Abstract

세포간지질을 함유하는 다층 라멜라 액정에멀견의 제조방법 【기술분야】
본 발명은 세포간지질을 함유하는 다층 라멜라 액정에멀전의 제조방법에 관한 것으로 더욱 구체적으로 세포간지질 성분, 비 (非) P0E계 비 (非)이온성 계면 활성제, 고급 지방알코을, 오일 및 왁스로 구성된 유상부와 물, 폴리올 및 기타 수상 성분으로 구성된 수상부를 함유하고, 급속냉각 방식을 이용한 공정을 통해 세포간지질을 함유하는 다층 라멜라 구조를 갖는 안정한 액정에멀견의 제조방법 에 관한 것이다.
【배경기술】
인체 피부의 최외각층인 각질층은 외부 유해환경 및 유해물질들로부터 피 부를 보호해주고, 인체 내의 수분이 증발하는 것을 차단하여 피부의 건조한 상태 를 막아주는 피부장벽기능을 수행한다. 이러한 각질층에서 지질 성분들은 특히 중요한 역할을 수행하는데, El i as 둥은 각질층 내의 지질성분이 형성하는 라멜라 구조가 피부장벽기능의 근원이라는 연구결과를 보고하였다 (J . Invest . Dermatol . 69: 535-546 , 1977) . 또한, 실험방법과 피부조직의 위치 등에 따라 차이는 있지 만, Wertz 등은 세포간지질은 대략적으로 세라마이드 약 50%, 콜레스테를 20 ~ 25%, 자유지방산 20 ~ 25%, 10% 정도의 콜레스테를 에스테르, 1 ~ 2% 콜레스테를 설페이트, 소량의 인지질, 소량의 파이토스핑고신 및 소량의 트리글리세라이드로 구성되어 있다고 보고하였다 (Adv. Drug. Del iv. Rev. 18 : 283-294, 1996) .
세포간지질 성분들, 특히 세라마이드와 콜레스테를 등은 거의 대부분 소수성이고, 결정화 경향이 큰 난용성 물질로 일반적인 수중유 (Oi l-in-water) 에멀견 제형에서 분산상인 오일상에 완전히 용해되지 않고, 연속상인 수상에 일부 존재하게 되는데, 이러한 문제로 인하여 겔화 (gel at ion)와 같은 제형 안정도 문제가 발생하게 된다.
이러한 문제를 해결하기 위해 세포간지질과 기타 성분으로 구성된 과립 (알갱이)을 만들어 에멀견 제조 시 맨 마지막 단계에서 후첨하는 방식으로 안정화 기술을 적용하기도 하지만 (대한민국 특허출원 2008—0072355), 캡술화 과정에서 사용되는 과량의 캡술화 기재로 인하여 실제 피부외용제에 사용될 수 있는 세포간지질의 함량이 감소하고, 당류계 혹은 고분자 계열의 캡슐화제는 피부외용제의 사용감을 해치는 단점이 있다.
세포간지질을 에멀견 내에서 안정화시키려는 노력의 일환으로 액정베이스 를 미리 제조하여 일반적인 수중유 에멀견의 맨 마지막 단계에서 후첨하는 방법 이 고안되기도 했다 (대한민국 특허등록 10-0452757, 10-0525842 , 10-0830153 , 10-0867679호) . 하지만, 이렇게 만들어진 액정베이스는 대부분 실온에서 매우 높 은 경도를 갖는 왁스상 또는 페이스트상으로 공정에서의 상용성이 쉽지 않다. 그 리고 액정베이스 제조를 위해서는 과량의 레시틴, 고급 지방알코을을 사용해야 하는데, 이러한 원료들은 대부분 뻑뻑한사용감을 갖고 있기 때문에 사용감 측면 에서 바람직하지 않다ᅳ 또한, 액정베이스를 사용하는 경우, 경시적으로 에멀젼의 점도나 경도가 증가하는 문제가 발생하기도 하는데, 이것은 에멀견에 존재하는 계면활성제 마이셀들이 액정베이스를 통해 만들어진 액정에멀견 오일입자를 공격 하여 궁극적으로는 액정에멀견의 안정도를 해치기 때문으로 추정된다. 이와 유사 한 예로 마이셀 형태로 존재하는 계면활성제 입자들이 수중유 에멀젼의 오일 입 자들 사이에서 물질전달을 일으킨다는 연구결과도 있다 (Food Hydrocol loids , 6(5): 415-422 , 1992) .
마지막으로 세포간지질을 에멀견 내에서 안정화시키기 위해 직접 유화공정 을 통해 액정에멀젼을 만들려는 시도가 있었다. 대한민국 특허등톡 10-0588743은 P0E 계열의 계면활성제와 세포간지질 및 고급 지방알코올 등으로 이루어진 다층 라멜라 구조를 갖는 액정에멀견 제조방법이다. 하지만, 이러한 P0E 계열의 계면 활성제로 이루어진 액정에멀견은 유화계에 팽윤을 일으켜 종국에는 유화계 전체 의 안정도에 영향을 줄 수 있다 (G. M. Eccleston, Mult iple-phase oi 1-in-water emulsions , J . Soc . Cosmet . Chem. , (41) 1-22 1990) . 또한, 상기 특허에서는 온 도조절을 이용한 상전이 유화공정을 이용하는데, 일반적으로 상전이 유화공정은 균일한 크기의 입자들을 얻을 수 있는 장점은 있지만, 제조공정이 쉽지 않고, 제
조공정 중 자연냉각 방식의 서냉공정을 적용해야 하기 때문에 제조시간이 오래 걸릴 수 있다는 단점이 있다. 대한민국 특허출원 2011-0093634에서는 글루코사이 드 계면활성제와 세라마이드 및 오일 등올 사용하여 액정에멀견을 제조하였으나, 제조 중 서냉방식을 사용하였다. 상기 특허에서는 입자가 매우 작아져 상품가치 가 떨어지는 급넁방식에 비해 서넁방식은 적당한 크기의 액정에멀견 입자를 만들 수 있다고 하였지만, 본 발명자들의 연구에 의하면 입자의 크기를 조절하는 것은 냉각방식 보다는 내용물의 조성비가 중요하다. 또한, 동종의 다른 특허 (대한민국 특허등록 10-0830153)에서는 액정의 형성과정이 오일상에 존재하는 고급 지방알 코올인 세토스테알릴알코올이 넁각과정을 통해 결정형태로 석출되면서 입자의 계 면으로 이동하고, 이 때 급넁조작을 가할 경우, 세토스테알릴알코올이 계면으로 이동하지 못하고 과포화상태로 오일내에 존재하여 액정 형성이 적어지고 점도가 낮아진다고 하였다. 하지만, 액정의 형성과정은 고온에서 오일상에 녹아있던 고 급 지방알코올이 넁각과정을 통해 결정형태로 석출되는 것이 아니라, 고체와 액 체의 두 가지 성질을 갖는 액정상으로 전이되면서 입자의 계면에서 일정한 방향 으로 계면활성제들과 함께 배향함으로써 형성되는 것이다. 또한, 급넁과정을 수 행하면 세토스테알릴알코올이 계면으로 이동하지 못한다고 하였지만, 세토스테알 릴알코올은 그 자체가 양친매성 물질로 오일상에 존재하기 보다는 계면으로 이동 하려는 성질을 가지고 있다. 따라서, 세토스테알릴알코올의 계면으로의 이동속도 는 사용되는 오일의 종류와 넁각속도로 조절할 수 있다. 또한, 상기 특허에서는 세토스테알릴알코올이 오일의 용해도 이상 함유해야 한다고 하는데, 과량 함유 시 제조 중 고급 지방알코을의 석출문제가 발생할 수 있고, 사용감 역시 문제될 수 있다.
이에, 본 발명자들은 상기 종래기술들의 문제점들을 극복하기 위하여 예의 연구 노력한 결과, 세포간지질 성분, 비 (非) P0E계 비 (非)이온성 계면활성제, 오 일 및 왁스로 구성된 유상부와 물, 폴리올, 기타 수상 성분으로 구성된 수상부를— 함유하고, 급속넁각 방식을 이용한 공정을 통해 세포간지질 성분들을 다량 함유 하는 다층 라멜라 구조를 갖는 안정한 액정에멀견을 만들 수 있다는 것을 확인하 고, 본 발명을 완성하게 되었다.
【발명의 상세한 설명】
【기술적 과제】
따라서, 본 발명의 주된 목적은 세포간지질 성분, 비 (非) P0E계 비 (非)이온성 계면활성제, 고급 지방알코을, 오일 및 왁스로 구성된 유상부와 물, 폴리을, 기타 수상 성분으로 구성된 수상부를 함유하고, 급속냉각 방식을 이용한 공정을 통해 세포간지질 성분들을 다량 함유하는 다층 라멜라 구조를 갖는 안정한 액정에멀견을 갖는 세포간지질올 함유하는 다층 라멜라 액정에멀견의 제조방법을 제공하는 데 있다.
본 발명의 다른 목적은 상기 세포간지질을 함유하는 다층 라멜라 액정에멀 견의 제조방법을 이용하여 제조된 액정에멀견을 포함하는 화장료 조성물을 제공 하는데 있다.
【기술적 해결방법】
본 발명의 한 양태에 따르면, 본 발명은 하기 단계들을 포함하는 액정에멀견의 제조방법을 제공한다:
a) 세포간지질 성분, 비 (非) P0E계 비 (非)이온성 계면활성제, 고급 지방알코올, 오일 및 왁스를 흔합하고 가온 용해한 유상부를 제조하는 단계;
b) 물, 폴리올 및 기타 수상 성분을 흔합하고 가온 용해한 수상부를 제조하는 단계;
c) 상기 유상부를 상기 수상부에 첨가한 후 교반하는 단계 ;
d) 상기 흔합물 용액을 교반시키면서 점도 조절제를 투입하는 단계;
e) 상기 흔합물 용액을 진공탈포하면서 급속냉각 시키는 단계;
f ) 상기 흔합물 용액을 여과지 또는 여과포로 여과하는 단계. 본 발명의 제조방법에서, 상기 유상부 (a)의 오일은 에스테르 오일, 트리글리세라이드류 오일과 같은 극성오일 또는 하이드로카본오일 및 광물성 오일과 같은 비극성 오일, 그리고 식물성 오일, 실리콘 오일 등으로 이루어진 군에서 선택된 1종 이상인 것일 수 있다.
왁스는 고급 지방알코을, 지방산, 식물성 왁스, 광물성 왁스, 합성 왁스,
오조케라이트, 세틸 팔미테이트, 솔비탄 팔미테이트, 비즈 왁스, 지질 및 지질 펩타이드로 이루어진 군에서 선택된 1종 이상인 것일 수 있다.
본 발명의 제조방법에서, 상기 수상부 (b)의 기타 수상 성분에는 수용성 효능성분을 더 포함할 수 있다. 상기 수용성 효능성분은 예를 들어, 주름개선 기능성 원료, 미백 기능성 원료 및 천연 추출물과 같이 물에 용해되어 수상부를 구성할 수 있는 것이면 어떤 것도 사용될 수 있다.

본 발명의 제조방법에서, 상기 e) 단계는 냉각단계로 여기에서는 급냉방식 을 이용한다. 오일상에 용해되어 있던 고급 지방알코올들은 계면으로 이동하면서 급넁과정에 의해 액상에서 액정상으로 전이하게 되고, 이렇게 액정상으로 전이된 고급 지방알코을들은 계면활성제와 양쪽성 성질을 가진 세포간지질 성분들과 함 께 계면부근에서 일정한 방향으로 배열을 하면서 다층 라멜라 구조를 형성하게 된다. 급넁과정을 거치면서 액정에멀젼은 도 1( f )에서와 같이 결국에는 안정한 다층 라멜라 구조를 갖는 액정에멀젼을 형성하게 된다.
본 발명의 실시예에 따르면, 도 2에서 나타나는 바와 같이, 급냉공정의 경 우, 40 ~ 45 °C 부근에서 고급 지방알코올들이 액상에서 액정상으로 전이되면서 계면에 잘 배향하여 5 ~ l /m 수준의 액정에멀견 입자를 잘 형성하였으나, 서넁 공정의 경우, 고급 지방알코올들이 분산상인 오일상에서 연속상인 수상으로 넘어
가면서 액정의 배열이 무너져 다층 라멜라 구조를 이루는 액정에멀젼 입자의 숫 자가 적고, 라멜라 구조의 밀도 또한 급넁공정에 비해 낮음을 알 수 있다. 따라 서, 본 발명의 급냉공정이 서냉공정 보다 더 안정한 다층 라멜라 구조를 형성한 다는 것을 알 수 있다 (도 2) .

이와 같은 문제점을 해결하기 위해 세포간지질을 에멀젼 내에서 안정화 시 키는 방법으로 액정베이스를 미리 제조하여 일반적인 수중유 에멀견의 맨 마지막 단계에서 후첨하는 방법, 직접 유화공정을 통해 액정에멀젼을 만드는 방법과 이 과정의 제조공정 중에서 자연넁각 방식을 채택하는 서냉공정을 적용하여 액정에 멀견이 만들어 질 수 있다.
그러나, 액정베이스를 후첨하는 방법은 액정베이스가 실온에서 매우 높은 경도를 갖기 때문에 공정상 상용성이 어렵고, 액정베이스를 사용하는 경우, 경시 적으로 에멀젼의 점도나 경도가 높아진다는 단점이 있다. 또한, 직접 유화공정을 통해 액정에멀견을 제조방법에서는 POE(polyoxyethylene) 계열의 계면활성제를 함유하고 있는데, 이러한 P0E 계열의 계면활성제로 이루어진 액정에멀젼은 유화 계에 팽윤을 일으켜 종국에는 유화계 전체의 안정도에 영향을 줄 수 있다. 팽윤 된 유화계는 계면이 파손되면서 내부에 배향되어진 각종 세포간지질과 고급 지방 알코올들이 연속상인 수상으로 빠져나와 경시적으로 점도와 경도의 상승을 일으 켜 에멀젼 전체의 안정성을 악화시킬 수 있다. 더욱이, 서넁공정을 적용하여 액 정베이스를 제조하는 경우 제조시간이 오래 걸릴 수 있다는 단점이 있기 때문에 이러한 액정에멀젼 제조방법은 바람직하지 못하다.
따라서, 본 발명의 세포간지질을 함유하는 다층 라멜라 구조를 갖는 액정 에멀젼을 제조함에 있어서, 액정베이스를 후첨하는 방법을 사용하지 않고, 비
(非) P0E 비 (非)이온성 계면활성제의 사용과 급넁공정을 적용하면 세포간지질을 함유하는 안정한 다층 라멜라 구조를 갖는 수중유 액정에멀견을 만들 수 있다는 점을 발견하고 본 발명을 완성한 것이다.
본 발명의 시험예에 따르면, 일반에멀견 (비교예 5)과 액정에멀견 (실시예 1) 에서의 세포간지질 성분들의 안정도를 확인한 결과, 액정에멀견 (실시예 1)의 경우 다층 라멜라 구조를 형성하기 때문에 양쪽성 성질을 가진 세포간지질 성분들이 라 멜라 구조내에서 안정하게 존재하여 점도 차이가 크지 않았으나, 일반에멀견 (비교 예 5)의 경우에는 냉동 및 넁장 조건에서는 에멀젼 내에서 내상에서 외상으로의 세포간지질 성분 이동속도가 느려지기 때문에, 상온 및 고온 조건에 비해 점도상 승의 폭이 낮았지만, 액정에멀견 (실시예 1)에 비해 점도가 3배 이상 증가하였고ᅳ 상온 및 고온 조건에서는 제조직후 내부에 존재하던 세포간지질 성분들이 계면을 통과하여 외상으로 이동하고, 그 후 겔화 (Gelat ion)화 과정을 거치면서 점도 측정 자체가 불가능해 질 정도로 점도가 급상승하였다. 이러한 결과로 볼 때, 본 발명 의 액정에멀젼은 여러 온도 조건에서도 세포간지질 성분을 함유하는 안정한 다층 라멜라구조를 갖는 액정에멀견을 제조할 수 있음을 알수 있다 (시험예 4) .
본 발명에 있어서, 상기 세포간지질은 종래에 알려진 어떠한 세포간지질 성분도 사용될 수 있으며, 바람직하게는 세라마이드, 콜레스테를, 트리글리세라 이드, 파이토스핑고신 및 자유지방산인 것을 특징으로 한다.
본 발명에서, 세라마이드는 천연 세라마이드와 유사 (pseudo) 세라마이드로 이루어진 군에서 선택되는 하나 이상인 것일 수 있다. 이 중 천연 세라마이드는 자연계에 존재하는 세라마이드로서 , 구체적으로는 세라마이드 -1, 세라마이드 -2, 세라마이드 -3, 세라마이드—4, 세라마이드 -5, 세라마이드 -6, 세라마이드 -7, 세라 마이드 -8 등이 포함된다.
본 발명에 있어서, 상기 비 (非)이온성 계면활성제는 종래에 알려진 어떠한 비 (非)이온성 계면활성제도 사용될 수 있으며, 바람직하게는 레시틴, 소르비탄계 계면활성제, 글루코사이드계 계면활성제로 구성된 군으로부터 하나 이상인 것을 특징으로 한다.
본 발명에 있어서, 상기 비 (非)이은성 계면활성제는 레시틴, 소르비탄계 계면활성제 및 글루코사이드계 계면활성제를 흔합하여 사용하는 것을 특징으로 한다.
본 발명에 따르면, 상기 계면활성제는 P0E계열의 비 (非)이온성 계면활성제 를 사용하지 않는다. 전술한 것처럼, P0E계열의 비 (非)이온성 계면활성제 사용 시 액정에멀견 입자가 팽윤되어 에멀견의 안정도를 해칠 수 있기 때문이다.
본 발명의 시험예에 따르면, 계면활성제는 레시틴 소르비탄계 계면활성제, 글루코사이드계 계면활성제의 흔합물을 이용하였다. 상기 계면활성제들이 각각 단독으로 사용될 경우, 도 3(a)에서와 같이 레시틴의 경우에는 액정의 안정도 문 제와 뻑뻑한 사용감 문제가 발생하고, 소르비탄계 계면활성제는 도 3(b)에서와 같이 입자크기가 너무 거대하여 제형 안정성이 떨어지며, 글루코사이드계 계면활 성제는 도 3(c)에서와 같이 상대적으로 액정입자가 너무 작아서 원하는 품질의 액정을 형성할 수 없다. 이에 반해, 세 가지의 계면활성제를 흔합한 경우 도 3(d) 와 같이 액정의 입자가 균일하여 원하는 품질의 액정에멀견을 제조할 수 있고, 액정의 안정도가 향상되었다. 이러한 결과로 볼 때, 안정도를 향상시키기 위해서 는 계면활성제를 단독으로 사용하는 것 보다 상기 세 가지의 계면활성제를 흔합 하여 사용하는 것이 가장 안정된 액정에멀젼을 제조하는데 바람직하다는 것을 알 수 있다 (시험예 1 및 도 3) .
본 발명에 있어서, 상기 레시틴은 총 중량 대비 0.05 - 1 중량 소르비탄 계 계면활성제는 총 중량 대비 0. 1 ~ 5 중량 % , 글루코사이드계 계면활성제는 총 중량 대비 0. 1 ~ 5 중량 % 함유하는 것을 특징으로 한다.
상기 수치보다 계면활성제 함량이 작을 경우, 액정에멀젼 입자가 거대하게 형성되거나 또는 유화자체가 불가능하게 될 수 있다. 또한, 상기 수치 이상의 계 면활성제가 사용될 경우에는 에멀견 입자가 극히 작게 형성되어 액정의 성질을 보여줄 수 없게 된다.
본 발명에 있어서, 상기 폴리을은 프로필렌글리콜, 디프로필렌글리콜, 1 , 3-부틸렌글리콜, 펜틸렌글리콜 및 1,2-핵산디을로 구성된 군으로부터 선택된 하나 이상인 것을 특징으로 하며 그 사용량은 에멀젼 총 중량에 대하여 1 ~ 30% 범위이다.
본 발명에 있어서, 상기 급속넁각은 5 ~ 15분 이내에 70°C ~ 90°C에서부터 40 ~ 50°C냉각하는 것을 특징으로 한다. 이것은 고급 지방알코을의 액정상으로의 전이온도가 40 ~ 50 °C 사이로 알려졌기 때문이다.
본 발명의 시험예에 따르면, 급넁공정에 의해 제조된 액정에멀견 (실시예 1) 은 모든 온도조건에서 3달간 안정한 다층 라멜라 구조를 보여주었으나, 서냉공정 에 의해 제조된 액정에멀젼 (비교예 4)은 3달 후 상온, 37°C , 45 °C , CYC 항온조 보관조건에서 액정상태가 약해지거나 액정이 붕괴되어 점도가 상승하는 등의 문 제를 일으키는 것을 확인하였다 (시험예 3) .
본 발명에 있어서, 상기 액정에멀젼은 다층 라멜라 구조를 갖는 것을 특징 으로 한다.
본 발명의 도 4에서 나타나는 바와 같이, 전자현미경으로 관찰한 본 발명 의 액정에멀견이 다층 라멜라 구조를 갖는 것을 확인 할 수 있었다. 본 발명의 다른 양태에 따르면, 본 발명은 상기 방법으로 제조된 액정에멀 젼을 포함하는 화장료 조성물을 제공한다.
본 발명의 시험예에 따르면, 본 발명의 실시예 1에서 제조된 액정에멀견으 로 피부 안전성 테스트를 한 결과, 피부 자극이 1 미만으로 무자극이라는 것을 확인하였다. 이러한 결과로 볼 때, 본 발명의 제조방법으로 만들어진 액정에멀견 을 포함하는 화장료의 조성물의 경우 피부에 사용하여도 안전하다는 것을 알 수 있다 (시험예 5) .
본 발명에 있어서, 상기 조성물은 용액, 현탁액, 유탁액, 페이스트, 겔, 크림, 로션, 파우더, 비누, 오일, 분말 파운데이션, 유탁액 파운데이션, 왁스 파 운데이션 및 스프레이로 구성된 군으로부터 선택되는 제형을 갖는 것을 특징으로 한다. 【유리한 효과】
본 발명은 세포간지질을 함유하는 다층 라멜라 구조를 갖는 안정한 액정에 멀견에 관한 것으로, 일반 수중유 에멀견에서는 안정화가 어려운 세포간지질 성분 들이 액정에멀견 내에서 질서있는 배향을 통해 제형내에서의 안정도를 향상시킨다.

도 1은 본 발명에 따라 제조된 액정에멀젼의 제조온도에 따른 편광현미경 사진으로 특히, 45°C 부근에서부터는 편광현미경 관측시 다층 라멜라 구조의 특 징인 Maltose cross 무늬를 관측할 수 있다. (a: 73 °C, b: 63 °C, c: 53°C , d: 45 °C, e: 35 °C, f: 25 °C)

이하, 실시예를 통하여 본 발명을 더욱 상세히 설명하기로 한다. 이들 실 시예는 단지 본 발명을 예시하기 위한 것이므로, 본 발명의 범위가 이들 실시예 에 의해 제한되는 것으로 해석되지는 않는다.

<제조방법>
1)상기 표 1에서 3 ~ 6은 세포간지질이고, 8 ~ 10은는 비 (非) P0E계 비 (非) 이온성 계면활성제이고, 2, 7은 고급 지방알코을이고, 11 ~ 13은 오일이며, 1은 왁스, 14 ~ 17은 수용성 성분으로, 이중 16, 17은 폴리을이고, 18, 19는 점도 조 절제이고 20은 보존쎄이며 21은 향 성분이다.
2) 상기 표 1에서 원료 1 - 13를 흔합하고, 70 - 80°C에서 용해하여 유상 용액을 제조하였다.
3) 별도의 용기에 상기 표 1의 원료 14 - 17을 혼합하고, 70 ~ 80°C까지 가온용해하여 수상용액을 제조하였다.
4) 상기 1)의 유상용액을 상기 2)의 수상용액에 첨가하고, 호모 믹서로 교 반하였다.
5) 상기 3)의 흔합용액에 표 1의 원료 18을 흔합하고, 이어서 원료 19을 흔합하였다.
6) 상기 4)의 과정이 끝나자마자, 급속넁각하여 10분 내에 40°C까지 넁각 하였다. 이옥고, 표 1의 원료 20과 21을 투입한 후, 기포를 제거하였다.
상기 표 1에 의해 만들어진 비교예 1 내지 3 및 실시예 1을 편광현미경으 로 관측한 결과, 도 3에 나타나는 바와 같이 계면활성제를 단독으로 사용하는 것 (도 3a , 도 3b 및 도 3c)보다 흔합하여 사용하는 것 (도 3d)이 액정의 안정도를 보다 향상시킬 수 있었다. 시험예 2 : 비교예 및 실시예 의 제조

<제조방법>
1)상기 표 1에서 3 ~ 6은 세포간지질이고, 8 ~ 10은 비 (非) P0E계 비 (非)이 온성 계면활성제이고, 2, 7은 고급 지방알코을이고, 11 - 13은 오일이며, 1은 왁 스, 14 ~ 17은 수용성 성분으로, 이중 16, 17은 폴리을이고, 18, 19는 점도 조절 제이고 20은 보존제이며 21은 향 성분이다.
2) 상기 표 2에서 원료 1 ~ 13를 흔합하고, 70 ~ 80°C에서 용해하여 유상 용액을 제조하였다.
3) 별도의 용기에 상기 표 2의 원료 14 ~ 17을 흔합하고, 70 ~ 80°C까지 가온용해하여 수상용액을 제조하였다.
4) 상기 1)의 유상용액을 상기 2)의 수상용액에 첨가하고, 호모 믹서로 교 반하였다.
5) 상기 3)의 흔합용액에 표 2 의 원료 18을 흔합하고, 이어서 표 2의 원 료 19을 흔합하였다.
6) 실시예 1은 상기 4)의 과정이 끝나자마자, 급속넁각하여 10분 내에 40°C 까지 냉각하였다. 이옥고, 표 2의 원료 20과 21을 투입한 후, 기포를 제거하였다. (시험예 2에서의 실시예 1은 시험예 1에서의 실시예 1과 동일하다. )
7) 비교예 4는 상기 4)의 과정이 끝난 후, 실온에 그대로 방치하다가 40°C 부근에서 표 2의 원료 20과 21을 투입한 후, 기포를 제거하였다. 상기 표 2에 따라 제도된 실시예 1 및 비교예 4를 편광현미경으로 측정한 결과, 도 2에 나타나는 바와 같이, 급넁방식이 서냉방식에 비해 안정도가 우수한 액정에멀견을 제조할 수 있음을 알 수 있다. 시험예 3 : 넁각속도에 따른 액정에멀젼 입자의 안정성 측정 시험
상기 실시예 1 및 비교예 4에서 넁각속도에 따른 액정에멀젼 입자의 크기 와 밀도, 안정성을 측정하기 위해 편광현미경을 이용하여 온도에 따른 액정에멀 견의 입자 상태를 관측하였다.
그 결과, 도 2에서 나타나는 바와 같이, 급넁공정의 경우 40 ~ 45 °C 부근 에서 고급 지방 알코을들이 액상에서 액정상으로 전이되면서 계면에 잘 배향하여 5 ~ 15 수준의 액정에멀견 입자를 잘 형성하였다. 이에 비해, 서넁공정의 경우, 고급 지방알코올들이 분산상인 오일상에서 연속상인 수상으로 넘어가면서 액정의 배열이 무너져 도 2에서 보듯, 다층 라멜라 구조를 이루는 액정에멀젼 입자의 숫 자가 적고, 다층 라멜라 구조의 밀도 역시 급넁공정에 비해 낮음을 알 수 있다. 또한, 실시예 1 및 비교예 4에서 액정에멀젼의 온도에 따른 안정도를 확인
하기 위해, 냉동 (_18°C ) , 냉장 (4°C ) , 상온, 37°C , 45°C , CYC (넁동, 상온, 45°C 각 12시간씩) 항온조에서 3달 동안 각각의 조성물에 대한 안정도를 하기 표 3에 나타내었다.

상기 표 3에서 나타나는 바와 같이, 급냉공정에 의해 쎄조된 액정에멀젼 (실시예 1)은 모든 온도조건에서 3달간 안정한 다층 라멜라 구조를 보여주었고, 서냉공정에 의해 제조된 액정에멀견 (비교예 1)은 3달 후 상은, 37°C , 45 °C 항은 조 보관조건에서 액정상태가 약해지거나 액정이 붕괴되어 점도가 상승하는 등의 문제를 일으키는 것을 확인하였다. 시험예 4 : 세포간지질 성분의 제형내 안정도 테스트
세포간지질 성분들의 제형내에서의 안정도를 알아보기 위해 일반에멀견과 액정에멀젼을 제조하였다. 일반에멀견은 시험예 2에서 원료 1과 원료 6을 제외한 후 제조하였고, 액정에멀젼은 시험예 2에서 실시예 1로 하였다. 이렇게 만들어진 일반에멀견을 비교예 5로 하고, 액정에멀젼은 실시예 1로 한다. 세포간지질 성분 들의 제형내에서의 안정도는 넁동, 넁장, 상온 및 고온에서의 점도변화를 근거로 측정하였다. 점도계 (Brookf ield, DV-I pr ime , USA) 측정조건은 상온 (25°C 기준) 에서 Spindle 64를 사용하여 30 rpm 조건에서 진행하였다. 점도측정의 단위는 cp 이다. 제조직후 및 3개월이 지난 시점에서 각 항온조 보관조건에서의 점도를 표 4에 기록하였다.

상기 표 4에서 나타나는 바와 같이, 액정에멀견 (실시예 1)의 경우에는 제 조직후와 3개월 후의 점도 차이가 1,000 ~ l,500cp 사이로 점도의 변화가 크지 않은 반면에, 일반에멀젼 (비교예 5)의 경우에는 제조직후와 3개월 후 냉동, 넁장 조건에서 점도 차이가 3,500cp 이상 차이가 발생하였고, 특히 상온 및 고온 보관 조건에서는 점도가 급상승하게 되어 측정 자체가 불가능했다. 시험예 5 : 액정에멀견의 피부 안전성 테스트
피부 질환이 없는 성인 남녀 10명을 대상으로 하여 상기 실시예 1 및 비교 예 4에 대하여 자극 정도를 다음과 같이 평가하였다. 시험자들의 팔 전박에 20μ1의 시료를 도포한 후, 시험 부위를 밀폐한 후 24시간 동안 첩포하고, 첩포 를 제거한 후 30분과 24시간 후에 피부에서의 반웅을 CTFMCosmetic, Toiletry and Fragrance Association) 가이드라인에 제시된 terminology에 의거하여 검사 한다. 판정기준은 다음과 같다. 판정기준에 의해 얻어진 시험.자들의 점수를 평균 하여 1미만이면 저자극, 3미만이면 경자극, 5미만이면 중자극, 5이상이면 강자극 으로 평가한다.

【청구항 1]
하기 단계들을 포함하는 액정에멀견의 제조방법:
a) 세포간지질 성분, 비 (非) P0E계 비 (非)이온성 계면활성제 , 고급 지방알코올, 오일 및 왁스를 흔합하고 가온 용해한 유상부를 제조하는 단계; b) 물, 폴리을 및 기타 수상 성분을 흔합하고 가온 용해한 수상부를 제조하는 단계 ;
c) 상기 유상부를 상기 수상부에 첨가한 후 교반하는 단계 ;
d) 상기 혼합물 용액을 교반시키면서 점도 조절제를 투입하는 단계; e) 상기 흔합물 용액을 진공탈포하면서 급속냉각 시키는 단계;
f ) 상기 흔합물 용액을 여과지 또는 여과포로 여과하는 단계.
【청구항 2】
제 1항에 있어서, 상기 세포간지질은 세라마이드, 콜레스테를, 파이토스핑 고신, 트리글리세라이드 및 자유지방산인 것을 특징으로 하는 액정에멀견 제조방 법.
