あるいは、本発明のアンモニア代謝促進方法はインビトロ方法であってもよく、該インビトロ方法を適用する対象としては、上述したヒトまたは非ヒト動物由来の肝臓組織および培養肝細胞(例えば肝臓初代培養細胞)等を挙げることができる。

当該対象のさらなる例としては、血中アンモニア濃度の上昇に伴う症状または状態、例えば、不明瞭言語、無気力、こん睡、意識障害、視力不鮮明、発達障害、タンパク質誘導性の嘔吐、乳幼児における哺乳力低下など、の予防もしくは改善を必要とする上述したヒトおよび非ヒト動物が挙げられる。
あるいは、本発明のアンモニア代謝促進方法はインビトロ方法であってもよく、該インビトロ方法を適用する対象としては、上述したヒトまたは非ヒト動物由来の肝臓組織および培養肝細胞(例えば肝臓初代培養細胞)等を挙げることができる。
当該本発明の方法における投与の有効量は、対象のアンモニア代謝促進を達成できる量であり得る。アンモニア代謝レベルは、対象の血中アンモニア濃度または尿素産生量を測定することによって評価することができる。例えば、有効量とは、カテキン類を投与した群のアンモニア代謝レベルを、未投与群と比べて統計的に有意に上昇させる量であり得る。また例えば、有効量とは、カテキン類を投与した個体群の血中アンモニア濃度を、未投与群の90%以下、好ましくは80%以下に低下させる量であり得る。また例えば、有効量とは、培養肝細胞(例えば肝臓初代培養細胞)において、カテキン類投与群の尿素産生量を、未投与群に対して統計学的に有意に、または未投与群の120%以上、好ましくは130%以上に上昇させる量であり得る。
あるいは、アンモニア代謝レベルは、肝臓のアンモニア代謝関連因子の発現量を測定することによって評価することができる。アンモニア代謝関連因子としては、NAGS(N-acetylglutamate synthase)、CPS1(Carbamoyl phosphate synthase-1)、Otc(Ornithine transcarbamoylase)、ASS(Argininosuccinate synthase)、ASL(Argininosuccinate lyase)、およびArg1(Arginase-1)からなる群より選択される少なくとも1つが挙げられる。上記の因子は、肝臓のアンモニア代謝に関わる酵素であり、その発現量の増加は、肝臓のアンモニア代謝を促進する。上記の因子の発現量は、例えばリアルタイムPCRによる当該因子の遺伝子(mRNA)の発現量の測定など、公知の手段に従って測定することができる。したがって、一実施形態において、本発明の方法における投与の有効量は、肝臓組織又は培養肝細胞(例えば肝臓初代培養細胞)におけるアンモニア代謝関連因子の発現量を、未投与群に対して統計学的に有意に増加させる量であり得る。
本発明において、カテキン類は、医薬品、医薬部外品または飲食品(非ヒト動物用飲食品を含む)等に対して、アンモニア代謝促進の機能を付与するための有効成分として、または高アンモニア血症、肝性脳症、慢性疲労症候群などのアンモニアの過剰な蓄積によって生じる疾患もしくは状態の予防もしくは改善機能を付与するための有効成分として、使用することができる。
当該医薬品(医薬部外品も含む)は、アンモニア代謝促進のため、または高アンモニア血症、肝性脳症、慢性疲労症候群などのアンモニアの過剰な蓄積によって生じる疾患もしくは状態を予防または改善するための医薬品であり、カテキン類を当該機能のための有効成分として含有する。さらに、該医薬品は、該有効成分の機能が失われない限りにおいて、必要に応じて薬学的に許容される担体、または他の有効成分、薬理成分等を含有していてもよい。
当該医薬品(医薬部外品も含む)の投与形態は、経口投与および非経口投与の何れであってもよいが、経口投与が好ましい。該医薬品の剤形は、経口または非経口的に投与可能な剤形であれば特に限定されず、例えば注射剤、坐剤、吸入剤、経皮吸収剤、各種外用剤、錠剤、カプセル剤、顆粒剤、散剤、液剤、シロップ剤等の何れでもよく、また、このような種々の剤形の製剤は、カテキン類を、薬学的に許容される担体(例えば賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、希釈剤等)、他の薬効成分等と適宜組み合わせて、定法に従って調製することができる。
当該飲食品は、アンモニア代謝促進、アンモニア蓄積による神経系の機能低下に伴う中枢性疲労の抑制、または該中枢性疲労による眠気もしくは倦怠感の抑制などの機能を提供するための飲食品であり、カテキン類を当該機能のための有効成分として含有する。該飲食品には、アンモニア代謝促進、中枢性疲労の抑制、眠気抑制、倦怠感抑制などの機能をコンセプトとし、必要に応じてその旨を表示した病者用飲食品、および栄養機能飲食品、特定保健用飲食品、機能性表示飲食品等の保健機能飲食品が包含される。
当該飲食品の形態は、固形、半固形または液状(例えば飲料)であり得る。飲食品の例としては、パン類、麺類、飯類、クッキー等の菓子類、ゼリー類、乳製品、スープ類、冷凍食品、インスタント食品、でんぷん加工製品、加工魚肉製品、その他加工食品、調味料、栄養補助食品、およびお茶やコーヒー飲料、果実飲料、炭酸飲料、ゼリー状飲料等の飲料、ならびにそれらの原料が挙げられる。あるいは、該飲食品は、錠剤、カプセル、顆粒、粉末、液剤、シロップなどの経口投与製剤の形態を有するサプリメントであってもよい。
当該飲食品は、カテキン類を、任意の飲食品材料または飲食品に許容される添加物(例えば溶剤、軟化剤、油、乳化剤、防腐剤、香科、甘味料、安定剤、着色剤、紫外線吸収剤、酸化防止剤、保湿剤、増粘剤、固着剤、分散剤、湿潤剤等)と適宜組み合わせて、定法に従って調製することができる。
当該医薬品(医薬部外品も含む)におけるカテキン類の含有量は、特に限定されないが、好ましくは0.01質量%以上、より好ましくは0.05質量%以上、さらに好ましくは0.1質量%以上であり、かつ好ましくは10質量%以下、より好ましくは5質量%以下、さらに好ましくは2.5質量%以下、なお好ましくは1質量%以下である。さらに、当該含有量の例として、0.01~10質量%、0.01~5質量%、0.01~2.5質量%、0.01~1質量%、0.05~10質量%、0.05~5質量%、0.05~2.5質量%、0.05~1質量%、0.1~10質量%、0.1~5質量%、0.1~2.5質量%、及び0.1~1質量%が挙げられる。
当該飲食品中におけるカテキン類の含有量は、特に限定されないが、好ましくは0.01質量%以上、より好ましくは0.05質量%以上、さらに好ましくは0.1質量%以上であり、かつ好ましくは5質量%以下、より好ましくは2.5質量%以下、さらに好ましくは1質量%以下である。さらに、当該含有量の例として、0.01~5質量%、0.01~2.5質量%、0.01~1質量%、0.05~5質量%、0.05~2.5質量%、0.05~1質量%、0.1~5質量%、0.1~2.5質量%、及び0.1~1質量%が挙げられる。
本発明において、カテキン類の投与量および投与計画は、対象の種、体重、性別、年齢、状態、またはその他の要因に従って当業者により適宜決定されればよい。経口投与の場合、本発明によるカテキン類の成人1人1日当たりの投与量は、好ましくは100~3000mg/60kg体重、より好ましくは250~2000mg/60kg体重、さらに好ましくは250~1000mg/60kg体重である。上記の用量を、例えば1日1回、または1日2回もしくは3回以上に分けて投与することが好ましい。
(3.カテキン類、シトルリンおよびアルギニンの併用によるアンモニア代謝促進および持久力向上)
別の一態様において、本発明は、優れたアンモニア代謝促進作用を有し、持久力向上および抗疲労に有効な素材を提供することに関する。本発明者らは、カテキン類と、シトルリンおよびアルギニンとを併用することにより、カテキン類単独またはシトルリンおよびアルギニンのみの併用の場合と比べて、顕著に高いアンモニア代謝促進作用および持久力向上作用が得られることを見出した。本発明によるカテキン類と、シトルリンおよびアルギニンとの併用は、体内のアンモニア代謝を促進し、疲労の軽減や持久力の向上をもたらす。さらに本発明による当該併用によれば、持久力の低下や疲労に悩む病弱者や中高年、高齢者、または運動機能の向上を必要とする者(例えばアスリート)などにおける運動の持久力を向上させ、または運動による疲労を軽減させることができる。
別の一態様において、本発明は、優れたアンモニア代謝促進作用を有し、持久力向上および抗疲労に有効な素材を提供することに関する。本発明者らは、カテキン類と、シトルリンおよびアルギニンとを併用することにより、カテキン類単独またはシトルリンおよびアルギニンのみの併用の場合と比べて、顕著に高いアンモニア代謝促進作用および持久力向上作用が得られることを見出した。本発明によるカテキン類と、シトルリンおよびアルギニンとの併用は、体内のアンモニア代謝を促進し、疲労の軽減や持久力の向上をもたらす。さらに本発明による当該併用によれば、持久力の低下や疲労に悩む病弱者や中高年、高齢者、または運動機能の向上を必要とする者(例えばアスリート)などにおける運動の持久力を向上させ、または運動による疲労を軽減させることができる。
本発明においては、カテキン類、シトルリンおよびアルギニンの3成分を併用することにより、各々単独での適用、またはシトルリンおよびアルギニンのみの併用の場合と比べて、顕著に高いアンモニア代謝促進作用および持久力向上作用が達成される。本発明において、カテキン類、シトルリンおよびアルギニンの3成分は、生体内で協働できる限りにおいて、それら3成分を含む1組成物として同時に投与されてもよく、または各成分が別個に投与されてもよい。
したがって、一態様において、本発明は、カテキン類、シトルリンおよびアルギニンを有効成分とするアンモニア代謝促進剤を提供する。また本発明は、カテキン類、シトルリンおよびアルギニンを有効成分とする持久力向上剤を提供する。また本発明は、カテキン類、シトルリンおよびアルギニンを有効成分とする抗疲労剤を提供する。
別の態様において、本発明は、アンモニア代謝促進剤、持久力向上剤、または抗疲労剤の製造のための、カテキン類、シトルリンおよびアルギニンの使用を提供する。
好ましい実施形態において、当該本発明のアンモニア代謝促進剤、持久力向上剤および抗疲労剤は、少なくともカテキン類、シトルリンおよびアルギニンの3成分を含有する組成物である。一実施形態において、当該本発明の剤は、カテキン類、シトルリンおよびアルギニンから本質的に構成され得る。
また別の態様において、本発明は、アンモニア代謝促進、持久力向上、または抗疲労のための、カテキン類、シトルリンおよびアルギニンの使用を提供する。好ましい実施形態において、該カテキン類、シトルリンおよびアルギニンは、それらを含有する組成物として使用される。
さらに別の態様において、本発明は、アンモニア代謝促進、持久力向上、または抗疲労に使用するための、カテキン類、シトルリンおよびアルギニンを含む組成物を提供する。
当該本発明によるカテキン類、シトルリンおよびアルギニンの3成分の使用は、治療的使用および非治療的使用を含み得る。非治療的使用としては、例えば、日常生活における身体活動による疲労の軽減、筋肉労作を伴う労働における持続性向上や疲労軽減、狭義の運動の際の持久力向上、狭義の運動中またはその後における疲労からの回復促進などの健康増進効果を得る目的で、医療行為としてではなく、カテキン類、シトルリンおよびアルギニンを投与するかまたは摂取することが挙げられる。また例えば、非治療的使用としては、他者に、医療行為としてではなくカテキン類、シトルリンおよびアルギニンを投与または摂取させるために、上記健康増進効果の目的を謳ってカテキン類、シトルリンおよびアルギニンを提供することが挙げられる。
治療的使用としては、慢性疲労症候群、高アンモニア血症、肝性脳症などのアンモニアの過剰な蓄積によって生じる疾患または状態の予防もしくは改善に適用することが挙げられる。
本発明において、カテキン類、シトルリンおよびアルギニンは、ヒトおよび非ヒト動物のいずれに対しても使用することができる。非ヒト動物としては、非ヒト哺乳動物、鳥類、爬虫類、両生類および魚類が挙げられ、非ヒト哺乳動物としては、例えば類人猿、その他霊長類、マウス、ラット、ウマ、ウシ、ブタ、ヒツジ、イヌ、ネコ、ハムスター、およびコンパニオン動物等が挙げられる。
なお別の態様において、本発明は、対象におけるアンモニア代謝促進方法を提供する。また本発明は、対象における持久力向上方法を提供する。また本発明は、対象における抗疲労方法を提供する。当該方法は、対象に、カテキン類、シトルリンおよびアルギニンを有効量で投与することを含む。好ましい実施形態においては、カテキン類、シトルリンおよびアルギニンを含む組成物が投与される。別の好ましい実施形態において、投与は経口投与である。
当該本発明の方法を適用する対象の例としては、アンモニア代謝促進、持久力向上、抗疲労、または慢性疲労症候群、高アンモニア血症もしくは肝性脳症のなどのアンモニアの過剰な蓄積によって生じる疾患もしくは状態の予防もしくは改善を必要とする、上述したヒトおよび非ヒト動物が挙げられる。
当該対象のさらなる例としては、日常生活における身体活動による疲労の軽減、筋肉労作を伴う労働における持続性向上や疲労軽減、狭義の運動の際の持久力向上、狭義の運動中またはその後における疲労からの回復促進などを必要とする、上述したヒトおよび非ヒト動物が挙げられる。
当該対象のさらなる例としては、高アンモニア血症によって引き起こされる状態、例えば、呼吸性アルカローシス(Respiratory alkalosis)、言語不明瞭(Slurred speech)、振戦(Tremors)、衰弱(Weakness)、筋緊張の増加または低下(Increased or decreased muscle tone)、運動失調(Ataxia)、低体温(Hypothermia)、発作(Seizure)、脳浮腫(Brain oedema)、昏睡(Coma)、脳幹圧迫(Brain stem compression)、頭痛を伴う錯乱(Confusion with headache)、悪心(Nausea)、嘔吐(Vomiting)、興奮(Agitation)、妄想と幻覚(Delusions and delirium)、過敏(irritability)、攻撃性(Aggression)、多動、奇行または自己傷害性行動(Hyperactive, bizarre or self-injurious behavior)、認知障害(Cognitive deficits)、タンパク質嫌悪(Protein aversion)、拒食(Anorexia)、成長遅延(Delayed growth)などの予防もしくは改善を必要とする上述したヒトおよび非ヒト動物が挙げられる。
当該対象のさらなる例としては、アンモニア蓄積による神経系の機能低下に伴う中枢性疲労の抑制、または該中枢性疲労による眠気もしくは倦怠感の抑制などを必要とする上述したヒトおよび非ヒト動物が挙げられる。
当該対象のさらなる例としては、血中アンモニア濃度の上昇に伴う症状または状態、例えば、不明瞭言語、無気力、こん睡、意識障害、視力不鮮明、発達障害、タンパク質誘導性の嘔吐、乳幼児における哺乳力低下など、の予防もしくは改善を必要とする上述したヒトおよび非ヒト動物が挙げられる。
あるいは、本発明のアンモニア代謝促進方法はインビトロ方法であってもよく、該インビトロ方法を適用する対象としては、上述したヒトまたは非ヒト動物由来の肝臓組織および培養肝細胞(例えば肝臓初代培養細胞)等を挙げることができる。
当該対象のさらなる例としては、日常生活における身体活動による疲労の軽減、筋肉労作を伴う労働における持続性向上や疲労軽減、狭義の運動の際の持久力向上、狭義の運動中またはその後における疲労からの回復促進などを必要とする、上述したヒトおよび非ヒト動物が挙げられる。

あるいは、本発明のアンモニア代謝促進方法はインビトロ方法であってもよく、該インビトロ方法を適用する対象としては、上述したヒトまたは非ヒト動物由来の肝臓組織および培養肝細胞(例えば肝臓初代培養細胞)等を挙げることができる。
当該本発明の方法における投与の有効量は、対象のアンモニア代謝促進を達成できる量、または対象の持久力向上もしくは抗疲労を達成できる量であり得る。アンモニア代謝レベルは、対象の血中アンモニア濃度または尿素産生量を測定することによって評価することができる。持久力や疲労耐性は、トレッドミル試験等によって実施することができる。一実施形態において、有効量とは、カテキン類、シトルリンおよびアルギニンの3成分を投与した群のアンモニア代謝レベルを、未投与群と比べて統計的に有意に上昇させる量であり得る。別の一実施形態において、有効量とは、該3成分を投与した個体群の血中アンモニア濃度を、未投与群の90%以下、好ましくは80%以下に低下させる量であり得る。また別の一実施形態において、有効量とは、該3成分を投与した個体群の血中アンモニア濃度を、未投与群と比べて統計学的に有意に低下させる量であり得る。さらに別の一実施形態において、有効量とは、培養肝細胞(例えば肝臓初代培養細胞)において、該3成分投与群の尿素産生量を、未投与群の2倍以上、好ましくは3倍以上に上昇させる量であり得る。なお別の一実施形態において、有効量とは、該3成分を投与した個体群のトレッドミル走行時間を、未投与群と比べて統計的に有意に延長させる量であり得る。
本発明において、カテキン類、シトルリンおよびアルギニンは、医薬品、医薬部外品または飲食品(非ヒト動物用飲食品を含む)等に対して、アンモニア代謝促進、持久力向上、または抗疲労の機能を付与するための有効成分として、または慢性疲労症候群、高アンモニア血症、肝性脳症などのアンモニアの過剰な蓄積によって生じる疾患または状態を予防または改善するための有効成分として、使用することができる。
当該医薬品(医薬部外品も含む)は、アンモニア代謝促進、持久力向上、または抗疲労のため、あるいは慢性疲労症候群、高アンモニア血症、肝性脳症などのアンモニアの過剰な蓄積によって生じる疾患もしくは状態を予防または改善するための医薬品であり、カテキン類、シトルリンおよびアルギニンを、当該機能のための有効成分として含有する。さらに、該医薬品は、該有効成分の機能が失われない限りにおいて、必要に応じて薬学的に許容される担体、または他の有効成分、薬理成分等を含有していてもよい。該医薬品は、カテキン類、シトルリンおよびアルギニンを含む1つの組成物として提供されてもよく、またはそれらの1種または2種を含む複数の組成物の組み合わせとして提供されてもよい。
当該医薬品(医薬部外品も含む)の投与形態は、経口投与および非経口投与の何れであってもよいが、経口投与が好ましい。該医薬品の剤形は、経口または非経口的に投与可能な剤形であれば特に限定されず、例えば注射剤、坐剤、吸入剤、経皮吸収剤、各種外用剤、錠剤、カプセル剤、顆粒剤、散剤、液剤、シロップ剤等の何れでもよく、また、このような種々の剤形の製剤は、カテキン類、シトルリンおよびアルギニンを、薬学的に許容される担体(例えば賦形剤、結合剤、増量剤、崩壊剤、界面活性剤、滑沢剤、分散剤、緩衝剤、保存剤、嬌味剤、香料、被膜剤、希釈剤等)、他の薬効成分等と適宜組み合わせて、定法に従って調製することができる。
当該飲食品は、アンモニア代謝促進、持久力向上、または抗疲労の機能を提供するための飲食品であり、カテキン類、シトルリンおよびアルギニンを、当該機能のための有効成分として含有する。該飲食品には、アンモニア代謝促進、持久力向上、または抗疲労の予防または改善をコンセプトとし、必要に応じてその旨を表示した病者用飲食品、および栄養機能飲食品、特定保健用飲食品、機能性表示飲食品等の保健機能飲食品が包含される。
当該飲食品の形態は、固形、半固形または液状(例えば飲料)であり得る。飲食品の例としては、パン類、麺類、飯類、クッキー等の菓子類、ゼリー類、乳製品、スープ類、冷凍食品、インスタント食品、でんぷん加工製品、加工魚肉製品、その他加工食品、調味料、栄養補助食品、およびお茶やコーヒー飲料、果実飲料、炭酸飲料、ゼリー状飲料等の飲料、ならびにそれらの原料が挙げられる。あるいは、該飲食品は、錠剤、カプセル、顆粒、粉末、液剤、シロップなどの経口投与製剤の形態を有するサプリメントであってもよい。該飲食品は、カテキン類、シトルリンおよびアルギニンを含む1つの組成物として提供されてもよく、またはそれらの1種または2種を含む複数の組成物の組み合わせとして提供されてもよい。

当該医薬品(医薬部外品も含む)中におけるシトルリンの含有量は、特に限定されないが、シトルリン遊離体換算で、好ましくは0.01質量%以上、より好ましくは0.05質量%以上、さらに好ましくは0.1質量%以上であり、かつ好ましくは10質量%以下、より好ましくは5質量%以下、さらに好ましくは2.5質量%以下、なお好ましくは1質量%以下である。さらに、当該含有量の例として、0.01~10質量%、0.01~5質量%、0.01~2.5質量%、0.01~1質量%、0.05~10質量%、0.05~5質量%、0.05~2.5質量%、0.05~1質量%、0.1~10質量%、0.1~5質量%、0.1~2.5質量%、及び0.1~1質量%が挙げられる。

当該医薬品(医薬部外品も含む)中におけるアルギニンの含有量は、特に限定されないが、アルギニン遊離体換算で、好ましくは0.01質量%以上、より好ましくは0.05質量%以上、さらに好ましくは0.1質量%以上であり、かつ好ましくは10質量%以下、より好ましくは5質量%以下、さらに好ましくは2.5質量%以下、なお好ましくは1質量%以下である。さらに、当該含有量の例として、0.01~10質量%、0.01~5質量%、0.01~2.5質量%、0.01~1質量%、0.05~10質量%、0.05~5質量%、0.05~2.5質量%、0.05~1質量%、0.1~10質量%、0.1~5質量%、0.1~2.5質量%、及び0.1~1質量%が挙げられる。
当該飲食品中におけるアルギニンの含有量は、特に限定されないが、アルギニン遊離体換算で、好ましくは0.01質量%以上、より好ましくは0.05質量%以上、さらに好ましくは0.1質量%以上であり、かつ好ましくは5質量%以下、より好ましくは2.5質量%以下、さらに好ましくは1質量%以下である。さらに、当該含有量の例として、0.01~5質量%、0.01~2.5質量%、0.01~1質量%、0.05~5質量%、0.05~2.5質量%、0.05~1質量%、0.1~5質量%、0.1~2.5質量%、及び0.1~1質量%が挙げられる。
本発明において、カテキン類、シトルリンおよびアルギニンの投与量および投与計画は、対象の種、体重、性別、年齢、状態、またはその他の要因に従って当業者により適宜決定されればよい。限定ではないが、経口投与の場合、本発明によるカテキン類、シトルリンおよびアルギニンの成人1人1日当たりの投与量の例は、以下のとおりである:

上記の用量を、例えば、1日1回、または1日2回もしくは3回以上に分けて投与することが好ましい。
[カテキン類]好ましくは100~3000mg/60kg体重、より好ましくは250~2000mg/60kg体重、さらに好ましくは250~1000mg/60kg体重;

上記の用量を、例えば、1日1回、または1日2回もしくは3回以上に分けて投与することが好ましい。
本発明において、シトルリンとアルギニンは、それぞれ遊離体換算で、好ましくは1:0.5~2、より好ましくは1:0.8~1.2の質量比で使用される。例えば、本発明のアンモニア代謝促進剤、持久力向上剤および抗疲労剤は、シトルリンとアルギニンを、好ましくは1:0.5~2、より好ましくは1:0.8~1.2の質量比で含有する。また例えば、本発明のアンモニア代謝促進、持久力向上または抗疲労方法において、シトルリンとアルギニンは、好ましくは1:0.5~2、より好ましくは1:0.8~1.2の質量比で投与される。
また本発明において使用されるカテキン類の量と、それぞれ遊離体換算でのシトルリン及びアルギニンの合計量との比は、質量比で、好ましくは1:0.1~10、より好ましくは1:0.2~8、さらに好ましくは1:0.5~2、なお好ましくは1:0.8~1.2である。例えば、本発明のアンモニア代謝促進剤、持久力向上剤および抗疲労剤におけるカテキン類と、シトルリン及びアルギニンの合計量との含有比は、質量比で、好ましくは1:0.1~10、より好ましくは1:0.2~8、さらに好ましくは1:0.5~2、なお好ましくは1:0.8~1.2である。また例えば、本発明のアンモニア代謝促進、持久力向上または抗疲労方法において投与されるカテキン類と、シトルリン及びアルギニンの合計量との用量比は、質量比で、好ましくは1:0.1~10、より好ましくは1:0.2~8、さらに好ましくは1:0.5~2、なお好ましくは1:0.8~1.2である。

以下、実施例を用いて本発明をさらに具体的に説明する。ただし、本発明の技術的範囲はこれら実施例に限定されるものではない。
実施例1 カテキン類が肝臓初代培養細胞のアンモニア代謝に及ぼす影響
(肝臓初代培養細胞の調製)
マウス(雄性Balb/c、18週齢;オリエンタルバイオ)を全身麻酔下にて開腹し、門脈より37℃に暖めたLiver perfusion medium(Invitrogen)を2mL/minの速度でペリスタポンプを用いて流し、15分間全身潅流した。続いて、Liver Digest Mediumで同様に15分間潅流した。その後、肝臓を取り出し、鋏で細切りした後にフィルター(BD falcon)を通して細胞を解離させた。50mLチューブに細胞を回収し、Complete DMEM(D6046,Sigma)10mLでピペッティング洗浄の後、400rpm、4℃で3分間遠心した。同様の操作を3回繰り返し、死細胞を除去した。その後、FBS free培地に細胞を懸濁し、24ウェルプレートに105cells/ウェルで播種した。37℃、5体積%CO2条件下にて24時間培養後、培地交換を行い下記実験に供した。
(肝臓初代培養細胞の調製)

(カテキン刺激および尿素定量)
播種24時間後の細胞をPBSで2回洗浄した後、Arginine、Leucine free DMEM培地(D9443,Sigma)に置き換え、茶カテキン類(ポリフェノン70S、三井農林(株))10μMまたは対照として水を添加し、37℃、5体積%CO2条件下にて24時間培養した後、培養上清を回収した。培養上清中に含まれる尿素をQuantiChrom Urea Assay Kit(BioAssay Systems)を用いて定量した。茶カテキン組成:GC6.1質量%、EGC20.0質量%、C1.8質量%、EC7.9質量%、EGCg32.3質量%、GCg3.2質量%、ECg8.8質量%、Cg0.8質量%。
播種24時間後の細胞をPBSで2回洗浄した後、Arginine、Leucine free DMEM培地(D9443,Sigma)に置き換え、茶カテキン類(ポリフェノン70S、三井農林(株))10μMまたは対照として水を添加し、37℃、5体積%CO2条件下にて24時間培養した後、培養上清を回収した。培養上清中に含まれる尿素をQuantiChrom Urea Assay Kit(BioAssay Systems)を用いて定量した。茶カテキン組成:GC6.1質量%、EGC20.0質量%、C1.8質量%、EC7.9質量%、EGCg32.3質量%、GCg3.2質量%、ECg8.8質量%、Cg0.8質量%。
(遺伝子発現解析)
培地から細胞を回収し、RNeasy Mini Kit(QIAGEN)を用いてRNAを抽出し、High-Capacity RNA-to-cDNA Kit(Applied Biosystems)を用いてcDNA合成を行った後、Realtime PCR(Taqman gene expression,Applied Biosystems)により下記アンモニア代謝関連因子の遺伝子発現の定量を行った。

尿素定量の結果を図1に、遺伝子発現解析の結果を図2に示す。対照群と比べて、カテキン刺激群では尿素産生量が統計学的に有意に増加し、かつCPS1、OtcおよびArg1の発現が統計学的に有意に亢進した。
実施例2 カテキン類摂取が肝臓のアンモニア代謝関連酵素遺伝子発現に及ぼす影響(動物および組織採取)
マウス(雄性Balb/c、6週齢;オリエンタルバイオ)を2群に分け、それぞれに表1記載の対照食および試験食(0.5%カテキン含有)を与えた。試験期間中、マウスは自由摂食、自由摂水とした。各群のマウスに1日30分間の軽度運動走行(トレッドミル、傾斜8度、15m/分を10分間→20m/分を30分間)を週4回施した。8週間後、安静時解剖を行い各マウスの腹部大静脈より採血後、肝臓を摘出して液体窒素にて速やかに凍結し、-80℃にて保存した。

結果を図3に示す。調べた6種の遺伝子全てにおいて、カテキン食摂取群では、対照群と比べて発現が増加した。
実施例3 カテキン類摂取が肝臓のアンモニア代謝に及ぼす影響
(動物および組織採取)
マウス(雄性Balb/c、8週齢;オリエンタルバイオ)は、1週間予備飼育した後、初期持久力平均値が同等になるよう2群に分けた。持久力はトレッドミル(傾斜8度、28m/分)走行にて測定した。各群のマウスに軽度運動走行(トレッドミル、傾斜6度、20m/分、30分間)を週5回、計4週間施した。各走行運動の1時間前には、各群に以下の試験物質のいずれかを経口ゾンデを用いて胃内投与した。4週間後、各群のマウスに経口ゾンデを用いて試験物質を胃内投与し、1時間後にトレッドミル走行(傾斜8度、25m/分)を1時間行った。運動直後に解剖を行い各マウスの腹部大静脈より採血後、肝臓を摘出して液体窒素にて速やかに凍結し、-80℃にて保存した。

Ammonia Assay Kit(abcam 83360)を用いて、肝臓内アンモニアの抽出および定量を行い、肝臓重量当たりのアンモニア量を算出した。

実施例4 カテキン類摂取が血中アンモニア濃度に及ぼす影響
実施例3と同様の条件で、マウスを2群に分け、それぞれに試験物質の投与と運動を行わせた後採血した。採取した血液サンプルについて血中アンモニア濃度を測定した。血中アンモニア濃度の測定にはAmmonia/Ammonium Assay Kit(Abcam)を用いた。

参考例 シトルリンおよびアルギニンが肝細胞の尿素産生に及ぼす影響
(肝臓初代培養細胞の採取)
実施例1と同様の手順でマウスの肝臓から細胞を回収し、死細胞を除去した。その後、DMEM培地に細胞を懸濁し、24ウェルプレートに105cells/ウェルで播種し、培養した。

(尿素の定量)
播種24時間後の細胞をPBSで2回洗浄した後、FBS、Arginine、Leucine free DMEM培地(D9443,Sigma)に置き換え、ここに塩化アンモニウム5mM(control)、塩化アンモニウム+L-アルギニン0.5mM+L-シトルリン0.5mM(Arg+Cit)、または塩化アンモニウム+L-シトルリン1mM(Cit)を添加した。24時間後に培養上清を回収し、含まれる尿素をQuantiChrom Urea Assay Kit(BioAssay Systems)を用いて定量した。
播種24時間後の細胞をPBSで2回洗浄した後、FBS、Arginine、Leucine free DMEM培地(D9443,Sigma)に置き換え、ここに塩化アンモニウム5mM(control)、塩化アンモニウム+L-アルギニン0.5mM+L-シトルリン0.5mM(Arg+Cit)、または塩化アンモニウム+L-シトルリン1mM(Cit)を添加した。24時間後に培養上清を回収し、含まれる尿素をQuantiChrom Urea Assay Kit(BioAssay Systems)を用いて定量した。
尿素定量結果を図6に示す。アルギニン+シトルリン添加群(Arg+Cit)、およびシトルリン単独添加群(Cit)のいずれにおいても肝細胞の尿素産生量は有意に増加し、アンモニア代謝が亢進したことが示された。
実施例5 カテキン類、シトルリンおよびアルギニンが持久力に及ぼす影響
マウス(雄性Balb/c、8週齢;オリエンタルバイオ)は、1週間予備飼育した後、初期持久力平均値が同等になるよう4群に分けた。試験期間中、マウスには試験食AIN76準拠粉末食(10%脂質)を自由摂食、自由飲水させた。持久力はトレッドミル(傾斜8度、28m/分)走行にて測定した。各群のマウスに軽度運動走行(トレッドミル、傾斜6度、20m/分、30分間)を週5回、計4週間施した。各走行運動の1時間前には、各群に以下の試験物質のいずれかを経口ゾンデを用いて胃内投与した。4週間後、再びトレッドミル(傾斜8度、28m/分)により各個体の持久力を測定した。

各群のマウスのトレッドミル走行時間を図7に示す。カテキン単独添加群(Cat)またはカテキン+シトルリン添加群(Cat+Cit)と比べて、カテキン、アルギニンおよびシトルリンの3成分を添加した群(Cat+Arg/Cit)では、走行時間が有意に延長し、持久力が向上したことが示された。
実施例6 カテキン類、シトルリンおよびアルギニンが血中アンモニア濃度に及ぼす影響

各群のマウスの血中アンモニア濃度を図8に示す。カテキン単独添加群(Cat)と比べて、カテキン、アルギニンおよびシトルリンの3成分を添加した群(Cat+Arg/Cit)では、血中アンモニア濃度が有意に低減した。

- カテキン類を有効成分とするアンモニア代謝促進剤。
- カテキン類を有効成分とする高アンモニア血症の予防もしくは改善剤。
- カテキン類を有効成分とする肝性脳症の予防もしくは改善剤。
- カテキン類を有効成分とする慢性疲労症候群の予防もしくは改善剤。
- カテキン類、シトルリンおよびアルギニンを有効成分とするアンモニア代謝促進剤。
- カテキン類、シトルリンおよびアルギニンを有効成分とする持久力向上剤。
- カテキン類、シトルリンおよびアルギニンを有効成分とする抗疲労剤。
- カテキン類と、シトルリンおよびアルギニンの合計量との質量比が1:0.1~10である、請求項5~7のいずれか1項記載の剤。
- シトルリンとアルギニンとの質量比が1:0.5~2である、請求項5~8のいずれか1項記載の剤。
- アンモニア代謝促進剤の製造のための、カテキン類の使用。
- 高アンモニア血症の予防もしくは改善剤の製造のための、カテキン類の使用。
- 肝性脳症の予防もしくは改善剤の製造のための、カテキン類の使用。
- 慢性疲労症候群の予防もしくは改善剤の製造のための、カテキン類の使用。
- アンモニア代謝促進剤の製造のための、カテキン類、シトルリンおよびアルギニンの使用。
- 持久力向上剤の製造のための、カテキン類、シトルリンおよびアルギニンの使用。
- 抗疲労剤の製造のための、カテキン類、シトルリンおよびアルギニンの使用。
- カテキン類と、シトルリンおよびアルギニンの合計量との質量比が1:0.1~10である、請求項14~16のいずれか1項記載の使用。
- シトルリンとアルギニンとの質量比が1:0.5~2である、請求項14~17のいずれか1項記載の使用。
- カテキン類が、カテキン、ガロカテキン、カテキンガレート、ガロカテキンガレート、エピカテキン、エピガロカテキン、エピカテキンガレート、およびエピガロカテキンガレートからなる群より選択される少なくとも1種である、請求項10~18のいずれか1項記載の使用。
- アンモニア代謝促進のための、カテキン類の使用。
- 高アンモニア血症の予防もしくは改善に使用するための、カテキン類。
- 肝性脳症の予防もしくは改善に使用するための、カテキン類。
- 慢性疲労症候群の予防もしくは改善に使用するための、カテキン類。
- アンモニア代謝促進のための、カテキン類、シトルリンおよびアルギニンの使用。
- 持久力向上のための、カテキン類、シトルリンおよびアルギニンの使用。
- 抗疲労のための、カテキン類、シトルリンおよびアルギニンの使用。
- カテキン類と、シトルリンおよびアルギニンの合計量との質量比が1:0.1~10である、請求項24~26のいずれか1項記載の使用。
