タウリンは、食品及び医薬品に汎用されている機能性成分であり、恒常性維持機能を有することが知られているが(非特許文献1参照)、近年皮膚機能の維持・改善や毛髪の成長促進にとっても重要な役割を担っていることが分かってきた。例えば、あれ肌改善効果(非特許文献2参照)、保湿への影響(非特許文献3参照)、マウスにおける損傷皮膚の治癒促進効果(非特許文献4参照)、毛包成長への影響(非特許文献5参照)が挙げられる。また、タウリンから誘導されるチオタウリンには皮脂酸化防止効果があること(非特許文献6参照)、塩化タウリン及び臭化タウリンには黄色ブドウ球菌やアクネ菌に対する抗菌作用があること(非特許文献7参照)が知られている。
もっとも、タウリンは水溶性であるため、皮膚の最外層で脂溶的性質を有する角層への浸透能が低く、外用剤に配合して皮膚や頭皮に適用した場合、その機能を十分に発揮させることできないという課題があった。
そこで、アミノ酸と脂肪酸とでイオンペアを形成させ、脂溶性を上げることで皮膚への吸収を増大させる方法(非特許文献8参照)、オレイン酸やazone(登録商標)を用いて経皮吸収を促進させる方法(非特許文献9参照)、炭素原子数4~8の1,2-アルカンジオール類の混合物により吸収を促進させる方法(特許文献1参照)が提供されている。
また、経皮吸収促進作用を有する成分としては、乳酸(特許文献2参照)といった有機酸、乳酸オクチルドデシル(特許文献3参照)といった有機酸エステル、エチルヘキサン酸2-エチルヘキシル(特許文献4参照)、ジエチレングリコールモノエチルエーテル(特許文献5参照)等が知られており、タウリンをこれらの成分と共に配合した外用剤を提供することも可能であるが、これらの成分は皮膚に対して刺激性を有したり、経皮吸収促進作用が充分でない場合もあった。

本発明は、水溶性であるタウリンの経皮吸収性を増大させた皮膚外用剤を提供することを課題とする。
本発明者らは、前記課題を解決すべく鋭意検討を重ねた結果、経皮吸収促進剤として知られる成分の中でも、アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルがタウリンの経皮吸収性を増大させることに関し、特異的に顕著な効果を奏することを見出した。
かかる知見により得られた本発明の態様は以下の通りである。
(1)タウリン、並びに、アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種を含有することを特徴とする皮膚外用剤。
(2)アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種の含有量が、タウリンの1質量部に対してアジピン酸ジイソプロピルでは0.3質量部以上、セバシン酸ジイソプロピルでは0.8質量部以上である(1)の皮膚外用剤。
(1)タウリン、並びに、アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種を含有することを特徴とする皮膚外用剤。
(2)アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種の含有量が、タウリンの1質量部に対してアジピン酸ジイソプロピルでは0.3質量部以上、セバシン酸ジイソプロピルでは0.8質量部以上である(1)の皮膚外用剤。
本発明により、タウリンの経皮吸収性を増大させた、タウリンを含有する皮膚外用剤を提供することが可能となった。これにより、皮膚(頭皮を含む)におけるタウリンの有効性(例えば、皮膚機能の維持・改善など)を充分に発揮させることが可能となった。本発明の皮膚外用剤は、皮膚刺激性が少なく安全性が高い点でも有利である。
本発明は、タウリン、並びに、アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種を含有することを特徴とする皮膚外用剤を提供する。
「タウリン」の含有(配合)量は、皮膚外用剤中、通常、0.1~10質量%であり、0.5~5.0質量%が好ましい。0.1質量%未満では充分な薬効が期待できない場合があり、10質量%を超えると製剤化が困難となる場合があるためである。
「アジピン酸ジイソプロピル」は、2-プロパノールのアジピン酸ジエステルを主とする無色澄明の液体であり、「セバシン酸ジイソプロピル」は、2-プロパノールのセバシン酸ジエステルを主とする無色澄明の液体である。
アジピン酸ジイソプロピルの含有(配合)量は、皮膚外用剤中、通常、1.0~10質量%であり、1.0~5.0質量%が好ましい。セバシン酸ジイソプロピルの含有(配合)量は、皮膚外用剤中、通常、2.5~10質量%であり、2.5~5.0質量%が好ましい。アジピン酸ジイソプロピルが1.0質量%未満であると、タウリンの経皮吸収性の増大効果が充分でないことがあり、10質量%を超えるとあると粘度の増加による使用感の悪化や皮膚刺激性が懸念されるためである。また、セバシン酸ジイソプロピルが2.5質量%未満であると、タウリンの経皮吸収性の増大効果が充分でないことがあり、10質量%を超えるとあると粘度の増加による使用感の悪化や皮膚刺激性が懸念されるためである。
そして、タウリンの経皮吸収を増大させるという観点からは、タウリンの1質量部に対するアジピン酸ジイソプロピルの含有(配合)量は、0.3質量部以上であり、好ましくは0.3~10質量部、より好ましくは0.8~10質量部、さらに好ましくは1.6~10質量部である。同様の観点から、タウリンの1質量部に対するセバシン酸ジイソプロピルの含有(配合)量は0.8質量部以上であり、好ましくは0.8~10質量部、より好ましくは1.6~10質量部である。
本発明の皮膚外用剤を適用する「皮膚」には、頭皮が含まれる。
本発明の「皮膚外用剤」は、水にタウリン並びにアジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種を配合する他、公知の有効成分、公知の外用基剤、エタノール等の有機溶剤を適宜に配合して、液剤、ローション剤、ゲル剤、エアゾール剤、クリーム剤等の各種皮膚外用剤として提供することができる。
以下に、実施例、比較例及び試験例を挙げて、本発明を更に詳細に説明する。
なお、表1に実施例1~5並びに比較例1~8の処方を示した。表中の数値の単位は「g」である。
実施例1~5並びに比較例3及び4は、アジピン酸ジイソプロピル又はセバシン酸ジイソプロピル、及びポリオキシエチレン硬化ヒマシ油60を表1に記載の処方量に従い秤量し、加温下で攪拌した後、精製水とタウリンを徐々に添加して、全量100gになるように皮膚外用剤を調製した。
比較例1は、タウリンを精製水に添加して、攪拌・溶解し、全量100gになるように皮膚外用剤を調製した。比較例2及び5~8は、各種成分及びポリオキシエチレン硬化ヒマシ油60を表1記載の処方量に従い秤量し、加温下で攪拌した後、精製水とタウリンを徐々に添加して、全量100gになるように皮膚外用剤を調製した。
[試験例1] in vitro ヘアレスラット摘出皮膚透過性試験
電気シェーバーで腹部を剃毛したヘアレスラット(雄、9週齢、(株)埼玉実験動物供給所)の腹部剃毛部より皮膚を摘出した。摘出した皮膚を平均有効面積0.98cm2の横型拡散セルに装着した後、ドナー側のセル内に表1に示した各外用剤(検体)3mLを加え、レシーバー側のセルにpH7.4リン酸緩衝液3mLを加えた。拡散セルの外層には37℃の恒温水を還流し、レシーバー液は撹拌子により撹拌した。

経時的にレシーバー液を一定量採取し、常法に従い液体クロマトグラフによりタウリン量を測定した。なお、タウリンは元々皮膚中に存在するため、検体を塗布しなくても経時的に僅かにレシーバー液に流出してくる。そこで、比較例1と比較例2について予備試験を行い、ポリオキシエチレン硬化ヒマシ油60の有無により経皮吸収促進効果に差が無いこと、すなわち、ポリオキシエチレン硬化ヒマシ油60にはタウリンの経皮吸収促進作用がないこと、及び再現性があることを確認した後、各実施例及び比較例の皮膚外用剤(検体)で試験を実施した。
そして、比較例2の8時間後の累積透過量(これを1.0とする)に対する実施例1~5並びに比較例3~8の8時間後の累積透過量を経皮吸収促進倍率として算出すると、実施例1では1.4倍、実施例2では1.8倍、実施例3では3.8倍、実施例4では1.4倍、実施例5では3.7倍であり、顕著な経皮吸収促進作用を示した。
一方、比較例3、4での配合量及び比較例5~8での一般的に経皮吸収促進を有する成分では、充分なタウリンの経皮吸収促進作用が認められなかった。
[試験例2]
ウサギ(雄、9~11週齢、日本白色種、北山ラベス(株))を用いた皮膚一次刺激性試験を実施した結果、セバシン酸ジイソプロピル及びアジピン酸ジイソプロピルの皮膚刺激スコアは0と非常に低値で安全な成分であることが確認された。

本発明により、タウリンの経皮吸収性を増大させることが可能となったので、タウリンの有する皮膚に対する諸機能を充分に発揮させることができる皮膚外用剤の提供を通じ、生活者の期待に応え、医薬品産業等の健全な発達に資することが期待される。
Claims (2)
- タウリン、並びに、アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種を含有することを特徴とする皮膚外用剤。
- アジピン酸ジイソプロピル及びセバシン酸ジイソプロピルの少なくとも1種の含有量が、タウリンの1質量部に対してアジピン酸ジイソプロピルでは0.3質量部以上、セバシン酸ジイソプロピルでは0.8質量部以上である請求項1に記載の皮膚外用剤。
