本発明 はポ リ ペプチ ド分泌発現用 ベ ク タ ー 、 該ベ ク タ 一 で形質転換 し た微生物及び該微生物 に よ る ポ リ ぺプチ ドの製造に 関 す る
背 景 技 術
遺伝子 工学的手法を用 い て 、 イ ン タ ー フ ェ ロ ン 、 成長 ホルモ ン等を始め と す る様々 なポ リ ペプチ ドを大腸菌,、 枯草菌 、 酵母等の宿主細胞の利用 に よ り 製造す る方法 は 既 に 確立さ れて い る 。 し か し なが ら 確立さ れた 方法 と い え ど も未だ幾つ かの未解決の 問題点を有 し て お り 、 天然 品 と 全 く 同一 の ポ リ ペプチ ド を製造す る こ と は必ず し も 容易 で は な い 。 上記問題点の 中で最も重要な も の の ひ と つ と し て は 、 ポ リ ペプチ ド の N 末端を 、 天然品 と 周一の 構造 と す る の が困難な こ と を挙げる こ と がで き る 。 即 ち 之等の ポ リ ペプチ ドを コ ー ド す る遣伝子を宿主細胞内で 直接発現 さ せ る と き 、 遺伝子か ら ポ リ ペプ チ ド への 翻訳 は 、 通常開始 コ ド ン ( A T G ) か ら 始 ま る た め 、 発現さ
れるポ リ ペプチ ドの N 末端は 、 ホル ミ ルメ チ才ニ ン残基 と なっ て し ま う 。 天然型ポ リ ペプチ ドの N 末端が ホル ミ ルメ チ才ニ ンであ る場合は少な く 、 他の N 末端を有する ポ リ ペプチ ド の製造に は こ の方法は利用 で きない 。 上記 ホル ミ ル メ チ才ニ ン残基 は 、 シ ァ ノ ゲ ンブ ロ マイ ドを用 い た化学反応に よ り ポ リ ペプチ ドか ら 除去す る こ と が で きる が 、 ポ リ ペプチ ドが N 末端の他 に もメ チ才 ニ ン残基 を有す る場合、 上記方法で は ポ リ ペプチ ド鏆自体が該個 所で切断さ れて し ま い 、 や は り 目 的の ポ リ ペプチ ド は製 造で きな い 。
ま た 目 的ポ リ ペプチ ド を コ ー ドす る遺伝子を 、 他の ポ リ ペプチ ド を コ ー ド する遣伝子 と結合さ せて 、 融合ポ リ ペプチ ド と し て 発現させ る方法も知 ら れて い る の方 法で は 、 得 ら れる融合ポ リ ペプ チ ド か ら の 目 的 ポ リ ぺ プ チ ド の分離は 、 卜 リ プシ ン等の酵素に よ る処理や シ ァ ノ ゲ ンブ ロ マイ ド等を用 い た化学的処理 に よ っ て い る 。 し か し な が ら こ の方法 に お いて も 目 的のポ リ ペプチ ド鎖内 に使用 す る酵素又 は化学薬品 の標的 と な る ア ミ ノ 酸が存 在 す れば 、 そ の個所でペプチ ド鎖が切断さ れ 、 結局 目 的 ポ リ ベ プ チ ド は 製造 はで き な い 。 ま た 、 上記方法に よ り 融合ポ リ ペプチ ド か ら の 目 旳ポ リ ペプチ ド の分離が可能
な場合 と い え ども 、 目 的ポ リ ペプチ ド の単離のた め に は 通常菌体粗抽出液か ら 融合ポ リ ペプチ ド を分離す る 工程 及び融合ポ リ ペプチ ド を 切 断 し た反応組成物か ら 目 的ポ リ ペプチ ド を分離す る 工程の 2 回 の精製操作が必要と な り 、 そ の操作及び工程自体煩雑 と な り 、 し かも 目 的物 の 収率の低下 は避け ら れな い 。
以上の よ う に 、 天然型 と全 く 周一の ポ リ ペプチ ドを遣 伝子 工学的手法で入手す る こ と は 、 従来非常に 困難で あ り 、 こ の天然型 と 全 く 同一の ポ リ ペプチ ド を宿主細胞か ら 直接製造で き る改良さ れ た方法の研究開発が 、 斯界で 要望さ れて い る現状 に あ る 。
本発明 は 、 上記斯界で 要望 さ れて い る天然型 と 全 く 周 一 の ポ リ ペプチ ド を宿主細胞か ら 直接製造でき る改良さ れた 方法を提供す る こ と を 目 的 と す る 。 ま た本発明 は 、 該方法の実施の た め の新 し い ベ ク タ ー 及び該べ ク タ 一 を 保有す る微生物 を提供す る こ と を も そ の 目 的 と し て い る 発 明 の 開 示
本発明 に よ れば、 シ グナルペ プ チ ド を コ ー ド す る D N A 塩基配列 と 目 的ポ リ ペプチ ド を コ ー ド す る D N A 塩基配列 と を直接連結 さ せ て 目 的ポ リ ペプ チ ド を分泌発 現さ せ る た め の 、 シ グ ナルペプチ ド を コ ー ド す る D N A
塩基配列を含む こ と を特徴 と する ポ リ ペプチ ド分泌発現 用 ベ ク タ ー 、 シグナルペプチ ド を'コ ー ド す る D N A塩基 配列 と 目 的ポ リ ペプチ ドを:コ ー ド する D N A塩基配列 と が直接連結さ れ て な る融合ポ リ ペプチ ドを コ一 ド す る D N A塩基配列 を含む上記ベ ク タ ー 、 該ベ ク タ ーで形質 転換さ れた微生物及び該微生物 を培養 し て 分泌 さ れる ポ リ ペプチ ド を採取する ポ リ ペプチ ド の製造法が提供さ れ る 。

本発明者 ら は 、 上記シグナルペプチ ドの特性 に着目 し 該シグナルペ プチ ドを利用 し て 、 目 的 と する外来蛋白質 を シグナルペプチ ド と の融合ポ リ ペプチ ド と し て宿主賴 胞内で生産さ せるこ と に よ り 、 外来蛋 白質を細胞外に成 熟ポ リ ペプチ ド と して分泌させ 、 か く し て 目 的ポ リ ぺプ チ ド を容易 に 製造入手 し得る と の着想か ら鋭意研究を重 ね た 。 そ の結果 、 実際に宿主細胞内 に導入 する こ と に よ つ て 、 目 的ポ リ ペプチ ド を成熟ポ リ ペプチ ド と し て 細胞 外 に 分泌さ せ得る新 し い ベ ク タ ー ( プラ ス ミ ド ) 及ぴ該 ベ ク タ ー のた め の ポ リ ペ プチ ド分泌発現用 ベ ク タ ー の構 築に 成功 する と共 に 、 こ のベ ク タ ー の利用 に よ る 目 的ポ リ ペプチ ド の 製造 に成功 し た 。

上記の よ う に し て 本発明ベ ク タ ー を導入 し て形質転換 し た細胞を培養す る と き に は 、 細胞内で融合ポ リ ぺ プチ ド が生産さ れ、 続い て細胞外又 はペ リ ブ ラズム に成熟ポ リ ペプチ ド が分泌蓄積さ れる 。 即 ち 、 ま ず 、 ベ ク タ 一 中 の融 合ポ リ ペプチ ドを コ ー ドす る遺伝子か ら 、 ベ ク タ ー 中 の転写調節因子並び に 宿主細胞中の諸因子の作用 で m R N A が生産さ れる 。 次いで 、 m R N A か ら 翻訳調節 因 子並び に 宿主細胞中'の 諸々 因子の作用 で融合ポ リ ぺ プ チ ド が生産 さ れる 。 更 に こ こ で生産さ れる融合ポ リ ぺプ チ ド は 、 シ グ ナルペプチ ド の作用 に よ り 、 細胞外又 は ぺ リ ブラズム に 分泌 さ れ 、 同 時に シグナルぺプチダーゼ の 作用 に よ り 、 融合ポ リ ペプ チ ド か ら シ グ ナルペプチ ド が 切 り 離さ れる ので あ る 。 そ の結果、 シグナルペ プチ ドも ま た 他の如 何な る不要な ア ミ ノ 酸配列 を も 含 ま な い成熟 ポ リ ペプチ ド が細 胞外又 はペ リ ブ ラズ ム に 分泌 、 蓄積さ れる 。
か く し て分泌、 蓄積さ れた成熟ポ リ ペプチ ド は 、 こ れ を常法に従い分離す る こ と がで き 、 ま た更に精製す る こ と ができる 。 こ の分離、 精製操作 と し て は例 えば培養上 澄又は浸透圧シ ョ ッ ク 法 に よ り 調整 し た ペ リ プラズム画 分か ら 、 ゲル泸過 、 吸着ク ロ マ ト グラ フ ィ ー 、 イ オ ン交 換ク ロ マ ト グ ラ フ ィ ー 、 高速液体ク ロ マ ト グラ フ ィ ー 等 を適宜組合せた方法を採用 する こ と がで き る 。 特に本発 明 に従い得 ら れる 目 的ポ リ ペ プチ ド は 、 分泌さ れた も の であ る た め 、 そ の分離、 精製が比較的容易で ある利点が ある 。
発明を実施する た め の最良の形態
以下、 本発明 を更 に 詳 し く 説明 す る た め実施例 を挙げ る 。 尚 、 各例 に お いて 用 い ら れて い る各方法及び操作は 、 特に 明記 し な い 限 り 、 以下の通 り 行なわ れた も ので あ る 。 1 . 制 限酵素 に よ る D N A の切 断操作
D N A の水溶液 ( 又は緩衝液溶液 ) 或い は粉末 に 、 下 記第 1 表 に示 し た 各緩衝液の濃縮液及び水を混和 し 、 次 いで制 限酵素を加 え 、 3 7 °C の水浴中で 3 時間静 置 し て 反応さ せ る 。 制 限酵素の標準的使用 量 は、 D N A 1 g に対 し て 1 ユ ニッ ト で あ り 、 最終液量 は 1 0 ^ 2 と な る よ う に す る 。

2 . フ エ ノ ー ル抽 出法
酵素反応の終了後 、 酵素を失活さ せ反応を停止さ せる た め に こ の抽 出法を行な っ た 。 即 ち 、 反応液に 、 そ の 液 量の半量 と な る T E飽和 フ エ ノ ー ル ( 1 m D T A を含む 1 O m M 卜 リ ス塩酸 ( P H 8 . 0 ) 緩衝液を フ エ ノ ー ル に 飽和 さ せ た も の ) を加 え て充分混和 し た 後 、 周 じ く 半量の ク ロ 口 ホルム を加 え て 更 に 混和 し 、 次い で遠 心分離 し て D N Aの 含 ま れる緩衝液層を取る 。 更 に 0 . 1 倍量の 3 M酢酸ナ ト リ ウ ム緩衝液 ( P H 5 . 0 ) と 2倍量の冷エ タ ノ ール と を加 え て 混和 し て 、 一 2 0 で 1 時間 以上放置 し て D N Aを 沈殺 と し て 回収す る こ と に よ り フ エ ノ ー ルを 完全 に 除去 す る 。
3 . D N Aポ リ メ ラ ーゼ I ( ク レ ノ ー 断片 ) に よ る D N Aのプラ ン 卜 エ ン ド化方法
4 0 m M リ ン酸カ リ ウム ( P H 7 . 4 ) 、 6 m M塩化 マ グネ シ ウ ム 、 1 m M 3 —メ ルカ プ 卜 エ タ ノ ー ル、 1 m A T P及び各 1 m Mの d A T P、 d C T P、 d G T P及び d T T Pを 含む水溶液中 に 、 D N Aを溶か し 、 D N A I ^ g に対 し て 1 ユニッ ト と な る量の D N A ポ リ メ ラ ー ゼ I ( ク レ ノ ー 断片、 宝酒造㈱製 ) を加 え 、 1 2でで 3 0分間反応さ せ る 。
4 . 丁 4 0 八 リ ガ ーゼ に ょ る 0 八 断片の結合 ( 環状 化 ) 操作
6 6 m M 卜 リ ス塩酸 ( P H 7 . 5 ) 、 6 . 6 m M塩化 マグネ シ ウ ム 、 1 0 m Mジ チ 才 ス レ イ 卜 ー ル及び 1 m A T Pに 0 . 0 1 % の牛血清アルブ ミ ン を添加 し た水溶 液中で 、 D N A断片 と 、 そ の Ί g 当 り 3 ユ ニ ッ ト と な る量の T 4 D N A リ ガ ーゼ ( 宝酒造㈱製 ) と を 、 1 2 °C で 5 時間以上反応さ せ る こ と に よ り D N Aを結合 ( 環状 化 ) さ せる 。
5 . 形質転換方法
宿主細胞 と し て は 、 大腸菌 K 1 2株由 来の H B 1 0 1 株を用 い る 。
H B 1 0 1 株を 、 L B培地 ( Ί %パ ク 卜 卜 リ プ ト ン 、 0 . 5 %パ ク 卜 ィ ー ス 卜 エ キ ス 、 0 . 5 %塩化ナ 卜 リ ウ ム ) で 、 3 7 °C下 、 6 1 0 nmの吸光度が 0 . 2 5 に なる ま で増殖さ せ 、 培養液 4 0 πιδを遠心分離 ( 6 0 0 0回 転 Ζ分 X 1 0分 ) し て 菌体を 回収 し 、 次いで氷冷 す る 。 れを 0 . 1 Μ塩化マ グネ シ ウ ム 2 0 mfiで洗净 し 、 続い て 氷冷 し た 0 . Ί iVI塩化カ ルシ ウ ム及び 0 . 0 5 M塩化マ グネ シ ゥ ム 溶液 2 0 πιβに懸濁 さ せ 、 1 時間氷冷す る 。 遠 心分離 ( 6 0 0 0回転 Ζ分 X Ί 0分 ) 後 、 菌体を氷冷 し た 0 . 1 Μ塩化 カ ル シ ウ ム及び 0 . 0 5 Μ塩化マグネ シ ゥ ム 溶液 2 ιπβに再懸濁 させ る <_ の 懸濁液 0 . 212 に 、
T i D N A リ ガ ー ゼ を用 い て 結合 さ せ た D N A断片の反 応組成液 0 . 0 1 mfiを加 え 、 Ί 時間 氷冷する 。 次いで 4 2 . 5 Cの 水浴で 9 0秒間加 温 し 、 L B培地 2 . 8 in3 を加 え 、 こ れを 3 7 'Όの水浴中で 1 時間 静置する 。
次 に 、 得 ら れる形質転換株 を以下の 抗生物質耐性で選 択 す る 。 即 ち 、 Ί . 5 %寒天 を含む L Β培地 に ア ン ピ シ リ ン 5 0 , g / ιπδ又 は テ 卜 ラ サイ ク リ ン 2 0 ^ g Z maを 添加 し て 調整 し た 平板培地 に 、 上記で得 た 反応耝成液の 溶液各 0 . 3 mSずつ を 拡げ 、 こ れを 3 7 で一晩培養 し 生 育す る大陽菌 コ ロ ニ ー を分離す る
6 . プラ ス ミ ドの単離
プラス ミ ドを保有する菌株を 、 ア ン ピ シ リ ン 5 0 〃 g Ζ ιπδ又 は テ 卜 ラ サイ ク リ ン 2 0 ^ g / πιδを添加 し た し B 培地 5 0 0 msで 、 6 1 0 nmで の吸光度が約 0 . 6 に な る ま で 3 7でで振盪培養す る 。 次いで ク ロ ラ ム フ エ ニ コ ー ル 8 0 mgを加 え 、 3 7 °Cで 1 2〜 1 6時間振盪培養する 。 こ れを遠心分離 ( 6 0 0 0回転 /分 X 1 0分 ) し て 菌体 を集め 、 0 . 8 5 %塩化ナ ト リ ウ ム水溶液で洗浄する 。 菌体を 2 096蔗糖及び 5 0 m M 卜 リ ス 塩酸 ( P H 8 . 0 ) 溶液 2 . 5 ιδに 懸濁させ 、 次 に 1 % リ ゾチー ム を 含む 0 . 2 5 Μ 卜 リ ス塩酸 ( Ρ Η 8 . 0 ) 溶液 0 . 5 13を加 え 、 分間水冷する 。 更 に 0 . 2 5 M E D T A
( Η 8 . 0 ) 1 ιπδを加 え 、 Ί 0分'間氷冷 す る 。 次に 6 m Μ 卜 リ ス塩酸 ( Ρ Η 8 . 0 ) 、 6 0 m M E D T A及 ぴ 0 . 1 % 卜 リ 卜 ン X— 1 0 0 の溶液 4 mfiを加 える 。 こ れを超遠心 ( 2 5 0 0 0回転 /分 X 9 0 分 ) し て上清を 採取す る 。 こ の上清 8 . 2 msに 塩化 セ シ ウ ム 9 . 0 g を 加 え て 溶か し 、 次いで Ί % ェチジ ゥ ムブ ロ マ イ ド 溶液 0 . 8 ι3を加 え る 。 こ れを遠心分離 ( 2 0 0 0回転 ./分 X 1 0 分 ) し て 浮遊物を 除き 、 溶液を超遠心
( 5 0 0 0 0回転 . Z分 X 1 5 時間 ) する 。 次いで紫外線
照射 に よ り 螢光を発するプラ ス ミ ド部分を分離す る 。 こ れを 5 M塩化 ナ ト リ ウ ム溶液で飽和 し た イ ソ プ ロ パ ノ ー ルで 5 〜 &回抽出 し て.こ れか ら ェ チジ ゥ ムプ ロ マ イ ド を 除去 す る 。 最後 に 1 0 m M 卜 リ ス塩酸 ( P H 8 . 0 ) 及 び Ί ϋΐ Μ E D T A に対 し て透析 し て 塩化セ シ ウ ム を 除 去す る 。
7 . 才 リ ゴヌ ク レ オ チ ドの合成
才 リ ゴヌ ク レ オ チ ド の合成 は 、 下記 に示す 固相合成法

即 ち 、 ま ず 1 %架橋ポ リ ス チ レ ン樹脂 S — X Ί
( 2 0 0 〜 4 0 0 メ ッ シ ュ 、 パ イ オ ラ ド ラ ボ ラ 卜 リ ー ズ 社製 ) を ァ ミ ノ メ チル化 し た も の と 、 5 ' — 0 — ジ メ 卜 キ シ 卜 リ チルヌ ク レ オ シ ド の モ ノ コ ハ ク 酸エ ス テル と を 反応 さ せ て 、 ヌ ク レ オ シ ド担持樹脂を得る 。 次 に 、 バ ー チ ェ ム社製 D N A合成機を用 い て 以下の操作を行な う 。
上記樹脂 4 O mgを反応管 に 入れ 、 Ί Μ臭化亜鉛のジ ク 口 口 メ タ ン 一 イ ソ プ ロ パ ノ ー ル ( 8 5 : 1 5 ) 溶液を 用 い て 5 ' 位の ジ メ 卜 キ シ 卜 リ チル基を脱離さ せ る 。 次 に 完全 に 保護 さ た ジ ヌ ク レ オ チ ド ( C . B roka et a I
N ucleic A cids R esearch, 8 , 5 4 6 1 - 5 4 7 1 ( 9 8 0 ) の方法 に よ り 調製 し た 〕 の 卜 リ エ チルアン モ ニ ゥ ム塩 5 0 mgを加え 、 縮合剤 ( メ シ チ レ ン スルホ 二 ル ー 5 — 二 卜 口 卜 リ アゾー ル ) を用 い て縮合させる 。 以 上の操作を繰返 し て 、 頫次鎖長をのば し て 、 保護さ れた 才 リ ゴヌ ク レ オ チ ドを担持 し た樹脂を得る 。 尚 、 最後の 縮合工程で は 、 必要に応 じ て ジヌ ク レ オチ ド の代 り に 、 前記文献に記載の方法に よ り 調製さ れる モ ノ ヌ ク レ オ チ ド の 卜 リ エ チル ア ン モ ニ ゥ ム塩 2 5 mgを使用 す る 。

次に 単一の無色プラ ー ク をパ ス ツ ール ピ ぺッ 卜 に て取 り 出 し 、 こ れ と J M 1 0 3株の培養液 0 . 0 1 と を 、 2 Χ Τ Υ培地 1 in8に加 え 、 約 5 時間 、 3 7 Χίで振盪培養 し て 耝換え フ ァ ー ジ を増殖さ せ 、 培養後、 遠心 に て菌体 を 除き 、 上清に 2 0 %ポ リ エチ レ ング リ コ ール 6 0 0 0 の 0 . 2 πώを混合 し 、 室温で 1 5 分以上静置 し た後、 遠 心 に て沈濺する フ ァ ー ジ を集め 、 フ エ ノ ール抽 出 に よ つ て 、 フ ァ ー ジ か ら 一本鎖 D N Aを抽 出 し 、 こ れを铸型一 本鎖 D Ν Α と し て 用 いる 。
錶型一本鎖 D N Aプ ラ イ マ ー ( 宝 酒造㈱製 、 M 1 3 の 1 5塩基プライ マ ー ) と のそれぞれ 0 . 5 P mo I ずつ を 混合 し 、 6 0 °Cで 2 0分間熱処理後 、 徐冷す る 。 。 次 に こ の混合液に α 3 2 Ρ - d C T P ( アマ シ ャ ム社製、 4 0 0 C i / m mo I ) 2 ^ S と D N Aポ リ メ ラ ー ゼ I ( ク レ ノ ー 、 宝酒造㈱製 ) 2 ユ ニ ッ ト と を加 え 、 充分に 混合 し た後 、 そ の 3 . 2 , " 2 ずつ を 、 下記第 2 表 に示 し
た 4種の(I N T P— ddN T P混合液のそれぞれ を 含む反応管 に加 える 。 室温で 2 0分間反応さ せ た後、 チ エ ー ス反応液 ( d A T P、 d C T P、 d G T P及び d T T Pの各 1 m ) の をそ れぞれ に加 え 、 更 に 2 0分間反応さ せる 。 ホルム ア ミ ド停止液 ( 9 5 % v / ホル ム ア ミ ド 、 0 . 1 % キ シ レ ン シ ァ ノ ー ル及び 0 . 1 %ブ ロ ム フ エ ノ ールブルー ) を ずつ加え 、 9 5 eCで 3分間加熱 し た 後、 急冷す る 。 次に サ ンプル 2 2 ずつ を 、 6 %又は 8 % ポ リ ア ク リ ルア ミ ドゲル に よ り 電気泳動 ( 1 8 0 0 、 3 0 111 八 、 2〜 3 時間 ) さ せ る 。 泳動後 、 ゲルを 紙 ( ヮ ッ 卜 マ ン 3 M M ) に移 し 、 ゲル乾燥器 に て 乾燥 し 、 ォ ー 卜 ラ ジ オ グラ ム を と り 、

③ 上記②で得た D N A断片を 、 上記①で調製さ れた リ ン 酸化 し た オ リ ゴヌ ク レ オ チ ド 〈 2 〉 及び 〈 4 〉 並び に リ ン酸化 し て いない 才 リ ゴヌ ク レ オ チ ド < Ί 〉 及び 〈 3 〉 のそ れぞれ約 ずっ と 合せ て 、 D N A リ ガ ー ゼ で結合反応さ せ た 。 反応終了後 、 こ の反応組成液で大腸 菌 K 一 1 2株由 来の Η Β Ί 0 1 株を形質転換さ せ た 。 得 ら れた テ 卜 ラサイ ク リ ン耐性を示す形質転換株の 中か ら 1 株を 選び 、 こ れか ら プ ラ ス ミ ド を 単離 し 、 目 的 と す る P G H 5 3 を得 た 。

① 上記 ( A ) で得た P G H 5 3 の 1 0 9 を制 眼酵素 N ael ( N E B社製 ) 及び A val ( 宝酒造㈱製 ) を用 い て 中塩濂度緩衝液中で切 断 し 、 次いで 1 . 0 %ァ ガ ロ ー スゲル電気泳動 を行なっ て 、 約 2 . 2 2 kbの D N A断片 《 A》 を分離 し た 。
し の 断片 は 、 合成オ リ ゴヌ ク レ オ チ ド 由 来の D M A配 列 の大部分 と プ ラ ス ミ ド の 複製開始領域を含ん で い る 。
② P B R 3 2 2 を制 限酵素 A val 及び H ind ffl ( いず れ も宝酒造㈱製 ) で 、 中塩濃度緩衝液を用 い て 切 断 し 、
1 . 0 % ァ ガ ロ ー スゲル電気泳動を行なっ て 、 約 1 . 4 〇 kbの D N A断片 《 B 》 を得 た 。
こ の 断片 に は 、 テ 卜 ラ サイ ク リ ン耐性遺伝子のプ ロ モ 一タ ー の一 部 と テ 卜 ラ サイ ク リ ン耐性の 構造遺伝子 の 全 て が 含ま れて いる 。
③ P B R 3 2 2 の 2 0 g を制 限酵素 F nu4 H I
( N E B社製 ) で低塩濃度緩衝液を用 い て切断 し 、 次い で S 1 ヌ ク レ ア ーゼ に よ り D N A断片末端の突出塩基を 分解除去 し た 。 こ れ は フ エ ノ ール抽出後の D N Aを 、 6 m M酔酸ナ ト リ ウ ム 、 4 0 m M塩化ナ 卜 リ ウ ム及び 1 m M硫酸亜鉛緩衝液 ( P H 4 . 5 ) 1 mSに 溶か し 、 こ れ に 2 0 0 0 ユニッ ト の S 1 ヌ ク レ ア ー ゼ ( B R L社製 ) を加 え て 2 0 で 3 0分間反応させ る こ と に よ り 行なつ た 。 次いで 、 フ エ ノ ール抽出後の 、 D N Aを制 眼酵素 H ind IEで 中塩寢度緩衝液を用 い て 切 断 し 、 6 %ポ リ ア ク リ ル ア ミ ドゲル電気泳動 を行な い 、 約 0 . 2 8 kbの D N A断片 《 C》 を得た 。

④ 上記で得た 3 つ の 断片 《 八 》 、 《 B 》 及び 《 C》 を 、 T 4 D N A リ ガ ーゼを用 い て結合させた 。 反応後 、 こ の 反応組成液で H B 1 0 1 株を形質転換 し た 。 得 ら れ fcテ 卜 ラ サイ ク リ ン耐性を示す形質転換株の 中 か ら 1 株を選 びプ ラ ス ミ ド を 単錐 し た 。 かく し て p G H 5 4 を得た 。

れも宝酒造㈱製 ) で切 断 し 、 フ エ ノ ール抽出後 、 D N A ポ リ メ ラ ーゼ I ( ク レ ノ ー 断片、 宝酒造㈱製 ) で切断断 片をブラ ン 卜 エ ン ド化 し た 。 次 に 1 . 096ァ ガ ロ ー スゲ ル電気泳動で約 3 . 7 2 の 0 八 断片を分離 し 、 し の 断片を T 4 D N Aリ ガ ーゼで環状化さ せ た 。 反応終了後 こ の反応組成液で H B 1 0 1 株を形質転換 し 、 得 ら れる ア ン ピ シ リ ン耐性及びテ 卜 ラサイ ク リ ン耐性を示す形質 転換株の 中 か ら 一株を選択 し て プラ ス ミ ドを単離 し p B R H 0 2を得た 。 得 ら れた P B R H 0 2 は 、 P B R 3 2 2 と は異なっ て 、 A valで も Ρ νιιϋでも切 断さ れな かっ た 。

合さ せた 。 反応後、 こ の反応組成液で H B 1 0 1 株を形 質転換 し 、 得 ら れる テ 卜 ラサイ ク リ ン耐性の形質転換株 の中か ら一株を選び、 プラ ス ミ ド p U G 2 0 1 を単離 し た 。

衝液 ( P H 8 . 0 ) 8 0 ιηδに懸濁 さ せ 、 0 . 2 5 Μ E D T A水溶液 ( Ρ Η 8 . 0 ) の Q . 3 2 m2を加 え 、 ロ ー タ リ ー シ ェ ー カ ー で 2 4で に て 1 8 0回転 /分で 1 0分間攬拌 し た 後 、 遠心分離 ( 9 0 0 0回転 /分 X 1 0分 ) し て菌体を集め 、 次いで こ の菌体を氷冷 し た水 8 012に 再懸濁 さ せ 、 氷中 に 1 0分間 静置 し て 時々 攬拌 し 、 遠心分離 ( 9 0 0 0回転 /分 X 1 0分 ) に よ り 菌体 と 上澄 と を分離 す る こ と に よ り 行なっ た も ので あ り 、 得 ら れ た 上澄が ペ リ ブラ ズム画分で あ る
次い で 、 上記ペ リ プ ラ ズ ム 画分を抽 出 し た後の 菌体を 前記 と 同一 の洗净緩衝液で洗净後 、 P B S ( 1 5 0 m M 塩化 ナ ト リ ウ ム を 含む 2 0 m M リ ン酸ナ ト リ ウ ム 、 p H 7 . 0 ) 6 に 懸濁 さ せ 、 迢音波破砕機 ( 大岳製作所製 5 2 0 2 型 ) を用 い て 出力 1 0 0 Wに て 3 0秒ず つ 3 回 超音波破砕処理 し 心分離 ( 1 8 0 0 0回転 Ζ分 X
2 〇 分 ) し て 上澄 を得た れを細胞内 画分 と し た
( C ) ラ ジ オ ィ ム ノ ア ッ セ ィ に よ る 3 ゥ ロ ガ ス 卜 ロ ン の 測定
上記 ( Β ) で 得た そ れぞれの 分 につ き 、 以下の通 り 3 ゥ ロ ガ ス 卜 ロ ン の 存在 を 、 ゥ ロ ガ ス 卜 ロ ン特異 ラ ジ オ イ ム ノ ア ツ.セ ィ に よ り 検討 し た 。 ラ ジ オ ィ ム ノ ア ッ セ
ィ の方法は次の通 り である 。 即 ち 、 精製 ヒ 卜 iS ゥ ロ ガス 卜 ロ ンを抗原 と し て 、 家兎を免疫 し 抗血清を作成 し た 。 3 ゥ 口 ガ ス 卜 ロ ン を蒸留水 0. 2 in2に 溶解後 5 0 % ポ リ ビ ニル ピ ロ リ ド ン液 1 . 5 ιπδを加 え室温で 2 時間撹拌 し た 。 コ ンプ リ ー ト · フ ロ イ ン ド ♦ ア ジ ュバ ン 卜 2 . Ο πιδを加 え て 乳化 し 、 家兎 3 匹の胸部に 皮下注射 し た 。 2週間毎に免疫を 4 回 く り 返 し た後、 さ ら に 5 0 g の抗原を静注 し 、 3 日 後 に全採血を行ない 、 血清を 分離 し た 。
次 に ア ツ セ ィ に 用 いる抗血清の希釈倍率を求める タ イ 卜 レ ー シ ヨ ン カ ー プ 、 ア ツ セ ィ 条件を最適化す る た め ィ ンキ ュ ベ ー シ ヨ ン 時間 、 抗体結合標識抗原 ( バ ウ ン ド ) と 遊離標識抗原 ( フ リ ー ) の分離方法等の検討を加 え 、 下記測定条件を設定 し た 。
即 ち 、 0 . 5 % ゥ シ血清 アルブ ミ ン ( B S A ) 、
1 4 0 m M塭化ナ ト リ ウ ム 、 2 5 m M D T Aニ ナ 卜 リ ウ 厶 を含む 1 0 m M リ ン酸緩衝液 ( P H 7 . 4 ) を希 釈液 と し て用 い 、 該希积液 4 0 0 ^ 2 、 測定試料又 は標 準 ヒ 卜 3 ゥ ロ ガ ス 卜 ロ ン 1 0 0 S 及び抗 ヒ 卜 3 ゥ ロ ガ ス 卜 ロ ン血清 1 0 0 ^ 2 を加 え て 4 °Cに て 2 4 時間 イ ン キ ュ ベ 一 卜 し た 後 、 1 2 5 I 標識 ヒ 卜 3 ゥ ロ ガ ス 卜 ロ ン
1 0 0 X 2 ( 約 5 〇 0 〇 cpm ) 加 え た 。 更 に 4 °C に て 4 8 時間 イ ン キ ュ ベ ー 卜 し fc後 、 第 2 抗体 ( 抗家兎 ァ ー グ ロ プ リ ン ャ ギ血清 ) ( 1 : 2 0 ) 1 0 0 ^ 2 、 正常家 兎血清 ( 1 : 2 0 0 ) 1 0 0 δ 及び 5 % ポ リ エ チ レ ン グ リ コ ールを含む 1 0 m M P B S 液 9 0 0 〃 2 を加 え て 4 °C に て 3 時間 イ ン キ ュ ベ ー ト し た 。 次 に 3 0 0 0 rpm で 3 0 分間遠心分離 し 、 上清を除き沈瀕物を カ ウ ン 卜 し た 。 標準 ヒ 卜 3 ゥ 口 ガ ス 卜 ロ ン よ り 得 ら れた 標準曲 線 よ り 試料中の ヒ 卜 3 ゥ ロ ガ ス 卜 ロ ン免疫活性物の含量 を 求め た 。
上記結果を下記第 5 表 に 示す 。 ま た 第 5 表に は 、 P U G 2 0 1 を保有す る H B 1 0 1 株に 代え て 、 周様 に し て 作成さ れた P G H 5 5 及び p U G 3 のそれぞれで形 質転換 さ れた H B 1 0 1 株を用 い て 同様 に し た結果を併 記 す る 。

( U G 2 0 ) 2 6 3 0 ._6 2一 上記第 5表 よ り 、 シ グナルペプチ ドの みを コ ー ド す る D N A塩基配列 を含むベ ク タ ー を保有する大腸菌 ( Η Β 1 0 1 ( G Η 5 5 ) ) 及び ゥ ロ ガ ス ト ロ ン の みを コ - ド す る D M A塩基配列 を含むプラ ス ミ ド を保有す る大 腸菌 ( H B 1 0 1 ( U G 3 ) で は 、 そ れ ら の培養抽 出液中 に 3 ゥ ロ ガ ス 卜 口 ン の免疫活性物質は実質的 に 検 出 さ れない が 、 シ グナルペプチ ド と 3 ゥ ロ ガ ス 卜 ロ ン の 融合ポ リ ペ プチ ド を コ ー ド す る D N A塩基配列 を含み 、 そ の前に プ ロ モ ー タ ー 及び リ ボゾー ム結 合部位が連結さ れた 本発明 の成熟 ポ リ ペプチ ド分泌発現ベ ク タ ー を 保有 す る大腸菌 ( H B 1 0 1 ( U G 2 0 1 :) で は 、 そ の
培養抽出液中 に 顕著な 5 ゥ ロ ガ ス 卜 口 ンの免疫活性が検 出 さ れる こ と が 明 ら かで あ る 。
ま た 本発明ベ ク タ ー で形質転換 し た上記微生物の培養 で は 、 3 ゥ ロ ガ ス 卜 ロ ンの免疫活性物質の殆ん ど す ベ て ( 9 9 . 8 % ) が 、 ペ リ ブラズ ム に 分布 し て い る こ と も 確認さ れる 。 こ の こ と か ら 、 シグ ナルペプチ ド と 3 ゥ ロ ガ ス 卜 ロ ン と の融合ポ リ ペプ チ ドの D N A 塩基配列 を利 用 し た 本発明 に よ れば、 /3 ゥ ロ ガ ス 卜 ロ ン は細胞膜を通 過 し て ペ リ プラ ズム に分泌さ れる こ と が判る 。
( D ) ポ リ ペプチ ド の精製
上記ペ リ ブラ ズム抽 出液中 の 3 ゥ 口 ガ ス 卜 ロ ン の免疫 活性物質を 、 プチル 卜 ョ パ ー ル ( 東洋曹達㈱製 ) を用 い た 吸着 ク ロ マ ト グ ラ フ ィ ー 、 C M — 卜 ョ パ ー ル ( 東洋曹 達株式会社製 ) を用 い た イ オ ン交換 ク ロ マ ト グラ フ ィ ー P e p R P C カ ラ ム ( フ ア ルマ シ ア社製 ) を用 い た 髙速液 体 ク ロ マ ト グ ラ フ ィ ー 操作 に よ り 、 そ れぞれ単一 の ポ リ ペプチ ド に なる ま で精製 し た 。
そ の結果、 精製さ れた ポ リ ペ プチ ド は 、 ポ リ ア ク リ ル ア ミ ドゲル電気泳動 、 S D S — ポ リ ア ク リ ル ア ミ ド ゲル 電気泳動 、 ア ミ ノ 酸分析 、 N 末端分析の す べ て に お い て ヒ 卜 尿 よ り 単離精製さ れた 3 ゥ 口 ガ ス 卜 ロ ン と 周一物質
であ る こ とが確認 された
