Abstract
【課題】 腐植物質に含まれるフルボ酸とI型アレルギーの発症機序との関係を解明し、フルボ酸によってI型アレルギーの発症を抑制する。
【解決手段】 抗原感作段階又は/及び抗体感作段階や細胞の脱顆粒段階をフルボ酸によって抑制するI型アレルギーの抑制剤や前記フルボ酸を少なくとも含む組成物を提供する。また、フルボ酸を用いて特異的減感作を行うことによりI型アレルギーの発症を抑制する方法、特に抗原感作段階又は/及び抗体感作段階をフルボ酸によってI型アレルギーの発症を抑制する方法を提供する。あるいは、フルボ酸を用いて非特異的減感作を行うことによりI型アレルギーの発症を抑制する方法を提供する。

フルボ酸を用いる I型アレルギー抑制剤及び I型アレルギーの発症抑制方 法
技術分野
[0001] 本発明は、 I型アレルギーの発症を抑制する技術に関する。より詳しくは、腐植物質 に含まれるフルボ酸の特異的減感作作用に基づく I型アレルギーの発症を抑制する 技術に関する。
背景技術
[0002] 「腐植物質」は、植物が腐植することによって生成する有機成分のことである。国際 腐植物質学会(International Humic Substances Society,略称 IHSS)における定義 によると、この腐植物質は、土壌を水酸ィヒナトリウム等のアルカリで抽出した分画、あ るレヽは天然水で XAD樹脂に吸着し、希アルカリ水溶液で溶出される分画のことであ り、その中で、酸により沈殿する分画を「フミン酸 (腐植酸)」、沈殿しない分画を「フル ボ酸(Fulvic Acid;)」、とそれぞれ呼ぶ。
[0003] ここで、本発明の主題に係わる前記フルポ酸は、鉄と強く結びつく性質を有するた め、海中への鉄供給源となること、あるいは、銅などの有害重金属に対する凝集能( 錯形成能)、発癌性物質である六価クロムの還元無毒化、有機ハロゲン化物の脱塩 素化などのような環境浄化機能を有することが知られている。しかしながら、フルボ酸 の多岐にわたると考えられる作用は、未だ詳しく解明されていない。
[0004] 近年では、上記した環境浄化機能以外に、フルボ酸の生体に及ぼす生理的作用 に関する研究も、徐々に始まっている。例えば、特許文献 1には、フルボ酸 (特に、ォ キシフルボ酸)、その塩、エステル又は誘導体を含む医薬組成物であり、特に、炎症 、ァクネ、湿疹、細菌又は真菌若しくはウィルスの感染症などの疾病に有効な医薬組 成物が開示されている。当該特許文献 1に示された実施例では、ォキシフルボ酸のヒ ト好中球によるオキシダント産生に及ぼす作用、マウス炎症部位へのクリーム剤塗布 による抗炎症性の評価、ネコ及びィヌの化膿性外傷性皮膚炎に対する評価、実験動 物による経口毒性確認試験、ォキシフルボ酸を含むフリーム剤を用いた抗微生物確

認試験などが行われており、概ね、ォキシフルボ酸の皮膚外用薬として抗炎症効果 を確認している。
[0005] 特許文献 2には、腐植土力 溶出される水溶性腐植物質であるフルボ酸、酵素、ビ タミン類、アミノ酸類、ミネラル類などを含む多成分系化粧品原料の抗菌、抗アレルギ 一作用、抗炎症作用、アレルギー肌やアトピー肌への低刺激性、かゆみ軽減などの 作用が開示されており、加えて、口中に使用する化粧品原料への用途も開示されて いる。しかし、当該特許文献 2に示された実施例では、皮膚への刺激を確認する試験 、抗菌力確認試験、経口投与毒性確認試験、活性酸素消去活性を確認する試験な どが開示されているのみである。
[0006] また、特許文献 3には、腐植土力も水や希アルコール等によって抽出される抽出液 であり、その pH値が 4. 0以下である抽出液を有効成分として含有する抗菌水が開示 されている。この特許文献 3には、前記抽出液が虫歯や歯周病の原因となる口腔細 菌ゃ黄色ブドウ状球菌などの食中毒原因菌に対する増殖抑制効果が示されている。 特許文献 1:特表 2002— 526407号公報。

発明の開示
発明が解決しょうとする課題
[0007] 上掲した特許文献 1〜3には、フルボ酸の抗菌作用ゃ抗炎症作用が主体的に開示 されているに留まっており、フルボ酸と I型アレルギーとの間の直接的な因果関係、よ り詳しくは、フルボ酸の I型アレルギーの発症機序における減感作に係わる作用につ いては、一切開示されておらず、示唆すらもされていない。
[0008] そこで、本発明は、腐植物質に含まれるフルボ酸と I型アレルギーの発症機序との 関係を解明することによって、フルボ酸による減感作に基づく I型アレルギーの発症 抑制技術を提供することを主な目的とする。
課題を解決するための手段
[0009] まず、本発明の主題である「I型アレルギー」は、アレルギー反応の一類型であり、ァ ナフイラキシー型又は即時型とも呼ばれる。この I型アレルギーは、抗原に曝露された
ことにより産生した IgE抗体などの抗体が肥満細胞又は好塩基球表面にあるレセプタ 一に対して Fc部分で強く結合すると、前記レセプターが架橋して細胞表面を変形し 、各種酵素の活性ィ匕ゃカルシウムイオン (Ca2+)の流入が起きて細胞外に顆粒を放 出し (脱顆粒現象)、その顆粒力もヒスタミン、セロトニン、 j8 -へキソサミニダーゼなど の酵素などのメディエーター (化学伝達物質)が放出される。これらのメディエーター は毛細血管の透過性を亢進させ、鼻粘膜、気管支粘膜、皮膚などで炎症を惹起する 。アレルギー性鼻炎、気管支喘息、アトピー性皮膚炎はこの I型アレルギーによって 発症する。
[0010] 本発明は、前記 I型アレルギーの発生機序とフルボ酸との関係を解明し、フルボ酸 による特異的減感作に基づく I型アレルギーの発症抑制技術を提供する。
[0011] 具体的には、本発明は、抗原感作段階又は Z及び抗体感作段階をフルボ酸によ つて抑制する I型アレルギーの抑制剤、ある 、は細胞の脱顆粒段階をフルボ酸によつ て抑制する I型アレルギーの抑制剤、特に、細胞へのカルシウムイオン (Ca2+)の流入 を阻害することにより前記脱顆粒段階を抑制する I型アレルギー抑制剤を提供する。 これらの抑制剤に用いるフルボ酸は、例えば、草炭力 抽出されたものを好適に採用 できる。
[0012] また、本発明では、抗原感作段階又は Z及び抗体感作段階にぉ 、て I型アレルギ 一を抑制するフルボ酸を少なくとも含む組成物を提供する。この組成物は、例えば、 洗顔料、クリーム、ノ ックなどのスキンケア化粧品などの化粧品、皮膚外用薬、貼薬 その他の医薬、医薬部外品、飲食物などに配合することによって、その効果を発揮さ せることができる。なお、商品の配合成分中には、当該商品の目的や用途に応じて、 フルボ酸含有組成物以外の成分を添加するのは自由であり、一例を挙げれば、毛穴 の汚れを吸着する成分とフルボ酸を組み合わせた洗顔料、コラーゲンなどの保湿成 分とフルボ酸を組み合わせたクリームやパック、その他の薬効成分とフルボ酸を組み 合わせた医薬、その他の機能性成分とフルボ酸を組み合わせた飲食物などである。
[0013] さらに、本発明は、フルボ酸を用いて特異的減感作を行うことにより I型アレルギー の発症を抑制する方法、特に抗原感作段階又は Z及び抗体感作段階をフルボ酸に よって I型アレルギーの発症を抑制する方法を提供する。あるいは、フルボ酸を用い
て非特異的減感作を行うことにより I型アレルギーの発症を抑制する方法、特に、フル ボ酸を用いて脱顆粒を阻害することにより I型アレルギーの発症を抑制する方法を提 供する。この方法の効果は、例えば、薬剤や飲食物などの形態としてフルボ酸を生 体内に投与したり、塗布、貼付、スプレーなどの方法でフルボ酸を皮膚へ接触させた りすること〖こよって得ることができる。
[0014] ここで、本発明に関連する主たる技術用語について説明する。
[0015] まず、「フルボ酸」とは、国際腐植物質学会 (IHSS)が定めた方法 (後述の実施例で 説明)に準じて、腐植物質である原料から得られる画分中に含まれるフルボ酸を意味 する。
[0016] 「草炭 (peat)」は、前記フルボ酸を抽出するための好適な原料であって、沼沢や湿 地帯において、諸種の植物が成長 *枯死し、酸素の供給が不十分な水中で比較的 低温の条件下で、数百年から数万年の間、分解不完全な状態で堆積したものである 。この草炭は、植物遺体が完全に水中に侵され空気に触れないため酸化され難ぐ また好気性菌類の作用を受けないし嫌気性菌類の作用があるにしても比較的微弱 で、きわめて分解し難い状態に長期間置かれた結果生成される。この過程を泥炭化 と呼び、その本質は十分に明らかにはされていないが化学的には加水分解が主であ り、脱炭酸も起こっていると考えられている。草炭の有機質成分は炭素、水素、酸素、 窒素、硫黄の五種類の元素カゝら主として構成されている。草炭の元素組成が草炭形 成過程における有機物の分解特性を反映している。一般には、草炭の分解が進め ば、草炭有機質成分の炭素、水素、硫黄および窒素含有量が増加し、酸素含有量 が減少する。草炭有機質成分の元素組成に関しては充分研究が行われ、木材と褐 炭の中間に位置することが確認されて 、る。分解が弱!ヽ草炭有機質成分の元素組 成が木材に近ぐ分解が進んだ草炭の有機質元素組成は褐炭に近い。
[0017] 「抗原感作段階」とは、上述した I型アレルギー発生機序過程における抗原抗体反 応の段階を意味する。
[0018] 「抗体感作段階」とは、上述した I型アレルギー発生機序過程にぉ 、て IgE抗体がレ セプターと結合する段階を意味する。
[0019] 「特異的減感作」とは、抗原によって産生した IgE抗体などの抗体力も肥満細胞や
好塩基球に対して刺激を伝える過程におけるいずれかの段階を抑制したり、遮断し たりすることを言う。
[0020] 「非特異的減感作」とは、肥満細胞や好塩基球の脱顆粒現象を抑制又は阻害する こと、即ち肥満細胞や好塩基球カゝらのメディエーター (化学伝達物質)の放出を抑制 又は阻害することを言う。
発明の効果
[0021] 本発明によれば、 I型アレルギーの発症を抑制することができる。特に、 I型アレルギ 一反応の抗原感作段階又は Z及び抗体感作段階をフルボ酸によって抑制したり、 細胞の脱顆粒段階をフルボ酸によって抑制したりすることができる。
[0022] このフルボ酸を含む組成物を、洗顔料、化粧品などの皮膚に作用する商品に使用 した場合は、 I型アレルギーの発症抑制効果に加えて、美白効果、保湿効果、化粧 水の水分浸透促進効果、化粧成分の浸透促進効果、乾燥防止効果などの効果も得 ることがでさる。
実施例 1
[0023] <フルボ酸の抽出方法 >。
[0024] 実験には、図 1に示す国際腐植物質学会 (IHSS)が定めた方法に準じて、草炭から XAD-7榭脂吸着画分のフルボ酸を抽出した。草炭としてカナダ産草炭 (Sphagnumdn oss) peat, Fisons Horticulture, Inc.ノ 用いた。
[0025] まず原料である前記カナダ産草炭を、 O.lmolじ1の塩酸 (HCL)で酸処理し、その不 溶成分を lmolじ1の水酸ィ匕ナトリウム (NaOH)水溶液で抽出し、続いて遠心分離により 得られた上澄み液に 6molじ1の塩酸をカ卩えてフミン酸 (HA)を析出させた。
[0026] その全ての過程で得られる水溶液を、 XAD— 7榭脂(ORGANO Co.)を用いて吸着 させ、 AG MP- 50陽イオン交換榭脂(Bio- Rad Laboratories)にて脱塩を行った後に凍 結乾燥させたものをフルボ酸とした。なお、原料である前記カナダ産草炭力 のフル ボ酸の抽出率を乾燥原料に対する抽出後の乾燥フルボ酸との重量比から算出した 結果、 6.4%であった。
実施例 2
[0027] <フルボ酸の分析〉。本発明に係るフルボ酸の性質を把握するために、排かん水
、風化炭、カナダ産草炭、黒ぼく土のそれぞれから得られるフルボ酸の化学分析を 行った。
[0028] (1)元素組成についての分析。
[0029] フルボ酸の炭素(C)、水素(H)、窒素(N)は、 CHN分析器 (YANACO CHN COR DER MT-5)を用いて測定し、全灰分を差し引いた (全)量当たりの (組成)重量百分 率として求め、酸素についてはその残りとして求めた。元素組成の値は、無水'無灰 ベース (d.a.l)で表示した。
[0030] 図 2には、元素組成に関する上記分析結果を示した。なお、図 2において、「KsW」 は排かん水、「WC」は風化炭、「CP」はカナダ産草炭、「AS」は黒ぼく土をそれぞれ 表している。
[0031] (2)赤外線吸収スペクトル (FT-IR)分析。
[0032] フルボ酸の赤外線吸収スペクトルの測定は、 JASCO FT/IR-3分光計により公知の KBr錠剤法に基づいて行った。試料 lmgと KBrlOOmgを用いて錠剤にし、 400-4000c m—1の吸収スペクトルを測定した。
[0033] 図 3に、異なる原料力 抽出されたフルボ酸 (FA)の赤外線吸収スペクトル (FT-IR) を示した。この図 3において、「Ks」は原かん水、「KsW」は排かん水、「WC」は風化 炭、「CP」はカナダ産草炭、「AS」は黒ぼく土をそれぞれ表している。
[0034] この図 3を見ると、 3400cm— 1付近の吸収は、水素結合した OH基に由来し殆どのフル ボ酸では強い吸収を示した力 風化炭 FA(WC-FA)の吸収が比較的弱いものであ つた。脂肪族炭素に起因すると思われる 2900cm— 1付近の吸収において、土壌 (AS)、 草炭 (CP)、風化炭 (WC)の各フルボ酸では大きな違いが認められな力つたのに対 し、かん水 FA (Ks-FA、 KsW-FA)のでは明らかな吸収が認められている。 1720cm" 1と 1620cm— 1付近に二つの吸収がみられる力 前者は主にカルボキシル基の C=0伸 縮による吸収を示し、若干カルボニル基による吸収も示すと考えられる。後者は、水 素結合したカルボニル基と芳香族構造の共役 C=Cに由来した吸収の二つの構造が それぞれ貢献していると思われる。 1720cm— 1付近の吸収は,草炭 FA(CP-FA)に比 ベ風化炭 FA(WC-FA)の方が若干強い吸収を示し、かん水 FAの吸収が著しいも のであった。 1620cm— 1付近の吸収は,ほとんどの試料で比較的弱い吸収し力認める
ことができなぐこれはフミン酸に比べフルボ酸にキノン基が少ない事実と一致してい る。 1400cm— 1付近の吸収スペクトルと 1300〜1200cm_1区域の比較的広いスペクトルは カルボキシル基の C-0伸縮振動と OHの変角振動に由来するものであると考えられて いる。全てのフルボ酸において吸収が認められている力 草炭、土壌および風化炭 F Aに比べかん水 FAの吸収強度が比較的強い。 800cm— 1付近の小さい谷間はほとんど の試料で明確にみられている。これは芳香環上の C-H変角振動ではないかと考えら れている。このように赤外線吸収スペクトルの結果から、異なる原料力 抽出されるフ ルポ酸 (FA)は、同一の基本的化学構造を有する一連の化合物の集合体であると結 論される。
[0035] 図 4は、原料の異なるフルボ酸(FA)の水素磁気共鳴 !" NMR)スペクトルを示し ている。スペクトルのパラメータは、 0-1.9ppmは脂肪族プロトン、 1.9-3.2ppmは主に力 ルボキシル炭素へつながる水素、 3.2— 6.2ppmは炭化水素炭素など帰属の水素、 6.2 -8.6ppmは芳香族炭素帰属水素をそれぞれ示している。
[0036] この測定の結果から、原カン水由来のフルボ酸 (Ks— FA)ゃ排かん水由来のフル ボ酸 (KsW— FA)は、 0-1.9ppm付近に現れる脂肪族水素帰属のピークが顕著であ るので、脂肪族が多く含まれているのに対して、風化炭由来のフルボ酸 (WC— FA) は、芳香族水素帰属を示す 6.2-8.6ppm付近のピークが比較的強ぐ芳香族が多く含 まれて 、ることが明らかになつた(図 4参照)。
[0037] また、カナダ産草炭由来のフルボ酸 (CP-FA)では、黒ぼく土由来のフルボ酸 (AS -FA)と同様に、比較的多くの複合スペクトルが得られた。したがって、カナダ産草炭 由来のフルボ酸(CP- FA)は、排かん水由来のフルボ酸 (KsW— FA)には及ばない 力 風化炭由来のフルボ酸 (WC— FA)に比べて脂肪族水素と炭水化物水素を多く 含む、腐植物資としては比較的若い分解段階のフルボ酸であることがわかった(図 4 参照)。
[0038] (3) 13C核磁気共鳴スペクトル(13C- NMR)分析。
[0039] 13C核磁気共鳴スペクトルは、 BRUKER AMX-400分光計を用いて、公知の交差分 極 ·マジック角回転 (CP/MAS)法で粉体測定を行った。測定周波数は炭素に対し 100 .0MHzであった。微粉砕試料約 200mgを専用セルに入れ 25〜26°Cの室温で、パルス
くり返し時間は 5秒とし、 10,000回の積算を行った。この分析結果を、図 5に示した。
[0040] この図 5は、カナダ産草炭と風化炭のそれぞれから抽出されたフミン酸 (HA)とフル ボ酸(FA)の13 C- CP/MAS NMR ^ベクトルを示す図である。この図 5では、結合型別 の炭素種の相対的含量を知るために、スペクトルの δ 0-230の範囲を炭素種別の 7 領域に区分している。即ち、未置換脂肪族性 C (アルカン、脂肪酸) ( δ 0-50)、 Ν-ァ ルキル (アミノ酸、ペプチド、タンパク性) +メトキシ C ( δ 50-60)、脂肪族 C-0 (特に炭 化水素) ( δ 60-110)、芳香族 C ( δ 110-150)、フエノール性 C ( δ 150- 160)、カルボキ シル C ( δ 160-190)、ケトン C=0 ( δ 190-230)である。
[0041] スペクトル曲線と上記各領域の境界線および基線で囲まれた面積の全面積に対す る百分率を計算して相対的炭素含量を求めると、原料の違いによってフミン酸 (ΗΑ) およびフルボ酸 (FA)の分子構造に多少の違!、が存在する。
[0042] カナダ産草炭由来のフミン酸(CP— ΗΑ)は、風化炭由来のフミン酸 (WC— HA)よ り脂肪族 Cが多ぐ芳香族 Cが少ない。カナダ産草炭由来のフルボ (CP— FA)は、風 化炭由来のフルボ酸 (WC—FA)より脂肪族 C- Oが多ぐフエノール性 C、カルボキ シル Cおよびケトン c = oが少ない。また、カナダ産草炭由来のフミン酸、フルボ酸と もに、風化炭由来のフミン酸、フルボ酸に比べてアルキル +メトキシ基が多い。これは 、草炭の腐植ィ匕過程と石炭の酸ィ匕過程が異なることに起因するものと考えられる。

化ナトリウムを用いた。試料は溶離液に溶解したもの(O.lmg mL を 10 L注入し、 分画液の吸光度は Waters 490E (Millipore Co.)を用いて波長 280nmと 400nmで測定 した。その測定結果を、図 6に示した。
[0045] この図 6は、原かん水、排かん水、風化炭、カナダ産草炭、黒ぼく土の各原料から の抽出フルボ酸を対象とする物性評価分析結果をまとめた図面代用表である。この 図 6に示すように、原かん水、排かん水、風化炭、カナダ産草炭、黒ぼく土の各原料 力らの抽出フノレボ酸の分子量 ίま、川頁に 740、 810、 920、 780、 680であった。
[0046] 原料の異なる各フルボ酸の分子量、元素組成、水素および炭素の帰属結果を基に 、フルボ酸を一つの分子と仮定し、平均化学構造式を求めた。フルボ酸は混合物で あり絶対分子量の測定が難 U、ことから、同一フルボ酸の分子量も測定方法によって 異なるので、いまのところ正確な化学構造は存在しない。したがって、図 6中の推定 平均化学構造式(Average molecular formura)は、あくまでも原料の異なるフルボ酸 を区別するときに有用である。

[0048] フルボ酸のカルボキシル基の定量は、公知の酢酸カルシウム法 (Blom et al., 1957) に基づいて行った。その結果、草炭フルボ酸のカルボキシル基含有量は 3.98mmol/g であった。
[0049] (6)電子スピン共鳴(ESR)スペクトル分析。
[0050] フルボ酸粉末を直径 0.5mmの石英管に採り、電子スピン共鳴(ESR)装置(BRUKER , ESR300E)を用いて、フルボ酸に含まれるフリーラジカル濃度を測定した。決定は M n2+標準マーカーを用い、スピン濃度の測定は DPPH (1.1- Diphenn- 2picrylhydrazyl, 分子量 394,ESR 3.1 X 102°spins/g)を用いた。その結果、草炭フルボ酸のフリーラジ カル含有量は 7.27 X 1015spins/gであった。
実施例 3
[0051] <フルボ酸の抗アレルギー効果検証実験 >。フルボ酸の抗アレルギー効果の有無 およびその具体的な作用機序の解明を目的とするバイオアツセィを行った。まず、本 バイオアツセィに用いたカナダ産草炭由来のフルボ酸の特性について説明する。
[0052] まず、図 7は、以下のバイオアツセィで用いたカナダ産草炭フルボ酸(CP— FA)の 特性を示した図面代用表である。
[0053] カナダ産草炭フルボ酸 (CP— FA)の元素組成 (d.a.f.%)を図 7に提示する。炭素 )は 47.8、水素(H)は 4.6、窒素(N)は 0.3、酸素(0)は 47.3、フ ノール性水酸基(Ph- OH)は 0.75mmolg— カルボキシル基(COOH)は 3.98mmolg— 'であった。

[0056] この図 8からは、原料であるカナダ産草炭 (Peat)に比べて、そのフミン酸 (HA)とフ ルポ酸 (FA)が多くの画分に分解されていることがわかる。これは、抽出段階におけ る塩ィ匕水素および水酸ィ匕ナトリウムなどの分解作用によるものであると思われる。
[0057] また、草炭の水溶性成分であるフルボ酸 (FA)がアルカリ可溶酸不溶成分であるフ ミン酸 (HA)に比べて、アルキルとメトキシ基を多く持ち、芳香族性及び炭水化物含 有量が高いことがスペクトル力も読み取ることができる。以下、実施例 3に関するバイ オアッセィの手順にっ 、て説明する。

[0059] 抽出したフルボ酸の濃度が lmg mじ1になるように蒸留水に溶かし、 0.22 mのフィ ルター(Milliporejapan)で滅菌した。

[0061] 細胞はラット肥満細胞 RBL2H3細胞を用いた。 RBL2H3細胞は 10%ゥシ胎児血清(F BS)、 2mML— glutamine、 60mg L カナマイシン含 ¾ Minimum Essential Medium Eagl e (MEM, Nissui)で培養した。

[0063] 肥満細胞で起こる I型アレルギー反応において、抗原と抗体の結合はヒスタミンや b- へキソサミニダーゼなど細胞内顆粒流出の直接原因になることが知られている。この ため、抗原と抗体の結合におけるフルボ酸 (CP— FA)の影響を、 β —へキソサミニ ダーゼの遊離阻害率を測定することにより確かめた。
[0064] まず、上記した MEM培地で予め前培養してお!、た RBL2H3細胞を 5.0 X 105 cells m じ1になるように調製し、ここに lmg mじ1マウス抗 DNP IgE抗体(Sigma Co.)を最終濃 度 0.3 g mじ1になるように加えて、よくサスペンドした。

[0068] この図 9の縦軸は、 13 -へキソサミニダーゼの遊離阻害率(%)、横軸がフルボ酸(C P— FA)の濃度 g mじ1)をそれぞれ示している。図 9中の「Keto.」は、比較のた めの臨床薬として実用されて 、るケトチフェン (対照区)を示して 、る。
[0069] この図 9で示したように、フルボ酸(CP— FA)は、高い j8 -へキソサミニダーゼ遊離 阻害効果を示した。特に、添加量 10 g mじ1において、ケトチフェンの倍近い抗ァレ ルギー効果を示し、添加濃度がそれより高 、あるいは低 、場合でもケトチフェンと同 等かそれよりも高 、抗アレルギー効果を示した。
[0070] したがって、フルボ酸 (CP— FA)は、 13 -へキソサミニダーゼ遊離阻害において濃 度依存性が高く、 I型アレルギー反応の抗原感作段階を抑制する抗アレルギー剤とし て有効であることを確認できた。

阻害による細胞内顆粒の放出抑制効果を調べた。

[0076] 図 10は、前記吸光度測定の結果を示した図(グラフ)である。この図 10で示したよう に、フルボ酸 (CP— FA)に肥満細胞力 の j8 -へキソサミニダーゼの遊離阻害がみ られたが、対照区であるケトチフェンに比べてその程度は低ぐ濃度依存性は認めら れな力つた。このことから、 I型アレルギー反応の抗体感作段階においても、フルボ酸 (CP - FA)にある程度の抗アレルギー効果を期待できることが明らかになつた。
[0077] (5)細胞内カルシウムイオン (Ca2+)濃度の測定。
[0078] 細胞内 Ca2+濃度の測定は、 Calcium Kit - Fluo 3を用いて行った。 MEM培地で予 め前培養しておいた RBL2H3細胞を 1.5 X 105cellsmじ1になるように調製し、ここに lmg mじ1マウス抗 DNP IgE抗体(Sigma, Co.)を最終濃度 0.3 L mじ1になるようにカロえよ くサスペンドした。 37°C、 5%COインキュベーターにー晚おき、細胞感作行った。

[0081] 図 11は、フルボ酸の抗原刺激による Ca2+流入阻害に与える影響を調べた結果を示 す図(グラフ群)である。図 11の縦軸がコントロール (滅菌水添加)の細胞内 Ca2+濃度 1.0に対する、抗原刺激による Ca2+の細胞内流入率を、横軸が抗原刺激からの時間( 秒)の経過を示している。
[0082] フルボ酸添カ卩は、いずれも抗原刺激による Ca2+流入を阻害することが明らかになつ た。また、その活性は、 Ca2+流入阻害剤であるケトチフェン (対照区)と同等なものであ り、抗原刺激直後にその Ca2+流入阻害効果が働くことが確認された。フルボ酸の抗原 刺激による Ca2+流入阻害効果に濃度依存性は認められな力つた。
[0083] 以上の実施例の結果から、フルボ酸に高い抗アレルギー効果があることがわ力つた 。図 12は、フルボ酸の I型アレルギーの発生機序との関係を模式的に説明するため の図である。
[0084] フルボ酸は、 I型アレルギーの発生機序において、三つの段階において作用してい る。まず、第一には、図 12中に矢印 Xで示す抗原感作段階、第二には、図 12中に矢 印 Yで示す抗体感作段階、第三には、カルシウムイオン流入によって起こる細胞の脱 顆粒段階(図 12の矢印 Zに示す段階)ある。フルボ酸は、これらの段階を抑制又は遮 断することによって I型アレルギーの発症を抑制する。
産業上の利用可能性
[0085] 本発明は、 I型アレルギーの発症予防や防止に有用である。例えば、 I型アレルギ 一の発症予防や防止を目的とする経口投与剤、注射液、皮膚外用薬、貼薬などの
医薬品、医薬部外品、あるいは I型アレルギーの発症予防や防止に係わる機能を有 する入浴剤、洗顔料、クリーム、ノ¾クなどのスキンケア化粧品などの化粧品、清涼飲 料などの飲料、インスタント食品、飴やガムなどのお菓子類、パン類、麵類、ベビーフ ード、調味料などの飲食品などに広く利用できる。なお、フルボ酸は、 日本国厚生労 働省から食品添加物成分として認可されている (食安基発第 0601001号)。

Claims
請求の範囲
[I] 抗原感作段階又は Z及び抗体感作段階をフルボ酸によって抑制する I型アレルギ 一の抑制剤。
[2] 細胞の脱顆粒段階をフルボ酸によって抑制する I型アレルギーの抑制剤。
[3] 細胞へのカルシウムイオン流入を阻害することにより前記脱顆粒段階を抑制するこ とを特徴とする請求の範囲第 3項記載の I型アレルギー抑制剤。
[4] 前記フルボ酸は草炭から抽出されたものであることを特徴とする請求の範囲第 1項 力 請求の範囲第 3項のいずれか一項に記載された I型アレルギー抑制剤。
[5] 抗原感作段階又は Z及び抗体感作段階において I型アレルギーを抑制するフルボ 酸を少なくとも含む組成物。
[6] 請求の範囲第 5項記載の組成物を配合したことを特徴とする洗顔料。
[7] 請求の範囲第 5項記載の組成物を配合したことを特徴とする化粧品。
[8] 請求の範囲第 5項記載の組成物を配合したことを特徴とする飲食物。
[9] 請求の範囲第 5項記載の組成物を配合したことを特徴とする皮膚外用薬。
[10] フルボ酸を用いて特異的減感作を行うことにより I型アレルギーの発症を抑制する 方法。
[II] 抗原感作段階又は Z及び抗体感作段階をフルボ酸によって抑制することを特徴と する請求の範囲第 10項記載の方法。
[12] フルボ酸を用いて非特異的減感作を行うことにより I型アレルギーの発症を抑制す る方法。
[13] フルボ酸を用いて脱顆粒を阻害することにより I型アレルギーの発症を抑制すること を特徴とする請求の範囲第 12項記載の方法。
