Abstract

Un vector de expresión para permitir la producción de alto nivel de una proteína derivada de gen foráneo en una célula huésped de mamífero, que comprende: (a) un casete del gen de la dihidrofolato reductasa de traducción alterada (casete del gen DHFR de traducción alterada), cuya expresión se atenúa mediante la alteración de los codones con los codones menos frecuentemente utilizados en un mamífero, en donde los codones han sido alterados con ACG para alanina, CGA para arginina, AAU para asparagina, GAU para ácido aspártico, UGU para cisteína, CAA para glutamina, GAA para ácido glutámico, GGU para glicina, CAU para histidina, UUA para leucina, AAA para lisina, CCA para prolina, UUU para fenilalanina, UCA para serina, ACU para treonina, UAU para tirosina, y/o GUA para valina, y en donde una región de codón alterado en el casete del gen DHFR de traducción alterada es 30% o más de la longitud completa del casete del gen; y (b) un casete de un gen que comprende un sitio de clonación para la integración de un gen foráneo entre un promotor altamente activo transcripcionalmente y una señal de poliadenilación altamente estable.
