Description
本発明は、エネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、アセチル-CoAカルボキシラーゼ2(ACC2)抑制剤及びピルビン酸デヒドロゲナーゼキナーゼ4(PDK4)抑制剤に関する。
肥満は、糖質や脂質等の摂取エネルギーが脂肪として皮下あるいは内臓周囲に過剰に蓄積され、通常に比べ体重が多くなった状態のことを言う。また、近年、メタボリックシンドロームという概念が注目されているが、これは、内臓脂肪の過剰蓄積(肥満)を基盤とし、耐糖能異常(高血糖)、高中性脂肪(高脂血症)、高血圧を合併する動脈硬化易発症状態のことで、生活習慣病への罹患リスクが高まっている状態のことをいう。
日本においては、2005年に発表されたメタボリックシンドロームの診断基準によれば、男性であればウエストが85cm以上、女性であれば90cm以上の者であって、(1)血中トリグリセリドが150mg/dl以上又はHDLコレステロールが40mg/dl未満であること、(2)高血糖(空腹時血糖が110mg/dl以上)であること、(3)高血圧(130/85mHg以上)であることの3項目のうち2項目以上が当てはまる者は、メタボリックシンドロームに該当する。そして、当該診断基準に基づく厚生労働省の調査(2006年5月)によれば、日本人の1300万人がメタボリックシンドロームに該当し、予備群も含めると2700万人に達する。
肥満やメタボリックシンドロームの該当者の増加は、医療費の増大を招くこともあり、肥満及びメタボリックシンドロームは、日本のみならず世界規模の大きな問題となっている。
肥満は、摂取エネルギー量が消費エネルギー量を上回ることにより誘導されることから、肥満を改善するためには、脂質や糖質などの摂取エネルギー量を減らすか、何らかの方法により生体の代謝を促進し、消費エネルギー量を増加させる方法が考えられる。すなわち、肥満やメタボリックシンドロームの予防・改善には、食生活の改善や運動が有効な方法と考えられている。
エネルギー消費を促進させる化合物としては、交感神経活性化作用を有するカフェインや唐辛子などが知られている(非特許文献1-3)。具体的には、コーヒー中のカフェインが熱産生を増加させることが知られている(非特許文献4)。しかしながら、カフェインには交感神経興奮作用やカフェイン中毒、利尿作用、胎児への影響など、種々の副作用も知られている他、カフェインは苦味を有することから、安全性や呈味等の実用性の点で満足できるものではない。
また、近年では、辛味の少ないカプサイシンの類縁体としてカプシエイトが見出され、エネルギー消費を亢進する作用が確認されている(非特許文献5)。

一方、運動は主に筋肉におけるエネルギー代謝を活性化し、エネルギー消費が増加することから、肥満やメタボリックシンドロームの予防・改善には有効な方法であるが、現代社会において、定期的に運動を行うことは現実的には難しく、それが肥満やメタボリックシンドローム人口増加の大きな要因となっている。従って、安全で連用でき、効果的にエネルギー消費を促進する物質や方法の開発が望まれている。
また、運動の作用を高めることが出来れば、肥満やメタボリックシンドロームの予防・改善に有効な手段になると考えられる。運動効果を高める素材としては茶カテキンが報告されているが(特許文献1)、それ以外には殆ど知られていない。
さらに、エネルギー代謝の活性化、糖質や脂質燃焼の促進は、運動機能の向上にも繋がると考えられる。
また、運動の作用を高めることが出来れば、肥満やメタボリックシンドロームの予防・改善に有効な手段になると考えられる。運動効果を高める素材としては茶カテキンが報告されているが(特許文献1)、それ以外には殆ど知られていない。

また、クロロゲン酸類が、グルコース-6-ホスファターゼ阻害作用(非特許文献8)、PPAR(ペルオキシソーム増殖剤活性化受容体)を活性化することによる脂質代謝活性化作用、高インスリン血症や高レプチン血症の予防改善作用(特許文献2)、酸化ストレスを低減することによる降圧作用(非特許文献9)を有すること、3,4-ジクロロゲン酸、3,5-ジクロロゲン酸、3,4,5-トリクロロゲン酸が、マルターゼ阻害作用(非特許文献10)を有すること、ネオクロロゲン酸及びフェルロイルキナ酸が、カルニチンパルミトイルトランスフェラーゼ活性化作用を有することが知られている(特許文献3)。

本発明は、安全性が高く、優れたエネルギー消費促進作用、脂質燃焼促進作用、糖質燃焼促進作用、運動効果向上作用又は運動機能向上作用を有し、肥満やメタボリックシンドロームの発症リスクの低下・予防・改善、運動機能の向上等に有効な医薬品、医薬部外品、飲食品、ペットフード等の飼料等を提供することに関する。
本発明者らは、エネルギー消費、脂質燃焼、糖質燃焼及び運動効果ついて検討したところ、クロロゲン酸類が、エネルギー消費、脂質燃焼、糖質燃焼を有意に促進すると共に、アセチル-CoAカルボキシラーゼ2(ACC2)抑制作用、ピルビン酸デヒドロゲナーゼキナーゼ4(PDK4)抑制作用を有し、運動効果及び運動機能を有意に向上することを見出した。
本発明のエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤及び運動機能向上剤は、優れたエネルギー消費促進作用、脂質燃焼促進作用、糖質燃焼促進作用、運動効果向上作用、運動機能向上作用を有する。従って、本発明によれば、肥満やメタボリックシンドロームの発症リスクの低下・予防・改善、運動効果向上、及び運動機能向上のための、医薬品、医薬部外品、飲食品、ペットフード等の飼料を提供できる。
本発明において、クロロゲン酸類としては、キナ酸の3位、4位及び5位から選ばれる1~2の水酸基がカフェ酸及び/又はフェルラ酸とエステル結合した化合物が挙げられ、具体的には、3-カフェオイルキナ酸、4-カフェオイルキナ酸、5-カフェオイルキナ酸(クロロゲン酸)、3,4-ジカフェオイルキナ酸、3,5-ジカフェオイルキナ酸、4,5-ジカフェオイルキナ酸等が挙げられ、これらを2種以上混合して使用しても良い。更に、3-フェルロイルキナ酸、4-フェルロイルキナ酸、5-フェルロイルキナ酸等のフェルロイルキナ酸類を含んでも良い。
クロロゲン酸類の塩としては、ナトリウム、カリウム等のアルカリ金属との塩、マグネシウム、カルシウム等のアルカリ土類金属との塩、モノエタノールアミン、ジエタノールアミン、トリエタノールアミン等の有機アミンとの塩、アルギニン、リジン、ヒスチジン、オルニチン等の塩基性アミノ酸との塩等が挙げられる。また、本発明のクロロゲン酸類は、カルボキシル基が遊離のものを含むものである。
クロロゲン酸類又はその塩は、一般的な化学合成によって製造されたもの、天然中から単離・精製された抽出物、或いはこれらを含有する天然物のまま使用しても良い。当該抽出は、常法により行えば良く、抽出物としては、例えば、コーヒー豆、リンゴ、ブドウ、タマネギ、サツマイモ等由来のものが挙げられ、コーヒー豆由来のものが好ましい。
クロロゲン酸類又はその塩を多く含む市販製剤としては、フレーバーホルダーRC-30R(長谷川香料(株))、生コーヒー豆エキスP(オリザ油化(株))、OXCH100(東洋醗酵(株))等が挙げられ、これら市販品をさらに精製し、クロロゲン酸類又はその塩の純度を高めることも可能である。クロロゲン酸類又はその塩の製造方法としては例えば、特開2006-241006公報、特開2006-306799公報、特開2008-94758公報、特開2008-94759公報等に開示された方法を用いることができる。
一般に、生体のエネルギー代謝は厳密に調節されており、エネルギー産生量/バランスの恒常性が保たれていることから、特定の分子の発現量や活性の変化が、個体レベルで見た場合のエネルギー代謝にそのまま反映されることは稀である。すなわち、特定の遺伝子発現量や酵素活性の結果のみでは、個体レベルのマクロなエネルギー状態を正確に理解することは出来ない。本発明は、個体レベルでエネルギー消費量を測定することにより真に有効な素材の探索を行い、クロロゲン酸類がエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤として有効であることを見出したことに基づくものである。
本発明においてエネルギー消費とは、栄養素(エネルギー源)が生体各組織において代謝され、化学エネルギーあるいは熱エネルギーに変換されることを言い、エネルギー消費量はその過程で消費される酸素量から算出されるものであり、個体レベルのマクロな物理化学エネルギー産生量を指す。すなわち、エネルギー消費促進作用とは、上記のように定義されるエネルギー消費量を増加させる作用のことを言う。脂質燃焼とは脂肪酸が生体各組織において代謝され、化学エネルギーあるいは熱エネルギーに変換されることを言う。
脂質燃焼量は、その酸化的代謝過程で消費される酸素量と排出される二酸化炭素量から例えば下記のPeronnetらの式により算出されるものであり、個体レベルの脂質由来のエネルギー産生量を指す。すなわち、脂肪燃焼促進作用とは、上記のように定義される脂肪燃焼量を増加させる作用のことを言う。
糖質燃焼とは、糖質が生体各組織において代謝され、化学エネルギーあるいは熱エネルギーに変換されることを言い、糖質燃焼量はその過程で消費される酸素量と排出される二酸化炭素量から例えば下記のPeronnetらの式により算出されるものであり、個体レベルの糖質由来のエネルギー産生量を指す。すなわち、糖質燃焼促進作用とは、上記のように定義される糖質燃焼量を増加させる作用のことを言う。

本発明において、運動効果向上作用とは、元来、運動によりもたらされる好ましい効果(エネルギー消費促進効果、脂質燃焼促進効果、糖質燃焼促進効果、抗肥満効果、抗メタボリックシンドローム効果等)を、運動のみ行う場合に比較し、より高める効果のことをいう。
また、運動機能向上作用とは、例えば嫌気性代謝閾値で表されるような運動時の有酸素代謝能等、身体能力を向上させる作用のことをいう。

本発明において、アセチル-CoAカルボキシラーゼ2抑制作用とは、具体的には、ACC2 mRNAの発現抑制作用、ACC2タンパク質の発現抑制作用、ACC2活性抑制作用等が挙げられる。ACC2は、筋肉や肝臓で多く発現し、アセチルCoAをマロニルCoAへ変換する作用を有している。ACC2ノックアウトマウスの研究から、ACC2の抑制は脂質及び糖質燃焼を促進し、エネルギー消費を促進することが明らかになっている(Choi CS. et al. Proc Natl Acad Sci USA 2007; 104:16480-16485)。また、ACCの阻害剤に関する研究知見(Savage DB et al. J Clin Invest 2006; 116:817-824)などとも考え合わせると、ACC2の抑制剤はエネルギー代謝の促進剤として有効であると云える。
本発明において、ピルビン酸デヒドロゲナーゼキナーゼ4(PDK4)抑制作用とは、具体的には、PDK4 mRNAの発現抑制作用、PDK4タンパク質の発現抑制作用、PDK4活性抑制作用等が挙げられる。PDK4はピルビン酸複合体をリン酸化することにより、生体のエネルギー源のスイッチングを担う酵素であり、PDK4の抑制は、ピルビン酸からアセチル-CoAへの脱炭酸反応を担うピルビン酸デヒドロゲナーゼ複合体の活性を高めることにより、グルコースからのアセチル-CoA産生を高める(Sugden, M.C., and Holness, M.J. (2003) Am. J. Physiol. Endocrinol. Metab. 284, E855-E862)。すなわち、PDK4の抑制は糖質の利用(酸化)の促進に繋がることが知られている。
これら作用は、これまで報告されている脂肪酸合成酵素阻害作用(肝臓や脂肪組織において脂肪酸合成を担う酵素である脂肪酸合成酵素をin vitroで阻害する作用)、血中グルコース吸収抑制作用(クロロゲン酸を含有するコーヒーをスクロースと同時に飲用すると、グルコースの吸収量が抑制される作用)、グルコース-6-ホスファターゼ阻害作用(糖新生を司る酵素であるグルコース-6-ホスファターゼの活性をin vitroで阻害する作用)、PPAR活性化作用(PPAR依存性の遺伝子転写を促進する作用)、高インスリン血症/高レプチン血症の予防改善作用(インスリン抵抗性やレプチン抵抗性の結果生じる高インスリン血症や高レプチン血症を予防改善する作用)、マルターゼ阻害作用(マルトース加水分解酵素であるマルターゼをin vitroで阻害する作用)、カルニチンパルミトイルトランスフェラーゼを活性化作用(カルニチンとアシル-CoAから、アシルカルニチンを生成する酵素である肝臓カルニチンパルミトイルトランスフェラーゼの酵素活性を促進する作用)とは本質的に異なる作用である。
クロロゲン酸類又はその塩は、後記実施例に示すように、エネルギー消費促進作用、脂質燃焼促進作用、糖質燃焼促進作用、運動効果向上作用、運動機能向上作用、ACC2抑制作用及びPDK4抑制作用を有する。従って、本発明のクロロゲン酸類又はその塩は、ヒトを含む動物に摂取又は投与して、エネルギー消費促進、脂質燃焼促進、糖質燃焼促進、運動効果向上、ACC2抑制、PDK4抑制、肥満やメタボリックシンドロームの発症リスクの低下・予防・改善、運動機能向上を図るための方法に使用することができる。また、本発明のクロロゲン酸類又はその塩は、エネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤及びPDK4抑制剤(以下、「エネルギー消費促進剤等」という)となり得、また、当該エネルギー消費促進剤等を製造するために使用することができる。このとき、当該エネルギー消費促進剤等には、当該クロロゲン酸類又はその塩を単独で、又はこれ以外に、必要に応じて適宜選択した担体等の、配合すべき後述の対象物において許容されるものを使用してもよい。なお、当該製剤は配合すべき対象物に応じて常法により製造することができる。斯かるエネルギー消費促進剤等は、エネルギー消費促進、脂質燃焼促進、糖質燃焼促進、運動効果向上、ACC2抑制、PDK4抑制、肥満やメタボリックシンドロームの発症リスクの低下・予防・改善、運動機能を向上するための医薬品、医薬部外品、飲食品、ペットフード等の飼料の有効成分として配合して使用可能である。
本発明のエネルギー消費促進剤等を医薬品の有効成分として用いる場合、当該医薬品は任意の投与形態で投与され得る。投与形態としては、経口、経腸、経粘膜、注射等が挙げられるが、経口投与が好ましい。経口投与のための製剤の剤型としては、例えば錠剤、被覆錠剤、カプセル剤、軟カプセル剤、顆粒剤、散剤、粉剤、徐放性製剤、懸濁液、エマルジョン剤、トローチ剤、内服液、糖衣錠、丸剤、細粒剤、シロップ剤、エリキシル剤、乳液等が挙げられる。非経口投与としては、静脈内注射、筋肉注射剤、吸入、輸液、坐剤、吸入薬、経皮吸収剤、点眼剤、点鼻剤、クリーム、ジェル、ローション、貼付剤、ゲル、ペースト等が挙げられる。
また、斯かる製剤では、本発明のエネルギー消費促進剤等を単独で、又は他の薬学的に許容される担体と組み合わせて使用してもよい。斯かる担体としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤、希釈剤、浸透圧調整剤、流動性促進剤、吸収助剤、pH調整剤、乳化剤、防腐剤、安定化剤、酸化防止剤、着色剤、紫外線吸収剤、保湿剤、増粘剤、光沢剤、活性増強剤、抗炎症剤、殺菌剤、矯味剤、矯臭剤、増量剤、界面活性剤、分散剤、緩衝剤、保存剤、固着剤、香料、被膜剤等が挙げられる。
上記担体の具体例としては、乳糖、カオリン、ショ糖、結晶セルロース、コーンスターチ、タルク、寒天、ペクチン、ステアリン酸、ステアリン酸マグネシウム、レシチン、塩化ナトリウム等の固形状坦体が挙げられ、グリセリン、落花生油、ポリビニルピロリドン、オリーブ油、エタノール、ベンジルアルコール、プロピレングリコール、水等の液状坦体が挙げられる。
また、上記製剤には、更にカフェイン等のエネルギー消費促進物質を配合することができる。カフェインを配合する場合、クロロゲン酸類又はその塩とカフェインの含有量の質量比率は、クロロゲン酸換算で、クロロゲン酸類/カフェインが、20/1~1/1であるのが好ましく、15/1~1/1がより好ましく、10/1~2/1がさらに好ましい。
上記製剤中のクロロゲン酸類又はその塩の含有量は、製剤の種類によっても異なるが、経口投与製剤の場合、クロロゲン酸換算で、製剤全質量の1~100質量%が好ましく、5~95質量%がより好ましく、10~70質量%がさらに好ましい。
本発明のエネルギー消費促進剤等を各種飲食品の有効成分として用いる場合、一般飲食品のほか、エネルギー消費促進、脂質燃焼促進、糖質燃焼促進、運動効果向上、ACC2抑制、PDK4抑制、肥満やメタボリックシンドロームの発症リスクの低下・予防・改善、運動機能向上をコンセプトとし、必要に応じてその旨表示した美容飲食品、病者用飲食品、栄養機能飲食品又は特定保健用飲食品等の機能性飲食品に応用できる。
飲食品の形態は、固形、半固形または液状であり得る。飲食品の例としては、パン類、麺類、クッキー等の菓子類、スナック類、ゼリー類、乳製品、冷凍食品、粉末コーヒー等のインスタント食品、でんぷん加工製品、加工肉製品、その他加工食品、コーヒー飲料等の飲料、缶コーヒー飲料や清涼飲料水、果汁飲料等の容器詰め飲料、スープ類、調味料、栄養補助食品等、及びそれらの原料が挙げられる。また、上記の経口投与製剤と同様、錠剤形態、丸剤形態、カプセル形態、液剤形態、シロップ形態、粉末形態、顆粒形態等であってもよい。
斯かる形態の飲食品は、クロロゲン酸類又はその塩の他、他の飲食品材料や、軟化剤、油、乳化剤、防腐剤、香科、安定剤、着色剤、酸化防止剤、増粘剤、固着剤、分散剤、湿潤剤等、更には前記と同様のカフェインを適宜組み合わせて配合し、調製することができる。
また、飲食品中におけるクロロゲン酸類又はその塩の含有量は、その使用形態により異なるが、クロロゲン酸類換算で、通常、飲料の形態では、0.01~5質量%であり、0.05~3質量%が好ましく、0.1~2質量%がより好ましく、0.1~1質量%がさらに好ましい。また、ゼリー類では、通常0.01~1質量%であり、0.02~0.5質量%が好ましく、0.05~0.3質量%がより好ましい。また、コーヒー飲料では、通常0.01~1質量%であり、0.05~1質量%が好ましく、0.1~1質量%がより好ましく、0.15~1質量%がさらに好ましい。
また、本発明のエネルギー消費促進剤等を飼料の有効成分として用いる場合には、当該飼料としては、例えば牛、豚、鶏、羊、馬等に用いる家畜用飼料、ウサギ、ラット、マウス等に用いる小動物用飼料、マグロ、ウナギ、タイ、ハマチ、エビ等に用いる魚介類用飼料、犬、猫、小鳥、リス等に用いるペットフード等が挙げられる。
尚、飼料を製造する場合には、エネルギー消費促進剤等を単独で、又はこの他に、牛、豚、羊等の肉類、蛋白質、穀物類、ぬか類、粕類、糖類、野菜、ビタミン類、ミネラル類等一般に用いられる飼料原料、更に一般的に飼料に使用されるゲル化剤、保型剤、pH調整剤、調味料、防腐剤、栄養補強剤等を必要に応じて配合し、常法により当該飼料を加工製造することができる。
尚、飼料を製造する場合には、エネルギー消費促進剤等を単独で、又はこの他に、牛、豚、羊等の肉類、蛋白質、穀物類、ぬか類、粕類、糖類、野菜、ビタミン類、ミネラル類等一般に用いられる飼料原料、更に一般的に飼料に使用されるゲル化剤、保型剤、pH調整剤、調味料、防腐剤、栄養補強剤等を必要に応じて配合し、常法により当該飼料を加工製造することができる。
本発明のエネルギー消費促進剤等を医薬品や機能性食品の有効成分として用いる場合の投与量・摂取量は、効果が得られる量であれば、特に限定されない。また、その投与量・摂取量は、対象者の状態、体重、性別、年齢又はその他の要因に従って変動し得るが、経口投与の場合の成人1人当たりの1日の投与量は、通常、クロロゲン酸類として100~3000mgが好ましく、300~2000mgがより好ましく、300~1000mgが特に好ましい。また、上記製剤は、任意の投与計画に従って投与され得るが、1日1回~数回に分けて投与することが好ましい。
本発明のエネルギー消費促進剤等の投与又は摂取対象者としては、それを必要としている者であれば特に限定されないが、肥満やメタボリックシンドローム者やその予備群が好ましい。肥満の基準としては、日本においてはBMI=25以上の者、欧米においてはBMI=30以上の者が主に該当する。メタボリックシンドロームの診断基準は、日本人の場合、男性であればウエストが85cm以上、女性であれば90cm以上の者であって、(1)血中トリグリセリドが150mg/dl以上又はHDLコレステロールが40mg/dl未満であること、(2)高血糖(空腹時血糖が110mg/dl以上)であること、(3)高血圧(130/85mHg以上)であることの3項目のうち1項目以上が当てはまる者が予備群に該当し、2項目以上が当てはまる者がメタボリックシンドロームに該当することから、これらの者が本発明の対象者として好ましい。米国の場合は、腹囲(男性で102cm以上、女性で88cm以上)、高中性脂肪、低HDL、高血圧、高空腹時血糖のうち、3つ以上を満たすものがメタボリックシンドロームに該当することから、2つ以上に該当する予備群のものを含め、これらの者が本発明の対象者として好ましい。
本発明のエネルギー消費促進剤等の投与又は摂取対象者としては、それを必要としている者であれば特に限定されないが、肥満やメタボリックシンドローム者やその予備群が好ましい。肥満の基準としては、日本においてはBMI=25以上の者、欧米においてはBMI=30以上の者が主に該当する。メタボリックシンドロームの診断基準は、日本人の場合、男性であればウエストが85cm以上、女性であれば90cm以上の者であって、(1)血中トリグリセリドが150mg/dl以上又はHDLコレステロールが40mg/dl未満であること、(2)高血糖(空腹時血糖が110mg/dl以上)であること、(3)高血圧(130/85mHg以上)であることの3項目のうち1項目以上が当てはまる者が予備群に該当し、2項目以上が当てはまる者がメタボリックシンドロームに該当することから、これらの者が本発明の対象者として好ましい。米国の場合は、腹囲(男性で102cm以上、女性で88cm以上)、高中性脂肪、低HDL、高血圧、高空腹時血糖のうち、3つ以上を満たすものがメタボリックシンドロームに該当することから、2つ以上に該当する予備群のものを含め、これらの者が本発明の対象者として好ましい。
製造例1
焙煎コーヒー粉砕豆(2kg)から熱水抽出により、固形物濃度約25%のコーヒー抽出物1.45kgを得た。得られたコーヒー抽出物を固形物濃度約35%まで減圧濃縮し、その後195℃で噴霧乾燥を行い、粉末コーヒー抽出物300gを得た。この粉末コーヒー抽出物270gを純水9Lに溶解した。得られたコーヒー溶解液9Lを、合成吸着剤(セパビーズSP70,三菱化学社製)を充填した3Lカラムに通液した。3Lの純水でカラムを通液洗浄後、0.1%水酸化ナトリウム水溶液33Lによりクロロゲン酸類を溶出させ、直ちにイオン交換樹脂(アンバーライト200CT:オルガノ社)を用いて弱酸性に中和・回収した。回収したクロロゲン酸類を含む調製液は減圧濃縮により固形分濃度が約10%になるまで濃縮し、その後噴霧乾燥を行い、高純度クロロゲン酸類製剤(40g)を得た。

製造したクロロゲン酸類製剤中のクロロゲン酸類の含有率(下記の各分子種含有量の総和)は77.1%で、組成は以下の通りであった。
3-カフェオイルキナ酸(7.7%)、4-カフェオイルキナ酸(15.5%)、5-カフェオイルキナ酸(31.9%)、3,4-ジカフェオイルキナ酸(9.0%)、3,5-ジカフェオイルキナ酸(8.9%)、4,5-ジカフェオイルキナ酸(5.3%)、3-フェルロイルキナ酸(7.6%)、4-フェルロイルキナ酸(5.6%)、及び5-フェルロイルキナ酸(8.5%)。なお、本クロロゲン酸類製剤にはカフェインは含まれていなかった。
3-カフェオイルキナ酸(7.7%)、4-カフェオイルキナ酸(15.5%)、5-カフェオイルキナ酸(31.9%)、3,4-ジカフェオイルキナ酸(9.0%)、3,5-ジカフェオイルキナ酸(8.9%)、4,5-ジカフェオイルキナ酸(5.3%)、3-フェルロイルキナ酸(7.6%)、4-フェルロイルキナ酸(5.6%)、及び5-フェルロイルキナ酸(8.5%)。なお、本クロロゲン酸類製剤にはカフェインは含まれていなかった。
製造例2
コーヒー生豆100gを1Lの98℃の熱水で4時間攪拌・抽出した。冷却後、固液分離を行い、抽出液を固形分濃度が20%(w/w)になるまで40℃にて減圧濃縮を行い、その後噴霧乾燥にてクロロゲン酸製剤を得た。得られたコーヒー豆抽出物のクロロゲン酸類量は、カフェオイルキナ酸(CQA)26.3質量%、フェルロイルキナ酸(FQA)5.1質量%、ジカフェオイルキナ酸(di-CQA)6.65質量%であった。

製造例3
上記クロロゲン酸製剤をクロロゲン酸類が1質量%になるようにイオン交換水で溶解させた後、溶解液165gに2N塩酸を添加しpHを1.3に調整した後、遠心分離により固液分離を行い、上層部分164gを得た。その後、採取した上層液54gを合成吸着剤(商品名セパビーズSP207:三菱化学社製)7.7mlの充填されたカラムに通液させた。その後、0.1%水酸化ナトリウム溶液116gをカラムに通液させ、イオン交換樹脂(アンバーライト200CT:オルガノ社)を用いて弱酸性に中和・回収した。回収したクロロゲン酸類を含む調製液は減圧濃縮を行い、高純度クロロゲン酸類製剤を得た。得られたクロロゲン酸類組成物は純度80%で、モノカフェオイルキナ酸類70%、フェルロイルキナ酸類14%、ジカフェオイルキナ酸類16%であった。

製造例4:9種クロロゲン酸類の調製
製造例1で得たクロロゲン酸類製剤から、中圧カラムクロマトグラフィーシステム(Yamazen:Ultra Pack ODS-A-40D column, UV detector PREP-UV-10V, fraction collector FR 50N, gradient mixer GR200, degassing unit, pump PUMP-600A)により、9種のクロロゲン酸類を調製した。クロロゲン酸類製剤(2g)を20mlのA液(酢酸-メタノール-水 =1:20:80)に溶解後、カラムにアプライし、10ml/minの流速で、0~100分までA液で、100~600分までB液(メタノール)でグラジエント(0→100%)をかけながら溶出させた。この操作により3-カフェオイルキナ酸、3、4-ジカフェオイルキナ酸、3,5-ジカフェオイルキナ酸、4,5-ジカフェオイルキナ酸を単一化合物として得た。4-カフェオイルキナ酸、5-カフェオイルキナ酸、3-フェルロイルキナ酸、4-フェルロイルキナ酸、5-フェルロイルキナ酸の混合物は更にInertsil ODS-3 column (GL Science Inc.)を用いるpreparative HPLC system (LC-908: Japan Analytical Industry Co., Ltd.)により精製した。溶出液はトリフルオロ酢酸-メタノール-水(1:300:700)を用い、流速は9ml/minとした。クロロゲン酸類の溶出は325nmにおける吸収を測定することによりモニターした。各化合物の構造は、1H-NMR(JEOL a500 NMR spectrometer:JEOL)にて確認した。

試験例1:継続摂取時のエネルギー消費促進作用、脂質燃焼促進作用及び糖質燃焼促進作用の評価
C57BL/6Jマウス(7週齢、雄:チャールズリバー)を1週間、標準固形飼料CE-2(オリエンタル酵母工業)を用いて飼育(室温:23±2℃、湿度:55±10%、明期を7時~19時、自由摂水)後、体重が等しくなるように2群に分けた。その後、コントロール食(25%コーン油、5%ラード、13%蔗糖、20%カゼイン、28.5%ポテトスターチ、4%セルロース、3.5%ビタミン(商品名:ビタミン混合AIN-76、オリエンタルバイオサービス)、1%ミネラル(商品名:ミネラル混合AIN-76、オリエンタルバイオサービス))またはクロロゲン酸類を含む試験食(25%植物油、5%ラード、13%蔗糖、20%カゼイン、27.5%ポテトスターチ、4%セルロース、3.5%ビタミン、1%ミネラル、1%クロロゲン酸類製剤(製造例1記載))を8週間与え、9週目に呼気分析に供した。
呼気分析用チャンバーにマウスを移し、Arco-2000system(アルコシステム)を用いて24時間に渡り呼気分析を行った。呼気分析では、チャンバー中の酸素濃度、二酸化炭素濃度を測定することにより、各マウスの酸素消費量(エネルギー消費量)を算出するとともに、Peronnetの式(Peronnet F, and Massicotte D (1991) Can J Sport Sci 16:23-29.)を用いて、脂質燃焼量並びに糖質燃焼量を算出した。図1に平均エネルギー消費量、平均脂質燃焼量及び平均糖質燃焼量の結果を示す。

クロロゲン酸類製剤を含む試験食摂取群では、コントロール群に比べ、エネルギー消費量、脂質燃焼量、糖質燃焼量が有意に増加した。従って、クロロゲン酸類は、エネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤として有効である。
試験例2:単回摂取時のエネルギー消費促進作用、脂質燃焼促進作用
C57BL/6Jマウス(7週齢、雄:チャールズリバー)を1週間、標準固形飼料CE-2(オリエンタル酵母工業)を用いて飼育(室温:23±2℃、湿度:55±10%、明期を7時~19時、自由摂水)後、体重が等しくなるように2群に分けた。8時間絶食の後、エーテル麻酔下、製造例1で製造したクロロゲン酸類製剤を含む試験水溶液(2% (w/v)総クロロゲン酸類)又はクロロゲン酸類を含まないコントロール水を、10ml/kg体重となるよう経口投与し、続けて下記の方法で調製した糖/脂質混合乳剤を20ml/kg体重となるよう経口投与した。マウスを直ちに呼気分析用チャンバーに移し、Arco-2000system(アルコシステム)を用いて1時間に渡り呼気分析を行った。呼気分析では、チャンバー中の酸素濃度、二酸化炭素濃度を測定することにより、各マウスの酸素消費量(エネルギー消費量)を算出するとともに、Peronnetの式(Peronnet F, and Massicotte D (1991) Can J Sport Sci 16:23-29)を用いて、脂質燃焼量を算出した。

<糖/脂質混合乳剤の調製>
予め水に溶解したスクロース溶液にレシチン(商品名:レシチン、卵製、和光純薬)とコーン油を以下の組成になるように加え、全量を10mlとし、超音波処理により乳化し、乳剤を得た。

クロロゲン酸類投与群においては、脂肪燃焼量が有意に多いことがわかる。従って、本発明のクロロゲン酸類は、脂質燃焼促進剤として有効である。また、本実験より、クロロゲン酸類は食餌性脂質の燃焼促進に有効であることがわかる。

試験例3:運動時のエネルギー消費促進作用、脂質燃焼促進作用、運動効果向上作用
C57BL/6Jマウス(6週齢、雄:チャールズリバー)を1週間、標準固形飼料CE-2(オリエンタル酵母工業)を用いて飼育(室温:23±2℃、湿度:55±10%、明期を7時~19時、自由摂水)し、環境に馴化させた。7週齢時に、トレッドミル走行運動に慣れさせるためのトレーニングを下記のとおりに5日間行った。
C57BL/6Jマウス(6週齢、雄:チャールズリバー)を1週間、標準固形飼料CE-2(オリエンタル酵母工業)を用いて飼育(室温:23±2℃、湿度:55±10%、明期を7時~19時、自由摂水)し、環境に馴化させた。7週齢時に、トレッドミル走行運動に慣れさせるためのトレーニングを下記のとおりに5日間行った。
1日目: 5m/min (10 min) → 10 m/min(10 min) → 15 m/min (10 min)
2日目:10m/min (10 min) → 15 m/min(10 min) → 20 m/min (10 min)

8週齢時に、体重が等しくなるよう、コントロール食 (Cont) 群及びクロロゲン酸群に群分けを行った。その後、コントロール食(5%コーン油、20%カゼイン、66.5%ポテトスターチ、4%セルロース、3.5%ビタミン(商品名:ビタミン混合AIN-76、オリエンタルバイオサービス)、1%ミネラル(商品名:ミネラル混合AIN-76、オリエンタルバイオサービス))またはクロロゲン酸類を含む試験食(5%コーン油、20%カゼイン、65.5%ポテトスターチ、4%セルロース、3.5%ビタミン、1%ミネラル、1%クロロゲン酸類製剤(製造例1記載))を8週間与えた。また、この飼育期間中には、18m/min (30min)の走行トレーニングを週3回行った。
試験食摂取開始8週目に、運動時呼気測定を実施した。運動時呼気測定試験は、Arco2000-system (アルコシステム社)を用いて行った。運動時呼気測定用のトレッドミル型チャンバーにマウスを1匹ずつ入れ、2時間安静状態で環境に馴化させた。その後、トレッドミルを始動し、30分間の運動時呼気測定を行った。トレッドミルのプログラムは、10m/min(5min)、その後15m/min(5min)、最後に18m/min(20min)とした。測定された酸素消費量と二酸化炭素排出量から呼吸交換比(RER)を求め、また、脂質燃焼量をPeronnetらの式により算出した(Peronnet F, and Massicotte D (1991) Can J Sport Sci 16:23-29.)。尚、測定中に走行を拒絶した個体及び走行不能となった個体に関しては、測定を中止して除外した。

運動時のエネルギー消費量(酸素消費量)、脂質燃焼量の結果を図3に示す。クロロゲン酸類を含む試験食群においては、エネルギー消費量、脂肪燃焼量が有意に多いことがわかる。従って、本発明のクロロゲン酸類は、エネルギー代謝促進剤、脂肪燃焼促進剤として有効である。また、本実験より、クロロゲン酸類は蓄積体脂肪の燃焼促進に有効であることがわかる。
更に、一般に運動はエネルギー消費を増加させるが、本結果からわかるように、クロロゲン酸類は運動と併用することにより、更に脂質燃焼量並びにエネルギー消費量を増加させることから、運動の効果をより高める作用があり、運動効果増強剤としても有効であると言える。このことは更に、肥満やメタボリックシンドロームの予防・改善のために運動を行う場合に、本発明のクロロゲン酸類を併用することで、より効果的に目的を達せられることを示している。
更に、一般に運動はエネルギー消費を増加させるが、本結果からわかるように、クロロゲン酸類は運動と併用することにより、更に脂質燃焼量並びにエネルギー消費量を増加させることから、運動の効果をより高める作用があり、運動効果増強剤としても有効であると言える。このことは更に、肥満やメタボリックシンドロームの予防・改善のために運動を行う場合に、本発明のクロロゲン酸類を併用することで、より効果的に目的を達せられることを示している。
試験例4:マウスにおけるクロロゲン酸類のエネルギー代謝関連分子発現調節作用

結果を図4に示す。クロロゲン酸類を摂取したマウスの肝臓においては、エネルギー代謝を負に制御するACC2 mRNAの発現が有意に低値を示した。ACC2の抑制はエネルギー消費、脂質燃焼、糖質燃焼の促進に繋がることから、クロロゲン酸類はエネルギー代謝促進剤として有効であると言える。また、クロロゲン酸類を摂取したマウスにおいては、糖質代謝を負に制御するPDK4 mRNAの発現が有意に低値を示したことから、クロロゲン酸類は糖質燃焼、エネルギー代謝を促進する作用を有することがわかる。
試験例5:肝細胞におけるクロロゲン酸類のエネルギー代謝関連分子発現調節作用
マウス培養肝細胞株(Hepa1-6細胞)は、10%ウシ血清を含むDulbeco’s Modified Eagle’s Medium(DMEM)で37℃、5% CO2 で培養を行った。細胞を6wellプレートに播き、サブコンフルエントに達した後、無血清培地(DMEM, -FBS)に交換し、更に12時間培養を行った。その後、各種サンプルを添加し(最終濃度:クロロゲン酸類(製造例1) 2.5 x 10-4%(w/v)、 各クロロゲン酸類(製造例4) 5 mM)、24時間培養後に定法に従い総RNAを調製した。逆転写反応を行いcDNAを調製後、Power SYBR Green Master Mix (Applied Biosystems)を用い、ABI PRISM 7000 Sequence Detection System (Applied Biosystems)により定量PCRを行った。また、各遺伝子の発現量は、内部標準として36B4の発現量で補正し、またコントロールの発現量を100とした相対発現量として表した。

結果を図5に示す。クロロゲン酸類はエネルギー代謝を負に制御するACC2mRNA、及び糖質代謝を負に制御するPDK4 mRNAの発現を有意に抑制した。また、クロロゲン酸類製剤中に含まれる9種のクロロゲン酸類(3-カフェオイルキナ酸(3-CQA)、4-カフェオイルキナ酸(4-CQA)、5-カフェオイルキナ酸(5-CQA)、3,4-ジカフェオイルキナ酸(3、4-diCQA)、3,5-ジカフェオイルキナ酸(3、5-diCQA)、4,5-ジカフェオイルキナ酸(4、5-diCQA)、3-フェルロイルキナ酸(3-FQA)、4-フェルロイルキナ酸(4-FQA)、及び5-フェルロイルキナ酸(5-FQA))について同様に評価した結果、3-カフェオイルキナ酸、4-カフェオイルキナ酸、5-カフェオイルキナ酸、3,4-ジカフェオイルキナ酸、3,5-ジカフェオイルキナ酸、4,5-ジカフェオイルキナ酸の6種のクロロゲン酸類が有意にACC2mRNAの発現を低下させた。ACC2の抑制はエネルギー消費、脂質燃焼、糖質燃焼の促進に繋がることから、クロロゲン酸類、特に上記6種化合物は、エネルギー代謝促進剤、脂質燃焼促進剤、糖質燃焼促進剤として有効であると言える。また、PDK4 mRNAの発現は、3-カフェオイルキナ酸、4-カフェオイルキナ酸、5-カフェオイルキナ酸、3,4-ジカフェオイルキナ酸により有意に抑制された。PDK4の抑制は糖質利用の促進に繋がることから、クロロゲン酸類、特に上記4種の化合物は、エネルギー代謝促進剤、糖質燃焼促進剤として有効であると言える。
試験例6:クロロゲン酸類を含有する容器詰め飲料のヒトにおけるエネルギー消費促進効果
試験飲料としてクロロゲン酸類を含む容器詰めコーヒー飲料を、対照飲料としてクロロゲン酸類を含まない容器詰めコーヒー様飲料を用い、クロロゲン酸類のヒトにおけるエネルギー代謝促進効果を検証した。試験は、ダブルブラインド、クロスオーバー試験として実施した。
・試験飲料(185ml):クロロゲン酸類(3-, 4-, 5-カフェオイルキナ酸 合計:306 mg(0.165質量%))、ヒドロキシヒドロキノン:0.11 mg、熱量:24 kcal
・対照飲料(185ml):クロロゲン酸類(0 mg)、ヒドロキシヒドロキノン:0.006 mg、熱量:24 kcal
上記飲料を7名の被験者を二群に分け、いずれかの飲料を一日一本、一週間飲用させた。一週間のウオッシュアウト期間の後、各被験者にもう一方の飲料を一日一本、一週間飲用させた。各飲料を一週間摂取した後、呼気分析装置(ARCO2000:アルコシステム)を用いて呼気分析(酸素消費量(VO2)並びに二酸化炭素排出量(VCO2)を測定)を行い、エネルギー消費量を評価した。
試験飲料としてクロロゲン酸類を含む容器詰めコーヒー飲料を、対照飲料としてクロロゲン酸類を含まない容器詰めコーヒー様飲料を用い、クロロゲン酸類のヒトにおけるエネルギー代謝促進効果を検証した。試験は、ダブルブラインド、クロスオーバー試験として実施した。
・試験飲料(185ml):クロロゲン酸類(3-, 4-, 5-カフェオイルキナ酸 合計:306 mg(0.165質量%))、ヒドロキシヒドロキノン:0.11 mg、熱量:24 kcal
・対照飲料(185ml):クロロゲン酸類(0 mg)、ヒドロキシヒドロキノン:0.006 mg、熱量:24 kcal
上記飲料を7名の被験者を二群に分け、いずれかの飲料を一日一本、一週間飲用させた。一週間のウオッシュアウト期間の後、各被験者にもう一方の飲料を一日一本、一週間飲用させた。各飲料を一週間摂取した後、呼気分析装置(ARCO2000:アルコシステム)を用いて呼気分析(酸素消費量(VO2)並びに二酸化炭素排出量(VCO2)を測定)を行い、エネルギー消費量を評価した。
呼気分析は、以下の通り行った:
15分間安静・環境順化した後、被検品(試験飲料または対照飲料)及びサンドイッチ(540 kcal)摂取し、30分後、安静状態で呼気分析(15分間)した。以降、30分毎に15分ずつ呼気分析を実施(2時間までの平均酸素消費量(エネルギー消費量)の測定結果を図6に示す)した。被検品摂取から4時間後、エルゴメーターにて3分間、運動のウオーミングアップ(20W、3分間)し、60%最大心拍数(最大心拍数=220-年齢(Achtew, Metabolism, 52:747-752, 2003))まで15W/分で運動負荷を漸増させながら運動中の呼気分析を実施した。呼気分析結果より算出した嫌気性代謝閾値(anaerobic threshold:AT)の結果を図7に示す。

図6に示すように、対照飲料群と比較し、試験飲料群の酸素消費(エネルギー消費)量は有意に多く、本発明のクロロゲン酸類を含有する容器詰め飲料のエネルギー消費促進効果が確認された。
嫌気性代謝閾値は運動強度を徐々に上げていくような運動をした場合に、筋肉への酸素供給が十分足りている状態から不足する状態に移行する変化点となる運動強度、すなわち、有酸素運動の上限の運動強度を指す。図7に示すように、嫌気性代謝閾値は試験飲料群で有意に高いことから、本発明のクロロゲン酸類を含有する容器詰め飲料を摂取することにより、運動時の有酸素代謝能が向上した、すなわち運動機能が向上したことがわかる。
嫌気性代謝閾値は運動強度を徐々に上げていくような運動をした場合に、筋肉への酸素供給が十分足りている状態から不足する状態に移行する変化点となる運動強度、すなわち、有酸素運動の上限の運動強度を指す。図7に示すように、嫌気性代謝閾値は試験飲料群で有意に高いことから、本発明のクロロゲン酸類を含有する容器詰め飲料を摂取することにより、運動時の有酸素代謝能が向上した、すなわち運動機能が向上したことがわかる。
このように、本発明のクロロゲン酸類は、エネルギー消費促進作用、脂質燃焼促進作用、糖質燃焼促進作用、運動効果向上作用、ACC2抑制作用、PDK4抑制作用、及び運動機能向上作用を有する。

実施例8:缶コーヒー飲料
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤及びPDK4抑制剤を配合したコーヒー組成物は次のように製造した。
カフェインレスインスタントコーヒー(ネスカフェ:ネスレ社製)5gを水に溶解し、ODS充填剤(オクタデシルシリル化シリカゲル)(YMC GEL ODS-A 細孔径6nm 粒子径150μm)1kgを充填したカラムに供し、0.5%酢酸水12Lで洗浄、その後メタノール12Lにてクロロゲン酸類を溶出した。そして、減圧濃縮することによりクロロゲン酸類製剤1.8gを得た。本製剤中のクロロゲン酸類含量は14.5%であった。また、酸化成分の一種、ヒドロキシヒドロキノンは含まれなかった。次いで、得られたクロロゲン酸類製剤を190mLの水に溶解し、缶に充填後121℃で10分間殺菌し缶コーヒー飲料を製造した。
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤及びPDK4抑制剤を配合したコーヒー組成物は次のように製造した。

実施例9:缶コーヒー飲料
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤及びPDK4抑制剤を配合した缶コーヒー飲料は次のように製造した。
製造例1に従い製造したクロロゲン酸類製剤10gを3Lの水に溶解した後、190mLのスチール缶に充填、レトルト殺菌機を用いて、121.5℃で10分間保持し、F値(殺菌指標)10となるような条件で熱処理を行い、缶コーヒー飲料を製造した。
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤及びPDK4抑制剤を配合した缶コーヒー飲料は次のように製造した。
製造例1に従い製造したクロロゲン酸類製剤10gを3Lの水に溶解した後、190mLのスチール缶に充填、レトルト殺菌機を用いて、121.5℃で10分間保持し、F値(殺菌指標)10となるような条件で熱処理を行い、缶コーヒー飲料を製造した。
実施例10:缶コーヒー飲料
焙煎コーヒー粉砕豆に8倍量の熱水を加え、コーヒー抽出液を得た。得られたコーヒー抽出液中のBrixに対して50質量%量の活性炭(白鷺:日本エンバイロケミカルズ)を充填したカラムに、25℃でコーヒー抽出液を通液し、活性炭処理して酸化成分(ヒドロキシヒドロキノン)を除去したコーヒー組成物を得た。得られたコーヒー組成物中のクロロゲン酸類含量を測定し、イオン交換水で希釈し、重曹にてpH調整を行った。得られたコーヒー組成物を190g缶に充填後、密封し、レトルト殺菌処理を施し、本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した缶コーヒー飲料を得た。クロロゲン酸類含量は0.176%であった。
焙煎コーヒー粉砕豆に8倍量の熱水を加え、コーヒー抽出液を得た。得られたコーヒー抽出液中のBrixに対して50質量%量の活性炭(白鷺:日本エンバイロケミカルズ)を充填したカラムに、25℃でコーヒー抽出液を通液し、活性炭処理して酸化成分(ヒドロキシヒドロキノン)を除去したコーヒー組成物を得た。得られたコーヒー組成物中のクロロゲン酸類含量を測定し、イオン交換水で希釈し、重曹にてpH調整を行った。得られたコーヒー組成物を190g缶に充填後、密封し、レトルト殺菌処理を施し、本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した缶コーヒー飲料を得た。クロロゲン酸類含量は0.176%であった。
実施例11:缶コーヒー飲料
焙煎コーヒー粉砕豆に8倍量の熱水を加え、コーヒー抽出液を得た。コーヒー抽出液(10L)にフレーバーホルダー(FH-1242、長谷川香料(株))(100mL)と水(80L)を加え、重曹にてpHを調整した。これを190mLのスチール缶に充填した。缶に充填してからレトルト殺菌(124℃、20分間)を行った。クロロゲン酸類含量は0.13%であった。
焙煎コーヒー粉砕豆に8倍量の熱水を加え、コーヒー抽出液を得た。コーヒー抽出液(10L)にフレーバーホルダー(FH-1242、長谷川香料(株))(100mL)と水(80L)を加え、重曹にてpHを調整した。これを190mLのスチール缶に充填した。缶に充填してからレトルト殺菌(124℃、20分間)を行った。クロロゲン酸類含量は0.13%であった。
実施例12:粉末コーヒー
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した粉末コーヒーは次のように製造した。焙煎コーヒー粉砕豆に8倍量の熱水を加え、コーヒー抽出液を得た。コーヒー抽出液1Lあたり製造例1のクロロゲン酸類製剤を1g加えた後凍結乾燥を行い、粉末コーヒーを得た。
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した粉末コーヒーは次のように製造した。焙煎コーヒー粉砕豆に8倍量の熱水を加え、コーヒー抽出液を得た。コーヒー抽出液1Lあたり製造例1のクロロゲン酸類製剤を1g加えた後凍結乾燥を行い、粉末コーヒーを得た。
実施例13
製造例1に従い製造したクロロゲン酸類製剤を水1L当り1g溶解し、フィルター滅菌した後500mLのペットボトルに充填し、エネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した清涼飲料水を製造した。無理なく連用できる味であった。
製造例1に従い製造したクロロゲン酸類製剤を水1L当り1g溶解し、フィルター滅菌した後500mLのペットボトルに充填し、エネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した清涼飲料水を製造した。無理なく連用できる味であった。
実施例14:缶コーヒー飲料
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合したコーヒー組成物は次のように製造した。
製造例1に従い製造したクロロゲン酸類製剤10gを3Lの焙煎コーヒー抽出液に溶解した後、190mLのスチール缶に充填、レトルト殺菌機を用いて、121.5℃で10分間熱処理を行い、缶コーヒー飲料を製造した。
本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合したコーヒー組成物は次のように製造した。
製造例1に従い製造したクロロゲン酸類製剤10gを3Lの焙煎コーヒー抽出液に溶解した後、190mLのスチール缶に充填、レトルト殺菌機を用いて、121.5℃で10分間熱処理を行い、缶コーヒー飲料を製造した。
実施例15
製造例1に従い製造したクロロゲン酸類製剤を以下の組成になるよう水に溶解し、フィルター滅菌した後100mLの褐色ボトルに充填し、本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した清涼飲料組成物を製造した。
製造例1に従い製造したクロロゲン酸類製剤を以下の組成になるよう水に溶解し、フィルター滅菌した後100mLの褐色ボトルに充填し、本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した清涼飲料組成物を製造した。

以下の各成分を水に溶解し、フィルター滅菌した後100mLの褐色ボトルに充填し、本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した清涼飲料組成物を製造した。
以下の各成分を水に溶解し、フィルター滅菌した後100mLの褐色ボトルに充填し、本発明のクロロゲン酸類を含有するエネルギー消費促進剤、脂質燃焼促進剤、糖質燃焼促進剤、運動効果向上剤、運動機能向上剤、ACC2抑制剤、及びPDK4抑制剤を配合した清涼飲料組成物を製造した。

- クロロゲン酸類又はその塩を有効成分とする脂質燃焼及び糖質燃焼促進剤。
- クロロゲン酸類又はその塩を有効成分とする運動効果向上剤。
- クロロゲン酸類又はその塩を有効成分とする運動機能向上剤。
- クロロゲン酸類又はその塩を有効成分とするアセチル-CoAカルボキシラーゼ2抑制剤。
- クロロゲン酸類又はその塩を有効成分とするピルビン酸デヒドロゲナーゼキナーゼ4抑制剤。
- クロロゲン酸類又はその塩を有効成分とするアセチル-CoAカルボキシラーゼ2及びピルビン酸デヒドロゲナーゼキナーゼ4抑制剤。
