Abstract

La présente invention concerne de nouvelles microparticules de polyaminoacides amphiphiles transporteurs de principe(s) actif(s) -PA-, en particulier protéinique(s) et peptidique(s), ainsi que de nouvelles formulations pharmaceutiques à libération modifiée contenant lesdites microparticules de PA. Le but de l'invention est de mettre au point de nouvelles microparticules chargées en PA obtenues par agrégation de nanoparticules de polyaminoacides amphiphiles, et ayant des propriétés améliorées, notamment sous forme solide sèche, au regard de leur aptitude à la dispersion, et, s'agissant de la suspension reconstituée, de sa stabilité et de son aptitude à être facilement manipulée et injectée. L'invention concerne dans un premier aspect, des microparticules de polyaminoacide amphiphile (PO) contenant au moins un PA (associé de façon non covalente), formant spontanément une suspension colloïdale de nanoparticules dans l'eau, à pH 7,0, en condition isotonique; lesdites microparticules étant caractérisées a. en ce qu'elles sont obtenues par atomisation d'une solution ou d'une suspension colloïdale de PO comprenant au moins un PA, b. par une taillecomprise entre 0,5 et 100 μm, c. et en ce qu'elles sont dispersables en suspension colloïdale. L'invention concerne également le procédé de préparation de ces microparticules, une formulation liquide comprenant une suspension de ces microparticules PO/PA, un procédé et un nécessaire de reconstitution de cette formulation, et une forme sèche de cette formulation.
