〔7〕〔1〕~〔6〕のいずれか1項に記載の発現ベクターに外来遺伝子を組み込む工程、および該発現ベクターによりジヒドロ葉酸還元酵素遺伝子欠損宿主細胞を形質転換させる工程を含む、外来遺伝子由来タンパク質の高レベル生産能およびHT不含培地生育能を有する形質転換体の製造方法。
〔8〕以下(a)~(d)の工程を含む、外来遺伝子由来タンパク質の生産方法。

〔9〕〔8〕(c)の工程において、Chemically Defined medium(CD培地)あるいはCD培地に非動物性の添加物を加えた培地で培養することを特徴とする、〔8〕に記載の方法。
〔10〕以下(a)~(c)の工程を含む、外来遺伝子由来タンパク質の高レベル生産能を有する形質転換体のスクリーニング方法。

本発明の発現ベクターは、バックボーンベクター上に、下記(a)および(b)を含むことによって構築される。
(a)コドンを哺乳動物において使用頻度の最も低いものに改変し発現を微弱化させた翻訳障害性ジヒドロ葉酸還元酵素遺伝子カセット(翻訳障害性DHFR遺伝子カセット)
(b)高転写活性プロモーターと高安定性ポリアデニレーションシグナルの間に外来遺伝子組み込み用クローニングサイトを含む遺伝子カセット
該「翻訳障害性DHFR遺伝子カセット」において、コドンが改変される領域は特に制限されるものではないが、遺伝子カセットの全長の30%以上(例えば、40%以上、50%以上、60%以上、70%以上、80%以上、90%以上、95%以上、又は100%)の領域におけるコドンが改変されることが好ましい。コドンの改変領域の範囲は、ベクターの他の条件を考慮して任意に決定することができる。
また、本発明は、上記発現ベクターに外来遺伝子を組み込む工程、および該発現ベクターによりジヒドロ葉酸還元酵素遺伝子欠損宿主細胞を形質転換させる工程を含む、外来遺伝子由来タンパク質の高レベル生産能およびHTを含まない培地における生育能を有する形質転換体の製造方法を提供する。
具体的には、発現させたいタンパク質をコードする外来遺伝子を本発明の発現ベクターのマルチクローニングサイト(以下、MCSと記載)に組み込んだ後、トランスフェクション法(ここで言うトランスフェクション法とはリポフェクチン法、エレクトロポーレーション法、リン酸カルシウム法、マイクロインジェクション法など当業者が周知の方法を挙げることができる)を利用して該発現ベクターによりジヒドロ葉酸還元酵素遺伝子欠損宿主細胞を形質転換し、HTを含まない培地における耐性でセレクションを行い、タンパク質の高生産性形質転換体を得る方法が挙げられる。
HT不含培地におけるセレクションで生存した形質転換細胞の多くは既に相対的にタンパク質の高い発現レベルを達成しているが、その中からさらに高レベル生産能をもつ形質転換細胞を選択するために、タンパク質の発現レベルの測定をしてもよい。

本発明において、上記(c)の工程においては、HT不含培地で培養を行うことにより、高効率のタンパク質発現を示す形質転換体(コロニー)を選択することが出来る。選択された形質転換体は、引き続き同じ培地において培養を続けてもよいし、他の培地、例えば大量発現用の培地に移して培養を行ってもよい。
本発明において、形質転換体を培養又は馴化させる培地は特に制限されないが、好ましくは無血清培地、より好ましくはCD培地あるいはCD培地に非動物性の添加物を加えた培地を例示することができる。
本発明において培養された形質転換体から外来遺伝子由来タンパク質を回収する際には、当業者に公知の方法(フィルトレーション(濾過)、遠心分離、およびカラム精製等)でタンパク質を精製してもよい。外来遺伝子由来タンパク質は、精製を容易にするなどの目的で、他のタンパク質との融合タンパク質として発現させることも可能である。

なお本明細書において引用された全ての先行技術文献は、参照として本明細書に組み入れられる。
当業者に周知の方法を用いて本発明のベクターであるpDC1, pDC2, pDC5, pDC6, pDC7を構築した。バックボーンベクターpDC1の全塩基配列を配列番号:1に記載する。pDC1は塩基配列No1784-No2347の間に野生型のDHFRのcDNAを有する(図1)。pDC2は、pDC1の塩基配列No1784-No2347の配列を、配列番号:2に記載の配列に置換したものである。pDC2の置換領域にはDHFRの全塩基配列のコドンを哺乳動物において使用頻度の最も低いものに全塩基配列を改変した翻訳障害性DHFR遺伝子が導入されている(図2)。
pDC5は、pDC1の塩基配列No1784-No2347の配列を、配列番号:3に記載の配列に置換したものである。pDC5の置換領域にはDHFRの塩基配列を5‘末端から90塩基の範囲で、コドンを哺乳動物において使用頻度の最も低いものに改変した翻訳障害性DHFR遺伝子が導入されている(図3)。
pDC6は、pDC1の塩基配列No1784-No2347の配列を、配列番号:4に記載の配列に置換したものである。pDC6の置換領域にはDHFRの塩基配列を5‘末端から180塩基の範囲で、コドンを哺乳動物において使用頻度の最も低いものに改変した翻訳障害性DHFR遺伝子が導入されている(図4)。
pDC7は、pDC1の塩基配列No1784-No2347の配列を、配列番号:5に記載の配列に置換したものである。pDC7の置換領域にはDHFRの塩基配列を5‘末端から270塩基の範囲で、コドンを哺乳動物において使用頻度の最も低いものに改変した翻訳障害性DHFR遺伝子が導入されている(図5)。
当業者に周知の方法を用いて本発明のベクターであるpDC1, pDC2, pDC5, pDC6, pDC7の塩基配列No1267-No1275を配列番号:6に記載のヒトマンナン結合レクチン(MBL)をコードするcDNA(以下hMBLと記載する)に置換し、pDC1/hMBL(図6)、pDC2/hMBL(図7)、pDC5/hMBL(図8)、pDC6/hMBL(図9)、pDC7/hMBL(図10)を構築した。
10μgのpDC1/hMBL, pDC2/hMBL, pDC5/hMBL, pDC6/hMBL, pDC7/hMBLをリポフェクチン法(Lipofectamine(登録商標) LTX, Invitrogenを使用)を用いて25cm2のカルチャーフラスコ中の500000個のCHO細胞(CHO DG44 cell)に遺伝子導入した。導入方法は製造業者の使用説明書に従った。遺伝子導入48時間後、細胞数を計測した後、細胞を4mM Gluta MAX(登録商標)-I (Invitrogen)を含むIS CHO-CD w/ Hydrolysate培地(IS Japan)にて希釈した。96ウェルマイクロタイタープレート中に1000 cells/well, 100 cells/wellの濃度で5枚ずつ計10枚(960ウェル)播き、5%炭酸ガス存在下で37℃、約3週間培養したところ、生存した細胞が見られた(HT不含培地生育セルライン)。生存細胞から任意に50株のHT不含培地生育セルラインを選択し、4mM Gluta MAX(登録商標)-I (Invitrogen)を含むIS CHO-CD w/ Hydrolysate培地(IS Japan)ともに24ウェルプレートに移し、細胞が各ウェルの1/3以上を占めるまで培養した。各株に0.4mLを滅菌チューブに取り、200g、2分間遠心した。上清を捨て、細胞を0.1mLの新しい培地(4 mM Gluta MAX(登録商標)-I ((Invitrogen)を含むIS CHO-CD w/ Hydrolysate培地(IS Japan))に懸濁し、細胞数を計測した後、細胞数を5 x 105 cells/mLになるように培地で希釈後、0.2mLを新しい24ウェルプレートに移し、5%炭酸ガス存在下で37℃、72時間培養し、9300g、2分間の遠心後に上清を回収した。続いて培養上清中のMBLの生産量を測定した。
生産量の検定はELISAにて実施した。コーティングバッフアー(15 mM, Na2CO3, 35 mM NaHCO3, 0.05 % NaN3, pH 9.6)で希釈した1μg/mL抗ヒトMBL抗体(日本・旭川医大・大谷博士より譲渡)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate,Cat no. 442404, Nunc)に4℃、16時間でコートした。4% Block Ace(大日本住友製薬株式会社)でブロッキングした後、72時間培養上清(1/1000-1/100000希釈)、精製したヒトMBL(旭川医大・大谷博士より譲渡)のCHO細胞用無血清培地IS CHO-CD w/ Hydrolysate培地(IS Japan)による2倍希釈系列(20~0.3125 ng/mL)およびIS CHO with Hydrolysate培地(IS Japan)をそれぞれ100μLずつアプライし、37℃で1時間インキュベートした。さらに0.1μg/mLのビオチン化ヒトMBLモノクローナル抗体(旭川医大・大谷博士より譲渡)と37℃、1時間インキュベートした。37℃で30分間インキュベートしたVECTASTAION Elite ABC kit STANDARD(Reagent A 2 drops,Regent B 2 drops / 5 mL, Vector)を100μL/wellずつアプライし、37℃で45分間反応させた。さらに室温で30分間インキュベートしたPEROXIDASE SUBSTRATE KIT TMB(2 drops of Buffer, 3 drops of TMB, 2 drops of HYDROGEN PEROXIDE / 5 mL, Vector)を100μL/wellずつアプライし、室温で15分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて行なった。表1にELISA法にて得られた結果、ヒトMBL生産量の高い上位3サンプルを示す。最も生産レベルの高かったセルラインは、コドン未改変のベクターと比較して優位に高い生産性が得られた。
本発明の発現ベクターpDC1, pDC5, pDC6, pDC7により発現されたhMBLの各セルラインの発現量の分布を表2に示す。
pDC1では50株のHT不含培地生育株のうち、28.0%はhMBLを0μg/mL以上5μg/mL未満で生産していた。また50株のうち36株(72.0%)は5μg/mL以上であった。また50株のうち19株(38.0%)は10μg/mL以上であった。また50株のうち12株(24.0%)は15μg/mL以上であった。また50株のうち2株(4.0%)は20μg/mL以上であった。最も高い生産レベルを示したものは20.5μg/mL/3dayであった。
pDC5では50株のHT不含培地生育株のうち70%はhMBLを0μg/mL以上5μg/mL未満で生産していた。また50株のうち15株(30.0%)は5μg/mL以上であった。また50株のうち3株(6.0%)は10μg/mL以上であった。最も高い生産レベルを示したものは11.8μg/mL/3dayであった。
pDC6では50株のHT不含培地生育株のうち、34.0%はhMBLを0μg/mL以上5μg/mL未満で生産していた。また50株のうち33株(66.0%)は5μg/mL以上であった。また50株のうち22株(44.0%)は10μg/mL以上であった。また50株のうち15株(30.0%)は15μg/mL以上であった。また50株のうち7株(14.0%)は20μg/mL以上であった。また50株のうち5株(10.0%)は25μg/mL以上であった。驚くべきことに50株のうち1株(2.0%)は50μg/mL以上であった。最も高い生産レベルを示したものは51.6μg/mL/3dayであった。
pDC7では50株のHT不含培地生育株のうち、56.0%はhMBLを0μg/mL以上5μg/mL未満で生産していた。また50株のうち22株(44.0%)は5μg/mL以上であった。また50株のうち16株(32.0%)は10μg/mL以上であった。また50株のうち13株(26.0%)は15μg/mL以上であった。また50株のうち8株(16.0%)は20μg/mL以上であった。また50株のうち6株(12.0%)は25μg/mL以上であった。また50株のうち3株(6.0%)は30μg/mL以上であった。また50株のうち2株(4.0%)は35μg/mL以上であった。また50株のうち1株(2.0%)は45μg/mL以上であった。最も高い生産レベルを示したものは33.2μg/mL/3dayであった。
これは文献等で報告されている代表的な発現ベクターによる遺伝子増幅前の初期クローンのデータと比較して最も高い水準であった(DNA,7,651頁,1988年;Biotechnology,10.1455頁,1992年;Biotechnology,8,662頁,1990年;Gene,76,19頁,1989年;Biotechnology,9,64頁,1991年)。
遺伝子増幅による組換細胞のスクリーニングには通常6ヶ月から1年を要し、かつ培養条件や増幅刺激剤濃度による差異が大きいため、発現ベクターのプライマリーな性能比較は増幅前の初期クローンの発現レベルで行うのが適当と考えられる。これにより本発明の発現ベクターの性能は極めて高いことが判明した。この結果、本発明のベクターは、得られるHT不含培地生育株の数が極めて低い一方、非常に高い効率で目的物質タンパク質の高生産性細胞株の確立を可能にすることが確かめられた。これにより、本発明の発現ベクターは非常に高いタンパク質発現レベルを可能にすることが証明された。
当業者に周知の方法を用いて本発明のベクターであるpDC6の塩基配列No1267-No1275を配列番号:7に記載のヒトエリスロポエチン(EPO)をコードするcDNA(以下hEPOと記載する)に置換し、pDC6/hEPO(図11)を構築した。
2.5μgのpDC6/hEPOをリポフェクチン法(Lipofectamine(登録商標) LTX, Invitrogenを使用)を用いて25cm2のカルチャーフラスコ中の4,000,000個のCHO細胞(CHO DG44 cell)に遺伝子導入した。導入方法は製造業者の使用説明書に従った。遺伝子導入48時間後、細胞数を計測した後、細胞を4mM Gluta MAX(登録商標)-I (Invitrogen)を含むIS CHO-CD w/ Hydrolysate培地(IS Japan)にて希釈した。96ウェルマイクロタイタープレート中に4000 cells/wellの濃度で5枚(480ウェル)播き、5%炭酸ガス存在下で37℃、約3週間培養したところ、生存した細胞が見られた(HT不含培地生育セルライン)。
産生量の検定はELISAにて実施した。固相抗体溶液(D-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ))で希釈した1μg/mL抗ヒトEPO抗体(rhEPO MAb R6K, 扶桑薬品工業社製)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate,Cat no. 442404, Nunc)に4℃、16時間でコートした。ブロッキング液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:3の比率で混合)でブロッキングした後、72時間培養上清(1/1000-1/100000希釈)、精製したヒトEPO(扶桑薬品工業社製)の抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)、D-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)による2倍希釈系列(500~15.6 mIU/mL)および抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)をそれぞれ100μLずつアプライし、25℃で2時間インキュベートした。さらに0.1μg/mLのペルオキシダーゼ標識ヒトEPOモノクローナル抗体(POD-rhEPO MAb R2C, 扶桑薬品工業社製)と25℃、2時間インキュベートした。Sure Blue TMB Microwell Peroxidase Substrate(KPL)を100μL/wellずつアプライし、25℃で30分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450nmの吸光度を測定した。表3にELISAにて得られた結果、ヒトEPO産生量の高い上位10サンプルを示す。最も産生量が高いセルラインでは3,727±109 IU/mL/3daysを示した。この値は、クローン化していない初期のセルラインであってしかも遺伝子増幅していない状態において、文献等で報告されている代表的なエリスロポエチンの産生量と比較して非常に高い水準を示していた(特開2002-45191、J Microbiol Biotechnol. 2008 Jul;18(7):1342-1351、 Biotechnol Appl Biochem. 2000 Dec;32 ( Pt 3):167-172、Proc Natl Acad Sci U S A. 1986 Sep;83(17):6465-6469)。これにより、本発明の発現ベクターは非常に高いタンパク質発現レベルを可能にすることが証明された。続いて培養上清中のhEPOのWesternblottingを行い、タンパク質の発現を確認した。
上記実施例8において採取したヒトEPO産生量上位10サンプルの3日間培養上清をウエスタンブロッティングによって解析した。培養上清10μLそれぞれに、5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。さらに標準品rhEPO(Recombinant human EPO, Cat 287-TC, R&D systems)100ng/10μLに5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。泳動層(DPE-1020、DAIICHI PURE CHEMICALS CO., LTD)に泳動緩衝液(Tris/Glycine/SDS、BIO-RAD)とSuper Sep(登録商標) 10~20% 17well(Wako)をセットし、加熱処理したサンプル溶液と標準品をSuper(登録商標)10~20% 17well(Wako)に20μL添加し、40mAで55分間電気泳動を行った(電源装置:MyRun、COSMO BIO CO., LTDを使用)。その後、ゲルをガラス板から外して転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に5分間振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸した。Immobilon-P Transfer Membrane(MILLIPORE)をそれぞれ8mLのメタノール(Wako)で15秒、8mLのMilliQ水(MILLIPORE)で2分間、8mLの転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で5分間の順に振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸して活性化させた。転写装置(TRANS-BLO、 SD SEMI-DRY TRANSFER CELL、BIO-RAD)に転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)、活性化させたImmobilon-P Transfer Membrane(MILLIPORE)、転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に浸した泳動後のゲル及び転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)を陰極側から順に敷き、カバーを被せて80mA(PowerPac HC(登録商標)、BIO-RAD)で1時間半電気泳動を行い、分離したタンパク質をImmobilon-P Transfer Membrane(MILLIPORE)上に転写させた。転写後、Immobilon-P Transfer Membrane(MILLIPORE)を8mLのImmunoBlock(登録商標、大日本住友製薬株式会社 ラボラトリープロダクツ部)に浸して4℃で18時間ブロッキングを行い、0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で1000倍希釈した10mLのEpo (H-162) rabbit polyclonal IgG (Santa Cruz Biotechnology)と膜上のタンパク質を室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で5000倍希釈したPeroxidase Conjugated Affinity Purified Anti-Rabbit IgG F(c)〔Goat〕(Rock Land)を10mL添加して室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に2mLのImmobilon(登録商標) Western Chemiluminescent HRP Substrate(MILLIPORE)を添加して化学発光させ、Light-Capture ATTO Cooled CCD Camera System(ATTO)のノーマル設定で30秒間写真を撮影した。図13にウエスタンブロットで得られた像を示す。標準品と同様のバンドが検出された。
高いEPO産生量を示しかつ増殖の速いセルライン(No 27, 36, 50, 78, 82)を選択し、さらに7日間培養と14日間培養を行い、培養上清中のhEPO濃度を測定した。培養方法は、まず細胞数を計測した後細胞数を0.5 x 105 cells/mLに合わせて培地で希釈後、7.5 mLを新しいT75フラスコに移し、5%炭酸ガス存在下で37℃、7日間および14日間培養し、9300g、2分間の遠心後に上清を回収した。産生量の検定はELISAにて実施した。固相抗体溶液(D-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ))で希釈した1μg/mL抗ヒトEPO抗体(rhEPO MAb R6K, 扶桑薬品工業社製)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate,Cat no. 442404, Nunc)に4℃、16時間でコートした。ブロッキング液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:3の比率で混合)でブロッキングした後、7日間および14日間培養上清(1/1,000-1/100,000希釈)、精製したヒトEPO(扶桑薬品工業社製)の抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)による2倍希釈系列(500~15.6 mIU/mL)および抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)をそれぞれ100μLずつアプライし、25℃で2時間インキュベートした。さらに0.1μg/mLのペルオキシダーゼ標識ヒトEPOモノクローナル抗体(POD-rhEPO MAb R2C, 扶桑薬品工業社製)と25℃、2時間インキュベートした。Sure Blue TMB Microwell Peroxidase Substrate(KPL)を100μL/wellずつアプライし、25℃で30分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450nmの吸光度を測定した。表4にELISAにて得られた結果、7日間および14日間培養の産生量を表に示す。最も産生量が高いセルラインでは31,590±444 IU/mL/14daysを示した。この値は、クローン化していない初期のセルラインであってしかも遺伝子増幅していない状態において、文献等で報告されている代表的なエリスロポエチンの産生量と比較して非常に高い水準を示していた(特開2002-45191、J Microbiol Biotechnol. 2008 Jul;18(7):1342-1351、Biotechnol Appl Biochem. 2000 Dec;32 ( Pt 3):167-172、Proc Natl Acad Sci U S A. 1986 Sep;83(17):6465-6469)。通常、クローン化後にMTXによる遺伝子増幅を行って産生量の増大を図るが、遺伝子増幅したクローンは産生量が安定しないこともある。また遺伝子増幅による組換細胞のスクリーニングには6ヶ月から1年を要する。その点、当該ベクターをトランスフェクトすることで得られた細胞は、遺伝子増幅の必要がない程高い産生量を示し、短期間で容易に高産生細胞を得ることに成功した。これにより本発明の発現ベクターの性能は極めて高いことが判明した。
さらに高産生能を有する細胞を作出するために、スクリーニングする細胞の数を増加させることで、hEPO高産生セルラインの作出を試みた。
2.5μgのpDC6/hEPOをリポフェクチン法(Lipofectamine(登録商標) LTX, Invitrogenを使用)を用いて25cm2のカルチャーフラスコ中の16,000,000個のCHO細胞(CHO DG44 cell)に遺伝子導入した。導入方法は製造業者の使用説明書に従った。遺伝子導入48時間後、細胞数を計測した後、細胞を4mM Gluta MAX(登録商標)-I (Invitrogen)を含むCD Opti CHO AGT培地(Invitrogen)にて希釈した。96ウェルマイクロタイタープレート中に16000 cells/wellの濃度で45枚(4320ウェル)播き、5%炭酸ガス存在下で37℃、約3週間培養したところ、生存した細胞が見られた(HT不含培地生育セルライン)。
産生量の検定はELISAにて実施した。固相抗体溶液(D-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ))で希釈した1μg/mL抗ヒトEPO抗体(rhEPO MAb R6K, 扶桑薬品工業社製)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate,Cat no. 442404, Nunc)に4℃、16時間でコートした。ブロッキング液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:3の比率で混合)でブロッキングした後、72時間培養上清(1/40000-1/160000希釈)、精製したヒトEPO(扶桑薬品工業社製)の抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)、D-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)による2倍希釈系列(500~15.6 mIU/mL)および抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)をそれぞれ100μLずつアプライし、25℃で2時間インキュベートした。さらに0.1μg/mLのペルオキシダーゼ標識ヒトEPOモノクローナル抗体(POD-rhEPO MAb R2C, 扶桑薬品工業社製)と25℃、2時間インキュベートした。Sure Blue TMB Microwell Peroxidase Substrate(KPL)を100 μL/wellずつアプライし、25℃で30分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450nmの吸光度を測定した。表5にELISAにて得られた結果、ヒトEPO産生量の高い上位40サンプルを示す。
当業者に周知の方法を用いて本発明のベクターであるpDC6の塩基配列No1267-No1275を配列番号:8に記載のダルベポエチンアルファ(D-EPO)をコードするcDNA(以下D-EPOと記載する)に置換し、pDC6/D-EPO(図14)を構築した。
2.5μgのpDC6/D-EPOをリポフェクチン法(Lipofectamine(登録商標) LTX, Invitrogenを使用)を用いて25cm2のカルチャーフラスコ中の4,000,000個のCHO細胞(CHO DG44 cell)に遺伝子導入した。導入方法は製造業者の使用説明書に従った。遺伝子導入48時間後、細胞数を計測した後、細胞を4mM Gluta MAX(登録商標)-I (Invitrogen)を含むIS CHO-CD w/ Hydrolysate培地(IS Japan)にて希釈した。96ウェルマイクロタイタープレート中に4000 cells/wellの濃度で5枚(480ウェル)播き、5%炭酸ガス存在下で37℃、約3週間培養したところ、生存した細胞が見られた(HT不含培地生育セルライン)。
生存細胞から任意に84株のHT不含培地生育セルラインを選択し、Dotblotにより発現の確認を行った。メンブレン(Nytran N, ITEM NO.10416196, SCHLEICHER & SCHUELL )に組換えヒトEPO標準品(Recombinat Human EPO, Cat. 287-TC, R & D systems)の2倍希釈系列(10~0.16 ng/mL)および任意に選択した84株の培養上清をそれぞれ1μLずつアプライし、室温で30分インキュベートした後、4% Block Ace(大日本住友製薬株式会社)で、室温で30分ブロッキングした。さらにPBST (D-PBS, 0.05% Tween20)で希釈した0.2μg/mL抗EPOウサギポリクローナル抗体(EPO(H-162): rabbit polyclonal IgG, Cat.sc-7956, Santa Cruz Biotechnology)を加えて室温で30分間震盪した。PBST (D-PBS, 0.05% Tween20)で希釈した0.2μg/mLペルオキシダーゼ標識抗ウサギIgG抗体(Peroxidase Conjugated Affinity Purified Anti-RABBIT IgG F8c, Cat. 611-1303, Rock Land)を加えて30分間震盪した。室温で5分間反応させたImmobilon(登録商標) Western Chemiluminescent HRP Substrate (2 mL Luminol Reagent, 2 mL Peroxide Solution, MILLIPORE, Cat.WBKLS0050, MILLIPORE)を加えて室温で5分インキュベートし、化学発光を検出器(ATTO Light-Capture, AE-6981FC, ATTO)によって撮影した。図15にDotblotで得られた像を示す。
産生量の検定はELISAにて実施した。固相抗体溶液(D-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ))で希釈した1μg/mL抗ヒトEPO抗体(rEPO MAb R6K, 扶桑薬品工業社製)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate, Cat no. 442404, Nunc)に4℃、16時間でコートした。ブロッキング液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:3の比率で混合)でブロッキングした後、3、7、14日間培養上清(1/1000-1/100000希釈)、ダルベポエチンアルファ(ネスプ静注用120μg/0.6mLプラシリンジ, 共和発光キリン)の抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)による2倍希釈系列(10~0.156 ng/mL)および抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)をそれぞれ100μLずつアプライし、25℃で2時間インキュベートした。さらに0.1μg/mLのペルオキシダーゼ標識ヒトEPOモノクローナル抗体(POD-rEPO MAb R2C, 扶桑薬品工業社製)と25℃、2時間インキュベートした。Sure Blue TMB Microwell Peroxidase Substrate(KPL)を100 μL/wellずつアプライし、25℃で30分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450nmの吸光度を測定した。表6にELISAにて得られた結果、D-EPO産生量の高い上位5サンプルを示す。もっとも産生量が高いラインでは22±0.3μg/mL/3days, 135±3.3μg/mL/7days, 170±3.7μg/mL/14daysを示した。続いて培養上清中のD-EPOのWesternblottingを行い、タンパク質の発現を確認した。
上記実施例15において採取したD-EPO産生量上位5サンプルの3、7、14日間培養上清をウエスタンブロッティングにて解析した。培養上清10μLそれぞれに、5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。さらに標準品ダルベポエチンアルファ(ネスプ静注用120μg/0.6mLプラシリンジ, 協和発酵キリン)100ng/10μLに5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。泳動層(DPE-1020、DAIICHI PURE CHEMICALS CO., LTD)に泳動緩衝液(Tris/Glycine/SDS、BIO-RAD)とSuper Sep(登録商標) 10~20% 17well(Wako)をセットし、加熱処理したサンプル溶液と標準品をSuper(登録商標) 10~20% 17well(Wako)に20μL添加し、40mAで55分間電気泳動を行った(電源装置:MyRun、COSMO BIO CO., LTDを使用)。その後、ゲルをガラス板から外して転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に5分間振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸した。

産生量の検定はELISAにて実施した。コーティングバッフアー(15 mM, Na2CO3, 35 mM NaHCO3, 0.05 % NaN3, pH 9.6)で希釈した0.5μg/mL抗ヒトG-CSF抗体(Anti-human G-CSF monoclonal Antibody, Cat No. MAB214, R&D system)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate,Cat no. 442404, Nunc)に4℃、16時間でコートした。ブロッキング液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:3の比率で混合)でブロッキングした後、3、7、14日間培養上清(1/10,000-1/200,000希釈)、組換えヒトG-CSF標準品(Recombinant human G-CSF, Cat 1001C, APOLLO)の抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)による2倍希釈系列(5~0.078125 ng/mL)および抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)をそれぞれ100μLずつアプライし、25℃で1時間インキュベートした。さらに0.25μg/mLのビオチン化ヒトG-CSF抗体(Biotinylated Anti-human G-CSF Antibody, Cat No BAF214, R&D system)と25℃、1時間インキュベートした。25℃で30分間インキュベートしたStandard Ultra-Sensitive ABC Staining kit(Reagent A 2 drops,Regent B 2 drops / 10 mL, Pro#32050, PIERCE)を100μL/wellずつアプライし、25℃で30分間反応させた。Sure Blue TMB Microwell Peroxidase Substrate(KPL)を100μL/wellずつアプライし、25℃で30分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450nmの吸光度を測定した。表7にELISAにて得られた結果、ヒトG-CSF産生量の高い上位10サンプルを示す。最も産生量が高いラインでは34.7±1.1 μg/mL/3days, 193.6±0.6 μg/mL/7days, 235.5±14.8 μg/mL/14daysを示した。この値は、クローン化していない初期のセルラインであってしかも遺伝子増幅していない状態において、文献等で報告されている代表的なG-CSFの産生量と比較して非常に高い水準を示していた(J Biosci Bioeng. 2000;89(6):534-538、Gene. 1996 Nov 21;180(1-2):145-150、Mol Biotechnol. 1997 Jun;7(3):231-240、特表平01-500483)。
これにより、本発明の発現ベクターは非常に高いタンパク質発現レベルを可能にすることが証明された。続いて培養上清中のhG-CSFのWesternblottingを行い、タンパク質の発現を確認した。
上記実施例19において採取したヒトG-CSF産生量上位5サンプルの3、7、14日間培養上清をウエスタンブロッティングにて解析した。培養上清10μLそれぞれに、5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98度で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。さらに組換えヒトG-CSF標準品(Recombinant human G-CSF, Cat 1001C, APOLLO)10 μg/10 μLに5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98度で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。泳動層(DPE-1020、DAIICHI PURE CHEMICALS CO., LTD)に泳動緩衝液(Tris/Glycine/SDS、BIO-RAD)とSuper Sep(登録商標) 10~20% 17well(Wako)をセットし、加熱処理したサンプル溶液と標準品をSuper(登録商標)10~20% 17well(Wako)に20μL添加し、40mAで55分間電気泳動を行った(電源装置:MyRun、COSMO BIO CO., LTDを使用)。その後、ゲルをガラス板から外して転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に5分間振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸した。Immobilon-P Transfer Membrane(MILLIPORE)をそれぞれ8mLのメタノール(Wako)で15秒、8mLのMilliQ水(MILLIPORE)で2分間、8mLの転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で5分間の順に振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸して活性化させた。転写装置(TRANS-BLO、 SD SEMI-DRY TRANSFER CELL、BIO-RAD)に転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)、活性化させたImmobilon-P Transfer Membrane(MILLIPORE)、転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に浸した泳動後のゲル及び転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)をマイナス側から順に敷き、カバーを被せて80mA(PowerPac HC(登録商標)、BIO-RAD)で1時間半電気泳動を行い、分離したタンパク質をImmobilon-P Transfer Membrane(MILLIPORE)上に転写させた。転写後、Immobilon-P Transfer Membrane(MILLIPORE)を8mLのImmunoBlock(登録商標、大日本住友製薬株式会社 ラボラトリープロダクツ部)に浸して4℃で18時間ブロッキングを行い、0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で2,000倍希釈した10mLの抗ヒトG-CSFマウスモノクローナル抗体 (Monoclonal Anti-human G-CSF Antibody, Cat MAB214, R & D)と膜上のタンパク質を室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で5,000倍希釈したペルオキシダーゼ標識抗マウスIgG抗体(Goat anti-Mouse IgG (H+L), Cat 115-036-062, Jackson)を10mL添加して室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に2mLのImmobilon(登録商標) Western Chemiluminescent HRP Substrate(MILLIPORE)を添加して化学発光させ、Light-Capture ATTO Cooled CCD Camera System(ATTO)のノーマル設定で30秒間写真を撮影した。図19にウエスタンブロットで得られた像を示す。標準品と同様のバンドが検出された。

これにより、本発明の発現ベクターは非常に高いタンパク質発現レベルを可能にすることが証明された。続いて培養上清中のhGM-CSFのWesternblottingを行い、タンパク質の発現を確認した。
上記実施例23において採取したヒトGM-CSF産生量上位6サンプルの3、7、14日間培養上清をウエスタンブロッティングにて解析した。培養上清10μLそれぞれに、5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。さらに標準品ヒトGM-CSF(Recombinant Human GM-CSF Expressed in Human Cell, Cat No. HZ-1001, HumanZyme Inc.)10 μg/10 μLに5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。泳動層(DPE-1020、DAIICHI PURE CHEMICALS CO., LTD)に泳動緩衝液(Tris/Glycine/SDS、BIO-RAD)とSuper Sep(登録商標) 10~20% 17 well(Wako)をセットし、加熱処理したサンプル溶液と標準品をSuper(登録商標)10~20% 17 well(Wako)に20μL添加し、40 mAで55分間電気泳動を行った(電源装置:MyRun、COSMO BIO CO., LTDを使用)。その後、ゲルをガラス板から外して転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に5分間振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸した。Immobilon-P Transfer Membrane(MILLIPORE)をそれぞれ8 mLのメタノール(Wako)で15秒、8 mLのMilliQ水(MILLIPORE)で2分間、8 mLの転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で5分間の順に振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸して活性化させた。転写装置(TRANS-BLO、 SD SEMI-DRY TRANSFER CELL、BIO-RAD)に転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)、活性化させたImmobilon-P Transfer Membrane(MILLIPORE)、転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に浸した泳動後のゲル及び転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)をマイナス側から順に敷き、カバーを被せて80mA(PowerPac HC(登録商標)、BIO-RAD)で1時間半電気泳動を行い、分離したタンパク質をImmobilon-P Transfer Membrane(MILLIPORE)上に転写させた。転写後、Immobilon-P Transfer Membrane(MILLIPORE)を8 mLのImmunoBlock(登録商標、大日本住友製薬株式会社 ラボラトリープロダクツ部)に浸して4℃で18時間ブロッキングを行い、0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で2,000倍希釈した10mLの抗ヒトGM-CSF抗体(Anti-human GM-CSF Neutralizing Antibody, Cat No.AB-215-NA, R & D systems)と膜上のタンパク質を室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で5,000倍希釈したペルオキシダーゼ標識済ウサギ抗ヤギIgG抗体(Peroxidase Conjugated Affinity Purified Anti-Goat IgG, Cat No.605-4302, ,Rock Land)を10mL添加して室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に2 mLのImmobilon(登録商標) Western Chemiluminescent HRP Substrate(MILLIPORE)を添加して化学発光させ、Light-Capture ATTO Cooled CCD Camera System(ATTO)のノーマル設定で5秒間写真を撮影した。図22にウエスタンブロットで得られた像を示す。標準品と同様のバンドが見られ、バンドの濃さも培養日数に比例して濃いことが示唆された。
当業者に周知の方法を用いて本発明のベクターであるpDC6の塩基配列No1267-No1275を配列番号:11 に記載のヒトインターフェロンα2b(hIFNα)をコードするcDNA(以下hIFNαと記載する)に置換し、pDC6/hIFNα(図23)を構築した。
2.5μgのpDC6/hIFNαをリポフェクチン法(Lipofectamine(登録商標) LTX, Invitrogenを使用)を用いて25cm2のカルチャーフラスコ中の4,000,000個のCHO細胞(CHO DG44 cell)に遺伝子導入した。導入方法は製造業者の使用説明書に従った。遺伝子導入48時間後、細胞数を計測した後、細胞を4 mM Gluta MAX(登録商標)-I (Invitrogen)を含むIS CHO-CD w/ Hydrolysate培地(IS Japan)にて希釈した。96ウェルマイクロタイタープレート中に4000 cells/wellの濃度で5枚(480ウェル)播き、5%炭酸ガス存在下で37℃、約3週間培養したところ、生存した細胞が見られた(HT不含培地生育セルライン)。
産生量の検定はELISAにて実施した。コーティングバッフアー(15 mM, Na2CO3, 35 mM NaHCO3, 0.05 % NaN3, pH 9.6)で希釈した0.5μg/mL抗ヒトIFNα抗体(Monoclonal Antibody to Human Interferon-α, Pro. 3423-3, MABTECH)で96ウェルプレート(F96 MAXI SORP Nunc-Immuno plate,Cat no. 442404, Nunc)に4℃、16時間でコートした。ブロッキング液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:3の比率で混合)でブロッキングした後、72時間培養上清(1/40,000-1/640,000希釈)、遺伝子組換え型インターフェロンα-2b(イントロンA注射用1,000, シェリング・プラウ株式会社)の抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)による2倍希釈系列(80~1.25 IU/mL)および抗原抗体希釈液(4% ブロックエース(大日本住友製薬株式会社)とD-PBS(ダルベッコリン酸緩衝液、シグマアルドリッチ)を1:9の比率で混合)をそれぞれ100μLずつアプライし、25℃で1時間インキュベートした。さらに0.5μg/mLのビオチン化ヒトIFNαモノクローナル抗体(Monoclonal Antibody to Human Interferon-α Biotin conjugate, Pro. 3423-6, MABTECH)と25℃、1時間インキュベートした。25℃で30分間インキュベートしたStandard Ultra-Sensitive ABC Staining kit(Reagent A 2 drops,Regent B 2 drops / 10 mL, Pro#32050, PIERCE)を100μL/wellずつアプライし、25℃で30分間反応させた。Sure Blue TMB Microwell Peroxidase Substrate(KPL)を100μL/wellずつアプライし、25℃で30分間反応させた後、1Mリン酸を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450 nmの吸光度を測定した。表9にELISAにて得られた結果、ヒトIFNα産生量の高い上位3サンプルを示す。もっとも産生量が高いラインでは(56.3±5.0) x104 IU/mL/3days, (219.3±11.1) x104 IU/mL/7days, (436.5±17.1)x104 IU/mL/14daysを示した。この値は、クローン化していない初期のセルラインであってしかも遺伝子増幅していない状態において、文献等(J Gen Virol. 1985 Apr;66 ( Pt 4):685-691、Nucleic Acids Res. 1983 Feb 11;11(3):555-573、Phil.Trans.R.Soc.Lond.B299,7-28(1982)、特表2003-530070)で報告されている代表的なインターフェロンαの産生量と比較して非常に高い水準を示していた。
これにより、本発明の発現ベクターは非常に高いタンパク質発現レベルを可能にすることが証明された。続いて培養上清中のhIFNαのWesternblottingを行い、タンパク質の発現を確認した。
上記実施例27において採取したヒトhIFNα産生量上位10サンプルの3、7、14日間培養上清をウエスタンブロッティングにて解析した。培養上清10μLそれぞれに、5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。さらに標準品ヒトIFNα (IFNα2B Expression in Human Cells, Cat HZ-1072, Human Zyme, IFN-α2b ) 10 μg/10 μLおよび標準品ヒトIFNα(iLite(登録商標) Alphabeta IFN stock solution (200 IU/ml), biomonitor)200 IU/ml/10 μLに5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98℃で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。泳動層(DPE-1020、DAIICHI PURE CHEMICALS CO., LTD)に泳動緩衝液(Tris/Glycine/SDS、BIO-RAD)とSuper Sep(登録商標) 10~20% 17well(Wako, )をセットし、加熱処理したサンプル溶液と標準品をSuper(登録商標)10~20% 17well(Wako)に20μL添加し、40mAで55分間電気泳動を行った(電源装置:MyRun、COSMO BIO CO., LTDを使用)。その後、ゲルをガラス板から外して転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に5分間振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸した。Immobilon-P Transfer Membrane(MILLIPORE)をそれぞれ8mLのメタノール(Wako)で15秒、8mLのMilliQ水(MILLIPORE)で2分間、8mLの転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で5分間の順に振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸して活性化させた。転写装置(TRANS-BLO、 SD SEMI-DRY TRANSFER CELL、BIO-RAD)に転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)、活性化させたImmobilon-P Transfer Membrane(MILLIPORE)、転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に浸した泳動後のゲル及び転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)をマイナス側から順に敷き、カバーを被せて80mA(PowerPac HC(登録商標)、BIO-RAD)で1時間半電気泳動を行い、分離したタンパク質をImmobilon-P Transfer Membrane(MILLIPORE)上に転写させた。転写後、Immobilon-P Transfer Membrane(MILLIPORE)を8mLのImmunoBlock(登録商標、大日本住友製薬株式会社 ラボラトリープロダクツ部)に浸して4℃で18時間ブロッキングを行い、0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で2,000倍希釈した10mLのRabbit polyclonal antibody against human interferon alpha (Cat No. 31130-1, PBL)と膜上のタンパク質を室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で5,000倍希釈したPeroxidase Conjugated Affinity Purified Anti-Rabbit IgG F(c)(Goat)(Rock Land)を10mL添加して室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に2 mLのImmobilon(登録商標) Western Chemiluminescent HRP Substrate(MILLIPORE)を添加して化学発光させ、Light-Capture ATTO Cooled CCD Camera System(ATTO)のノーマル設定で3秒間写真を撮影した。図25にウエスタンブロットで得られた像を示す。標準品と同様のバンドが検出された。
当業者に周知の方法を用いて本発明のベクターであるpDC6の塩基配列No1267-No1275を配列番号:12 に記載のヒトオステオポンチン(OPN)をコードするcDNA(以下hOPNと記載する)に置換し、pDC6/hOPN(図26)を構築した。
2.5μgのpDC6/hOPNをリポフェクチン法(Lipofectamine(登録商標) LTX, Invitrogenを使用)を用いて25cm2のカルチャーフラスコ中の4,000,000個のCHO細胞(CHO DG44 cell)に遺伝子導入した。導入方法は製造業者の使用説明書に従った。遺伝子導入48時間後、細胞数を計測した後、細胞を4mM Gluta MAX(登録商標)-I (Invitrogen)を含むIS CHO CD w/H (IS Japan)にて希釈した。96ウェルマイクロタイタープレート中に4000 cells/wellの濃度で5枚(480ウェル)播き、5%炭酸ガス存在下で37℃、約3週間培養したところ、生存した細胞が見られた(HT不含培地生育セルライン)。
各株15 mLを15 mLチューブに取り、1,100rpm、7分間遠心した。上清を捨て、細胞を15 mLの新しい培地(4 mM Gluta MAX(登録商標)-I ((Invitrogen)を含むIS CHO CD w/H (IS Japan))に懸濁し、細胞数を計測した後、細胞数を5 x 105 cells/mLになるように培地で希釈後、7.5 mLを新しいT75Flaskに移し、5%炭酸ガス存在下で37℃、14日間培養した。培養3、7、14日に培養液を1 mL ずつ採取し、9300g、2分間の遠心後に上清を回収した。産生量を測定した。
産生量の検定はELISA Kit (Human Osteopontin Assay Kit, Code 27158, IBL) にて実施した。抗体プレート (抗Human OPN (O-17) Rabbit IgG A.P.固相)に対し、3、7、14日間培養上清(1/10-1/8,000希釈)、組換えヒトOPN標準品(Recombinant human OPN)の希釈用緩衝液 (1 % BSA, 0.05 % Tween-20 含有PBS)による2倍希釈系列(5~320 ng/mL)をそれぞれ100μLずつアプライし、37℃で1時間インキュベートした。さらに標識抗体 (HRP 標識抗Human OPN (10A16) Mouse IgG MoAb Fab A.P.)と4℃、1時間インキュベートした。TMB 基質液を100μL/wellずつアプライし、25℃で30分間反応させた後、停止液 (1N H2SO4)を100μL/wellずつ入れて反応を停止させた。タンパク質濃度の測定はマイクロプレートリーダー(Model680, BioRad社製)を用いて、マイクロプレートリーダーで波長655nmを対照として波長450nmの吸光度を測定した。ライン32では1.12±0.07 μg/mL/3days, 7.40±0.24 μg/mL/7days, 13.75±0.03 μg/mL/14daysを示した。続いて培養上清中のhOPNのWesternblottingを行い、タンパク質の発現を確認した。
上記実施例31において採取したライン32の3、7、14日間培養上清をウエスタンブロッティングにて解析した。培養上清10μLそれぞれに、5%の2-メルカプトエタノール(wako)を含むLaemmli Sample Buffer(BIO-RAD)10μLを混合して98度で5分間加熱(TaKaRa PCR Thermal Cycler PERSONAL、TaKaRa BIOMEDICALS)して還元した。泳動層(DPE-1020、DAIICHI PURE CHEMICALS CO., LTD)に泳動緩衝液(Tris/Glycine/SDS、BIO-RAD)とSuper Sep(登録商標) 10~20% 17well(Wako, )をセットし、加熱処理したサンプル溶液と標準品をSuper(登録商標)10~20% 17well(Wako)に20μL添加し、40mAで55分間電気泳動を行った(電源装置:MyRun、COSMO BIO CO., LTDを使用)。その後、ゲルをガラス板から外して転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に5分間振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸した。Immobilon-P Transfer Membrane(MILLIPORE)をそれぞれ8mLのメタノール(Wako)で15秒、8 mLのMilliQ水(MILLIPORE)で2分間、8 mLの転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で5分間の順に振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら浸して活性化させた。転写装置(TRANS-BLO、 SD SEMI-DRY TRANSFER CELL、BIO-RAD)に転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)、活性化させたImmobilon-P Transfer Membrane(MILLIPORE)、転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))に浸した泳動後のゲル及び転写緩衝液(メタノール(Wako)を30%含んだTris/Glycin Buffer(BIO-RAD))で浸したろ紙(Extra Thick Blot Paper Criterion(登録商標) Size、BIO-RAD)をマイナス側から順に敷き、カバーを被せて80mA(PowerPac HC(登録商標)、BIO-RAD)で1時間半電気泳動を行い、分離したタンパク質をImmobilon-P Transfer Membrane(MILLIPORE)上に転写させた。転写後、Immobilon-P Transfer Membrane(MILLIPORE)を8mLのImmunoBlock(登録商標、大日本住友製薬株式会社 ラボラトリープロダクツ部)に浸して4℃で18時間ブロッキングを行い、0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で2,000倍希釈した10mLの抗ヒトOPNマウスモノクローナル抗体 (Anti-Human Osteopontin (10A16) Mouse IgG MoAb, Cat 10011, IBL)と膜上のタンパク質を室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に0.05%Tween20(Polyoxyethylene (20) Sorbitan Mocolaurate、Wako)を含むD-PBS(Wako)で5,000倍希釈したペルオキシダーゼ標識抗マウスIgG抗体(Goat anti-Mouse IgG (H+L), Cat 115-036-062, Jackson)を10mL添加して室温で振とう(ROTO-SHAKE GENIE、Scientific Industries)させながら1時間反応させた。結合しなかった抗体を除いた後に2 mLのImmobilon(登録商標) Western Chemiluminescent HRP Substrate(MILLIPORE)を添加して化学発光させ、Light-Capture ATTO Cooled CCD Camera System(ATTO)のノーマル設定で30秒間写真を撮影した。図27にウエスタンブロットで得られた像を示す。
さらに、本発明によるタンパク質の生産方法は、ウイルスや微生物を使用しないため、安全性の高いタンパク質生産が可能である。
Claims (10)
- 下記(a)および(b)を含む、哺乳動物宿主細胞において外来遺伝子由来タンパク質の高レベル生産を可能にするための発現ベクター。
(a)コドンを哺乳動物において使用頻度の最も低いものに改変し発現を微弱化させた翻訳障害性ジヒドロ葉酸還元酵素遺伝子カセット(翻訳障害性DHFR遺伝子カセット)
(b)高転写活性プロモーターと高安定性ポリアデニレーションシグナルの間に外来遺伝子組み込み用クローニングサイトを含む遺伝子カセット - 請求項1(a)に記載の翻訳障害性DHFR遺伝子カセットのコドンが、ヒトにおいて使用頻度が最も低いコドンに改変されたものであることを特徴とする請求項1に記載の発現ベクター。
- 請求項1(a)に記載の翻訳障害性DHFR遺伝子カセットのコドンが、アラニンはGCA、アルギニンはCGA、アスパラギンはAAU、アスパラギン酸はGAU、システインはUGU、グルタミンはCAA、グルタミン酸はGAA、グリシンはGGU、ヒスチジンはCAU、ロイシンはUUA、リシンはAAA、プロリンはCCA、フェニルアラニンはUUU、セリンはUCA、トレオニンはACU、チロシンはUAU、および/又はバリンはGUAに改変されたものであることを特徴とする請求項1に記載の発現ベクター。
- 請求項1(a)に記載の翻訳障害性DHFR遺伝子カセットが、プロモーターとして発現誘導性の低いプロモーターを使用していることを特徴とする請求項1に記載の発現ベクター。
- 前記活性の低いプロモーターとして、哺乳動物細胞ではほとんど発現しない遺伝子を由来とするプロモーター、又はエンハンサー部分を除去したプロモーターを使用することを特徴とする請求項4に記載の発現ベクター。
- 請求項1(a)に記載の翻訳障害性DHFR遺伝子カセットにおいてコドンの改変領域が、遺伝子カセットの全長の30%以上であることを特徴とする、請求項1に記載の発現ベクター。
- 請求項1~6のいずれか1項に記載の発現ベクターに外来遺伝子を組み込む工程、および該発現ベクターによりジヒドロ葉酸還元酵素遺伝子欠損宿主細胞を形質転換させる工程を含む、外来遺伝子由来タンパク質の高レベル生産能を有する形質転換体の製造方法。
- 以下(a)~(d)の工程を含む、外来遺伝子由来タンパク質の生産方法。
(a)請求項1~6のいずれか1項に記載の発現ベクターに外来遺伝子を組み込む工程
(b)該発現ベクターによりジヒドロ葉酸還元酵素遺伝子欠損宿主細胞を形質転換させる工程
(c)該形質転換体をヒポキサンチン・チミジン不含培地で培養する工程
(d)培養された形質転換体から外来遺伝子由来タンパク質を回収する工程 - 請求項8(c)の工程において、Chemically Defined medium(CD培地)あるいはCD培地に非動物性の添加物を加えた培地で培養することを特徴とする、請求項8に記載の方法。
- 以下(a)~(c)の工程を含む、外来遺伝子由来タンパク質の高レベル生産能を有する形質転換体のスクリーニング方法。
(a)請求項1~6のいずれか1項に記載の発現ベクターに外来遺伝子を組み込む工程
(b)該発現ベクターによりジヒドロ葉酸還元酵素遺伝子欠損宿主細胞を形質転換させる工程
(c)該形質転換体をヒポキサンチン・チミジン不含培地で培養する工程
