Due to the chemical properties of the guanidinium groups, and in particular their high pKa, these cationic lipids are capable of protecting DNA molecules against degradation due to variations in pH. The results presented in the examples show that these compounds allow the transfection of numerous cell types, with great efficiency. The efficiency of the transfection depends in particular on the cationic lipid / DNA ratio in the complexes formed.

- Table 1 Expression of luciferase in different cell lines of mammals transfected with either BGTC, calcium phosphate or Transfectam ®.
- the BGSC compound being less soluble in an aqueous medium than the BGTC, the applicant used the first in the form of liposomes in the presence of a neutral lipid, DOPE (dioleoylphosphatidylethanolamine).
- DOPE dioleoylphosphatidylethanolamine
- the formulations are prepared in a 3/2 molar ratio. A second preparation this time containing
- BGTC was also prepared with DOPE under the same conditions.
- the Applicant first determined the lipid / DNA ratio during a transfection experiment on HeLa cells.
- the curve obtained is represented in FIG. 4.
- the transfection optima are obtained for BGSC / DNA liposome ratios between 1.5 and 5 with a center at 2.5-3. Since the guanidinium groups are protonated at neutral pH, the best BGSC-DOPE / DNA aggregates have a positive charge of approximately 3 (FIG. 4).
- the Applicant has carried out transfection experiments in different cell lines using formulations of cationic liposomes BGSC-DOPE and BGTC-DOPE in 3/2 molar ratios.
- the giianidinium charge ratio of lipids / DNA phosphates is approximately 2.5 (-3) for these two gene transfer systems.
- a transfection with Lipofectin® serves as a control. The results are set out in Table 2 below.
- Table 2 Expression of luciferase in different eukaryotic cell lines transfected using BGSC / DOPE liposomes. BGTC / DOPE liposomes and Lipofectin ®.
- transfection compositions based on cationic lipids exhibit reduced effectiveness in the presence of serum.
- the purpose of this example is to test the sensitivity of the compounds of the invention to the serum effect.
- the lipids described in the invention are dissolved in ethanol.
- solutions of “liposomes” are obtained in the presence of DOPE as follows: DOPE (AVANTI) in solution in chloroform is added to the solutions of lipid in ethanol, in a cationic lipid / DOPE molar ratio of 3/2 . After dry evaporation the mixtures are taken up in water and heated at 50 ° C for 30 minutes;
- the solutions are all adjusted to 10 mM in cationic lipid.
- the nucleic acid used is the plasmid pXL2774.
- the DNA is diluted to 20 ⁇ g / ml in 150 mM NaCl and the various formulations of cationic lipid are diluted in water to 40, 80,120 and 160 ⁇ M.
- the DNA and lipid solutions are mixed volume to volume, which gives ratios in nmoles / ⁇ g DNA of 2, 4, 6 and 8 respectively; the salt concentration is 75 mM
- the cells are cultured under appropriate conditions in 24-well microplates (2 cm 2 / well) and are transfected while they are in the exponential growth phase and at 50-70% of the confluence.
- the cells are washed with twice 500 ⁇ l of medium devoid of serum proteins and returned to growth either in a medium without serum [transfection in the absence of serum], or in a complete medium [transfection in the presence of serum] and 50 ⁇ l of cytofectant mixture [ 0.5 ⁇ g DNA / well] are added to the cells [3 wells / lipid-DNA condition].
- the growth medium is supplemented with the appropriate quantity of serum 2 hours after transfection.
- the transfection efficiency is evaluated by measuring the expression of luciferase according to the recommendations given for the use of the Promega kit [Luciferase Assay System].
- the toxicity of the cytofectant mixtures is estimated by measuring the protein concentrations in the cell lysates.
- Liposomes are more effective than those in the form of “micelles”. Furthermore, contrary to what is observed with most cytofectant preparations containing cationic lipids, no significant inhibitory effect linked to the presence of BGTC serum in the form of “liposomes” or “micelles” could not be demonstrated in our transfection conditions.
- X-gal cells are detected in the epithelium of the respiratory tract of mice treated 48 hours after transfection into the cells of the LacZ plasmid using the BGTC / DOPE liposomes. The results are presented in Figure 6.
- transfected cells are located in the trachea and only a few X-gal cells are seen in the secondary respiratory tract. This type of distribution has already been reported in other cell transfections.
- Transgenic expression is also demonstrated in the submucosal glands by immunoperoxidase staining with an antibody directed against the luciferase protein after transfection of p RSV-Luc with BGTC / DOP (FIG. 6D).
- Negative controls are performed without antibodies (Figure 6E). These results are of particular interest for gene therapy directed against muscovidosis. Indeed, to treat this pathology, it is necessary to target the submucosal glands which are the main site for the expression of CFTR in human bronchi.
- mice treated with BGTC / DOPE liposome and pRSV-Luc plasmid under the conditions described in Example 8, are removed 48 hours after transfection and the luciferase activity assayed according to the protocol described in material and method.
- Luciferase activity is detected systematically in tracheal homogenates but not in those of the lungs. This observation is in agreement with the previous results obtained during the examination of the distribution of Xgal positive cells. It is noted that the luciferase expression is directly linked to the expression plasmid since no activity could be detected in the control containing a LacZ plasmid. As a witness, a transfection of the plasmid without vector. No expression of luciferase was observed in this control test.
- BalbC mice aged 5 weeks are injected intravenously (tail vein) or intraperitoneally with 200 ⁇ l of transfectam mixture dissolved in 37.5 mM NaCl, 5% glucose and the expression of luciferase is sought after 24 hours post -transfection in the lung after intravenous injection, and in the liver and spleen after intraperitoneal injection.
- the organs are removed cold in lysis buffer [Promega El 53 A] supplemented with protease inhibitors [Boehringer 1697498] and homogenized with a Heidolph DIAX600 mill. Luciferase activity is sought in the supernatant 14000 g of the tissue extracts.
- Each mouse receives 50 ⁇ g of DNA complexed by different concentrations of
- BGTC in the form of "liposomes".
- the optimum ratio of cationic lipid nanomoles / ⁇ g DNA observed is 9 (FIG. 7); no toxicity could be demonstrated for doses up to 900 nanomoles of BGTC injected.
- BGTC Intraperitoneal injection: BGTC in the form of “liposomes” was injected with 100 ⁇ g of DNA per mouse. Both in the liver ( Figure 8B) and in the spleen ( Figure 8A), the maximum expression is obtained for 1.5 nanomoles BGTC / ⁇ g of DNA.
