코피루왁은 알라미드라는 다른 이름으로도 불리는데 '루왁' 은 인도네시아어이고, '알마미드' 는 필리핀어로 모두 '사향고양이' 를 뜻한다. 둘은 이름만 다르고 맛과 향은 똑같다. 코피루왁의 제조과정은 야생 긴꼬리 사향고양이를 매개체로한 발효공정을 통해 이루어진다. 동남아에 주로 서식하는 야생 긴꼬리 사향고양이는 뛰어난 후각을 이용해 잘 익고 맛있는 팜너츠라는 커피 열매를 따 먹는다. 과육부분은 소화가 되고 소화가 되지 않은 커피 열매 씨앗은 배설물로 나온다. 이때 사향고양이의 침과 위액 등이 소화과정에서 섞이며, 소화기관을 거쳐서 발효되어, 커피 특유의 쓴 맛을 줄이고 특유의 맛과 향을 내게 된다. 꿈의 커피원두가 바로 사향고양이의 배설물 속에 있는 것이다. 코피루왁이 소량으로 생산되는 이유도 사향고양이를 통해 생산될 수밖에 없는 이 과정 때문이다. 수확기가 되면 인근 주민들은 해뜨기 전에 고양이 배설물을 거둬들인다. 그리고 여기서 커피 생두를 골라내 깨끗이 세척하고 햇빛에 잘 말린다. 말린 커피 생두의 속껍질을 벗겨낸 후 로스팅(볶기) 과정을 거치면 커피원두 완제품이 된다. 그러나 코피루왁은 한 잔에 7만원 이상의 고가에 거래되므로, 일반인이 일상적으로 커피를 즐기기에는 무리가 있어 커피 특유의 쓴 맛을 줄이고 특유의 맛과 향을 내는 발효 커피를 개발하여 대중화할 필요가 있다.
이에, 본 발명자는 상기 종래기술들의 문제점을 극복하기 위하여 예의 연구 노력한 결과, 코피루왁의 장내세균을 이용하여 커피 생두의 커피콩을 발효시킨 후 진공처리하여 루왁커피를 생산하는 경우, 카페인 농도가 낮고 커피콩 생두 자체에서 고함량의 가바(GABA,γ- aminobutyric acid)를 함유하였고 커피 특유의 쓴 맛을 줄이고 특유의 맛과 향을 내는 루왁커피를 제조할 수 있음을 확인하고 본 발명을 완성하게 되었다.
따라서, 본 발명의 주된 목적은 고함량의 가바(GABA, γ-aminobutyric acid)를 함유하고, 카페인 농도가 저하되며, 커피콩 커피 특유의 쓴 맛을 줄이고 독특한 맛과 향을 내는 코피루왁의 장내세균을 이용한 발효커피 제조방법을 제공하는데 있다.
본 발명의 일 실시예에 따른 코피루왁의 장내세균을 이용한 발효커피 제조방법은 사향고양이가 커피생두를 따 먹고 배설한 코피루왁을 채취하여 9%의 생리식염수 ⅓과 코피루왁 생두 ⅔를 비커에 넣고 필터 페이퍼로 밀봉한 후 인큐베이터에 넣고 18℃에서 4~5일간 배양하여 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 포함하는 락토바실러스 종균을 배양하는 종균배양단계; 말린 커피 생두의 속껍질을 벗겨낸 커피콩을 물로 세척한 후 상기 종균배양단계에서 배양된 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 포함하는 락토바실러스 종균을 적어도 3%(v/v) 접종한 다음 인큐베이터에 넣어 37℃에서 20시간 발효시키는 발효단계; 및 상기 발효단계에서 발효된 루왁커피를 진공기에 넣어 10-2mmgH으로 25℃에서 4시간 동안 진공처리하는 진공처리단계를 포함하는 것을 특징으로 한다.
본 발명의 방법에 의하면, 향이 캐러멜, 초콜릿, 풀냄새 등의 특성이 있고 쓴 맛이 덜하고 신맛이 적절하게 조화를 이루어 깊고 중후한 맛을 가지며, 고함량의 가바(GABA, γ-aminobutyric acid)를 함유하고, 카페인 농도가 감소된 루왁커피를 제조할 수 있다.

본 발명은 코피루왁의 장내세균을 이용한 발효커피 제조방법에 관한 것으로, 사향고양이가 커피생두를 따 먹고 배설한 코피루왁을 채취하여 9%의 생리식염수 ⅓과 코피루왁 생두 ⅔를 비커에 넣고 필터 페이퍼로 밀봉한 후 인큐베이터에 넣고 18℃에서 4~5일간 배양하여 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 포함하는 락토바실러스 종균을 배양하는 종균배양단계; 상기 배양된 종균을 500 내지 1500rpm에서 5 내지 15분간, 7000 내지 12000rpm에서 5 내지 15분간 및 20000 내지 50000rpm에서 20 내지 40분간 원심분리하여 코피루왁 고초균 중 바실러스 서버틸리스(Bacillus subtilis), 코피루왁 황국균 중 아스퍼질러스 타마리(Aspergillus tamaril), 코피루왁 유산균 중 락토바실러스 플란타룸(Lactobacillus plantarum), 코피루왁 효모 중 지고삭차로미세스 마조르(Zygosaccharomyces major) 각각을 분리하는 단계; 말린 커피 생두의 속껍질을 벗겨낸 커피콩을 물로 세척한 후 불려서 커피콩을 70℃로 열풍건조하여 커피콩의 습도를 50%로 유지시킨 후 상기 분리된 바실러스 서버틸리스(Bacillus subtilis), 아스퍼질러스 타마리(Aspergillus tamaril), 락토바실러스 플란타룸(Lactobacillus plantarum), 지고삭차로미세스 마조르(Zygosaccharomyces major) 각각의 함량을 조절하여 커피콩에 접종한 다음 인큐베이터에 넣어 37℃에서 20시간 발효시키는 발효단계; 및 상기 발효단계에서 발효된 루왁커피를 진공기에 넣고 10-2mmgH으로 25℃에서 4시간 동안 진공처리하는 진공처리단계를 포함할 수 있다.
이하, 본 발명의 바람직한 실시예를 첨부된 도면을 참조하여 상세히 설명하기로 한다.
본 발명에서 '코피루왁 장내세균'은 루왁커피 생두의 발효 중에 발견되는 세균을 일컫는 말로, 코피루왁 장내세균의 대부분은 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모로 이루어진다.
종균배양단계
피나무 열매에서 사향고양이가 커피생두를 따 먹고 배설한 코피루왁을 채취하여 9%의 생리식염수 ⅓량과 코피루왁 ⅔양을 비커에 넣고 필터 페이퍼로 밀봉한 후 인큐베이터에 넣어 15~21℃, 바람직하게는 18℃에서 4~5일간 배양하여 종균(락토바실러스) 즉, 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 추출한다.
여기서, 상기 코피루왁 고초균은 바실러스 서버틸리스(Bacillus subtilis), 바실러스 낫토(Bacillus natto) 중 선택되는 어느 하나 이상이 될 수 있다.
또한, 상기 코피루왁 황국균은 아스퍼르질러스 오리제(Aspergilus oryzae), 아스퍼르질러스 소제(Aspergilus sojae), 아스퍼질러스 타마리(Aspergillus tamaril) 중 선택되는 어느 하나 이상이 될 수 있다.
또한, 상기 코피루왁 유산균은 류코노스톡크 메센터로이데스(Leusonost mesenteroides), 락토바실러스 플란타룸(Lactobacillus plantarum), 락토바실루스 베레비스(Lactobacillus brevis), 류코노스톡 기시코미티톰(Leuconostoc gastcomitaum), 류코노스톡 락티스(Leuconostoc lactis), 락토바실러스 엑소도필루스(Lactobacillus lactis), 비피도박테리움 론굼(Bifidobacterium longum), 스트렙토코커스 써머필러스(Streptococctus thermophilus) 중 선택되는 어느 하나 이상이 될 수 있다.
또한, 상 코피루왁 효모는 지고삭차로미세스 마조르(Zygosaccharomyces major), 시키로미세스 세리비지애(Saccharomyces cerevisiae) 중 선택되는 어느 하나 이상이 될 수 있다.

종균분리단계
배양된 종균을 원심분리기에 넣고 500 내지 1500rpm에서 5 내지 15분간, 7000 내지 12000rpm에서 5 내지 15분간 및 20000 내지 50000rpm에서 20 내지 40분간 원심분리하여 코피루왁 고초균 중 바실러스 서버틸리스(Bacillus subtilis), 코피루왁 황국균 중 아스퍼질러스 타마리(Aspergillus tamaril), 코피루왁 유산균 중 락토바실러스 플란타룸(Lactobacillus plantarum), 코피루왁 효모 중 지고삭차로미세스 마조르(Zygosaccharomyces major) 각각을 분리한다. 이러한 종균분리단계는 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 포함하는 락토바실러스 종균 중 발효커피의 맛이 실제 코피루왁 커피와 가장 유사한 맛이 되도록 종균 중 바실러스 서버틸리스, 아스퍼질러스 타마리, 락토바실러스 플란타룸, 지고삭차로미세스 마조르를 분리하는 과정이다.

발효단계
커피의 생두를 물에 세척한 후 멸균한 배지: MRS+2.0mg/ℓ 각각 0.5∼2%, 0.05∼0.2% 정도를 혼합한 다음 여기에 전술한 종균배양단계에서 배양되어 추출된 종균을 3%(v/v) 이상 되게 접종한 다음 인큐베이터에 넣어 35~40℃에 18~21시간 발효한다. 즉, 말린 커피 생두의 속껍질을 벗겨낸 커피콩을 생수로 세척한 후 불려서 생두커피를 65~75℃로 열풍건조하여 커피콩의 습도를 40~60%로 유지시킨 후 커피콩의 온도가 35~40℃에서, 분리된 바실러스 서버틸리스(Bacillus subtilis), 아스퍼질러스 타마리(Aspergillus tamaril), 락토바실러스 플란타룸(Lactobacillus plantarum), 고삭차로미세스 마조르(Zygosaccharomyces major)를 배지: MRS+2.0mg/ℓ를 혼합하여 커피콩에 직접 접종한 후 인큐베이터에 넣어 35~40℃에서 18~21시간 발효시킨다.

도 1 및 도 2는 아라비카 커피콩 및 로부스타 커피콩과 본 발명에 따른 코피루왁의 장내세균을 이용하여 발효된 커피콩을 나타낸 도면으로, 커피콩이 팽창하여 크기가 커졌음을 알 수 있다.

제조된 발효커피 (루왁커피)의 성분을 발효 "전" 의 커피와 비교하여 분석하였다.

발효 후의 커피 성분을 발효 전과 비교하면, 발효 커피의 경우, 카페인은 발효 '전'의 약 12% 수준으로 감소하였고(0.15g/100g), 가바(GABA,γ-aminobutyric acid)는 발효 '전'의 커피에 비해 그 농도가 5배 이상 증가하였다. 또한, 발효 후 진공처리 한 발효진공 커피의 경우, 카페인은 발효 '전'의 약 8% 수준으로 감소하였고(0.1g/100g), 가바(GABA,γ-aminobutyric acid)는 발효 '전'의 커피에 비해 그 농도가 10배 이상 증가하였다.

본 발명에 있어 '저농도의 카페인'이라 함은 발효 전 커피 생두의 카페인 함량과 비교하여 발효 후에 12.6% 이하로 감소한 카페인 함량을 갖는 것을 의미하며, 보다 바람직하게는 8% 정도로 감소한 카페인 함량을 의미한다.
한편, 본 발명에서 커피 생두를 멸균하는 방법은 당 업계에서 어떠한 멸균 방법도 사용가능하지만, 생두를 물에 세척하여 침지한 후 꺼내여 종균을 접종하는 것도 무방하다.
그리고 커피 생두를 코피루악 유산균으로 발효하기 위해서는 액상 발효를 해야 하며 유산균의 빠른 생장을 위해 추가적인 배지 성분을 넣을 수도 있다,
본 발명의 코피루왁 유산균의 배양에 있어서, 커피 생두와 함께 발효배지에 첨가될수 있는 물질로는 코피루왁 유산균의 배양시 일반적으로 첨가되는 당 업계에 공지된 어떠한 물질도 될수 있으나, 일반적으로는 커피 생두의 2~3배 무게의 물과 포도당, 정백당, 식이섬유 둥의 탄수화물과 효모추출물, 펩톤 등의 단백질을 함께 커피 생두와 첨가하는 것이 바람직하다,
본 발명에 있어서 상기 유산균은 바람직하게 류코노스톡크 메센터로이데스(Leusonost mesenteroides), 락락토바실러스 플란타룸(Lactobacillusns plantarum), 락토바실루스 베레비스(Lactobacillus brevis), 류코노스톡 기시코미티톰(Leuconostoc gastcomitaum), 류코노스톡 락티스(Leuconostoc lactis), 락토바실러스 엑소도필루스(Lactobacillus lactis), 비피도박테리움 론굼(Bifidobacterium longum), 스트렙토코커스 써머필러스(Streptococctus thermophilus) 중 선택되는 어느 하나 이상일 수 있다.
본 발명에서와 같이 코피루왁의 장내세균인 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 생두에 접종하여 발효시킨 후 진공처리하는 경우 발효진공에 의해 카페인 제거의 효과뿐만 아니라 고농도의 가바를 함유하는 발효진공 커피를 생산할 수 있으므로 본 발명의 발효진공커피가 숙면에 큰 도움을 줄 수 있다.
본 발명의 발효커피는 숙면효과를 갖는 가바를 고함량으로 함유함으로써, 각성작용이 있는 카페인의 함량을 저하시켜 기존의 커피가 갖는 문제점을 해소할 수 있다. 이와 동시에 본 발명은 루왁커피 유산균을 접종하여 커피 생두를 발효시킴으로써 숙면에 도움을 주면서 맛과 향이 변하지 않고 오히려 맛과 향이 증가된 커피를 제공하여 종래의 유기용매를 이용한 카페인 커피나 효소를 이용한 가용성 커피의 제조에서 발생되었던 품질의 저하로 인한 문제점을 극복할 수 있었다.
본 발명의 방법에 의하면, 향이 캐러멜, 초콜릿, 풀냄새 등의 특성이 있고 쓴 맛이 덜하고 신맛이 적절하게 조화를 이루어 깊고 중후한 맛을 가지며, 고함량의 가바(GABA, γ-aminobutyric acid)를 함유하고, 카페인 농도가 감소된 루왁커피를 제조할 수 있다.
Claims (1)
- 사향고양이가 커피생두를 따 먹고 배설한 코피루왁을 채취하여 9%의 생리식염수 ⅓과 코피루왁 생두 ⅔를 비커에 넣고 필터 페이퍼로 밀봉한 후 인큐베이터에 넣고 18℃에서 4~5일간 배양하여 코피루왁 고초균, 코피루왁 황국균, 코피루왁 유산균, 코피루왁 효모를 포함하는 락토바실러스 종균을 배양하는 종균배양단계;상기 배양된 종균을 500 내지 1500rpm에서 5 내지 15분간, 7000 내지 12000rpm에서 5 내지 15분간 및 20000 내지 50000rpm에서 20 내지 40분간 원심분리하여 코피루왁 고초균 중 바실러스 서버틸리스(Bacillus subtilis), 코피루왁 황국균 중 아스퍼질러스 타마리(Aspergillus tamaril), 코피루왁 유산균 중 락토바실러스 플란타룸(Lactobacillus plantarum), 코피루왁 효모 중 지고삭차로미세스 마조르(Zygosaccharomyces major) 각각을 분리하는 단계;말린 커피 생두의 속껍질을 벗겨낸 커피콩을 물로 세척한 후 불려서 커피콩을 70℃로 열풍건조하여 커피콩의 습도를 50%로 유지시킨 후 상기 분리된 바실러스 서버틸리스(Bacillus subtilis), 아스퍼질러스 타마리(Aspergillus tamaril), 락토바실러스 플란타룸(Lactobacillus plantarum), 지고삭차로미세스 마조르(Zygosaccharomyces major) 각각의 함량을 조절하여 커피콩에 접종한 다음 인큐베이터에 넣어 37℃에서 20시간 발효시키는 발효단계; 및상기 발효단계에서 발효된 루왁커피를 진공기에 넣고 10-2mmgH으로 25℃에서 4시간 동안 진공처리하는 진공처리단계를 포함하는 것을 특징으로 하는 코피루왁의 장내세균을 이용한 발효커피 제조방법.
