Abstract

Vector de expresión que induce alta productividad de proteínas genéticamente recombinante en células huésped de animales, que comprende un promotor de expresión altamente inducible, de antígeno principal inmediatamente precoz de citomegalovirus humano, o un quimérico sintético CMV5, de beta-actina o precoz de SV40, un sitio de clonación múltiple para inserción del gen y una secuencia de senales de poliadenilación en este orden desde arriba y debajo de ellos un gen resistente a las drogas, como ser a la neomicina o higromicina o zeocina o blaticina, o disoptimizado de codon, que no tiene un promotor capaz de actuar en células animales y además un gen dihidrofolato reductasa. Transformante que comprende dicho vector, método para obtenerlo en un medio si cuero, introduciéndolo en células animales huésped, CHO con déficit de dihidrofolato reductada y aclimatándolo al medio. Es útil para expresar proteína, no obtenible como activa cuando se usa como huésped una bacteria, microorganismo eucariótico o célula de insecto.
