Abstract
Description
ADAM17と同じくADAMファミリーに属する膜結合型タンパク質分解酵素であるADAM10は、その基質として、癌の進行に関わるノッチ(Notch)、E-カドヘリン、上皮成長因子(Epidermal Growth Factor:EGF)、Erythroblastic leukemia viral oncogene homolog 2(ErbB2)などが知られており、また、ADAM17と共通してTNF-α、HB-EGFをも遊離させることも報告されている。これらのことから、複数の因子が複雑に絡む癌や炎症などの疾患において、ADAM10も病態の進行に促進的に関与していることが示唆される。特許文献18には、ADAM10阻害剤が開示されているものの、特許文献18で開示された化合物と本願化合物とはその構造が大きく異なる。

なお、以下の説明では、一般式(I)で表される化合物、もしくはその塩、またはそれらの溶媒和物を総称して「本発明のN-ヒドロキシホルムアミド誘導体」という。
まず、前記一般式(I)における各置換基について説明する。各置換基の説明における「C1~C6」、「C6~C14」は、それぞれ、炭素数が1~6、6~14の範囲内にあることを意味する。
R2,R3,R4,R5およびZにおける「置換されていてもよいC1~C6アルキル基」の「C1~C6アルキル基」とは、直鎖または分岐状のC1~C6アルキル基を意味し、具体例としては、メチル基、エチル基、n-プロピル基、イソプロピル基、n-ブチル基、イソブチル基、tert-ブチル基、sec-ブチル基、n-ペンチル基、tert-アミル基、3-メチルブチル基、ネオペンチル基、n-ヘキシル基などがあげられる。
前記、「置換されていてもよいC1~C6アルキル基」における置換基とは、水酸基、ハロゲン原子、シアノ基、ニトロ基、C1~C6アルコキシ基、カルボキシル基、C1~C6アルコキシカルボニル基などがあげられる。これらは、全ての可能な位置で一個以上置換しうる。複数個置換する場合は、それらの置換基は同一であっても異なっていてもよいし、同一の炭素原子に置換していてもよいし異なる炭素原子に置換していてもよい。
「ハロゲン原子」とは、フッ素原子、塩素原子、臭素原子、ヨウ素原子を意味する。
「C1~C6アルコキシ基」とは、アルキル部分が、前記「C1~C6アルキル基」と同義であるアルコキシ基を意味し、たとえば、メトキシ基、エトキシ基、n-プロポキシ基、イソプロポキシ基、n-ブトキシ基、イソブトキシ基、tert-ブトキシ基、sec-ブトキシ基、n-ペンチルオキシ基、tert-アミルオキシ基、3-メチルブトキシ基、ネオペンチルオキシ基、n-ヘキシルオキシ基などの直鎖または分岐状のアルコキシ基があげられる。
「C1~C6アルコキシカルボニル基」とは、オキシカルボニル部分を除いたアルキル部分が直鎖または分岐状のC1~C6アルキル基であるものを意味し、たとえば、メトキシカルボニル基、エトキシカルボニル基、n-プロポキシカルボニル基、イソプロポキシカルボニル基、n-ブトキシカルボニル基、イソブトキシカルボニル基、tert-ブトキシカルボニル基、sec-ブトキシカルボニル基、n-ペンチルオキシカルボニル基、tert-アミルオキシカルボニル基、3-メチルブトキシカルボニル基、ネオペンチルオキシカルボニル基、n-ヘキシルオキシカルボニル基などがあげられる。
R2における「置換されていてもよいアリール基」の「アリール基」とは、芳香族炭素環を表わし、C6~C14の芳香族炭素環が好ましく、たとえば、フェニル基、ナフチル基などがあげられる。
前記、「置換されていてもよいアリール基」における芳香環上の置換基とは、水酸基、ハロゲン原子、シアノ基、ニトロ基、トリフルオロメチル基、置換されていてもよいC1~C6アルキル基、C1~C6アルコキシ基、カルボキシル基、C1~C6アルコキシカルボニル基などがあげられる。これらは、全ての可能な位置で一個以上置換しうる。複数個置換する場合は、それらの置換基は同一であっても異なっていてもよいし、同一の炭素原子に置換していてもよいし異なる炭素原子に置換していてもよい。ここで、「ハロゲン原子」、「置換されていてもよいC1~C6アルキル基」、「C1~C6アルコキシ基」および「C1~C6アルコキシカルボニル基」は前記と同じ意味を示す。
R5における「C1~C6アルキルスルホニル基」とは、アルキル部分が、前記「C1~C6アルキル基」と同義であるアルキルスルホニル基を意味し、たとえば、メタンスルホニル基、エタンスルホニル基などがあげられる。
一般式(I)で表される化合物の塩としては、薬理学的に許容される塩であればとくに制限されず、たとえば、無機塩基との塩、有機塩基との塩、有機酸との塩、無機酸との塩およびアミノ酸との塩などがあげられる。無機塩基との塩の例としては、リチウム塩、ナトリウム塩、カリウム塩、カルシウム塩、マグネシウム塩などのアルカリ金属塩およびアルカリ土類金属塩などがあげられる。有機塩基との塩の例としては、トリエチルアミン塩、ピリジン塩、エタノールアミン塩、シクロヘキシルアミン塩、ジシクロヘキシルアミン塩、ジベンジルエタノールアミン塩などがあげられる。有機酸との塩の例としては、ギ酸塩、酢酸塩、酒石酸塩、マレイン酸塩、コハク酸塩、乳酸塩、リンゴ酸塩、アスコルビン酸塩、シュウ酸塩、グリコール酸塩、フェニル酢酸塩、メタンスルホン酸塩などがあげられる。無機酸との塩の例としては、塩酸塩、臭化水素酸塩、リン酸塩、スルファミン酸塩、硝酸塩などがあげられる。また、アミノ酸との塩の例としては、グリシン塩、アラニン塩、アルギニン塩、グルタミン酸塩、アスパラギン酸塩などがあげられる。
一般式(I)で表される化合物はプロドラッグの形態をとり得る。プロドラッグの例としては、一般式(I)の化合物のカルボキシル基のメチルエステル、エチルエステル、アミノアルキルエステル誘導体、一般式(I)の化合物の水酸基およびアミン官能基のアセテート、ホルメート、およびベンゾエート誘導体などがあげられるが、これらに限定されない。
上記の一般式(I)で表わされる化合物は、種々の方法で製造できるが、以下に示す製造方法により、効率よく製造することができる。
以下の製造方法において使用される「保護基」の具体例としては、tert-ブチル基、ベンジル基、o-メチルベンジル基、p-ニトロベンジル基、p-メトキシベンジル基、o-クロロベンジル基、2,4-ジクロロベンジル基、p-ブロモベンジル基、アリル基、tert-ブトキシカルボニル基、ベンジルオキシカルボニル基、o-メチルベンジルオキシカルボニル基、p-ニトロベンジルオキシカルボニル基、p-メトキシベンジルオキシカルボニル基、o-クロロベンジルオキシカルボニル基、2,4-ジクロロベンジルオキシカルボニル基、p-ブロモベンジルオキシカルボニル基、アリルオキシカルボニル基、tert-ブチルジメチルシリル基、tert-ブチルジフェニルシリル基、トリエチルシリル基、トリメチルシリル基、トリイソプロピルシリル基、メトキシメチル基、テトラヒドロピラニル基、カルボニルの保護基(たとえば、エタンジオール、プロパンジオール、メルカプトエタノール、メルカプトプロパノール、エタンジチオール、プロパンジチオールなどによる保護基)などがあげられる。
工程1においては、ヒドロキシルアミンまたはその塩を化合物(II)に付加し、一般式(III)で表される化合物を製造する。この付加反応では、ヒドロキシルアミンが塩(塩酸塩や酢酸塩など)の場合、炭酸カリウム、炭酸ナトリウム、炭酸水素ナトリウム、水酸化ナトリウム、水酸化カリウム、水酸化リチウムなどの無機塩基の存在下に行なわれる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されないが、水、テトラヒドロフラン、シクロペンチルメチルエーテル、アセトニトリル、1,4-ジオキサン、ジエチルエーテルまたはそれらの混合溶媒などが好ましい。
反応温度はとくに限定されず、通常、0~100℃で行なわれ、反応時間は、2時間から1週間が好ましい。
工程2においては、工程1で得られた化合物(III)を、一般式(IV)で表される中間体と縮合し、一般式(I)で表される化合物を製造する。中間体(IV)は、ギ酸の混合酸無水物(ギ酸と酢酸の混合酸無水物など)、ギ酸ペンタフルオロフェニルまたはギ酸とカルボジイミド(ジシクロヘキシルカルボジイミド、ジイソプロピルカルボジイミドまたは水溶性カルボジイミド)から得られる活性中間体である。反応を円滑に行なうためにトリエチルアミン、ジイソプロピルエチルアミン、ピリジン、ルチジン、コリジン、ジメチルアミノピリジンなどの有機塩基を共存させてもよい。これらの場合の幾つか(とくにカルボジイミドから活性中間体を得る場合)は1-ヒドロキシベンゾトリアゾールおよび/または4-ジメチルアミノピリジンを加えることにより反応が促進される。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されないが、クロロホルム、塩化メチレン、テトラヒドロフラン、アセトニトリル、シクロペンチルメチルエーテル、1,4-ジオキサン、ジメチルホルムアミド、ジメチルスルホキシド、ピリジンなどが好ましい。反応温度はとくに限定されず、通常、0~100℃で行なわれ、反応時間は、1~24時間が好ましい。なお、本工程において、原料の化学的性質により、ヒドロキシルアミノ基の水酸基にもCHO基が付加する場合があるが、これは、酸性、塩基性または中性条件下において低級アルコールで処理することにより目的物である化合物(I)に変換できる。低級アルコールとしては、メタノール、エタノール、プロパノールなどが好ましい。補助溶媒を使用してもよく、その場合使用する補助溶媒はとくに限定されない。
X、Y、およびZの性質によっては、前記工程1~工程2の反応において事前に保護基を使用することおよび事後に保護基の除去を行なうことが必要であることは言うまでもない。保護されていない場合、次工程あるいは次次工程の収率が低下したり、中間体の取り扱いが困難な場合がある。
工程3においては、一般式(V)で表される化合物を塩基によりアニオンにしたのち、一般式(VI)で表される化合物と反応させて化合物(VII)を製造する。使用する塩基としては、リチウムジイソプロピルアミド、リチウム(ビストリメチルシリル)アミド、リチウムテトラメチルピペラジド、ナトリウム(ビストリメチルシリル)アミド、カリウム(ビストリメチルシリル)アミド、n-ブチルリチウム、sec-ブチルリチウム、tert-ブチルリチウムなどがあげられる。これらの塩基は、単独で用いるか、または場合によって2つ以上を組み合わせて使用する。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されないが、テトラヒドロフラン、シクロペンチルメチルエーテル、テトラヒドロピラン、ジエチルエーテル、tert-ブチルメチルエーテルまたはそれらの混合溶媒などが好ましい。
反応温度は、通常-100~40℃で行なわれ、反応時間は、1~12時間が好ましい。なお、この工程においては、化合物(VI)の化学的性質により化合物(II)が得られる場合があるが、本製造目的を斟酌すると問題にならない。
工程4においては、工程3で得られた化合物(VII)を一般式(VIII)で表される化合物と反応させ、一般式(IX)で表される化合物を製造する。反応溶媒としては、化合物(VIII)が水素化ホウ素ナトリウムまたは水素化ホウ素リチウムの場合には、メタノール、エタノール、イソプロパノール、テトラヒドロフラン、シクロペンチルメチルエーテル、ジクロロメタン、クロロホルムまたはそれらの混合溶媒などが好ましく、化合物(VIII)が前記2種以外の場合は、テトラヒドロフラン、シクロペンチルメチルエーテル、テトラヒドロピラン、ジエチルエーテル、tert-ブチルメチルエーテルまたはそれらの混合溶媒などが好ましい。反応温度は、通常-100~30℃で行なわれ、反応時間は、1~12時間が好ましい。
なお、工程4の反応中あるいは反応後に、生成した化合物(IX)の水酸基が自然脱離し、一部あるいは全部が化合物(II)になる場合がある。一部の場合、それらを分離することなく工程5を行なうことができ、全部の場合、工程5を省略することができる。
工程5においては、工程4で得られた化合物(IX)を脱水反応により化合物(II)を製造する。脱水反応は、水酸基を活性化する試薬と有機塩基の組み合わせにより行なわれ、水酸基を活性化する試薬としては、塩化メタンスルホニル、塩化p-トルエンスルホニル、塩化ベンゼンスルホニル、塩化メタンスルホニル、塩化チオニル、塩化スルフリル、五塩化リンなどがあげられる。有機塩基としては、トリエチルアミン、ジイソプロピルエチルアミン、ジアザビシクロウンデセン、ジアザビシクロノネン、ピリジン、ジメチルアミノピリジン、ルチジン、コリジンなどがあげられる。好適な組み合わせとして、塩化メタンスルホニルとトリエチルアミンがあげられる。また、別の脱水試薬として、トリフェニルホスフィン-ジエチルアゾジカルボキシラート、トリフェニルホスフィン-ジイソプロピルアゾカルボキシラート、トリn-ブチルホスフィン-ジエチルアゾジカルボキシラート、トリn-ブチルホスフィン-ジイソプロピルアゾカルボキシラートなどがあげられる。反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されないが、クロロホルム、塩化メチレン、テトラヒドロフラン、シクロペンチルメチルエーテル、アセトニトリル、1,4-ジオキサン、ジメチルホルムアミドなどが好ましい。反応温度はとくに限定されず、通常、0~100℃で行なわれ、反応時間は、1~24時間が好ましい。
X,Y、およびZの性質によっては、前記工程3~工程5の反応において事前に保護基などを使用することおよび事後に保護基などの除去を行なうことが必要であることは言うまでもない。
工程6においては、一般式(X)で表される化合物またはその塩と一般式(XI)で表される化合物を無機塩基の存在下縮合させ、化合物(V)を製造する。好適な無機塩基としては、水酸化ナトリウム、水酸化カリウム、水酸化リチウム、炭酸ナトリウム、炭酸カリウム、炭酸セシウム、水酸化カルシウムなどがあげられる。ただし、J1が水酸基の場合は、一般式(X)で表される化合物またはその塩の水酸基を試薬により活性化し、一般式(XI)で表される化合物を縮合させ、化合物(V)を製造する。水酸基の活性化に適した試薬としては、アゾジカルボン酸ジエチル(DEAD)-トリフェニルホスフィン、アゾジカルボン酸ジイソプロピル-トリフェニルホスフィン、シアノメチレントリブチルホスホラン、シアノメチレントリメチルホスホラン、ブチルリチウム-クロロジフェニルホスフィンなどがあげられる。また、ブチルリチウム-クロロジフェニルホスフィンで活性化する場合は、2,6-ジメチル-1,4-ベンゾキノン、テトラフルオロ-1,4-ベンゾキノンなどのキノン類を添加する。
反応溶媒としては、反応を著しく阻害しない溶媒であればとくに限定されないが、水、メタノール、エタノール、tert-ブタノール、テトラヒドロフラン、シクロペンチルメチルエーテル、アセトニトリル、ジエチルエーテル、ジメチルエーテル、ジクロロメタン、1,4-ジオキサン、2-メトキシエタノール、N,N-ジメチルホルムアミドまたはそれらの混合溶媒などが好ましい。反応温度はとくに限定されず、通常、-80~120℃で行なわれ、反応時間は、1~24時間が好ましい。
<工程7>
工程7においては、工程3と同様にして、一般式(XII)で表される化合物を塩基によりアニオンとした後、化合物(VI)と反応させて化合物(XIII)を製造する。
<工程8>
工程8においては、工程4と同様にして、一般式(XIII)で表される化合物を一般式(VIII)で表される化合物と反応させ、化合物(XIV)を製造する。
XおよびYの性質によっては、前記工程7~工程8の反応において事前に保護基などを使用することおよび事後に保護基などの除去を行うことが必要であることは言うまでもない。
<工程9>
工程9においては、工程5と同様にして、化合物(XIV)を脱水反応により化合物(XV)を製造する。
なお、工程9において保護基P1が自然脱離し、一部あるいは全部が化合物(XVI)になる場合がある。一部の場合、それらを分離することなく工程10を行なうことができ、全部の場合、工程10を省略する事ができる。
<工程10>
工程10においては、一般式(XV)で表される化合物に使用されている保護基P1の種類にあわせて、公知の方法によりその脱保護を行い、一般式(XVI)で表される化合物を製造する。
<工程11>
工程11においては、工程6と同様にして、化合物(XVI)と一般式(X)で表される化合物またはその塩を縮合させ、化合物(II)を製造する。
本発明のN-ヒドロキシホルムアミド誘導体は、優れたADAM17阻害作用を示し、ADAM17が関与する種々の疾患の治療薬として有用であり、たとえば、リウマチ性関節炎(RA)、変形性関節症(OA)、全身性エリテマトーデス、多発性硬化症、ベーチェット病、シェーグレン症候群などの自己免疫疾患、およびそれらに伴う各種臓器炎症、喘息、アトピー性皮膚炎、鼻閉、鼻炎などのアレルギー性疾患、クローン病などを含む炎症性腸疾患、腎炎、肝炎、中枢神経系の炎症性疾患、乾癬、強皮症、サルコイドーシスなどの皮膚炎関連疾患、歯周病、心臓血管性疾患、動脈硬化症、糖尿病、重症筋無力症、急性感染症、発熱、貧血、敗血症、虚血後再灌流障害、マラリア、マイコバクテリア感染、髄膜炎、うっ血性心不全、線維症、悪液質、移植片拒絶反応、血管新生に関する疾患、強直性脊椎炎、乾癬性関節炎、成人スティル病、ウェゲナー肉芽腫症、多発性筋炎、皮膚筋炎、坐骨神経痛、複合性局所疼痛症候群、放射線障害、高酸素症肺胞傷害、HIV、緑内障、特発性肺線維症、気管支肺形成異常、網膜疾患、骨粗鬆症、腎虚血、心筋梗塞、脳卒中、脳虚血、糸球体腎炎、特発性線維化性肺胞炎、脈管炎、可逆性気道閉塞、成人呼吸促進症候群、慢性閉塞性肺疾患(COPD)、気管支炎、種々の癌、移植臓器の損傷予防、腫瘍増殖または転移阻止などがあげられる。
また、本発明のN-ヒドロキシホルムアミド誘導体のうち、ADAM17の阻害作用に加え、ADAM10の阻害作用を併せ持つものは、ADAM17とADAM10の両方が関与する疾患(例えば、種々の癌やリウマチ性関節炎等の炎症性疾患など)の治療薬としてより有用である。
本発明のN-ヒドロキシホルムアミド誘導体の投与量は、投与対象、投与ルート、症状などの条件によって適宜決定すればよく、たとえば、成人の患者に対して経口投与する場合、有効成分である本化合物を通常1回量として、約0.1~100mg/kg、好ましくは1~40mg/kgの範囲であればよく、1日1~3回投与するのが好ましい。
また、本発明のN-ヒドロキシホルムアミド誘導体のADAM17阻害効果は、50%抑制濃度(IC50)で0.01nM~1000nMが好ましい。
なお、以下に示す1H-NMRスペクトルは、重クロロホルム(CDCl3)または重ジメチルスルホキシド(DMSO-d6)を溶媒とし、テトラメチルシラン(TMS)を内部標準として、ECA400型スペクトルメーター(400MHz、日本電子株式会社製)で測定したものである。化学シフトの測定結果は、δ値をppmで示し、結合定数のJ値をHzで示した。略号のsはsinglet、dはdoublet、tはtriplet、qはquartet、ddはdoublet doublet、mはmultiplet、brはbroadを意味する。低分解能質量スペクトル(高速原子衝撃法:FAB-MS)測定には、日本電子JMS-HX-110A型を、質量スペクトル(エレクトロスプレーイオン化法:ESI-MS)測定には、サーモフィッシャーサイエンティフィック(株)のExactiveをそれぞれ使用した。
N-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(4-ジエチルアミノメチルフェニル)エチル]-N-ヒドロキシホルムアミド(I-1)
1-ブロモ-2-ブチン 2.12g(15.9mmol)のジメチルスルホキシド溶液(30mL)に4-メチルスルホニルフェノール 2.88g(15.9mmol)を加えて溶解させた後、炭酸カリウム 2.64g(19.1mmol)を加えた。6時間撹拌した後、飽和食塩水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した。1-ブト-2-イニルオキシ-4-メタンスルホニルベンゼン(V-1)3.39g(15.11mmol)を粗精製物として得た(収率95%)。物性値を以下に示す。
MS (FAB) m/z : 225 (M+H)+.
1H-NMR (CDCl3):δ 7.88(2H, m), 7.10(2H, m), 4.73(2H, m), 3.04(3H, s), 1.87(3H, t, J = 2.3 Hz).
前記(1-1)で得た化合物(V-1)1.36g(6.08mmol)のテトラヒドロフラン(70mL)溶液に、アルゴン雰囲気下-78℃でリチウムジイソプロピルアミドの2.0Mヘキサン-ヘプタン-エチルベンゼン溶液3.65mL(7.29mmol)を加え、30分間撹拌した後、リチウムヘキサメチルジシラジドの1.0M-テトラヒドロフラン溶液12.16mL(12.16mmol)と、4-(tert-ブトキシカルボニルアミノメチル)安息香酸メチル(VI-1)1.61g(6.08mmol)のテトラヒドロフラン溶液5mLを加えた。-78℃にて5分間撹拌した後、徐々に室温まで昇温し、1時間撹拌した。飽和食塩水を加えた後、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄した。無水硫酸マグネシウムで乾燥した後、溶媒を減圧下留去した。シリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=2:1→1:1)にて精製し、{4-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)アセチル]ベンジル}カルバミン酸tert-ブチルエステル(VII-1)2.02g(4.41mmol)を無色飴状物質として得た(収率72%)。物性値を以下に示す。

前記(1-2)で得た化合物(VII-1)2.02g(4.41mmol)のメタノール(50mL)溶液に、0℃で水素化ホウ素ナトリウム 167mg(4.41mmol)を加えた。2時間30分間撹拌した後、飽和食塩水と飽和塩化アンモニウム水溶液を加えた。メタノールを減圧下留去した後、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥した。溶媒を減圧下留去し、{4-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-ヒドロキシエチル]ベンジル}カルバミン酸tert-ブチルエステル(IX-1)2.04g(4.41mmol)を無定形状固体の粗精製物として得た(収率99%)。物性値を以下に示す。

前記(1-3)で得た化合物(IX-1)2.04g(4.41mmol)とトリエチルアミン 3.1mL(22.1mmol)のジクロロメタン(45mL)溶液に0℃で塩化メタンスルホニル 0.7mL(8.8mmol)を加えた。3時間30分間撹拌した後、飽和食塩水を加え、クロロホルムで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した。溶媒を減圧下留去した後、残渣をシリカゲルカラムクロマトグラフィー(クロロホルム:酢酸エチル=3:1)で精製し、{4-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)ビニル]ベンジル}カルバミン酸tert-ブチルエステル(II-1a)1.77g(4.00mmol)を無色無定形状固体として得た(収率91%)。物性値を以下に示す。

前記(1-4)で得た化合物(II-1a)295mg(0.67mmol)のメタノール(5mL)溶液に、0℃で4M-塩酸/ジオキサン2mLを加えて10分間撹拌した後、室温まで昇温し、2時間撹拌した。溶媒を減圧下留去した後、メタノール20mLを加えて再び、溶媒を減圧下留去し、4-[(E)-2-(4-ブト-2-イニルオキシベンゼンスルホニル)ビニル]ベンジルアミン塩酸塩(II-1b)を無色固体として得た。物性値を以下に示す。

前記(1-5)で得た化合物(II-1b)のメタノール(6mL)溶液に、0℃でアセトアルデヒド1mL、トリアセトキシヒドロほう酸ナトリウム212mg(1.00mmol)および酢酸3滴をそれぞれ加えた後、室温にて1時間30分間撹拌した。飽和食塩水と飽和重曹水を加えた後、溶媒を減圧下留去した。クロロホルムで抽出し、有機層を飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥した。溶媒を減圧下留去した後、残渣をシリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=20:1→10:1)にて精製し、{4-[(E)-2-(4-ブト-2-イニルオキシベンゼンスルホニル)ビニル]ベンジル}ジエチルアミン(II-1c)111.7mg(0.28mmol)を淡黄色無定形状固体として得た(2段階収率42%)。物性値を以下に示す。

前記(1-6)で得た化合物(II-1c)108mg(0.27mmol)のテトラヒドロフラン(8mL)溶液に、50%ヒドロキシルアミン溶液(3mL)を加え、室温にて25時間攪拌した。反応溶液を減圧下留去した後、水を加え、クロロホルムで抽出した。有機層を飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥し、減圧濃縮した。N-[2-(4-ブト-2-イニルオキシ-ベンゼンスルホニル)-1-(4-ジエチルアミノメチルフェニル)エチル]ヒドロキシルアミン(III-1)92.9mg(0.22mmol)を淡黄色無定形状固体として得た(収率81%)。物性値を以下に示す。

ギ酸1mLを0℃に冷却し、無水酢酸0.3mLを滴下した後、30分間撹拌することでギ酸―酢酸の混合酸無水物溶液を調製した。前記(1-7)で得た化合物(III-1)92mg(0.21mmol)のテトラヒドロフラン(3mL)溶液に、0℃で、先に調製したギ酸―酢酸の混合酸無水物溶液0.6mLを加え、室温で4時間撹拌した。反応混合物を減圧濃縮した後、トルエンで共沸した。得られた油状物をクロロホルム 2mLおよびメタノール10mLに溶解し、12時間撹拌した。この溶液を減圧濃縮し、得られた油状物をクロロホルムに溶解し、飽和重曹水を加えて中和した。クロロホルムで抽出した後、飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した。中圧シリカゲルカラムクロマトグラフィー(クロロホルム:メタノール=95:5→75:25)により精製し、N-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(4-ジエチルアミノメチルフェニル)エチル]-N-ヒドロキシホルムアミド(I-1)53.9mg(0.11mmol)を淡黄色無定形状固体として得た(収率52%)。物性値を以下に示す。

前記実施例1(1-5)で得た化合物(II-1b)283mg(0.75mmol)のピリジン溶液(5mL)に、室温でメトキシアセチルクロリド0.2mL(1.85mmol)を滴下した。2日後、飽和食塩水を加え、酢酸エチルで抽出し、飽和食塩水で洗浄した。無水硫酸マグネシウムで乾燥し、溶媒を減圧下留去した後、シリカゲルクロマトグラフィー(クロロホルム:メタノール=20:1→10:1)にて精製し、化合物(II-3)296mg(0.71mmol)を淡黄色固体として得た(収率94%)。物性値を以下に示す。

4-メチルスルホニルフェノール10g(58.07mmol)のN,N-ジメチルホルムアミド溶液(150mL)にイミダゾール9.9g(145.18mmol)を加えて溶解させた後、tert-ブチルジメチルクロロシラン10.5g(69.7mmol)を加えて撹拌した。反応終了後、飽和食塩水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で3回洗浄し、無水硫酸マグネシウムで乾燥した。シリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=5:1→3:1)にて精製し、tert-ブチル-(4-メタンスルホニルフェノキシ)ジメチルシラン(XII-6)15.97g(55.75mmol)を得た(収率96%)。物性値を以下に示す。

前記(6-1)で得たtert-ブチル-(4-メタンスルホニルフェノキシ)ジメチルシラン(XII-6)2.0g(6.98mmol)のテトラヒドロフラン溶液に、アルゴン雰囲気下-78℃でリチウムジイソプロピルアミドの2.0Mヘキサン-ヘプタン-エチルベンゼン溶液4.19mL(8.37mmol)を加え、リチウムヘキサメチルジシラジドの1.0M-テトラヒドロフラン溶液6.98mL(6.98mmol)と、4-モルホリン-4-イルメチル安息香酸メチル(IX-6)1.6g(6.98mmol)のテトラヒドロフラン溶液5mLを加えた。その後、撹拌しながら徐々に室温まで昇温した。反応終了後、飽和食塩水を加え、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄した。無水硫酸マグネシウムで乾燥した後、溶媒を減圧下留去した。2-[4-(tert-ブチルジメチルシラニロキシ)ベンゼンスルホニル]-1-(4-モルホリン-4-イルメチルフェニル)エタノン(XIII-6)3.74gを粗精製物として得た。
前記(6-2)で得た化合物(XIII-6)3.74g(6.98mmol)のメタノール(50mL)溶液に、0℃で水素化ホウ素ナトリウム 264mg(6.98mmol)を加えた。1時間撹拌した後、飽和食塩水を加えた。メタノールを減圧下留去した後、酢酸エチルで抽出し、有機層を飽和食塩水で洗浄した後、無水硫酸マグネシウムで乾燥した。溶媒を減圧下留去し、残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル→酢酸エチル:メタノール=10:1)で精製し、2-[4-(tert-ブチルジメチルシラニロキシ)ベンゼンスルホニル]-1-(4-モルホリン-4-イルメチルフェニル)エタノール(XIV-6)を2.19g(4.48mmol)得た。物性値を以下に示す。

前記(6-3)で得た2-[4-(tert-ブチルジメチルシラニロキシ)ベンゼンスルホニル]-1-(4-モルホリン-4-イルメチルフェニル)エタノール(XIV-6)2.19g(4.48mmol)のジクロロメタン溶液(45mL)にトリエチルアミン3.10mLを加え、0℃にて撹拌した。塩化メタンスルホニル0.61mL(8.91mmol)を加えた後、室温にて8時間撹拌した。さらにトリエチルアミン3.10mL、塩化メタンスルホニル0.61mLをそれぞれ加え、4時間撹拌した。飽和食塩水を加えてクロロホルムで抽出し、有機層を飽和食塩水で洗浄した。無水硫酸マグネシウムで乾燥した。溶媒を減圧下留去し、残渣をシリカゲルカラムクロマトグラフィー(ヘキサン:酢酸エチル=1:2→酢酸エチル→酢酸エチル:メタノール=10:1)で精製し、4-[(E)-2-(4-モルホリン-4-イルメチルフェニル)エテンスルホニル]フェノール(XVI-6)を0.757g(2.11mmol)得た(収率47%)。物性値を以下に示す。

前記(6-4)で得た4-[(E)-2-(4-モルホリン-4-イルメチルフェニル)エテンスルホニル]フェノール(XVI-6)187mg(0.520mmol)のN,N-ジメチルホルムアミド溶液に炭酸カリウム108mg(0.780mmol)および1-ブロモ-2-ブチン0.094mL(1.04mmol)を加え、4時間撹拌した。飽和食塩水を加え、酢酸エチルで抽出した。有機層を飽和食塩水で洗浄し、無水硫酸マグネシウムで乾燥した。溶媒を減圧下留去し、残渣をシリカゲルカラムクロマトグラフィー(酢酸エチル:クロロホルム=1:1→1:2→酢酸エチル)で精製し、4-[4-[(E)-2-(4-ブト-2-イニルオキシベンゼンスルホニル)ビニル]ベンジル]モルホリン(II-6)を0.0238g(0.0578mmol)得た(収率11%)。物性値を以下に示す。

ADAM17阻害実験
ADAM17の塩基配列はモス(Moss)らにより報告されている(Moss, M. L. et al., Nature 1997, 385, 733-736)。したがって、ヒト単球細胞株THP-1細胞から定法にしたがってcDNAを取得し、これを発現ベクターに組み込み、次いでこのベクターを哺乳動物細胞あるいは昆虫細胞に形質転換し、ADAM17を発現させ取得した。
ADAM17阻害実験は、前記のようにして得られたADAM17を酵素として用い、また基質としては、膜結合型TNFのADAM17切断配列を含んだ蛍光合成基質 Nma(N-メチルアントラニル酸)-Leu-Ala-Gln-Ala-Val-Arg-Ser-Ser-Lys-Dnp(ジニトロフェニル)-D-Arg-NH2 を用いて、被験物質存在下または非存在下におけるADAM17の活性を測定することにより行なった。以下にADAM17阻害実験の方法を示した。
すなわち、アッセイバッファーA(200mM塩化ナトリウム、5mM塩化カルシウム、10μM硫酸亜鉛、0.004%アジ化ナトリウム、2mg/mL牛血清アルブミンを含む、50mMトリス-塩酸緩衝液(pH7.5))にて調整した14unitsの酵素液(25℃の条件下で1pmolの基質を1分間で分解する酵素量を1unitと定義した)90μL、およびアッセイバッファーB(200mM塩化ナトリウム、5mM塩化カルシウム、10μM硫酸亜鉛、0.004%アジ化ナトリウム、0.05%PLURONIC F-68を含む、50mMトリス-塩酸緩衝液(pH7.5))にて20μMに調製した蛍光合成基質90μLを混合し、37℃で1.5時間反応させた。この後、蛍光強度計(Fluoroskan Ascent)にて、励起波長355nm、測定波長460nmの条件で測定し、酵素活性を求めた。
被験化合物の存在下および非存在下の酵素活性より阻害率を求め、50%抑制濃度(IC50)を算出した。
本試験により得られた、本発明のN-ヒドロキシホルムアミド誘導体のADAM17に対する50%阻害濃度を表1に示す。
ADAM10阻害実験
THP-1細胞から定法にしたがってADAM10のcDNAを取得し、これを発現ベクターに組み込み、次いでこのベクターを哺乳動物細胞あるいは昆虫細胞に形質転換し、ADAM10を発現させ取得した。ADAM10阻害実験は、前記のようにして得られたADAM10を酵素として用い、また基質としては、蛍光合成基質 Nma(N-メチルアントラニル酸)-Leu-Ala-Gln-Ala-Val-Arg-Ser-Ser-Lys-Dnp(ジニトロフェニル)-D-Arg-NH2 を用いて、被験物質存在下または非存在下におけるADAM10の活性を測定することにより行なった。以下にADAM10阻害実験の方法を示した。

被験化合物の存在下および非存在下の酵素活性より阻害率を求め、50%抑制濃度(IC50)を算出した。
本試験により得られた本発明のN-ヒドロキシホルムアミド誘導体のADAM10に対する50%阻害濃度を表2に示す。

- 一般式(I)で表される化合物が、
N-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(4-ジエチルアミノメチルフェニル)エチル]-N-ヒドロキシホルムアミド、
N-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(4-ジメチルアミノメチルフェニル)エチル]-N-ヒドロキシホルムアミド、
N-{4-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(ホルミルヒドロキシアミノ)エチル]ベンジル}-2-メトキシアセトアミド、
N-{4-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(ホルミルヒドロキシアミノ)エチル]ベンジル}メタンスルホンアミド、
N-{4-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(ホルミルヒドロキシ-アミノ)エチル]ベンジル}ベンズアミド、
N-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(4-モルホリン-4-イルメチルフェニル)エチル]-N-ヒドロキシホルムアミド、
N-ヒドロキシ-N-[1-(4-モルホリン-4-イルメチルフェニル)-2-(4-ペント-2-イニルオキシベンゼンスルホニル)エチル]ホルムアミド、
N-[2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-(4-ジメチルアミノフェニル)エチル]-N-ヒドロキシホルムアミド、

N-{2-(4-ブト-2-イニルオキシ-ベンゼンスルホニル)-1-[4-(3-モルホリン-4-イルプロピル)フェニル]エチル}-N-ヒドロキシホルムアミド、
N-{2-(4-ブト-2-イニルオキシベンゼンスルホニル)-1-[4-(4-モルホリン-4-イルブチル)-フェニル]-エチル}-N-ヒドロキシホルムアミド、
N-{4-[1-(ホルミルヒドロキシアミノ)-2-(4-ペント-2-イニルオキシベンゼンスルホニル)エチル]ベンジル}メタンスルホンアミド、または、
N-{4-[1-(ホルミルヒドロキシアミノ)-2-(4-オクト-2-イニルオキシベンゼンスルホニル)エチル]ベンジル}メタンスルホンアミド
である、請求項1に記載の化合物、もしくはその塩、またはそれらの溶媒和物。 - 請求項1あるいは2に記載の化合物、もしくはその塩、またはそれらの溶媒和物を有効成分として含有してなる医薬。
- ADAM17阻害剤である請求項3に記載の医薬。
- リウマチ性関節炎、全身性エリテマトーデス、クローン病、ベーチェット病、多発性硬化症、シェーグレン症候群、敗血症、急性感染症、喘息、アトピー性皮膚炎、乾癬もしくは癌の予防または治療剤である請求項3に記載の医薬。
