発明が解決しょうとする課題
[0008] 本発明は、様々な薬理作用を有するマンゴスチン誘導体を安全性が高!、溶媒を用 いて、効率良く精製できるマンゴスチン誘導体の単離方法と、安全性の高いマンゴス チン誘導体を含有する医薬品、及び健康食品を提供することである。
課題を解決するための手段
[0009] 効率の良!ヽ方法で、安全性の高!ヽマンゴスチン誘導体を提供するために鋭意研究 を重ねた結果、マンゴスチン果皮を水洗後、含水エタノールで抽出して、マンゴスチ ンの粗結晶を得て、含水エタノールに再溶解後、結晶化と溶解を一度、又は数回繰 り返しながら結晶化させることによって、純度の高いマンゴスチン誘導体を得ることに 成功した。
[0010] 本発明の第一の特徴は、マンゴスチン果皮に水をカ卩えて 90°C以上で加熱洗浄後、 洗浄した果皮に 25〜75% (vZv)エタノールを加えて 50〜85度で加熱抽出後、抽 出物を濾過して、得られた抽出液を冷却して晶出する粗マンゴスチン結晶を濾取し

発明の効果
[0016] マンゴスチン果皮に水をカ卩えて 90°C以上、好ましくは 95〜100°Cで加熱洗浄する こと〖こよって、マンゴスチン果皮に含まれる赤色物質、粘性物質、吸湿性物質等の不 純物質が多量に溶出されて、加熱洗浄後のマンゴスチン果皮から純度の高 、マンゴ スチン結晶を含有する組成物を得ることが出来る。
[0017] 安全性の高 、エタノールを溶媒として用 ヽてマンゴスチンを抽出及び晶出を行うこ とにより、最終製品としても安全性の高い高純度のマンゴスチン誘導体を含有する組 成物を得ることができる。他の有機溶媒を用いた分画操作やシリカゲルまたはイオン 交換榭脂を用いたカラムクロマトグラフィーを使用しないため、安全でかつ工程を短く することができ、低コストで高純度のマンゴスチン誘導体を含有する組成物を得ること ができる。
[0018] 本発明の単離方法で得られるマンゴスチン誘導体を含有する組成物は純度の高!、 a一マンゴスチンと γ—マンゴスチンを一定の割合で含有している。この (X一マンゴ スチンと γ—マンゴスチンの含有割合は、薬理効果を最も発揮できる割合であって、 癌の予防治療薬として利用することができる。さらに、各種の食物の形態や飲料の形 態で健康食品として摂取することができる。
発明を実施するための最良の手段
[0019] 以下の実施例では、本発明を実施するいくつかの好ましい形態を例示する力 特 許請求の範囲に記載された発明の範囲を限定することは意図していない。
[0020] マンゴスチン果皮に対し 2〜 10倍量の水をカ卩えて 90°C以上で、加熱洗浄する。マ ンゴスチン果皮に含まれる赤色物質、粘性物質、吸湿性物質等のマンゴスチンの精 製に障害となる不純物質が、本発明方法における加熱洗浄によって多量に溶出除 去される利点を有し、しカゝも、本発明の洗浄方法でマンゴスチン果皮を洗浄してもマ ンゴスチンの溶出は殆ど認められな力つた。
[0021] 高温の水で洗浄したマンゴスチン果皮に濃度が 25〜75% (v/v)の熱エタノール を加えて、 1時間抽出する。抽出液を冷却して析出する粗マンゴスチン結晶を濾過し
て集め、この粗マンゴスチン結晶をエタノールに再溶解して、水に滴下してエタノー ル濃度を薄めて再結晶化する。再結晶化は一度でも良いが繰り返すことによって、よ り純度の高いマンゴスチン結晶を得ることができる。このようにして得られたマンゴスチ ン結晶は at一マンゴスチンを 75〜85%、 γ一マンゴスチンを 7〜 15%の割合で含 み、かつ a一マンゴスチンと γ—マンゴスチンの含有量の合計は 90%以上である。
[0022] マンゴスチン果皮力 アルコールで抽出され結晶化された ex一マンゴスチンと γ— マンゴスチンの含有割合が 75〜85%: 7〜15%の割合で含有する本発明のマンゴ スチン含有組成物は、前記のマンゴスチンを治療的有効量で含む薬学的に許容し 得る製剤として、 1つまたは複数の薬学的に許容し得る担体 (添加剤)および Ζまた は希釈剤とともに薬学的組成物として処方される。以下で詳細に説明するように、本 発明のマンゴスチン含有組成物は、以下のことに適応したものを含めて、固体または 液体での投与のために具体的に処方され得る:(1)経口投与、例えば、水薬 (水溶 液もしくは非水溶液または懸濁液)、錠剤、巨丸剤、粉末薬、顆粒剤、舌に塗布する ためのペースト;(2)非経口投与、例えば滅菌溶液もしくは懸濁液として例えば皮下 、筋内もしくは静脈内注射による;(3)局所応用、例えば皮膚に応用されるクリーム、 軟膏またはスプレーとして;または (4)膣内または直腸内に、例えば膣座薬、クリーム または発泡剤として。しかし、いくつかの実施形態では、本発明の化合物を単に滅菌 水に溶解または懸濁してもよ 、。

[0029] 本発明の剤形は、経口、経鼻、局所(口内および舌下を含む)、直腸、膣および Z または非経口投与に好適なものを含む。剤形は、単位投薬形態で都合よく差し出さ れてもよぐ薬学分野で周知のいかなる方法によって調製されてもよい。担体材料と 組み合わせて単一投薬形態を作製することができる活性成分の量は、治療されるホ スト、特定の投与方式に応じて変わるであろう。担体材料と組み合わせて単一投薬形 態を作製することができる活性成分の量は一般に、治療効果を生じる化合物の量で あろう。一般に、 100パーセントのうち、この量は、約 1%力も約 99%まで、好ましくは 約 5%から約 70%まで、最も好ましくは約 10%から約 30%までの範囲の活性成分で ある。

[0048] さらに、マンゴスチン果皮力もアルコールで抽出されたマンゴスチン結晶は、 α—マ ンゴスチンと Ί一マンゴスチンの含有割合が 75〜85%: 7〜15%の割合で含有する 本発明組成物を含む健康食品について例示する。本発明のマンゴスチン含有組成 物は、適当な賦形剤等と共に加工して健康食品とすることができる。前記賦形剤とし ては、例えば、食品素材、食品添加物を挙げることができる。例えば、糖質 (糖類、糖 アルコール)、甘味料、酸味料、酸化防止剤、日持ち向上剤、保存料等を挙げること ができる。この中、糖質、甘味料、酸味料は、それぞれ、 0. 01〜90重量%の範囲内 で配合するのが適当であり、 0. 1〜70重量%の範囲内が好ましい。酸ィ匕防止剤、日 持ち向上剤、保存料は、それぞれ 0. 001〜: LO重量%の範囲内で配合するのが適 当であり、 0. 01〜1重量0 /0が好ましい。その他、ビタミン類、アミノ酸類、ミネラル類、 増粘安定剤、着色料、食物繊維、香料、乳化剤、 ΡΗ調整剤等を適当量配合すること ができる。

[0060] 本発明の組成物を含有する健康食品は、例えば、クッキー、キャンディー、ゼリー、 グミ、シャーベット、ケーキ、饅頭、ガム、アイスクリーム、シュークリーム、ワッフル、煎 餅、錠菓等の菓子、果汁入り飲料、果汁なし飲料、炭酸飲料、乳酸菌入り飲料、コー ヒー、茶等の飲料、固形状の、ゲル状の若しくは液状の又はそれらを組み合わせたス ポーッフーズゃスポーツドリンク、カプセル型、錠剤型、又は散剤型の健康食品、タレ 、醤油、ソース、カレー粉、カレールー等の調味料又は調理用素材、その他、麵、パ ン、シリアル、レトルト食品等を挙げることができる。
実施例 1
[0061] 図 1に示す工程に従って、マンゴスチン果皮カゝらマンゴスチンを抽出精製した。まず 、マンゴスチン果皮の乾燥物 60kgに対して水 600Lをカ卩えて、 90°C以上、好ましくは 95°C〜100°Cで攪拌ししながら 1時間洗浄を行った。加熱洗浄工程で、マンゴスチ
ン果皮に含まれる赤色物質、粘性物質、吸湿性物質等の不純物質が多量に溶出さ れて、以後の抽出工程で純度の高いマンゴスチン結晶を含有する組成物を得ること が出来る。
[0062] 加熱洗浄したマンゴスチン果皮を濾過して、その残渣に 50% (v/v)エタノールを 6 OOLカ卩えて、 80°C〜85°Cで 1時間攪拌しながら抽出して、抽出液を濾過した。
[0063] 濾液を常温にまで徐々に温度を下げた後、常温で 1夜放置して、粗マンゴスチン結 晶を析出させた。粗マンゴスチン結晶の析出した液を濾過して粗マンゴスチン結晶を 分離した。分離した粗マンゴスチン結晶を 50%VZVエタノール 10Lで洗浄後、 60 °Cで真空乾燥して、粗マンゴスチン結晶粉末を 2. 4kgを得た。
[0064] 粗マンゴスチン結晶 2. 4kgに対してエタノール 24Lをカ卩えて、常温で 1時間攪拌し て溶解した。この溶解液を常温で攪拌しながら水 36Lに滴下し、その後 80°C〜85°C で 1時間攪拌した。この液を常温まで徐々に冷却し、常温で 1夜攪拌してマンゴスチ ン結晶を析出させ濾過後、得られたマンゴスチン結晶を 50%VZVエタノールで洗 浄した後、真空乾燥してマンゴスチン結晶を含有するマンゴスチン含有組成物を得 た。
[0065] マンゴスチンの定量法
上記方法により得られたマンゴスチン含有組成物の定量は、マンゴスチン含有組成 物 20mgを正確に計り、エタノールをカ卩えて正確に 100mlに溶解して試料とした。別 に ex一マンゴスチン標準品 20mgを量り取り、エタノールをカ卩えて正確に 100mlに溶 解して、標準溶液一 1とした。別に γ—マンゴスチン標準品 lOmgを正確に計り取り、 エタノールをカ卩えて正確に 50mlに溶解した。この液 5mlを正確に計り、エタノールを 加えて正確に 50mlとして、これを標準溶液一 2とした。
[0066] 上記各試料の分析は高速液体クロマトグラフィーを用いて分析した。使用したカラ ムは Develosil ODS- 5( φ 4. 6 X 250mm,野村化学製)を用いて、移動相にァセトニ トリル Ζθ. 05Mリン酸水溶液(7 : 3)を用い、流量 1. OmlZminで、 40°Cで展開し、 波長 365nmで測定を行った。高速液体クロマトグラフィーでマンゴスチン含有組成 物を分析した結果、本願発明方法で得られたマンゴスチン含有組成物は OC一マンゴ スチンが 82. 1 %、 γ—マンゴスチンが 9. 0%の割合で含まれていた。上記方法で
分離定量したマンゴスチン (キサントン)誘導体の化学構造を図 2に示す。
実施例 2
[0067] マンゴスチンの薬理作用
図 3は.ヒト大腸癌細胞株 DLD— 1細胞に対するマンゴスチン (キサントン)誘導体 の細胞増殖抑制効果について検討した結果を示している。 DLD— 1細胞は、牛胎児 血清 10%を含む、 RPMI1640培地中で DLD— 1細胞を l〜2xl05Zmlの濃度で、 3 7°Cの C02インキュベーターで培養した。 DLD— 1の細胞 l〜2xl05/ml濃度に 対して、種々濃度のマンゴスチン (キサントン)誘導体をカ卩えて、 DLD— 1細胞の増殖 に及ぼすマンゴスチン誘導体の効果について検討した。その結果、図 3に示すように 、 bM- OMe以外は DLD— 1細胞の増殖抑制効果が認められた。この増殖抑制効果 は g一マンゴスチン(γ—M) > α一マンゴスチン(a— Μ) > β一マンゴスチン(b— Μ) の順で活性が高力つた。
[0068] 図 4はマンゴスチン (キサントン)誘導体によるアポ一トシスの誘導を調べた結果であ る。各マンゴスチン(キサントン)誘導体 20 Mを含む培地で、 DLD— 1細胞を 72時 間処理後、へキスト 33342核染色法により染色したところ DLD 1細胞死が確認され た。マンゴスチン (キサントン)誘導体処理によって死んだ細胞は核の分断、クロマチ ンの濃縮が観察され、形態学的にアポ一トシスを誘導していることが示された (A図参 照)。さらに、生化学的にも DNAラダーが認められた (B図参照)。これらの事実から、 マンゴスチン (キサントン)誘導体による DLD— 1細胞の増殖抑制効果はアポ一トシス によるものであることが確認された。
[0069] 図 5は、マンゴスチン (キサントン)誘導体による DLD—1細胞の増殖抑制効果がァ ポートシスによるものであることから、アポトーシスのメカニズムを調べるために DLD 1細胞の細胞周期に対するマンゴスチンの影響について検討した。アポトーシスの メカニズムを調べる方法として、 Propiodium Iodide (PI)を用いて核を染色して、 FACS にて DNA量をモニターした。その結果、 a-M ( a一マンゴスチン)と b— M ( j8—マンゴ スチン)は G1/S期, g— Μ ( γ—マンゴスチン)は G2/M期の細胞周期の停止を誘導し た。以上の結果力もマンゴスチン (キサントン)誘導体は DLD— 1細胞の細胞周期を 停止させて、アポ一トシスを誘導して 、ることが示された。
[0070] 図 6は、上記のように、 DLD— 1細胞の細胞周期に対して a一マンゴスチンと γ— マンゴスチンは作用点が異なることが明ら力となった。このように作用点の異なる α— マンゴスチンと Ί一マンゴスチンを併用することで抗腫瘍活性がより高くなることが期 待される。そこで a一マンゴスチンと γ—マンゴスチンの併用効果について、 Κ562 細胞の細胞増殖抑制効果にっ 、て検討をカ卩えた。ここで用いた白血病細胞株 Κ562 は種々の化学物質により赤芽球系細胞や巨核球系細胞に分化することが知られて いる細胞である。その結果、 Κ562細胞に対して、それぞれ 10 Μ濃度で 72時間処 理した場合、 a一マンゴスチンまたは γ—マンゴスチンの単独で処理した場合に比 較して a一マンゴスチンと γ—マンゴスチンを併用することによって、細胞増殖抑制 効果がより高くなることが示された。その a一マンゴスチンと γ—マンゴスチンの混合 割合は、 80: 14の方が 50: 50よりも活性が高 、傾向が示された。
[0071] 図 7は、マンゴスチンの外皮力も種々の溶媒で抽出したマンゴスチン誘導体の細胞 増殖抑制効果にっ 、て検討した結果を示す。図 7に示すようにマンゴスチンの抽出 物の濃度を 10 gZmlで 48時間処理した場合、抽出物中の a一マンゴスチンと γ 一マンゴスチンの含有比率と細胞増殖抑制効果は正の相関を示して 、た。このこと は α一マンゴスチンと Ί一マンゴスチンが腫瘍細胞の細胞増殖抑制活性に対して、 主たる役割を果たして 、ることを示唆して 、る。
実施例 3
[0072] 実施例 1で得られたマンゴスチン含有組成物を以下に示す割合で配合して、マンゴ スチンを含む錠剤を調製した例を示す。

上記の製剤例におけるマンゴスチンの含量は、適用する対象疾患、重傷度等、投 与する患者に対応できるように適宜変更し、含有量の異なるものを調製することがで きる。
実施例 4
[0073] 実施例 1で得られたマンゴスチン含有組成物を以下に示す割合で配合したマンゴ スチンを含有する健康食品 (キャンディー)を調製した例を示す。

上記の健康食品におけるマンゴスチンの含量は、適用する食品の種類、摂食期間 、摂食量により異なるので、それぞれ目的に応じて対応できるように適宜変更し、食 品の種類やマンゴスチンの含有量の異なるものを調製することができる。

Claims
[1] マンゴスチン果皮に水を加えて 90°C以上で加熱洗浄後、洗浄した果皮に 25〜75%
(vZv)エタノールをカ卩えて 50〜85度で加熱抽出後、抽出物を濾過して、得られた 抽出液を冷却して晶出する粗マンゴスチン結晶を濾取した後、さらにこの結晶を 25 〜75% (vZv)エタノールに溶解後、再結晶させてマンゴスチン結晶を得ることを特 徴とするマンゴスチンの単離方法。
[2] マンゴスチン果皮に水を加えて 90°C以上で加熱洗浄後、洗浄した果皮に 25〜75%
(vZv)エタノールをカ卩えて 50〜85度で加熱抽出後、抽出物を濾過して、得られた 抽出液を冷却して晶出する粗マンゴスチン結晶を濾取した後、さらにこの結晶を 25 〜75% (vZv)エタノールに溶解後、再結晶させたマンゴスチン結晶を含有すること を特徴とするマンゴスチン含有組成物。
[3] a一マンゴスチンと γ—マンゴスチンの含有割合が 75〜85%: 7〜15%で、かつ oc 一マンゴスチンと Ί一マンゴスチンの含有量の合計が 90%以上であることを特徴と する請求項 1に記載のマンゴスチンの単離方法。
