実際の測定条件は個々に異なるが、公知の文献中に各種アマドリアーゼの熱安定性に関する開示がみられる:Aspergillus terreus GP1株由来の真核型アマドリアーゼは、45℃、10分間の熱処理で約40%の残存活性を示す(例えば、非特許文献4参照。)。Fusarium oxysporum S-1F4株由来の真核型アマドリアーゼの45℃、5分間の熱処理後における残存活性は約10%である(例えば、非特許文献12参照。)。また、Coniochaetidium savoryi ATCC36547株由来の真核型アマドリアーゼは、37℃、10分間の熱処理で80%の残存活性を示している(例えば、特許文献9参照。)。Arthrinium sp. TO6株、Pyrenochaeta sp. YH807株、Leptosphaeria nodorum NBRC7480株、Pleospora herbarum NBRC32012株、Ophiobolus herpotrichus NBRC6158株由来の各真核型アマドリアーゼは、40℃、30分間の熱処理後に80%の残存活性を示している(例えば、特許文献9参照。)。Neocosmospora vasinfecta NBRC7590株由来の真核型アマドリアーゼは、45℃、30分間の熱処理で80%の残存活性を示している(例えば、特許文献9参照。)。Curvularia clavata YH923株由来の真核型アマドリアーゼは、50℃、30分間の熱処理で80%の残存活性を示している(例えば、特許文献9参照。)。カルボキシル末端領域の34~39アミノ酸残基を欠損させたCryptococcus neoformas由来アマドリアーゼは、45℃、10分間の熱処理で40%の残存活性を示している(例えば、特許文献12参照。)。Eupenicillium terrenum ATCC 18547株またはConiochaeta sp. NISL9330株由来のアマドリアーゼは、45℃、10分間の熱処理で80%以上の残存活性を示している(例えば、特許文献8参照。)。

(アマドリアーゼ)
アマドリアーゼは、ケトアミンオキシダーゼ、フルクトシルアミノ酸オキシダーゼ、フルクトシルペプチドオキシダーゼ、フルクトシルアミンオキシダーゼ等とも称され、酸素の存在下で、イミノ2酢酸またはその誘導体(アマドリ化合物)を酸化して、グリオキシル酸またはα-ケトアルデヒド、アミノ酸またはペプチドおよび過酸化水素を生成する反応を触媒する酵素のことをいう。アマドリアーゼは、自然界に広く分布しており、微生物や、動物または植物起源の酵素を探索することにより、得ることができる。微生物においては、例えば、糸状菌、酵母または細菌等から得ることができる。
熱安定性の向上をもたらすアミノ酸の置換として、配列番号1に示すアミノ酸配列における以下の位置のアミノ酸に対応する位置のアミノ酸の置換が挙げられる。
(1)カルボキシル末端からの3アミノ酸残基の欠失。
(2)151位のアラニンの置換、例えば、システインへの置換。

これらのアマドリアーゼをコードする本発明の遺伝子(以下、単に「アマドリアーゼ遺伝子」ともいう。)を得るには、通常一般的に用いられている遺伝子のクローニング方法が用いられる。例えば、アマドリアーゼ生産能を有する微生物菌体や種々の細胞から常法、例えば、Current Protocols in Molecular Biology (WILEY Interscience,1989)記載の方法により、染色体DNAまたはmRNAを抽出することができる。さらにmRNAを鋳型としてcDNAを合成することができる。このようにして得られた染色体DNAまたはcDNAを用いて、染色体DNAまたはcDNAのライブラリーを作製することができる。
本発明において用いることのできるベクターとしては、上記プラスミドに限定されることなくそれ以外の、例えば、バクテリオファージ、コスミド等の当業者に公知の任意のベクターを用いることができる。具体的には、例えば、pBluescriptII SK+(STRATAGENE社製)等が好ましい。
アマドリアーゼ遺伝子の変異処理は、企図する変異形態に応じた、公知の任意の方法で行うことができる。すなわち、アマドリアーゼ遺伝子あるいは当該遺伝子の組み込まれた組換え体DNAと変異原となる薬剤とを接触・作用させる方法;紫外線照射法;遺伝子工学的手法;またはタンパク質工学的手法を駆使する方法等を広く用いることができる。
上記変異処理に用いられる変異原となる薬剤としては、例えば、ヒドロキシルアミン、N-メチル-N’-ニトロ-N-ニトロソグアニジン、亜硝酸、亜硫酸、ヒドラジン、蟻酸または5-ブロモウラシル等を挙げることができる。
上記接触・作用の諸条件は、用いる薬剤の種類等に応じた条件をとることが可能であり、現実に所望の変異をアマドリアーゼ遺伝子において惹起することができる限り特に限定されない。通常、好ましくは0.5~12Mの上記薬剤濃度において、20~80℃の反応温度下で10分間以上、好ましくは10~180分間接触・作用させることで、所望の変異を惹起可能である。紫外線照射を行う場合においても、上記の通り常法に従い行うことができる(現代化学、p24~30、1989年6月号)。
上述の如くして得られたアマドリアーゼ遺伝子を、常法により、バクテリオファージ、コスミド、または原核細胞もしくは真核細胞の形質転換に用いられるプラスミド等のベクターに組み込み、各々のベクターに対応する宿主を常法により、形質転換または形質導入をすることができる。例えば、得られた組換え体DNAを用いて、任意の宿主、例えば、エッシェリシア属に属する微生物、具体例としては大腸菌K-12株、好ましくは大腸菌JM109株、大腸菌DH5α株(ともにタカラバイオ社製)や大腸菌B株、好ましくは大腸菌SHuffle株(NEW ENGLAND BioLabs社製)、大腸菌BL21株(ニッポンジーン社製)等を形質転換またはそれらに形質導入してそれぞれの菌株を得ることができる。
アミノ酸配列の相同性は、GENETYX-Mac(Software Development社製)のマキシマムマッチングやサーチホモロジー等のプログラムまたはDNASIS Pro(日立ソフト社製)のマキシマムマッチングやマルチプルアライメント等のプログラムにより計算することができる。
「アミノ酸に対応する位置」とは、配列番号1に示すConiochaeta属由来のアマドリアーゼのアミノ酸配列の特定の位置のアミノ酸に対応する他の生物種由来のアマドリアーゼのアミノ酸配列における位置をいう。
「アミノ酸に対応する位置」を特定する方法としては、例えばリップマン-パーソン法等の公知のアルゴリズムを用いてアミノ酸配列を比較し、各アマドリアーゼのアミノ酸配列中に存在する保存アミノ酸残基に最大の相同性を与えることにより行うことができる。アマドリアーゼのアミノ酸配列をこのような方法で整列させることにより、アミノ酸配列中にある挿入、欠失にかかわらず、相同アミノ酸残基の各アマドリアーゼ配列における配列中の位置を決めることが可能である。相同位置は、三次元構造中で同位置に存在すると考えられ、対象となるアマドリアーゼの特異的機能に関して類似した効果を有することが推定できる。
なお、本発明において、「配列番号1記載のアミノ酸配列の43位のフェニルアラニンに対応する位置」とは、確定したアマドリアーゼのアミノ酸配列を、配列番号1に示されるConiochaeta属由来のアマドリアーゼのアミノ酸配列と比較した場合に、配列番号1のアマドリアーゼの43位のフェニルアラニンに対応するアミノ酸を意味するものである。これにより、上記の「相当する位置のアミノ酸残基」を特定する方法でアミノ酸配列を整列させた図1により特定することができる。
「配列番号1記載のアマドリアーゼのカルボキシル末端からの3アミノ酸残基に対応する位置」とは、確定したアマドリアーゼのアミノ酸配列を、配列番号1に示されるConiochaeta属由来のアマドリアーゼのアミノ酸配列と比較した場合に、配列番号1記載のアミノ酸配列のカルボキシル末端からの3アミノ酸残基を意味するものである。Coniochaeta属由来のアマドリアーゼにおける、この位置の3残基の配列は、435位のプロリン、436位のリジンおよび437位のロイシンからなり、これらに対応する位置のアミノ酸配列も、上記の方法でアミノ酸配列を整列させた図1より特定することができる。
上記のようにして得られた熱安定性の優れたアマドリアーゼの生産能を有する菌株を用いて、当該アマドリアーゼを生産するには、この菌株を通常の固体培養法で培養してもよいが、可能な限り液体培養法を採用して培養するのが好ましい。
なお、培地の初発pHは、pH7~9に調整するのが適当である。
また、培養は任意の条件を用いることができるが、例えば、20~42℃の培養温度、好ましくは37℃前後の培養温度で4~24時間、さらに好ましくは37℃前後の培養温度で4~8時間、通気攪拌深部培養、振盪培養、静置培養等により実施することができる。
上記のような手段で得られる本発明のアマドリアーゼは、遺伝子改変等により、そのアミノ酸配列に変異を生じた結果、改変前のものと比較して熱安定性が向上していることを特徴とする。具体的には、改変前のものと比較して、本明細書中に記載の活性測定方法および熱安定性評価方法に記載した反応条件下で、所定の熱処理、例えば、「55℃、30分間」あるいは「60℃、30分間」等の熱処理後の残存活性(%)が、本発明の変異を導入する前と比較して向上していることを特徴とする。
アマドリアーゼの活性の測定方法としては、種々の方法を用いることができるが、一例として、以下に、本発明で用いるアマドリアーゼ活性の測定方法について説明する。
本発明におけるアマドリアーゼの酵素活性の測定方法としては、酵素の反応により生成する過酸化水素量を測定する方法や酵素反応により消費する酸素量を測定する方法などが主な測定方法として挙げられる。以下に、一例として、過酸化水素量を測定する方法について示す。
フルクトシルバリン等の糖化アミノ酸、およびフルクトシルバリルヒスチジン等の糖化ペプチドは、阪上らの方法に基づき合成、精製することができる(特許文献3参照)。
(1)試薬1:POD-4-AA溶液
4.0kUのパーオキシダーゼ(キッコーマン社製)、100mgの4-アミノアンチピリン(東京化成社製)を0.1Mのリン酸カリウム緩衝液(pH7.0)に溶解し、1Lに定容する。
(2)試薬2:TOOS溶液
500mgのTOOS(同仁化学社製)をイオン交換水に溶解し、100mlに定容する。
(3)試薬3:基質溶液(150mM;終濃度 5mM)
フルクトシルバリン417mgをイオン交換水に溶解して10mlに定容する。
2.7mlの試薬1,100μlの試薬2、および100μlの酵素液を混和し、37℃で5分間予備加温する。その後、試薬3を100μl加えてよく混ぜた後、分光光度計(U-3010、日立ハイテクノロジーズ社製)により、555nmにおける吸光度を測定する。測定値は、555nmにおける1分後から3分後の1分間あたりの吸光度変化とする。なお対照液は、100μlの試薬3の代わりに100μlのイオン交換水を加える以外は前記と同様に調製する。37℃、1分当たりに生成される過酸化水素のマイクロモル数を酵素液中の活性単位(U)とし、下記の式に従って算出する。

アマドリアーゼ粗酵素液、またはアマドリアーゼ精製標品を約0.5U/mlとなるように、10% キシリトールを含む0.1M リン酸緩衝液(pH7.0)で希釈し、55℃にて30分間加温する。上述のB.の方法を用いて加熱前と加熱後のサンプルの酵素活性を測定し、加熱前の活性を100とした場合の残加熱後の活性の割合、すなわち、残存活性(%)を求めることにより、熱安定性を評価する。
CFP-T9遺伝子(配列番号2)を含む組換え体プラスミドを有する大腸菌JM109(pKK223-3-CFP-T9)株(特許文献16参照)を、LB-amp培地[1%(W/V) バクトトリプトン、0.5%(W/V) ペプトン、0.5%(W/V) NaCl、50μg/ml Ampicilin]100mlに接種して、37℃で20時間振とう培養し、培養物を得た。
この培養物を7,000rpmで、5分間遠心分離することにより集菌して菌体を得た。次いで、この菌体よりQIAGEN tip-100(キアゲン社製)を用いて組換え体プラスミドpKK223-3-CFP-T9を抽出して精製し、組換え体プラスミドpKK223-3-CFP-T9のDNA100μgを得た。
上記組換え体プラスミドpKK223-3-CFP-T9 DNA100μgのうち、20μgを用いて、大腸菌XL1-Red(STRATAGENE社製)(増殖の際、プラスミドの複製にエラーを起こしやすく、改変を生じやすい)をD.M.Morrisonの方法(Method in Enzymology,68,326~331,1979)に従って形質転換し、約15,000株の形質転換株を得た。
まず、得られた上記形質転換体の全てを、ビロード生地を用いて新しいLB-amp寒天培地にレプリカした。レプリカプレート上のコロニーをHybond-N+(Amersham社製)に転写し、BugBuster Protein Extraction Reagent(Novagen社製)に浸した。このHybond-N+を55℃で1時間処理した後、2mM フルクトシルバリン、4U/ml パーオキシダーゼ(キッコーマン社製)、1mg/ml 4-アミノアンチピリン(東京化成社製)、10mg/ml TOOS(同仁化学社製)を含む0.1Mのリン酸カリウム緩衝液(pH7.0)に浸したところ、少数の強い発色を示す株が認められた。
この強い発色に相当するコロニーをマスタープレート上より選択し、2mlのLB-amp培地で液体培養することにより、プラスミドにコードされる改変アマドリアーゼを生産させた。
得られた8株をLB-amp培地2ml中、37℃で18時間振盪培養し、この培養液からQIAGEN tip-100(キアゲン社製)を用いてプラスミドを単離した。該プラスミドをそれぞれpKK223-3-CFP-T11、pKK223-3-CFP-T12、pKK223-3-CFP-T13、pKK223-3-CFP-T14、pKK223-3-CFP-T15、pKK223-3-CFP-T17、pKK223-3-CFP-T18、pKK223-3-CFP-T19と命名し、各プラスミド中のアマドリアーゼをコードするDNAの塩基配列を、マルチキャピラリーDNA解析システムCEQ2000(ベックマン・コールター社製)を用いて決定した。
その結果、pKK223-3-CFP-T11中には、配列番号1記載のアミノ酸配列の196番目のグルタミン酸がアスパラギン酸に、pKK223-3-CFP-T12には、配列番号1記載のアミノ酸配列の299番目のセリンがスレオニンに、pKK223-3-CFP-T13には、配列番号1記載のアミノ酸配列の323番目のバリンがグルタミン酸に、pKK223-3-CFP-T14中には、配列番号1記載のアミノ酸配列の43番目のフェニルアラニンがチロシンに、pKK223-3-CFP-T15には、配列番号1記載のアミノ酸配列の53番目のヒスチジンがアスパラギンに、pKK223-3-CFP-T17には、配列番号1記載のアミノ酸配列の185番目のアラニンがセリンに、pKK223-3-CFP-T18には、配列番号1記載のアミノ酸配列の267番目のフェニルアラニンがチロシンに、pKK223-3-CFP-T19には、配列番号1記載のアミノ酸配列の350番目のスレオニンがアラニンに置換する変異が導入されていることが明らかとなった。
特許文献17によると配列番号1記載のアミノ酸配列の53位のヒスチジンに対応する位置であるAspergillus nidulans由来のフルクトシルアミノ酸オキシダーゼにおける52位のチロシンをヒスチジンへ置換すると耐熱性が向上すると示されている。そこで、CFP-T9においても同様の効果があるか検証するために、配列番号1記載のアミノ酸配列の53位のヒスチジンをチロシンへ置換したアマドリアーゼの作製を試みた。組換え体プラスミドpKK223-3-CFP-T9 DNAを鋳型として、CFP-T9の53位のヒスチジンをチロシンに置換することを意図して、配列番号3、4のDNA配列よりなるプライマーを、常法により合成した。次に、上記で得た組換え体プラスミドpKK223-3-CFP-T9のDNAを鋳型として、配列番号3、4のプライマー、KOD-Plus-(東洋紡績社製)を用い、以下の条件でPCR反応を行った。
すなわち、10×KOD-Plus-緩衝液を5μl、dNTPが各2mMになるよう調製されたdNTPs混合溶液を5μl、25mMのMgSO4溶液を2μl、鋳型となるpKK223-3-CFP-T9 DNAを50ng、上記合成オリゴヌクレオチドをそれぞれ15pmol、KOD-Plus-を1Unit加えて、滅菌水により全量を50μlとした。調製した反応液をサーマルサイクラー(エッペンドルフ社製)を用いて、94℃で2分間インキュベートし、続いて、「94℃、15秒」-「50℃、30秒」-「68℃、6分」のサイクルを30回繰り返した。
タンパク質の熱安定性を向上させるひとつの手法として、タンパク質分子内に新規のジスルフィド結合を構築させる手法が知られている(例えば、Science 226,555-557,1984参照)。そこで、CFP-T9タンパク質分子内に新たにジスルフィド結合を構築することを意図し、そのための参考情報を得る目的で、既に結晶構造が報告されているタンパク質の中で、CFP-T9と最もアミノ酸配列の相同性が高いAmadoriase IIの立体構造(例えば、J.Biol.Chem. 283,27007-27016,2008参照)を参考にして、CFP-T9の立体構造に関する予測を行うことを試みた。なお、Amadoriase IIとCFP-T9は、アミノ酸配列において34%の同一性を有している。
Coniochaeta属由来のアマドリアーゼのアミノ酸配列のカルボキシル末端からの3アミノ酸残基の配列は、435位のプロリン、436位のリジンおよび437位のロイシンからなる。公知の情報を参考にすれば、この配列は、真核生物においてタンパク質をペルオキシソームへ輸送するためのシグナル配列であり、カルボキシル末端の3アミノ酸から構成されるモチーフであるPTS1に相当する可能性が示唆される。CFP-T9においてもこの配列が存在しており、この領域がPTS1に相当する可能性も示唆され得る。
得られた大腸菌SHuffle(pKK223-3-CFP-T11)株、大腸菌SHuffle(pKK223-3-CFP-T12)株、大腸菌SHuffle(pKK223-3-CFP-T13)株、大腸菌SHuffle(pKK223-3-CFP-T14)株、大腸菌SHuffle(pKK223-3-CFP-T15)株、大腸菌SHuffle(pKK223-3-CFP-T16)株、大腸菌SHuffle(pKK223-3-CFP-T17)株、大腸菌SHuffle(pKK223-3-CFP-T18)株、大腸菌SHuffle(pKK223-3-CFP-T19)株、大腸菌SHuffle(pKK223-3-CFP-T20)株、大腸菌SHuffle(pKK223-3-CFP-T21)株を、LB-amp培地で30℃、20時間培養した。その後、各菌体をpH8.0の0.1Mリン酸緩衝液で洗浄、超音波破砕、15,000rpmで10分間遠心分離し、各粗酵素液1.5mlを調製した。
このようにして調製した各粗酵素液をサンプルとし、上記の熱安定性測定法に従って、各種改変型アマドリアーゼの熱安定性評価を行った。結果を表1に示す。表1において、CFP-T9は、大腸菌SHuffle(pKK223-3-CFP-T9)株由来のアマドリアーゼを示す。なお、本実施例では大腸菌SHuffle(pKK223-3-CFP-T9)株由来のアマドリアーゼであるCFP-T9を変異元酵素としたため、表中に記載の「アミノ酸変異」の記載には、CFP-T9に既に導入済みの各種変異点は含めていない。
これまでに、いわゆるPTS1配列に相当する領域を欠失させることにより、生産されるタンパク質の熱安定性を向上させることができるという知見は全く知られていない。従って、今回の結果は、発明者らとしても予測し得ない顕著な効果であり、驚くべき結果であった。アマドリアーゼタンパク質のカルボキシル末端の数アミノ酸が実際にいわゆるPTS1配列に相当し得る領域であるか否かは現状定かではなく、また、この領域を削除することが熱安定性を向上させる具体的な作用機序も現時点では不明であるが、実際に、図1に示す複数のアマドリアーゼをみてもわかるように、アミノ酸としては同一でなくとも、いわゆるPTS1配列モチーフに相当し得るアミノ酸残基の配列を有しているという視点で見た場合、複数のアマドリアーゼにおいてカルボキシル末端の3アミノ酸の削除によって、同様なシグナル配列領域に相当し得る配列が削除され、これにより各種のアマドリアーゼに関しても効果的に熱安定性を付与できる可能性が示唆され得る。
実施例1で見出された11の熱安定性向上変異の知見に基づき、これらを組み合わせて変異を蓄積させることにより、さらに熱安定性を高めたアマドリアーゼを取得することを目的として、多重変異体(2重変異体、3重変異体、4重変異体)の作製による耐熱性向上型変異の蓄積を試みた。
そして、上記と同様の条件で大腸菌SHuffle株を形質転換し、大腸菌SHuffle(pKK223-3-CFP-T22)株、大腸菌SHuffle(pKK223-3-CFP-T23)株、大腸菌SHuffle(pKK223-3-CFP-T24)株、大腸菌SHuffle(pKK223-3-CFP-T25)株、大腸菌SHuffle(pKK223-3-CFP-T26)株を得た。
そして、上記と同様の条件で大腸菌SHuffle株を形質転換し、大腸菌SHuffle(pKK223-3-CFP-T27)株、大腸菌SHuffle(pKK223-3-CFP-T28)株、大腸菌SHuffle(pKK223-3-CFP-T29)株、大腸菌SHuffle(pKK223-3-CFP-T30)株、大腸菌SHuffle(pKK223-3-CFP-T31)株、大腸菌SHuffle(pKK223-3-CFP-T32)株を得た。
これに対し、実施例1で確認された各種の単独変異を組み合わせて作製した各種多重変異体においては、残存活性がいずれも顕著に向上していた。具体的には、43位のフェニルアラニンがアラニンに置換され、かつカルボキシル末端の3アミノ酸残基が欠失した二重変異体における残存活性は2.5%であり、CFP-T9と比較して向上した。さらに、この変異に350位のスレオニンがアラニンに置換された三重変異体における残存活性は4.8%であり、CFP-T9と比較してさらに向上した。また、151位のアラニンがシステインに置換されたものにおける残存活性は26%であり、CFP-T9と比較して顕著に向上した。さらに、この変異に別の変異を順次加えて変異を蓄積して得た二重変異体、三重変異体および四重変異体でも、CFP-T9と比較して顕著に残存活性が向上し、これらの多重変異体が、熱安定性が向上したアマドリアーゼの熱安定性を向上させる変異点であることが確認された。
しかも、151位のアラニンがシステインに置換された変異体や43位のフェニルアラニンがアラニンに置換され、かつカルボキシル末端の3アミノ酸残基が欠失した二重変異体に変異を蓄積する都度、得られたさらなる多重変異体の熱安定性は段階的に向上しており、実施例1で確認された本発明の変異点を適宜組み合わせることによって、一層優れた熱安定性を有するアマドリアーゼが作出され得ることが明らかとなった。
配列番号16は熱安定性向上型変異(G184D、N272D、H388Y)を導入したEupenicillium terrenum由来アマドリアーゼのアミノ酸配列であり、配列番号16のアミノ酸配列をコードする遺伝子(配列番号17)を挿入した組換え体プラスミドpUTE100K’-EFP-T5を大腸菌で発現させることにより、Eupenicillium terrenum由来アマドリアーゼの活性が確認されている(特許文献16参照)。
そして、実施例1と同様の条件で大腸菌SHuffle株を形質転換し、大腸菌SHuffle(pUTE100K’-EFP-T5-G151C)株を得た。
上記のようにして得られた改変型アマドリアーゼ生産能を有する大腸菌SHuffle(pUTE100K’-EFP-T5-G151C)株を、実施例1の方法で培養して、各種改変型アマドリアーゼの粗酵素液1.5mlを調製した。得られた各粗酵素液をサンプルとし、実施例1に準じた熱安定性測定方法に従って、各種改変型アマドリアーゼの熱安定性評価を行った。結果を表5に示す。
Phaeosphaeria nodorum由来フルクトシルペプチドオキシダーゼを大腸菌で発現させることを試みた。すでに明らかになっているPhaeosphaeria nodorum由来フルクトシルペプチドオキシダーゼのアミノ酸配列を配列番号20に示した(例えば、Biotechnology and Bioengineering,106,358-366,2010参照)。この配列番号20で示した437アミノ酸をコードし、且つ大腸菌発現用にコドンを最適化した、配列番号21で示す1314bpの遺伝子(終止コドンTAAを含む)を、定法である遺伝子断片のPCRによる全合成によりcDNAを全合成することで取得した。このとき、配列番号21の5’末端、3’末端にはそれぞれEcoRIサイトとHindIIIサイトを付加した。また、クローニングした遺伝子配列から予想されるアミノ酸配列全長は図1のPhaeosphaeria nodorum由来フルクトシルペプチドオキシダーゼの配列と一致していることを確認した。
Phaeosphaeria nodorum由来フルクトシルペプチドオキシダーゼに耐熱性向上型変異を導入するために、組換え体プラスミドpKK223-3-PnFXを鋳型にして、配列番号22、23の合成オリゴヌクレオチド、KOD-Plus-(東洋紡績社製)を用い、実施例1と同様の条件でPCR反応、大腸菌JM109の形質転換および生育コロニーが保持するプラスミドDNA中のアマドリアーゼをコードするDNAの塩基配列決定を行った。その結果、配列番号20記載のアミノ酸配列の149位のアラニンがシステインに置換されたPhaeosphaeria nodorum由来フルクトシルペプチドオキシダーゼ遺伝子をコードする組換え体プラスミド(pKK223-3-PnFX-A149C)を得た。
そして、上記と同様の条件で大腸菌SHuffle株を形質転換し、大腸菌SHuffle(pKK223-3-PnFX-A149C)株を得た。
そして、上記と同様の条件で大腸菌SHuffle株を形質転換し、大腸菌SHuffle(pKK223-3-PnFX-ΔPTS1)株を得た。
上記のようにして得られた改変型フルクトシルペプチドオキシダーゼ生産能を有する大腸菌SHuffle(pKK223-3-PnFX-A149C)株および大腸菌SHuffle(pKK223-3-PnFX-ΔPTS1)株を、上記の方法で培養して、各種改変型フルクトシルペプチドオキシダーゼの粗酵素液1.5mlを調製した。得られた各粗酵素液をサンプルとし、実施例1に準じた熱安定性測定方法に従って、各種改変型フルクトシルペプチドオキシダーゼの熱安定性評価を行った。結果を表6に示す。

のいずれかに記載されるアミノ酸残基へと置換されており、前記置換または欠失を有する位置以外の位置で数個のアミノ酸の欠失、挿入、付加および置換のうちの1以上が導入されたアミノ酸配列を有し、前記置換または欠失を有する前のアマドリアーゼと比較して、pH7.0において55℃、30分間の熱処理後の残存活性(%)が向上しているアマドリアーゼ。 - 配列番号1に示すアミノ酸配列において、以下の(u)から(ae):
(u)151位のアラニンに対応する位置のアミノ酸のシステインへの置換および299位のセリンに対応する位置のアミノ酸のスレオニンへの置換;
(v)43位のフェニルアラニンに対応する位置のアミノ酸のチロシンへの置換および151位のアラニンに対応する位置のアミノ酸のシステインへの置換;
(w)43位のフェニルアラニンに対応する位置のアミノ酸のチロシンへの置換およびカルボキシル末端からの3アミノ酸残基に対応する位置のアミノ酸残基の欠失;
(x)151位のアラニンに対応する位置のアミノ酸のシステインへの置換、196位のグルタミン酸に対応する位置のアミノ酸のアスパラギン酸への置換および299位のセリンに対応する位置のアミノ酸のスレオニンへの置換;
(y)151位のアラニンに対応する位置のアミノ酸のシステインへの置換、299位のセリンに対応する位置のアミノ酸のスレオニンへの置換およびカルボキシル末端からの3アミノ酸残基に対応する位置のアミノ酸残基の欠失;
(z)43位のフェニルアラニンに対応する位置のアミノ酸のチロシンへの置換、350位のスレオニンに対応する位置のアミノ酸のアラニンへの置換およびカルボキシル末端からの3アミノ酸残基に対応する位置のアミノ酸残基の欠失;
(aa)43位のフェニルアラニンに対応する位置のアミノ酸のチロシンへの置換、151位のアラニンに対応する位置のアミノ酸のシステインへの置換およびカルボキシル末端からの3アミノ酸残基に対応する位置のアミノ酸残基の欠失;
(ab)43位のフェニルアラニンに対応する位置のアミノ酸のチロシンへの置換、151位のアラニンに対応する位置のアミノ酸のシステインへの置換および350位のスレオニンに対応する位置のアミノ酸のアラニンへの置換;
(ac)151位のアラニンに対応する位置のアミノ酸のシステインへの置換、196位のグルタミン酸に対応する位置のアミノ酸のアスパラギン酸への置換、299位のセリンに対応する位置のアミノ酸のスレオニンへの置換および323位のバリンに対応する位置のアミノ酸のグルタミン酸への置換;
(ad)151位のアラニンに対応する位置のアミノ酸のシステインへの置換、196位のグルタミン酸に対応する位置のアミノ酸のアスパラギン酸への置換、299位のセリンに対応する位置のアミノ酸のスレオニンへの置換およびカルボキシル末端からの3アミノ酸残基に対応する位置のアミノ酸残基の欠失;
(ae)43位のフェニルアラニンに対応する位置のアミノ酸のチロシンへの置換、151位のアラニンに対応する位置のアミノ酸のシステインへの置換、350位のスレオニンに対応する位置のアミノ酸のアラニンへの置換およびカルボキシル末端からの3アミノ酸残基に対応する位置のアミノ酸残基の欠失
よりなる群から選択されるアミノ酸残基の置換または欠失を有し、前記置換または欠失を有する位置以外の位置で数個のアミノ酸の欠失、挿入、付加および置換のうちの1以上が導入されたアミノ酸配列を有し、前記置換または欠失を有する前のアマドリアーゼと比較して、pH7.0において60℃、30分間の熱処理後の残存活性(%)が向上しているアマドリアーゼ。 - 請求項1~3のいずれか1項に記載のアミノ酸配列をコードするアマドリアーゼ遺伝子。
- 請求項4記載のアマドリアーゼ遺伝子を含む組換えベクター。
- 請求項5記載の組換えベクターを含む宿主細胞。
- 以下の工程:
(i)請求項5記載の宿主細胞を培養する工程;
(ii)宿主細胞に含まれるアマドリアーゼ遺伝子を発現させる工程;及び
(iii)培養物からアマドリアーゼを単離する工程
を含む、アマドリアーゼを製造する方法。 - 請求項1~3のいずれかに記載のアマドリアーゼを含む、糖化タンパク質の測定に用いるためのキット。
- 請求項1~3のいずれかに記載のアマドリアーゼを含む、糖化ヘモグロビンの測定に用いるためのキット。
